Purpose: To present our experiences and surgical efficacy of the three-dimensional(3D)visualization system in endophthalmitis vitrectomy. Methods: 11 cases of endophthalmitis had undergone NGENUITY® 3D Visualization System assisted23-gauge transconjunctival vitrectomy, by a same surgeon at Xuzhou First People’s Hospital between 1 March 2020 and 31 March 2022. The efficacy of vitrectomy , best-corrected visual acuity (BCVA), intraoperative illumination, and comfort of the surgeon was investigated. Results: The vitrectomy was finished in all cases with lower intensity of intraoperative illumination on the 3D visualizationsystem without any complications attributed to the visualization system, and none of them were abdicated or converted to the conventional microscope. All of the patients were cured after operation combined with antibiotics therapy, consistent with significantly improved BCVA(t = 4.039, p = 0.0024). Conclusion: 3D visualizationsystem with advantages of clearer intraoperation visualization and optimized ergonomic design, is suitable for a variety of vitreoretinal surgery with good safety and efficacy, especially in the complicated and long-lasting endophthalmitis surgeries.

The possibility of using drug loaded bovine serum albumin (BSA) porous films as therapeutic contact lenses is investigated. Kaempferol (KAE), a hydrophobic antioxidant and anti-inflammatory agent, is incorporated into BSA porous films to form BSA/KAE films. The BSA/KAE films are transparent in the visible wavelength range of the human eye, possessing high water content and good cytocompatibility. A prolonged and sustained drug release is observed, and the in vivo efficacy of BSA/KAE films is better than the individual KAE. BSA/KAE films promoted the corneal re-epithelialization, inhibited neovascularization, and reduced the inflammation of an alkali burn induced corneal injury model. The study demonstrates the promising potential of BSA/KAE films as therapeutic contact lenses for the treatment of corneal injury, builds an available ocular drug delivery platform for ocular diseases.

Background To analyze the level changes of 28 cytokines in aqueous humor of patients with proliferative diabetic retinopathy (PDR) coexisting neovascular glaucoma (NVG) after intravitreal injection of conbercept (IVC), and to investigate whether these cytokines are associated with intraoperative bleeding (IOB). Methods Totally 34 eyes with NVG secondary to PDR were enrolled. Patients were randomized into two groups, and all of them underwent 25-gauge pars plana vitrectomy (PPV) combined with trabeculectomy. Group I, 18 eyes received IVC 3 days before PPV, and 100 µL aqueous humor was collected at the time of IVC pretreatment and 3 days later at the beginning of PPV respectively. Group II, 16 eyes received IVC after PPV, and 100 µL aqueous humor was collected only at the beginning of PPV. Aqueous humor from 19 eyes with age-matched cataract patients served as controls. Luminex bead-based multiplex array was used to measure the levels of 28 cytokines in aqueous humor. The baseline cytokine levels were compared among the three groups. All NVG patients were divided into IOB and non-bleeding (INB) groups. The cytokine levels of aqueous humor at the beginning of PPV were compared between group I and II, also between IOB and INB groups. IOB in NVG patients was graded according to vitreous bleeding amount. The correlation between cytokine levels and the grades of IOB were analyzed. Results Compared with controls, the baseline levels of 18 cytokines associated with inflammation and angiogenesis showed significantly increased in group I and group II (all, P P P rs > 0.4, P Conclusions IVC 3 days before PPV combined with trabeculectomy reduces IOB in NVG patients, in which the downregulation of IL-4, Ang-2, PLGF and VEGF-A after IVC may be an underlying mechanism. The lack of kinetic parameters of VOC photochemical conversion especially in the presence of NO and O3, and product information, will result in the incomplete understanding for the occurrence of haze. In the present study, the photochemical conversion of toluene, one of most significant hydrocarbons of VOCs, in toluene, toluene/NO, toluene/O3 and reaction systems was assessed for up to 4 days in a smog chamber at 278 K and 308 K. The results indicated that the addition of NO and O3 promoted the conversion of toluene. The first-order kinetic rate constants of toluene assessed in the toluene reaction system at 278 K and 308 K were 0.12 and 0.19 day−1, respectively. The second-order kinetic rate constants of toluene assessed in the toluene/NO were 0.1 × 10−13 cm3 molecule−1 day−1 (at 278 K) and 0.9 × 10−13 cm3 molecule−1 day−1 (at 308 K), and those in the toluene/O3 reaction systems were 0.06 × 10−12 cm3 molecule−1 day−1 (at 278 K) and 0.11 × 10−12 cm3 molecule−1 day−1 (at 308 K). The small difference of second-order kinetic rate constants between 278 K and 308 K obtained for the toluene/O3 system when compared with the toluene/NO system indicated the reduced reaction sensitivity of O3 to the temperature. Several dozens of large molecule products containing up to O6 heteroatoms were identified by positive ion atmospheric pressure photoionization (+APPI) coupled with Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS), with double bond equivalents up to 18, and carbon numbers ranging within 12–38, respectively. The findings presented herein may provide a new train of thought for the photochemical reaction process of toluene and its conversion in ambient air.

