This study was conducted to determine the effects of rhizobacteria having high phosphorus solubilizing and nitrogen fixating ability, obtained from soil rhizosphere regions of tomato plants, on causal agent of tomato bacterial cancer and wilt disease [Clavibacter michiganensis subsp. michiganensis (Smith) Davis et al.] (Cmm). Twelve different rhizobacteria isolates were used in the study. In vitro tests revealed that antagonistic effects of the isolates on Crum ranged from 13.16 to 88.44%. Based on the results of in vitro tests, rhizobacteria isolates with high impact on Cmm were selected and pot experiments as well as seed treatments were performed with these isolates. In pot experiments, combination of Pseudomonas koreensis + P. thivervalensis resulted in 52.78% disease suppression. Similarly, combination of P. koreensis + P. thivervalensis + P. koreensis was more effective than the rest of the treatments and resulted a disease suppression ratio of 72.97%. Further field experiments should be performed with the rhizobacteria isolates having the highest impact on Cmm in current study. Purpose: Present study, we were carried out to evaluate the antifungal activities of extracts of Grindelia robusta Nutt., Tanacetum praeteritum subsp. praeteritum and Alchemilla vulgaris L. against Fusarium oxysporum f. sp. lycopersici, Botrytis cinerea Macrophamina phaseolina, Rhizoctania solani, Alternaria alternata, Cylindrocarpon destructans and Phytopthora infestans.Material and Methods: The screening of the antifungal potential of three extract forms from three different plant species was performed in vitro by using Agar-diffusion method on Potato Dextrose Agar media (PDA). One doses (1.0%, 1.5% and 2,0%) of the three diffent extracts were tested. Experimental results were obtained in three (n=3) replicates.Results: All plant extracts prepared in the study exhibited significant activity against plant pathogenic fungi. In particular, Tanacetum praeteritum subsp. praeteritum prevented the growth of micelles between 73.5 - 100% in five different fungal species, while Grindelia robusta Nutt. had a 100% effect on two of the four different fungus species.Conclusion: The extracts exhibited an antifungal activity against all the tested fungi. The analysis shown that the all extracts caused significantly inhibited the mycelium growth of fungi. This study paves the way for the development of new methods for environmentally friendly and economically viable phytosanitary practices.

Orta Karadeniz Bölgesi Samsun, Tokat ve Amasya ili fasulye üretim alanlarında 2013-2014 yıllarında fasulye hale yanıklığı hastalığı etmeni Pseudomonas savastonoi pv. phaseolicola (Psp) ve adi yaprak yanıklığı hastalığı etmeni Xanthomonas axonopodis pv. phaseoli (Xap)’nin belirlenmesi için surveyler yapılmıştır. Güdümlü örnekleme yöntemiyle toplam 154 fasulye tarlası gezilerek şüpheli semptom gösteren yaprak ve meyve örnekleri toplanmıştır. İzolasyon çalışmaları sonucu 72 adet saf bakteri izolatı elde edilmiştir. Patojenisite, morfolojik, biyokimyasal testler, tanıyı destekleyici LOPAT testleri ve moleküler analizler (PCR) sonucunda 30 izolat Psp ve 17 izolat Xap olarak tanılanmıştır. Psp izolatları 8 farklı fasulye çeşidinden oluşan ırk ayrım setindeki bitkilere inokule edilmiş ve çeşitlerin verdiği reaksiyonlara göre Psp’nın ırk/ırkları belirlenmiştir. Çeşit reaksiyonlarına göre Psp izolatlarının tamamının 1 nolu ırk olduğu tespit edilmiştir. Bu çalışmada, Türkiye fasulye üretiminin % 16’sının gerçekleştiği Orta Karadeniz bölgesinde evrensel ırk ayrım seti kullanılarak Psp izolatlarının ırk düzeyinde ayrımı ilk kez yapılmıştır., In 2013-2014, surveys were conducted in order to investigate the presence of Pseudomonas savastonoi pv. phaseolicola (Psp) halo blight and Xanthomonas axonopodis pv. phaseoli (common bacterial blight) (Xap) in common bean production areas of Samsun, Tokat, Amasya at Middle Black Sea region. Total, 154 common bean fields were surveyed and their leaf and bean samples were collected with typically resembling symptoms using guided sampling method. Total of 72 purified bacterial isolates were obtained. All the isolates assayed in pathogenicity, morphological, biochemical tests. Further identification methods were conducted with LOPAT tests and molecular analyses (PCR), their results have revealed that 30 isolates were Psp and 17 isolates were Xap. Psp isolates were separately inoculated on 8 bean varieties to determine their races according to phenotypic reactions. Their reactions on 8 bean varieties were revealed that all Psp isolates were Psp race 1. This work are proved that races of Psp isolates using universal separation set at Middle Black Sea region in which supplies about 16 % of the total bean production in Turkey.

