The white-lipped peccary (WLP; Tayassu pecari) and the collared peccary (CP; Pecari tajacu) are social ungulates of huge ecological and economic importance. We determined the habitat occupancy for peccaries in a rural community bordering the Calakmul Biosphere Reserve in Campeche, Mexico. Twenty-eight stratified sampling points were randomly placed in the study area during a rainy and a dry season, where the presence of both peccary species was measured in relation to three ecological variables (habitat type, presence of natural predators, and water availability) and one anthropogenic variable (hunting). Ten sampling points were established with camera traps (total days/camera traps = 2268), and 18 sampling points were plots of 20 m2 to detect feces and/or footprints. To assess the relationship between both species’ presence and the selected variables, models of habitat occupancy were constructed using a binary matrix of detection/non-detection using the PRESENCE program. For WLP and CP, the respective estimated occupancy probabilities were 30 and 40% in the rainy season and 88 and 44% in the dry season. Our results suggest that occupancy by WLP is affected by habitat type and predators in the rainy season and by water availability and hunting in the dry season. Conversely, for CP, occupancy is affected by water availability and hunting in the rainy season, and by habitat type and predators in the dry season. Thus, hunting negatively affects the way that WLP occupy specific habitats, such as floodplain forest and medium sub-deciduous forest, particularly during periods of water scarcity when the species is more vulnerable to hunting.

The Wolbachia-based approach is under evaluation as a control strategy against Aedes aegypti in Mexico. From 2017 to 2019, we performed a pilot study to evaluate an open-field mass-release of wAlbB-infected Ae. aegypti male mosquitoes, as part of an integrated vector management (IVM) plan led by the Ministry of Health in Mexico to suppress natural populations of Ae. aegypti in southern Mexico. Community engagement and social evaluation were part of the key activities conducted. Overall, results showed the positive benefits of this Wolbachia-based method in the reduction of Aedes mosquitoes (90%). Mosquito’s nuisance at bedtime and the increasing circulation of mosquitoes during the releasing days were reported as the negative perceptions of this method. Importantly, participants understood the difference between wild mosquitoes and those released as part of the project, as well as the importance of the IVM. A significant number of the population accepted and supported the project, and feedback was given to improve future mosquito-releasing activities. The social license was a key factor in the success of the intervention and should be part of innovative paradigms for mosquito-vector control strategies involving community engagement. We outline the Mexican experience of community engagement and social assessment in implementing a Wolbachia-based strategy.

The ability of the maternally transmitted endosymbiotic bacterium Wolbachia to induce cytoplasmic incompatibility (CI) and virus blocking makes it a promising weapon for combatting mosquito-borne diseases through either suppression or replacement of wild-type populations. Recent field trials show that both approaches significantly reduce the incidence of dengue fever in humans. However, new questions emerge about how Wolbachia-mosquito associations will co-evolve over time and whether Wolbachia-mediated virus blocking will be affected by the genetic diversity of mosquitoes and arboviruses in the real world. Here, we have compared the Wolbachia density and CI expression of two wAlbB-infected Aedes aegypti lines transinfected 15 years apart. We have also assessed wAlbB-mediated virus blocking against dengue (DENV), Zika (ZIKV), and Chikungunya (CHIKV) viruses and examined whether host genetic backgrounds modulate viral blocking effects by comparing ZIKV infection in mosquitoes with a Mexican genetic background to those with a Singaporean background. Our results show that over 15 years, wAlbB maintained the capacity to form a stable association with Ae. aegypti in terms of both density and CI expression. There were variations in wAlbB-induced virus blocking against CHIKV, DENV, and ZIKV, and higher inhibitory effects on ZIKV in mosquitoes on the Singaporean genetic background than on the Mexican background. These results provide important information concerning the robustness and long-term stability of Wolbachia as a biocontrol agent for arbovirus disease control.

