Optical logic gates based on nonlinear optical property of material with ultrafast response speed and excellent computational processing power can break the performance bottleneck of electronic transistors. As one of the layered 2D materials, Ta2NiS5 exhibits high anisotropic mobility, exotic electrical response, and intriguing optical properties. Due to the low-symmetrical crystal structures, it possesses in-plane anisotropic physical properties. The optical absorption information of Ta2NiS5 is investigated by anisotropic linear absorption spectra, femtosecond laser intensity scanning (I-scan), and non-degenerate pump-probe technology. The I-scan results show a distinct maximum of ∼4.9 % saturable absorption (SA) and ∼4 % reverse saturable absorption (RSA) at different polarization directions of the incident laser. And, these unique nonlinear optical (NLO) properties originate from the anisotropic optical transition probability. Furthermore, the novel Ta2NiS5-based all-optical logic gates are proposed by manipulating the NLO absorption processes. And, the all-optical OR and NOR logic gates possess an ultrafast response speed approaching 1.7 THz. Meanwhile, an all-optical information transmission method with higher security and accuracy is achieved, which has promising potential to avoid the disclosure of information. This work provides a new path for designing versatile and novel optical applications based on Ta2NiS5 materials.

Abstract Background During the aging process, the number and functional activity of endothelial progenitor cells (EPCs) are impaired, leading to the unsatisfactory efficacy of transplantation. Previous studies demonstrated that Yiqihuoxue decoction (Ginseng-Sanqi-Chuanxiong, GSC) exerts anti-vascular aging effects. The purpose of this study is to evaluated the effects of GSC on D-galactose (D-gal)induced senescence and the underlying mechanisms. Methods The levels of cellular senescence-related markers P16, P21, P53, AMPK and p-AMPK were detected by Western blot analysis (WB). SA-β-gal staining was used to evaluate cell senescence. EPCs function was measured by CCK-8, Transwell cell migration and cell adhesion assay. The morphological changes of mitochondria were detected by confocal microscopy. The protein and mRNA expression of mitochondrial fusion fission Drp1, Mff, Fis1, Mfn1, Mfn2 and Opa1 in mitochondria were detect using WB and RT–qPCR. Mitochondrial membrane potential, mtROS and ATP of EPCs were measured using IF. H&E staining was used to observe the pathological changes and IMT of the aorta. The expressions of AGEs, MMP-2 and VEGF in aorta were measured using Immunohistochemical (IHC). The levels of SOD, MDA, NO and ET-1 in serum were detected by SOD, MDA and NO kits. Results In vitro, GSC ameliorated the senescence of EPCs induced by D-gal and reduced the expression of P16, P21 and P53. The mitochondrial morphology of EPCs was restored, the expression of mitochondrial Drp1, Mff and Fis1 protein was decreased, the levels of mtROS and ATP were decreased, and mitochondrial function was improved. Meanwhile, the expression of AMPK and p-AMPK increased. The improvement effects of GSC on aging and mitochondrial morphology and function were were hindered after adding AMPK inhibitor. In vivo, GSC improved EPCs efficiency, ameliorated aortic structural disorder and decreased IMT in aging mice. The serum SOD level increased and MDA level decreased, indicating the improvement of antioxidant capacity. Increased NO content and ET-1 content suggested improvement of vascular endothelial function. The changes observed in SOD and MMP-2 suggested a reduction in vascular stiffness and the degree of vascular damage. The decreased expression of P21 and P53 indicates the delay of vascular senescence.

