Abstract The frequent occurrence of extreme climate events disrupts the regional water budget balance and leads to changes in the dry and wet conditions of the surface, making the water surplus and deficit more complex and variable. To explore the quantitative relationship between the spatiotemporal evolution of dry and wet conditions and meteorological factors in the Hexi Corridor under changing environmental conditions, the relative moisture index (MI) and FAO Penman–Monteith (FAO P–M) model were combined to construct a partial differential quantitative attribution model for dry and wet variations affected by climate factors in the Hexi Corridor. The results show that: (1) MI in the Hexi Corridor increased significantly (Z = 2.341) during 1960–2019, showing a wet-trend change, and the degree of drought increased from southeast to northwest in the Hexi Corridor. (2) The order of drought degree in four seasons is as follows: winter (− 0.95), spring (− 0.93), autumn (− 0.89) and summer (− 0.83). (3) The frequency of extreme drought, severe drought, moderate drought, and mild drought within 60 years of 21 meteorological stations accounted for 28.38%, 50.48%, 8.85%, and 7.38%, respectively, and the frequency above severe drought was the highest. (4) The sensitivity of meteorological factors gradually increased from northwest to southeast, and MI was the most sensitive to the change of precipitation (P), followed by net radiation (R n), wind speed (u 2), mean temperature (T mean), relative humidity (RH) and maximum temperature (T max). MI was the least sensitive to the change of minimum temperature (T min). P is the most important meteorological variable that contributes to the increase of MI, followed by u 2, T mean, and T min. R n, T max, and RH have the least influence, and the total contribution of the seven meteorological factors is 85.59%. Compared with the reference evapotranspiration, P is the main factor affecting the dry and wet variations in Hexi Corridor.

Abstract The sediment content and transport rate of rivers are crucial indicators reflecting soil erosion, water quality, and water resource management in a region. Studying changes in river sediment transport rates within a basin is essential for evaluating water quality, restoring water ecosystems, and implementing soil and water conservation measures. This study focused on the Shule River Basin and utilized various methods such as moving average, cumulative anomaly, Mann–Kendall mutation test, Mann–Kendall (M–K) trend test, Sen’s slope estimation, Correlation analysis, wavelet analysis, R/S analysis, ARCGIS10.7 interpolation, non-uniformity coefficient, and concentration to analyze data from hydrologic stations at Changmapu (CMP), Panjiazhuang (PJZ), and Dangchengwan (DCW). The research examined the temporal and spatial characteristics of sediment transport rates and identified key driving factors. Findings revealed significant increases in annual sediment transport rates at CMP and PJZ by 12.227 and 4.318 kg/s (10a)−1, respectively, while DCW experienced a decrease of 0.677 kg/s (10a)−1. The sediment transport rate of the three stations had a sudden change around 1994. The average annual sediment transport rates displayed distinct cycles, with CMP, PJZ, and DCW showing cycles of 51a, 53a, and 29a respectively. Additionally, while CMP and PJZ exhibited a continuous upward trend in sediment transport rates, DCW showed a consistent decline. The annual average sediment transport rates of CMP, PJZ, and DCW were 1305.43 kg/s, 810.06 kg/s, and 247.80 kg/s, respectively. These research findings contribute to enhancing the comprehension of sediment dynamics in the arid region of northwest China and offer a theoretical basis for the restoration and management of ecological environments in similar areas in the future.