We conducted a baseline characterization of the abundance and seasonality of Aedes aegypti (Linnaeus, 1762)—a vector of dengue, chikungunya, and Zika—in two suburban localities of Yucatan, Mexico, as the first step in the implementation of an integrated vector management (IVM) plan combining ‘traditional Aedes control’ (source reduction/truck-mounted ultra-low volume [ULV] spraying) and incompatible insect technique/sterile insect technique for population suppression in Yucatan, Mexico. Weekly entomological collections with ovitraps and BG-sentinel traps were performed in 1-ha quadrants of both localities for 1 yr. Three distinct periods/phases were identified, closely associated with precipitation: 1) a phase of low population abundance during the dry season (weekly average of Aedes eggs per ovitrap and adults per BG trap = 15.51 ± 0.71 and 10.07 ± 0.88, respectively); 2) a phase of population growth and greatest abundance of Aedes (49.03 ± 1.48 eggs and 25.69 ± 1.31 adults) during the rainy season; and finally 3) a phase of decline among populations (20.91 ± 0.97 eggs and 3.24 ± 0.21 adults) after the peak of the rainy season. Seasonal abundance and dynamics of Ae. aegypti populations suggest that it is feasible to develop and implement time-specific actions as part of an IVM approach incorporating integrating novel technologies (such as rear-and-release of Wolbachia-infected males) with classic (insecticide-based) approaches implemented routinely for vector control. In agreement with the local vector control program, we propose a pilot IVM strategy structured in a preparation phase, an attack phase with traditional vector control, and a suppression phase with inundative releases, which are described in this paper.

In Mexico, Aedes aegypti (L.) is the primary dengue vector, chikungunya, and Zika viruses. The continued use of synthetic pyrethroids has led to the development of resistance in target populations, which has diminished the effectiveness of vector control programs. Resistance has been associated with disadvantages that affect the biological parameters of resistant mosquitoes compared to susceptible ones. In the present study, the disadvantages were evaluated by parameters related to survival and reproduction (‘fitness cost’) after selection with deltamethrin for five generations. The parameters analyzed were the length of the development cycle, sex ratio, survival, longevity, fecundity, egg viability, preoviposition, oviposition and postoviposition periods, and growth parameters. In the deltamethrin-selected strain, there was a decrease in the development cycle duration, the percentage of pupae, the oviposition period, and eggs viability. Although mean daily fecundity was not affected after the selection process, this, together with the decrease in the survival and fecundity levels by specific age, significantly affected the gross reproductive rate (GRR), net reproductive rate (Ro), and intrinsic growth rate (rm) of the group selected for five generations with deltamethrin compared to the group without selection. Identifying the ‘cost’ of resistance in biological fitness represents an advantage if it is desired to limit the spread of resistant populations since the fitness cost is the less likely that resistant individuals will spread in the population. This represents an important factor to consider in designing integrated vector management programs.

Aedes

In recent years, Aedes albopictus has become the most important invasive mosquito species worldwide. In 2018, Ae. albopictus was found in a suburban area of Merida, one of the cities with the highest number of arbovirus cases in Mexico in the last 10 years. As Ae. albopictus continues its range expansion, there is a need to monitor its susceptibility to existing insecticide classes, since countries like Mexico currently do not consider Ae. albopictus in its insecticide management programs. In order to determine its susceptibility to the insecticides usually applied by the vector control program in Mexico, the Centers for Disease Control and Prevention bottle bioassays were performed on individuals from established population of Ae. albopictus from Merida, Yucatan, Mexico. Results suggested that the population recently found in the suburban area of Merida is susceptible to permethrin, deltamethrin, chlorpyrifos, malathion, bendiocarb, and propoxur. Further studies of insecticide resistance using biochemical and molecular tools together with more knowledge of the biology and ecology of this species are necessary to generate specific and efficient control strategies in Mexico.

A survey was carried out in 51 households within a suburban area of Merida, Yucatan, Mexico, for 5 consecutive days. Adult collections were performed using Prokopack aspirators (indoors) and human-landing mosquito catches (HLC) outdoors, and larval sites (artificial containers) were revised for larvae collection. A total of 259 Aedes albopictus were collected, 246 from artificial larval sites, 8 by indoor aspiration, and 5 by HLC. This is the first record of Ae. albopictus in Merida, Yucatan, Mexico.