Abstract Background DIR (Dirigent) proteins play important roles in the biosynthesis of lignin and lignans and are involved in various processes such as plant growth, development, and stress responses. However, there is less information about VvDIR proteins in grapevine (Vitis vinifera L). Results In this study, we used bioinformatics methods to identify members of the DIR gene family in grapevine and identified 18 VvDIR genes in grapevine. These genes were classified into 5 subfamilies based on phylogenetic analysis. In promoter analysis, various plant hormones, stress, and light-responsive cis-elements were detected. Expression profiling of all genes following Colletotrichum gloeosporioides infection and phytohormones (salicylic acid (SA) and jasmonic acid (JA)) application suggested significant upregulation of 17 and 6 VvDIR genes, respectively. Further, we overexpressed the VvDIR4 gene in Arabidopsis thaliana and grapes for functional analysis. Ectopic expression of VvDIR4 in A. thaliana and transient expression in grapes increased resistance against C. gloeosporioides and C. higginsianum, respectively. Phenotypic observations showed small disease lesions in transgenic plants. Further, the expression patterns of genes having presumed roles in SA and JA signaling pathways were also influenced. Lignin contents were measured before and after C. higginsianum infection; the transgenic A. thaliana lines showed higher lignin content than wild-type, and a significant increase was observed after C. higginsianum infection. Conclusions Based on the findings, we surmise that VvDIR4 is involved in hormonal and lignin synthesis pathways which regulate resistance against anthracnose. Our study provides novel insights into the function of VvDIR genes and new candidate genes for grapevine disease resistance breeding programs.

The use of weak passwords is a significant vulnerability in computer information system security, and this issue is also present in the digital applications of grain storage information systems. Simple hashing algorithms have obvious limitations when dealing with weak passwords. Based on the actual application of grain storage software systems, this paper proposed a method to enhance weak password protection by using a hashing algorithm as the foundation, combined with the introduction of salt values and high-order reflections for strengthening, along with multiple rehashing operations. Validation and analysis showed that this method increased the hashing strength of weak passwords, and effectively prevented dictionary attacks or rainbow table attacks, which could achieve the effective protection for the transmission and storage of weak passwords. Additionally, it had low complexity, and was easy to implement. It also had an average computational latency of approximately 2.5 ms, which was imperceptible to users. This method is suitable for the construction and management of digital grain storage information systems, providing robust security protection.

BackgroundPrimary malignant melanoma (MM) of skin threatens health, especially in the older population, causing a significant risk of early death. The purpose of this study was to establish a diagnostic nomogram to predict the early mortality risk in older patients with primary skin MM and to determine the independent risk factors of cancer-specific early death in such patients.MethodsThe Surveillance, Epidemiology and End Results (SEER) database provided the clinical and pathological characteristics of older patients with primary skin MM from 2000 to 2019. Initially, a 7:3 random assignment was used to place the recruited patients into training and validation cohorts. Then, the independent risk variables of cancer-specific early death in those individuals were determined using univariate and multivariate logistic regression analysis. Those patients’ diagnostic nomograms were constructed using the acquired independent risk variables. Ultimately, the performance of the newly created diagnostic nomogram was verified using calibration curves, receiver operating characteristic (ROC), and decision curve analysis (DCA) curves.ResultsIn this study, 2,615 patients in total were included. Age, histology, liver metastasis, tumor stage, surgery, therapy, and radiation were found to be independent risk factors following statistical analysis, with a special emphasis on early death in older patients with primary skin MM. A diagnostic nomogram for the cancer-specific early death risk was created and validated based on these variables. High agreement was reported between the expected and actual probabilities in the calibration curves. Area under the curves (AUC) of the novel created diagnostic nomogram was greater than that of each independent risk factor, with AUCs for the training and validation cohorts being 0.966 and 0.971, respectively. The nomogram had a high value for its applicability in clinical settings, according to DCA.ConclusionIn older patients with primary skin MM, the current study created a diagnostic nomogram to predict the probability of cancer-specific early death. Because of the nomograms’ good performance, physicians will be better able to identify older patients who are at a high risk of early death and treat them individually to increase their survival benefit.

