Objective: Eremurus spectabilis M. Bieb. is grown in East and Southeast Mediterranean regions of Turkey, and is commonly used as an edible plant in these regions. The aim of the current study was to determine the antioxidant activities of different E. spectabilis M. Bieb. extracts and some sulfur compounds. Materials and Methods: In this study, the antioxidant activities of aqueous, ethanolic, and ethyl acetate extracts of E. spectabilis M. Bieb. and sulfur compounds (α-lipoic acid, cysteamine, cysteine, diallyl sulfide, glutathione, homocysteine, N-acetyl cysteine, vitamin U and 1,4-dithioerythritol) were assessed in several antioxidant tests. These tests included reducing power, cupric ion reducing antioxidant capacity, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging, 2,2-diphenyl-1-picrylhydrazyl radical scavenging, N,N-dimethyl-4-phenylenediamine dihydrochloride radical scavenging, superoxide anion radical scavenging, hydroxyl radical scavenging, metal chelating activities and ferric ion reducing antioxidant power. The phytochemical profiles and total phenolic and flavonoid contents of all extracts were investigated. Results: It was found out that the antioxidant activities of all the extracts and sulfur compounds generally increased with concentration increases, the highest antioxidant activity was found in ethyl acetate extract, while the aqueous extract was found to have the lowest antioxidant activity. Diallyl sulfide was found to have the highest antioxidant activity, whereas vitamin U had the lowest antioxidant activity. It was observed that all extracts of E. spectabilis M. Bieb. and the sulfur compounds showed antioxidant activity. Conclusion: The E. spectabilis M. Bieb. and sulfur compounds investigated in this study could be considered as a source of antioxidants.

Amaç: Bitkilerden elde edilen doğal bileşiklerin (özellikle yenilebilir olanlar), kanser ve gastrointestinal bozukluklar gibi belirli rahatsız-lıklar üzerinde tedavi edici/önleyici etkilerinin olup olmadığı araştırılmaya devam etmektedir. Çalışmanın temel amacı, Eremurus (E) spectabilis M. Bieb. ekstrelerinin histon deasetilaz (HDAC), ksantin oksidaz (XO) ve üreaz aktiviteleri üzerindeki inhibitör etkilerini araştırmaktır.Gereç ve Yöntem: E. spectabilis’in taze yaprakları, Eyüp/İstanbul’daki, yerel bir pazardan temin edilmiştir. Etanol ve etil asetat ekstrelerinin HDAC, XO ve üreaz üzerindeki inhibitör aktiviteleri incelenmiştir.Bulgular: Elde edilen sonuçlara göre, E. spectabilis'in hem etanol hem de etil asetat ekstrelerinin incelenen tüm enzimler üzerinde belirgin inhibitör etkiye sahip olduğu bulunmuştur. Etanol ekstresi, HDAC üzerinde etil asetat ekstresinden daha iyi bir inhibisyon etkisi göstermiştir. Etil asetat ekstresinin ise, hem XO hem de üreaza karşı inhibisyon etkisinin, etanol ekstresinden daha yüksek olduğu görülmüştür.Sonuç: Sonuç olarak, incelenen bitkinin gösterdiği inhibitör etkiler nedeni ile, bu bitkinin tıbbi amaçla, ham ekstre şeklinde veya biyoaktif bileşik kaynağı olarak kullanılabileceği ileri sürülebilir., Objective: Natural ingredients derived from plants (especially ed-ible ones) continue to be investigated for understanding whether they have therapeutic/preventive effects on certain ailments, such as cancer and gastrointestinal disorders. The main goal of the current work is to investigate the inhibitory potential of Eremurus (E) spectabilis M. Bieb. extracts on histone deacetylase (HDAC), xanthine oxidase (XO) and urease activities. Material and Method: Fresh E. spectabilis leaves were obtained from a local market in Eyup/Istanbul. Inhibitory activities of ethanolic and ethyl acetate extracts on HDAC, XO and urease were examined. Results: According to our findings, ethanolic and ethyl acetate extracts of E. spectabilis both have revealed apparent inhibitory effects against all examined enzymes. Ethanolic extract showed a better inhibitory effect on HDAC than ethyl acetate extract. On the other hand, the inhibitory effects of ethyl acetate extract against both XO and urease were higher than the ethanolic extract. Conclusion: Consequently, due to the inhibitory effects of the plant being studied, it might be suggested that it can be used for medicinal purposes, in raw extract form, or as a source of bioactive compounds.

