Aims: Excessive neuroinflammation mediated mainly by microglia plays a crucial role in ischemic stroke. AZD1390, an ataxia telangiectasia mutated (ATM) specific inhibitor, has been shown to promote radio‐sensitization and survival in central nervous system malignancies, while the role of AZD1390 in ischemic stroke remains unknown. Methods: Real‐time PCR, western blot, immunofluorescence staining, flow cytometry and enzyme‐linked immunosorbent assays were used to assess the activation of microglia and the release of inflammatory cytokines. Behavioral tests were performed to measure neurological deficits. 2,3,5‐Triphenyltetrazolium chloride staining was conducted to assess the infarct volume. The activation of NF‐κB signaling pathway was explored through immunofluorescence staining, western blot, co‐immunoprecipitation and proximity ligation assay. Results: The level of pro‐inflammation cytokines and activation of NF‐κB signaling pathway was suppressed by AZD1390 in vitro and in vivo. The behavior deficits and infarct size were partially restored with AZD1390 treatment in experimental stroke. AZD1390 restrict ubiquitylation and sumoylation of the essential regulatory subunit of NF‐κB (NEMO) in an ATM‐dependent and ATM‐independent way respectively, which reduced the activation of the NF‐κB pathway. Conclusion: AZD1390 suppressed NF‐κB signaling pathway to alleviate ischemic brain injury in experimental stroke, and attenuated microglia activation and neuroinflammation, which indicated that AZD1390 might be an attractive agent for the treatment of ischemic stroke. In many cases, 1D inversion is still an important step in transient electromagnetic data processing. Potential issues may arise in the calculation of apparent resistivity using induced electromotive force (EMF) due to overshoot and the presence of multi-valued functions. Obtaining reliable and consistent inversion results using a uniform half-space as the initial model is challenging, especially when aiming for efficient inversion. Focusing on these problems, we use the land-based transient electromagnetic (TEM) sounding data, which was acquired by using a large fixed-loop transmitter, and adopt a quasi-2D inversion scheme to generate improved images of the subsurface resistivity structure. First, we have considered directly using magnetic field data or converting induced EMF into magnetic field, and then calculating the apparent resistivity over the whole zone. Next, a resistivity profile that varies with depth is obtained through fast smoke ring imaging. This profile serves as the initial model for the subsequent optimal inversion. The inversion scheme uses a nonlinear least-squares method, incorporating lateral and vertical constraints, to produce a quasi-2D subsurface image. The potentiality of the proposed methodology has been exemplified through the interpretation of synthetic data derived from a 3D intricate resistivity model, as well as field data obtained from a TEM survey conducted in a coalmine field. In both cases, the inversion process yields quasi-2D subsurface images that exhibit a reasonable level of accuracy. Objective: To observe the characteristics of gene methylation in obese rats with phlegm-dampness syndrome induced by the high-fat diet, and to study the effect of Wen Dan Decoction on gene methylation after the intervention. Methods: Methylation sites of genes were detected by the MeDIP-seq method. Bioinformatics method was used to analyze the gene methylation characteristics of obesity with phlegm- dampness syndrome and the effect of Wen Dan Decoction. Results: (1) There were 3 242 methylation differential loci in diet- induced obesity with phlegm-dampness syndrome, of which 1 243 were down-regulated and 1 999 were up-regulated, involving 1 579 differential genes. GO analysis showed that “offactory receptor activity” and others were enriched. The possible signal pathways involved were “Olfactory transduction” “Tuberculosis” “Systemic lupus erythematosus” and “Ribosome”. (2) After the intervention of Wen Dan Decoction in obesity with phlegm- dampnesssyndrome,4046differentmethylationlociwereobtained, including 1 067 down-regulated loci and 2 979 up-regulated loci, involving 2 068 genes. GO analysis showed that ''offactory receptor activity'' and others were enriched. These genes involved seven signaling pathways, such as “Metabolic pathways”. (3) Between diet-induced obesity with phlegm-dampness syndrome and Wen Dan Decoction intervening obesity with the phlegm-dampness syndrome, 582 common genes of methylation differential genes were obtained. After the intervention of Wen Dan Decoction, the number of GO enrichment items was more than that of obesity with phlegm-dampness syndrome, and even the same GO enrichment items involved more genes. Conclusions: The phlegm-dampness syndrome of obesity- induced by diet had the characteristics of gene methylation changes, and the intervention of Wen Dan Decoction could also affect the status of gene methylation. The genes affected by Wen Dan Decoction were closely related to the methylation gene of phlegm-dampness syndrome of obesity-induced by diet but covered a wider range.
