1. Low Radon Concentration Measurement with a Radon-Dissolved Liquid Scintillator Pilot Detector

Abstract

We construct a high sensitivity radon pilot detector using liquid scintillator dissolved radon for the CDEX rare-event searches program. The CDEX-10 project employs a germanium detector array immersed in a massive liquid nitrogen tank. However, radon emanated from the surface of the tank can contribute background. As a result, radon contamination in the liquid nitrogen tank must be regulated and monitored, which necessitates the use of a low-level Rn measuring device. The radon-dissolved liquid scintillator (LS) detectors utilizes cascade decay selections, yielding a unique signature. All background events occurring in the LS, with regard to the system and co-efficiency of Rn absorption, have been studied. Background activity is measured to be 59.8$\pm$18.4 $\mu$Bq per 300 mL LS. Meanwhile, it shows that radon concentration absorbed by LS will be saturated and is linearly related to radon concentration in nitrogen gas. Furthermore, the co-efficiency factor can be optimized by lowering temperature or raising pressure. Abstract

The China Dark Matter Experiment (CDEX) is located at the China Jinping underground laboratory (CJPL) and aims to directly detect the WIMP flux with high sensitivity in the low mass region. Here we present a study of the predicted photon and electron backgrounds including the background contribution of the structure materials of the germanium detector, the passive shielding materials, and the intrinsic radioactivity of the liquid argon that serves as an anti-Compton active shielding detector. A detailed geometry is modeled and the background contribution has been simulated based on the measured radioactivities of all possible components within the GEANT4 program. Then the photon and electron background level in the energy region of interest (<10^-2 events kg-1 day-1 keV-1 (cpkkd)) is predicted based on Monte Carlo simulations. Abstract

The CDEX Collaboration has been established for direct detection of light dark matter particles, using ultra-low energy threshold p-type point-contact germanium detectors, in China JinPing underground Laboratory (CJPL). The first 1 kg point-contact germanium detector with a sub-keV energy threshold has been tested in a passive shielding system located in CJPL. The outputs from both the point-contact p+ electrode and the outside n+ electrode make it possible to scan the lower energy range of less than 1 keV and at the same time to detect the higher energy range up to 3 MeV. The outputs from both p+ and n+ electrode may also provide a more powerful method for signal discrimination for dark matter experiment. Some key parameters, including energy resolution, dead time, decay times of internal X-rays, and system stability, have been tested and measured. The results show that the 1 kg point-contact germanium detector, together with its shielding system and electronics, can run smoothly with good performances. Abstract

Weakly Interacting Massive Particles (WIMPs) are the candidates of dark matter in our universe. Up to now any direct interaction of WIMP with nuclei has not been observed yet. The exclusion limits of the spin-independent cross section of WIMP-nucleon which have been experimentally obtained is about 10^{-7}pb at high mass region and only 10^{-5}pb} at low mass region. China Jin-Ping underground laboratory CJPL is the deepest underground lab in the world and provides a very promising environment for direct observation of dark matter. The China Dark Matter Experiment (CDEX) experiment is going to directly detect the WIMP flux with high sensitivity in the low mass region. Both CJPL and CDEX have achieved a remarkable progress in recent two years. The CDEX employs a point-contact germanium semi-conductor detector PCGe whose detection threshold is less than 300 eV. We report the measurement results of Muon flux, monitoring of radioactivity and Radon concentration carried out in CJPL, as well describe the structure and performance of the 1 kg PCGe detector CDEX-1 and 10kg detector array CDEX-10 including the detectors, electronics, shielding and cooling systems. 23. Nitrogen fixation and phytohormone stimulation of sugarcane plant through plant growth promoting diazotrophic Pseudomonas.

Abstract

Diazotrophic microorganisms are free-living groups of organisms that can convert atmospheric nitrogen (N) into bioavailable nitrogen for plants, which increases crop development and production. The purpose of the current study was to ascertain how diazotrophic plant growth promoting (PGP) Pseudomonas strains (P. koreensis CY4 and P. entomophila CN11) enhanced nitrogen fixation, defense activity, and PGP attributes of sugarcane varieties; GT11 and G×B9. A 15N isotope-dilution study was conducted to confirm the sugarcane strains' capacity to fix nitrogen, and the results indicated that between 21 to 35% of plant, nitrogen is fixed biologically by selected rhizobacteria. In comparison to the control, after 30, 60, and 90 days, both CY4 and CN11 strains significantly increased defense-related enzymes (catalase, peroxidase, phenylalanine ammonia-lyase, superoxide dismutase, glucanase, and chitinase) and phytohormones (abscisic acid, ABA, cytokinin, etc.) in GT11 and GXB. Additionally, the expression of SuCHI, SuGLU, SuCAT, SuSOD, and SuPAL genes was found to be elevated in Pseudomonas strains inoculated plants using real-time quantitative polymerase chain reaction (RT-qPCR). Both bacterial strains increased all physiological parameters and chlorophyll content in sugarcane plants more than their control. The effects of P. koreensis CY4 and P. entomophila CN11 strains on sugarcane growth promotion and nitrogen fixation under greenhouse conditions are described here for the first time systematically. The results of confirmation studies demonstrated that P. koreensis CY4 and P. entomophila are PGP bacterial strains with the potential to be employed as a biofertilizer for sugarcane growth, nitrogen nutrient absorption, and reduced application of chemical nitrogenous fertilizers in agricultural fields. Abstract

Sugarcane (Saccharum spp. hybrid) is the chief source of sugar and biofuel globally and is prominent among cash crops. Sucrose is the main required product in sugarcane, and many studies have been performed to understand the phenomena of sucrose synthesis, metabolism, and accumulation in sugarcane. However, none of the studies concluded that a single gene is responsible for the sucrose content. Instead, a complex mechanism consisting of several genes, such as sucrose phosphate synthase genes (SPS1, SPS2, SPS4, SPS5), sucrose synthase genes (SuSy1, SuSy2, SuSy4), invertase genes (INV, CWIN, NIN1, CINV2), and phytohormone, trehalose, transcription factor (TF), protein kinase, and sugar transporter genes are working spatiotemporally in sugarcane. Currently, omics approaches like transcriptomics, proteomics, and metabolomics are also being used to explore the sugar metabolism in sugarcane, but integrated transcriptomic, proteomic, and metabolomic studies have been less reported. The results obtained from the integrated analysis of transcriptomics, proteomics, and metabolomics are more reliable because the strong gene expression, received in the form of abundant mRNA, does not guarantee the plentiful existence of associated proteins or their particular activity in the target cells or tissues, which discloses the restraint of single interpretation and stresses the significance of the integrated analysis of transcriptomics, proteomics, and metabolomics. This review highlights different genes and molecules contributing to sugar metabolism at different stages and the significance of omics approaches in explaining sucrose metabolism, especially sucrose accumulation in sugarcane.
