Abstract Background Blood-brain barrier (BBB) disruption after stroke is closely associated with brain tissue edema and neuronal injury, which requires accurate assessment. However, there is a lack of appropriate BBB imaging modality in vivo. As albumin in the blood could cross the damaged BBB into brain tissue after stroke, it serves as a biomarker for BBB disruption. Therefore, we aimed to develop an albumin-seeking near-infrared (NIR) probe to assess BBB disruption in stroke. Results We proposed a chemoselective strategy for seeking albumin with NIR dyes and identified an optimal probe to evaluate BBB disruption in stroke. The probe combined a NIR fluorescent dye with inherent albumin-targeting moieties and exhibited high affinity and selectivity for binding to albumin. Using a mouse stroke model, the probe displayed a high-resolution visualization of the location and extent of BBB disruption in vivo and correlated well with BBB leakage measured by Evans blue ex vivo. A dual-channel NIR-II imaging was successfully used to simultaneously assess BBB disruption and cerebral perfusion after stroke. Furthermore, we applied this method to dynamically evaluate the BBB disruption process and reperfusion of thrombolytic therapy in a stroke model in real time, which showed excellent application value. Conclusions We developed an albumin-seeking NIR probe that accurately evaluated BBB disruption in a safe, non-invasive and real-time manner in various stroke models, and has a great potential guiding stroke treatment in a real-time manner. Abstract Aiming at problems such as slow cutting speed, poor cutting quality and inability to cut thick plates effectively with traditional single-layer nozzle structure, it is proposed to add an outer ring core to form a double-layer nozzle structure on the basis of the single-layer nozzle structure to improve its cutting performance. The three-dimensional entity of single- and double-layer nozzle is established, and the simulation model of auxiliary gas flow field is established by using N-S equation and SST k-ω turbulence model assisted by laser cutting in this paper. The simulation results show that under the same cutting parameters, the auxiliary gas flow rate, oxygen concentration, mass flow rate, shear force and dynamic pressure of the double-layer nozzle structure are higher than those of the single-layer nozzle structure. In addition, compared with the single-layer nozzle structure, the double-layer nozzle structure can effectively improve the attenuation of oxygen concentration in the direction of sheet thickness. Two nozzle structures were used to laser cutting carbon steel plates with a thickness of 20 mm in the experiment, and the experimental parameters of cutting speed were 1.2–1.6 m/min. The comparison results show that the cutting speed and cutting section quality of the double-layer nozzle are better than those of the single-layer nozzle, and the results of simulation analysis are consistent with the cutting test results.

Abstract Synaptojanin 2 (SYNJ2) has crucial role in various tumors, but its role in papillary thyroid carcinoma (PTC) remains unexplored. This study first detected SYNJ2 protein expression in PTC using immunohistochemistry method and further assessed SYNJ2 mRNA expression through mRNA chip and RNA sequencing data and its association with clinical characteristics. Additionally, KEGG, GSVA, and GSEA analyses were conducted to investigate potential biological functions, while single‐cell RNA sequencing data were used to explore SYNJ2's underlying mechanisms in PTC. Meanwhile, immune infiltration status in different SYNJ2 expression groups were analyzed. Besides, we investigated the immune checkpoint gene expression and implemented drug sensitivity analysis. Results indicated that SYNJ2 is highly expressed in PTC (SMD = 0.66 [95% CI: 0.17–1.15]) and could distinguish between PTC and non‐PTC tissues (AUC = 0.74 [0.70–0.78]). Furthermore, the study identified 134 intersecting genes of DEGs and CEGs, mainly enriched in the angiogenesis and epithelial‐mesenchymal transition (EMT) pathways. Subsequent analysis showed the above pathways were activated in PTC epithelial cells. PTC patients with high SYNJ2 expression showed higher sensitivity to the six common drugs. Summarily, SYNJ2 may promote PTC progression through angiogenesis and EMT pathways. High SYNJ2 expression is associated with better response to immunotherapy and chemotherapy.

