Myocardial infarction (MI) is caused by the formation of plaques in the arterial walls, leading to a decrease of blood flow to the heart and myocardium injury as a result of hypoxia. Ferroptosis is a crucial event in myocardial injury, and icariin (ICA) exerts protective effects against myocardial injury. Here, we investigated the protective mechanism of ICA in hypoxia/reoxygenation (H/R)‐induced ferroptosis of cardiomyocytes. H9C2 cells were subjected to H/R induction. The content of lactate dehydrogenase and the levels of oxidative stress and intracellular ferrous ion Fe2+ were measured. The levels of ferroptosis markers (ACSL4 and GPX4) were detected. H/R‐induced H9C2 cells were cultured with ICA in the presence or absence of ferroptosis inducer (erastin). Znpp (an HO‐1 inhibitor) was added to ICA‐treated H/R cells to verify the role of the Nrf2/HO‐1 pathway. H/R‐induced H9C2 cells showed reduced viability, enhanced oxidative stress and lactate dehydrogenase content, increased levels of Fe2+ and ACSL4, and decreased levels of GPX4. ICA inhibited H/R‐induced ferroptosis and oxidative stress in cardiomyocytes. Erastin treatment reversed the inhibitory effect of ICA on ferroptosis in H/R cells. The expression of Nrf2 and HO‐1 in H/R‐induced H9C2 cells was reduced, whereas ICA treatment reversed this trend. Inhibition of the Nrf2/HO‐1 pathway reversed the protective effect of ICA on H/R‐induced ferroptosis. Collectively, our results suggest that ICA attenuates H/R‐induced ferroptosis of cardiomyocytes by activating the Nrf2/HO‐1 signaling pathway., Hypoxia/reoxygenation can induce ferroptosis in cardiomyocytes. Here, we report that icariin can activate the Nrf2/HO‐1 signaling pathway and inhibit hypoxia/reoxygenation‐induced ferroptosis in cardiomyocytes.

The effect of Pulsatilla saponin A (PsA) on acute myocardial infarction (AMI) was unknown. This study targeted to examine the roles of PsA on hypoxia-triggered toxicity to H9c2 cells and reveal the potential mechanism. H9c2 cells were maintained under a hypoxic environment for 12 h to construct the AMI cell model and the cells were pretreated by PsA. Hypoxia triggered toxicity to H9c2 cells and the anti-toxicity effect of PsA was evaluated by CCK8, TUNEL, and Western blot. The levels of miR-24-3p and p16 in H9c2 cells, AMI group tissues, and their respective controls were assessed using qRT-PCR. The dual-luciferase assay was applied to verify the targeting mechanism of miR-24-3p on p16. Then the effects of miR-24-3p inhibitor or/and si-p16 on H9c2 cells treated with PsA under hypoxia were detected by CCK8, TUNEL, and Western blot. Flow cytometry was executed to determine the cell cycle. Hypoxia decreased viability and proliferation and increased apoptosis of H9c2 cells, which were ameliorated by PsA pretreatment. The level of miR-24-3p was diminished, but p16 expression was elevated in hypoxia-treated cells and AMI group tissues. MiR-24-3p could sponge p16 in hypoxia-treated cells. Furthermore, the impact of applying miR-24-3p inhibitor on PsA and hypoxia-treated cells could be reversed by si-p16. PsA relieved hypoxia-triggered cell toxicity via miR-24-3p/p16 axis. These findings provided some fresh insights into the potential therapeutic effects of the application of PsA in AMI.

Background The study aims to evaluate the effectiveness and safety of bloodletting therapy for herpes zoster. Methods The following electronic databases will be searched from PubMed (1966 to March 2020), the Cochrane Central Register of Controlled Trials (update to March 2020), EMBASE (1980 to March 2020), China National Knowledge Infrastructure (1979 to March 2020), Wan Fang Data (1980 to March 2020), Chinese Scientific Journal Database (1989 to March 2020), Chinese Biomedical Database (1978 to March 2020) and traditional Chinese medicine Literature Analysis and Retrieval Database (1949 to March 2020). All randomized controlled trials without any limitation of blinding or publication language about this topic will be included, exclude cohort studies and case reports. Two independent researchers will operate article retrieval, duplication removing, screening, quality evaluation, and data analyses by Review Manager (V.5.3.5). Meta-analyses, subgroup analysis, and/or descriptive analysis will be performed based on the included data conditions. Results High-quality synthesis and/or descriptive analysis of current evidence will be provided from cure rate, converting to clinical diagnosis rate, and side effects of bloodletting. Conclusion This study will provide the evidence of whether bloodletting is an effective and safe intervention for herpes zoster. Prospero registration number CRD42020171976.