Our study is on testing of different concentrations (5, 10, 15, 20, 30 and 40 µg/ml) of twelve the newly synthesized bimetallic cyanido complexes ([Ni(hydeten)2Ag(CN)2] [Ag(CN)2].H2O (K1), [Cd2(hydeten)2Ag4(CN)8].H2O (K2), [Ni(bishydeten)2Ag(CN)2] [Ag(CN)2].H2O (K3), [Cu(bishydeten)2Ag2(CN)4] (K4), [Cd(bishydeten)0.5]2[Ag(CN)2]4.3H2O (K5), [Ni(N-bishydeten)Ag3(CN)5] (K6), [Cu(Nbishydeten)Ag3(CN)5] (K7), [Zn(N-bishydeten)Ag3(CN)5] (K8), [Cd(N-bishydeten)]4[Ag(CN)2]8[Ag(CN)]2 (K9), [Cu(edbea)Ag2(CN)4].H2O (K11), [Cd(edbea)2][Ag(CN)2]2.H2O (K12) and [Cd(edbea) Ag3(CN)5] (K13) for their antibacterial activity against Pseudomonas syringae pv. tomato (Pst), Clavibacter michiganensis subsp. michiganensis (Cmm) and Xanthomonas axonopodis pv. vesicatoria (Xav). Antibacterial activity determined using the agar well diffusion assay method.showed that compound K9 has by 90% inhibition ratio on the bacterial growth . Eight out of 12 compounds has over 90% inhibition on Cmm. Inhibitory effects varying between 72% and 100% ratio by K11 and K12 on selected bacterial strains were less than the effects of K9 and K3 which showed highly significant antibacterial activity by 93.83% and 85.71% inhibitions against Pst respectively, whereas the lowest activity was by K4 (33%) treatment at 40 µg/ml against whole bacterial pathogens tested in vitro conditions. On the other hand compound K13 exhibited the highest antibacterial activity against Xav by 99% growth inhibition at 40 µg/ml. Most of these compounds showed promising activities against the tested bacteria. Our further studies will continue on testing of their efficiency under field conditions.

This study was carried out to evaluate the antibacterial effects of methanolic extracts of Grindelia robusta, Rhus coriaria, Alchemilla vulgaris, Juglans spp., Malva sylvestris, Saponaria officinalis, Prunella vulgaris, and Tanacetum vulgare against Clavibacter michiganensis subsp. michiganensis (Cmm) and Pseudomonas syringae pv. tomato (Pst). The plant extracts were added to King B medium before being autoclaved to reach concentrations of 0,06, 0,125, 0,25, 0,5, 1, 1,5, 2%. The solidified King B medium was inoculated with the 100 µl of inoculums (1x106 cells/ml) and incubated at 27 °C for two days. Control was maintained in pure King B. The experiment was repeated twice with five replication. At the end incubation periods, bacterial concentrations were measured in the spectrophotometer. The obtained results indicated that 1,5% and 2% extracts of all plants were inhibited both of the test bacteria. Especially Grindelia robusta and Rhus coriaria extracts caused the highest effect on Cmm and Pst. Also, 0,25% extract of Alchemilla vulgaris displayed 76% and 96% growth inhibition on Cmm and Pst, respectively. Also, in the other plants inhibition rates increased depending of increase of consantration. As a result, it was determined that plant extracts used in the study had antibacterial effect on Cmm and Pst and these extracts of plants were found as promising in control of these disease., Bu çalışma, Grindelia robusta, Rhus coriaria, Alchemilla vulgaris, Juglans spp., Malva sylvestris, Saponaria officinalis, Prunella vulgaris ve Tanacetum vulgare bitkilerinden elde edilen metanol ekstraktlarının Clavibacter michiganensis subsp. michiganensis (Cmm) ve Pseudomonas syringae pv. tomato (Pst) üzerindeki antibakteriyal etkilerini belirlemek amacı ile yürütülmüştür. Bitki ekstraktları son konsantrasyon %0,.06, 0.125, 0.25, 0.5, 1, 1.5, 2 olacak şekilde otoklav edilmeden önce King B besi yerine eklenmiştir. Katılaşan King B besi yerine, 1x106 hücre/ml yoğunluğunda hazırlanan Cmm ve Pst bakteri süspansiyonlarının ekimi yapılmış ve 27 °C’de iki gün inkübasyona bırakılmıştır. Kontrol grubu olarak bitki ekstraktlarının olmadığı King B besi yerlerine patojenlerin ekimi yapılmıştır. Çalışma 5 tekerrürlü olarak kurulmuş olup, 2 kez tekrarlanmıştır. İnkübasyon periyodu sonunda, besi yerlerindeki bakteri kolonileri toplanarak spektrofotometrede yoğunlukları ölçülmüştür. Elde edilen verilere göre, tüm bitki ekstraktlarının %1,5 ve %2’lik konsantrasyonları testlenen her iki bakteri üzerinde etkili olmuştur. Özellikle Grindelia robusta ve Rhus coriaria ekstraktları Cmm ve Pst üzerinde en yüksek etkiye sahip olmuştur. Aynı zamanda Alchemilla vulgaris’in %0,25’lik düşük dozu bile %76 ve %96 oranında sırasıyla Pst ve Cmm üzerinde etkili olmuştur. Diğer bitki ekstraktlarında ise doz artışına bağlı olarak patojenler üzerindeki engelleme oranı da artmıştır. Sonuç olarak, çalışmada kullanılan bitki ekstraktlarının Cmm ve Pst etmenleri üzerinde antibakteriyal etkiye sahip olduğu ve hastalıkların mücadelesinde ümit vaat edici olarak değerlendirilmiştir.