Asian seabass (Lates calcarifer) is an important foodfish species. It has been cultured in Singapore since the 1980s. A selective breeding program to improve growth, meat quality and disease resistance has been ongoing in Singapore since 2004. In 2012, outbreaks of the big belly disease (BBD) occurred in two batches of offspring generated by a mass-cross among F1 brooders in a hatchery. After the outbreaks, only 437 survived in 6000 F2 juveniles. Molecular parentage analysis with 10 microsatellites revealed that four parents (F1) produced high percentage of offspring that survived the BBD outbreaks. Eighty-two BBD survivor offspring (F2) were selected based on their growth performance at 90 days post hatch (dph), family origin and allelic diversity. They were used as brooders to generate F3. In a period of five years, these F2 brooders produced six batches of offspring, where no outbreak of BBD occurred. In the same period, outbreaks of BBD occurred in 4 out of 18 batches produced by F2 brooders selected for growth, with mortality rates ranging from 85.1% to 95.4%. In F3, the average body weight (57.1 ± 1.09 g, n = 3068) of the line for BBD survival was slightly lower than that (60.6 ± 2.29 g, n = 4280) of the line for growth at 90 dph. In both lines, the body weight of F3 was much higher than that (46.90 ± 0.19 g, n = 12117) of F2. The genetic diversity in the line for BBD survival (F3) was much lower than that in F3 line for growth due to small founder population. These data indicate that the selection of BBD survival during BBD outbreaks using molecular parentage analysis is effective to reduce BBD outbreaks. Avoiding further reduction of genetic diversity is essential in the line selected for increased survival against BBD.

Abstract Background The latent tuberculosis infection (LTBI) burden is still unclear in schoolchildren and adolescents in China. Previous study and daily surveillance data indicate a LTBI detection gap. The research objective was to evaluate the LTBI burden and detection gap among schoolchildren and adolescents in China. Methods A cross-sectional study was conducted among 69,667 schoolchildren and adolescents in Chongqing, China between September 2022 and December 2023 implemented by Chongqing Municipal Institute of Tuberculosis using tuberculin skin test (TST) and creation tuberculin skin test (C-TST). To evaluate the LTBI detection gap, the pulmonary tuberculosis (PTB) screening data implemented by Chongqing Municipal Institute of Tuberculosis have been compared with the data in 2021 implemented by community-level medical and health care institutions. Results The LTBI prevalence rate using TST and C-TST implemented by Chongqing Municipal Institute of Tuberculosis was 12.8% (95%CI, 12.5-13%) and 6.4% (95%CI, 6-6.8%) respectively. In Hong Kong, studies on trading and human exploitation of adult Asian horseshoe crabs (Carcinoscorpius rotundicauda and Tachypleus tridentatus) are limited, with the last study dating back to 2005–2006. To identify how trading activities (e.g., for consumption as food, bycatch, and mercy release activities) may threaten horseshoe crab populations in recent times, market surveys were conducted at five potential horseshoe crab trade outlets in Hong Kong in 2020 – 2021 in tandem with phone interviews with Hong Kong citizens to measure their knowledge/awareness on horseshoe crabs and relevant conservation measures. We also conducted in-depth interviews with 15 representatives from fishers’ associations and 20 from religious organisations on their views on horseshoe crab status in fisheries and mercy release. The market survey recorded a total of 80 adult horseshoe crabs for sale in seafood restaurants or fish stalls out of the 98 shops surveyed. In addition, phone interviews revealed a low level of awareness among Hong Kong citizens (N = 1005 respondents), with 53 % having no prior knowledge and never heard of “horseshoe crab”. However, such knowledge improved with an increased education level, particularly among those in the 50–59 age group and those who visited horseshoe crab habitats. Nearly all respondents (975) pledged not to consume horseshoe crabs upon being informed of their conservation status. Representatives from the fishers’ association reported that while there were no targeted fisheries for horseshoe crabs in Hong Kong, a small quantity of horseshoe crabs were captured as bycatch. Among the 12 religious organisations surveyed, 30 % of the respondents still conducted mercy release activities in Hong Kong. Some strongly disagreed with the need for government interventions on mercy release and were not willing to accept alternative official approaches. The above results revealed a considerable knowledge and awareness gap among the public and identified anthropogenic threats to Asian horseshoe crabs. To alleviate the situation, a pilot public awareness campaign was launched to increase public awareness and promote the conservation of horseshoe crabs in Hong Kong. We recommend strengthening the protection of local Asian horseshoe crabs through a combination of conservation management and public education.

