4. Effects of heavy ion mutation induction breeding on growth and physiological characteristics of broccoli

Abstract

Brassica oleracea is one of the important vegetable crops in China. Its seeds are primarily imported. Hence, developing innovative varieties belonging to China is an urgent requirement. Since broccoli is a cross-pollinated crop systematic selection and cross-breeding are challenging, and the application of radiation mutagenesis breeding in broccoli has not been reported. In this study, the seeds of Brassica oleracea were irradiated using carbon ion beams to investigate the biological effects of these beams on broccoli. The growth indices, antioxidant enzyme activity, photosynthetic indices, and chlorophyll fluorescence were detected at the seedling stage. The results revealed that irradiation with 100–500 Gy exhibited no significant effect on the germination of Brassica oleracea, while the germination of seeds was significantly inhibited at 600 Gy. After irradiation with 100–600 Gy, the root length, shoot length, seedling height, and leaf area decreased with the increase in irradiation dose. The Median lethal dose (LD50) of Brassica oleracea irradiated using a carbon ion beam was 415.89 Gy, and the dose that halved the root length was 495.12 Gy. After irradiation, the activities of superoxide dismutase (SOD) and peroxidase (POD) were higher than that of the control plants, while the activity of catalase (CAT) was lower than that of the control plants. The content of malondialdehyde (MDA) increased significantly at a dose of 400 Gy. The photosynthetic pigments (chlorophyll a, chlorophyll b, and carotenoids) showed an increasing trend initially, and then a decreasing trend with an increasing irradiation dose, and the highest value was detected at a dose of 300 Gy. Net photosynthesis, transpiration rate, and stomatal conductance exhibited a negative correlation with irradiation dose, while the non-photosynthetic quenching coefficient exhibited a significant increase after irradiation. The findings indicated that heavy ion beam radiation inhibited the growth of broccoli plants and affected the activity of antioxidant enzymes and photosynthesis. The study provided basic data for the radiation mutation breeding of Brassica oleracea.

12. Atomistic aspects of load transfer and fracture in CNT-reinforced aluminium

Abstract

This paper describes atomistic simulations of deformation and fracture of Al reinforced with carbon nanotubes (CNTs). We use density functional theory (DFT) to understand the energetics of Al-graphene interfaces and gain reference data for the parameterization of Al-C empirical potentials. We then investigate the load transfer between CNTs and Al and its effect on composite strengthening. To this end, we perform uniaxial tensile simulations of an Al crystal reinforced with CNTs of various volume fractions. We also study the interaction of the embedded CNTs with a crack. We show that the interaction between CNTs and Al is weak such that, under tensile loading, CNTs can easily slide inside the Al matrix and get pulled out from the cracked surface. This effect is almost independent of CNT length and volume fraction. Little load transfer and consequently no crack bridging are observed during the simulation of pristine CNTs threading the crack surfaces. CNTs that are geometrically fixated inside Al, on the other hand, can increase the fracture stress and enhance plastic dissipation in the matrix. CNTs located in front of a growing crack blunt the crack and induce plastic deformation of the Al matrix. Abstract

Irreversible cardiomyopathy was caused by the therapeutic of anthracyclines in the chemotherapy of cancers. The cell apoptosis and autophagy were induced by anthracyclines in AC16 cells. MiR-223-3p ascends in anthracycline-treated AC16, but the expression of nuclear factor I-A (NFIA) was specifically down-regulated. However, the underlying molecular mechanism between NFIA and miR-223-3p is unclear now in AC16 cells. In our research, NFIA expression was dampened in AC16 cells by miR-223-3p mimics. Additionally, miR-223-3p knockdown hindered the apoptosis and autophagy in anthracycline-treated AC16. Furthermore, NFIA was predicted and verified as a miR-223-3p’s downstream target and rescued the functions of miR-223-3p. These findings illustrated that miR-223-3p advances anthracycline-stimulated cardiomyocyte damage progression by targeting NFIA, implying the promising therapeutic function of miR-223-3p on cardiomyocyte damage in cancer patients.

Abstract

The most common subtype of lung cancer is non-small cell lung cancer (NSCLC) , accounting for 80% to 85% of lung cancer. During the development process, the tumor is easily metastasized to the brain, seriously affecting the treatment prognosis and quality of life of patients. At present, the main treatment options for brain metastasis of lung cancer are surgery, chemotherapy, radiotherapy, targeted therapy, and immunotherapy. In recent years, breakthroughs have been made in the application of immune checkpoint inhibitors, and some combination immunotherapies have been clinically approved. According to the Chinese Experts Consensus on Immune Checkpoint Inhibitors for Non-small Cell Lung Cancer, there have been certain studies showing that immunotherapy can be applied to the treatment of advanced NSCLC. Immunotherapy is like a double-edged sword, and its practical clinical significance still needs further exploration. This article reviews the progress of immunotherapy for brain metastasis of NSCLC and explores more reasonable clinical treatment plans for patients with brain metastasis of lung cancer.

Abstract

We report results of a multiscale simulation study of multilayer structures consisting of graphene sheets with embedded Pt nanoparticles. Density functional theory is used to understand the energetics of Pt-graphene interfaces and provide reference data for the parameterization of a Pt-graphene interaction potential. Molecular dynamics simulations then provide the conformation and energetics of graphene sheets with embedded Pt nanoparticles of varying density, form and size. These results are interpreted using a continuum mechanical model of sheet deformation, and serve to parameterize a meso-scale Monte Carlo model to investigate the question under which conditions the free volume around the Pt nanoparticles forms a percolating cluster, such that the structures can be used in catalytic applications.