The optimal allocation of water resources is crucial for addressing regional water scarcity, adapting to climate change, and promoting sustainable development. This study focused on the Shule River Basin and considered three climate scenarios (SSP1–2.6, SSP2–4.5, SSP5–8.5), which is combinations of Shared Socioeconomic Pathways (SSPs) and Representative Concentration Pathways (RCPs). A water resources optimal allocation model including water supply and demand prediction, multi-objective optimization, decision-making and analysis on optimal water allocation schemes was constructed. This model aims to analyze the optimal allocation of water resources in the Shule River Basin over three planning years (2025, 2030, 2035), considering guarantee rates of 50 % and 75 % under various climate scenarios. The obtained results indicate that: (1) An increasing trend in water demand in the planning year. For instance, under the SSP1–2.6 scenario, the basin's water demand is projected to reach 1.920×10^9 m3, 2.037×10^9 m3, and 2.103×10^9 m3 in 2025, 2030, and 2035, respectively. (2) The total water supply with a guarantee rate of 50 % (75 %) in the planning years is projected to be 1.939 (1.483)×10^9, 1.957(1.502) ×10^9, and 1.974 (1.519) ×10^9 m3, respectively. (3) In the three scenarios of SSP1–2.6, SSP2–4.5, and SSP5–8.5, the water demand for domestic and ecology is met in each planning year. There is a slight water shortage in the industrial sector, with a shortage rate ranging from 0 % to 13.41 %. The agricultural sector experiences the highest water shortage, with a rate of 1.93–44.77 %. (4) The water supply structure of each optimal allocation scheme of water resources was optimized. The economic benefits mainly depend on the industrial and ecological sectors. These findings offer valuable insights for optimizing the distribution of regional water resources in response to changing climatic conditions.

Background: To evaluate the efficacy and safety of allogenic CD8 + natural killer T (CD8+ NKT) immunotherapy combined with gefitinib in the treatment of advanced or metastatic EGFR mutant non-small cell lung cancer (NSCLC). Methods: This study is prospective. The NSCLC patients with exon 19 (Ex19del) or exon 21 L858R point mutations, and response to gefitinib treatment were enrolled into the trial to be randomly assigned into the gefitinib arm and the gefitinib/NKT arm. Allogenic CD8+ NKT cells were cultured in vitro and adaptive transferred into the patients via vein in the gefitinib/NKT arm. The primary endpoint was progression-free survival (PFS). Secondary endpoint analysis included time to disease progression (TTP), overall survival (OS), levels of serum tumour markers for carcinoembryonic antigen (CEA) and alanine aminotransferase (ALT) in the blood, the response rate and safety. From July 2017 to June 2021, 19 patients were randomly assigned to the gefitinib arm (n = 8) and the gefitinib/NKT arm (n = 11). Results: The estimated median survival PFS in the gefitinib/NKT arm was significantly longer than that of the gefitinib arm (12 months vs 7 months). Similar results were also observed for the median TTP. Moreover, the gefitinib/NKT arm had better CEA control than the gefitinib arm. Clinical grade 3 adverse reactions occurred in 64% and 39% of patients in the gefitinib/NKT arm and the gefitinib arm, respectively. The most common grade 3 adverse events in the gefitinib/NKT arm included abnormal liver function in 8 cases (73%) and diarrhoea in 1 case (9%), both of which resolved after drug intervention. Conclusion: The PFS of EGFR-mutated advanced NSCLC treated with allogenic CD8+ NKT cells combined with gefitinib was longer than that of gefitinib alone. No obvious serious adverse reactions occurred, and the patients compliance and survival status were good.

The synthesis of alternative fuels from biomass has emerged to be a vital strategy to maintain sustainable development. Long-chain levulinate esters (LEs) are a class of good biofuel candidates because they have similar structures to biodiesels and superior heating values. The synthesis of long-chain LEs with effective heterogeneous catalysts has always been challenging. In this work, a few heterogeneous phosphotungstate catalysts were facilely prepared, among which the V3+ exchanged phosphotungstic acid catalyst (VPW) was the most active for the alcoholysis of furfuryl alcohol (FA) and 1-hexanol into 1-hexyl levulinate (HL). A maximum HL yield of 63% was achieved at 180 °C. The VPW catalyst could be reused without obvious decrement of activity. The system was also applicable to the alcoholysis of FA with other hexanols and 1-octanol into various long-chain LEs.

