Este trabalho objetiva identificar a percepção dos profissionais cadastrados numa Universidade do Sul do Brasil sobre nível de orientação empreendedora (OE) nas organizações em que atuam. O arcabouço teórico incluiu a reflexão sobre o empreendedorismo corporativo e a orientação empreendedora. O processo de coleta dos dados consistiu com a utilização de um questionário estruturado com base nas variáveis de França, Saraiva e Hashimoto (2012) sobre a temática. Foram enviados 12.000 e-mails para executivos matriculados numa Universidade do Sul do Brasil. Destes, 383 questionários se enquadraram ao perfil da amostra pretendida para o estudo, tendo em vista que aqueles que não trabalhavam regularmente numa organização, foram eliminados da amostra. Os resultados apontaram que o nível de orientação empreendedora não está associado com o gênero ou o segmento das organizações que os respondentes estavam vinculados, porém pode-se concluir que o nível OE está relacionado ao cargo ocupado pelos respondentes.

Objective To investigate a mechanistic determinant of left atrial (LA) reservoir function in patients presenting left ventricular (LV) hypertrophy, and clarify diagnostic and prognostic values of LA reservoir strain in patients with cardiac amyloidosis (CA). Methods Three-hundred sixty patients (median age: 68 years, male gender: 65%) with left ventricular hypertrophy (LVH) assessed by echocardiography were retrospectively included. The LVH etiologies were diagnosed by any of biopsy, cardiac magnetic resonance imaging or 99mTc-PYP scintigraphy. LV segmental longitudinal strain was estimated from apical three views, and LA reservoir strain was measured from an apical 4-chamber view. Results The LVH etiologies were confirmed with CA in 81 patients, hypertensive heart disease in 87 patients, hypertrophic cardiomyopathy in 143 patients, and miscellaneous disorders in 49 patients. The median (25th, 75th percentile) value of LV ejection fraction was 59% (48–67). LV basal longitudinal strain and LA reservoir strain were significantly reduced in patients with CA compared with those with other etiologies; LV basal strain: 5.4% (3.9–8.7) vs. 11.9% (9.3–14.6), LA reservoir strain: 9.2% (6.3–12.3) vs. 17.5% (11.3–24.1), p Conclusions The decrease of LV longitudinal shortening at cardiac base could worsen LA reservoir function especially in patients with CA. LA reservoir strain might be an alternative measure to identify CA etiology and have a predictive value of heart failure hospitalization in patients with LV hypertrophy. Objectives: Mainstays of contemporary therapies for this arterial occlusive disease include angioplasties, stents, endarterectomies and bypass surgery. However, these treatments suffer from high failure rates due to re-occlusive vascular wall adaptations, namely intimal hyperplasia (IH). IH develops in response to endothelium injury, leading to inflammation, vascular smooth muscle cells (VSMC) dedifferentiation, migration and proliferation at the site of injury. Hydrogen sulfide (H 2 S) is a ubiquitous signaling gazotransmitter, which exhibits antioxidant, anti-inflammatory, and vaso-relaxant properties. Thus, we hypothesized that H 2 S could reduce IH formation. Methods: WT male C57BL6/J mice submitted to femoral wire injury surgery to induce IH were treated with an H 2 S donor (NaHS) in the drinking water. IH was measured 28 days post-surgery by histology. In addition, segments of great saphenous vein obtained from patients undergoing bypass surgery were maintained in culture ex-vivo for 7days in presence of various H 2 S donors (NaHS, GYY4137, diacyltrisulfide). Finally, primary human umbilical vein endothelial cells (HUVEC) and primary human VSMC were treated in-vitro with the same H 2 S donors to study cellular proliferation and migration. Results: NaHS treatment significantly reduced IH development in the mouse model of femoral wire injury ( Figure 1 ). Similarly, the various H 2 S donors prevented the development of IH in vein segments ex-vivo. In vitro , the same H 2 S donors stimulated human endothelial cells (HUVEC) migration and proliferation, while inhibiting migration and proliferation of primary VSMC ( Figure 2 ). Conclusions: Exogenous H 2 S prevents IH formation in mice in-vivo and in human veins ex-vivo . Importantly, H 2 S reduces VSMCs but stimulates ECs proliferation and migration. These data suggest that exogenous H 2 S therapy could be used in human to minimize IH, thus limiting vascular reconstruction failure.