Background and Aims: Organosulfur compounds modulate the activities of plurality of metabolic enzymes, especially those that activate (cytochrome P450s) or detoxify (glutathione-S-transferases) carcinogens. They also inhibit the formation of DNA adducts in different target tissues. The aim of the present study was to investigate the effect of some sulfur compounds on urease and trypsin activities in vitro. Methods: In the present study, the inhibitory effect of sulfur compounds on the activities urease and trypsin were determined according to the method of Hanif et al. (2012) and Ribeiro et al. (2010), respectively. Results: In comparison to the reference standard thiourea (IC50= 53.81±0.68 μg/mL), S-allyl-L-cysteine (IC50= 0.88±0.01 μg/ mL) and D, L-methionine (IC50= 0.91±0.02 μg/mL) had the highest urease inhibitor activity, corresponding to the lowest IC50 values among the sulfur compounds. Among the sulfur compounds used in this study, D,L-methionine (IC50= 0.13±0.01 mg/ mL) exhibited the lowest IC50 value for trypsin inhibitor, though its activity was less than that of tannic acid which was used as a standard (IC50= 0.06±0.01 mg/mL). Conclusion: The present outcome suggests that sulfur compounds are potential inhibitors of urease and trypsin activities, and may find importance in medicine and agriculture.

Amaç: Aritmi tedavisinde kullanılan amiodaronun çeşitli organlarda yan etkilere neden olduğu gösterilmiştir. Beyaz lahana (Brassica oleracea L. var. capitata) yaygın olarak tüketilen bir sebzedir. Amiodaron maruziyeti ve beyaz lahana tüketiminin kalp dokusu üzerindeki etkileri araştırılmamıştır. Beyaz lahana ekstraktının amiodaron uygulanan sıçanların kalp dokuları üzerindeki etkilerini araştırmayı amaçladık. Gereç ve Yöntem: Sıçanlara yedi gün boyunca gavajla beyaz lahana ekstresi ve amiodaron verildi. Beyaz lahana ekstresinin uygulanmasından 1 saat sonra amiodaron uygulandı. Sekizinci günde, hayvanların kalpleri alındı ve homojenize edildi. Lipit peroksidasyon (LPO), indirgenmiş glutatyon (GSH), siyalik asit (SA) düzeyleri ile glutatyon-S-transferaz (GST), katalaz, süperoksit dismutaz (SOD) ve doku faktörü (TF) aktiviteleri belirlendi. Bulgular: Amiodaron verilen grupta GSH, CAT, SOD aktiviteleri azalırken LPO ve SA seviyeleri arttı. Beyaz lahana, LPO ve SA miktarını azaltarak amiodaron zararını önledi. Ayrıca GSH düzeyleri ile SOD, CAT ve GST aktivitelerini artırarak antioksidan savunma sistemini güçlendirdi. Sonuç: Beyaz lahana tüketiminin, amiodaronun kalp dokusunda olumsuz etkilerini önlemek için alternatif bir yaklaşım olabileceği ileri sürülebilir., Objective: Amiodarone is used in the treatment of arrhythmia and has been shown to cause side effects in various organs. White cabbage (Brassica oleracea L. var. capitata) is a widely consumed vegetable. The effects of exposure to amiodarone and white cabbage consumption on heart tissue has not previously been investigated. We aimed to investigate the effects of white cabbage extract on amiodarone given to rats’ cardiac tissues. Material and Method: Rats were given white cabbage extract and amiodarone by gavage for seven days. Amiodarone was applied 1 hour after administration of the white cabbage extract. On the eighth day, the hearts of the animals were taken and homogenized. The levels of lipid peroxidation (LPO), reduced glutathione (GSH), sialic acid (SA) and activities of glutathione-S-transferase (GST), catalase, superoxide dismutase (SOD) and tissue factor (TF) were determined. Results: The GSH level, CAT and SOD activities decreased, while the LPO and SA levels increased in the amiodarone treated group. The White cabbage extract prevented any damage of the amiodarone by decreasing the amount of LPO and SA and also strengthened the antioxidant defence system by increasing the amount of the GSH and SOD activities and the CAT and GST activities. Conclusion: It is suggested that the consumption of white cabbage could be an alternative approach to the prevention of the adverse effects of amiodarone on heart tissue.