Surgical site infections (SSIs) following colorectal surgery is common, and local application of gentamicin for SSIs in the surgery remains controversial. To identify whether local application of gentamicin reduces incidence of SSIs in colorectal surgery. PubMed, Embase, the Cochrane Library, and Science Citation Index were searched for relevant randomized controlled trials (RCTs) and reference list up to November 2014. Two independent reviewers screened the records from the electronic databases, selected relevant studies, assessed the methodological quality, and extracted the data from included articles. Stata 12.0 was used to conduct a pooled analysis for main outcomes. Eight relevant randomized controlled trials with a total of 1685 patients were included in the meta-analysis. All included studies were of moderate to high quality by the Cochrane Collaboration’s tool for assessing risk of bias. There was no significant difference being found in the total pooled results for wound infection (relative risk (RR) 0.73, 95 % confidence interval (CI) 0.47 to 1.12) and organ space infection (RR 0.90, 95 % CI 0.51 to 1.59). However, subgroup analysis showed that the significant decrease of wound infection was associated with the population in the Western Europe (RR 0.60, 95 % CI 0.42 to 0.87) and follow-up periods of 30 days (RR 0.63, 95 % CI 0.42 to 0.94). Local application of gentamicin significantly reduced incidence of wound infection following colorectal surgery in Western Europe, and it was also associated with lower risk of wound infection during follow-up period of 30 days. However, its effectiveness on prophylaxis of perineal wound infection and organ space infection still lacked evidence.

In overlaid handwriting, multiple characters are written sequentially in the same area. This needs special consideration for segmenting the stroke sequence into characters. We propose a learning-based model for scoring the candidate stroke cuts and segments for online overlaid Chinese handwriting recognition. Based on stroke cut classification using support vector machine (SVM), strokes are grouped into segments, and consecutive segments are concatenated into candidate characters. The likeliness of candidate characters (unary geometry) and the compatibility between adjacent characters (binary geometry) are measured by combining the stroke cut score and the between-segment geometric score, and are integrated with the character classification score and linguistic context for character string recognition. Experiments on a large database of online Chinese handwriting demonstrate the effectiveness of the proposed method.

Event surveillance especially the composite event surveillance is a crucial task for wireless sensor networks applications. However, sensor nodes are vulnerable by harsh environmental conditions, which cause sensor nodes fallibility and affect the quality of surveillance. In order to improve the surveillance quality of composite event, one straightforward method is to increase the number of wake-up nodes in event surveillance, but this result in an immense consumption of precious energy. This paper focuses on improving surveillance quality of composite event in an energy-efficient manner. We use a three-layer statistically reliability model (StaR) to quantify the surveillance quality of composite event. Given the detection quality parameter and a k-out-of-n voting strategy, StaR-based scheduling scheme can dynamically schedule the nodes' states within a composite event detection group, i.e., inactivate low detection reliability node and activate high detection reliability node, to satisfy the user-defined composite event detection quality and realize energy conservation meanwhile.

Radio Frequency Identification (RFID) has gained a lot of attention as a promising technology that facilitates pervasive computing and the gradual proliferation of RFID-based applications will produce a large amount of urgent and complicated event streams. The RFID middleware system is expected to offer the further complex event processing service to satisfy the high-level query semantics and performance demands, and therefore, how to evaluate the service capability will be a key problem in mission-critical monitoring scenarios. In this paper, we analyze the RFID primitive event arrival and complex event processing model in a novel perspective. Furthermore, based on our proposed models, service capability evaluation method is introduced to estimate whether and how the RFID middleware system can afford the query requirements in a deterministic or statistical manner, which is believed to be very helpful in RFID-based applications. Our experiments demonstrate the utility and feasibility of our models and methods.