Background Acute myeloid leukemia (AML), which is characterized by the uncontrolled proliferation of myeloid leukemia cells in the bone marrow and other hematopoietic tissues, and is highly heterogeneous. While with the progress of sequencing technology, understanding of the AML-related biomarkers are still incomplete. The purpose of this study is to identify potential biomarkers for diagnosis of AML. Methods Based on WGCNA analysis of gene mutation expression, methylation level distribution, mRNA expression and AML-related genes in public databases, were employed for investigating potential biomarkers for prognosis of AML. Results This study screened a total of 6,383 genes by analyzing various changes in 103 acute myeloid leukemia (AML) samples, including gene mutation expression, methylation level distribution, mRNA expression, and AML-related genes in public databases. Moreover, seven AML-related co-expression modules were mined by WGCNA analysis and twelve biomarkers associated with the AML prognosis were identified from each top 10 genes of the seven co-expression modules. The AML samples were then classified into two subgroups, the prognosis of which is significantly different, based on the expression of these twelve genes. The differentially expr essed genes are mainly involved in glucose metabolism, glutathione biosynthesis, small G protein-mediated signal transduction, and the Rap1 signaling pathway. Conclusions With the utilization of WGCNA mining, seven gene co-expression modules were identified from TCGA database and there are unreported genes that may as potential driven genes of AML and may be the direction to identify the possible molecular signatures to predict survival of AML patients and help guide experiments for potential clinical drag targets.

NixCo1-xFe2Oz composite oxide nanosphere was successfully prepared, to degrade 2-monochlorobiphenly (CB-1) in continuous-flow fixed-bed microreactor at GHSV of 20000 h−1. The five cycles of temperature-dependent run experiments between 150 and 350 °C showed its superior activity with a CB-1 conversion of more than 95% above 300 °C over Ni0.5Co0.5Fe2Oz. Importantly, the sustainable higher reactivity could be observed over prolonged 600 min reaction times after the 5th run test. The degradation products detected as biphenyl and monochlorobenzene with yield ratio of 129, account for 0.24% and 0.0011% of initial CB-1 respectively. This indicated the weak occurrence of hydrodechlorination and breakage of C C bridge bond during the degradation of CB-1. The possibly dominant occurrence of oxidative degradation probably follows Mars–van Krevelen mechanism, resulting in the generation of the formic, acetic, propanoic and butyric acids and so on. Due to the high oxygen mobility over Ni0.5Co0.5Fe2Oz nanosphere, the consumed oxygen species could be compensated rapidly by the gas phase oxygen via O2 → O2− → 2O− → 2O2−. The interaction among different elements in Ni0.5Co0.5Fe2Oz nanosphere confirmed by the derivation of the electron cloud, enhanced the mobility of the reactive oxygen species, which would be beneficial for the oxidation of chlorinated biphenyls.