Abstract Background To investigate factors associated with different reproductive outcomes in patients with Caesarean scar pregnancies (CSPs). Methods Between May 2017 and July 2022, 549 patients underwent ultrasound-guided uterine aspiration and laparoscopic scar repair at the Gynaecology Department of Hubei Maternal and Child Health Hospital. Ultrasound-guided uterine aspiration was performed in patients with type I and II CSPs, and laparoscopic scar repair was performed in patients with type III CSP. The reproductive outcomes of 100 patients with fertility needs were followed up and compared between the groups. Results Of 100 patients, 43% had live births (43/100), 19% had abortions (19/100), 38% had secondary infertility (38/100), 15% had recurrent CSPs (RCSPs) (15/100). The reproductive outcomes of patients with CSPs after surgical treatment were not correlated with age, body mass index, time of gestation, yields, abortions, Caesarean sections, length of hospital stay, weeks of menopause during treatment, maximum diameter of the gestational sac, thickness of the remaining muscle layer of the uterine scar, type of CSP, surgical method, uterine artery embolisation during treatment, major bleeding, or presence of uterine adhesions after surgery. Abstract The bioactivity of interferon-γ (IFN-γ) in cancer cells in the tumor microenvironment (TME) is not well understood in the current immunotherapy era. We found that IFN-γ has an immunosuppressive effect on colorectal cancer (CRC) cells. The tumor volume in immunocompetent mice was significantly increased after subcutaneous implantation of murine CRC cells followed by IFN-γ stimulation, and RNA sequencing showed high expression of B7 homologous protein 4 (B7H4) in these tumors. B7H4 promotes CRC cell growth by inhibiting the release of granzyme B (GzmB) from CD8+ T cells and accelerating apoptosis in CD8+ T cells. Furthermore, interferon regulatory factor 1 (IRF1), which binds to the B7H4 promoter, is positively associated with IFN-γ stimulation-induced expression of B7H4. The clinical outcome of patients with CRC was negatively related to the high expression of B7H4 in cancer cells or low expression of CD8 in the microenvironment. Therefore, B7H4 is a biomarker of poor prognosis in CRC patients, and interference with the IFN-γ/IRF1/B7H4 axis might be a novel immunotherapeutic method to restore the cytotoxic killing of CRC cells.

AbstractBackground Anxiety and depression are common comorbidities in idiopathic pulmonary fibrosis (IPF) that impair health-related quality of life. However, there is a lack of studies focusing on the mental disorder of IPF after antifibrotic treatment and their related predictive factors.Methods Patients with an initial diagnosis of IPF were enrolled. Data on demographics, lung function, Generalized Anxiety Disorder-7 (GAD-7) Scale, Patient Health Questionnaire 9 (PHQ-9), Patient Health Questionnaire-15 (PHQ-15), and St. George's Respiratory Questionnaire total score(SGRQ-T) were collected. Changes in anxiety, depression, somatic symptoms, and quality of life scores before and after nintedanib treatment were compared, and the related predictive factors were analyzed.Results A total of 56 patients with a first diagnosis of IPF were enrolled, with 42 and 35 patients suffering from anxiety and depression, respectively. The GAD-7, PHQ-9, PHQ-15, and SGRQ scores were higher in the anxiety and depression groups. Abstract Silver sulfide thin film, with excellent thermoelectric properties, is few reported due to the complex and time‐consuming high‐temperature or high‐pressure synthesis process. Here, a fast ionic conductor n‐type Ag2S film with good crystallinity and uniform density is prepared by sputtering metal Ag films of different thicknesses on glass and then reacting in S precursor solution at low temperature. At 450 K, β‐Ag2S films can be obtained and underwent a phase transition from α‐Ag2S monoclinic, which had a significant effect on their electrical and thermal properties. The grain size of Ag2S films increases with the increase of film thickness. Before and after the phase transition, the carrier concentration and mobility cause obvious changes in the electrical properties of Ag2S. The carrier concentration of body‐centered cubic phase β‐Ag2S is about three orders of magnitude higher than that of monoclinic phase α‐Ag2S, and the mobility is also 2–3 times that of the latter. Especially, after the phase transition, the conductivity of β‐Ag2S rises exponentially from the zero conductivity of α‐Ag2S and increases with the increase of temperature. The Ag2S film shows the highest figure of merit of 0.83 ± 0.30 at 600 K from the sample with ∼1600 nm thickness, which is the highest record among Ag2S‐based thermoelectric materials reported so far.

