The microseismic first arrival time picking is an important prerequisite for the high-precision positioning of the microseismic source. The traditional manual picking method is inefficient. The automatic picking method is difficult to pick the arrival time of the first wave accurately under the condition of low signal-to-noise ratio. In order to solve the above problems, an automatic picking method of microseismic first arrival time based on improved support vector machine (SVM) is proposed. Firstly, the method carries out normalization processing, linear correction and proper clipping on original microseismic data. The method marks different categories of the data by taking the amplitude, the energy and the energy ratio of adjacent moments of the microseismic data as features. Secondly, the method adopts a particle swarm optimization (PSO) algorithm and a grid search method to optimize the penalty parameters and the kernel function parameters of the SVM. The method carries out large-range fast positioning on the parameters by using the PSO algorithm to obtain a preliminary optimal solution. Then the method re-constructs a parameter search interval by taking the solution as an initial position, sets a small-step grid search method to carry out fine searching on the parameters to obtain the optimal parameters. The method substitutes the optimal parameters into the SVM model to train, and obtains the improved SVM model. Finally, the microseismic data are classified and identified according to the improved SVM model. The time corresponding to the first sampling point of the microseismic wave is defined as the arrival time of the first wave. The microseismic monitoring data from a mine shaft is used for the experiment. The results show that the accuracy of the method for picking the microseismic first arrival time is 96.5%, and the average picking error is 3.8 ms. Under the condition of low signal-to-noise ratio, the microseismic first arrival time can still be picked accurately. The picking precision is higher than the short term average/long term average (STA/LTA) method commonly used in automatic picking methods.

The regulation of fruit development is a complex process and a core issue in the fruit tree industry. To investigate the role of PbGIF1 in pear fruit development, we identified a transcription factor PbbHLH137 that regulates pear (Pyrus bretschneideri) fruit development by screening a yeast library constructed from fruit cDNA. Yeast two‐hybrid (Y2H), bimolecular fluorescence complementation (BiFC), and split luciferase complementation (split‐LUC) assays were performed to confirm the PbbHLH137–PbGIF1 interaction. By tracing the complete fruit development process, we found that PbbHLH137 expression was closely related to fruit size and highly involved at the late pear fruit development stage. Transgenic experiments showed that heterologous expression of PbbHLH137 or PbGIF1 promoted fruit enlargement. PbbHLH137 promoted mainly the expansion of fruit cell volume, whereas PbGIF1 mainly increased the number of cells. Further LUC experiments demonstrated that PbGIF1 promoted the transcriptional activation ability of PbbHLH137. Our work identified PbbHLH137 as a transcription factor that regulates fruit development, and showed that PbGIF1 played an ongoing role during fruit development, making it a candidate gene for genetic improvement of pear fruit development. The red color of the pear peel in red-skinned European pear is due to the accumulation of anthocyanins. Numerous transcription factors play pivotal roles in anthocyanin biosynthesis, with zinc finger proteins frequently implicated in the regulation of this process via MYB10, as documented in earlier studies. In this article, we identified a zinc finger protein gene, named PbZAT12, that does not rely on PbMYB10, to regulate anthocyanin biosynthesis. The PbZAT12 protein was localized within the nucleus and exhibited a positive influence on the accumulation of anthocyanins in the peel of 'Red Zaosu' pears. Moreover, overexpression of PbZAT12 resulted in a significant up-regulation of PbDFR, PbANS, and PbUFGT expression levels in pear fruitlets. Y1H assays demonstrated a direct binding ability of PbZAT12 to proPbDFR, proPbANS, and proPbUFGT, which was supported by a dual luciferase assay, indicating its potential to activate the transcriptional activity of these promoters. However, in