Flowerlike LiαTiOx micro/nanostructures were successfully synthesized to degrade 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47) at 250–350 °C. The pseudo-first-order kinetics rate constant of the reaction at 300 °C was in the range of 0.034–0.055 min–1. The activation energy was as low as 39.9–48.1 kJ/mol. The excellent performance attained over LiαTiOx was attributed to Li dopant having the electron-donating effect, which enhanced the oxygen species mobility. The oxidative reaction was believed to be the dominant degradation pathway following the Mars–van Krevelen mechanism, being accompanied by the weak hydrodebromination occurrence generating the trace mono- to tri-BDEs. However, the precise mechanism of action of the drug remains to be fully elucidated. Tumor-infiltrating macrophages presented in the tumor microenvironment have been shown to promote development and progression of B-cell lymphomas through crosstalk mediated by secreted cytokines and chemokines. Because Btk has been implicated in Toll-like receptor (TLR) signaling pathways that regulate macrophage activation and production of proinflammatory cytokines, we investigated the immunomodulatory effects of Btk inhibitor on macrophages. Our results demonstrate that Btk inhibition efficiently suppresses production of CXCL12, CXCL13, CCL19, and VEGF by macrophages. Furthermore, attenuated secretion of homeostatic chemokines from Btk inhibitor-treated macrophages significantly compromise adhesion, invasion, and migration of lymphoid malignant cells and even those not driven by Btk expression. The supernatants from Btk inhibitor-treated macrophages also impair the ability of endothelial cells to undergo angiogenic tube formation. Mechanistic analysis revealed that Btk inhibitors treatment downregulates secretion of homeostatic chemokines and cytokines through inactivation of Btk signaling and the downstream transcription factors, NF-κB, STAT3, and AP-1. Taken together, these results suggest that the encouraging therapeutic efficacy of Btk inhibitor may be due to both direct cytotoxic effects on malignant B cells and immunomodulatory effects on macrophages present in the tumor microenvironment. This novel mechanism of action suggests that, in addition to B-cell lymphomas, Btk inhibitor may also have therapeutic value in lymphatic malignancies and solid tumors lacking Btk expression.

Three crystalline forms of Al2O3 featuring rod-like fibers are synthesized, to perform the gaseous degradation of 1-chloronaphthalene (CN-1), a model compound. The degradation efficiency (DE) decreases as follows: micro/nano-γ-Al2O3 > micro/nano-η-Al2O3 > regular α-Al2O3. The high DE of 92.2% attained over γ-Al2O3 is attributed to its specific defective spinel structure, resulting in its large surface area and pore volume, large amounts of surface-adsorbed oxygen species, and Bronsted and Lewis acid sites. The degradation products, naphthalene and 1,4/1,6-dichloronaphthalene (DiCN), with yield ratios of 7.3–13.3, are detected, indicating the occurrence of hydrodechlorination and side chlorination reactions. 1,6-DiCN is believed to be the most favored DiCN isomer product based on the calculated ΔHr and ΔGr. However, the percentage values of intermediates formed via these two reactions, in the degradation of CN-1, are 9.1%, 4.5% and 4.4%, over α-, η- and γ-Al2O3, respectively. The oxidative degradation pathway possibly dominantly occurs especially over micro/nano-γ-Al2O3 with oxidative species such as O2−, O− and O2−, following the Mars–van Krevelen mechanism. Oxidation intermediates containing –CH2–, –CH3, and C–O groups are produced, which can be subsequently oxidized to low-molecular-weight products, such as formic, acetic and propanoic acids, and eventually completely oxidized to CO2.