One of themost important diseases of wheat affecting yield and quality negatively ispowdery mildew. It is seen very often in spring in regions with high humiditiyand using pesticides to control the disease is not economical. In this study,49 durum wheat land races collected from different regions of Turkey were used.In addition, five registered wheat cultivars (Çeşit–1252, Gediz–75,Bezostaja–1, Tahirova–2000 and Sofu) were used as controls. Reaction types ofthe land races infected with a collection of powdery mildew spores obtainedfrom different locations and representing possible races of the pathogen weredetermined. As a result, only one land race (TR 31729), was found to have racespecific resistance., Buğdayın verimve kalitesini olumsuz yönde etkileyen en önemli hastalıklardan biriside Blumeria graminis f. sp. tritici’ninneden olduğu külleme hastalığıdır. Külleme, bahar döneminde nemin en yüksekolduğu bölgelerde sık görülen, ekonomik olarak ilaçlı mücadelesi olmayan birhastalıktır.Makarnalıkbuğdayın önemli gen kaynaklarından olan bazı yerel makarnalık buğdayçeşitlerinin küllemeye karşı dayanıklılıklarının belirlenmesi amacıylayürütülen bu araştırmada Türkiye’nin farklı bölgelerden toplanmış olan 49 adetyerel makarnalık buğday çeşidi ve kontrol amacıyla Çeşit 1252, Gediz 75,Bezostaja, Tahirova-2000 ve Sofu çeşitleri kullanılmıştır. Farklılokasyonlardan toplanan ve Türkiye’deki muhtemel ırkları temsil eden küllemesporları ile inoküle edilmiş yerel çeşitlerin hastalığa karşı gösterdiklerireaksiyon tipleri belirlenmiştir. Sonuç olarak küllemeye karşı bir yerelçeşidin (TR 31729) tek gen dayanıklılığına sahip olduğu tespit edilmiştir.

This study aimed to determine Clavibacter michiganensis subsp. michiganensis (Cmm) caused tomato bacterial cancer and wilt disease on tomato seedling used by farmers in Tokat province. In 2013, 2015, 2016, 2017 and 2018 years 104, 82, 94, 102 and 169 tomato seedling samples were taken from seedling providers located in Central, Turhal, Pazar, Niksar ve Erbaa in Tokat, respectively. The disease agent was detected on tomato seedling by specific polymerase chain reaction (PCR) assay after streaking on semiselective medium and extracting DNA of the bacteria growing on the medium. In 2013, 2015 and 2016, 19 out of 104, 7 out of 82 and 4 out of 94 tomato seedlings were infected with pathogen, respectively. On the other hand the disease agent was not detected in tomato seedlings samples collected in 2017 and 2018 years. According to results of this study, in seedling entered to the province, contamination ratio with Cmm decreased at important level., Bu çalışma,üreticiler tarafından Tokat ilinde kullanılan, hastalık belirtisi göstermeyendomates fidelerinde Domates bakteriyel kanser ve solgunluk hastalığı etmeni Clavibactermichiganensis subsp. michiganensis (Cmm)’in varlığının belirlenmesi amacıyla yapılmıştır. TokatMerkez, Turhal, Pazar, Niksar ve Erbaa ilçelerinden 2013 yılında 104, 2015yılında 82, 2016 yılında 94, 2017 yılında 102 ve 2018 yılında 169 adet domatesfide örneği toplanmıştır. Toplanan örneklerden hazırlanan süspansiyonlar yarıseçici besi yerine ekilmiş, gelişen bakteriler toplanarak DNA izolasyonundansonra etmene spesifik primerler ile polimeraz zincir reaksiyonu (PCR) analiziyapılmıştır. Yapılan çalışmasonucunda, 2013 yılında Tokat Merkez ve ilçelerinden toplanan 104 fide örneğinin19’unda, 2015 yılında 82 fide örneğinin 7’sinde, 2016 yılında 94 fide örneğinin4’ünde etmene rastlanılırken, 2017 yılında toplanan 102 fide örneğinde ve 2018yılında toplanan 169 fide örneğinde hastalık etmenine rastlanılmamıştır. Buçalışma sonuçlarına göre, bölgeye giriş yapan fidelerde Cmm ilebulaşıklık oranı önemli düzeyde azalmıştır.

