2. Predictions and Uncertainty Estimates of Reactor Pressure Vessel Steel Embrittlement Using Machine Learning

Abstract

An essential aspect of extending safe operation of the active nuclear reactors is understanding and predicting the embrittlement that occurs in the steels that make up the Reactor pressure vessel (RPV). In this work we integrate state of the art machine learning methods using ensembles of neural networks with unprecedented data collection and integration to develop a new model for RPV steel embrittlement. The new model has multiple improvements over previous machine learning and hand-tuned efforts, including greater accuracy (e.g., at high-fluence relevant for extending the life of present reactors), wider domain of applicability (e.g., including a wide-range of compositions), uncertainty quantification, and online accessibility for easy use by the community. These improvements provide a model with significant new capabilities, including the ability to easily and accurately explore compositions, flux, and fluence effects on RPV steel embrittlement for the first time. Furthermore, our detailed comparisons show our approach improves on the leading American Society for Testing and Materials (ASTM) E900-15 standard model for RPV embrittlement on every metric we assessed, demonstrating the efficacy of machine learning approaches for this type of highly demanding materials property prediction.

Abstract

The effects of transcription factors on the maintenance and differentiation of human-induced or embryonic pluripotent stem cells (iPSCs/ESCs) have been well studied. However, the importance of posttranscriptional regulatory mechanisms, which cause the quantitative dissociation of mRNA and protein expression, has not been explored in detail. Here, by combining transcriptome and proteome profiling, we identified 228 posttranscriptionally regulated genes with strict upregulation of the protein level in iPSCs/ESCs. Among them, we found 84 genes were vital for the survival of iPSCs and HDFs, including 20 genes that were specifically necessary for iPSC survival. These 20 proteins were upregulated only in iPSCs/ESCs and not in differentiated cells derived from the three germ layers. Although there are still unknown mechanisms that downregulate protein levels in HDFs, these results reveal that posttranscriptionally regulated genes have a crucial role in iPSC survival.

Abstract

In vitro fertilization (IVF) and intracytoplasmic sperm injection (ICSI) are 2 major assisted reproductive techniques (ARTs) used widely to treat infertility. Recently, spermatogonia! transplantation emerged as a new ART to restore fertility to young patients with cancer after cancer therapy. To examine the influence of germ cell manipulation on behavior of offspring, we produced [F.sub.1] offspring by a combination of two ARTs, spermatogonial transplantation and ICSI. When these animals were compared with [F.sub.1] offspring produced by ICSI using fresh wild-type sperm, not only spermatogonial transplantation- ICSI mice but also ICSI-only control mice exhibited behavioral abnormalities, which persisted in the [F.sub.2] generation. Furthermore, although these [F.sub.1] offspring appeared normal, [F.sub.2] offspring produced by IVF using [F.sub.1] sperm and wild- type oocytes showed various types of congenital abnormalities, including anophthalmia, hydrocephalus, and missing limbs. Abstract

Nanostructured ferritic alloys (NFAs) contain an ultra-high density of 2–4 nm fcc pyrochlore Y2Ti2O7 nano-oxides (NOs) embedded in a bcc Fe-14Cr ferritic matrix. Characterization of helium interactions with NOs and associated Fe-Y2Ti2O7 interfaces is important to the development of structural materials for nuclear fusion and fission applications. A benchmark 14YWT NFA was first annealed to coarsen the NOs, then insoluble helium was implanted at 700 °C to produce a high number density of bubbles. High-resolution scanning transmission electron microscopy characterization shows two dominant Fe-Y2Ti2O7 crystallographic orientation relationships (cube-on-edge and cube-on-cube). The smallest NOs (≈ 2 nm) are associated with the smaller bubbles (≈ 1.5 nm), while some of the largest NOs (> 6 nm) have larger, and sometimes multiple, bubbles. NO corner {111} facets are the preferred sites for He bubble nucleation. A refined sequence of events for He trapping and bubble formation is presented. These observations offer new insight on He management in NFAs, and provide a foundation for detailed modeling studies.