Trauma is a leading cause of death and disability worldwide. Corneal tissue donors generally are those who suffered an injury to the brain or fatal trauma caused by stroke, vehicle/motorbike accidents, gunshot wounds, and drowning or cardiovascular death. In Brazil, the Distrito Federal (DF) Eye Bank, located within a trauma center hospital, and the Secretariat of Public Security have collaborated with the aim of increasing the overall number of cornea donations from fatal trauma victims. The purpose of this study was to determine the suitability of cornea tissue for transplantation derived from trauma-related death. The records of eyes donated in the DF Eye Bank were analyzed retrospectively for the period from 2004-2013. We had 3388 cornea donors, the majority of which were between 21 and 30 years old (17.4%), which were derived from violent death (84.1%; P = .00) and were predominately male (73.5%). Among the donated corneas, 54.0% were used for optic purposes. Mechanical trauma caused by gunshot, stabbing or blunt force (23.7%), and road traffic injuries (11%) were the main causes of violent death. Another common cause of death was cardiovascular disease (26.3%). Donor tissue derived from violent death had no statistical interference on tissue suitability for transplantation (P = .06). Because of the large waiting lists, and waiting times for transplants, it is advisable to increase the available tissue from corneas donors derived from violent death through the implementation of this interagency model of collaboration and by the practicing of active tissue donor screening in trauma center hospitals.

In recent years, due to the development of laser-measurement technology, 3D laser-scanned point clouds for cultural assets are often used as the recording format of digital archiving. The acquired point clouds are large-scale and precisely record complex 3D internal structures of the scanned objects. Visualization quality of such point clouds highly depends on density distributional uniformity, that is, the uniformity of the inter-point distances. The visualization quality can be improved by making point distances uniform. In addition, the visibility further improves by emphasizing edges. In this study, first the uniformity and flexible tuning of the point density based on Poisson disk sampling are examined. The resultant high-quality point clouds are then applied to transparent visualization. Moreover, edge emphasis visualization is realized by combining the principal component analysis calculating feature amount and Poisson disk sampling.

No. of bitstreams: 0 Previous issue date: 2012-04-01 The search for molecular markers to improve diagnosis, individualize treatment and predict behavior of tumors has been the focus of several studies. This study aimed to analyze homeobox gene expression profile in oral squamous cell carcinoma (OSCC) as well as to investigate whether some of these genes are relevant molecular markers of prognosis and/or tumor aggressiveness. Homeobox gene expression levels were assessed by microarrays and qRT-PCR in OSCC tissues and adjacent non-cancerous matched tissues (margin), as well as in OSCC cell lines. Analysis of microarray data revealed the expression of 147 homeobox genes, including one set of six at least 2-fold up-regulated, and another set of 34 at least 2-fold down-regulated homeobox genes in OSCC. After qRT-PCR assays, the three most up-regulated homeobox genes (HOXA5, HOXD10 and HOXD11) revealed higher and statistically significant expression levels in OSCC samples when compared to margins. Patients presenting lower expression of HOXA5 had poorer prognosis compared to those with higher expression (P=0.03). Additionally, the status of HOXA5, HOXD10 and HOXD11 expression levels in OSCC cell lines also showed a significant up-regulation when compared to normal oral keratinocytes. Results confirm the presence of three significantly upregulated (>4-fold) homeobox genes (HOXA5, HOXD10 and HOXD11) in OSCC that may play a significant role in the pathogenesis of these tumors. Moreover, since lower levels of HOXA5 predict poor prognosis, this gene may be a novel candidate for development of therapeutic strategies in OSCC. A alelopatia caracteriza-se pelos efeitos danosos ou beneficos que metabolitos secundarios produzidos por plantas, microrganismos ou fungos liberados no ambiente exercem sobre o desenvolvimento de sistemas biologicos naturais ou implantados. O objetivo do trabalho foi avaliar o efeito alelopatico de extratos aquosos de eucalipto ( Eucalyptus globulus Labill.) e guacatonga ( Casearia sylvestris Sw.) na germinacao e no crescimento inicial de mostarda ( Brassica campestris L.), repolho ( Brassica oleracea L. cv. capitata), brocolis ( Brassica oleracea L. cv. italica), couve ( Brassica pekinensis L.), alface ( Lactuca sativa L. cv. grand rapids), tomate ( Lycopersicum esculentum Miller), nabo ( Brassica rapa L.), rucula ( Eruca sativa L.) e rabanete ( Raphanus sativus L.). Foram testadas seis concentracoes de cada extrato aquoso (10, 30, 50, 70, 90 e 100%) e comparadas com controle (agua destilada), com cinco repeticoes cada, sendo distribuidas dez sementes das especies cultivadas em cada repeticao. Os extratos aquosos de E. globulus e C. sylvestris reduziram significativamente o percentual de germinacao das sementes, o indice de velocidade de germinacao, o crescimento inicial da parte aerea e do sistema radicular de todas as especies cultivadas, com reducao nestes parâmetros com aumento das concentracoes utilizadas e, acarretando raizes mais espessas, atrofiadas e com maior numero de pelos absorventes. Assim, os resultados indicam a existencia de potencial alelopatico de E. globulus e C. sylvestris.