Bu çalışma, Clavibacter michiganensis subsp. michiganensis (Cmm)’in tohum, toprak ve bitki artıklarındaki yaşam süresini belirlemek amacıyla yapılmıştır. Patojenin tohumdaki yaşam süresi için, rifampicin’e dayanıklı Cmm izolatı ile bulaştırılan tohumlar aylık periyotlarda besi yerlerine ekilerek, enfekteli tohum oranı belirlenmiştir. Ayrıca, tohumlardaki inokulumun hastalığı başlatma yeteneğini belirlemek amacıyla tohumlardan gelişen bitkilerde hastalık gözlemi yapılmıştır. Cmm’in topraktaki yaşam süresi için, Cmm ile bulaştırılan toprak, kutular içerisinde deneme alanına gömülmüş, yaz ve kış ayları boyunca alınan örneklerde bakteri yoğunluğu hesaplanmıştır. Bitki artıklarındaki yaşam süresi için, Cmm ile enfekteli bitki parçaları toprağa gömülmüş ve aylık periyotlarda örneklemeler yapılarak bakteri yoğunluğu hesaplanmıştır. Yapılan çalışma sonuçlarına göre, Cmm ile bulaştırılmış tohumlarda 0. gün %100’lük bir bulaşıklık belirlenirken, bu oran aylık periyotlarda düzenli olarak azalmıştır. En son 370. günde %17’lik bir bulaşıklık tespit edilirken, 400. günde etmene rastlanılmamıştır. Etmenin bulaşık topraklarda, yaz aylarında 15 gün, kış aylarında 30 gün canlı kalabildiği, toprakta bulunan bitki artıklarında ise 30 gün canlı kalabildiği belirlenmiştir. 12 aylık periyotta ilk ay patojen tespit edilirken, diğer aylarda etmen elde edilememiştir. Sonuç olarak, Tokat ilinde etmenin toprak ve bitki artıklarında canlılığını sürdüremediği, bulaşık tohumların birincil inokulum kaynağı oluşturduğu ve tohumlardaki inokulumun hastalığı başlatacak yeteneğe sahip olduğu belirlenmiştir. Bundan dolayı hastalık ile mücadelede başlangıç inokulumunu azaltmak için temiz tohum kullanımı önem arz etmektedir. As a result, it has been determined that Cmm could not survive in soil and plant residues in Tokat province. However, it has been determined that the infected seeds are primary inoculum source and that inoculum in the seeds has ability to initiate disease. Therefore it is important to use clean seeds to reduce initial inoculum for disease management.

The reaction of certain metal salts with N,N-bis(2-hydroxyethyl)ethylenediamine (bishydeten) and [Ag(CN)(2)](-) resulted in [Ni(bishydeten)(2)][Ag(CN)(2)](2)H2O (C1), [Cu(bishydeten)(2)Ag-2(CN)(4)] (C2) and [Cd(bishydeten)(0.5)](2)[Ag(CN)(2)](4)3H(2)O (C3), characterized by elemental, magnetic (C1 and C2), FT-IR, EPR (C2), ESI-MS, thermal, spectroscopic (C1, C2 or C3), and X-ray diffraction (C3) techniques. A detailed study of the antiproliferative and apoptotic properties of all the compounds and [Ag(CN)(2)](-) was carried out using the BrdU Cell Proliferation Assay (BCPA), DNA laddering assay and TUNEL method on HT29, HeLa, C6, and Vero cell lines. These compounds exhibiting a very strong antiproliferative effect even at low micromolar concentrations were found to have cytostatic activity, whereas [Ag(CN)(2)](-) had cytotoxic activity. The cells exposed to the compounds showed DNA laddering formations and the appearance of apoptotic bodies. Also, application of the test compounds at low concentrations was associated with a pronounced loss of the adhesive, migratory, and topoisomerase I activity of the treated tumor cell lines. Immunohistochemical staining analysis demonstrated that these complexes significantly reduced the level of Bcl-2 while causing the accumulation of P53 in HeLa and HT29 cells. Absorbance and emission spectral data reveal that the complexes interact with the DNA via a groove binding mode of interaction.

