Urothelial damage and barrier dysfunction emerge as the foremost mechanisms in Hunner‐type interstitial cystitis/bladder pain syndrome (HIC). Although treatments aimed at urothelial regeneration and repair have been employed, their therapeutic effectiveness remains limited due to the inadequate understanding of specific cell types involved in damage and the lack of specific molecular targets within these mechanisms. Therefore, we harnessed single‐cell RNA sequencing to elucidate the heterogeneity and developmental trajectory of urothelial cells within HIC bladders. Through reclustering, we identified eight distinct clusters of urothelial cells. There was a significant reduction in UPK3A+ umbrella cells and a simultaneous increase in progenitor‐like pluripotent cells (PPCs) within the HIC bladder. Pseudotime analysis of the urothelial cells in the HIC bladder revealed that cells faced challenges in differentiating into UPK3A+ umbrella cells, while PPCs exhibited substantial proliferation to compensate for the loss of UPK3A+ umbrella cells. The urothelium in HIC remains unrepaired, despite the substantial proliferation of PPCs. Thus, we propose that inhibiting the pivotal signaling pathways responsible for the injury to UPK3A+ umbrella cells is paramount for restoring the urothelial barrier and alleviating lower urinary tract symptoms in HIC patients. Subsequently, we identified key molecular pathways (TLR3 and NR2F6) associated with the injury of UPK3A+ umbrella cells in HIC urothelium. Finally, we conducted in vitro and in vivo experiments to confirm the potential of the TLR3–NR2F6 axis as a promising therapeutic target for HIC. These findings hold the potential to inhibit urothelial injury, providing promising clues for early diagnosis and functional bladder self‐repair strategies for HIC patients. © 2024 The Pathological Society of Great Britain and Ireland. Purpose: To design a quick checklist for urodynamic study (UDS), aiming to reduce the occurrence of errors in the process, which may help to increase the quality of UDS. And further to analyze the effectiveness of this quick checklist for UDS quality control. Methods: First, a quick checklist for uroflow study and pressure‐flow study was developed, based on the International Continence Society‐Good Urodynamic Practice standards, our previous studies, and recent literature, as well as expert suggestions. Then, patients who underwent UDS between January 2023 to February 2023 were randomly assigned to a study group or a control group. For the study group, the quick checklist was used throughout the UDS process, while the control group did not. The main artefacts were chosen to verify the effectiveness of the quick checklist for improving the UDS quality. Results: The quick checklist comprised three subtypes: checklist for patients, checklist for environment and device, and checklist for UDS test process. 38 UDS traces per group were included. The incidence of missing the standard cough test decreased significantly from 18.4% to 0 (p = 0.012), with the checklist implementation. The baseline drift frequency rate also declined significantly from 39.5% to 5.3% (p < 0.05). Volume < 150 mL on uroflow study occurred in 68.4% of cases and its frequency rate decreased significantly with checklist implementation (p < 0.05). Conclusion: A quick checklist for quality control of UDS was developed. The quick checklist as a convenient, quick, and easy used urodynamic quality control method, may help to reduce the technical artefacts and improve fundamental urodynamic quality control. To control the meshing performance of hypoid gear drive, current local conjugate theory and local synthesis method establish a relationship between the relative motion and local geometrical properties of the mating surfaces at one reference point. Theoretically, both local conjugate theory and local synthesis method cannot ensure the contact performance on the entire tooth surface resulting in uncontrolled contact pattern and undesirable transmission error. A global synthesis approach consisting of a machine-tool settings calculation model, a tooth contact analysis model, a meshing quality assessment function, and a swarm intelligence algorithm are proposed. The direction, area, and shape of the contact pattern and the amplitude of the intersection of two adjacent transmission error curves are taken as the evaluation indexes. Three curvatures of the design pitch cone of the pinion are taken as the control variables. A global selection space is then established within the reasonable range of the curvatures. An improved multiple population genetic algorithm is employed to find the optimal set of the curvatures to achieve the target values of the evaluation indexes. To avoid the edge contact and corner contact condition, a feasible region is defined on tooth surface and the contact patterns on the gear and pinion surfaces are all confined within the area. The optimized contact patterns obtained by the loaded tooth contact analysis method are similar to those obtained by the proposed approach, thus demonstrating the effectiveness of this methodology. The main novelty of this approach is to translate the derivation of the mathematical relation between the curvatures of the mating surfaces and the contact properties to solving a multi-objective optimization problem of the meshing quality indexes by the intelligence algorithm. To enhance the property of YG8 hard alloy, two elements of carbon and vanadium were implanted into its surface by using Metal Vapor Vacuum Arc (MEVVA) source with several different doses. The structure of sample surface is characterized and analyzed with metallurgical microscope, X-ray diffraction and Raman spectrum, respectively. The results show that there exist new phases of C, VC, and V7O3 in the sample surface, and that C + V ion implantation actively promotes the formation of DLC structure. Moreover, it can be found that the performances of sample surface (such as microhardness, coefficient of friction and corrosion resistance) are evidently improved. In conclusion, the surface modification of YG8 hard alloy by C + V ion implantation is not only affected by the appearance of new phase, but also related to the implantation mechanism. In order to provide control and prevention measures against an influenza pandemic, we investigated the epidemic characteristics of influenza from January 2001 in some surveillance sites of Fujian province, especially in Fuzhou city. We isolated 42 influenza viruses in 2001, 32 of them were H1N1, 10 isolates belonged to the B/Yamagata lineage. We did not isolate H3N2 in 2001. But in 2002, we isolated 44 influenza viruses; 22 of them were H3N2, only one isolate was H1N1, the other 21 isolates were B type. Five of the B type were B/Yamagata and 16 were B/Victoria. Eight isolates of B/Victoria were isolated from a primary school in Fuzhou in December 2002 for an influenza outbreak. In Fuzhou, the epidemic was predominated by H1N1 in 2001. But in 2002, the epidemic was predominated by H3N2 and B/Victoria. There were two epidemic peaks in 2001 and 2002, but the peaks'' time and predominant strains were different. It is so strange that we isolated 32 isolates of the H1N1 influenza virus with embryonated chicken eggs in 2001, but in 2002 we only isolated 1 isolate of H1N1 with embryonated chicken eggs. In 2002, most influenza viruses, especially H3N2, were isolated by MDCK cells. The epidemic strain of the influenza virus changed all the time, and the influenza virus also changed in its way. This study aims to investigate the characteristics of genomic variation of pandemic A/H1N1/2009 influenza virus isolated in Fujian Province, China. Complete genome sequence analysis was performed on 14 strains of pandemic A/H1N1/2009 influenza virus isolated from Fujian during 2009-2012. All virus strains were typical low-pathogenic influenza viruses, with resistance to amantadine and sensitivity to neuraminidase inhibitors. Eight genome fragments of all strains were closely related to those of A/California/07/2009 (H1N1) vaccine strain, with > or = 98.2% homology. Compared with the vaccine strain, the influenza strains from Fujian had relatively large variation, and variation was identified at 11 amino acid sites of the HA gene of A/Fujiangulou/SWL1155/2012 strain, including 4 sites (H138R, L161I, S185T, and S203T) involved inthree antigen determinants (Ca, Sa, and Sb). In conclusion, the influenza vaccine has a satisfactory protective effect on Fujian population, but the influenza strains from Fujian in 2012 has antigenic drift compared with the vaccine strain, more attention should therefore be paid to the surveillance of mutations of pandemic A/H1N1/2009 influenza virus.