The degradation of 1,2,3,4-tetrachloronaphthalene (CN-27) featuring a one-side fully-chlorinated aromatic ring, was evaluated over three of the prepared rod-like Fe–Al composite oxides (FeAl-1, FeAl-5 and FeAl-10). The results showed that their reactive activities were in the order of FeAl-5 ≈ FeAl-10 ≫ FeAl-1, which could be attributed to their different pore structural properties and reactive sites caused by the different phase interaction between iron species and the γ-Al2O3. The generation of trichloronaphthalenes (1,2,3-TrCN and 1,2,4-TrCN, i.e. CN-13 and CN-14), dichloronaphthalenes (1,2-DiCN, 1,3-DiCN, 1,4-DiCN and 2,3-DiCN, i.e. CN-3, CN-4, CN-5 and CN-10) and monochloronaphthalenes (1-MoCN and 2-MoCN, i.e. CN-1 and CN-2) suggested the occurrence of successive hydrodechlorination reactions. The amount of CN-14 exceeded that of CN-13 from 71.5% to 77.7% across the three different systems, revealing the preferred occurrence of the first hydrodechlorination step at the β-position. This is dissimilar to the preference at the α-position observed during the dechlorination of octachloronaphthalene (CN-75) over micro/nano Fe3O4. The structural differences between one-side and two-side fully-chlorinated aromatic rings would have a pronounced impact on the reactivity of the chlorine substitution position. The major hydrodechlorination pathway was judged to be CN-27 → CN-14 → CN-4 → CN-2. Additionally, the detected 1,2,3,4,6-pentachloronaphthalene (CN-50) and 1,2,4,6/7-tetrachloronaphthalenes (CN-33/34) suggested the reverse chlorination reaction also happened while the hydrodechlorination reaction was occurring. The C–Cl bond dissociation energies (BDEs) of the parent and daughter polychlorinated naphthalene (PCN) congener were calculated using density functional theory (DFT), to achieve a deeper understanding of a different product yield distribution.

The near-solid wall multi-bubble cavitation is an extremely complex phenomenon, and cavitation has strong erosiveness. The melting point (about 3410 °C) of tungsten is highest among all pure metals, and its hardness is also very high (its yield strength is greater than 1 GPa). What would happen to pure tungsten wire under extreme conditions caused by collapsing cavitation bubbles at high hydrostatic pressure? In this paper, we have studied the fracture process of pure tungsten wire with diameter of 0.2 mm mounted at the focus of a standing acoustic wave produced by a spherical cavity transducer with two open ends placed in a near spherical pressure container, and also studied the macro and micro morphological characteristics of the fracture and the surface damage at different fracture stages of tungsten wire under various hydrostatic pressures and driving electric powers. The results have shown that the fracture time of tungsten wire is inversely proportional to avitation intensity with hydrostatic pressure and driving electric power, the higher the acoustic pressure caused by higher electric power, the shorter the fracture time. The possible fracture mechanisms of tungsten wire in this situation we found mainly contributed to asymmetrically bubbles collapse near the surface of tungsten wire, leading to tearing the surface apart; consequently cracks along the radial and axial directions of a tungsten wire extend simultaneously, classified as trans-granular fracture and inter-granular fracture, respectively. With the increase of cavitation intensity, the cracks tend to extend more radially and the axial crack propagation path becomes shorter, that is, mainly for trans-granular fracture; with the decrease of cavitation intensity, intergranular fracture becomes more obvious. When the hydrostatic pressure was 10 MPa and the driving electric power was 2 kW, the fibers became softener due to the fracture of the tungsten wire. The fracture caused by acoustic cavitation was different from conventional mechanical fracture, such as tensile, shear, fatigue fracture, on macro and micro morphology.