Viticulture is one of the most economically important agricultural sectors in Tokat province. One of the major problems of viticulture is virus diseases. The viral diseases can cause severe yield losses in grape production. In this study, the samples were collected from grapevines showing virus like symptoms from the vineyards in Tokat central, Zile, Erbaa, Niksar, Pazar and Turhal districts to identify and determine the prevalence of viruses included in the grapevine certification system and causing yield and quality losses in the grape varieties grown in Tokat province in 2015. The “Double Antibody Sandwich Enzyme Linked Immunosorbent Assay” (DAS-ELISA) technique was used to determine the presence of the following viruses caused significant yield losses in grapevines Strawberry latent ring spoty virus (SLRSV), Grapevine leafrol –associated virus 1(GLRAV-1), Grapevine fanleaf virus (GFLV), Arabis mosaic nepovirus (ArMV), Grapevine virus A (GVA), Grapevine fleck virus (GFkV) on the samples collected. Serological test revealed that the presence of GLRaV-1 (9.7%), GFLV (0.3%), ArMV (5.7%), SLRSV (4.7%) GFkV (0.7%) and GVA (5%) in the samples tested. The tests have also showed that the presence of mixed infection in a few samples collected from Central and Zile districts. Based on the results of the study, even if the incidence rates of viral agents are low, it is necessary to take measures to prevent the spread of viral agents and to raise the awareness of the producers. Bağcılık Tokat ilinde ekonomik açıdan önemli tarım kollarından biridir. Bağcılığın önemli sorunlarından biride virüs hastalıklarıdır. Virüs hastalıklarından dolayı üzümlerde önemli verim kayıpları yaşanmaktadır. Tokat ilinde yetiştiriciliği yapılan üzüm çeşitlerinde verim ve kalite kayıplarına neden olan ve sertifikasyon sistemi içerisinde yer alan bazı virüslerin Tokat ilinde varlığının belirlenmesi için 2015 yılında yapılan bu çalışmada, bağcılığın yoğun olarak yapıldığı Tokat Merkez, Zile, Erbaa, Niksar, Pazar ve Turhal ilçelerinde bağcılık yapılan işletmelerden hastalık belirtisi gösteren bitkilerden örnekler toplanmıştır. Toplanan örnekler, bağlarda önemli verim kayıplarına neden olan Grapevine leafroll-associated virus 1 (GLRaV-1), Grapevine fanleaf virus (GFLV), Strawberry latent ringspot virus (SLRSV),Arabis mosaic nepovirus (ArMV), Grapevine virus A (GVA), Grapevine fleck virus (GFkV) viral etmenlerinin varlığını belirlemek amacıyla “Double Antibody Sandwich Enzyme-Linked Immunosorbent Assay” (DAS-ELISA) yöntemi ile testlenmiştir. Test sonuçlarına göre toplanan örneklerde %9.7 GLRaV-1, %5.7 ArMV, %5 GVA, %4.7 SLRSV, %0.7 GFkV ve %0.3 GFLV oranlarında viral etmenlerin varlığı tespit edilmiştir. Yine Tokat Merkez ve Zile ilçelerinden alınan az sayıda örnekte de birden fazla virüsün yer aldığı karışık enfeksiyonlar belirlenmiştir. Çalışma sonuçlarına göre söz konusu viral etmenlerin enfeksiyon oranları düşük olmasına rağmen bölgede yayılmasını önlemeye yönelik tedbirlerin alınması ve üreticilerin bu konuda bilinçlendirilmesi gerekmektedir.

Karadeniz ve İç Anadolu bölgeleri arasında geçitkuşağında yer alan Tokat Türkiye sebze üretiminde önemli bir paya sahip olup,yörede üreticiler için sebzeler önemli bir gelir kaynağını oluşturmaktadır.Ancak son yıllarda yörede artan virüs enfeksiyonlarından dolayı sebzeüretiminde problemler yaşanmaktadır. Bu çalışmada Tokat ilinde yoğun olarakkabakgil (karpuz, kavun, kabak ve hıyar) yetiştirilen alanlarda sorun olanvirüslerin belirlenmesi amaçlanmıştır. Bu amaçla Tokat ilinin Merkez, Erbaa,Niksar, Pazar ve Turhal ilçelerindeki kabakgil alanlarında 2014 yılındagerçekleştirilen surveylerde; yapraklarda iplikleşme, kabarcıklaşma ve mozaik;bitkinin genelinde rozetleşme, gelişme geriliği ve bodurlaşma; meyvede şekilbozukluğu belirtisi gösteren bitkilerden toplam 571 yaprak örneği toplanmıştır.Alınan örnekler, Kabak sarı mozaik virüsü (Zucchini yellow mosaic virus;ZYMV), Hıyar mozaik virüsü (Cucumber mosaic virus; CMV), Karpuz mozaikvirüsü (Watermelon mosaic virus; WMV), Papaya halkalı leke virüsü (Papayaring spot virus; PRSV), Kabak mozaik virüsü (Squash mosaic virus; SqMV),Domates mozaik virüsü (Tomato mosaic virus; ToMV), Patates Y virüsü (Potatovirus Y; PVY) ve Tütün mozayik virüsü (Tobacco mosaic virus; TMV)’nespesifik antiserumlar kullanılarak double antibody sandwich enzyme-linkedimmunosorbent assay (DAS-ELISA) yöntemi ile test edilmiştir. Test sonuçlarına göre 571 örneğin %37’sinin WMV, %12’sinin ZYMV, %11’inin CMV, %3,4’ünün PRSV ve%0,2’sinin SqMV ile enfekteli olduğu saptanmıştır. ToMV, TMV ve PVY için toplam287 kabakgil örneği test edilmiş ve yaklaşık %14’ünün ToMV ile bulaşık olduğubelirlenmiştir. TMV ve PVY etmenlerine rastlanmamıştır. Tüm kabakgil türleriarasında virüsle bulaşıklık oranları;kabak %74, kavunda %72,4, karpuz %47,7 ve hıyar %31,7 olarakbelirlenmiştir.

