Abstract

Breast cancer that overexpresses Human Epidermal Growth Factor Receptor 2 (HER2+) due to gene amplification or overexpression constitutes 15-20% of all breast cancer cases. Trastuzumab, the first FDA-approved monoclonal antibody targeting HER2, serves as the standard first-line treatment for HER2-positive advanced breast cancer, as recommended by multiple clinical guidelines.Currently, accumulated clinical evidence reveals a considerable degree of variability in the response of HER2+ breast cancer to trastuzumab treatment. Specifically, over 50% of patients either do not respond to or develop resistance against trastuzumab.The specific mechanisms of resistance to trastuzumab are currently unclear. This paper aims to review the existing research on the resistance mechanisms of trastuzumab, based on its target, from aspects such as genetic loci, molecular structure, signaling pathways, and the tumor microenvironment and to outline current research progress and new strategies. Abstract

Arbuscular mycorrhizal fungi (AMF) play important roles in the dynamics of soil organic carbon (SOC), as they can promote its accumulation and the formation of soil aggregates, thereby increasing soil carbon storage. However, the impact of carbon input through AMF inoculation on SOC sequestration is still unclear. In this study, the effects of AMF on photosynthetic carbon transport and SOC accumulation in two types of black soils with either high or low SOC soils were analyzed by an outdoor pot experiment using isotope 13C labeling, thus, revealing the mechanism of action of AMF in stabilizing SOC fixation. The results showed that AMF symbiosis increased the allocation of photosynthetic carbon to the roots of the maize plant and soils. Inoculation with AMF also increased the proportions of soil macro-aggregates and the soil microbial biomass carbon content in low SOC soil, promoted the accumulation of soil aggregates, and enhanced the chemical composition of SOC. After returning the harvested labeled straw to the original pots the following year after planting, inoculation with AMF was found to increase the contents of hemicellulose and lignin at the time when maize kernels attained a plump appearance. AMF significantly increased glomalin-related soil protein in high SOC soil. In addition, AMF had a promoting effect on the decomposition of cellulose, hemicellulose, and lignin in the straw, which could subsequently increase the accumulation of carbon. 36. Influence of exogenous 24‐epibrassinolide on improving carotenoid content, antioxidant capacity and gene expression in germinated maize seeds.

Abstract

Background: Carotenoids have various physiological functions, such as immune regulation and cancer prevention. Germination could further improve the content of carotenoids in maize seeds. In this study, yellow maize seeds (Suyu 29) were soaked and germinated with different concentrations of 24‐epibrassinolide. The changes of germination percentage, sprout length, bioactive components, antioxidant capacity and carotenoid content of the maize seeds were analyzed. Additionally, the relative expression of key genes in the carotenoid synthesis pathway was investigated. Results: The results showed that the sprout length, germination percentage, soluble protein, free amino acids, proline, endogenous abscisic acid, vitamin C, total phenolics and carotenoids displayed a significant increasing trend compared with the control group (P < 0.05). The activity of superoxide dismutase and peroxidase increased by 55.1% and 58.5% versus the control group, and the antioxidant capacity of 2,2‐diphenyl‐1‐picrylhydrazyl, 2,2‐azinobis(3‐ethylbenzothiazoline‐6‐sulfonic acid) and ferric reducing antioxidant power was 19.8%, 13.4% and 44.1% higher than that of the control group (P < 0.05). Compared with the control group, the expression of genes was significantly up‐regulated (P < 0.05). Under the treatment of 0.1 mg L−1 of 24‐epibrassinolide, carotenoid content reached the highest value. The carotenoids showed a positive correspondence with antioxidant enzyme activity, antioxidant capacity and total phenolics content (P < 0.05). Conclusion: This study showed that 0.1 mg L−1 of exogenous 24‐epibrassinolide promoted the accumulation of carotenoids and improved the antioxidant capacity and the quality of germinated maize seeds.