This study reports the results of a molecular screening for Wolbachia (Wb) infection in Aedes albopictus (Skuse) populations recently established in the Yucatan Peninsula, Mexico. To do so, collections of free-flying adults with BG traps and emerged adults from eggs after ovitrap field collections were performed in three suburban localities of the city of Merida, Yucatan. Overall, local populations of Ae. albopictus present a natural Wb infection rate of ~40% (18 of 45). Wb infection was detected in both field-collected adults (76.5%, 13 of 17) and eggs reared (17.8%, 5 of 28) and in 37.9% (11/29) of females and 43.7% (7/16) of male Ae. albopictus mosquitoes. An initial screening for Wolbachia strain typing showed that native Ae. albopictus were naturally coinfected with both wAlbA and wAlbB strains. The knowledge of the prevalence and diversity of Wolbachia strains in local populations of Aedes mosquitoes is part of the baseline information required for current and future Wolbachia-based vector control approaches to be conducted in Mexico.

Vector control methods that mobilize and impact rapidly during dengue, Zika, and chikungunya outbreaks are urgently needed in urban contexts. We investigated whether one person using a handheld aerosolized insecticide could achieve efficacy levels comparable to targeted indoor residual spraying (TIRS), using pyrethroid-resistant Aedes aegypti in a semi-field setting with experimental houses in Mexico. The insecticide product (H24, a carbamate and pyrethroid mixture), available over-the-counter locally, was sprayed only on known Ae. aegypti–resting surfaces, for example, walls less than 1.5 m and dark hidden areas. In six identical houses with paired bedrooms, one bedroom was treated, and the other remained an untreated control. Each week for 8 weeks, 100 female pyrethroid-resistant Ae. aegypti were released in each bedroom and followed up daily. Mortality rates in treated bedrooms exceeded 90% for at least 2 weeks, and more than 80% (89.2; 95% CI: 79.98–98.35) for 3 weeks or more. Mortality rates in control houses were zero. Results demonstrate that the immediate impact of TIRS can be delivered by one person using existing products, at an estimated cost for the average household in Mexico of under US$3 per month. Triggered by early outbreak signs, dissemination via community hubs and mass/social media of instructions to treat the home immediately, with monthly re-treatment thereafter, provides a simple means to engage and empower householders. Compatible with integrated vector management strategies, it enables self-protection even if existing agencies falter, a situation exemplified by the potential impact on vector control of the restrictions imposed during the 2020 COVID-19 pandemic.

Introduction: The vector-borne diseases program in México has an established network of ovitraps for entomological surveillance of Aedes spp. In response to reports of Aedes albopictus in the periphery of Mérida, the state capital of Yucatán, the Ministry of Health increased the specificity of this surveillance. Objective: To describe the presence and distribution of Ae. albopictus in Mérida and its relative abundance compared to Aedes aegypti in ovitraps of the vector control program. Materials and methods: During October, 2019, 91 ovitraps were randomly selected from 31 neighborhoods of Mérida. Mosquitoes were reared at the insectary of the Collaborative Unit for Entomological Bioassays of the Autonomous University of Yucatán from eggs collected in the field. Relative abundance was determined for adult individuals of each identified species and neighborhood. Results: 32 % of the neighborhoods were positive for Ae. albopictus and 100 % for Ae. aegypti. A total of 28 adults of Ae. albopictus (10 females and 18 males) were obtained from ovitraps. No correlation was observed between the abundance of Ae. aegypti and Ae. albopictus for both adults and females (p0.05) at the neighborhood level. Conclusions: The results confirm that Ae. albopictus coexisted with Ae. aegypti in Mérida at the time of the study. The low relative abundance suggests that Ae. albopictus was in the initial phase of invasion.Introducción. El programa de enfermedades transmitidas por vectores en México tiene una red establecida de ovitrampas para la vigilancia entomológica de Aedes spp. Los servicios de salud del estado de Yucatán, en respuesta a reportes de Aedes albopictus en la periferia de Mérida, capital del estado, incrementaron la especificidad de dicha vigilancia. Objetivo. Describir la presencia y distribución de Ae. albopictus en Mérida y su abundancia relativa comparada con Aedes aegypti, en ovitrampas del programa de control de vectores. Materiales y métodos. Durante octubre de 2019, se seleccionaron al azar 91 ovitrampas en 31 barrios de Mérida. Los mosquitos adultos se obtuvieron del insectario de la Unidad Colaborativa para Bioensayos Entomológicos de la Universidad Autónoma de Yucatán a partir de huevos recolectados en campo. Se determinó la abundancia relativa de individuos adultos de cada especie identificada y por barrios evaluados. Resultados. En el 32 % de los barrios muestreados, se detectó Ae. albopictus y, en todos ellos, Ae. aegypti. Se recolectaron 28 adultos de Ae. albopictus (10 hembras y 18 machos) en las ovitrampas. No se observó correlación entre la abundancia de adultos ni de hembras Ae. aegypti y Ae. albopictus por barrio (p0,05). Conclusiones. Los resultados confirmaron que Ae. albopictus estaba coexistiendo con Ae. aegypti en Mérida en el momento del estudio. La baja abundancia relativa sugiere que Ae. albopictus se encontraba en la fase inicial de invasión.