An eco-friendly material, activated carbon cloth (ACC) was used as the heterogeneous catalyst in activation of peroxydisulfate (PDS) for the efficient degradation of organic pollutant in water. Besides, the effects of several parameters in the ACC/PDS process including initial pH, PDS concentration, reaction temperature, stirring speed and co-existing anions were investigated. Under optimum conditions, almost complete removal (98.6%) of AO7 in 60 min and 67.4% of total organic carbon (TOC) removal within 180 min were obtained, accompanied by the remarkable destruction of azo band and naphthalene ring on AO7. The electron paramagnetic resonance and radical quenching experiments were carried out to identify the reactive radicals in the ACC/PDS process. Surface characteristic techniques such as XRD, BET, SEM, FTIR, XPS were applied to analysis the change of crystal structure, surface area, surface morphology, functional groups on the surface of fresh and spent ACC samples. Hydroxyl groups (C‒OH) and π-π transitions significantly affected the catalytic activity of ACC. The intermediate products of AO7 oxidation were identified by LC-MS and the corresponding degradation pathway was proposed.

Background Angioimmunoblastic T cell lymphoma (AITL) is a distinct subtype of peripheral T cell lymphoma and associated with poor outcomes. The activation status of T cell receptor (TCR) signaling has recently become a focus of attention in terms of the therapeutic targets. However, the molecular pathogenesis mechanisms and novel therapeutic targets are largely unknown. Methods Antibodies specific to phosphorylated ZAP70, ITK and PLCγ1 were used to identify the activation status of intracellular proteins involved in TCR signaling in AITL patients. Malignant T cell lymphoma cells were transduced with a lentiviral construct containing ITK shRNA for cellular and functional assays. The antitumor effects of the selective ITK inhibitor BMS-509744 were determined in vitro and in vivo. Results Immunohistochemistry staining showed that more than half of the AITL patients (n = 38) exhibited continuously activated intracellular TCR signaling pathway. Patients positive for phosphorylated ITK showed a lower rate of complete response (20% vs. 75%, P = 0.004) and a shorter progression-free survival (5.17 months vs. 25.1 months, P = 0.022) than patients negative for phosphorylated ITK. Genetic and pharmacological cellular ITK inhibition significantly compromised the proliferation, invasion and migration of malignant T cells. The selective ITK inhibitor BMS-509744 also induced the pro-apoptotic effects and G2/M phase cell cycle arrest in vitro and in vivo. Finally, inhibition of ITK synergistically enhanced the antitumor effect of vincristine and doxorubicin on malignant T cell lymphoma cell lines. Conclusions Our findings suggest that ITK may be a novel candidate therapeutic target for the treatment of patients with ITK-expressing malignant T-cell lymphomas. Objective: The aim of the study was to analyze the clinical characteristics of pediatric patients with ocular toxocariasis. Methods: Ocular toxocariasis was diagnosed and treated in 46 children from Shanghai and surrounding provinces. The diagnosis of ocular toxocariasis was confirmed immunologically by performing an enzyme-linked immunosorbent assay on serum and/or intraocular fluid. All pediatric patients and their guardians completed a questionnaire concerning their cases and living habits. Results: The mean age of onset was 6 ± 3 years. Most children (85%) resided in rural areas, and 91% of the children had contact with adult dogs or puppies. Primary gastric lymphoma (PGL) is the most common extranodal non-Hodgkin lymphoma. However, uniform criteria for treatment and outcome evaluation are lacking. MicroRNA-16 (miRNA-16) has been shown to exhibit tumor-suppressive properties in many cancers and is a good biomarker of prognosis. We detected the expression level of miRNA-16 in 91 patients with PGL by qPCR and analyzed the prognostic significance. The proportion of patients with high expression of miRNA-16 was greater in patients with disease onset at age ≤ 60 years (P = 0.027), early-stage disease (P = 0.06), and marginal zone B-cell lymphoma (P = 0.0031). Furthermore, the proportion of patients with high expression of miRNA-16 was greater in those with a complete response than in those without a complete response (P = 0.054). However, miRNA-16 expression was not a statistically significant predictor of survival. These results suggest that miRNA-16 is a useful biomarker of disease prognosis in patients with PGL.