ObjectiveTo develop a model that integrates radiomics features and clinical factors to predict upper gastrointestinal bleeding (UGIB) in patients with decompensated cirrhosis.Methods104 decompensated cirrhosis patients with UGIB and 104 decompensated cirrhosis patients without UGIB were randomized according to a 7:3 ratio into a training cohort (n = 145) and a validation cohort (n = 63). Radiomics features of the abdominal skeletal muscle area (SMA) were extracted from the cross-sectional image at the largest level of the third lumbar vertebrae (L3) on the abdominal unenhanced multi-detector computer tomography (MDCT) images. Clinical-radiomics nomogram were constructed by combining a radiomics signature (Rad score) with clinical independent risk factors associated with UGIB. Nomogram performance was evaluated in calibration, discrimination, and clinical utility.ResultsThe radiomics signature was built using 11 features. Plasma prothrombin time (PT), sarcopenia, and Rad score were independent predictors of the risk of UGIB in patients with decompensated cirrhosis. The clinical-radiomics nomogram performed well in both the training cohort (AUC, 0.902; 95% CI, 0.850–0.954) and the validation cohort (AUC, 0.858; 95% CI, 0.762–0.953) compared with the clinical factor model and the radiomics model and displayed excellent calibration in the training cohort. Decision curve analysis (DCA) demonstrated that the predictive efficacy of the clinical-radiomics nomogram model was superior to that of the clinical and radiomics model.ConclusionClinical-radiomics nomogram that combines clinical factors and radiomics features has demonstrated favorable predictive effects in predicting the occurrence of UGIB in patients with decompensated cirrhosis. This helps in early diagnosis and treatment of the disease, warranting further exploration and research.

Immune cells are key players in acute tissue injury and inflammation, including acute kidney injury (AKI). Their development, differentiation, activation status, and functions are mediated by a variety of transcription factors, such as interferon regulatory factor 8 (IRF8) and IRF4. We speculated that IRF8 has a pathophysiologic impact on renal immune cells in AKI and found that IRF8 is highly expressed in blood type 1 conventional dendritic cells (cDC1s), monocytes, monocyte-derived dendritic cells (moDCs) and kidney biopsies from patients with AKI. In a mouse model of ischemia‒reperfusion injury (IRI)-induced AKI, Irf8−/− mice displayed increased tubular cell necrosis and worsened kidney dysfunction associated with the recruitment of a substantial amount of monocytes and neutrophils but defective renal infiltration of cDC1s and moDCs. Mechanistically, global Irf8 deficiency impaired moDC and cDC1 maturation and activation, as well as cDC1 proliferation, antigen uptake, and trafficking to lymphoid organs for T-cell priming in ischemic AKI. Moreover, compared with Irf8+/+ mice, Irf8−/− mice exhibited increased neutrophil recruitment and neutrophil extracellular trap (NET) formation following AKI. IRF8 primarily regulates cDC1 and indirectly neutrophil functions, and thereby protects mice from kidney injury and inflammation following IRI. Our results demonstrate that IRF8 plays a predominant immunoregulatory role in cDC1 function and therefore represents a potential therapeutic target in AKI.