Background and objective Hepatic portal venous gas(HPVG) represents a rare radiographic phenomenon frequently linked to intestinal necrosis, historically deemed to need immediate surgical intervention. The pivotal query arises about the imperative of urgent surgery when a patient manifests HPVG after gastrointestinal surgery. This inquiry seeks to elucidate whether emergent surgical measures remain a requisite in such cases.Methods The investigation into 14 cases of HPVG after gastrointestinal procedures was conducted through a comprehensive review of relevant literature. This methodological approach contributes to a nuanced understanding of HPVG occurrences following gastrointestinal surgery, informing clinical considerations and potential therapeutic strategies.Results Among the 14 patients, 12 recovered and 2 died. 6 patients underwent surgical exploration, 4 with negative findings and recovered. 8 cases received conservative treatment, resulting in improvement for 5, and 1 initially treated conservatively, revealed perforation during later surgical exploration, leading to improvement, 1 case ended in mortality.Conclusion After gastrointestinal surgery, in Computed Tomography (CT) imaging, the coexistence of HPVG and gastrointestinal dilatation, without signs of peritoneal irritation on abdominal examination, may suggest HPVG due to acute gastrointestinal injury, intestinal gas, and displacement of gas-producing bacteria. These patients can be managed conservatively under close supervision. In cases where HPVG coexists with gastrointestinal dilatation and Pneumatosis intestinalis (PI) without signs of peritoneal irritation, conservative treatment may be continued under close supervision. However, if progressive exacerbation occurs despite close monitoring and the aforementioned treatments, timely surgical exploration is deemed necessary. When HPVG is combined with signs of peritoneal irritation, prompt laparotomy and exploration are preferred.

Background: Disruption of the blood-brain barrier (BBB) is a major contributor to hemorrhagic transformation (HT) in patients with acute ischemic stroke (AIS) following intravenous thrombolysis (IVT). However, the clinical therapies aimed at BBB protection after IVT remain limited. Methods: One hundred patients with AIS who underwent IVT were enrolled (42 with HT and 58 without HT 24 h after IVT). Based on the cytokine chip, the serum levels of several AIS-related proteins, including LCN2, ferritin, matrix metalloproteinase-3, vascular endothelial-derived growth factor, and X-linked inhibitor of apoptosis, were detected upon admission, and their associations with HT were analyzed. After finding that LCN2 was related to HT in patients with IVT, we clarified whether the modulation of LCN2 influenced BBB dysfunction and HT after thrombolysis and investigated the potential mechanism. Results: In patients with AIS following IVT, logistic regression analysis showed that baseline serum LCN2 (p = 0.023) and ferritin (p = 0.046) levels were independently associated with HT. A positive correlation between serum LCN2 and ferritin levels was identified in patients with HT. In experimental studies, recombinant LCN2 (rLCN2) significantly aggravated BBB dysfunction and HT in the thromboembolic stroke rats after thrombolysis, whereas LCN2 inhibition by ZINC006440089 exerted opposite effects. Further mechanistic studies showed that, LCN2 promoted endothelial cell ferroptosis, accompanied by the induction of high mobility group box 1 (HMGB1) and the inhibition of nuclear translocation of nuclear factor E2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) proteins. Ferroptosis inhibitor ferrostatin-1 (fer-1) significantly restricted the LCN2-mediated BBB disruption. Transfection of LCN2 and HMGB1 siRNA inhibited the endothelial cell ferroptosis, and this effects was reversed by Nrf2 siRNA. Conclusion: LCN2 aggravated BBB disruption after thrombolysis by promoting endothelial cell ferroptosis via regulating the HMGB1/Nrf2/HO-1 pathway, this may provide a promising therapeutic target for the prevention of HT after IVT.

Abstract We investigate a generalized poly-Laplacian system with a parameter on weighted finite graphs, a generalized poly-Laplacian system with a parameter and Dirichlet boundary value on weighted locally finite graphs, and a ( p , q ) $(p,q)$ -Laplacian system with a parameter on weighted locally finite graphs. We utilize a critical points theorem built by Bonanno and Bisci [Bonanno, Bisci, and Regan, Math. Comput. Model. 52(1-2):152–160, 2010], which is an abstract critical points theorem without compactness condition, to obtain that these systems have infinitely many nontrivial solutions with unbounded norm when the parameters locate some well-determined range.

