The real-world safety and effectiveness outcomes were assessed at baseline and at 1, 3, 6, 12, and 18 months post-treatment or the last day of treatment.Results: Most patients received tadalafil 5 mg per day throughout the observation period. Among 1393 patients analyzed for safety, the overall incidence of adverse drug reactions was 8.3%. These adverse drug reactions were generally consistent with the known safety profile of tadalafil and no new safety risks were identified in long-term use. There was no statistical difference in the frequency of adverse drug reactions between patients aged < 75 and ≥ 75 years. The mean change in total International Prostate Symptom Score (IPSS) and IPSS-quality of life subscore was significantly improved at each timepoint. 34. Brain uptake and metabolism of [1-C]octanoate in rats: Pharmacokinetic basis for its application as a radiopharmaceutical for studying brain fatty acid metabolism.

Abstract

The uptake of octanoate in rat brain and its metabolism were investigated by means of intravenously injecting [1-C] or [1-C]octanoate as a tracer. The radioactivity in the cerebrum was increased by an injection of [1-C]octanoate, and reached its peak level (0.33% ID/g) in about 2 to 5 min, and then decreased slowly. The cerebrum-to-blood ratio of the radioactivity increased with time over a period of 30 min. At 30 sec, [ 1-C]octanoate that remained unchanged in the cerebrum accounted for only 8% of the total radioactivity, in spite of there being about 90% in the blood. By means of an injection of [ 1-C]octanoate, more than 70% of the total radioactivity in the cerebrum was found to be attributable to radiolabeled glutamate and glutamine at each time point measured between 30 sec and 30 min. The results show that [1-C]octanoate enters rat brain easily and is trapped in the cerebrum, probably in the form of glutamate and glutamine, and the usefulness of [ 1-C] octanoate as a radiopharmaceutical for studying brain fatty acid metabolism by positron emission tomography is therefore suggested. 35. Automated synthesis of radiopharmaceuticals for positron emission tomography: an apparatus for [1-11 C] labeled carboxylic acid

Abstract

A fully automated apparatus for the synthesis and formulation of short-lived 11 C (t1/2 = 20 min)-labeled carboxylic acids for positron emission tomograpy (PET) has been developed. Injectable solutions of [1-11 C]acetic acid, [1-11 C]octanoic acid and [1-11 C]palmitic acid wilh radioactivities of 6.36-8.29 GBq, 0.070-1.43 GBq and 0.42-0.89 GBq were obtained. The preparation time was under 40 min after the end of bombardment. An automatic washing function means that labeled compound of the same or different kinds can be produced several times a day without any maintenance of the system. The control system is sited away from the ‘hot laboratory’, so operator exposure to radiation is minimized.