Climate change and human activities exert significant impact on the mechanism of runoff generation and confluence. Comprehending the reasons of runoff change is crucial for the sustainable development of water resources. Taking the Upper Shule River as the research area, the M-K test and the moving t test were used to diagnose the runoff mutation time. Furthermore, the slope changing ratio of cumulative quantity method (SCRCQ), climate elasticity method, and Budyko equation were utilized to quantitatively evaluate the impacts and contribution rates of climate change and human activities. The following results were obtained: (1) The Upper Shule River experienced a significant increase in runoff from 1972 to 2021, with 1998 marking the year of abrupt change. (2) The runoff sensitivity showed a downward trend from 1972 to 2021. The main factor affecting the decrease in runoff sensitivity was the characteristic parameters of underlying surface (n), followed by precipitation (P), while the influence of potential evapotranspiration (ET0) was the weakest. (3) The response of runoff changes to runoff sensitivity and influencing factors were 90.32% and 9.68%, respectively. (4) The results of three attribution methods indicated that climate change was the primary factor causing the alteration of runoff in the Upper Shule River. The research results supplement the hydrological change mechanisms of the Upper Shule River and provide a scientific basis for future water resources management and flood control measures.

Abstract Background To survive harsh environmental conditions, desert plants show various adaptions, such as the evolution of trichomes, which are protective epidermal protrusions. Currently, the morphogenesis and function of trichomes in desert plants are not well understood. Salsola ferganica is an annual halophyte distributed in cold deserts; at the seedling stage, its rod-shaped true leaves are covered with long and thick trichomes and are affected by habitat conditions. Therefore, we evaluated the trichomes on morphogenesis and cell wall composition of S. ferganica compared to Arabidopsis thaliana and cotton, related gene expression, and preliminary function in salt accumulation of the leaves. Results The trichomes of S. ferganica were initiated from the epidermal primordium, followed by two to three rounds of cell division to form a multicellular trichome, while some genes associated with them were positively involved. Cell wall composition analysis showed that different polysaccharides including heavily methyl-esterified and fully de-esterified pectins (before maturation, probably in the primary wall), xyloglucans (in the mid-early and middle stages, probably in the secondary wall), and extensin (during the whole developmental period) were detected, which were different from those found in trichomes of Arabidopsis and cotton. Moreover, trichome development was affected by abiotic stress, and might accumulate salt from the mesophyll cells and secrete outside. Conclusions S. ferganica has multicellular, non-branched trichomes that undergo two to three rounds of cell division and are affected by abiotic stress. They have a unique cell wall composition which is different from that of Arabidopsis and cotton. Furthermore, several genes positively or negatively regulate trichome development. Our findings should contribute to our further understanding of the biogenesis and adaptation of plant accessory structures in desert plant species.

Background: Exposure to particulate matter (PM) has been a major public health threat, but the potentially differential effects on asthma of PM remain largely unknown in high altitude settings. We evaluated the effects of ambient PM on asthma in high altitude settings. Methods: The study recruited a representative sample from high altitude settings using a multistage stratified sampling procedure. Asthma was defined by a self-reported history of diagnosis by a physician or by wheezing symptoms in the preceding 12 months. The annual mean PM2.5 and PM10 concentrations were calculated for each grid cell at 1-km spatial resolution based on the geographical coordinates. Results: We analyzed data for participants (mean age 39.1 years, 51.4% female) and 183 (3.7%, 95% confidence interval (CI): 3.2–4.2) of the participants had asthma. Prevalence was higher in women (4.3%, 95% CI 3.5–5.1) than in men (3.1%, 2.4–3.8) and increasing with higher concentration of PM exposures. For an interquartile range (IQR) difference (8.77 μg/m3) in PM2.5 exposure, the adjusted odds ratio (OR) was 1.64 (95% CI 1.46–1.83, P

ABSTRACT: Flexible photodetectors (PDs) comprised of low-dimensional organic-inorganic hybrid perovskites with perovskite quantum dots are expected to be the next generation wearable optoelectronic devices. A flexible Vis-NIR PD which contains 2D Dion-Jacobson (DJ) perovskite (4AMP)(MA)2Pb3I10 (4AMP = 4-(aminomethyl)piperidinium, MA = methylammonium) (n3) and micro concentration of CsPbI3 perovskite quantum dots (QDs) layered heterostructures was designed and synthesized in the current work. Controlled by the optimal concentration of QDs, the device response under 660 nm light was increased to 615%. The device combination as per mass of QDs exhibited strong photosensitivity and high-power output. The band gap between the two is minimal, which formed a matching structure and lowered the energy barrier of carrier transport process. QDs layer filled the gap of perovskite film, forming an almost defect-free heterostructure. QDs layer isolated water and passivated the perovskite layer, which therefore contributed to the high-performance of optoelectronic devices. Under the optimal concentration of QDs with up to 5000 bending cycles and different bending angles, the degradation of PDscouldbe ignored, and the devices tended to show a self-healing phenomenon with increasing bending cycles. The optimized strategy will be conducive to developing flexible, wearable, high-performance and low-cost PDs.

