Abstract

Osteosarcoma, one of primary bone tumor in children and young adults, has poor prognosis and drug resistances to chemotherapy. In order to reinforce the conventional therapies and antagonize the osteosarcoma in patients, a novel strategy is required for developing a new treatment. In this study, surfactin, a natural product from Bacillus subtilis, showed the efficiency of cell death in osteosarcoma, but not in normal cells. Surfactin triggers ER stress mechanism by promoting the aberrant Ca2+ release from ER lumen and ER‐signaling to mitochondrial dysfunction following caspases activation mediating cell apoptosis. Surfactin‐induced ER stress not only upregulated of glucose‐regulated protein 78/94 and IRE1‐ASK1‐JNK pathway but also leading to calpains and Bcl‐2 proteins family involving the release of cytochrome c. The releases into cytosol trigger the cleavage of caspase‐9 and caspase‐3 to induce cell apoptosis. In this study, surfactin demonstrated the potential functions to trigger the ER stress, ER stress‐associated IRE1‐ASK1‐JNK signaling pathway, mitochondrial dysfunction, and caspase activations leading to programmed cell apoptosis. Importantly, implicating the signaling pathway that regulates the connection between ER stress and mitochondrial dysfunction causing apoptosis associated with surfactin. Abstract

Mineral trioxide aggregate (MTA) is well known as an effective root canal filling material for endodontics therapy. Within MTA, bismuth oxide (Bi2O3) serving as the radiopacifier still has biocompatibility concerns due to its mild cytotoxicity. In the present study, we tried to modify the Bi2O3 radiopacifier by doping hafnium ions via the sol-gel process and investigated the effects of different doping ratios (Bi2-xHfxO3+x/2, x = 0–0.3) and calcination temperatures (400–800 °C). We mixed various precursor mixtures of bismuth nitrate (Bi(NO3)3·5H2O) and hafnium sulfate (Hf(SO4)2) and controlled the calcination temperatures. The as-prepared Hf-doped Bi2O3 radiopacifier powders were investigated by thermogravimetric analysis (TGA), X-ray diffraction (XRD), field-emission scanning electron microscopy (FESEM), and transmission electron microscopy (TEM). Portland cement/radiopacifier/calcium sulfate (75/20/5) were mixed and set by deionized water (powder to water ratio = 3:1). Changes in radiopacity, diametral tensile strength (DTS), and in vitro cell viability of the hydrated MTA-like cement were carried out. The experimental results showed that the group containing radiopacifier from sol-gelled Bi/Hf (90/10) exhibited significantly higher radiopacity (6.36 ± 0.34 mmAl), DTS (2.54 ± 0.29 MPa), and cell viability (84.0±8.1%) (p < 0.05) when compared to that of Bi/Hf (100/0) powders. It is suggested that the formation of β-Bi7.78Hf0.22O12.11 phase with hafnium addition and calcining at 700 °C can prepare novel bismuth/hafnium composite powder that can be used as an alternative radiopacifier for root canal filling materials. 41. The preferential accumulation and the settling velocity of small heavy particles in Taylor–Couette flows

Abstract

In a gap between two concentric cylinders, we discover that the small heavy particle settling velocity (Vs) is greater than its associated terminal velocity (Vt) in featureless turbulence that is generated by counter-rotating inner and outer cylinders simultaneously. The peak enhancement of Vsoccurs at St× (Vt/u′) ∼ 1, where u′and Stare, respectively, the r.m.s. turbulence fluctuation velocity and the Stokes number defined as the ratio between the particle response time to the Kolmogorov turbulence time. The present (Vs− Vt)/u′ data as a function of St measured by the high-speed particle imaging velocimetry are compared with previous experimental results obtained from different turbulent flow configurations in an attempt to gain further understandings of the complicated interactions between small heavy particles and featureless turbulence with negligible mean velocities. Moreover, it is found that small heavy particles are preferentially accumulated around the edges of an array of counter-rotating turbulent Taylor vortices with the inner cylinder rotation alone when a time ratio between particle response and Taylor vortices is near unity. These experimental results are interesting that may deserve to disseminate in our fluid mechanics community.

This catalytic effect also makes possible its cytotoxicity toward microorganisms at room temperature. To study this cytotoxicity, we synthesized the doped and undoped ceria particles of 8–9 nm in size using an inexpensive precipitation method and evaluated their disinfecting aptitudes with the turbidimetric and plate count methods. Among the samples being analyzed, the silver-doped ceria exhibits the highest sterilization ability, yet the undoped ceria is the most intriguing. The disinfection effect of undoped ceria is moderate in magnitude, demanding a physical contact between the ceria surface and bacteria cell wall, or the redox catalysis that can damage the cell wall and result in the cell killing. Evidently, this effect is short-range and depends strongly on dispersion of the nanoparticles. In contrast, the disinfection effects of silver-doped ceria reach out several millimeters since it releases silver ions to poison the surrounding microorganisms. Additionally, the aliovalent silver substitution creates more ceria defects. The synergetic combination, silver poisoning and heterogeneous redox catalysis, lifts and extends the disinfecting capability of silver-doped ceria to a superior level.
