To solve the oxygen dependence problem of photodynamic therapy (PDT), it is critical to explore photosensitizers that do not rely on O2 molecule to generate reactive oxygen species (ROS). Herein, a stable cationic metal‐organic cage [Pd6(RuLoz3)8](BF4)28 (MOC‐88) that possesses high +28 charges is designed. The cage‐confined positive microenvironment enables efficient generation of hydroxyl radicals and improved yield of the singlet oxygen under one‐/two‐photon excitation, showing excellent performance to concurrently enhance Type‐II and O2‐independent‐Type‐I PDT. Moreover, the effective ROS production and robust lipid peroxidation trigger a series of signaling pathways (inflammasome, cyclic guanosine monophosphate–adenosine monophosphate synthase stimulator of interferon genes, and NF‐κB) to evoke PANoptosis and ferroptosis in tumor cells, enabling MOC‐88 to simultaneously cause the loss of cell membrane integrity, release a series of inflammatory cytokines and damage‐associated molecular patterns, stimulate the maturation and antigen presentation ability of dendritic cells, and ultimately activate T‐cell‐dependent adaptive immunity in vivo to inhibit tumor growth.

Objective: The purpose of this research was to examine the potential anticancer properties of Xiao Tan San Jie Fang (XTSJF) and its potential mechanism of action against colorectal cancer. Materials and Methods: HCT116 cells were induced into HCT116 spheres in DMEM/F12 medium by treatment with epidermal growth factor + fibroblast growth factor + leukemia inhibitory factor + B27. The proliferation ability and stemness of HCT116 spheres was examined. Various concentrations of XTSJF were used to treat HCT116 spheres to observe the impact on proliferation, apoptosis, and expression of stem cell markers. Next, Wnt/β-catenin pathway-related factor proteins were detected. Results: The findings revealed that XTSJF suppressed cell growth and induced cell death in HCT116 cells in a dosage-dependent manner. Similarly, XTSJF promotes apoptosis, inhibits cell proliferation, prolongs survival, and maintains the expression of stem cells through the Wnt/-catenin/TCF4 axis. XTSJF also inhibits AKT activity and subsequently activates glycogen synthesis kinase-3β expression, inhibiting Wnt/beta-catenin pathway activity and downstream target gene transcript expression. The Wnt/β-catenin signaling pathway is inhibited by the XTSJF, leading to the suppression of colon cancer stem cell proliferation. Conclusion: Xiaotan Sanjie Prescription inhibited colon cancer stem cell growth through Wnt/β-catenin signaling pathway.

Diabetic retinopathy(DR)is a neurovascular disease caused by the neurovascular unit(NVU)impairment. Immune imbalance and inflammation are key factors that affect the normal function of NVU and lead to the progression of DR. Nucleotide-binding oligomerization domain-like receptor protein 3(NLRP3)inflammasome is indicated as an important component of the inflammatory response, and it can identify endogenous danger signals, leading to the activation of caspase-1 and then activating a series of inflammatory cytokines and pyroptosis. Early activation of inflammasome maintains and promotes innate immunity against bacterial and viral infections, while excessive inflammasome activation results in excessive expression and ongoing action of inflammatory proteins, which in turn triggers off immune disorders and an inflammatory cascade that seriously harms the body. This review summarizes the recent research progress on the mechanism of NLRP3 inflammasome in NVU impairment of DR, including the related drugs targeting NLRP3 pathways.

Secondary flowering is the phenomenon in which a tree blooms twice or more times a year. Along with the development of blueberry (Vaccinium corymbosum L.) fruits in spring, a large number of secondary flowers on the strong upright spring shoots were noticed in blueberries planted in the greenhouse. To reveal the cause and possible regulatory mechanism of the phenomenon, we clarified the phenological characteristics of flower bud differentiation and development on the spring shoots by combining phenological phenotype with anatomical observation. Furthermore, the changes in carbohydrates, trehalose-6-phosphate (Tre6P), and the relationship among the key enzyme regulatory genes for Tre6P metabolism and the key regulatory genes for flower formation during the differentiation process of apical buds and axillary buds were investigated. The results showed that the process of flower bud differentiation and flowering of apical and axillary buds was consistent, accompanied by a large amount of carbohydrate consumption. This process was positively correlated with the expression trends of VcTPS1/2, VcSnRK1, VcFT, VcLFY2, VcSPL43, VcAP1, and VcDAM in general, and negatively correlated with that of VcTPP. In addition, there is a certain difference in the differentiation progress of flower buds between the apical and axillary buds. Compared with axillary buds, apical buds had higher contents of sucrose, fructose, glucose, Tre6P, and higher expression levels of VcTPS2, VcFT, VcSPL43, and VcAP1. Moreover, VcTPS1 and VcTPS2 were more closely related to the physiological substances (sucrose and Tre6P) in axillary bud and apical bud differentiation, respectively. It was suggested that sucrose and trehalose-6-phosphate play a crucial role in promoting flower bud differentiation in strong upright spring shoots, and VcTPS1 and VcTPS2 might play a central role in these activities. Our study provided substantial sight for further study on the mechanism of multiple flowering of blueberries and laid a foundation for the regulation and utilization of the phenomenon of multiple flowering in a growing season of perennial woody plants.

AbstractObjective The present study aims to investigate the clinical and histopathological features of peritoneal endometriosis (PEM) and deep infiltrating endometriosis (DIE).Methods A total of 100 patients with PEM and DIE admitted to Dalian Women and Children’s Hospital/Dalian Women and Children’s Medical Center between October 2018 and December 2021 were selected as the study subjects. Morinda officinalis oligosaccharides (MOO) are an oral drug approved in China for the treatment of depression in China. However, MOO is hardly absorbed so that their anti-depressant mechanism has not been elucidated. Here, we show that oral MOO acted on tryptophan → 5-hydroxytryptophan (5-HTP) → serotonin (5-HT) metabolic pathway in the gut microbiota. MOO could increase tryptophan hydroxylase levels in the gut microbiota which accelerated 5-HTP production from tryptophan; meanwhile, MOO inhibited 5-hydroxytryptophan decarboxylase activity, thus reduced 5-HT generation, and accumulated 5-HTP. The raised 5-HTP from the gut microbiota was absorbed to the blood, and then passed across the blood–brain barrier to improve 5-HT levels in the brain. Additionally, pentasaccharide, as one of the main components in MOO, exerted the significant anti-depressant effect through a mechanism identical to that of MOO. This study reveals for the first time that MOO can alleviate depression via increasing 5-HTP in the gut microbiota.