Abstract Acute kidney injury (AKI) is a common kidney disease associated with excessive reactive oxygen species (ROS). Unfortunately, due to the low kidney targeting and undesired side effects, the existing antioxidant and anti-inflammatory drugs are unavailable for AKI management in clinic. Therefore, it’s essential to develop effective nanodrugs with high renal targeting and biocompatibility for AKI treatment. Herein, we reported a novel nanodrug for AKI treatment, utilizing poly(ursolic acid) (PUA) as a bioactive nanocarrier and resveratrol (RES) as a model drug. The PUA polymer was synthesized form ursolic acid with intrinsic antioxidant and anti-inflammatory activities, and successfully encapsulated RES through a nanoprecipitation method. Subsequently, we systemically investigated the therapeutic potential of RES-loaded PUA nanoparticles (PUA NPs@RES) against AKI. In vitro results demonstrated that PUA NPs@RES effectively scavenged ROS and provided substantial protection against H2O2-induced cellular damage. In vivo studies revealed that PUA NPs significantly improved drug accumulation in the kidneys and exhibited favorable biocompatibility. Furthermore, PUA NPs alone exhibited additional anti-inflammatory and antioxidant effect, synergistically enhancing therapeutic efficacy in AKI mouse models when combined with RES. Overall, our study successfully developed an effective nanodrug using self-therapeutic nanocarriers, presenting a promising option for the treatment of AKI. Graphical abstract

Impact of hardware impairment on the performance of intelligent reflecting surface (IRS) aided downlink two-user short-packet communication (SPC) systems using non-orthogonal multiple access over Rician channels was analyzed.Phase adjustments at the IRS were based on statistical channel state information.Firstly, the analytical expressions for average block error rates of two users were derived by using central limit theorem, moment matching method, SPC related theory, and Gaussian-Chebyshev quadrature method.Secondly, to minimize the transmission latency under the reliability constraints of two users, the optimal power allocation scheme for the considered system was given by using the proposed bisection search algorithm and one-dimensional search algorithm.Finally, simulation results verified the correctness of the theoretical analysis and showed that, compared to long-packet communication, hardware impairments have a greater impact on SPC.

Abstract Background NRG1 fusions are rare oncogenic drivers in solid tumors, and the incidence of NRG1 fusions in non-small cell lung cancer (NSCLC) was 0.26%. It is essential to explore potential therapeutic strategies and efficacy predictors for NRG1 fusion-positive cancers. Case presentation We report an advanced lung adenocarcinoma patient harboring a novel NPTN-NRG1 fusion identified by RNA-based next-generation sequencing (NGS), which was not detected by DNA-based NGS at initial diagnosis. Transcriptomics data of the tissue biopsy showed NRG1α isoform accounted for 30% of total NRG1 reads, and NRG1β isoform was undetectable. The patient received afatinib as fourth-line treatment and received a progression-free survival (PFS) of 14 months. Conclusions This report supports afatinib can provide potential benefit for NRG1 fusion patients, and RNA-based NGS is an accurate and cost-effective strategy for fusion detection and isoform identification.

Pimpinella candolleana is known as Miao ethnic herbal medicine in Guizhou for the treatment of icteric hepatitis, acute cholecystitis and other diseases. To investigate the chemical constituents of P. candolleana and their anti-inflammatory activities, the chemical constituents from the 70% ethanol extract of P. candolleana were separated by silica gel, Sephadex LH-20, Toyopearl HW-40F, Toyopearl HW-40C, ODS and other column chromatography technologies, and their structures were elucidated by extensive spectroscopic analysis such as nuclear magnetic resonance (NMR) and mass spectrum (MS). The inflammatory cell model, built by LPS-induced RAW264.7 macrophage cells, was used to evaluate the anti-inflammatory activity. The results were as follows: (1) Twenty compounds from P. candolleana were isolated and identified of including vanillin (1), sesamin (2), 2-methyl-2-hydroxy-5-methoxy berz (d) hydrofuran-3-one (3), procatechin (4), 1,5-dihydroxy-2,3-dimethoxyxanthone (5), isorhamnetin (6), kaempferol (7), 8-hydroxy-2-methylchromone (8), luteolin (9), quercetin (10), 1-O-β-D-glucopyranosyl-(2S,3S,4R,8E)-2-［(2′R)-2′-hydroxypalmitoylamino］-8-octadecene-1,3,4-triol (11), isorhamnetin-3-O-β-D-galactopyranoside (12), isoquercitrin (13), norswertianolin (14), luteolin-6-C-α-L-arabinoside (15), kaempferol-3-O-β-D-galactopyranoside (16), kaempferol-7-O-β-D-glucopyranoside (17), luteolin-7-O-β-D-glucopyranoside (18), isovitexin (19), rutin (20). Vascular aging is an independent risk factor for age-related diseases and a specific type of organic aging. Endothelial progenitor cells (EPCs), a type of bone marrow stem cell, has been linked to vascular aging. The purpose of this study is to investigate if Ginseng-Sanqi-Chuanxiong (GSC) extract, a traditional Chinese medicine, can delay aortic aging in mice by enhancing the performance and aging of EPCs in vivo and to analyze the potential mechanisms through a d-Galactose (D-gal)-induced vascular aging model in mice. Our study revealed that GSC extracts not only enhanced the aortic structure, endothelial function, oxidative stress levels, and aging in mice, but also enhanced the proliferation, migration, adhesion, and secretion of EPCs in vivo, while reducing the expression of p53, p21, and p16. To conclude, GSC can delay vascular senescence by enhancing the function and aging of EPCs, which could be linked to a decrease in p16 and p53/p21 signaling. Consequently, utilizing GSC extracts to enhance the function and senescence of autologous EPCs may present a novel avenue for enhancing autologous stem cells in alleviating senescence.