Bu çalışma, hastalık belirtisi göstermeyen domates fidelerinde Domates bakteriyel solgunluk hastalığına sebep olan Clavibacter michiganensis subsp. michiganensis (Cmm)’in varlığı belirlenirken kullanılacak en uygun yöntemin belirlenmesi amacıyla yapılmıştır. Farklı Cmm konsantrasyonları ile domates fideleri bulaştırılmış ve farklı zaman dilimlerinde örneklemeler yapılarak patojenin varlığı genel bir besi yeri olan King B, yarı seçici besi yeri olan Selective Clavibacter Medium (SCM)’a ekim, serolojik (DAS-ELISA) ve moleküler (PCR) yöntemlerkullanılarak belirlenmeye çalışılmıştır. Çalışma sonunda, Cmm’in iletim demetlerinde ve yeşil aksamda bulaşık olduğu halde, genel besi yerine ekim ve DAS-ELISA testi olumlu sonuç vermemiştir. SCM’ye ekim ve gelişen bakterilerin toplanarak etmene spesifik PCR testleriyle, patojenin varlığı 24 saat sonra başarılı bir şekilde belirlenmiştir. Çalışma sonucunda belirlenen bu yöntemle domates fideleri bu patojenin varlığı yönünden incelenmiş olup, hastalığın bölgesel epidemiyolojisinde epifitik bulaşıklığın rolü ortaya konulmuştur., Thisstudy was conducted to determine the most appropriate method for determiningthe presence of causal agent of tomato bacterial wilt disease Clavibacter michiganensis subsp. michiganensis (Cmm) in symptomless tomato seedlings. Tomato seedlings wereinfected with different concentrations of Cmmand sampled at different time intervals. The presence of the pathogen ininfected seedling tried to determine by plating on King B medium and asemi-selective Clavibacter Medium (SCM) and by using serological (DAS-ELISA)and molecular (PCR) methods. At the end of the study, both methods (plating onKing B and DAS-ELISA) did not give any positive results even though the presenceof the pathogen in leaves and vascular bundles of infected tomato seedlings.The presence of the pathogen is determined successfully in infected tomatoseedling 24 hours post-inoculation by plating on SCM and performing specificPCR tests with bacteria grown on the medium. As a result of the study, tomatoseedlings were tested with this method in terms of the presence of thepathogen, revealing the role of epiphytic infection in the regionalepidemiology of the disease.

Bu çalışma, Antalya’nın modern seraalanları bakımından önemli bir konumda olan Serik ilçesinde yürütülmüştür.İlçede örtü altı yetiştiriciliği yapan üreticilerin, üretim sorunları ve busorunlara yönelik çözüm üretme sürecindeki bilgi düzeyleri ile ürünlerdekihastalık ve zararlılarla mücadele yöntemleri incelenmiştir.Araştırma Serikmerkez ilçe ve merkez ilçeye bağlı 10 köyde bulunan toplam 100 örtü altıüretici ile yüz yüze yapılan anketlerden elde edilen bilgileredayanmaktadır.Çalışmada ayrıca, Antalya ilinde örtü altı sebze üretiminde öneçıkan domates, biber, patlıcan ve hıyar ürünlerinde sorun olan hastalık vezararlılar ile mücadele yöntemleri hakkında bilgiler sunulmuştur.Araştırmabulgularından bazılarına göre, üreticilerin dörtte biri entegre mücadeleyöntemlerinden yararlandıklarını belirtirken dörtte üçü ise kimyasal mücadeleyaptıklarını ifade etmişlerdir.Diğer yandan üreticilerin, kimyasal mücadeleuygulamalarındaki güvenlik önlemlerinin alınması ve çevre konularındaçoğunlukla bilinçsiz oldukları belirlenmiştir.