Measuring the landscape pattern from a three-dimensional perspective is of great significance for comprehensively revealing the complex spatial characteristics of island ecosystems. However, the archipelago composed of rocky islands has received little attention as its three-dimensional landscape characteristics are difficult to quantify. This study took the largest archipelago in China, the Zhoushan Archipelago, as the study area and constructed an island landscape pattern evaluation model from a dual-three-dimensional (dual-3D) perspective. The model divided the island into upper and lower layers, namely the surface landscape based on topography and the landscape elements above the surface (i.e., vegetation and buildings), and then evaluated their landscape patterns from a three-dimensional perspective, respectively. The landscape pattern model based on a dual-3D perspective and multiple scales achieved excellent results in the archipelago. First, the island landscape pattern was evaluated from three-dimensional perspectives, including human interference, landscape fragmentation, vegetation space, and building space. Second, landscape indices such as the human interference three-dimensional index (HITI), the landscape fragmentation three-dimensional index (LFTI), the vegetation three-dimensional index (VTI), and the building three-dimensional index (BTI) established at multiple spatial scales revealed spatial heterogeneity within and between islands. Environmental factors such as elevation, slope, and island area exhibited significant correlations with them. There were significant differences in landscape pattern indices between the two-dimensional (2D) and the three-dimensional (3D) perspectives, and high values were mainly distributed in areas with significant topographic changes and larger islands. In addition, as the evaluation unit increased, the landscape indices increased, and HITI became more responsive to the transitions from 2D to 3D, while LFTI was the opposite. Therefore, the multiscale landscape pattern measurement of China’s largest archipelago based on high-resolution remote sensing was carried out from three-dimensional perspectives to accurately reveal the spatial heterogeneity.

Abstract Fibroblast-like synoviocytes (FLS) play a pathogenic role in rheumatoid arthritis (RA). STAT3 signaling is activated in FLS of RA patients (RA-FLS), which in turn causes RA-FLS hyperproliferation. RL is a traditional remedy for treating inflammatory diseases in China. It comprises Rosae Multiflorae Fructus and Lonicerae Japonicae Flos. A standardized ethanolic extract of RL (RLE) has been shown to exert anti-arthritic effects in collagen-induced arthritis (CIA) rats. Some constituents of RLE were reported to inhibit JAK2/STAT3 signaling in rat FLS. Here, we determined whether RLE inhibits FLS hyperproliferation, and explored the involvement of STAT3 signaling in this inhibition. In joints of CIA rats, RLE increased apoptotic FLS. In IL-6/sIL-6R-stimulated RA-FLS, RLE reduced cell viability and evoked cell apoptosis. In synovial tissues of CIA rats, RLE lowered the protein level of phospho-STAT3. In IL-6/sIL-6R-stimulated RA-FLS, RLE inhibited activation/phosphorylation of STAT3 and JAK2, decreased the nuclear localization of STAT3, and downregulated protein levels of Bcl-2 and Mcl-1. Over-activation of STAT3 diminished RLE’s anti-proliferative effects in IL-6/sIL-6R-stimulated RA-FLS. In summary, RLE inhibits hyperproliferation of FLS in rat and cell models, and suppression of STAT3 signaling contributes to the underlying mechanisms. This study provides further pharmacological groundwork for developing RLE as a modern anti-arthritic drug.

AIM: To determine the risk factors of age-related macular degeneration(ARMD)in high altitude areas and establish Nomoto prediction model.METHODS:Using the method of cluster random sampling, the subjects were selected from some communities in Xining City, with an average altitude of 2 100m. The subjects were ≥40 years old, and the investigation method was field investigation. Single factor and multi factor analysis were used to determine the risk factors of ARMD, and R software was used to draw Nomoto.RESULTS: The actual number of subjects in this survey is 2 595. Age, cataract, living time at high altitude, smoking, drinking, high blood pressure and mobile phone use are risk factors of ARMD. Old age was the most risk factor for ARMD(OR: 53.078, 95% CI: 28.405-77.183, POR: 9.142, 95% CI: 1.906-43.846, PCONCLUSION: The risk factors of ARMD are old age, high altitude living time, cataract, smoking, drinking, high blood pressure and mobile phone using time, especially the old people who live in high altitude for a long time. Nomo scoring system can accurately predict ARMD and provide theoretical basis for clinicians to intervene ARMD in high altitude areas.

Introduction Gastric cancer is the fifth most common cancer worldwide and the detection rate of proximal gastric cancer has been increasing. Currently, surgical resection using gastrectomy and proper perigastric lymphadenectomy is the only treatment option to enhance the survival rate of patients with gastric cancer. Laparoscopic total gastrectomy (LTG) is increasingly performed for adenocarcinoma of the oesophagogastric junction. However, totally LTG (TLTG) is only performed by a few surgeons due to difficulty associated with oesophagojejunostomy (OJ), in which there is no consensus on a standardised anastomosis technique. We propose a randomised trial to compare functional end-to-end anastomosis (FETE) and side-to-side anastomosis (Overlap) for OJ.Methods and analysis A prospective, randomised, open-label, single-centre, interventional trial has been designed to evaluate the quality of life (QoL) outcomes and safety of FETE and Overlap, with a 1-year follow-up as the primary endpoint. The trial began in 2020 and is scheduled to enrol 96 patients according to a previous sample size calculation. Patients were randomly allocated to the FETE or Overlap groups with a follow-up of 1 year to assess QoL after the procedure. All relevant clinical data including biological markers were collected. The primary indicator is the D-value between the postoperative and preoperative QoL. Student’s t-tests will be used to compare continuous variables, while χ2 tests or Fisher’s exact tests will be used to compare categorical variables. Potato (Solanum tuberosum L.), a species of the family Solanaceae, is the fourth most important food crop worldwide. Solanum tuberosum L. cv. Shepody is a long, smooth, white-skinned potato cultivar with medium green leaves. It has good specific gravity and boils and bakes well. To support more molecular data for breeding of S. tuberosum, the complete chloroplast (cp) genome sequence of S. tuberosum L. cv. Shepody was determined using the next-generation sequencing. In leaves, the chloroplast genome accounts for 3.88% of the total genome. The entire cp genome was determined to be 155,296 bp in length. It contained large single-copy (LSC) and small single-copy (SSC) regions of 85,737 and 18,373 bp, respectively, which were separated by a pair of 25,593 bp inverted repeat (IR) regions. The genome contained 132 total genes, including 87 protein-coding genes, 37 tRNA genes, and 8 rRNA genes. The overall GC content of the genome is 37.9%. A phylogenetic tree reconstructed by 60 chloroplast genomes reveals that S. tuberosum L. cv. Shepody was closely related to S. tuberosum L. cv. Desiree with bootstrap support values of 100%.