Background: Differentiating adrenal adenomas from metastases poses a significant challenge, particularly in patients with a history of extra-adrenal malignancy. This study investigates the performance of three-phase computed tomography (CT) based robust federal learning algorithm and traditional deep learning for distinguishing metastases from benign adrenal lesions. Material and methods: This retrospective analysis includes 1187 instances who underwent three-phase CT scans between January 2008 and March 2021, comprising 720 benign lesions and 467 metastases. Utilizing the three-phase CT images, both a Robust Federal Learning Signature (RFLS) and a traditional Deep Learning Signature (DLS) were constructed using the Least Absolute Shrinkage and Selection Operator (LASSO) logistic regression. Their diagnostic capabilities were subsequently validated and compared using metrics such as the Areas Under the Receiver Operating Curve (AUCs), Net Reclassification Improvement (NRI), and Decision Curve Analysis (DCA). Objective This research aimed to assess the levels of cognitive function and its contributing factors among individuals experiencing cancer pain (CP) in mainland China.Design A descriptive, cross-sectional study.Setting The investigation was undertaken within three tertiary oncology hospitals.Participants We included 220 hospitalised individuals who reported experiencing cancer-related pain and consented to complete the research questionnaires.Outcome measures The collected data encompassed sociodemographic and clinical variables, augmented by results from validated questionnaires. Cognitive impairment (CI) was evaluated using the Functional Assessment of Cancer Therapy-Cognitive (FACT-Cog) scale, with scores ranging from 0 to 148. Sleep quality, depression and anxiety were assessed through the Pittsburgh Sleep Quality Index, the Patient Health Questionnaire-9 and the Generalised Anxiety Disorder-7, respectively. A binary logistic regression model was used to identify factors associated with CI in individuals with CP.Results Of the 225 individuals approached, 220 (97.8%) participated in the study. The mean FACT-Cog score for those with CP was 101.29 (SD=25.24; range=25–148). The prevalence of CI among these individuals was 35.90%. Sleep quality was rated below medium in 45% of participants with CP. More than moderate pain was reported by 28.2%, with 64.6% experiencing depression and 38.6% experiencing anxiety. Increased odds of developing CI were observed in those with CP (OR 1.422, 95% CI 1.129 to 1.841), depression (OR 1.119, 95% CI 1.029 to 1.2117), anxiety (OR 1.107, 95% CI 1.005 to 1.220), advancing age (OR 1.042, 95% CI 1.013 to 1.073), poor sleep quality (OR 1.126, 95% CI 1.013 to 1.252) and a history of smoking (OR 3.811, 95% CI 1.668 to 8.707).Conclusions CI associated with CP is notably prevalent in China. Those older, with a smoking history, inadequate sleep, more severe pain, depression and anxiety, have a heightened risk of CI. Consequently, interventions need to be personalised, addressing these key determinants.

