Since the first plasma realized in 2020, a series of key systems on HL-3 (known as HL-2M before) tokamak have been equipped/upgraded, including in-vessel components (the first wall, lower divertor, and toroidal cryogenic/water-cooling/baking/glow discharge systems, etc.), auxiliary heating system of 11 MW, and 28 diagnostic systems (to measure the plasma density electron temperature, radiation, magnetic field, etc.). Magnet field systems were commissioned firstly for divertor plasma discharges. During the 2nd experimental campaign of HL-3 tokamak, several great progresses have been achieved. Firstly, the successful operation with plasma current larger than 1 MA was achieved under a divertor configuration. Secondly, the advanced divertor concept with two distinct snowflake configurations was realized. It is found that the distribution of ion saturation current and heat flux on bottom plate becomes wide due to magnetic surface expansion, demonstrating the advantage of such configuration in the heat flux mitigation. In addition, using the combination of NBI, ECRH and LHCD, the standard sawtoothing high confinement mode of megampere plasma was firstly accessed on the HL-3. The successful commissioning of HL-3 is beneficial for the initial operation of ITER.

Enhancements of residual Reynolds stresses and symmetry breaking with resonant magnetic perturbations (RMPs) are presented. These experiments were performed using a Langmuir probe array in the edge plasmas of the J-TEXT tokamak. This study aims to explain the mechanisms for generating residual stresses and intrinsic rotations. It is observed that with RMPs, peaked residual stresses, steeper pressure gradients and stronger shear layers all appear around the resonant surface. The toroidal rotation is accelerated in the direction of the plasma current and shows a correlation with turbulence and the measured residual stresses near the resonant surfaces of RMPs. Symmetry breaking induced by magnetic islands is proposed as a new mechanism, which qualitatively explains the enhanced residual stresses and the acceleration of the toroidal rotation. The changes to the residual stresses and symmetry breaking with RMPs near the last closed surface are also discussed.

The mission of HL-2A is to explore the key physical topics relevant to ITER and to the advanced tokamak operation (e.g. the operation of future HL-2M), such as the access of H-mode, energetic particle physics, edge-localized mode (ELM) mitigation/suppression and disruption mitigation. Since the 2016 Fusion Energy Conference, the HL-2A team has focused on the investigations on the following areas: (i) pedestal dynamics and L-H transition, (ii) techniques of ELM control, (iii) turbulence and transport, (iv) energetic particle physics. The HL-2A results demonstrated that the increase of mean shear flow plays a key role in triggering L-I and I-H transitions, while the change of flow is mainly induced by the ion pressure gradient. Both mitigation and suppression of ELMs were realized by laser blow-off seeded impurity (Al, Fe, W). The 30% Ne mixture supersonic molecular beam injection seeding also robustly induced ELM mitigation. The ELMs were mitigated by low-hybrid current drive. The stabilization of m/n = 1/1 ion fishbone activities by electron cyclotron resonance heating was found on the HL-2A. A new m/n = 2/1 ion fishbone activity was observed recently, and the modelling indicated that passing fast ions dominantly contribute to the driving of 2/1 fishbone. The non-linear coupling between the toroidal Alfven eigenmode and tearing mode (TM) led to the generation of a high frequency mode with the toroidal mode number n = 0. The turbulence was modulated by TM when the island width exceeds a threshold and the modulation is localized merely in the inner area of the islands. Meanwhile, turbulence radial spread took place across the island region.

The temporal-spatial structures of plasma flows and turbulence around tearing mode islands are presented. The experiments were performed using Langmuir probe arrays in the edge plasmas of J-TEXT tokamak. The correlation analyses clearly show that the flows have similar structures of m/n = 3/1 as the magnetic island does (m and n are the poloidal and toroidal mode numbers, respectively). The sign of the potential fluctuations for the flows inverses and the powers significantly reduce at q = 3 surface. Approaching to the last closed flux surface for the magnetic islands, the radially elongated flow structure forms. The flows are concentrated near separatrix and show quadrupole structures. The turbulence is concentrated near X-point and partly trapped inside the magnetic islands.

The synchronization of geodesic acoustic modes (GAMs) and magnetic fluctuations is identified in the edge plasmas of the HL-2A tokamak. Mesoscale electric fluctuations (MSEFs) having components of a dominant GAM, and m/n=6/2 potential fluctuations are found at the same frequency as that of the magnetic fluctuations of m/n=6/2 (m and n are poloidal and toroidal mode numbers, respectively). The temporal evolutions of the MSEFs and the magnetic fluctuations clearly show the frequency entrainment and the phase lock between the GAM and the m/n=6/2 magnetic fluctuations. The results indicate that GAMs and magnetic fluctuations can transfer energy through nonlinear synchronization. Such nonlinear synchronization may also contribute to low-frequency zonal flow formation, reduction of turbulence level, and thus confinement regime transitions.

The acceleration of the co-current toroidal rotations around resonant surfaces by resonant magnetic perturbations (RMPs) through turbulence is presented. These experiments were performed using a Langmuir probe array in the edge plasmas of the J-TEXT tokamak. This study aims at understanding the RMP effects on edge toroidal rotations and exploring its control method. With RMPs, the flat electron temperature T e profile, due to magnetic islands, appears around resonant surfaces (Zhao et al 2015 Nucl. Fusion 55 073022). When the resonant surface is closer to the last closed flux surface, the flat T e profile vanishes with RMPs. In both cases, the toroidal rotations significantly increase in the direction of the plasma current around the resonant surfaces with RMPs. The characteristics of turbulence are significantly affected by RMPs around the resonant surfaces. The turbulence intensity profile changes and the poloidal wave vector k θ increases with RMPs. The power fraction of the turbulence components in the ion diamagnetic drift direction increases with RMPs. The measurements of turbulent Reynolds stresses are consistent with the toroidal flows that can be driven by turbulence. The estimations of the energy transfer between the turbulence and toroidal flows suggest that turbulence energy transfers into toroidal flows. The result has the implication of the intrinsic rotation being driven by RMPs via turbulence.