The southern house mosquito, Culex quinquefasciatus Say (Diptera: Culicidae), transmits pathogens and is one of the most important vectors of human disease. The carboxylesterase mosquito genes encoded esterase enzyme involved in the mechanism that provides resistance to insecticides. We amplified by RTPCR four alternative transcripts from the esterase gene not reported previously (A, B, C, and D) from Cx. quinquefasciatus larvae from Monterrey, N.L., northeastern Mexico. Through cDNA sequences, we predicted amino acid sequences used to model the three-dimensional structure of protein isoforms. We determined dN and dS ratios to identify evolutionary forces which lead the family. PCR products were obtained from 5A esterases (1839, 1898, 1894, and 1953 bp). The translation sequence of the PCR products showed polypeptides of 612, 473, 282, and 282 amino acids in length for isoforms A, B, C, and D, respectively. Three exons and two introns were obtained in the analyzed sequences. The study sequences were compared with those previously published showing 98.7, 98.5, 91.5, and 91.5% amino acid similarity for isoforms A, B, C, and D, respectively. The threedimensional models showed that only ORF-D had the three amino acids corresponding to the catalytic triad, while ORF-A and ORF-C lacked one or two of the three amino acids. Esterase genes are under purifying of selection; a clue the members are functional. The processes might be useful in monitoring evolutionary dynamics of known carboxylesterase genes and identifying new carboxylesterase splicing.

BACKGROUND Resistance to insecticides through one or several mechanisms has a cost for the insect in various parameters of its biological cycle. The present study evaluated the effect of deltamethrin on detoxifying enzymes and biological parameters in a population of Aedes aegypti selected for 15 generations. The enzyme activities of alpha- and beta-esterases, mixed-function oxidases and glutathione-S-transferases were determined during selection, along with biological parameters. RESULTS Overexpression of mixed-function oxidases as a mechanism of metabolic resistance to deltamethrin was found. There were decreases in percentages of eggs hatching, pupation, and age-specific survival and in total survival at the end of the selection (F16). Although age-specific fecundity was not affected by selection with deltamethrin, total fertility, together with lower survival, significantly affected gross reproduction rate (GRR), gradually decreasing due to deltamethrin selection. Similarly, net reproductive rate (Ro) and intrinsic growth rate (rm) were affected by selection. CONCLUSION Alterations in life parameters could be due to the accumulation of noxious effects or deleterious genes related to detoxifying enzymes, specifically those coding for mixed-function oxidases, along with the presence of recessive alleles of the V1016I and F1534C mutations, associating deltamethrin resistance with fitness cost in Ae. aegypti.