Bu araştırma Erzincan’da M9 anacı üzerine aşılı 12 elma çeşidinin (Vista Bella, Williams Pride, Summer Red, Propolis, insanlar tarafından bin yıldır antimikrobiyal ve farmasötik özellikleri için kullanılmaktadır. Bununla birlikte, tarımsal alanda antimikrobial ajan olarak kullanımı son yıllarda ele alınmaya başlamıştır. Bu çalışmada Tokat ve Ankara’dan getirilen propolisin etanolik ekstraktının (PEE) ayva kahverengi çürüklük hastalığı (Monilinia fructigena Honey in Whetzel)'nın kontrolünde kullanılabilirliği araştırılmıştır. PEE'nin antifungal aktivitesi, propolis ekstraktı içeren Patates Dekstroz Agar ortamı ve meyve kaplama yöntemleri ile test edilmiştir. Çalışmada PEE üç farklı konsantrasyonu (%1, 3 ve 5) kullanılmıştır. Elde edilen sonuçlar doğrultusunda PEE konsantrasyonu artıkça etkinliğinde paralel olarak artığı görülmüştür. Tokat ve Ankara PEE %3 konsantrasyonuda M. fructigena’nın miselyum gelişimini %100 oranında engellemiştir. Tokat ve Ankara PEE %1 konsantrasyonuda patojenlerin gelişimini sırasıyla %84.2 ve %85.6 arasında değişen oranlarda engellemiştir. Ankara ve Tokat PEE, %5 konsantrasyonda patojenlerin gelişimini tamamen (%100) durdurmuştur. Benzer sonuçlar, in vivo çalışmada da elde edilmiştir. Her iki PEE %5 konsantrasyonunda, meyve enfeksiyonları tamamen engellenmiştir. Elde edilen sonuçlar, propolisin etanolik ekstraktlarının (PEE) %3 ve %5 lik konsantrasyonlarının, hasat sonrası ayva kahverengi çürüklük hastalığı (Monilinia fructigena)'nın kontrolü için ayvada yenilebilir kaplama olarak kullanılabileceğini göstermiştir Propolis has been used by man for millennia due to its antimicrobial and pharmaceutical properties. However, its use as an agricultural antimicrobial agent has only recently been assessed. This study assessed the use of an ethanolic extract of Tokat and Ankara propolis (PEE) for the control of quince fruit Brown rot disease. Antifungal activity of EEP was evaluated by amended Potato Dextrose Agar medium and fruit coating methods. Propolis Ethanolic Extract (PEE) in three concentrations 1, 3 and 5% were used. Based on the results of the study, increase in extract concentration resulted in increase of the extract efficacy. Tokat, and Ankara propolis ethanol extract at 3% concentration inhibited the growths of Monilinia fructigena at the rate of 100%. Tokat, and Ankara propolis ethanol extract at 1% concentration inhibited the growths of the pathogens between 84.2 and 85.6%. Ankara and Tokat propolis extracts at 5% concentration inhibited the growths of the pathogens at highest proportion (100%). Similar results were obtained in invivo study. At the 5% extract concentrations of both propolis extracts, fruit infections were inhibited completely. Present results indicated that propolis extracts (3% and 5% concentrations) could be used as an edible coating of quince fruits to eliminate postharvest brown rot disease.

Elma ürününde önemli kayıplara neden olan pek çok zararlı vardır. Yaprakbitleri elmanın önemli zararlıları arasındadır. Kırmızı gal yaprakbiti, Dysaphis devecta Walk. (Hemiptera: Aphididae), elmada önemli bir zararlı yaprakbiti türüdür. Mücadelesinde yoğun olarak insektisit kullanılmakta ve insektisitlere karşı kısa sürede dayanıklılık kazanmaktadırlar. İnsektisitlere alternatif mücadele yöntemerinden biride entomopatojen funguslardır. Bu çalışmada Tokat ilinden topraktan izole edilen entomopatojen fungus Beauveria bassiana Balls. (Vull.) izolatlarının kırmızı gal yaprakbiti üzerine etkisinin belirlenmesi amaçlanmıştır. Bu çalışmada 12 B. bassiana izolatının 1x108 spor/ml dozu püskürtme yöntemi ile uygulanmış ve 25±2 °C sıcaklık % 65±5 ortalama nem ve 16:8 Işık:Karanlık şartlarında üç gün süreyle inkübasyona bırakılmıştır. Birinci günde %81 ölüm oranı ile GOPT-297 nolu izolat öne çıkmıştır. Bunu %41’lik ölüm oarnı ile GOPT-64 nolu izolat takip etmiştir. İnkübasyon süresi arttıkça ölüm oranları da artmıştır. İki günlük inkübasyon süresi sonunda GOPT-297 nolu izolat %100 ölüme neden olurken bunu %79 ve %75 ile GOPT114 ve GOPT-282 nolu izolatlar takip etmiştir. Üç günlük inkübasyon süresi sonunda kontrol uygulamasında %11,6 oranında ölüm gerçekleşirken GOPT-332 nolu izolat hariç bütün izolatların neden olduğu ölüm oranı kontrolden istatistiki olarak önemli düzeyde yüksek bulunmuştur. Üç günlük inkübasyon süresi sonunda GOPT-297 nolu izolat %100 ölüm gerçekleştirirken, bunu %92 ile GOPT-282 ve %89.5 ile GOPT-284 nolu izolatlar takip etmiştir. İzolatların neden olduğu mikosis oranları da %6.2 ile 46,.8 arasında değişmiştir. Sonuç olarak entomopatojen funguslar Dysaphis devecta mücadelesinde kullanılma potansiyeline sahip olduğu ortaya konulmuştur. There are many pests that cause significant loss of apple yield. The aphids are among the major pests of the apple orchards. Dysaphis devecta Walk. (Hemiptera: Aphididae), rosy leaf curling aphid, an important pest species in apple. Insecticide is used extensively in the control of them and they gain resistance in a short time against insecticides. Alternative methods to insecticides in management of D. devecta is entomopathogenic fungi. The aim of the present study was to determine the effect of entomopathogenic fungus Beauveria bassiana Balls. (Vull.) isolates on rosy leaf curling aphid. In this study, 12 B. bassiana isolates were applied at 1x108 spores/ml concentration by sprey method and left to incubate for three days at 25±2°C temperature, %65±5 humidity and16:8 Light:Dark conditions. On the first day, the isolate GOPT-297 caused highest mortality with 81% mortality rate. This was followed by isolate GOPT-64 with 41% mortality. As the duration of incubation increased, the mortality rates increased. At the end of two days of incubation, GOPT-297 isolate resulted in 100% mortality, followed by isolates GOPT-114 and GOPT-282 with 79% and 75%, respectively. At the end of the three-day incubation period, the mortality rate of all isolates except for GOPT-332 were statistically significantly higher than control. At the end of the three-day incubation period, GOPT-297 isolate had 100% mortality followed by GOPT-282 with 92% and GOPT-284 with 89.5%. The percentage of mycosis caused by the isolates ranged from 6.2% to 46.58%. In conclusion, it can be said that the entomopathogenic fungi has the potential to be used in the rosy leaf curling aphid control.