The knowledge units of gear transmission scheme digital design are established，and the motion，dynamic and structural attributes of the knowledge units are extracted. From the state space theory of mechanical motion scheme design，the motion and dynamic attributes of the knowledge units are transformed into feature vectors，so that the solution of gear transmission scheme is converted into a matrix calculation. The relationship between dynamic attributes of knowledge units and 3D parametric model is established to realize the visualization of transmission scheme. The knowledge unit database of the gear transmission scheme is established and transformed into the corresponding data structure. The solution of hybrid combination schemes of knowledge units is implemented by considering the series-parallel combination of knowledge units. The gear transmission scheme design module，which is developed by VS，is used to output the three-dimensional model of the gear transmission scheme meeting the user's requirements and the relevant details of the scheme.

Variations in runoff and sediment discharge are important characteristic variables for revealing the coupled effects of climate change (including both the natural variability of climate and anthropogenic climate change) and human activities (including soil and water conservation measures, land use changes, and hydraulic engineering construction). Based on the meteorological data from 19 meteorological stations and the hydrological data from the watershed control station of Hongqi Station, the temporal and spatial evolution of runoff and sediment discharge and the water–sand relationship were analyzed, and the response mechanisms of runoff and sediment discharge changes were clarified using Mikhail Budyko’s theory and other qualitative and quantitative methods. The results determined that: (1) The runoff and sediment discharge showed significant downward trends, with linear change rates of −0.28 × 108 m3/a and −46.10 × 104 t/a, respectively. The change points of the runoff and sediment discharge occurred in 1987 and 1996, respectively. (2) The spatial distribution of water and sediment was different, and the upper and middle reaches produced water, while the downstream produced sediment. (3) Comparing potential evapotranspiration and rainfall based on Budyko theory and the regression relationship, runoff is more closely related to rainfall, and runoff changes are more affected by it. The change in sediment discharge is most closely related to sediment concentration, followed by rainfall and potential evaporation. (4) The contribution rates of runoff and sediment discharge changes influenced by climate change were 24% and 3%, respectively, and the contribution rates by human activities were 76% and 97%, respectively. Human activities, including soil and water conservation measures, land use changes, and hydraulic engineering construction were the main influencing factors, and the impacts of human activities increased from 1960 to 2019. The research results are of great significance for erosion control and ecological restoration in the Tao River Basin under the conditions of the changing environment.

To explore the mechanisms driving variation in ET0 (reference evapotranspiration) in an arid inland region of Northwest China, daily meteorological data from 1960 to 2019 from 19 meteorological stations in the Taohe River basin and its surrounding areas were used to analyze the temporal and spatial distributions of ET0 and meteorological variables. Various qualitative and quantitative analysis methods were used to reveal the correlation between ET0 and meteorological variables. The degree of sensitivity of ET0 variations to meteorological variables and the contribution from each meteorological variable were clarified, and the mechanisms driving variation in ET0 were fully revealed. These are the results: (1) ET0 in the Taohe River basin presented a significant upward trend with a linear change rate of 0.93 mm/a, and a sudden change occurred in 1994. The spatial variation in ET0 ranged from 779.8 to 927.6 mm/a, with low values in the upper and middle reaches and high values in the lower reaches. The ET0 at 14 stations (73.68% of the total) was significantly increased (p < 0.05), and that at 5 stations (26.32% of the total) was not significantly increased (p > 0.05). (2) RH, Rn, and u2 did not change significantly, while Tmax and Tmin showed a significant upward trend. (3) Rn is a meteorological variable closely related to variations in ET0, and is the most sensitive variable for variations in ET0, followed by Tmax and u2. (4) Tmax is the meteorological variable that contributes most to the variation in ET0 (30.98%), followed by Tmin (29.11%), u2 (6.57%), Rn (2.22%), and RH (0.05%). The research results provide a scientific basis for the rational and efficient utilization of water resources and the maintenance of ecosystem health.

