Abstract

Background: Only scattered information is available on the tillage and nutrient management information for the sesame crop following rice in the literature. Sesame as an edible oil yielding crop with high levels of unsaturated fatty acids has high international demand due to superior health benefits. Being a small seeded crop, it requires standard tillage and nutrient management to obtain optimum productivity under rice fallow ecologies. As a sequential crop after rice harvest, the tillage and nutrient management practices followed for the preceding rice have astounding effects on the succeeding sesame crop. To better understand and manipulate the agro ecology in the rice fallow culture, it is necessary to study the behaviour of sesame cultivars, in relation to the tillage requirements and macro nutrient factors that have a bearing on the productivity. Methods: The aim of this work was to evaluate the productivity of rice fallow sesame in the southern plateau and hills regions of the Indian sub-continent (Tamil Nadu) with a hypothesis that tillage and nutrient management would immensely benefit the sesame crop. Field experiments were conducted at TNAU, Tamil Nadu Rice Research Institute, Aduturai, Tamil Nadu during 2019–2020 and 2020–2021 with tillage practices (reduced tillage, conventional tillage and zero tillage) and fertilizer doses (zero percent RDF, 25% RDF, 50% RDF, 75% RDF and 100% RDF) in a split plot design replicated thrice. Results: The results have clearly indicated that the performance of rice fallow sesame was poor under zero till conditions as the sesame crop is poorly adapted leading to a yield penalty up to 68%. A total of 75% RDF has yielded statistically similar yield to that of 100% RDF to the rice fallow sesame. 36. Host defense responses during powdery mildew (Golovinomyces latisporuscomb. nov.) infection in sunflower (Helianthus annuusL.)

Abstract

The infection by a plant pathogen results in the production of the reactive oxygen intermediates (ROIs) in the plant. ROIs threshold levels determine the fate of the resistance in the host; higher than the threshold levels result in the appearance of the resistant or susceptible reaction on the infected plants. ROIs below the threshold levels have been shown to be important in eliciting disease resistance-related signaling pathways that result in the production of antimicrobial compounds, pathogenesis-related proteins, cell wall cross-linking. Golovinomyces latisporusis a very important biotrophic pathogen on sunflower causing powdery mildew disease, which causes serious yield losses. A set of genotypes confirmed earlier to be susceptible (PS 2023 and Morden) and resistant (TX 16R and ID-25) were analyzed for disease progression after the pathogen infection and the development of oxidative burst. In comparison, susceptible genotypes exhibited higher levels of ROIs than resistant genotypes, thus indicating the importance of these molecules in the infection process. The higher levels of ROIs in susceptible genotypes lead to cell damage. Concomitantly, the anti-oxidant enzyme activities are significantly higher in the resistant genotypes than in the susceptible genotypes. The resistant genotypes displayed higher levels of transcripts of hsr203J, which has proven roles in cell death, and NPR1, the master regulator of the salicylic acid-based resistance signaling that acts in plants resistance against biotrophic pathogens.

Abstract

A field experiment was conducted to investigate the effect of deficit moisture stress on eleven sesame genotypes viz., IC-132171, IC-132186, IC-204445, IC-131500, IC-132207, IC-205471, IC-203962, IC-205353, IC-96229, and IC-204966 along with a national check (GT-10). The deficit moisture stress (WS) was imposed for a period of 50–55 days from flowering to physiological maturity. The morphological, physiological, and yield traits under WS significantly decreased in comparison with irrigated conditions (WW). Based on the mean trait performance studied among the genotypes, IC-204966 was found significantly superior with respect to plant height, capsules number, seed weight, total dry matter, leaf area, relative water content and reduced canopy temperature under WS conditions. The correlation analysis of different traits with seed yield indicated that the plant height and number of capsules/plant were highly and positively correlated with seed yield under both WW and WS conditions. The principal component analysis (PCA) revealed that among the traits studied, number of capsules/ plant, total dry matter, stomatal conductance, transpiration rate and fatty acid i.e., palmitic acid were the most important traits that accounted for more than half of the total variation among the sesame genotypes studied. Furthermore, the scatter plot revealed that two genotypes, IC 132,207 and IC 205,471 were stable performers under both stress and irrigated conditions, whereas the performance of check GT-10 varied depending upon conditions.