Bu çalışmada, Manisa İli üzüm üretiminde karşılaşılan sorunlar ve üreticilerin hastalıklarla mücadele konusundaki bilgi ve tutumları araştırılmıştır. Araştırmada Manisa İli Turgutlu ilçesinde üzüm üretimi ile uğraşan 100 üretici ile anket çalışması yapılmıştır. Üreticilere yapılan anketlerle, bağ işletmelerindeki nüfus ve işgücü durumu, arazi varlıkları, yetiştiricilik, hastalık, zararlı, yabancı otları ve mücadelesi konularındaki bilgileri toplanmıştır. Anket sonuçları tablo ve grafiklerle sunularak yüzdelerle yorumlanmıştır. Araştırma sonucunda ortaya çıkan bulgulara göre üreticinin karşılaştıkları sorunlar tespit edilmiş çözüm önerileri getirilmiştir. Üreticilerle yapılan anket çalışması sonucunda; üreticilerin %49‟uüzümde zarar oluşturan etmenler hakkında hiçbir bilgiye sahip olmadığını bildirmiştir. Diğer taraftan %42‟si ilaçları ambalajları üzerinde belirtilen dozlarda kullandığını, %28‟i ise teknik elemanların önerdiği dozları kullandığını belirtmiştir. Üreticilerin %89‟u ilaçların uygulandığı zamanla hasat arasında geçmesi gereken süreye dikkat ettiklerini belirtmişlerdir. Üreticilerin çoğunluğu pestisitlerin insan ve çevreye olan olumsuz etkilerini bildiklerini ve bu olumsuz etkileri azaltmak için gerekli önlemleri aldıklarını belirtmişlerdir, Present study was aimed at evaluating the knowledge and attitudes of the farmers on plant protection issues encountered in vineyards of Manisa province. Qestionnaire survey was performed with 100 farmers dealing with vineyard production in Turgutlu, Manisa. With the questionnaire survey conducted with vineyard growers, information was collected on the growers‟ and labor population, land assets, production practices, disease, pest and pest management issues of the vineyard enterprises. Survey results are presented in tables and graphs and interpreted in percentage. According to the results of present study, the problems encountered by the vineyard growers have been identified and a solutions are proposed. Some of the results of the qestionnaire survey are; 49% of the producers stated that they did not have knowledges about the pests causing damages to vineyards. On the other hand, 42% of the growers use pesticide dosage, which is indicated on the label, and 28% of them use pesticide dosage according to comments of experts, 89% of them harvested their products obeying post harvest intervals which is indicated on the label instructions. Most of the growers know the negative effects of pesticides on human and environment and were taking necessary precautions to reduce their adverse effects

Bu çalışma Veratrum album L. (Adi çöpleme)’unbiyolojik mücadelesinde Stemphyliumvesicarium (Wallr.) Simmons’un etkinliğini belirlemek için yapılmıştır.Trabzon İli mera alanlarında 2009 ve 2010 yıllarında yapılan sürveyçalışmalarında V. album’unyapraklarında hastalık belirtileri gözlemlenmiştir. Hastalıklı bitkikısımlarından yapılan izolasyon sonucunda hastalık etmeninin S. vesicarium olduğu tespit edilmiştir. S. vesicarium’un biyolojik mücadelepotansiyelinin belirlenmesi için konukçuya özelleşme testleri ve biyolojiketkinlik çalışmaları yapılmıştır. Biyolojik etkinlik çalışmalarında ise S. vesicarium’un 5×105spor/mL konsantrasyonu V. album’a 3-4yapraklı döneminde uygulanmıştır. Bir aylık inkubasyon süresi sonunda S. vesicarium’un V. album üzerinde %75,25 oranında etkili olduğu tespit edilmiştir., This study wascarried out to determine the efficacy of the Stemphylium vesicarium (Wallr.) Simmons as a biological controlagent on the Veratrum album L.(falsehelleborine). In the survey study which is carried out on grasslands of Trabzonprovince in 2009-2010, symptoms of a disease were observed on V. album leaves. As a result of theisolation from diseased plant parts, S.vesicarium was identified. Host selectivity and biological efficiency testswere performed to determine biological control potential of S. vesicarium on false hellebore. V. albumseedling with 3-4 leaves stage has been sprayed with S. vesicarium 5×105spores/mL. Effectiveness of S. vesicariumon V. album at the endof one-month incubation period was 75.25%.
