Abstract

This article discusses a case report of a successful bone marrow transplantation in a patient with Griscelli syndrome type 2 (GS2), a rare genetic disorder characterized by silver-gray hair and hypopigmentation. The patient underwent transplantation using a non-busulfan-based regimen and post-transplantation cyclophosphamide administration, resulting in a successful outcome with minimal adverse events. The article also reviews the literature on GS2 patients who received transplantation with non-busulfan-based regimens, highlighting the lower incidence of veno-occlusive disease in this group compared to those who received busulfan-based regimens. The study suggests that avoiding busulfan-based regimens may improve outcomes for transplantation in GS2 patients. [Extracted from the article]

Abstract

Although cancer personalized profiling by deep sequencing (CAPP‐Seq) of cell‐free DNA (cfDNA) has gained attention, the clinical utility of circulating tumour DNA (ctDNA) in extramammary Paget's disease (EMPD) has not been investigated. In this study, genomic alterations in the cfDNA and tumour tissue DNA were investigated in seven patients with metastatic EMPD. CAPP‐Seq revealed mutations in 18 genes, 11 of which have not yet been reported in EMPD. The variant allele frequency of some of the mutated genes reflected the disease course in patients with EMPD. In one patient, the mutation was detected even though imaging findings revealed no metastasis. In another patient with triple EMPD (genital area and both axilla), cfDNA sequencing detected the mutation in a rib metastatic lesion, which was also detected in both axilla lesions but not the genital region. Investigations of the ctDNA may be useful towards the elucidation of clonal evolution in EMPD. [ABSTRACT FROM AUTHOR]

Abstract

Purpose: The present study aimed to evaluate the effects of zinc deprivation on the properties of membrane and spike-discharge features of mesencephalic trigeminal neurons (MTNs), which are important sensory neurons for oral-motor reflexes and rhythmical jaw movements.Methods: Neonatal Sprague Dawley rats (P10-12) were distributed equally into a normal diet group and a zinc-deficient diet (ZD) group. Whole cell patch-clamp recordings were obtained from MTNs from coronal brain slices.Results: Passive membrane properties showed a modest depolarized membrane potential and decreased cell capacitance in the ZD group. Zinc deprivation decreased the minimal current amplitude, which induced an action potential and increased the amplitude of afterhyperpolarization following the action potential. Negligible changes were observed for other action potential properties. A decreased burst duration was observed, accompanied by hastened spike frequency adaptation in the burst discharge. There was no difference in the resonant properties at both the subthreshold depolarized potential and hyperpolarized membrane potential between the control and ZD groups.Conclusion: These results suggests that neither the persistent sodium conductance nor slow inwardly rectifying conductance were altered; however, there appeared to be an increase in Ca2+-dependent K+ conductance in zincdeficient MTNs. Abstract

The present study examined whether the optimum slurry conditions in which the green body with the highest packing fraction was fabricated were the same regardless of whether tape or slip casting methods were used. Additionally, we investigated the optimum slurry evaluation method to predict the packing fraction of each green body. Specifically, aqueous dense alumina slurries, whose particle dispersion state was changed by varying pH and the amount of dispersant additive, were prepared. After changing the pH, the optimum slurry conditions were achieved regardless of the casting method. The most suitable amount of additive dispersant differed due to the changing solution conditions during the casting and drying process; the medium of the slurry permeated into the plaster during slip casting and dried on the substrate during tape casting. Thus, in slip casting, non‐adsorbed polymers could also be permeated. Hence, it is crucial for slurry evaluation to predict the packing fraction of the slip cast body. A sedimentation test, which involved particle condensation as well as casting, was valid for the green sheet. Conversely, it was experimentally shown that the constant pressure filtration test, which involved the permeation of the medium in addition to particle condensation, was suitable for slip casting. 34. Atypical lipomatous tumor of the buccal mucosa: A case report and review of literature

Abstract

Atypical lipomatous tumor (ALT), one of the histologic subtypes of liposarcoma, is locally aggressive with intermediate malignant potential and is categorized as a synonymous entity with well-differentiated liposarcoma (ALT/WDLPS). Although ALT/WDLPS is the most common subtype of liposarcoma in the oral and maxillofacial region, its occurrence is extremely rare, and asymptomatic clinical behavior often delays definitive diagnosis and treatment. We present an extremely rare case of ALT/WDLPS in the buccal mucosa of an elderly Japanese woman. A provisional diagnosis of benign tumor was made from clinical symptoms that involved findings on magnetic resonance imaging. Microscopic examination revealed mature adipocytes with substantial variation in cell size, fewer multivacuolated lipoblasts, and positive stain with MDM-2 and P16, which resulted in a definitive diagnosis of ALT/WDLPS. Conservative surgical resection resulted in locoregional recurrence; however, no evidence of dedifferentiation was detected, and repeat conservative resection finally allowed the patient to avoid esthetic and functional disorder and to obtain an uneventful clinical prognosis over the long-term follow-up period. We also review previous reports of cases of ALT/WDLPS arising from the cheek/buccal mucosa.