The first comprehensive measurements of plasma flows and fluctuations nearby static magnetic islands driven by resonant magnetic perturbations are presented. These experiments were performed using multiple Langmuir probe arrays on the edge plasmas of the J-TEXT tokamak. Controlled variations of the island size and location are explored. This study aims to understand the interaction between turbulence and magnetic islands, and to elucidate magnetic island effects on edge turbulence and flow intensity profiles, edge electric fields, and thus confinement regime transitions. Turbulence and low frequency flows (LFFs) all drop inside the magnetic island, but increase at its boundary, as island width increases. The geodesic acoustic mode is damped in most of the edge area with magnetic islands. The sign of the radial electric field changes from negative to positive within the islands. The gradient of turbulent stresses vanishes at the island center, and becomes steeper at the boundaries of the islands. The particle transport induced by the turbulence is reduced inside the magnetic islands. The magnetic island effects on flows and turbulence can lead to an increase in LFFs and enhance Reynolds stresses near the last closed flux surface (LCFS). A stronger radial electric field layer can be formed near the LCFS when magnetic islands are present. The results suggest that magnetic islands can be used as a tool to enhance edge turbulence and flows, edge electric fields, and thus to trigger confinement regime transitions.

We report recent experimental results from HL-2A and KSTAR on ELM mitigation by supersonic molecular beam injection (SMBI). Cold particle deposition within the pedestal by SMBI is verified in both machines. The signatures of ELM mitigation by SMBI are an ELM frequency increase and ELM amplitude decrease. These persist for an SMBI influence time τI. Here, τI is the time for the SMBI influenced pedestal profile to refill. An increase in fELMSMBI/fELM0 and a decrease in the energy loss per ELM ΔWELM were achieved in both machines. Physical insight was gleaned from studies of density and vΦ (toroidal rotation velocity) evolution, particle flux and turbulence spectra, divertor heat load. The characteristic gradients of the pedestal density soften and a change in vΦ was observed during a τI time. The spectra of the edge particle flux Γ ∼ 〈ṽrñe〉 and density fluctuation with and without SMBI were measured in HL-2A and in KSTAR, respectively. A clear phenomenon observed is the decrease in divertor heat load during the τI time in HL-2A. Similar results are the profiles of saturation current density Jsat with and without SMBI in KSTAR. We note that τI/τp (particle confinement time) is close to ∼1, although there is a large difference in individual τI between the two machines. This suggests that τI is strongly related to particle-transport events. Experiments and analysis of a simple phenomenological model support the important conclusion that ELM mitigation by SMBI results from an increase in higher frequency fluctuations and transport events in the pedestal. © 2014 IAEA, Vienna.

Transitions between low (L-mode) and intermediate (I-phase) confinement regimes triggered by sawteeth are investigated using multiple Langmuir probe arrays in the edge plasmas of the HL-2A tokamak. The I-phase is characterized by limit-cycle oscillations (LCOs). Repeated L-I-L transitions induced by sawtooth heat pulses are also observed. Statistical analyses show that the delay time of the L-I transition relative to sawtooth crashes is less than ∼1 ms. The intensity of turbulence bursts in the I-phase is stronger than those observed in L-mode plasmas. Measurements are consistent with zonal flow reducing turbulence intensity in the frequency band ∼20-100 kHz, while the LCO turbulence of frequency higher than 100 kHz has more power than that in the L-mode. The analyses of time-resolved power transfers between turbulence and zonal flows suggest that power transfers exist between turbulence and zonal flows. The direction of the power transfers is from turbulence into geodesic acoustic modes in ohmic plasmas. © 2013 IAEA, Vienna.

For the first time supersonic molecular beam injection (SMBI) and cluster jet injection (CJI) were applied to mitigate edge-localized modes (ELMs) in HL-2A successfully. The ELM frequency increased by a factor of 2-3 and the heat flux on the divertor target plates decreased by 50% on average after SMBI or CJI. Energetic particle induced modes were observed in different frequency ranges with high-power electron cyclotron resonance heating (ECRH). The high frequency (200-350 kHz) of the modes with a relatively small amplitude was close to the gap frequency of the toroidicity-induced Alfvén eigenmode. The coexistent multi-mode magnetic structures in the high-temperature and low-collision plasma could affect the plasma transport dramatically. Long-lived saturated ideal magnetohydrodynamic instabilities during strong neutral beam injection heating could be suppressed by high-power ECRH. The absolute rate of nonlinear energy transfer between turbulence and zonal flows was measured and the secondary mode competition between low-frequency (LF) zonal flows (ZFs) and geodesic acoustic modes (GAMs) was identified, which demonstrated that ZFs played an important role in the L-H transition. The spontaneously generated E × B shear flow was identified to be responsible for the generation of a large-scale coherent structure (LSCS), which provided unambiguous experimental evidence for the LSCS generation mechanism. New meso-scale electric potential fluctuations (MSEFs) at frequency f ∼ 10.5 kHz with two components of n = 0 and m/n = 6/2 were also identified in the edge plasmas for the first time. The MSEFs coexisted and interacted with magnetic islands of m/n = 6/2, turbulence and LF ZFs. © 2013 IAEA, Vienna.

The radial structures of multiple sheared flow populations and fluctuations are simultaneously measured using combinations of Langmuir probe arrays in the edge plasmas of the HL-2A tokamak with ohmic and electron cyclotron resonance heating (ECRH). The maximum of geodesic acoustic mode (GAM) amplitude locates at ∼2-3 cm inside the last close flux surface (LCFS). The low-frequency zonal flow (LFZF) and GAM tend to coexist in the inner region. The dependences of the amplitudes of the multiple sheared flows and turbulence on ECRH heating power are also investigated. The measured turbulent Reynolds stresses are shown to be strongly correlated with the sheared flows, as predicted by theory. The turbulence is modulated at the each frequency of the multiple sheared flows simultaneously, and the particle fluxes induced by the turbulence are significantly reduced near the LCFS and in the GAM peaking region. The analyses also show that the spatial structures of the turbulent envelopes at the LFZF and GAM frequencies are similar to those observed in LFZF and GAM. © 2013 IAEA, Vienna.