Abstract The skin is intrinsically a cell-membrane-compartmentalized hydrogel with high mechanical strength, potent antimicrobial ability, and robust immunological competence, which provide multiple protective effects to the body. Methods capable of preparing hydrogels that can simultaneously mimic the structure and function of the skin are highly desirable but have been proven to be a challenge. Here, dual structurally and functionally skin-mimicking hydrogels are generated by crosslinking cell-membrane compartments. The crosslinked network is formed via free radical polymerization using olefinic double bond-functionalized extracellular vesicles as a crosslinker. Due to the dissipation of stretching energy mediated by vesicular deformation, the obtained compartment-crosslinked network shows enhanced mechanical strength compared to hydrogels crosslinked by regular divinyl monomers. Biomimetic hydrogels also exhibit specific antibacterial activity and adequate ability to promote the maturation and activation of dendritic cells given the existence of numerous extracellular vesicle-associated bioactive substances. In addition, the versatility of this approach to tune both the structure and function of the resulting hydrogels is demonstrated through introducing a second network by catalyst-free click reaction-mediated crosslinking between alkyne-double-ended polymers and azido-decorated extracellular vesicles. This study provides a platform to develop dual structure- and function-controllable skin-inspired biomaterials.

Abstract Follicle-stimulating hormone (FSH) is involved in mammalian reproduction via binding to FSH receptor (FSHR). However, several studies have found that FSH and FSHR play important roles in extragonadal tissue. Here, we identified the expression of FSHR in human and mouse pancreatic islet β-cells. Blocking FSH signaling by Fshr knock-out led to impaired glucose tolerance owing to decreased insulin secretion, while high FSH levels caused insufficient insulin secretion as well. In vitro, we found that FSH orchestrated glucose-stimulated insulin secretion (GSIS) in a bell curve manner. Mechanistically, FSH primarily activates Gαs via FSHR, promoting the cAMP/protein kinase A (PKA) and calcium pathways to stimulate GSIS, whereas high FSH levels could activate Gαi to inhibit the cAMP/PKA pathway and the amplified effect on GSIS. Our results reveal the role of FSH in regulating pancreatic islet insulin secretion and provide avenues for future clinical investigation and therapeutic strategies for postmenopausal diabetes.

Abstract Microbes well-adapted to the Arctic Ocean are promising for producing novel compounds, due to their fancy strategies for adaptation and being under-investigated. Two new phenazine alkaloids (1 and 2) and one new phenoxazine (3) were isolated from Nocardiopsis dassonvillei 502F, a strain originally isolated from Arctic deep-sea sediments. AntiSMASH analysis of the genome of Nocardiopsis dassonvillei 502F revealed the presence of 16 putative biosynthetic gene clusters (BGCs), including a phenazine BGC. Most of the isolated compounds were evaluated for their antibacterial, antiallergic, and cytotoxic activities. Among them, compounds 4 and 5 exhibited potent in vitro cytotoxic activities against osteosarcoma cell line 143B with IC50 values 0.16 and 20.0 μM, respectively. Besides, the results of antiallergic activities of compounds 6–8 exhibited inhibitory activities with IC50 values of 10.88 ± 3.05, 38.88 ± 3.29, and 2.44 ± 0.17 μg/mL, respectively (IC50 91.6 μM for the positive control loratadine). AIM: To explore the correlation between the expression levels of microRNA-377-3p(miR-377-3p)and microRNA-365-3p(miR-365-3p)in serum and aqueous humor and the degree of diabetes macular edema(DME).METHODS: A total of 60 DME patients(60 eyes)admitted to 363 Hospital from February 2021 to February 2022 were selected in this prospective study(the severe eye was selected if both eyes had DME, while the right eye was selected if the same degree of DME), including 24 mild eyes, 21 moderate eyes and 15 severe eyes. In addition, another 60 patients(60 eyes)with type 2 diabetes(without fundus disease)admitted to our hospital during the same period were selected as the control group. ObjectiveAt present, the structure of knowledge in the field of childhood thyroid cancer is not clear enough, and scholars lack a sufficient understanding of the developing trends in this field, which has led to a shortage of forward-looking outputs. The purpose of this research is to help scholars construct a complete knowledge framework and identify current challenges, opportunities, and development trends.MethodsWe searched the literature in the Web of Science Core Collection database on August 7, 2023 and extracted key information from the top 100 most cited articles, such as the countries, institutions, authors, themes, and keywords. We used bibliometric tools such as bibliometrix, VOSviewer, and CiteSpace for a visualization analysis and Excel for statistical descriptions.ResultsThe top 100 most cited articles fluctuated over time, and the research was concentrated in European countries, the United States, and Japan, among which scientific research institutions and scholars from the United States made outstanding contributions. Keyword analysis revealed that research has shifted from simple treatment methods for pediatric thyroid cancer (total thyroidectomy) and inducing factors (the Chernobyl power station accident) to the clinical applications of genetic mutations (such as the BRAF and RET genes) and larger-scale genetic changes (mutation studies of the DICER1 gene). The thematic strategy analysis showed an increasing trend towards the popularity of fusion oncogenes, while the popularity of research on traditional treatments and diagnostics has gradually declined.ConclusionExtensive research has been conducted on the basic problems of pediatric thyroid cancer, and there has been significant outputs in the follow-up and cohort analysis of conventional diagnostic and treatment methods. However, these methods still have certain limitations. Therefore, scholars should focus on exploring fusion genes, the clinical applications of molecular targets, and novel treatment methods. This study provides a strong reference for scholars in this field.