Abstract Background Epidermal growth factor receptor tyrosine kinase inhibitors (EGFR-TKIs), such as gefitinib, have achieved good efficacy in EGFR mutation-positive non-small-cell lung cancer (NSCLC) patients, but eventual drug resistance is inevitable. Thus, new TKI-based combination therapies should be urgently explored to extend the overall survival time of these patients. CD8 + CD56+ natural killer T (NKT) cells are a natural and unique subset of lymphocytes in humans that present characteristics of T and NK cells and exert cytotoxicity on tumour cells in a granzyme B-dependent manner. The aim of this trial was to explore the efficacy and safety of CD8 + CD56+ NKT cell immunotherapy combined with gefitinib in patients with advanced EGFR-mutated NSCLC. Methods The study was designed as a prospective, randomized, controlled, open-label, phase I/II trial that includes 30 patients with EGFR mutation-positive stage III/IV NSCLC. All patients will be randomized in blocks at a 1:1 ratio and treated with gefitinib 250 mg/day monotherapy or combination therapy with allogeneic CD8 + CD56+ NKT cell infusions twice per month for 12 cycles or until disease progression occurs. The effectiveness of this treatment will be evaluated based on by progression-free survival (PFS), the time to progression (TTP), overall response rate (ORR), disease control rate (DCR) and overall survival (OS). The safety of the trail is being assessed based on adverse events (AEs). Recruitment and data collection, which started in December 2017, are ongoing. Discussion Although immunotherapy, including programmed death-1/programmed death-1 ligand (PD-1/PD-L1) immunotherapy, has been used for NSCLC treatment with or without EGFR-TKIs, its clear efficacy still has not been shown. Assessing the safety and therapeutic potential of allogeneic CD8 + CD56+ NKT killer cells in combination with EGFR-TKIs in NSCLC will be of great interest. Trial registration This trial (Phase I/II Trails of NKT Cell in Combination With Gefitinib For Non Small Cell Lung Cancer) was registered on 21 November 2017 with www.chictr.org.cn , ChiCTR-IIR-17013471 .

Background: Chronic obstructive pulmonary disease (COPD) is a public health challenge globally. The burden of COPD is high in never-smokers but little is known about its causes. We aimed to find the prevalence and correlates of COPD in never-smokers, with a special focus on solid fuel exposure.Methods: We conducted a cross-sectional study in Western China. COPD was defined by FEV1/FVC < lower limits of normal (LLN). Descriptive statistics and multivariable logistic regression were used for analyses.Results: Six thousand two hundred and seventy one patients were enrolled between June 2015 and August 2016. The prevalence of COPD in never-smokers was 15.0% (95% confidence interval 14.1–15.9). The common independent predictors of COPD in never-smokers included age ≥60 years, exposure to solid fuel, living in a rural area and a history of tuberculosis. Participants with solid fuel exposure were 69% more likely to have COPD (adjusted odds ratio 1.69, 95% CI 1.41–2.04) than those without such exposure. In addition, we found a positive association between small airway dysfunction and solid fuel exposure (OR 1.35, 95% CI 1.18–1.53).Conclusions: This study confirmed the substantial burden of COPD among never-smokers and also defined the risk factors for COPD in never-smokers. Furthermore, we found a positive association between solid fuel exposure and COPD or small airway dysfunction.

