Long-term exposure to ultraviolet (UV) radiation induces skin photoaging, which manifests as oxidative stress, inflammation, and collagen degradation. Multiple approaches (topical or systemic retinoids, antioxidants, alpha-hydroxy acids, laser, surgery) are used in the treatment of photoaged skin, and the use of topical retinoids is currently a primary clinical treatment. Previous studies revealed that retinoic acid promotes keratinocyte proliferation and reduces melanin deposition and matrix metalloproteinase (MMP) secretion; it also causes potential allergic and inflammatory damage to the skin. This study aimed to investigate the therapeutic effects and mechanisms of trifarotene, a functional retinoic acid analog, on UV-irradiated photoaging ICR and BALB/c nude mice and UVB photodamaged human epidermal keratinocyte (HaCaT) cells by examining indicators such as collagen, oxidoreductase, and inflammatory factor presence through histochemical staining, Western blot, and ELISA. Results suggested that trifarotene significantly reduced UV-induced photoaging in mouse skin tissue, potentially by reducing oxidative stress damage and inflammatory factor release, and inhibiting melanin deposition and collagen degradation by downregulating MMP expression. Concentrations of malondialdehyde, tyrosinase, interleukin-6, interleukin- 12, and tumor necrosis factor-alpha in photoaged skin decreased, while SOD content in photodamaged HaCaT cells significantly increased. Trifarotene (3.3 μmol L–1) inhibited phosphorylated JNK and c-Jun expression both independently and collaboratively with the JNK activator anisomycin, demonstrating that trifarotene mitigates UV-induced collagen degradation and apoptosis through inhibition of the JNK/c-Jun/MMPs signaling pathway.

Deep-sea minerals are regarded as the most economically viable and promising mineral resource. Vertical hydraulic lifting represents one of the most promising methods for deep-sea mining lifting systems. To mitigate the potential for clogging due to the aggregation of particles in vertical pipe transport during deep-sea mining operations, this paper employs numerical simulations utilizing the computational fluid dynamics and discrete element method (CFD-DEM) model to investigate the swirling flow transportation of mineral particles. The characteristics of the swirling flow field and the motion law of double-size particles at different swirling ratios are investigated. The findings demonstrate that, in comparison to axial transport within the pipeline, the particle movement observed in swirling flow transport exhibits an upward spiral trajectory. This phenomenon facilitates the orderly movement of particles, thereby enhancing the fluidization of particles within the pipeline. An increase in the swirling ratio (SR) has a considerable impact on the velocity within the pipe. The tangential velocity distribution undergoes a gradual transition from centrosymmetric to non-centrosymmetric as the distance from the inlet increases. An increase in the SR results in an enhanced aggregation of particles at the wall, accompanied by a notable rise in the local particle concentration. The value of SR = 0.3 represents a critical threshold. When SR exceeds this value, the distribution of particles in the cross-section reaches a relatively stable state, rendering it challenging to further alter the distribution and concentration of particles, even if the SR is augmented. Furthermore, the maximum local particle concentration in the vicinity of the wall tends to be stable. Background: Tripartite motif containing 59 (TRIM59) is a ubiquitin ligase and is involved in the pathogenesis of various diseases, including cancers, sepsis, and other immune-related diseases. However, it has not been defined whether TRIM59 plays a role in ischemic stroke in mice. Methods: This study determined the influence of Trim59 deficiency on experimental stroke outcomes and the cerebral proteomic profile using myeloid cell Trim59 conditional knockout (Trim59-cKO) mice and a label-free quantitative proteomic profiling technique. The possible mechanisms by which TRIM59 affected stroke onset were elucidated by in vivo and in vitro experiments. Results: Immunofluorescence staining results showed that TRIM59 expression was up-regulated after cerebral ischemia and co-localized with macrophages. Myeloid cell Trim59 deficiency exacerbated ischemic injury on day 3 after experimental stroke. In proteomic analysis, 23 differentially expressed proteins were identified in ischemic brain of Trim59-cKO mice as compared to Trim59flox/flox mice. Kyoto Encyclopedia of Genes and Genomes pathway analysis revealed that the differentially expressed proteins were enriched in complement and coagulation cascades. Protein–protein interaction analysis suggested the central role of clusterin in the interaction network. ELISA and Western blot assays confirmed the reduced levels of clusterin protein in the ischemic brains of Trim59-cKO mice. Further experimental results showed that clusterin was expressed in neurons. Conditional co-culture experiments of primary neurons and bone marrow-derived macrophages demonstrated that LPS stimulated macrophages to secrete complement C3. In addition, TRIM59 may affect the changes in clusterin expression in an indirect manner by influencing the secretion of complement C3 in macrophages. In vivo experiments also proved a significant increase in C3 levels in the brains of Trim59-cKO mice after ischemia. Conclusion: Myeloid cell Trim59 deficiency aggravated ischemic stroke outcomes in conjunction with a distinct cerebral proteomic profile, and the underlying mechanism may be related to the regulation of macrophage C3 expression by TRIM59. Arrow bamboo (Fargesia qinlingensis) is endemic to the Qinling Mountains and has remarkable adaptive resilience to changing climates. However, its complete mitogenome remains unknown. Using the Illumina and PacBio HiFi sequencing platforms, we found that the mitogenome assembly of the F. qinlingensis has a multi-branched skeleton comprising three linear molecules (M1, M2, and M3), with a length of 442,368 bp and a GC content of 44.05%. Thirty-five unique PCGs were identified in the complete mitogenome, including twenty-four core structural genes, eleven noncore structural genes, three rRNAs, and sixteen tRNAs. The GCU for alanine and CAA for glutamine represented the most significant frequency (RSCU = 1.55) in the codon usage preference. A total of 51, 28, and 14 SSRs were determined on M1, M2, and M3, respectively. The mitogenome contained 149 pairs of dispersed repeats with lengths greater than 30 bp, the most abundant of which were 82 forward and 67 palindromic repeats. A long repeat sequence (14,342 bp) was characterized in mediating mitogenome recombination. DNA transfer analyses suggested that 44 MTPTs (30,943 bp, 6.99%) originated from the plastome. Among the 482 potential C-U/T RNA-editing sites predicted in 35 PCGs, ccmFn (38 times) and ccmC (36 times) shoed the highest frequency. Collinearity and phylogenetic trees revealed the close relationship between F. qinlingensis and Bambusa oldhamii. The primary features of the mitogenome of F. qinlingensis will help decipher the functional mitochondrial traits related to growth performance and climate resilience. In the process of oil production, wells containing gas can impact the efficiency of electric submersible pump (ESP), potentially causing gas lock. This issue can lead to the loss of lifting capacity in ESP, affecting the normal production of oil wells. To address this problem, the concept of gas separation before mixing transportation has been proposed, and a gas–liquid separation–mixing device has been designed. Experimental tests on the gas–liquid two-phase flow under various working conditions were conducted. A numerical model of the physical process was developed and validated with the experimental results. The results indicate that when the inlet flow rate exceeds 8.75 m3/h, the gas phase can be effectively accumulated in the center of the main pipeline after flowing through the guide vanes, thereby achieving efficient gas–liquid separation. Centrifugal number, which is defined as the ratio of axial flux of centrifugal force to axial flux of gravity, was proposed for evaluating the flow characteristics. When the centrifugal number exceeds 6.5, a high-quality gas core is formed in the pipe. At high inlet gas content, the volume fraction of gas in the main pipe initially decreases to 2% as the flow rate increases to 15 m3/h. However, at a flow rate of 30 m3/h, the volume fraction gradually rises to 30%, which results in a significant amount of gas being forced into the main pipe.