Abstract In this paper, for the first time, a rationally designed strategy for synthesis of Co nanoparticles encapsulated by boron and nitrogen co‐doped carbon nanosheets (CNs), namely B, N‐Co/CNs, as highly efficient electrocatalyst for oxygen reduction reaction (ORR) is demonstrated. The generated Co nanoparticles not only create well‐defined heterointerfaces with high conductivity to overcome the poor ORR activity but also promote the formation of robust graphitic carbon. The co‐existence of boron and nitrogen atoms can increase the highest occupied molecular orbital energy of sp2 hybridization, activating π electrons in graphitic CNs, thereby enhancing the activity of the catalyst. The B, N‐Co/CNs exhibit a comparable half‐wave potential (E1/2 = 0.83 V) to that of commercial Pt/C catalyst (E1/2 = 0.85 V) with a larger current density for ORR. Importantly, the homemade disposable Zn‐air battery (ZAB) is able to deliver excellent performance, including a peak power density of 93.93 mW cm−2 and a specific capacity of 727.5 mAh g−1, outperforming the Pt/C catalyst. The findings highlight a new guideline for constructing B, N‐Co/CNs catalyst with a rationally designed structure toward superior property for advanced metal‐air cathode materials.

Snakeheads are economically important freshwater fish in China, of which males grow much faster than females, and the individual size determines the trading price. All-male NBS (northern snakehead (Channa argus, NS) ♀ × blotched snakehead (Channa maculata, BS) ♂) were produced by combining sex control, sex-specific molecular marker and hybridization of YY super-male BS and normal XX female NS. In this study, the culture performance of all-male NBS was evaluated, including yield, survival rate, growth rate, sex ratio, size uniformity, feed conversion ratio, and low-temperature resistance. The results showed that all-male NBS exhibited superior productivity traits compared to the existing cultured snakehead varieties. The average body weight of all-male NBS was heavier than that of mixed-sex NBS by 23.1–38.8 %, or inbred NS by 16.8–34.0 %; the average daily gains of all-male NBS were faster than those of mixed-sex NBS by 28.4–39.3 %, or inbred NS by 18.6–34.1 %. The male ratio of all-male NBS was more than 93.0 %, and there was a small proportion of female or bisexual individuals. The rates of body weight above 1 kg of all-male NBS were 91.3–96.4 %, which were far higher than those of mixed-sex NBS or inbred NS, which were only 62.0–75.8 %. Therefore, all-male NBS individuals are larger and more uniform. The feed conversion ratios of all-male NBS decreased 12.5–17.6 % compared to mixed-sex NBS fed artificial compound feed and 72.3 % compared to inbred NS fed iced fresh fish. In addition, the low-temperature resistance of all-male NBS was high, with only 4.1 % mortality vs. 28.5–37.4 % in other hybrids. In snakeheads trading, large individuals are more favored, and the price of snakeheads above 1 kg is approximately 40 % higher than that of individuals in the range of 0.5–2 kg. Therefore, the large and uniform all-male NBS population could bring tremendous economic benefits, because input costs are falling, while output profits are increasing. The large and uniform all-male NBS population is thus quite promising in snakeheads market applications.

Objective To investigate the effect of psychological intervention on the prognosis of patients with C4 dislocation and spinal cord injury. Methods We investigated target patients admitted between 2010 and 2018. Patients’ mental state, quality of life and neurological function at different time points were evaluated to examine the relationship between psychological intervention and recovery and prognosis of acute and critical spinal cord injury. Results All patients showed improvements in clinical symptoms, neurological function and quality of life. Psychological intervention significantly improved Zung Self-Rating Anxiety Scale score, Zung Self-Rating Depression Scale score and SF-36 Mental Component Summary score within 3 months. Japanese Orthopaedic Association neurological function score and SF-36 Physical Component Summary score were significantly improved after 1-year follow-up. Psychological intervention did not improve 2-year survival. Conclusion Timely and professional psychological intervention can eliminate the psychological disorders of C4 dislocation patients with spinal cord injury. This has a positive effect on their quality of life and prognosis.

Abstract Background Rhizoma Paridis (Chonglou) is a commonly used and precious traditional Chinese medicine. Paris polyphylla Smith var. yunnanensis (Franch.) Hand. -Mazz. and Paris polyphylla Smith var. chinensis (Franch.) Hara are the two main sources of Chonglou under the monograph of Rhizoma Paridis in Chinese Pharmacopoeia. In the local marketplace, however, this medicine is prone to be accidentally contaminated, deliberately substituted or admixed with other species that are similar to Rhizoma Paridis in shape and color. Consequently, these adulterations might compromise quality control and result in considerable health concerns for consumers. This study aims to develop a rapid and sensitive method for accurate identification of Rhizoma Paridis and its common adulterants. Methods DNA barcoding coupled with high resolution melting analysis was applied in this research to distinguish Rhizoma Paridis from its adulteration. The internal transcribed spacer 2 (ITS2) barcode was selected for HRM analysis to produce standard melting profile of the selected species. DNA of the tested herbal medicines was isolated and their melting profiles were generated and compared with the standard melting profile of P. polyphylla var. chinensis. Results The results indicate that the ITS2 molecular regions coupled with HRM analysis can effectively differentiate nine herbal species, including two authentic origins of Chonglou and their seven common adulterants. Ten herbal medicines labeled “Chonglou” obtained from a local market were collected and identified with our methods, and their sequence information was analyzed to validate the accuracy of HRM analysis. Conclusions DNA barcoding coupled with HRM analysis is a accurate, reliable, rapid, cost-effective and robust tool, which could contribute to the quality control of Rhizoma Paridis in the supply chain of the natural health product industry (NHP).