In this paper, we report the study of the process of fabricating a multi-layer metal micro-structure using UV-LIGA overlay technology, including mask fabrication, substrate treatment, and UV-LIGA overlay processes. To solve the process problems in the masking procedure, the swelling problem of the first layer of SU-8 thick photoresist was studied experimentally. The 5 μm line-width compensation and closed 20 μm and 30 μm isolation strips were designed and fabricated around the micro-structure pattern. The pore problem in the Ni micro-electroforming layer was analyzed and the electroforming parameters were improved. The pH value of the electroforming solution should be controlled between 3.8 and 4.4 and the current density should be below 3 A/dm2. To solve the problems of high inner stress and incomplete development of the micro-cylinder hole array with a diameter of 30 μm, the lithography process was optimized. The pre-baking temperature was increased via gradient heating and rose every 5 °C from 65 °C to 85 °C and then remained at 85 °C for 50 min – 1 h. In addition, the full contact exposure was used. Finally, a multi-layer metal micro-structure with high precision and good quality of micro-electroforming layer was fabricated using UV-LIGA overlay technology. Abstract Background Pectobacterium spp. are necrotrophic bacterial plant pathogens of the family Pectobacteriaceae, responsible for a wide spectrum of diseases of important crops and ornamental plants including soft rot, blackleg, and stem wilt. P. carotovorum is a genetically heterogeneous species consisting of three valid subspecies, P. carotovorum subsp. brasiliense (Pcb), P. carotovorum subsp. carotovorum (Pcc), and P. carotovorum subsp. odoriferum (Pco). Results Thirty-two P. carotovorum strains had their whole genomes sequenced, including the first complete genome of Pco and another circular genome of Pcb, as well as the high-coverage genome sequences for 30 additional strains covering Pcc, Pcb, and Pco. In combination with 52 other publicly available genome sequences, the comparative genomics study of P. carotovorum and other four closely related species P. polaris, P. parmentieri, P. atrosepticum, and Candidatus P. maceratum was conducted focusing on CRISPR-Cas defense systems and pathogenicity determinants. Our analysis identified two CRISPR-Cas types (I-F and I-E) in Pectobacterium, as well as another I-C type in Dickeya that is not found in Pectobacterium. The core pathogenicity factors (e.g., plant cell wall-degrading enzymes) were highly conserved, whereas some factors (e.g., flagellin, siderophores, polysaccharides, protein secretion systems, and regulatory factors) were varied among these species and/or subspecies. Notably, a novel type of T6SS as well as the sorbitol metabolizing srl operon was identified to be specific to Pco in Pectobacterium. Conclusions This study not only advances the available knowledge about the genetic differentiation of individual subspecies of P. carotovorum, but also delineates the general genetic features of P. carotovorum by comparison with its four closely related species, thereby substantially enriching the extent of information now available for functional genomic investigations about Pectobacterium.

Aim of Study: Four hundred million people live at high altitude worldwide. Prevalence and risk factors for COPD in these populations are poorly documented. We examined the prevalence and risk factors for COPD in residents living at an altitude of 2,100–4,700 m.Methods: We performed a cross-sectional survey in Xinjiang and Tibet autonomous region. A multistage stratified sampling procedure was used to select a representative population aged 15 years or older from eight high altitude regions. All participants underwent pre- and post-bronchodilator measurement of forced expiratory volumes. COPD was diagnosed according to 2019 Global Initiative for Chronic Obstructive Lung Disease (GOLD) criteria.Results: Between June, 2015 and August 2016, 4,967 subjects were included. Median age was 38.0 years (range: 15–91 years; inter-quartile range: 28–49 years); 51.4% participants were female. Overall prevalence of spirometry-defined COPD was 8.2% (95% CI 7.4–8.9%): 9.3% in male (95% CI 8.2–10.4%), and 7.1% in female (95% CI 6.1–8.2%). By multivariable logistic regression analysis, COPD was significantly associated with being aged ≥40 years (odds ratio: 2.25 [95% CI 1.72–2.95], P < 0.0001), exposure to household air pollution (OR: 1.34 [95% CI 1.01–1.79], P = 0.043), and a history of tuberculosis (OR: 1.79 [95% CI 1.23–2.61], P = 0.030), while living at a higher altitude (OR: 0.45 [95% CI 0.33–0.61], P < 0.0001) and having a higher educational level (OR: 0.64 [95% CI 0.43–0.95], P = 0.025) were associated with a lower prevalence of COPD.Conclusions: Our results show that the spirometry-defined COPD is a considerable health problem for residents living at high altitudes and COPD prevalence was inversely correlated with altitude. Preventing exposure to household air pollution and reducing the incidence of tuberculosis should be public health priorities for high altitude residents.

