1. Combined transcriptome and metabolome analysis provides insight into the ERF073 – Malic acid network in pakchoi under submergence stress

Abstract

Pakchoi is sensitive to oxygen deficiency caused by submergence; however, the molecular mechanisms underlying its response to these conditions remains unclear. To determine these mechanisms, two pakchoi cultivars, Heiyoudong (HYD) and Suzhouqing (SZQ), were subjected to submergence for 1 h and 5 h, respectively. Following this treatment, the plants were transferred back to the control conditions for recovery. HYD exhibited higher survival rate than SZQ after recovery. Leaves from 1 h stress, 5 h stress, and subsequent 5-h recovery phases were harvested for metabolome and transcriptome analyses. Data analysis revealed that 206 differentially accumulated metabolites (DAMs) were identified between SZQ and HYD after 1 h stress (S1 vs H1) and 330 DAMs were identified between SZQ and HYD after 5 h stress (S5 vs H5) at stress stage. During recovery stage, 124 and 310 DAMs were found in S1 vs H1 and S5 vs H5, respectively. Additionally, 553 commonly differentially expressed genes (DEGs) were found both in S1 vs H1 and S5 vs H5 at the stress stage, while 458 DEGs were commonly found in the two comparison groups at the recovery stage. Among these DEGs, ERF073 was down-regulated, as further confirmed by qRT-PCR. KEGG pathway analysis revealed that DAMs and DEGs were mainly enriched in metabolic pathways. Taken together, these findings indicate that ERF073 may regulate malic acid accumulation in pakchoi, increasing its resistance to submergence.

Abstract

Growing-dimensional data with likelihood unavailable are often encountered in various fields. This paper presents a penalized exponentially tilted likelihood (PETL) for variable selection and parameter estimation for growing dimensional unconditional moment models in the presence of correlation among variables and model misspecifica- tion. Under some regularity conditions, we investigate the consistent and oracle proper- ties of the PETL estimators of parameters, and show that the constrainedly PETL ratio statistic for testing contrast hypothesis asymptotically follows the central chi-squared distribution. Theoretical results reveal that the PETL approach is robust to model mis- specification. We also study high-order asymptotic properties of the proposed PETL estimators. Simulation studies are conducted to investigate the finite performance of the proposed methodologies. An example from the Boston Housing Study is illustrated.

Abstract

Neuroinflammation is associated with poor outcomes in patients with spinal cord injury (SCI). Recent studies have demonstrated that stimulator of interferon genes (Sting) plays a key role in inflammatory diseases. However, the role of Sting in SCI remains unclear. In the present study, it is found that increased Sting expression is mainly derived from activated microglia after SCI. Interestingly, knockout of Sting in microglia can improve the recovery of neurological function after SCI. Microglial Sting knockout restrains the polarization of microglia toward the M1 phenotype and alleviates neuronal death. Furthermore, it is found that the downregulation of mitofusin 2 (Mfn2) expression in microglial cells leads to an imbalance in mitochondrial fusion and division, inducing the release of mitochondrial DNA (mtDNA), which mediates the activation of the cGas‐Sting signaling pathway and aggravates inflammatory response damage after SCI. A biomimetic microglial nanoparticle strategy to deliver MASM7 (named MSNs‐MASM7@MI) is established. In vitro, MSNs‐MASM7@MI showed no biological toxicity and effectively delivered MASM7. In vivo, MSNs‐MASM7@MI improves nerve function after SCI. The study provides evidence that cGas‐Sting signaling senses Mfn2‐dependent mtDNA release and that its activation may play a key role in SCI. Abstract

This paper proposes an integral sliding mode guidance law with the constraints of the line-of-sight angle and its rate for the interception of a non-cooperative maneuvering target. The analytical guidance law is derived using an integral sliding mode control and an inertial delay control allowing for unknown uncertainties. The guidance law is further advanced to achieve exponential convergence and high terminal guidance accuracy with autopilot lag. This is accomplished by applying dynamic surface control and fractional order filters. Finally, the finite-time stability of the guidance law with and without missile autopilot lag is demonstrated. In addition to its robustness and precision, there are other benefits of this proposed guidance law. First, the proposed guidance law can effectively attenuate an abrupt change in guidance commands during the terminal homing phase in the presence of uncertainties without prior information. Second, the design parameters in the inertial delay control are decoupled with the integral sliding mode control. This decoupling supplies more degrees of freedom to regulate the performance of guidance commands. Third, the finite-time convergence of the proposed guidance law is guaranteed while accounting for the approximated second-order dynamics and uncertainties of the missile autopilot. Theoretical analysis and numerical simulations were conducted to demonstrate the efficiency of the proposed guidance law.

Abstract

Rapid prototyping aims to provide rapid design, validation and testing methods for physical prototypes. The paper designed rapid prototyping simulation system based on RTX real-time kernel and VPM (Virtual Prototyping and Modeling) development tools, and discussed the system's operation environment, system architecture, system components and major functions. Through combination of VPM modeling and simulation software and RTX real-time kernel, the system is able to implement hardware in loop simulation flexibly. Furthermore, algorithms designing, evaluating and its performance testing with physical devices in loop can be achieved.

Laser Trimming System is an equipment to adjust the resistance of chip resistor accurately by laser trimming. The measure precision of resistance measure system affects the precision of resistance directly. In order to reduce the influence of the measure error coming from measure system, a software method based on Wheatstone bridge is introduced to compensate measure error of Laser Trimming System. With its simple principle, the method is easy to realize and has good circumstance adaptability. Through measuring and compensating the measure error of high-precision standard resistors, the compensating values of measure error are gained automatically.
