6. Integrating molecular pathway with genome-wide association data for causality identification in breast cancer

Abstract

Abstract Objective The study purpose was to explore the causal association between pyruvate metabolism and breast cancer (BC), as well as the molecular role of key metabolic genes, by using bioinformatics and Mendelian randomization (MR) analysis. Methods We retrieved and examined diverse datasets from the GEO database to ascertain differentially acting genes (DAGs) in BC via differential expression analysis. Following this, we performed functional and pathway enrichment analyses to ascertain noteworthy molecular functions and metabolic pathways in BC. Employing MR analysis, we established a causal association between pyruvate metabolism and the susceptibility to BC. Additionally, utilizing the DGIdb database, we identified potential targeted medications that act on genes implicated in the pyruvate metabolic pathway and formulated a competing endogenous RNA (ceRNA) regulatory network in BC. Results We collected the datasets GSE54002, GSE70947, and GSE22820, and identified a total of 1127 DEGs between the BC and NC groups. GO and KEGG enrichment analysis showed that the molecular functions of these DEGs mainly included mitotic nuclear division, extracellular matrix, signaling receptor activator activity, etc. Metabolic pathways were mainly concentrated in PI3K−Akt signaling pathway, Cytokine−cytokine receptor binding and Pyruvate, Tyrosine, Propanoate and Phenylalanine metabolism, etc. In addition, MR analysis demonstrated a causal relationship between pyruvate metabolism and BC risk. Finally, we constructed a regulatory network between pathway genes (ADH1B, ACSS2, ACACB, ADH1A, ALDH2, and ADH1C) and targeted drugs, as well as a ceRNA (lncRNA-miRNA-mRNA) regulatory network for BC, further revealing their interactions. Conclusions Our research revealed a causal association between pyruvate metabolism and BC risk, found that ADH1B, ACSS2, ACACB, ADH1A, ALDH2, and ADH1C takes place an important part in the development of BC in the molecular mechanisms related to pyruvate metabolism, and identified some potential targeted small molecule drugs.

35. Construction and Effectiveness Evaluation of Smart Classroom for Civic and Political Education

Abstract

A smart classroom is a new type of teaching mode based on dynamic data analysis, which is a major change in the teaching mode, and the application of smart classrooms in ideological education can significantly enhance the relevance and effectiveness of ideological and political education in colleges and universities. This paper constructs a smart classroom teaching model for college ideology and politics based on the theory and technology of smart classrooms and designs a smart classroom teaching evaluation model to assess its practical application effect. The model design is combined with the expert consultation method to establish a comprehensive evaluation index system and calculate the weights of each index. Finally, the evaluation of the actual application effect of smart classroom teaching is carried out. The results show that the weights of the two first-level indexes, namely, “scientific method” and “patriotism”, are the highest, respectively 0.5 and 0.5, and the weights of the two first-level indexes are the highest. The results show that among the first-level indicators, the two first-level indicators of “scientific method” and “patriotism” have the highest weights, respectively 0.214 and 0.186. The weights of “classroom performance”, “homework,” and “practical activities” are lower, respectively 0.077, 0.073, and 0.073. Applying this paper's Civics Smart Classroom Teaching Model to actual Civics teaching, the model evaluation results show that the scores of the secondary indicators have increased, of which “asking questions” has increased from 3.54 to 3.98, an increase of 12.43%. In contrast, the indicator of “ extracurricular activities” has risen from 3.54 to 3.98, an increase of 12.43%, and the indicator of “extracurricular activities” has increased from 0.077 to 3.98. Extracurricular activities” indicator rose from 4.36 to 4.47, an increase of 2.52%. This paper designs an intelligent classroom teaching model for college civic education with good practical application effects.

Abstract

In this paper, we fill the research gap by adopting state-of-the-art computer vision techniques for the data extraction stage in a data mining system. As shown in Fig.1, this stage contains two subtasks, namely, plot element detection and data conversion. For building a robust box detector, we comprehensively compare different deep learning-based methods and find a suitable method to detect box with high precision. For building a robust point detector, a fully convolutional network with feature fusion module is adopted, which can distinguish close points compared to traditional methods. The proposed system can effectively handle various chart data without making heuristic assumptions. For data conversion, we translate the detected element into data with semantic value. A network is proposed to measure feature similarities between legends and detected elements in the legend matching phase. Furthermore, we provide a baseline on the competition of Harvesting raw tables from Infographics. Some key factors have been found to improve the performance of each stage.