Experiments on HL-2A, DIII-D and EAST show that turbulence just inside the last closed flux surface acts to reinforce existing sheared E × B flows in this region. This flow drive gets stronger as heating power is increased in L-mode, and leads to the development of a strong oscillating shear flow which can transition into the H-mode regime when the rate of energy transfer from the turbulence to the shear flow exceeds a threshold. These effects become compressed in time during an L-H transition, but the key role of turbulent flow drive during the transition is still observed. Abstract Density profiles in the pedestal region (H-mode) are measured in HL-2A and the characteristics of the density pedestal are described. Cold particle deposition by supersonic molecular beam injection (SMBI) within the pedestal is verified. Edge-localized mode (ELM) mitigation by SMBI into the H-mode pedestal is demonstrated and the relevant physics is elucidated. The sensitivity of the effect to SMBI pressure and duration is studied. Following SMBI, the ELM frequency increases and the ELM amplitude decreases for a finite duration. Increases in ELM frequency of are achieved. This experiment argues that the ELM mitigation results from an increase in higher frequency fluctuations and transport events in the pedestal, which are caused by SMBI. These inhibit the occurrence of large transport events which span the entire pedestal width. The observed change in the density pedestal profiles and edge particle flux spectrum with and without SMBI supports this interpretation. An analysis of the experiment and a model shows that ELMs can be mitigated by SMBI with shallow particle penetration into the pedestal.

The absolute rate of nonlinear energy transfer among broadband turbulence, low-frequency zonal flows (ZFs) and geodesic acoustic modes (GAMs) was measured for the first time in fusion-grade plasmas using two independent methods across a range of heating powers. The results show that turbulent kinetic energy from intermediate frequencies (20-80 kHz) was transferred into ZFs and GAMs, as well as into fluctuations at higher frequencies (>80  kHz). As the heating power was increased, the energy transfer from turbulence into GAMs and the GAM amplitudes increased, peaked and then decreased, while the energy transfer into the ZFs and the ZFs themselves increased monotonically with heating power. Thus there exists a competition between ZFs and GAMs for the transfer of turbulent energy, and the transfer into ZFs becomes dominant as the heating power is increased. The poloidal-radial Reynolds stress and the mean radial electric field profiles were also measured at different heating powers and found to be consistent with the energy transfer measurement. The results suggest that ZFs play an important role in the low-to-high (L-H) plasma confinement transition.

The present status of experiments on zonal flows in magnetic confinement experiments is examined. The innovative use of traditional and modern diagnostics has revealed unambiguously the existence of zonal flows, their spatio-temporal characteristics, their relationship to turbulence and their effects on confinement. In particular, a number of observations have been accumulated on the oscillatory branch of zonal flows, named geodesic acoustic modes, suggesting the necessity for theories to give their proper description. In addition to these basic properties of zonal flows, several new methods have elucidated the processes of zonal flow generation from turbulence. Further investigation of the relationship between zonal flows and confinement is strongly encouraged as cross-device activity including low temperature, toroidal and linear devices. © 2007 IAEA.

The imbalance of redox homeostasis is a major characteristic of aging and contributes to the pathogenesis of various aging-related diseases. As a regulatory hub of redox homeostasis, nuclear factor erythroid 2-related factor 2 (NRF2) can attenuate oxidative stress by activating the transcription of many antioxidant enzymes. China is the birthplace of traditional Chinese medicine (TCM) which has been wildly used as medicine for thousands of years. Recently, TCM as anti-aging medicine has attracted enormous attention. Focusing on the NRF2 signaling pathway, this paper summarizes the correlation between various anti-aging TCM and the NRF2 signaling, and discusses the common key mechanisms by which TCM slows the aging process by targeting the NRF2 signaling network.

To explore the situation of 8 common respiratory pathogens in children with acute respiratory infection (ARI) from 2021 to 2022.The retrospective study selected 8 710 ARI patients from September 2021 to August 2022 in the Maternal and Child Health Hospital of Gansu Province as the study object, patients aged 0 to 17 years old, including 5 048 male children and 3 662 female children. Indirect immunofluorescence was used to detect 8 common respiratory pathogens, including influenza virus A (FluA), influenza virus B (FluB), parainfluenza virus (PIV), respiratory syncytial virus (RSV), adenovirus (ADV), Mycoplasma pneumoniae (MP), Chlamydia pneumoniae (CP), and Coxsackie virus group B (CoxB) IgM antibodies. χ 2 test was used to analyze the results. The results showed that 1 497 of 8 710 children with ARI were positive, with a positive rate of 17.19%. The detection rate of MP among 8 common respiratory pathogens was 11.34%, accounting for 66.0%, followed by FluB, CoxB, PIV, RSV, ADV, FluA and CP, accounting for 13.83%, 9.55%, 6.01%, 2.61%, 1.47%, 0.40% and 0.13%, respectively. Respiratory tract viruses (FluA, FluB, RSV, ADV, PIV, CoxB) accounted for 33.86%.There were significant differences in the detection rates of PIV, ADV and MP among children of different genders (χ 2 =6.814, 5.154 and 17.784, P <0.05). The detection rate of school-age children (6-17 years old) was the highest, accounting for 33.27% (184/553). The detection rates of 8 common respiratory pathogens in patients with ARI were higher in spring and winter and lower in summer and autumn. To sum up, from 2021 to 2022, MP and FluB infection were dominant in ARI patients in our hospital. The peak period of 8 common respiratory pathogens was in spring and winter. The physical examination rate of 8 common respiratory pathogens in ARI patients aged 6-17 years old was the highest.