Neuropathic pain often occurs during chemotherapy with oxaliplatin. AG490 has been shown to exert an antagonistic effect on inflammatory pain, but its effect on oxaliplatin-induced neuropathic pain remains poorly understood. This study sought to observe the analgesic effect of AG490 on acute neuropathic pain induced by a single oxaliplatin treatment and to address the possible mechanism. In this study, we established a model of oxaliplatin-induced acute neuropathic pain by intraperitoneal injection of 6 mg/kg oxaliplatin. On day 2 after injection, models were intraperitoneally injected with 1, 5, or 10 mg/kg AG490. Paw withdrawal threshold to mechanical stimuli and tail withdrawal latency to cold stimuli were determined. Western blot assay was performed to detect the expression of spinal phosphorylated signal transducer and activator of transcription 3 (p-STAT3). Immunohistochemistry was used to determine the immunoreactivity of p-STAT3 and interleukin-6. Results demonstrated that paw withdrawal threshold and tail withdrawal latency were significantly increased by the treatment of AG490 in rats. There was no significant difference in the effect among the different doses of AG490. AG490 10 mg/kg decreased the expression of p-STAT3, the immunoreactivity of p-STAT3 and interleukin-6 in spinal cord of acute neuropathic pain rats. These findings confirm that AG490 can attenuate oxaliplatin-induced acute neuropathic pain and is associated with the inhibition in the JAK/STAT3 signaling pathway.

Objective: To evaluate the clinical features of Chinese women with idiopathic hypogonadotropic hypogonadism (IHH). Methods: We retrospectively reviewed the clinical characteristics, laboratory and imaging findings, therapeutic management and fertility outcomes of 138 women with IHH. All patients had been treated and followed up at an academic medical centre during 1990–2016. Results: Among the 138 patients, 82 patients (59.4%) were diagnosed with normosmic IHH and 56 patients (40.6%) were diagnosed with Kallmann syndrome (KS). The patients with IHH experienced occasional menses (4.3%), spontaneous thelarche (45.7%) or spontaneous pubarche (50.7%). Women with thelarche had a higher percentage of pubarche (P < 0.001) and higher gonadotropin concentrations (P < 0.01). Olfactory bulb/sulci abnormalities were found during the magnetic resonance imaging (MRI) of all patients with KS. Most patients with IHH had osteopenia and low bone age. Among the 16 women who received gonadotropin-releasing hormone treatment, ovulation induction or assisted reproductive technology, the clinical pregnancy rate was 81.3% and the live birth rate was 68.8%. Conclusions: The present study revealed that the phenotypic spectrum of women with IHH is broader than typical primary amenorrhoea with no secondary sexual development, including occasional menses, spontaneous thelarche or pubarche. MRI of the olfactory system can facilitate the diagnosis of KS. Pregnancy can be achieved after receiving appropriate treatment.

Uncoordinated carbon-nitrogen ratio in raw materials will lead to excessive contents of higher alcohols in alcoholic beverages. The effect of GAT1 gene, the GATA transcription activator, on higher alcohol biosynthesis was investigated to clarify the mechanism of Saccharomyces cerevisiae regulating higher alcohol metabolism under high concentrations of free amino nitrogen (FAN). The availability of FAN by strain SDT1K with a GAT1 double-copy deletion was 28.31% lower than that of parent strain S17, and the yield of higher alcohols was 33.91% lower. The transcript levels of the downstream target genes of GAT1 and higher alcohol production in the double-copy deletion mutant suggested that a part of the effect of GAT1 deletion on higher alcohol production was the downregulation of GAP1, ARO9, and ARO10. This study shows that GATA factors can effectively regulate the metabolism of higher alcohols in S. cerevisiae and provides valuable insights into higher alcohol biosynthesis, showing great significance for the wheat beer industry.

A series of Ln3+-metal centered complexes, Ln(TTA)3(DPPI) (Ln = La, 1; Ln = Eu, 2; Ln = Tb, 3; or Ln = Gd, 4) [(DPPI = N-(4-(1H-imidazo [4,5-f][1,10]phenanthrolin-2-yl)phenyl)-N-phenylbenzenamine) and (TTA = 2-Thenoyltrifluoroacetone)] have been synthesized and characterized. Among which, the Eu3+-complex shows efficient purity red luminescence in dimethylsulfoxide (DMSO) solution, with a Commission International De L’ Eclairage (CIE) coordinate at x = 0.638, y = 0.323 and ΦEuL = 38.9%. Interestingly, increasing the amounts of triethylamine (TEA) in the solution regulates the energy transfer between the ligand and the Eu3+-metal center, which further leads to the luminescence color changing from red to white, and then bluish-green depending on the different excitation wavelengths. Based on this, we have designed the IMPLICATION logic gate for TEA recognition by applying the amounts of TEA and the excitation wavelengths as the dual input signal, which makes this Eu3+-complex a promising candidate for TEA-sensing optical sensors.

