16. Analysis of sulfur in deposited aerosols by thermal decomposition and sulfur dioxide analyzer

Abstract

A thermal decomposition method that measures aerosol sulfur at the nanogram level directly from the collection substrate is described. A thermal decomposition apparatus was designed. A stainless steel strip was used as the aerosol collection substrate. A 0.1 mol/L Mn[Cl.sub.2] solution was added as the thermal decomposition catalyst. Currents were passed through the strip where aerosol particles had been deposited. In this way, the strip was heated at 780 [+ or -] 10 [degrees]C, and particulate sulfur was evaporated. A sulfur dioxide analyzer (SDA) with flame photometric detector (FPD) was used to detect gaseous sulfur. High sulfur recoveries from [(N[H.sub.4]).sub.2]S[O.sub.4] and other inorganic sulfates, such as N[H.sub.4]HS[O.sub.4], [K.sub.2]S[O.sub.4], MgS[O.sub.4], and Ca[SO.sub.4], were obtained. From the sulfur blank and the calibration, a lower limited detection of 0.2 ng of sulfur and the determination range of 3.3-167 ng of sulfur were estimated. The method is effective for measuring the sulfate size distributions of urban aerosols in a small sample air volume of 50-60 L. The method is applicable to measuring the sulfur in aqueous extracts of size-segregated urban aerosols collected by impactor and comparing the results with the sulfate data measured by ion chromatography.

Abstract

Thermal decomposition-chemiluminescence was developed to determine nanogram quantities of aerosol nitrate rapidly. Standard nitrates placed on a stainless steel strip were decomposed by resistance heating of the strip and converted to NOx, which was measured with a chemiluminescent NOx analyzer. By adding 1 micron L of 0.1 N NaOH and 0.1 mol/L CrO3 prior to heating, ammonium interferences was eliminated and about 97-99% NOx conversion from nitrate was achieved. For standard NaNO3 and NH4NO3, a determination range of 5-2000 ng and a detection limit of 1.1 ng of nitrate were obtained. Interferences from the particle nitrite and the gaseous HNO3, HNO2, NO, and NO2 in sampling process were almost negligible. Other inorganic nitrates such as KNO3, Ca(NO3)2, and Fe(NO3)3 were examined. Among aqueous extracts of atmospheric aerosols, the nitrates quantified using this method agreed well with the results obtained by means of ion chromatography. The method is effective for measuring the size distribution of a nitrate aerosol in ambient air, when air volumes as small as 30 L are sampled.

Abstract

Multicellular organisms repair injured epithelium by evolutionarily conserved biological processes including activation of c-Jun N-terminal kinase (JNK) signaling. Here, we show in Drosophila imaginal epithelium that physical injury leads to the emergence of dying cells, which are extruded from the wounded tissue by JNK-induced Slit-Roundabout2 (Robo2) repulsive signaling. Reducing Slit-Robo2 signaling in the wounded tissue suppresses extrusion of dying cells and generates aberrant cells with highly upregulated growth factors Wingless (Wg) and Decapentaplegic (Dpp). The inappropriately elevated Wg and Dpp impairs wound repair, as halving one of these growth factor genes cancelled wound healing defects caused by Slit-Robo2 downregulation. Our data suggest that JNK-mediated Slit-Robo2 signaling contributes to epithelial wound repair by promoting extrusion of dying cells from the wounded tissue, which facilitates transient and appropriate induction of growth factors for proper wound healing.

Abstract

Although many attempts have been made to study human colonic motility, the colonic transit is still poorly understood. Both spontaneous and neostigmine-induced peristalsis of the colon were studied with scintigraphy. A polythene tube was inserted into the cecum through a colonofiberscope. 37 MBq of ⁹⁹ᵐTc-DTPA and 75 ml of saline were instilled and dynamic scan was begun. Eight healthy volunteers were examined by the method above mentioned. The sampling time was set at fifteen seconds in six persons and three seconds in the rest. 0.5 mg of neostigmine was injected intravenously to stimulate the paristalsis when no peristalsis occurred within thirty minutes after the study was begun. Dynamic scanning was performed for sixty to ninety minutes. This scintigraphic study revealed that the spontaneous and induced peristalsis were almost identical on colonogram. ⁹⁹ᵐTc-DTPA solution was propelled from the cecum and ascending colon to the sigmoid colon or the rectum for about fifteen seconds during mass peristalsis. Colonogram (time-activity curve) enables us to analyze mass peristalsis easily and more objectively than colonoscintigram. The spontaneous and neostigmine-induced peristalsis seemed to be almost identical in all but one of eight subjects.