Objective: To investigate the effect of phorbol-12-myristate-13-ace-tate (TPA) on the proliferation and apoptosis of acute promyelocytic leukemia cell line NB4 and its molecular mechanism., Methods: The effect of different concentrations of TPA on the proliferation of NB4 cells at different time points was detected by CCK-8 assay. The morphological changes of NB4 cells were observed by Wright-Giemsa staining. The cell cycle and apoptosis of NB4 cells after TPA treatment were detected by flow cytometry. The mRNA expressions of NB4 cells after TPA treatment were analyzed by high-throughput microarray analysis and real-time quantitative PCR. Western blot was used to detect the protein expression of CDKN1A, CDKN1B, CCND1, MYC, Bax, Bcl-2, c-Caspase 3, c-Caspase 9, PIK3R6, AKT and p-AKT., Results: Compared with the control group, TPA could inhibit the proliferation of NB4 cells, induce the cells to become mature granulocyte-monocyte differentiation, and also induce cell G 1 phase arrest and apoptosis. Differentially expressed mRNAs were significantly enriched in PI3K/AKT pathway. TPA treatment could increase the mRNA levels of CCND1, CCNA1 , and CDKN1A , while decrease the mRNA level of MYC . It could also up-regulate the protein levels of CDKN1A, CDKN1B, CCND1, Bax, c-Caspase 3, c-Caspase 9, and PIK3R6, while down-regulate MYC, Bcl-2, and p-AKT in NB4 cells., Conclusion: TPA induces NB4 cell cycle arrest in G 1 phase and promotes its apoptosis by regulating PIK3/AKT signaling pathway.

Ovarian cancer (OC) represents one of the most detrimental gynecological malignancies. RNA-binding protein eukaryotic translation initiation factor 4A isoform 3 (EIF4A3) is well-regarded as a definitive oncogene that contributes to the development of multiple malignant tumors. This study sought to elucidate the molecular mechanism of EIF4A3 in OC growth and aerobic glycolysis by regulation of pyruvate dehydrogenase kinase 4 (PDK4) mRNA stability. We determined the EIF4A3 and PDK4 expression levels in OC cell lines and normal ovarian epithelial cells, and subsequently evaluated the cell viability and colony formation by cell counting kit-8 and colony formation assays. The degree of cell aerobic glycolysis was evaluated by measurements of lactic acid production, glucose intake, adenosine triphosphate level, extracellular oxygen consumption, and protein levels of pyruvate kinase isozymes M2 and hexokinase-2. Afterwards, we verified the binding of EIF4A3 and PDK4 mRNA via RNA immunoprecipitation, and determined the mRNA stability after actinomycin D treatment. Finally, a series of rescue experiments was performed with pcDNA3.1-PDK4. EIF4A3 and PDK4 were upregulated in OC cells. Silencing EIF4A3 obstructed cell proliferation and aerobic glycolysis, while the same was annulled by EIF4A3 overexpression. Mechanically, EIF4A3 could bind to PDK4 mRNA to stabilize its mRNA and upregulate its protein levels. PDK4 overexpression inverted the inhibitory role of silencing EIF4A3 in proliferation and aerobic glycolysis. Overall, our findings highlighted that EIF4A3 induced OC progression by stabilizing PDK4 mRNA., (© 2023 The Authors. Traumatic painful neuroma is an intractable clinical disease characterized by improper extracellular matrix (ECM) deposition around the injury site. Studies have shown that the microstructure of natural nerves provides a suitable microenvironment for the nerve end to avoid abnormal hyperplasia and neuroma formation. In this study, we used a decellularized nerve matrix scaffold (DNM-S) to prevent against the formation of painful neuroma after sciatic nerve transection in rats. Our results showed that the DNM-S effectively reduced abnormal deposition of ECM, guided the regeneration and orderly arrangement of axon, and decreased the density of regenerated axons. The epineurium-perilemma barrier prevented the invasion of vascular muscular scar tissue, greatly reduced the invasion of α-smooth muscle actin-positive myofibroblasts into nerve stumps, effectively inhibited scar formation, which guided nerve stumps to gradually transform into a benign tissue and reduced pain and autotomy behaviors in animals. These findings suggest that DNM-S-optimized neuroma microenvironment by ECM remodeling may be a promising strategy to prevent painful traumatic neuromas., Competing Interests: None

The wide-angle view imaging system, in terms of a tangential view diagnostic with field of view (FOV) of 56.8° and a downward-looking diagnostic from the top of the machine with FOV of 94.7°, has been newly constructed for the first plasma of the HL-2M tokamak achieved in December 2020. Its mission in this stage is to monitor the plasma evolution during its startup, sustainment, and disruption in the visible spectral range as well as the plasma-wall interaction. For the latter ultrawide view diagnostic, nearly three-quarters of the divertor region and half the area of the inner wall are in the view range. Both the diagnostics are characterized by a similar optical structure, i.e., the light emission from the plasma is collected by a front-end lens and transferred through an imaging fiber bundle to the camera. This optical structure is suitable for application in the complex tokamak environment mainly because the fiber bundle is flexible. Photos of glow discharges are acquired prior to the first plasma for testing the FOVs in the vacuum vessel. The spatial resolution is ∼4 m m for the tangential view diagnostic and ∼10 m m for the downward-looking diagnostic. The temporal resolutions, ranging from 90 to 360 Hz by changing the region of interest or binning acquisition mode of the color camera, are applied to record the plasma evolutions and/or dust creation events during the first plasma campaign.