ObjectiveTo establish a reverse osmosis technology-based method for enrichment of virus in water samples. MethodsTwo inactivated virus vaccines and two viral nucleic acid plasmids were added into simulated water samples and detected with real-time quantitative reverse transcriptase PCR (qRT-PCR) after reverse osmosis-based enrichment. Reverse osmosis membrane material, membrane pore size, and filtration time were optimized in the enrichments. ResultsThe enrichment efficiency increased with the decrease of pore size of the reverse osmosis membrane. The average virus enrichment efficiency of reverse osmosis membrane with the pore size of 0.10 μm was 22.86 ± 0.84% for 2 simulated water samples. The virus enrichment efficiency of nylon, cellulose acetate and polyethersulfone membranes were similar between each other. The enrichment efficiency goes up with the increase of filtration times, but the difference in the efficiency was not significant between two and three times of filtration. ConclusionThe efficiency can be up to 32.64% for the enrichment of polio virus in water samples using cellulose acetate filter membrane with 0.10 μm aperture and two times of filtration.

Aim: To explore the potential categories and characteristic differences of maternal postpartum depression literacy. Design: Cross-sectional survey. Methods: From February 2023 to April 2023, convenience sampling was used to survey 278 women attending postnatal visits to three tertiary level A hospitals. The study included general demographic characteristics, postpartum depression literacy scale, and family caring index scale. Latent profile analysis was performed to identify the categories of maternal postpartum depression literacy, and multiple disordered logistic regression was used to analyze the influencing factors of different categories. Results: Maternal postpartum depression literacy was divided into three categories: low literacy (41.0 %), moderate literacy (32.4 %), and high literacy (26.6 %). The results showed that work status, education level, whether the pregnancy was planned, whether or not they had participated in mental health-related courses, and family functioning status were factors influencing the category of maternal postpartum depression literacy (P

The development of urbanization and the transformation of green lands into impermeable land increase temperature and create urban heat islands (UHIs). Our observations with remote sensing instruments of Landsat platforms show considerable changes in land use types in Beijing city with the shrinking of green lands, expansion of built environments, and a slight increase in the temperature during the recent four decades. Using remote sensing instruments of Landsat platforms and registered data from two meteorological stations in Beijing, this study finds the relationship between land surface temperature (LST) and the increasing conversion of cultivated lands into built-up areas. This article presents innovative research that shows the mutual correlation well and recommends revisions in the land use policies for better weather. The geographically weighted regression model (GWR) with a Gaussian weighting kernel function analyzes the impact of various urban land use types on the LST and the increase UHIs. In Beijing city, green lands show fewer standard deviations (SD) in the average temperatures equal to 0.109, while the industrial spaces exhibit a high SD equal to 0.212. The outcomes of this paper contribute to finding optimal land use policies everywhere in the world with the increasing urbanization through simulating its model for a more comfortable life.

