Abstract

Poplar stands as one of the primary afforestation trees globally. We successfully generated transgenic poplar trees characterized by enhanced biomass under identical nutrient conditions, through the overexpression of the pivotal nitrogen assimilation gene, pxAlaAT3. An environmental risk assessment was conducted for investigate the potential changes in rhizosphere soil associated with these overexpressing lines (OL). The results show that acid phosphatase activity was significantly altered under ammonium in OL compared to the wild-type control (WT), and a similar difference was observed for protease under nitrate. 16SrDNA sequencing indicated no significant divergence in rhizosphere soil microbial community diversity between WT and OL. Metabolomics analysis revealed that the OL caused minimal alterations in the metabolites of the rhizosphere soil, posing no potential harm to the environment. With these findings in mind, we anticipate that overexpressed plants will not adversely impact the surrounding soil environment. [ABSTRACT FROM AUTHOR]

11. High-Efficiency Direct Esterification of CO2and Methanol over Co-Doped CeO2Catalysts

Abstract

The direct esterification reaction of CO2with methanol to produce dimethyl carbonate (DMC) provides a sustainable pathway for the resourceful utilization of CO2. However, the chemical stability of CO2makes the reaction kinetically difficult with low methanol conversion and DMC yields. In this study, a series of catalysts (denoted as M-CeO2, M = Fe, Cu, Co, La, Zr, Ni, and Al) with oxygen vacancy defects were constructed by a doping strategy, which provided critical sites for the adsorption and activation of methanol and CO2. Additionally, 2-cyanopyridine (2-CP) was employed as a dehydrating agent to facilitate the esterification reaction through dehydration. Under the optimized experimental conditions, the conversion of methanol was 61.9% and the DMC yield was 61.6%. The experimental and characterization results show that the extensive oxygen vacancies and base sites on the surface of the 5% Co-CeO2catalyst are crucial for activating methanol and CO2, which is the main reason for the high catalytic activity of the catalyst. In situ diffuse reflectance infrared Fourier transform spectroscopy (DRIFTS) effectively demonstrated the efficient activation of methanol and CO2on the catalyst and further confirmed the formation of monomethyl carbonate species during the reaction process. The high catalytic performance was also demonstrated in the direct esterification of other monohydric alcohols, thus confirming the broad applicability of the catalyst. This study presents a viable approach for the efficient utilization of CO2resources, offering a promising solution for the sustainable management of greenhouse gas.

Abstract

Study Design: Retrospective Cohort Study Objectives: To explore whether classification of the increased signal intensity (ISI) on magnetic resonance imaging (MRI) correlates with clinical presentations and outcomes in symptomatic thoracic ossification of ligamentum flavum (T-OLF) patients. Methods: All patients with symptomatic T-OLF who underwent laminectomy at four institutions were reviewed. The ISI on preoperative T2-weighted MRI was divided into 3 groups, Grade 0, none; Grade 1, light (obscure); and Grade 2, intense (bright). Neurological function before surgery and at follow-up was evaluated by the revised Japanese Orthopedic Association (JOA) score. Patients' demographics, clinical manifestations, and surgical outcomes were compared. Results: A total of 94 patients were involved. Preoperative MRI showed 32 patients in Grade 0, 39 patients in Grade 1, and 23 patients in Grade 2. Low extremities numbness, weakness, and clinical signs were less frequent in Grade 0 patients. The grade of ISI was correlated with the duration of symptoms and cord compression. Grade 0 patients had a better preoperative JOA score than those with ISI changes, while Grade 2 patients showed worse neurological recovery, longer duration of operation, more intraoperative blood loss, and a higher incidence of perioperative complications. Conclusion: The classification of ISI is an effective parameter for preoperatively assessing cord compression, clinical severity, and surgical outcomes in T-OLF patients. Grade 0 patients have relatively mild neurological impairment but are more likely to be misdiagnosed. Grade 2 indicates the worst clinical impairment and neurological recovery, and implies a risky and challenging surgery. [ABSTRACT FROM AUTHOR]

Abstract

Background: Osteosarcoma is a common malignant tumor, with relatively lower survival rates in adolescents. Dihydrotanshinone I (DHI) was extracted from the traditional Chinese medicine Salvia miltiorrhiza and was shown to inhibit several types of cancer. Purpose: To explore the effect of DHI on the proliferation, migration, invasion, and apoptosis of osteosarcoma cells, as well as the possible molecular mechanism. Methods: The effect of DHI on the proliferation of osteosarcoma was detected by crystal violet assay, MTT assay, colony formation assay. The effects of DHI on the migration and invasion of osteosarcoma were detected by wound healing assays, cell migration and invasion assays. The effect of DHI on apoptosis of osteosarcoma was detected by cell apoptosis assay and Hoechst apoptosis staining. The protein expression levels were detected by Western blotting assay. The activity of Wnt/β-Catenin signaling pathway was detected by luciferase reporter assay and Western blot. The inhibitory effect of DHI on osteosarcoma in vivo was analyzed by an orthotopic OS tumor animal model and immunohistochemistry. Result: DHI may inhibit the proliferation, decrease the migration, reduce the invasion, and promote the apoptosis of osteosarcoma cells. In vivo mouse model, DHI can inhibit the formation of osteosarcoma. In terms of mechanism, DHI may inhibit both the transcriptional activity and the total protein level of β-catenin. Conclusion: DHI may inhibit the proliferation, migration, and invasion as well as induce the apoptosis of osteosarcoma cells, possibly through suppressing the Wnt/β-catenin signaling pathway.

Hoechst 33258 staining and flow cytometry were used to observe apoptosis. The migration and invasion role of OS cells were evaluated using transwell assays and wound healing assays. Western blotting was used to analyse the protein expression levels. Nude mice were inoculated with 143B cells to establish an orthotopic OS tumor animal model and to investigate the effects of CA on OS tumors.Results: According to crystal violet assay, MTT assay and colony-forming assay, CA significantly inhibited cell proliferation. Hoechst 33258 staining and flow cytometry analysis showed that CA-induced apoptosis in a concentration-dependent manner. In addition, transwell assays and wound healing assays showed that CA inhibited the migration and invasion of osteosarcoma cells. In vivo mouse models, CA inhibited the growth of osteosarcoma. The potential mechanisms could be that CA inhibited the transcriptional activity of Wnt/β-catenin and PI3K/Akt of the osteosarcoma.Conclusion: CA may inhibit the proliferation, migration, invasion and promote apoptosis of OS cells by inhibiting Wnt/β-catenin and PI3K/Akt signaling pathways.