Abstract

Ascorbate peroxidase (EC 1.11.1.11; APX) was purified from ripe ber ( Ziziphus mauritiana L.) fruits var. Illaichi using conventional techniques of ammonium sulphate fractionation, gel filtration through Sephadex G-100 and ion-exchange chromatography on DEAE-cellulose. The enzyme was purified about 47.4 fold with 34.6% recovery. The molecular weight as determined by gel filtration was found to be 58.08 kDa. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) yielded a single major protein band with molecular weight of 29.79 kDa indicating that the enzyme was a homodimer. Native PAGE revealed a single prominent band suggesting that enzyme was purified to near homogeneity. The optimum pH for APX was found to be 7.8. It exhibited the Michaelis-Menten kinetics with Km values for ascorbate and H 2 O 2 of 1.82 and 2.85 mM, respectively. Mn 2+ , NO 3 - , SO 4 2- and Co 2+ were found to be potent inhibitors of APX while K + , Na + , Ca 2+ and Cl - stimulated the enzyme activity. Diethylpyrocarbonate (DEPC), dithiothreitol (DTT), NaBH 4 and mercaptoethanol inhibited the enzyme activity while iodoacetate and 5, 5’-dithiobis-2-nitrobenzene (DTNB) had no inhibitory effect. Based on the inhibition studies, histidine and tryptophan have been suggested to be present at the active site. Keywords. 42. Defense responses to Fusarium oxysporumf. sp. riciniinfection in castor (Ricinus communisL.) cultivars

Abstract

The biophysical and biochemical basis of resistance in castor to wilt caused by Fusarium oxysporumf. sp. riciniwas studied. Entry and colonization of fungal mycelium in susceptible (JI-35) and resistant (48-1) cultivars was studied histopathologically by employing bright field, scanning and transmission electron microscopy. Browning of xylem vessels of vascular bundles in susceptible cultivar can be taken as an anatomical parameter for early detection of the wilt disease. Electron microscopy revealed more mycelium and spores in the susceptible cultivar. Cell wall was thick in resistant cultivar than the susceptible cultivar. Biochemical activity of antioxidant enzymes and pathogenesis related (PR) proteins in the roots of resistant and susceptible genotypes of castor were studied at 24, 48, 72 and 96 h after inoculation till 7 days. Studies pertaining to the activity of defense enzymes viz, superoxide dismutase (SOD), peroxidase (POX), catalase (CAT), ascorbate peroxidase (APX), glutathione reductase (GR) and β-1,3-glucanase revealed that the level of defense related enzyme activity of SOD, GR and β-1,3-glucanase was higher in resistant cultivar. However, the activity of CAT reached a maximum at 4 days post inoculation and decreased by 7 days. In contrast, the activity of APX enzyme was higher in susceptible cultivar. Release of these enzymes related to expression of resistant mechanism restricted the browning of xylem vessels in resistant cultivar. The restricted growth of mycelium, absence of browning in xylem vessels and increase in activity of defense related enzymes in resistant cultivar indicates the resistance mechanism in the plant.

Abstract

Two ber varieties differing in their shelf lives (Umran, 8 to 9 and Illaichi, 4 to 5 days) were analyzed for cell wall components, cell wall degrading enzymes and their isoenzyme profile at immature green, mature green, turning colour, ripe and overripe stages of ripening. Cellulose and pectin contents decreased during ripening in both varieties. This decrease was accompanied by a corresponding increase in pectin methylesterase, polygalacturonase and cellulase. The increase in the activity of pectin methylesterase was about 20- and 10-fold, that of polygalacturonase about 8.4- and 5.7-fold, and of cellulase 5.5- and 4.4-fold in Umran and Illaichi, respectively. The basal level of activities of all these enzymes was higher at all the stages of ripening in Illaichi variety, having short shelf life, as compared to Umran, with long shelf life. This was further confirmed by the intensity of isoenzyme bands of these enzymes. The isoenzyme profile also revealed that two isoenzymes of each, pectin methylesterase and polygalacturonase, and one of cellulase could be considered as isoenzymes responsible for the softening of cell wall during ripening and could be potential targets for manipulation to delay softening during ripening. Abstract

The present investigations were undertaken to ascertain the extent of lipid peroxidation, oxidative stress and antioxidative system during ripening of two varieties of ber fruits (Ziziphus mauritiana Lam.) differing in their shelf lives, viz. Umran (8-9 days) and Kaithali (4-5 days). Based on visual observations, fruits were categorized as immature green (IG), mature green (MG), turning colour (TC), ripe (R) and overripe (OR). Lipoxygenase (LOX) activity, malondialdehyde (MDA) value and H2O2 content increased significantly during ripening in both varieties, obtaining 3-, 3- and 2.5-fold increase in Umran and 4-, 3.5- and 3-fold increase in Kaithali, respectively. Kaithali being short-lived variety, exhibited higher values of these parameters at almost all the stages of fruit ripening. Activity of superoxide dismutase (SOD), glutathione reductase (GR) and ascorbate peroxidase (APX) increased during initial stages but thereafter decreased, whereas that of peroxidase (POX) decreased continuously during ripening. On the other hand, catalase (CAT) activity increased continuously as ripening progressed. In general, antioxidative enzymes exhibited comparatively higher activity in Umran. Total ascorbic acid content increased from IG to TC stage, while total glutathione increased up to MG stage and thereafter decreased in both varieties. Kaithali had higher content of ascorbate than Umran. β-Carotene and α-tocopherol also decreased as ripening progressed. However, the decrease was more pronounced in Umran as compared to Kaithali.