Background Ferroptosis, which is caused by an iron-dependent accumulation of lipid hydroperoxides, is a type of cell death linked to diabetic kidney disease (DKD). Previous research has shown that fatty acid binding protein 4 (FABP4) is involved in the regulation of ferroptosis in diabetic retinopathy. The present study was constructed to explore the role of FABP4 in the regulation of ferroptosis in DKD. Methods We first detected the expression of FABP4 and proteins related to ferroptosis in renal biopsies of patients with DKD. Then, we used a FABP4 inhibitor and small interfering RNA to investigate the role of FABP4 in ferroptosis induced by high glucose in human renal proximal tubular epithelial (HG-HK2) cells. Results In kidney biopsies of DKD patients, the expression of FABP4 was elevated, whereas carnitine palmitoyltransferase-1A (CP-T1A), glutathione peroxidase 4, ferritin heavy chain, and ferritin light chain showed reduced expression. In HG-HK2 cells, the induction of ferroptosis was accompanied by an increase in FABP4. Inhibition of FABP4 in HG-HK2 cells changed the redox state, sup-pressing the production of reactive oxygen species, ferrous iron (Fe2+), and malondialdehyde, increasing superoxide dismutase, and reversing ferroptosis-associated mitochondrial damage. The inhibition of FABP4 also increased the expression of CPT1A, reversed lipid deposition, and restored impaired fatty acid β-oxidation. In addition, the inhibition of CPT1A could induce ferroptosis in HK2 cells. Conclusion Our results suggest that FABP4 mediates ferroptosis in HG-HK2 cells by inhibiting fatty acid β-oxidation.

Abstract Deep‐Learning‐based Fault Localisation (DLFL) leverages deep neural networks to learn the relationship between statement behaviour and program failures, showing promising results. However, since DLFL uses program failures as labels to conduct supervised learning, a labelled dataset is a requisite of applying DLFL. A failure is detected by comparing program output with a test oracle which is the standard answer for the given input. The problem is, test oracles are often difficult, or even impossible to acquire in real life, and that has severely restricted the application of DLFL since we have only unlabelled datasets in most cases. Thus, MetaFL: Metamorphic Fault Localisation Using Weakly Supervised Deep Learning is proposed, to provide a weakly supervised learning solution for DLFL. Instead of using test oracles, MetaFL uses metamorphic relations to prescribe expected behaviour of a program, and defines labels of metamorphic testing groups by verifying integrity in each group of test cases. Hence, a coarse‐grained labelled dataset can be built from the originally unlabelled one, with which DLFL can work now, utilising a weakly supervised learning paradigm. The experiments show that MetaFL yields a performance comparable to plain DLFL under ideal condition (i.e. the labels of datasets are available). MetaFL successfully extends the methodology of DLFL from supervised learning to weakly supervised learning, and a fully labelled dataset is no longer mandatory for applying DLFL.

Aiming at the problem of low accuracy of high voltage line loss prediction, a line loss prediction model is proposed based on improved BP neural network and Equalization optimizer (EO) algorithm. Firstly, in order to improve the optimization ability of EO algorithm, a variety of chaotic mapping relations is used to initialize the population to increase the population diversity, then the global search ability could be improved. At the same time, the EO algorithm is improved by using the natural selection probability jump strategy, so that the model could jump out of the local optimization according to the probability and converge to the global optimal solution. Secondly, the improved EO algorithm is used to optimize the weight and bias of BP neural network, and the prediction effect of BP neural network for high voltage line loss is improved. Finally, the experimental results show that the proposed line loss prediction model has the highest prediction accuracy compared with regression model, BP neural network model, simulated annealing optimized BP neural network model and EO optimized BP neural network model.