Background: Chronic kidney disease (CKD) with moderate-to-severe renal dysfunction usually exhibits an irreversible course, and available treatments for delaying the progression to end-stage renal disease are limited. This study aimed to assess the efficacy and safety of the traditional Chinese medicine, Niaoduqing particles, for delaying renal dysfunction in patients with stage 3b-4 CKD. Methods: The present study was a prospective, randomized, double-blind, placebo-controlled, multicenter clinical trial. From May 2013 to December 2013, 300 CKD patients with an estimated glomerular filtration rate (eGFR) between 20 and 45 ml·min−1·1.73 m−2, aged 18–70 years were recruited from 22 hospitals in 11 Chinese provinces. Patients were randomized in a 1:1 ratio to either a test group, which was administered Niaoduqing particles 5 g thrice daily and 10 g before bedtime for 24 weeks, or a control group, which was administered a placebo using the same methods. The primary endpoints were changes in baseline serum creatinine (Scr) and eGFR after completion of treatment. The primary endpoints were analyzed using Student's t-test or Wilcoxon's rank-sum test. The present study reported results based on an intention-to-treat (ITT) analysis. Results: A total of 292 participants underwent the ITT analysis. At 24 weeks, the median (interquartile range) change in Scr was 1.1 (−13.0–24.1) and 11.7 (−2.6–42.9) μmol/L for the test and control groups, respectively (Z = 2.642, P = 0.008), and the median change in eGFR was −0.2 (−4.3–2.7) and −2.2 (−5.7–0.8) ml·min−1·1.73 m−2, respectively (Z = −2.408, P = 0.016). There were no significant differences in adverse events between the groups. Conclusions: Niaoduqing particles safely and effectively delayed CKD progression in patients with stage 3b-4 CKD. This traditional Chinese medicine may be a promising alternative medication for patients with moderate-to-severe renal dysfunction. This is a study on the relationship between the protective effect of ginsenoside Rg1 on senescent neural stem cells and Wnt-β/catenin signaling pathway. Background. Recent studies have shown that overactivation of the Wnt/β-catenin signaling pathway is closely related to stem cell senescence. Whether Rg1 delays the senescence of NSCs is related to the regulation of this signaling pathway. Methods. The whole brain of Nestin-GFP transgenic newborn rat was extracted, and NSCs were extracted and cultured to P3 generation. The following indicators were detected: (1) NSC culture identification, (2) the effect of LiCl on the proliferation and survival rate of NSCs, (3) the effect of ginsenoside Rg1 on the proliferation and survival of NSCs, (4) the growth of NSCs in each group observed by an optical microscope, (5) the cell cycle of each group detected by flow cytometry, (6) the proliferative ability of each group detected by BrdU, (7) the fluorescence intensity of Nestin and Sox2 of NSCs in each group observed by a fluorescence microscope, (8) the positive rate of senescence staining analyzed by SA-β-Gal staining, (9) the localization of β-catenin in NSCs observed by laser confocal microscopy, and (10) the changes of the Wnt/β-catenin pathway-related proteins in each group detected by Western blotting. Results. LiCl activates the Wnt/β-catenin pathway and promotes mouse neural stem cell senescence. Ginsenoside Rg1 promotes proliferation of neural stem cells and inhibits Wnt/β-catenin pathway activation. Conclusions. LiCl can activate the Wnt/β-catenin signaling pathway of NSCs, and ginsenoside Rg1 can antagonize the senescence of NSCs caused by activation of the Wnt/β-catenin signaling pathway and delay brain aging.

Hepatocellular carcinoma (HCC), the major form of primary liver cancer, is a common cause of cancer-related death worldwide. Signal transducer and activator of transcription 3 (STAT3) signaling is constantly activated in HCC and has been proposed as a chemotherapeutic target for HCC. Antrodia camphorata (AC), a medicinal mushroom unique to Taiwan, is traditionally used for treating HCC. Whereas natural AC is scarce, cultured AC mycelia are becoming alternatives. In this study, we investigated the anti-HCC effects of the ethyl acetate fraction of an ethanolic extract of AC mycelia (EEAC), particularly exploring the involvement of STAT3 signaling in these effects. We found that EEAC reduced cell viability, induced apoptosis, and retarded migration and invasion in cultured HepG2 and SMMC-7721 cells. Immunoblotting results showed that EEAC downregulated protein levels of phosphorylated and total STAT3 and JAK2 (an upstream kinase of STAT3) in HCC cells. Real-time PCR analyses showed that STAT3, but not JAK2, mRNA levels were decreased by EEAC. EEAC also lowered the protein level of nuclear STAT3, decreased the transcriptional activity of STAT3, and downregulated protein levels of STAT3-targeted molecules, including anti-apoptotic proteins Bcl-xL and Bcl-2, and invasion-related proteins MMP-2 and MMP-9. Over-activation of STAT3 in HCC cells diminished the cytotoxic effects of EEAC. In SMMC-7721 cell-bearing mice, EEAC (100 mg/kg, i.g. for 18 days) significantly inhibited tumor growth. Consistent with our in vitro data, EEAC induced apoptosis and suppressed JAK2/STAT3 activation/phosphorylation in the tumors. Taken together, EEAC exerts anti-HCC effects both in vitro and in vivo; and inhibition of STAT3 signaling is, at least in part, responsible for these effects. We did not observe significant toxicity of EEAC in normal human liver-derived cells, nude mice and rats. Our results provide a pharmacological basis for developing EEAC as a safe and effective agent for HCC management.

