The aim of this study was to prepare and characterize folic acid conjugated nanostructure lipid carriers (NLCs) as nanocarriers for parenteral delivery of gemcitabine (GEM) and paclitaxel (PTX). Folic acid-conjugated GEM-PTX-NLCs were prepared by solvent emulsion-evaporation method and characterized for particle size, polydispersity index and zeta potential which were found to be 134.62±4.11 nm, 0.189±0.05, +28.4±0.6 mV, respectively. Percent entrapment efficiency of GEM and PTX in FA-conjugated GEM-PTX- NLCs were recorded as 89.1±0.6 % and 86.6±0.8 % respectively, which was within an acceptable range for injectable formulation. In vitro drug release study of optimized formulation was carried out using the dialysis tube method. FA-conjugated GEM-PTX-loaded NLCs showed cumulative drug release 78.6±0.93 % and 75.3±0.7 % of GEM and PTX respectively in 72 h in PBS pH 7.4: methanol (7:3) while FA-conjugated GEM-PTX- NLCs showed cumulative drug release 85.5±0.075% and 82.2±0.72% of GEM and PTX respectively in 72 h in PBS pH 4.0 : methanol (7:3). Furthermore, stability testing data indicated that NLCs formulations stored at 5°C±3°C were more stable than those stored at 25 °C±2 °C/60% RH ± 5% RH. These results suggest that folic acid-conjugated GEM-PTX- NLCs have the potential for serving as a novel carrier system for delivery of antineoplastic agents. [ABSTRACT FROM AUTHOR]

The study emphases on polyherbal antidiabetic formulations of different plants used in the treatment of diabetes mixed in different concentrations. In the current study four medicinal plants with proven antidiabetic were selected for the preparation of formulation. The efficacy of prepared mixture has been tested on streptozotocin- (STZ-) induced diabetic rats and compared with a commercially existing drug glibenclamide. The polyherbal formulation was formulated using the ethanol extracts ofleaves of Gymnema sylvestre bark of Pterocarpus marsupium, stem of Tinospora cordifolia and seeds of Trigonella foenum-garecumin the ratio of 3:3:2:2.The investigational drug was administered for 21 consecutive days, and the effect of the polyherbal formulation on blood glucose levels was studied at regular intervals. The elevated level of SGPT, SGOT, and ALP in the diabetic controlled group reflected the significant alteration of liver function by STZ induction and was found to be equipotent to glibenclamide in restoration of the elevated enzyme levels to normal. The elevated lipid levels (triglyceride and total cholesterol) were restored to near normal by these mixtures for all the estimated parameters. The results of the mixtures on treated group were found to restore the glycemic level to the near normal level thereby indicating antihyperglycemic activity of the formulated mixtures. Paclitaxel (Ptxl) is currently the most common first-line therapeutic option for cancer. However, adverse side effects and problems associated with chemo-resistance, limits its use in clinical settings. Nanoparticle mediated delivery can improve Ptxl delivery and its activity at the tumor site, and considered to be an attractive strategy for cancer therapy. Therefore, we aim to generate aimprovedPtx medical nanoparticle formulation (PMNPs) using a polyphenol as a carrier, and evaluate its efficacy against Cancer cells. Physico-chemical characterization of this PMNPs was performed using TEM, DLS, FT-IR, TGA, X-RD, and SAXS methods. Its internalization and cellular availability was examined using HPLC method. Cell proliferation and colony formation assays were utilized to evaluate therapeutic efficacy of this exclusivenanoformulation in clinically relevant cell line models (MCF7 and MDA-MB-231). Additionally, molecular effects of this formulation on apoptosis, anti-apoptosis, and drug resistant associated proteins were evaluated using immunoblotting assays. A tubulin stabilization study was performed using confocal immunofluorescence microscopy analysis. The PMNPs formulation showed optimal particle size and zeta potential, which can be efficiently internalized in Cancer cells. PMNPs exhibited potent anti-cancer efficacy via induction of the expression of apoptosis associated proteins (Bax and Bad, cleaved PARP, and caspase 3) and downregulation of anti-apoptotic proteins (Bcl-2 and Bcl-xL) in Cancer ell lines. Further, the expression of chemoresistance-associated proteins were also decreased with the treatment of PMNPs. Thus PMNPs formulation can efficiently be internalized in Cancer cells and induce enhanced therapeutic potential of Ptxl. Development of a targeted drug delivery system would not only reduce the dose of paclitaxel required to kill cancer cells but also minimize long-term paclitaxel associated systemic toxicity and drug-resistance. It also looks at how well they work as therapeutic candidates and finds promising peptide-based strategies for better outcomes. Furthermore, it underscores the need for further studies to support their clinical utility and suggests that results from such investigations will enhance our understanding of the pathophysiology of these conditions.