Objective: To study the effects of miR-16-5p on proliferation, migration and invasion of osteosarcoma cells and its mechanism. Methods: Quantitative polymerase chain reaction (qPCR) and Western blotting were used to detect the mRNA and protein expression of miR-16-5p and TSPAN15 in human normal osteoblasts hFOB 1.19 and osteosarcoma cells MG63, Saos2 and HOS. The miR-16-5p or si-TSPAN15 was transfected into MG63 cells to observe its role in cell proliferation, migration and invasion. Cell proliferation was measured with MTT assay, cell migration and invasion were examined by Transwell, and the protein expression of CyclinD1, matrix metalloproteinase 2 (MMP-2), MMP-9, tetraspanin 15 (TSPAN15), phospha-tidylinositol3-kinase(p-PI3K) and phospha-protein kinase B(p-AKT) were determined by using Western blotting. The starbase website prediction combined with dual luciferase gene reporter assay was performed to analyze the targeting relationship between miR-16-5p and TSPAN15. miR-16-5p and pcDNA-TSPAN1 were co-transfected to assess the effect of high expression of TSPAN15 on overexpression of miR-16-5p-induced proliferation, migration and invasion of MG63 cells. Data comparison between the two groups was performed by using t test. Results: Compared with hFOB 1.19 cells (1.00±0.12), the expression of miR-16-5p was significantly decreased in MG63, Saos2 and HOS cells (0.32±0.05, 0.40±0.04, 0.45±0.06, respectively)( F= 156.204, P< 0.05), and TSPAN15 mRNA and protein levels were greatly increased ( F= 71.718, 110.350, both P< 0.05). Overexpression of miR-16-5p obviously reduced the expression of CyclinD1, MMP-2, MMP-9 protein, cell viability, cell migration and invasion ( F= 150.136,117.228, 154.971, 89.479, 98.373, 130.880, all P< 0.05) in MG63 cells. Knockdown of TSPAN15 greatly reduced CyclinD1, MMP-2, MMP-9 protein levels, cell survival rate, cell migration, and invasion number ( F= 93.206, 107.030, 109.326, 115.625, 146.113, 139.300, all P< 0.05). Overexpression of miR-16-5p markedly decreased the expression of p-PI3K and p-AKT protein in MG63 cells ( F= 156.755, 181.419, both P< 0.05). miR-16-5p targeted to regulate the expression of TSPAN15. High expression of TSPAN15 partially reversed the inhibitory effect of miR-16-5p on TSPAN15, CyclinD1, MMP-2, MMP-9, p-PI3K, p-AKT protein expression, cell viability, cell migration number and invasion number in MG63 cells. Conclusion: miR-16-5p inhibits the proliferation, migration and invasion of osteosarcoma cells by targeting the TSPAN15 gene and regulating the PI3K/AKT signaling pathway.

A Fast Charge eXchange Recombination Spectroscopy (CXRS) diagnostic with eight radial channels has been implemented on a HuanLiu-2A (HL-2A) tokamak with a time resolution of up to 10 kHz monitoring helium II spectra or 1 kHz monitoring carbon VI spectra. The crucial aspects of the fast CXRS are to improve the spectral intensity and the acquisition frequency. The spectral intensity has been greatly enhanced by customized fiber bundles. The main boost in optimizing the acquisition frequency is achieved by binning more pixel rows of the charge coupled device (CCD) representing one radial channel and by reducing the effective image area of the CCD. Consequently, the sawtooth oscillations of ion temperature and rotation velocity are continuously observed for the first time in the HL-2A tokamak.

Objective: The use of a biofabrication nerve scaffold, which mimics the nerve microstructure, as an alternative for autologous nerve transplantation is a promising strategy for treating peripheral nerve defects. This study aimed to design a customized biofabrication scaffold model with the characteristics of human peripheral nerve fascicles., Methods: We used Micro-MRI technique to obtain different nerve fascicles. A full-length 28 cm tibial nerve specimen was obtained and was divided into 14 two-centimetre nerve segments. 3D models of the nerve fascicles were obtained by three-dimensional reconstruction after image segmentation. The central line of the nerve fascicles was fitted, and the aggregation of nerve fascicles was analysed quantitatively. The nerve scaffold was designed by simulating the clinical nerve defect and extracting information from the acquired nerve fascicle data; the scaffold design was displayed by 3D printing to verify the accuracy of the model., Result: The microstructure of the sciatic nerve, tibial nerve, and common peroneal nerve in the nerve fascicles could be obtained by three-dimensional reconstruction. The number of cross fusions of tibial nerve fascicles from proximal end to distal end decreased gradually. By designing the nerve graft in accordance with the microstructure of the nerve fascicles, the 3D printed model demonstrated that the two ends of the nerve defect can be well matched., Conclusion: The microstructure of the nerve fascicles is complicated and changeable, and the spatial position of each nerve fascicle and the long segment of the nerve fascicle aggregation show great changes at different levels. ArgR, a transcriptional regulator belonging to the AraC/XylS family, plays a key role in arginine metabolism regulation. ArgR has also been found to repress the transcription of a lipase gene, but its molecular mechanism is still unknown. In this study, we investigated the molecular mechanism acting on the expression of intracellular lipase gene lipA regulated by ArgR in Pseudomonas protegens Pf-5 through knockout and overexpression of argR, detection of DNA-protein interaction in vivo, determining whole-cell lipase activities of various strains derived from Pf-5, and examining β-galactosidase activities of various lacZ fusions. The results demonstrated that ArgR inhibits lipA expression at the transcriptional level. Further results showed that the inhibition of lipA transcription by ArgR is mediated by binding to the ArgR binding site of lipA promoter to produce steric hindrance, in which the common sequence, TGTCGC is crucial for the ArgR binding. Besides, arginine inhibits lipA expression in both wild-type and argR mutant, and shows a synergistic inhibition on lipA expression when combined with ArgR. The intermingling of regenerated nerve fibers inside nerve grafts is the main reason for mismatched nerve fibers. This is one of the key factors affecting limb function recovery after nerve injury. Previous research has shown that the accuracy of axon regeneration can be improved by a bionic structural implant. To this aim, iodine and freeze-drying high-resolution micro-computed tomography was performed to visualize the 3D topography of the New Zealand rabbit sciatic nerve (25 mm). A series of 1-, 2-, 3-, and 4-custom anatomy-based nerve conduits (CANCs) were fabricated based on the anatomical structure of the nerve fascicle. The match index, luminal surface, and mechanical properties of CANCs were evaluated before implanting in a 10-mm gap of the sciatic nerve. Recovery was evaluated by histomorphometric analyses, electrophysiological study, gastrocnemius muscle weight recovery ratio, and behavioral assessments at 12 and 24 weeks postoperatively. The accuracy of nerve regeneration was determined by changes in fluorescence-labeled profile number during simultaneous retrograde tracing. Our results showed that the optimal preprocessing condition for high-resolution micro-computed tomography visualization was treatment of the sciatic nerve with 40% Lugol's solution for 3 days followed by lyophilization for 2 days. In vitro experiments demonstrated that the match index was highest in the 3-CANC group, followed by the 2-, 1-, and 4-CANC groups. The luminal surface was lowest in the 1-CANC group. Mechanical properties (transverse compressive and bending properties) were higher in the 3- and 4-CANC groups than in the 1-CANC group. In vivo experiments demonstrated that the recovery (morphology of regenerated fibers, compound muscle action potential, gastrocnemius muscle weight recovery ratio, pain-related autotomy behaviors, and range of motion) in the 3-CANC group was superior to the other CANC groups, and achieved the same therapeutic effect as the autograft. The simultaneous retrograde tracing results showed that the percentages of double-labeled profiles of the 2-, 3-, and 4-CANC groups were comparatively lower than that of the 1-CANC group, which indicates that regenerated nerve fascicles were less intermingled in the 2-, 3-, and 4-CANC groups. These findings demonstrate that the visualization of the rabbit sciatic nerve can be achieved by iodine and freeze-drying high-resolution micro-computed tomography, and that this method can be used to design CANCs with different channels that are based on the anatomical structure of the nerve. Compared with the 1-CANC, 3-CANC had a higher match index and luminal surface, and improved the accuracy of nerve regeneration by limiting the intermingling of the regenerated fascicles. A 7-channel motional Stark effect diagnostic based on dual photo-elastic modulators is installed and operated routinely for rather low beam energy and magnetic field on the HL-2A tokamak, with a spatial resolution of ∼3 cm and a temporal resolution of 10 ms. The instrument observes the σ component of the full energy D α from the first or the fourth ion source of a neutral beam injector. However, the change in beam energy during a discharge causes variation of the Doppler shift with the maximum of 1 Å, which leads to the polarization fraction drop from 30%-40% to 10% and then makes the signal-to-noise ratio of the system become very poor. Therefore, a real-time wavelength matching system is designed to promote polarization fraction. The beam emission spectra are filtered by using a monochrometer in real time. And a narrowband filter is tilted by using an absolutely calibrated rotator through beam energy in order to make sure that the deviation of wavelength matching is less than 0.1 Å.