Background: The efficacy and safety of telmisartan combined with clopidogrel, leflunomide, or both drugs for immunoglobulin A nephropathy (IgAN) are unclear. This study was designed to evaluate the efficacy and safety of telmisartan combined with clopidogrel, leflunomide, or both drugs for IgAN. Methods: It is a multicenter, prospective, double-dummy randomized controlled trial. Primary IgAN patients were recruited in 13 renal units across Beijing, China, from July 2010 to June 2012. After a 4-week telmisartan (80 mg/d) wash-in, 400 patients continuing on 80 mg/d telmisartan were randomly assigned to additionally receive placebo (Group A), 50 mg/d clopidogrel (Group B), 20 mg/d leflunomide (Group C), or 50 mg/d clopidogrel and 20 mg/d leflunomide (Group D). The 24-week intervention was completed by 360 patients. The primary endpoint was change in 24-h proteinuria at 24 weeks. A linear mixed-effect model was used to analyze the changes at 4, 12, and 24 weeks. Generalized estimating equations were used to evaluate changes in hematuria grade. This trial was registered at the Chinese Clinical Trial Registry. Results: The effects of telmisartan combined with leflunomide on changes in proteinuria (0.36 [95% confidence interval (CI) 0.18–0.55] g/d, P < 0.001), in serum uric acid (76.96 [95% CI 57.44–96.49] μmol/L, P < 0.001), in serum creatinine (9.49 [95% CI 6.54–12.44] μmol/L, P < 0.001), and in estimated glomerular filtration rate (−6.72 [95% CI−9.46 to −3.98] ml·min−1·1.73 m−2, P < 0.001) were statistically significant, whereas they were not statistically significant on changes in systolic and diastolic blood pressure and weight (P > 0.05). Telmisartan combined with clopidogrel had no statistical effect on any outcome, and there was no interaction between the interventions. No obvious adverse reactions were observed. Conclusions: Telmisartan combined with leflunomide, not clopidogrel, is safe and effective for decreasing proteinuria in certain IgAN patients. Background: It is now recognized that Cimicifuga foetida (C. foetida) extract is effective in alleviating menopausal symptoms. But the durations reported were usually short. The aim of this study was to investigate the effects of C. foetida extract therapy and different estrogen and progesterone sequential therapies, on the breasts of early postmenopausal women. Methods: This was a prospective randomized trial. Ninety-six early menopausal women were recruited and randomly assigned into three groups treated with different therapies for 2 years. Patients were given C. foetida extract in Group A, estradiol valerate and medroxyprogesterone acetate in Group B, and estradiol valerate and progesterone in Group C. Ultrasonography was used to monitor changes in breast during treatment. Results: In comparing breast glandular section thickness before and after 1 and 2 years of treatment, no significant difference was observed in Group A (11.97 ± 2.84 mm vs. 12.09 ± 2.58 mm and 12.61 ± 3.73 mm, P > 0.05); in Group B glandular section thickness had increased significantly (10.98 ± 2.34 mm vs. 11.84 ± 2.72 mm and 11.90 ± 3.33 mm, P < 0.05) after treatment, the same as Group C (11.56 ± 3.03 mm vs. 12.5 ± 3.57 mm and 12.22 ± 4.39 mm P < 0.05). In comparing breast duct width before and after 1 and 2 years of treatment, no significant difference was seen in Group A (1.07 ± 0.19 mm vs. 1.02 ± 0.18 mm and 0.98 ± 0.21 mm, P > 0.05); in Group B the duct width had a downward trend after treatment (0.99 ± 0.14 mm vs. 0.96 ± 0.22 mm and 0.90 ± 0.18 mm, P < 0.05), the same as Group C (1.07 ± 0.20 mm vs. 1.02 ± 0.17 mm and 0.91 ± 0.19 mm, P < 0.05). The nodules detected before treatment had disappeared after 1-year of treatment or exhibited no distinct changes in the three groups. However, new breast nodules had appeared after 2 years of treatment: There was one case in Group A, two cases in Group B and four cases in Group C, with breast hyperplasia after the molybdenum target check. Conclusions: In early postmenopausal patients, C. foetida extract therapy and estrogen and progesterone therapy at low doses did not increase the incidence of malignant breast tumors

hMLH1 is one of the mismatch genes closely related to the occurrence of gastric cancer. Epigenetic regulation may play more important roles than gene mutations in DNA damage repair genes to drive carcinogenesis. In this article, we discuss the role of epigenetic changes, especially histone modifications in the regulation of hMLH1 alternative splicing. Our results showed that hMLH1 del Ex10, del Ex11, del Ex10–11, del Ex16 and del Ex17 transcripts were ubiquitous in sporadic Chinese gastric cancer patients and gastric cancer cell lines. Lower level of H4K16ac and H3ac was detected in hMLH1 exon 10–11 region in gastric cancer cell lines when compared with human gastric mucosal epithelial cell line GES-1. A significant decrease of hMLH1 del Ex11 and del Ex10–11 was observed in gastric cancer cell lines after trichostatin A treatment. H3K36me3 and H3K4me2 levels were lower in hMLH1 exon 10–11 and exon 16–17 regions in gastric cancer lines when compared with GES-1. Aberrant transcripts such as hMLH1 del Ex11 and del Ex10–11 were significantly higher in gastric cancer cell lines after small interfering RNA–mediated knockdown of SETD2 (the specific methyltransferase of H3K36). The hMLH1 del Ex10 and del Ex10–11 transcripts were increased after interference of SRSF2. Taken together, our study demonstrates that lower level of histone acetylation and specific histone methylation such as H3K36me3 correlate with aberrant transcripts in hMLH1 exon 10–11 region. SRSF2 may be involved in these specific exons skipping as well.

Abstract.: The main purpose of this study is to evaluate the effect of echinacoside (ECH) on hypoxia-induced proliferation of rat pulmonary artery smooth muscle cells (PASMCs) and the underlying mechanism. PASMCs were incubated under normoxia (nor), hypoxia (hyp), hypoxia + 0.35 mM ECH (hyp + ECH0.35), or hypoxia + 0.4 mM ECH (hyp + ECH0.4) for 24 h. Cell viability was assessed by MTS assays. The morphology of apoptosis was observed by DAPI staining, and apoptosis was quantified by flow cytometric analysis. Caspase-3 activity was determined by immunohistochemistry and real-time PCR, and the expressions of HIF-1α, Bax, Bcl-2, and Fas were determined by real-time PCR. Hypoxia induced significant proliferation of PASMCs, which could be inhibited by ECH in a concentration-dependent manner. This was associated with apoptosis of PASMCs. Z-DEVD-FMK could partly reduce the suppression effect of ECH; protein and gene expression of caspase-3 were significantly higher in the hyp + ECH0.4 and hyp + ECH0.35 groups. ECH significantly increased the expressions of Bax and Fas, but decreased the expressions of Bcl-2 and HIF-1α. ECH could inhibit hypoxia-induced proliferation of rat PASMCs, which is associated with apoptosis of PASMCs and improvement of hypoxia. ECH might be a potential agent for prevention and treatment of hypoxia-induced PAH. BACKGROUND:We investigated whether there had been an improvement in the quality of reporting for randomised controlled trials of acupuncture and moxibustion published in Chinese journals. We compared the compliance rate for the quality of reporting following the publication of both the STRICTA and CONSORT recommendations in China. METHODS:Four Chinese databases were searched for RCTs of acupuncture from January 1978 through to December 2012. The CONSORT and STRICTA checklists were used to assess the quality of reporting. Data were collected using a standardised form. All included RCTs were divided into three distinct time periods based on the time that CONSORT and STRICTA were introduced in China, respectively. Pearson's χ2 test and/or Fisher's exact test were used to assess differences in reporting among three groups. PRINCIPAL FINDINGS:A total of 1978 RCTs were identified. Although the percentage of all the items has increased over time with the introduction of CONSORT and STRICTA in China, the actual compliance in several important methodological components, including sample size calculation (0% vs. 0% vs. 1.2%, for pre-CONSORT and pre-STRICTA, post-CONSORT but pre-STRICTA, and post-CONSORT and post-STRICTA, respectively), randomisation sequence generation (1.4% vs. 15% vs. 26.3%) and implementation (0% vs. 0% vs. 1.3%), allocation concealment (0% vs. 1.4% vs. 4.9%), and blinding (0% vs. 5.7% vs. 9.1%), remains low. Moreover, no RCTs have reported the setting and context of treatment and no descriptions of the participating acupuncturists have been provided thus far. CONCLUSIONS:Overall, the quality of the reporting of RCTs of acupuncture and moxibustion published in Chinese journals has improved since CONSORT and STRICTA were introduced in China, though the actual compliance rate of some important items were still low as of 2012. In the future, Chinese journals should enhance the adoption of the CONSORT and STRICTA statement to improve the reporting quality of the RCTs of acupuncture and moxibustion and to ensure the truth and reliability of the conclusions.