AimTo analyse the effect of breast crawl on neonatal breastfeeding within 5 months of delivery.DesignProspective Cohort Study.MethodsNeonates were divided into successful and failed groups, according to whether the newborn crawled to the breast and began sucking for the first time within 1 h after delivery. The initiation of lactation and breastfeeding duration of the two groups were analysed at 24 h, 48 h, 72 h, the feeding practices were followed-up on the 7th day, 42nd day, and 5th month in order to explore the long-term benefits of breast crawl on breastfeeding.ResultsA total of 163 neonates were included. The initiation time and the duration of first feeding, the lactation initiation in the successful group was earlier, the scores of first and in-hospital breastfeeding scales were higher.Public ContributionBreast crawl is the preferred method for mothers to begin breastfeeding. The delivery room is the place where the first breast crawl occurs immediately after delivery. The midwife is the key person to protect this valuable behavior. Therefore, the midwife needs provide valuable opportunities for the breast crawl of the newborn and promote this behavior.

Choroidal vascular diseases, such as age-related macular degeneration, are the leading cause of vision impairment and are characterized by pathological angiogenesis. Verteporfin-mediated photodynamic therapy is a current strategy that selectively occludes choroidal neovasculature. However, the clinically used large-dose systemic administration increases the risk of systemic adverse events, such as phototoxicity to superficial tissues. In this study, we developed an in situ verteporfin delivery system with a photoswitching synergistic function that disassembles in response to intraocular inflammatory enzymes. Under light-on conditions, verteporfin-mediated photodynamic therapy effectively occurs and this leads to vascular occlusion. Under light-off conditions, non-photoactive verteporfin negatively regulates vascular endothelial growth factor-induced angiogenesis as a yes-associated protein inhibitor. Taken together, our system serves as an intraocular verteporfin reservoir to improve the bioavailability of verteporfin by innovatively exploiting its photochemical and biological functions. This work provides a promising strategy with synergistic antiangiogenic effects for the treatment of choroidal vascular diseases.

Neutron induced nuclear reaction on light nuclei is an important subject in nuclear data research. The reaction of the n+d three nucleons system is also an important platform for examining the nucleonnucleon interaction theories. The progress on calculations of neutron induced nuclear reactions on deuterons with the FaddeevAGS equations was introduced in this paper. The equations were solved with the wave packet continuum discretization method in momentum space. The calculated results include the angular distributions of elastic scattering, doubledifferential cross sections of the emitting neutrons and protons from breakup reactions, excitation functions of the total elastic scattering cross sections, and the excitation function of the total breakup reaction cross sections. The elastic scattering cross sections were calculated with the separable Paris nucleonnucleon interaction for incident energy below 20 MeV and with the Nijmegen potential for incident energy above 20 MeV. A comparison between results with the Paris potential, which consists of both s and pwave nucleonnucleon (NN) interactions, and those with the swave Yamaguchi potential for the n+d elastic scattering suggest that NN interactions that are beyond the swave are necessary to describe the experimental data, especially at higher incident energy. Due to practical reasons, calculations of the breakup cross sections, namely, the double differential cross sections of the emitting neutrons and protons are limited with a separable swave Yamaguchitype nucleonnucleon interaction, which limited the calculation of the double differential cross sections to below 20 MeV. Fair agreements are found between the results in this paper and the experimental data and with the evaluated data in CENDL32, ENDF/BⅧ.0, JENDL5, and JEFF33.

Abstract Nanoparticle (NP) is an emerging tool applied in the biomedical field. With combination of different materials and adjustment of their physical and chemical properties, nanoparticles can have diverse effects on the organism and may change the treating paradigm of multiple diseases in the future. More and more results show that nanoparticles can function as immunomodulators and some formulas have been approved for the treatment of inflammation-related diseases. However, our current understanding of the mechanisms that nanoparticles can influence immune responses is still limited, and systemic clinical trials are necessary for the evaluation of their security and long-term effects. This review provides an overview of the recent advances in nanoparticles that can interact with different cellular and molecular components of the immune system and their application in the management of inflammatory diseases, which are caused by abnormal immune reactions. This article focuses on the mechanisms of interaction between nanoparticles and the immune system and tries to provide a reference for the future design of nanotechnology for the treatment of inflammatory diseases.