The most common methods for three-dimensional reconstruction of peripheral nerve fascicles include histological and radiology techniques. Histological techniques have many drawbacks including an enormous manual workload and poor image registration. Micro-magnetic resonance imaging (Micro-MRI), an emerging radiology technique, has been used to report results in the brain, liver and tumor tissues. However, micro-MRI usage for obtaining intraneural structures has not been reported. The aim of this study was to present a new imaging method for three-dimensional reconstruction of peripheral nerve fascicles by 1 T micro-MRI. Freshly harvested sciatic nerve samples from an amputated limb were divided into four groups. Two different scanning conditions (Mannerist Solution/GD-DTPA contrast agent, distilled water) were selected, and both T1 and T2 phases programmed for each scanning condition. Three clinical surgeons evaluated the quality of the images via a standardized scale. Moreover, to analyze deformation of the two-dimensional image, the nerve diameter and total area of the micro-MRI images were compared after hematoxylin-eosin staining. The results show that rapid micro-MRI imaging method can be used for three-dimensional reconstruction of the fascicle structure. Nerve sample immersed in contrast agent (Mannerist Solution/GD-DTPA) and scanned in the T1 phase was the best. Moreover, the nerve sample was scanned freshly and can be recycled for other procedures. MRI images show better stability and smaller deformation compared with histological images. Background/objectives: Complex extremity trauma commonly involves both soft tissue and vascular injuries. Traditional two-stage surgical repair may delay rehabilitation and functional recovery, as well as increase the risk of infections. We report a single-stage reconstructive surgical method that repairs soft tissue defects and vascular injuries with flow-through free flaps to improve functional outcomes., Methods: Between March 2010 and December 2016 in our hospital, 5 patients with severe upper extremity trauma received single-stage reconstructive surgery, in which a flow-through anterolateral thigh free flap was applied to repair soft tissue defects and vascular injuries simultaneously. Cases of injured artery were reconstructed with the distal trunk of the descending branch of the lateral circumflex femoral artery. A segment of adjacent vein was used if there was a second artery injury. Patients were followed to evaluate their functional recoveries, and received computed tomography angiography examinations to assess peripheral circulation., Results: Two patients had post-operative thumb necrosis; one required amputation, and the other was healed after debridement and abdominal pedicle flap repair. The other 3 patients had no major complications (infection, necrosis) to the recipient or donor sites after surgery. All the patients had achieved satisfactory functional recovery by the end of the follow-up period. Computed tomography angiography showed adequate circulation in the peripheral vessels., Conclusions: The success of these cases shows that one-step reconstructive surgery with flow-through anterolateral thigh free flaps can be a safe and effective treatment option for patients with complex upper extremity trauma with soft tissue defects and vascular injuries., (Copyright © 2017. Silver nanorods/polyimide (Ag-NRs/PI) nanocomposites with high conductivity (An order of magnitude higher than pure PI), frequency-independent dielectric permittivity (3.8-4.2) and low dielectric loss (<0.05) were prepared by an in-situ polymerization process. Ag-nanorods with a mean width of approximately 300 nm and an average length over 8 microm were synthesized in the presence of polyvinylpyrrolidone (PVP) and NaCl by polyol process. SEM images showed that metallic Ag-nanorods were well dispersed in PI matrix. The structure of Ag-NRs was not destroyed or changed in nanocomposite films and the order of PI molecular chains was maintained as well. The orientation of the Ag-NRs in the PI matrix improved the mechanical properties of nanocomposite films. TGA results showed that the thermal property of nanocomposite films was almost as good as pure PI.