The prevalent and increasing situations such as the expansion of areas for agricultural use, livestock raising and hunting represent the most complex issues for the conservation of wild species. Mammals of the Order Carnivora are key species in ecosystems because they respond differently to changes in habitat. This study explored the species richness and composition of trophic guilds of the assemblage of the Order Carnivora in an area adjacent to Calakmul Biosphere Reserve, Mexico. From February 2014 to February 2015, 10 stations with camera traps were set within the sub-evergreen tropical forest near water bodies, roads and trails. The structure of the carnivorous assemblage was evaluated through the species richness and trophic guild of species. The recording efficiency of traps was evaluated through the latency to first detection (LTD). A total of 76 photographic records were obtained for eight species of the Order Carnivora, as well as direct observations of three additional species, so the assemblage recorded was composed of 11 species. Four trophic guilds were identified: carnivores (5 species), omnivores (4), insectivores (1), and frugivores (1). The Order consists of two species of large predators (Panthera onca and Puma concolor) and three generalist species (Urocyon cinereoargenteus, Puma yagouaroundi, and Procyon lotor). Subsistence hunting for obtaining wild meat is a common practice in rural neotropical communities. Like other peasant practices disrupted by the COVID-19 pandemic, subsistence hunting could exacerbate pressure on wild mammals whose greater size contributes to feeding the hunter and his family. Thus, in the context of the pandemic, we assessed the subsistence hunting of the white-tailed deer ( Odocoileus virginianus), one of the main traditional game species in the Calakmul region, Campeche, Mexico, and compared this activity with its pre-pandemic levels of such vertebrate species. Based on ethnographic information and hunting records from three rural communities, we found that in one trimester (July-September 2020) of the rainy season, a total of 26 white-tailed deer (923 kg of game biomass) were obtained by local peasant-hunters carrying out hunting mostly alone. Most peasant-hunters interviewed (36 of 51) stated that they hunted daily, and only a few hunted once a week or once a month (8 and 3%, respectively). This hunting activity and modalities were carried out at night (68%) versus day, stalking (21%) and opportunist (11%) near their community. The game biomass and hunting frequency in the studied communities were twice as high during the pandemic, compared to similar pre-pandemic periods in the region. Our survey highlights the need to expand monitoring and evaluation (during and after the pandemic) of subsistence hunting on key species, such as white-tailed deer, in order to ensure conservation and sustainable use of wildlife in this important Mesoamerican region.

Background There is a need for effective methods to control Aedes aegypti and prevent the transmission of dengue, chikungunya, yellow fever and Zika viruses. Insecticide treated screening (ITS) is a promising approach, particularly as it targets adult mosquitoes to reduce human-mosquito contact. Methodology/Principal findings A cluster-randomised controlled trial evaluated the entomological efficacy of ITS based intervention, which consisted of the installation of pyrethroid-impregnated long-lasting insecticide-treated netting material fixed as framed screens on external doors and windows. A total of 10 treatment and 10 control clusters (100 houses/cluster) were distributed throughout the city of Merida, Mexico. Cross-sectional entomological surveys quantified indoor adult mosquito infestation at baseline (pre-intervention) and throughout four post-intervention (PI) surveys spaced at 6-month intervals corresponding to dry/rainy seasons over two years (2012–2014). A total of 844 households from intervention clusters (86% coverage) were protected with ITS at the start of the trial. Significant reductions in the indoor presence and abundance of Ae. aegypti adults (OR = 0.48 and IRR = 0.45, P, Author summary An ITS based intervention, screened houses with insecticide-treated netting (long-lasting insecticidal nets permanently fixed with aluminium frames on external doors and windows), in Merida city, Mexico. Screened houses had >50% less chance of having Ae. aegypti mosquitoes indoors and importantly, >50% fewer female Ae. aegypti in comparison with non-screened houses (control houses). The impact was sustained over 2 years. House screening could provide a feasible sustainable method to reduce human-vector contact inside houses and could potentially be scaled up to protect against mosquito-borne diseases, particularly in locations where the simultaneous transmission, or threat, of dengue, chikungunya, yellow fever and Zika is occurring.
