Abstract

0000-0002-0602-0665, The characteristics of a gas-temperature-controllable atmospheric-pressure helium plasma jet and the development of an experimental system for cell viability assays of yeasts (fission yeast Schizosaccharomyces pombe and budding yeast Saccharomyces cerevisiae) are reported. The physicochemical properties of the plasma plume, which can maintain the temperature of the irradiated object at a temperature suitable for yeast, were not significantly different from those of a typical helium plasma jet. Furthermore, good reproducibility of cell viability was observed when gas temperature, gas flow rate, applied high voltage, and irradiation distance remained fixed, and only irradiation time was used as a parameter. This experimental system allows us to carry out various experiments, such as the search for plasma-resistant mutants that will contribute to the identification of genes involved in resistance to direct plasma irradiation.

38. Distribution of aquaporin genes and selection of individual reference genes for quantitative real-time RT-PCR analysis in multiple tissues of the mouse

Abstract

Aquaporins (AQPs) are a family of water-transporting proteins that are selectively expressed in epithelial, endothelial, and many other cell types of various tissues, where they play important physiological functions. However, the accurate distribution of AQP gene expression has not yet been examined in various tissues of the mouse. We first evaluated the tissue distribution of AQP gene expression using tongue, nasal epithelium, bronchus, trachea, lung, esophagus, stomach, ileum, transverse colon, liver, pancreas, whole blood, thigh muscle, spinal cord, brain, thoracic aorta, heart, kidney, thymus, spleen, skin, eye, and testis of the mouse. Furthermore, for a quantitative analysis, we selected appropriate reference genes for normalized qRT-PCR data in various tissues. The stability of the reference genes was assessed using NormFinder. The stably expressed genes identified in the present study were 18s rRNA. When 18s rRNA was used, as the best reference gene in the present study, the genes for AQPs 0,1,2,3,4,5,6,7,8,9,11, and 12 were notably expressed in the eye, lung, testis, eye, spinal cord, trachea, kidney, testis, testis, testis, testis, and pancreas. These results, regarding the distribution of AQPs, suggest that AQPs may be involved in various physiological and pathophysiological processes. Les aquaporines (AQP) consistent en une famille de proteines transporteuses d'eau qui sont exprimees de maniere selective dans les cellules epitheliales et endotheliales, de meme que dans differents autres types de cellules de divers tissus, ou elles exercent des fonctions physiologiques importantes. Cependant, la distribution precise de l'expression genique des AQP n'a pas encore ete examinee dans divers tissus chez la souris. Les auteurs ont d'abord evalue la distribution tissulaire de l'expression genique des AQP dans la langue, l'epithelium nasal, les bronches, la trachee, les poumons, l'resophage, l'estomac, l'ileon, le colon transverse, le pancreas, le sang total, le muscle de la cuisse, la moelle epiniere, le cerveau, l'aorte thoracique, le creur, les reins, le thymus, la rate, la peau, l'reil et les testicules de souris. De plus, pour les fins de l'analyse quantitative, les auteurs ont selectionne des genes de reference appropries afin de normaliser les donnees en qRT-PCR dans les divers tissus. La stabilite des genes de reference a ete evaluee par NormFinder. Le gene ARNr 18S etait celui qui etait exprime de fa^on stable dans la presente etude. Lorsque ARNr 18S etait utilise, etant le meilleur gene de reference, les genes AQP 0,1, 2, 3, 4, 5, 6, 7, 8, 9,11 et 12 etaient particulierement exprimes dans l'reil, le poumon, le testicule, l'reil, la moelle epiniere, la trachee, le rein, le testicule, le testicule, le testicule, le testicule et le pancreas, respectivement. Ces resultats en ce qui concerne la distribution des AQP suggerent que les AQP peuvent etre impliquees dans differents processus physiologiques et patho-physiologiques. Abstract

The suitable temperature condition for fission yeast growth is around 30 °C, whereas the gas temperature of commonly used atmospheric-pressure helium plasma jets tends to increase with discharge time. To avoid the damage of fission yeast from heat, a Peltier device was used to control the feeding gas temperature of low-frequency helium plasma jet. The temperature of a thermistor directly irradiated by the plasma jet plume remained almost constant at the acceptable temperature to fission yeast for more than 200 s. The morphology of fission yeast cells exposed to the gas-temperature-controlled plasma jet plume was examined. Abstract

平成30年度, To investigate differences in chronological variations in characteristics and outcomes of out-of-hospital cardiac arrests (OHCAs) between elderly and non-elderly patients., We retrospectively analyzed bystander-witnessed OHCAs without prehospital involvement of physicians between January 2007 and December 2014 in Japan. We considered the following time periods: night-time (23:00-5:59) and non-night-time; we further divided non-night-time into dinnertime (18:00-20:29) and other non-night-time. Subsequently, we analyzed chronological variations in factors associated with OHCA survival using univariate and multivariable logistic regression analyses for unmatched and propensity-matched pairs, respectively., For elderly (≥65 years old, N = 201,073) and non-elderly (≥10, Dinnertime, particularly in winter, is associated with lower survival in elderly OHCA patients.