This paper studies the Czerny-Turner optical structure which is used for the application in imaging spectrometers. To obtain the perfect astigmatism-corrected condition, the Czerny-Turner system has been analyzed and advanced. The basic structure of optical system is still as the traditional form which is composed by the spherical collimating mirror, the plane grating and the spherical focusing mirror. However, an off-the-shelf cylindrical lens is added after the focusing mirror to remove astigmatism differences between the tangential direction and the sagittaI direction. It makes the advanced optical system presents high resolution over the full bandwidth and decreases the cost. An example of the imaging spectrum system in the waveband of 380-760 nm has been designed to prove our theory. A system owns that NA equals to 0.05, and the modulation transfer functions (MTF) of all fields of view are more than 0.59 over the broadband under the required Nyquist frequency (20 lp x mm(-1)). It certificates that the optical system theory can be applied to the small scale imaging spectrometer with high resolution in spectral broadband.

A 32/64-channel Charge eXchange Recombination Spectroscopy (CXRS) and a 7-channel motional Stark effect (MSE) polarimeter have been developed on the HL-2A tokamak. To extract the maximum time resolution of the system, the incidence fibers of the spectrometer are pitch-controlled; and the double-slit fiber bundle can increase the spatial channels with one charge-coupled device detector. The ion temperature and plasma rotation with time and spatial resolutions up to 5 ms and 1 cm are obtained. Sawtooth oscillation, transition from intermediate phase (I phase) to high confinement mode (H mode) can be clearly observed by the CXRS. The spectrometer can be utilized as the main component of the MSE polarimeter, which can effectively overcome the weak Stark effect. The pitch angles of magnetic field are obtained for 7 spatial points covering 24 cm along major radius with time resolution of 40 ms.

A 32/64-channel charge exchange recombination spectroscopy (CXRS) diagnostic system is developed on the HL-2A tokamak (R = 1.65 m, a = 0.4 m), monitoring plasma ion temperature and toroidal rotation velocity simultaneously. A high throughput spectrometer (F/2.8) and a pitch-controlled fiber bundle enable the temporal resolution of the system up to 400 Hz. The observation geometry and an optimized optic system enable the highest radial resolution up to ∼1 cm at the plasma edge. The CXRS system monitors the carbon line emission (C VI, n = 8-7, 529.06 nm) whose Doppler broadening and Doppler shift provide ion temperature and plasma rotation velocity during the neutral beam injection. The composite CX spectral data are analyzed by the atomic data and analysis structure charge exchange spectroscopy fitting (ADAS CXSFIT) code. First experimental results are shown for the case of HL-2A plasmas with sawtooth oscillations, electron cyclotron resonance heating, and edge transport barrier during the high-confinement mode (H-mode).

The design of the probe is based on the concept of the α-particle detectors on Tokamak Fusion Test Reactor (TFTR) using scintillator plates. The probe is capable of traveling across an equatorial plane port and sweeping the aperture angle rotationally with respect to the axis of the probe shaft by two step motors, in order to optimize the radial position and the collimator angle. The energy and the pitch angle of the lost fast ions can be simultaneously measured if the two-dimensional image of scintillation light intensity due to the impact of the lost fast ions is detected. Measurements of the fast-ion losses using the probe have been performed during HL-2A neutral beam injection discharges. The clear experimental evidence of enhanced losses of beam ions during disruptions has been obtained by means of the SLIP system. A detailed description of the probe system and the first experimental results are reported.

A 7-channel motional Stark effect polarimeter based on four polarizers and a spectrometer has been developed in the HL-2A tokamak, which is the first time successful utilizing this kind of polarimeter on a tokamak. The accuracy of the angle can reach ±0.25° in the calibration experiments. Pilot experiments of measuring the magnetic pitch angle have been successfully carried out in the weak motional Stark effect plasma discharge with toroidal magnetic field of ~1.3 T and beam energy of ~25 keV/amu. The pitch angles of magnetic field are obtained for 7 spatial points covering 24 cm along major radius with time resolution of 40 ms; the profiles of safety factor are obtained by combining with the Equilibrium and Reconstruction Fitting Code. The core value of safety factor (q) is less than 1 during the sawtooth oscillation and the position of q = 1 surface is well consistent with the results measured by soft X-ray array.

The paper designs and improves a telecentric imaging spectrometer, the Dyson imaging spectrometer. The optical structure of the imaging spectrometer is simple and compact, which is only composed of a hemispherical lens and a concave grating. Based on the Rowland circle and refraction theory, the broadband anastigmatic imaging condition of Dyson imaging spectrometer which is the ratio of the grating radius and hemispherical lens radius has been analyzed. By imposing this condition for two different wavelengths, the parameters of the optical system presenting low aberrations and excellent imaging quality are obtained. To make the design spectrometer more suitable for the engineering application, the paper studies the method making the detector not to attach the surface of the hemispherical lens. A design example using optimal conditions was designed to prove our theory. The Dyson imaging spectrometer of which the imaging RMS radii are less than 2.5 microm and the advanced spectrometer of which the imaging RMS radii are less than 8 microm, with NA 0.33, waveband 0.38-1.7 microm and the slit length 15 mm, have been obtained. The design method and results are more feasible and predominant, and can be applied in the areas of the industry and remote sensing.

