4. What's Race Got to do with it? Predicting Youth Depression Across Racial Groups Using Machine and Deep Learning

Abstract

Depression is a common yet serious mental disorder that affects millions of U.S. high schoolers every year. Still, accurate diagnosis and early detection remain significant challenges. In the field of public health, research shows that neural networks produce promising results in identifying other diseases such as cancer and HIV. This study proposes a similar approach, utilizing machine learning (ML) and artificial neural network (ANN) models to classify depression in a student. Additionally, the study highlights the differences in relevant factors for race subgroups and advocates the need for more extensive and diverse datasets. The models train on nationwide Youth Risk Behavior Surveillance System (YRBSS) survey data, in which the most relevant factors of depression are found with statistical analysis. The survey data is a structured dataset with 15000 entries including three race subsets each consisting of 900 entries. For classification, the research problem is modeled as a supervised learning binary classification problem. Factors relevant to depression for different racial subgroups are also identified. The ML and ANN models are trained on the entire dataset followed by different race subsets to classify whether an individual has depression. The ANN model achieves the highest F1 score of 82.90% while the best-performing machine learning model, support vector machines (SVM), achieves a score of 81.90%. 17. Exploring α, β-unsaturated carbonyl compounds against bacterial efflux pumps viacomputational approach

Abstract

AbstractAntibiotic resistance has become a pressing global health crisis, with bacterial infections increasingly difficult to treat due to the emergence of multidrug resistance. This study aims to identify potential chalcone molecules that interact with two key multidrug efflux pumps, AcrB and EmrD, of Escherichia coli, using advanced computational tools. In silicoADMET (absorption, distribution, metabolism, excretion, and toxicity), drug-likeness prediction, molecular docking, and molecular dynamics simulation analyses were conducted on a ligand library comprising 100 chalcone compounds against AcrB (PDB: 4DX5) and EmrD (PDB: 2GFP). The results demonstrated that Elastichalcone A (PubChem CID 102103730) exhibited a remarkable binding affinity of −9.9 kcal/mol against AcrB, while 4'-methoxy-4-hydroxychalcone (PubChem CID 5927890) displayed a binding affinity of −9.8 kcal/mol against EmrD. Both ligands satisfied drug-likeness rules and possessed favorable pharmacokinetic profiles. Molecular dynamics simulation of the AcrB-Elastichalcone A complex remained stable over 100 ns, with minimal fluctuations in root-mean-square deviation and root-mean-square fluctuation. The screened ligand library demonstrated good drug-likeness and pharmacokinetic properties. Moreover, the MM/PB(GB)SA calculation indicated the tight binding and thermodynamic stability of the simulated protein-ligand complexes. Overall, this study highlights the potential of chalcones as promising candidates for targeting multidrug efflux pumps, offering a potential strategy to overcome antibiotic resistance. Further exploration and optimization of these compounds may lead to the development of effective therapeutics against multidrug-resistant bacterial infections.Communicated by Ramaswamy H. Sarma

Abstract

Background: Peri-operative shivering remains a common issue often encounteredduring surgery. Various pharmacological and nonpharmacological methods are employed to control shivering. Nalbuphine and tramadol are shown to possess potent anti-shivering effects. Hence the present study was undertaken to compare the effect of Inj. Nalbuphine Hydrochloride and Inj. Tramadol Hydrochloride for treatment of perioperative shivering in regional anaesthesia. Method: Total 90 patients of either sex, aged 18-65years, ASA I/II, who scheduled for elective surgeries under regional anaesthesia were randomly divided into three groups of 30 patients in each. Group N received 0.08µg/kg Nalbuphine diluted in 5 ml of 0.9% normal saline, Group T received 0.5 mg/kg Tramadol diluted in 5 ml of 0.9% normal saline and Group C (Controlled) received 5ml of 0.9% normal saline. Results: In group T, mean score according to Filos sedation was found to be 1.10±0.31, and in group N it was 1.10±0.31 whilein controls it was 1.10±0.30, (p>0.05). Tramadol shows lesser cessation time of shivering (3.73±1.43min) as compared to nalbuphine (4.08±1.28 min), though the difference was not statistically significant, (p=0.36). Theinsignificant difference found between the response intramadol and nalbuphine groups, (p=0.36) but significant difference was observed between the response of normal saline and tramadol (p<0.0001) and between the response of normal saline and nalbuphine (p<0.0001). The groups were found to be matched regarding recurrence (p=0.120), hypotension(p=0.200), nausea (p=0.133), vomiting (p=0.133) and pruritis (p=0.364). Conclusion: Tramadol administration in dose of 0.5mg/kg or Nalbuphine administration in dose of 0.08mg/kg is recommended to counteract perioperative shivering in subjects undergoing surgery under regional anaesthesia with central neuraxial blockade. Abstract

Background: Hyperlipidemia plays a major role in the pathogenesis of cardiovascular diseases (CVD). Statins are considered to be the most efficacious drugs in the management of hyperlipidemia and this class of drugs is most commonly prescribed for the treatment of hyperlipidemias. Although statin therapy has transformed the management of hyperlipidemia, it is associated with the skeletal muscle, neurological, and metabolic adverse effects. This study was conducted to evaluate the adverse effects of statin therapy in a geriatric population which may help in understanding whether these effects are dose-dependent. Methods: The study was conducted on 200 patients receiving statin therapy (atorvastatin and rosuvastatin) for hyperlipidemias. They were divided into four groups depending on the prescribed dose of atorvastatin and rosuvastatin. All study subjects were followed up for 6 months. The adverse effects reported by them during the statin therapy were documented and analyzed. Results: All patients reported adverse effects after the initiation of statin therapy. Headache and muscle symptoms were among the most commonly reported adverse effects. There was no serious adverse effect (SAE). None of the adverse effects led to the discontinuation of the statin therapy. Conclusion: The results of this study suggest that all patients receiving statin therapy experience one or more adverse effects during the therapy.
