Abstract

Research was conducted to examine the phylogenetic diversity of methanogens in Reticulitermes speratus termites collected in the Japan archipelago using the PCR approach. The sequencing of 60 clones of archaeal small-subunit rRNA genes amplified from the termite showed that majority, 56 clones, clustered in the genus Methanobrevibacter. Results indicate that Methanobrevibacter-related methanogens were the main species colonizing the hindgut of the R. speratus.

18. Symbiosis in Termites.

Abstract

Termites are divided into two types, the lower termite, which possesses symbiotic protozoa and several species of bacteria in the intestine and the higher termite, which has no protozoa but several species of symbiotic bacteria in the intestine. The digestive system for cellulose differs in each. In the higher termites, the digestive system is complex, since several problems remain unresolved. In the lower termites, cellulose digestion depends on the intestinal protozoa and symbiotic relationships between the termite and its intestinal protozoa, first published by Cleveland in 1924 (xcCleveland, 1924A). Recent reports, however, show that the symbiotic relationship in the digestive system involving cellulose is more complex. xcYamaoka and Nagatani (1975) reported that the termites themselves produce a cellulase which differs from the cellulase produced by the intestinal protozoa. Since the concept of the cellulose digestion system has changed significantly, the role of intestinal bacteria and their diversity in termites are presented here. [ABSTRACT FROM AUTHOR]

26. Establishment of MIN-Lymphosarcoma as a Permanent Cell Strain in Tissue Culture

Abstract

Three tissue culture strains were established from MN-lymphosarcoma cells originally carried intraperitoneally in NA2 female mice. They have been maintained by serial subcultivation for more than 16 months. There were no apparent differences except for cell size in morphological features among the 3 trains. When inoculated into mice intermittetly after prolonged maintenance in vistro, strain No. 1 and No. 3 cells induced ascites tumor formation and a mortality of 80～100%. Strain No. 2 cells seemed to have lost their transplantability. When the cultured stain No. 1 cells were longer in the first passage, was shortened within a few geberations to that of tha original mouse-to-mouse passage. This result might be discussed on the hupothesis of a gradual transformaion of cells for adaptaion to growth in altered enviroment.

27. Osmium-impregnation patterns of the golgi complex in the epididymal epithelial cells of castrated and testosterone-injected mice

Abstract

After prolonged exposure of mouse epididymal epithelial cells to a solution of osmium tetroxide, reduced osmium compounds were detected in the Golgi cisternae and in the cytoplasm adjacent to the Golgi complex. Their appearance changed in time under certain conditions. Eight days after castration each cisterna of the regularly arranged Golgi lamellae fragmented into small vesicles, in which deposits of reduced osmium compound were in reduced amounts or completely absent, but no notable decrease of the fine reduced osmium particles in the cytoplasm adjacent to the Golgi complex occurred. The amount of deposit in the Golgi cisternae in castrated mice recovered to the normal level after the subcutaneous injection of testosterone for two weeks. On the other hand, the osmium particles observed in the cytoplasm adjacent to the Golgi complex during the recovery process of the lamellar structure increased in amount. This study showed that the reduced osmium compounds were of two kinds, and that the deposits contained in the Golgi cisternae were related to secretory products, and might be controlled by hormonal factors, but the fine reduced osmium particles that appeared in the cytoplasm adjacent to the Golgi complex might be regulated by other factors.