contrast to its effect on the aforementioned promoters, PbZAT12 did not exhibit an activation of PbMYB10. In summary, our findings suggest that a zinc finger transcription factor, PbZAT12, exerts a positive influence on anthocyanin biosynthesis in pear fruit through direct upregulation of the expression levels of PbDFR, PbANS, and PbUFGT. Anthocyanins are common secondary metabolites in plants that impart red coloration to fruits and flowers. The important WRKY transcription factor family plays multifaceted roles in plant growth and development. In this study, we found a WRKY family gene, Pyrus bretschneideri WRKY75, that may be involved in anthocyanin synthesis in pear. Unlike Arabidopsis thaliana WRKY75, PbWRKY75 may be a positive regulator of anthocyanin synthesis. A transient expression assay indicated that PbWRKY75 promoted pear anthocyanin synthesis. The structural genes (PbANS, PbDFR, and PbUFGT) and positive regulators (PbMYB10 and PbMYB10b) of anthocyanin synthesis were significantly upregulated in the fruitlet skins of PbWRKY75‐overexpressing "Zaosu" pears. Subsequently, yeast one‐hybrid and dual‐luciferase assays indicated that PbWRKY75 promoted PbDFR, PbUFGT, and PbMYB10b expression by activating their promoters. These results revealed that PbWRKY75 may promote the expression of both PbMYB10b and anthocyanin late biosynthetic genes (PbDFR and PbUFGT) by activating their promoters, thereby inducing anthocyanin synthesis in pear. Fruit set and development occur following successful fertilization. Parthenocarpy, a valuable trait in some self-incompatible species, produces seedless fruit without fertilization. Gibberellin (GA) is a crucial hormone in fruit-set regulation and development. While investigating the development of parthenocarpy in pear (Pyrus bretschneideri Rehd.), we determined that GA 20-oxidases (GA20ox) may play key roles in seedless pear fruit development. Sequence analysis revealed three PbGA20ox genes: PbGA20ox1, PbGA20ox2 , and PbGA20ox3. We analyzed the expression patterns of candidate genes and found that PbGA20ox2 levels significantly changed in pollinated fruits. Tissue-specific expression assays revealed that PbGA20ox2 is highly expressed in young fruit and leaves. Subcellular localization assays showed it was located in the cytoplasm, nucleus, and plasma membrane. Overexpressed PbGA20ox2 tomato plants were taller and had longer hypocotyls and internodes, and the emasculated flowers produced parthenocarpic fruit. In pear, the transient overexpression of PbGA20ox2 promoted fruit development and delayed the drop of nonpollinated fruit. Furthermore, the fruit of PbGA20ox2- overexpressing tomato and transient PbGA20ox2- overexpressing pear had increased GA4 (but not GA3 and GA1) contents. This result provided evidence that PbGA20ox2 was necessary for GA4-dependent pear fruit development. Our study revealed that PbGA20ox2 altered the GA biosynthetic pathway and enhanced GA4 synthesis, thereby promoting fruit set and parthenocarpic fruit development. [ABSTRACT FROM AUTHOR]

Parthenocarpy, the productions of seedless fruit without pollination or fertilization, is a potentially desirable trait in many commercially grown fruits, especially in pear, which is self‐incompatible. Phytohormones play important roles in fruit set, a process crucial for parthenocarpy. In this study, 2,4‐dichlorophenoxyacetic acid (2,4‐D), an artificially synthesized plant growth regulator with functions similar to auxin, was found to induce parthenocarpy in pear. Histological observations revealed that 2,4‐D promoted cell division and expansion, which increased cortex thickness, but the effect was weakened by paclobutrazol (PAC), a gibberellin (GA) biosynthesis inhibitor. Phenotypic differences in pear may therefore be due to different GA contents. Hormone testing indicated that 2,4‐D mainly induced the production of bioactive GA4, rather than GA3. Three key oxidase genes function in the GA biosynthetic pathway: GA20ox, GA3ox and GA2ox. In a pear group treated with only 2,4‐D, PbGA20ox2‐like and PbGA3ox‐1 were significantly upregulated. When treated with 2,4‐D supplemented with PAC, however, expression levels of these genes were significantly downregulated. Additionally, PbGA2ox1‐like and PbGA2ox2‐like expression levels were significantly downregulated in pear treated with either 2,4‐D only or 2,4‐D supplemented with PAC.