The frequency modulated continuous wave reflectometer was developed for the first time on the HL-2A tokamak. The system utilizes a voltage controlled oscillator and an active multiplier for broadband coverage and detects as heterodyne mode. Three reflectometers have been installed and operated in extraordinary mode polarization on HL-2A to measure density profiles at low field side, covering the Q-band (33-50 GHz), V-band (50-75 GHz), and W-band (75-110 GHz). For density profile reconstruction from the phase shift of the probing wave, a corrected phase unwrapping method is introduced in this article. The effectiveness of the method is demonstrated. The density profile behavior of a fast plasma event is presented and it demonstrates the capability of the reflectometer. These diagnostics will be contributed to the routine density profile measurements and the plasma physics study on HL-2A.

The dynamic features of the low-intermediate-high-(L-I-H) confinement transitions on HL-2A tokamak are presented. Here we report the discovery of two types of limit cycles (dubbed type-Y and type-J), which show opposite temporal ordering between the radial electric field and turbulence intensity. In type-Y, which appears first after an L-I transition, the turbulence grows first, followed by the localized electric field. In contrast, the electric field leads type-J. The turbulence-induced zonal flow and pressure-gradient-induced drift play essential roles in the two types of limit cycles, respectively. The condition of transition between types-Y and -J is studied in terms of the normalized radial electric field. An I-H transition is demonstrated to occur only from type-J.

The Choi-Williams distribution (CWD) technique is introduced as a time-frequency tool for processing data measured from the new developed homodyne and the fixed frequency reflectometry in the HL-2A tokamak. The comparison between spectrogram and CWD for the simulated signal is presented. It indicates that the CWD can greatly improve the representation of the time-frequency content of the multi-components signal. Its effectiveness is demonstrated through two applications in HL-2A, which are the extraction of beat frequencies from the frequency modulated-continuous wave reflectometry (FM-CW) and the characterizing of the fluctuations. The density profile inversed from the group delay of the FM-CW and the density fluctuations deduced from the fixed-frequency reflectometry would be more reliable and accurate by using the CWD technique.

A recent study shows that the dipole elongation in the extended dipole model plays a significant role in the phase transitions of liquid crystal phases. In this paper, molecular dynamics (MD) simulations were performed for the dipole model with different distances between the two charges keeping the total dipole moment the same. The potential energy consists of the Lennard-Jones potential and the site-site electrostatic contribution of partial charges. Detailed analyses were made with respect to the average order parameters (P(1)) and (P(2)) as functions of density along with other thermodynamic properties. When the reduced dipole elongations are 0.16 and 0.32, respectively, it is shown that the chainlike structures in the low density regime, liquid phases with columnar and smectic orders, and solid phases are formed; the phase with nematic order is not present anymore. At 0.64, the phases with antiferroelectric order were favored. The transition is found at the reduced elongation 0.55. It shows that the phase transitions are quite sensitive to the molecular charge distribution; this simple system could exhibit rather rich phase behaviors, which represents a significant advance in identifying molecular and state parameters of the future ferroelectric liquids.

Using the profile analysis, the density perturbation transport analysis, and the Doppler reflectometry measurement, for the first time a spontaneous and steady-state particle-transport barrier has been evidenced in the Ohmic plasmas in the HL-2A tokamak with no externally applied momentum or particle input except the gas puffing. A threshold in density has been found for the observation of the barrier. The particle diffusivity is well-like, and the convection is found to be inward outside the well and outward inside the well. The formation of the barrier coincides with the transition between the trapped electron mode and the ion temperature gradient driven mode.

Spherical 2-diazo-4,6-dinitrophenol (DDNP) with good flowability and controlled bulk density (0.65-0.95 g/cm(3)) has been prepared at factory scale by the modified method using 4-methylphenol as crystal control ingredient. Results showed that the yield of product was increased by 5-10%, and the waste water was significantly decreased due to circulation use of waste water compared with traditional method. Synthesized spherical DDNP was characterized by IR, laser granularity measurement, SEM, HPLC and XRD. IR spectrum confirmed the structural features of spherical DDNP. The particle analysis revealed that the modified method could offer spherical DDNP with average particle size of 350 microm and high purity (>98.52%). The XRD peaks of spherical DDNP have similar diffraction angles as those of traditional DDNP. The DSC profile of spherical DDNP showed the exothermic decomposition in the temperature range of 161.2-188.5 degrees C. The product can be pressed at over 40MPa without dead pressed phenomenon, and the minimum detonating charge of spherical DDNP was measured to be about 0.15g. A method of the particle transport study using supersonic molecular beam injection (SMBI) and microwave reflectometry is reported in this paper. Experimental results confirm that pulsed SMBI is a good perturbation source with deeper penetration and better localization than the standard gas puffing. The local density modulation is induced using the pulsed SMBI and the perturbation density is measured by the microwave reflectometry. Using Fourier transform analysis for the local density perturbation, radial profiles of the amplitude and phase of the density modulation can be obtained. The experimental results in HL-2A show that the particle injected by SMBI is located at about r/a=0.65-0.75. The position of the main particle source can be determined through three aspects: the minimum of the phase of the first harmonic of the Fourier transform of the modulated density measured by microwave reflectometry; the H(a) intensity profile and the local density increase ratio. The maximum of the amplitude of the first harmonic shifts often inward relative to the particle source location, which indicates clearly there is an inward particle pinch in this area. Good agreement has been found between the experimental results and the simulation using analytical transport model. The particle diffusivity D and the particle convection velocity V have been obtained by doing this simulation. The sensitivity in the transport coefficients of the amplitude and the phase of the density modulation has been discussed.

A low-frequency (<4 kHz), poloidally and toroidally symmetrical potential structure that peaks near zero frequency is observed in the edge plasma of the HL-2A tokamak. The axisymmetry structure exhibits a radial coherence length less than 1 cm. These characteristics are consistent with the theoretically predicted low-frequency zonal flows (LFZF). The radial wave-number frequency spectra of the LFZF show that the LFZF packets propagate both outwards and inwards. The geodesic acoustic mode (GAM) is found to coexist with the LFZF, and the LFZF is found to modulate the GAM and ambient turbulence with in-phase and antiphase relations, respectively, through an envelope analysis.