In recent years, non-rigid structure from motion (NRSFM) has become one of the hottest issues in computer vision due to its wide applications. In practice, the number of available high-quality images may be limited in many cases. Under such a condition, the performances may not be satisfactory when existing NRSFM algorithms are applied directly to estimate the 3D coordinates of a small-size image sequence. In this paper, a sub-sequence-based integrated algorithm is proposed to deal with the NRSFM problem with small sequence sizes. In the proposed method, sub-sequences are first extracted from the original sequence. In order to obtain diversified estimations, multiple weaker estimators are constructed by applying the extracted sub-sequences to a recent NRSFM algorithm with a rotation-invariant kernel (RIK). Compared to other first-order statistics, the trimmed mean is a relatively robust statistic. Considering the fact that the estimations of some weaker estimators may have large errors, the trimmed means of the outputs for all the weaker estimators are computed to determine the final estimated 3D shapes. Compared to some existing methods, the proposed algorithm can achieve a higher estimation accuracy, and has better robustness. Experimental results on several widely used image sequences demonstrate the effectiveness and feasibility of the proposed algorithm.

The researches in the dynamic changes of the progress of HSCs aging are very limited and necessary. In this study, male C57BL/6 mice were divided into 5 groups by age. We found that the superoxide damage of HSPCs started to increase from the middle age (6 months old), with notably reduced antioxidation ability. In accordance with that, the senescence of HSPCs also started from the middle age, since the self-renewal and differentiation ability remarkably decreased, and senescence-associated markers SA-β-GAL increased in the 6-month-old and the older groups. Interestingly, the telomere length and telomerase activity increased to a certain degree in the 6-month-old group. It suggested an intrinsic spontaneous ability of HSPCs against aging. It may provide a theoretical and experimental foundation for better understanding the senescence progress of HSPCs. And the dynamic biological characteristics of HSPCs senescence may also contribute to the clinical optimal time for antiaging drug intervention.

We investigated the effects of apocynin on high-fat diet- (HFD-) induced insulin resistance in C57BL/6 mice. After 12 weeks of HFD, the mice that exhibited insulin resistance then received 5 weeks of apocynin (2.4 g/L, in water). Following apocynin treatment, fasting glucose, insulin, and glucose tolerance test showed significant improvement in insulin sensitivity in HFD-fed mice. We demonstrated that serum levels of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and leptin were remarkably reduced with apocynin treatment. We also found that mRNA expression of TNF-α, IL-6, and monocyte chemoattractant protein-1 (MCP-1) in the liver and mRNA expression of TNF-α, IL-6, MCP-1, and leptin in adipose tissue were suppressed by apocynin. Furthermore, the activity of transcription factor NF-κB in the liver was significantly suppressed with apocynin treatment. These results suggest that apocynin may reduce inflammatory factors in the blood, liver, and adipose tissue, resulting in amelioration of insulin resistance in HFD-fed mice.