Variations of photosynthetic structures in different tissues or cells are in coordination with changes in various aspects, e.g. physiology, biochemistry, gene expression, etc. Most C4 plant species undergo developmental enhancement of the photosynthetic system, which may present different modes of changes between anatomy and physiology/biochemistry. In the current study, we investigated a Kranz-type C4 species Salsola ferganica with the progressive development of photosynthetic (PS) structure, performance of PS physiology, induction of PS enzymes, and transcriptional and translational regulation of PS genes, results revealed that S. ferganica presented C3 type anatomy in cotyledons but C4 type in leaves (C3/L4), with the C4 system separation of initial carbon fixation in the palisade mesophyll (M) cells and the following incorporation into triosephosphates and sugars in the bundle sheath (BS) cells, respectively. The BS cells continuously surrounded the vascular bundles and water storage cells in leaf anatomic structure. Compared to the single-cell C4 species Suaeda aralocaspica, S. ferganica exhibited similar developmental enhancement of C4 syndrome temporally and spatially in anatomic structures, enzyme activities, and gene expression, which suggests that completion of differentiation of the photosynthetic system is necessary for a C4 assimilation pathway. Besides, S. ferganica also displayed some different characteristics compared to S. aralocaspica in photosynthetic physiology, e.g. a more flexible δ13C value, much lower phosphoenolpyruvate carboxylase (PEPC) activity, and an insensitive response to stimuli, etc., which were not typical C4 characteristics. We speculate that this may suggest a different status of these two species in the evolutionary process of the photosynthesis pathway. Our findings will contribute to further understanding of the diversity of photosynthesis systems in Kranz-type C4 species and the Salsola genus.

Diabetic nephropathy (DN) is characterized by inflammatory responses and extracellular matrix (ECM) accumulation. Astilbin is an active natural compound and possesses anti-inflammatory activity. The aim of this study was to evaluate the anti-inflammatory effect of astilbin on high glucose (HG)-induced glomerular mesangial cells and the potential mechanisms. The results showed that HG induced cell proliferation of HBZY-1 cells in a time-dependent manner, and astilbin inhibited HG-induced cell proliferation. The expression and secretion of inflammatory cytokines, including interleukin-6 (IL-6) and tumor necrosis factor alpha (TNF-α), and ECM components, including collagen IV (Col IV) and fibronectin (FN), were induced by HG. Moreover, TGF-β1 and CTGF were also induced by HG. The induction by HG on inflammatory response and ECM accumulation was inhibited after astilbin treatment. Astilbin treatment also attenuated HG-induced decrease in expression of matrix metalloproteinase (MMP)-2 and MMP-9. The TLR4/MyD88/NF-κB pathway was activated by HG, and the inhibitor of TLR4 exhibited the same effect to astilbin on reversing the induction of HG. TLR4 overexpression attenuated the effect of astilbin on HG-induced inflammatory cytokine production and ECM accumulation. The results suggested that astilbin attenuated inflammation and ECM accumulation in HG-induced rat glomerular mesangial cells via inhibiting the TLR4/MyD88/NF-κB pathway. This work provided evidence that astilbin can be considered as a potential candidate for DN therapy.

Seed heteromorphism is a common characteristic of halophyte and an adaptation to the spatial and temporal variations of natural habitats. Differences in dormancy and germination requirements have been documented in heteromorphic seeds of many species, but the mechanisms for maintenance between different status in various populations have not been well-understood. Salsola ferganica is a typical annual halophyte in Chenopodiaceae distributed in cold desert, in the present study, we found that it could produce three distinct types of seed according to the shape and size of winged perianth (WP), which differed in dispersal ability, dormancy and germination behaviors. Our further investigation revealed that light could significantly promote germination of heteromorphic seeds of S. ferganica, and WP inhibited while GA3 enhanced germination, which suggests that S. ferganica seeds possessed a photo-sensitive combined with morphological and non-deep physiological dormancy type, in which light was the dominant factor. Not like other desert plant species, the germinability of heteromorphic seeds of S. ferganica could not sustain for long (only 1–2 years), especially the small seeds, and was affected by storage time, temperature, salinity, even the environmental conditions of the maternal plant. Thus, the differences of characteristics existed among heteromorphic seeds and variations of heteromorphic ratio among different calendar years were presumed as diverse adaptation strategies integrated in the individual mother plant, and might apply important ecological significance for successful reproduction of the species in the unpredictable cold desert.