Background Rituximab combined with cyclophosphamide, doxorubicin, vincristine and prednisone (CHOP) is used as standard frontline regimen for diffuse large B-cell lymphoma (DLBCL). The landscape of somatic mutations in DLBCL revealed that inactivation of EP300 plays an important role in lymphomagenesis. A common EP300 single nucleotide polymorphism (SNP) rs20551 results in the substitution of valine for isoleucine at codon 997 close to the Bromodomain. However, the association between SNP rs20551 and clinical prognosis in DLBCL patients treated with R-CHOP is unknown. Methods In this study we analyzed the EP300 SNP rs20551 and prognosis of 226 DLBCL patients who treated with R-CHOP or R-CHOP-like regimes from 2002 to 2013. Determination of the EP300 SNP rs20551 from genomic DNA was obtained by Sanger chain termination sequencing. Result In this study, the frequency of the A and G allele of the EP300 SNP rs20551 in 226 patients were 92.5 and 7.5%, respectively. We did not observe obvious correlation between patients’ disease features and the EP300 SNP rs20551. But the patients with genotype AA had a higher 5-year overall survival rate than those with genotype GA (77.0% vs. 64.7%, p = 0.045). Furthermore, multivariate Cox regression analysis showed that the GA genotype of EP300 SNP rs20551 was an independent poor prognostic factor for DLBCL patients treated with Rituximab-chemotherapy (p = 0.009, HR 2.956, 95% CI 1.315–6.645). Conclusion This study suggests that EP300 SNP rs20551 might be a useful biomarker to predict the long-term outcome of R-CHOP in DLBCL patients.

Background TP53 Arg72Pro (SNP rs1042522) is associated with risk of non-Hodgkin lymphoma (NHL). Diffuse large B-cell lymphoma (DLBCL) is the most common subtype of NHL. However, the relationship between this SNP and prognosis of DLBCL in Asians is unknown. Methods Genotyping of TP53 Arg72Pro was done in 425 Chinese DLBCL patients. Two hundred and eighty-nine patients were treated with R-CHOP, and 136 patients received CHOP or CHOP-like as frontline regimen. Three hundred and ninety-six patients were assessable for the efficacy. Results Patients with Arg/Arg and Arg/Pro at codon 72 of TP53 had a higher complete response rate (61% vs. 44%, P = 0.007) than those with Pro/Pro. In the subgroup treated with CHOP or CHOP-like therapy, patients with Arg/Arg and Arg/Pro showed a higher 5-year overall survival (OS) rate than those with Pro/Pro (68.8% vs. 23.2%, P = 0.001). Multivariate Cox regression analysis revealed TP53 Arg72 as a favorable prognostic factor in this group. However, the combination of rituximab with CHOP significantly increased the 5-year OS rate of patients with Pro/Pro to 63%. Conclusion This study revealed TP53 Arg72 as a favorable prognostic factor for Chinese DLBCL patients treated with CHOP or CHOP-like as frontline therapy.

In present paper, a novel smoothing function method for nonlinear complementarity problems is proposed. Then the property of the function is discussed. And the conditions of convergences of this iteration algorithm are given, so that the reference conclusions are extended. Theory analysis and primary numerical results illustrate that this method is feasible and effective.

The standard 2-norm support vector machine (SVM for short) is known for its good performance in classification and regression problems. In this paper, the 1-norm support vector machine is considered and a novel smoothing function method for Support Vector Classification(SVC) and Regression (SVR) are proposed in an attempt to overcome some drawbacks of the former methods which are complex, subtle, and sometimes difficult to implement. First, using Karush-Kuhn-Tucker complementary condition in optimization theory, unconstrained non-differentiable optimization model is built. Then the smooth approximation algorithm basing on differentiable function is given. Finally, the paper trains the data sets with standard unconstraint optimization method. This algorithm is fast and insensitive to the initial point. Theory analysis and numerical results illustrate that the smoothing function method for SVMs are feasible and effective.