Background: Gravity-driven depositional processes play a pivotal role in shaping the geomorphology of subaqueous deltas worldwide, particularly by eroding the seafloor, leading to the formation of rugged submarine channels (SCs) and triggering various subaqueous geohazards. A comprehensive understanding of SCs is crucial for elucidating these depositional mechanisms and mitigating the risks associated with submarine geohazards. Although SCs in the Huanghe delta front have been previously identified, often described as seafloor gullies or subsurface "disturbed strata," most studies have primarily concentrated on their engineering geological properties. However, there has been limited research on the scale, morphology, and development of these SCs, especially those that are buried within the stratigraphy. Methods: This study integrates high-resolution sub-bottom data, sediment core analyses, and historical bathymetric data to investigate the morphology, distribution, and formation mechanisms of SCs in the Huanghe subaqueous delta. Results: The SCs, both buried and exposed, are widespread along the middle and upper delta front, predominantly located at or near the landward flank of sediment gravity flow (SGF) -related accumulations. The buried SCs are characterized by cut-and-fill structures with transparent to semi-transparent fills, indicating rapid infilling processes as SGF energy dissipates. These buried channels were preserved within different sets of delta front deposits formed during 1855-1964 CE, 1964-1976 CE, and 1976-2007 CE, with the latter two periods separated by a significant lobe-switching event in 1976 CE. This event, combined with prevailing southeastward sediment transport and erosional regimes, appears to have controlled the preservation of SCs along the delta front: SCs in the Diaokou Lobe's delta front (pre-1976) suffered significant erosion, leaving only one set of channel (erosional remnants) preserved, while the SCs in the Qingshuigou Lobe's delta front (post-1976) are characterized by well-preserved, multi-phase channels at different horizons. The cross-section morphology of the SCs reveal three primary types: symmetrical, asymmetrical, and composite, corresponding respectively to (1) idealized SGF incision, (2) uneven incision intensity on either side of the SCs, and (3) the merging of two or more symmetrical/asymmetrical SCs. Conclusions: We describe in detail the morphology, distribution and development of SCs in the modern Huanghe subaqueous delta. These findings provide insights into the formation and distribution of SCs in other shallow marine settings, particularly in delta front areas, and potentially offer information for disaster prevention and engineering development in such regions. Wave- and current-supported fluid mud on gently sloped continental shelves represents a type of sediment gravity flow capable of rapidly transporting substantial sediment over short periods, significantly contributing to coastal geomorphic evolution. To investigate the dynamics of intertidal fluid mud events, in-situ observations were conducted from May 9 to May 18, 2017, using a seabed tripod system at the lower intertidal flat of the central Jiangsu coast, China. Fluid mud was observed following a medium wind-wave event, with a maximum significant wave height of 0.42 m. The liquefied seabed, loosened by liquefaction, facilitated bed erosion and sediment resuspension. Fluid mud layers, with mean suspended sediment concentrations (SSCs) exceeding 10 g/L, periodically formed during high slack tide, early flood, and late ebb phases. These layers varied in thickness from 4 cm to 20 cm and exhibited strong stratification caused by suspended sediment. Fluid mud disappeared when the bottom turbulence kinetic energy exceeded a threshold of 0.00045 m2/s2 due to an increase in current velocity. The downslope movement of intertidal fluid mud was estimated using a theoretical buoyancy-friction model and validated by observed offshore-directed velocity jets at 0.1 m above the seabed during high slack tide phases. Additionally, onshore winds favored fluid mud formation during early flood phases, while offshore winds favored it during late ebb phases. These observations suggest that fluid mud can form on intertidal flats under conditions where tidal flows and winds align, contributing to a deeper understanding of the formation mechanisms of shallow gravity flows and the improvement of sediment transport models. Waves and currents are responsible for sedimentmovement around and off coral reefs, affecting reef growth at both geological and modern timescales. Given the wide distribution and limited hydrodynamic information of reefs in the South China Sea, we carried out observations on tidal-cycle hydrodynamics in the Nansha Islands with tripod stationary instruments on the seafloor in order to fill the gap in our understanding of these processes. It was found that the magnitudes of near-bed orbital velocity were comparable with that of the mean tidal current, despite generally calm wave conditions. Waves dominated the combined wave-current skin-friction shear velocities acting on reef sediment, which were significantly higher than those generated by currents alone. Due to the large physical roughness of reef, drag coefficient and hydrodynamic roughness length estimated from logarithmic velocity profiles were two orders of magnitude higher than that in macro-tidal-estuary or inner shelf areas covered with siliceous muds or sands. The combined sinusoidal wave and asymmetric tidal current, along with the physical reef roughness, shaped velocity profile structures in the bottom boundary layer, which exhibited a logarithmic profile during the flood tide and a potential flow during the ebb. In absence of wave breaking, strong turbulence dissipation was observed across the rough reef, promoting strong mixing of water, which is crucial for delivering nutrients for coral growth. These findings imply the need to consider the unique characteristics of rough reef structure and combined effects of waves and currents to model the hydrodynamics in reef environment correctly. This understanding is critical for predicting energy and material transport in reef environments, which is essential for maintaining healthy coral ecosystems, and opens new paths for managing and preserving coral reefs in the face of environmental change. Hence, the extent to which oil exploration can be extended, and the prediction of the depth limit of oil accumulation (DLOA), are issues that have attracted significant attention in petroleum geology. Since it is difficult to characterize the evolution of the physical properties of the marine carbonate reservoir with burial depth, and the deepest drilling still cannot reach the DLOA. Hence, the DLOA cannot be predicted by directly establishing the relationship between the ratio of drilling to the dry layer and the depth. In this study, by establishing the relationships between the porosity and the depth and dry layer ratio of the carbonate reservoir, the relationships between the depth and dry layer ratio were obtained collectively. The depth corresponding to a dry layer ratio of 100% is the DLOA. Based on this, a quantitative prediction model for the DLOA was finally built. The results indicate that the porosity of the carbonate reservoir, Lower Ordovician in Tazhong area of Tarim Basin, tends to decrease with burial depth, and manifests as an overall low porosity reservoir in deep layer. The critical porosity of the DLOA was 1.8%, which is the critical geological condition corresponding to a 100% dry layer ratio encountered in the reservoir. The depth of the DLOA was 9,000 m. This study provides a new method for DLOA prediction that is beneficial for a deeper understanding of oil accumulation, and is of great importance for scientific guidance on deep oil drilling. With sea level rise (SLR), tidal nuisance flooding has become a growing threat, especially around estuaries with large tidal amplitudes. This study investigated how sea level change affects tides in Hangzhou Bay, a macro-tidal estuary with high SLR rate. By downscaling climate projections to a regional hydrodynamic model, the amplitude of primary tidal constituent (M2) was predicted to increase by 0.25 m in the upper bay, where the amplitude of major diurnal tide (K1) was also predicted to increase by 15%. In addition, the sensitivity of tidal amplitude to mean sea level was examined by a set of numerical simulations with different SLR. It was found that the increase of tidal amplitude is nonlinear to SLR, and the tidal amplitudes almost cease to increase when SLR is over 1.5 m. Although predictions show less amplitude changes in the lower bay, Zhoushan Archipelago around the bay mouth strongly modulates the incoming tidal energy, thus affecting the tidal amplitude in the upper bay. Energy budget analysis revealed that the complex topography, such as narrow channels, in the archipelago area leads to strong horizontal shear, which dissipates approximately 25% of total tidal energy in the bay. On the other hand, around 60% of the energy is dissipated in the bottom boundary layer. However, the bottom dissipation decreases by 4% due to reduced friction, while horizontal dissipation increases by 10% due to enhanced horizontal shear with SLR. This suggests that the strong horizontal shear in the Zhoushan archipelago region can play a more important role in the tidal energy budget in the future. [ABSTRACT FROM AUTHOR]