Allelopathy is characterized by the harmful or benefic effects caused by secondary metabolites, that are produced by plants, microorganisms or fungi and are released in the environment, on the development of natural biological systems or implemented ones. This study aimed to evaluate the allelopathic effects of aqueous extracts of eucalypt (Eucalyptus globulus Labill.) and wild coffee (Casearia sylvestris Sw.) on the germination and initial development of mustard (Brassica campestris L.), cabbage (Brassica oleracea L. cv. capitata), broccoli (Brassica oleracea L. cv. italica), kale (Brassica pekinensis L.), lettuce (Lactuca sativa L. cv. grand rapids), tomato (Lycopersicum esculentum Miller), turnip (Brassica rapa L.), rucola (Eruca sativa L.) and radish (Raphanus sativus L.). Six concentrations of each aqueous extract were tested (10, 30, 50, 70, 90 and 100%) and compared to control (distilled water), with five replicates of each concentration, being ten seeds of each crop distributed in each replicate. The aqueous extracts of E. globulus and C. sylvestris reduced significantly the percentage of seed germination, the index of germination speed and the initial growth of the above ground part and roots of all cultivated species, being the reduction of these parameters higher with the increment of the aqueous extracts concentration, which led to thicker and atrophied roots with a higher number of absorbent hairs. The relation between the properties of polyampholytes in aqueous solution and their adsorption behaviors on silica and cellulose surfaces was investigated. Four polyampholytes carrying different charge densities but with the same nominal ratio of positive to negative segments and two structurally similar polyelectrolytes (a polyacid and a polybase) were investigated by using quartz crystal microgravimetry using silica-coated and cellulose-coated quartz resonators. Time-resolved mass and rigidity (or viscoelasticity) of the adsorbed layer was determined from the shifts in frequency (Deltaf) and energy dissipation (DeltaD) of the respective resonator. Therefore, elucidation of the dynamics and extent of adsorption, as well as the conformational changes of the adsorbed macromolecules, were possible. The charge properties of the solid surface played a crucial role in the adsorption of the studied polyampholytes, which was explained by the capability of the surface to polarize the polyampholyte at the interface. Under the same experimental conditions, the polyampholytes had a higher nominal charge density phase-separated near the interface, producing a soft, dissipative, and loosely bound layer. In the case of cellulose substrates, where adsorption was limited, electrostatic and polarization effects were concluded to be less significant.

O objetivo deste trabalho foi analisar e quantificar a variabilidade genética entre e dentro das populações de arnica por meio de marcadores RAPD. Foram amostradas quatro populações na região geoeconômica do Distrito Federal: Parque Nacional de Brasília (2), Fazenda Água Limpa - UnB (1) e Reserva do Instituto Brasileiro de Geografia e Estatística (IBGE) (1). Folhas de 24 indivíduos de cada região foram coletadas, totalizando 96 indivíduos. Num total de 105 iniciadores testados foram selecionados 15, totalizando 60 bandas polimórficas. Marcadores RAPDs selecionados foram analisados com a utilização dos programas NTSYS e Amova. O dendrograma obtido pelo método UPGMA e coeficiente de dissimilaridade Dice evidenciou quatro agrupamentos consistentes, com índice de dissimilaridade variando entre 62 a 71%. O teste de Mantel aplicado estabeleceu uma correlação cofenética com valores de r = 0.82, significando que as distâncias geográficas entre as populações amostradas estão correlacionadas com a distância genética. A análise de AMOVA mostrou uma percentagem variabilidade genética entre populações de 35,7% e dentro de populações de 64,3%, evidenciando uma alta variação entre populações, sendo um importante resultado para definição de uma estratégia de conservação da espécie que se encontra em situação vulnerável à extinção. The main objective of this research was to analyze and quantify the genetic variability within and between populations of arnica using RAPD markers. Four populations from Federal District area, Brazil were sampled: Parque Nacional de Brasília - (2 ), Fazenda Água Limpa -UnB (1), and Reserva do Instituto Brasileiro de Geografia e Estatística (IBGE) (1). Leaves from twenty-four individuals from each population were collected and preserved under refrigeration. Fifteen primers were selected from 105 tested, totalizing 60 polymorphic bands. Scored RAPD markers were analyzed using NTSYS and Amova. The results indicated four consistent clusters, with dissimilarity index varying from 62 to 71%. The Mantel test indicates a cophenetic correlation (r-0.82), which means that the geographic distances are correlated to the genetic distances. An AMOVA analysis presented 35.7% variation among populations, and 64.3% within populations, showing a high variation among populations. This is an important result for conservation strategy for such species considered vulnerable to extinction.

Neste artigo, apresenta-se o desenvolvimento de um aplicativo computacional, de caráter educativo, realizado por alunos e orientado por professores do Centro Universitário Nove de Julho (Uninove), que resolve problemas de transferência de calor. Para exemplificar a utilização do aplicativo, apresenta-se um caso empregado.

The objective of the present study was to test the validity of a semiologic maneuver on the diagnosis of peritrochanteric bursitis. Thirty patients with a clinical diagnosis of great trochanter bursitis were assessed. All patients were submitted to X-ray studies at AP and LP. Whenever necessary, other imaging tests were performed, intending to rule out any associated diseases; the patients were submitted to two semiologic maneuvers on both hips, the affected and the normal one. TEST 1: with patient at supine position, the examined limb is adduced in extension and crossing it over the contralateral limb, with the calcaneus touching the bed, a hip flexion is produced at approximately 90o; at the end of this maneuver, the hip will be flexed, adduced and slightly externally rotated; during this maneuver, the patient may report pain at major trochanter region. TEST 2: the final position of TEST 1 is the initial position of TEST 2, with a forced hip adduction where pain or exacerbation may be reported. The maneuver results were submitted to statistical analysis, with both tests showing to be different from each other while supplementary. Test 2 was shown to be positive in 96.6%. Semiologic maneuvers referred as TEST 1 and TEST2 may be used to assist on providing a clinical diagnosis of peritrochanteric bursitis.

In western countries breast cancer is still the leading cause of death of women. Very promising results have been obtained by combining vinorelbine and doxorubicin, two of the most active drugs in metastatic breast cancer. However, despite the activity reported, this combination has shown a 10% rate of grade 2-4 cardiac toxicity, mainly due to the total cumulative doses of anthracycline delivered. The aim of this study was to divide the total dose of doxorubicin into two administrations on days 1 and 8, in order to cut down its toxicity while maintaining the same activity. Fifty-two chemotherapy naïve patients with metastatic breast cancer entered into the study and were treated with vinorelbine 25 mg/m2 plus doxorubicin 25 mg/m2 both on days 1 and 8 every three weeks. Fifty-one patients were eligible and evaluable for toxicity while 47 of them were evaluable for activity. Haematological toxicity was predominantly related to neutropenia, with grade 3/4 in 16% of cycles. Non-haematological toxicity was represented by alopecia grade 3 (which affected 65% of the patients), local phlebitis and severe constipation. No clinically significant cases of neuropathy or cardiac dysfunction were seen. With regard to activity, 38 out of 47 patients (80%) responded to therapy, nine of them achieving complete responses (19%). Median response duration was 16 months and the median overall survival was 22.7 months. We conclude that the fractionated administration of vinorelbine and doxorubicin is associated with excellent haematological and non-haematological tolerability (especially as regards cardiac toxicity), coupled with high levels of activity comparable to those observed using regimens based on unfractionated administration of treatment.

Background Pancreatic cancer represents a malignancy with very poor clinical prognosis and limited therapeutic potential. Recent developments of gene transfer technology offer new therapeutic avenues by delivering recombinant genes directly into normal or neoplastic tissue in vivo. Methods Here we show that the LacZ marker gene, complexed to cationic liposomes, can be introduced into the pancreas by either intraductal or intra-arterial injection. Expression of the β-galactosidase gene product was monitored by polymerase chain reaction and histochemistry. Results Up to 28 days after in vivo gene transfer, β-galactosidase activity could be demonstrated in the pancreas. Intraductal application induced gene expression in lining duct cells preferentially. Twenty-four hours after intraductal injection of liposomes, a dose-dependent, transient increase in serum amylase levels was detected. Nevertheless, no histological signs of pancreatitis were evident. Intra-arterial injection resulted in β-galactosidase expression in endothelial cells of intrapancreatic arteries, as well as in the spleen, lymph nodes and liver, but not in ductal cells of the pancreas. Only occasionally were acinar cells positive for blue staining by either type of treatment. Conclusion These experiments demonstrate that in vivo gene transfer into the pancreas is feasible using DNA–liposome complexes. Furthermore, the route of administration largely determines cell type specificity and side-effects. This technique might have an impact for the development of gene therapy strategies for pancreatic diseases.