Ethanol as a renewable fuel has been applied in fuel vehicles (FVs), and it is promising in hybrid electric vehicles (HEVs). This work aims to investigate the emission characteristics of ethanol applied in both FVs and plug-in hybrid electric vehicles (PHEVs). The paper conducted a real-road test of an internal combustion FV and PHEV, respectively, based on the world light vehicle test cycle (WLTC) by using gasoline and regular gasoline under different temperature conditions. The use of E10 and E20 in FVs has been effective in reducing the conventional emissions of the vehicles. At 23 °C, E10 and E20 reduced the conventional emissions including carbon monoxide (CO), total hydrocarbon compound (THC), non-methane hydrocarbon compound (NMHC), particulate matter (PM), and particulate number (PN) by 15.40–31.11% and 11.00–44.13% respectively. At 6 °C, E10 and E20 reduced conventional emissions including THC, CO, and PM by 2.15–8.61% and 11.02–13.34%, respectively. However, nitrogen oxide (NOX) emissions increased to varying degrees. The reduction trend of non-conventional emissions including methane (CH4), nitrous oxide (N2O), and carbon dioxide (CO2) from FVs fueled with E10 and E20 is not significant for vehicles. Overall, the emission reduction effect of E20 is better than that of E10, and the emission reduction effect of ethanol gasoline on vehicle emissions is reduced at low temperatures. Lower ambient temperatures increase vehicle emissions in the low-speed segment but decrease vehicle emissions in the ultra-high-speed segment. HEV emissions of THC, CO, PN, and PM are reduced by 25.28%, 12.72%, 77.34%, and 64.59%, respectively, for E20 compared to gasoline, and the use of E20 in HEVs contributes to the reduction of overall vehicle emissions.

Background: Yuquan Pill (YQW) is a modern concentrated pill preparation of six herbs, namely, Ge Gen (Pueraria lobata Ohwi), Di huang (Rehmannia glutinosa Libosch.), Tian Huafen (Trichosanthes kirilowii Maxim.), Mai Dong (Ophiopogon japonicus (L. f.) Ker Gawl.), Wu Weizi (Schisandra chinensis (Turcz.) Baill.) and Gan Cao (Glycyrrhiza uralensis Fisch.). It is extensively used to treat type 2 diabetes-related glucose and lipid metabolism disorders. But what’s the pharmacodynamic substance and how it works in the treatment of Type 2 diabetes mellitus (T2DM) are still unclear.Purpose: The purpose of this study is to determine the likely pharmacological components and molecular mechanism of YQW’s intervention on T2DM by combining serum pharmacochemistry, network analysis and transcriptomics.Methods: The efficacy and prototypical components of blood entry were determined after oral administration of YQW aqueous solution to T2DM rats induced by high-fat feed and low-dose streptozotocin (STZ), and the key targets and pathways for these compounds to intervene in T2DM rats were predicted and integrated using network analysis and transcriptomics techniques.Results: In diabetic rats, YQW can lower TG, CHO, NO, and MDA levels (p < 0.05) while increasing HDL-C levels (p < 0.01), and protecting the liver and kidney. 22 prototype components (including puerarin, daidzein, 3′-methoxypuerarin, and liquiritigenin, among others) were found in the serum of rats after oral administration of YQW for 90 min, which might be used as a possible important ingredient for YQW to intervene in T2DM rats. 538 YQW pharmacodynamic components-related targets and 1,667 disease-related targets were projected through the PharmMapper database, with 217 common targets between the two, all of which were engaged in regulating PI3K-Akt, MAPK, Ras and FoxO signal pathway. Finally, the mRNA expression profiles of liver tissues from rats in the control, model, and YQW groups were investigated using high-throughput mRNA sequencing technology. YQW can regulate the abnormal expression of 89 differential genes in a disease state, including 28 genes with abnormally high expression and 61 genes with abnormally low expression. Five common genes (Kit, Ppard, Ppara, Fabp4, and Tymp) and two extensively used regulatory pathways (PI3K-Akt and MAPK signaling pathways) were revealed by the integrated transcriptomics and network analysis study.Conclusion: The mechanism of YQW’s intervention in T2DM rats could be linked to 22 important components like puerarin, daidzein, and glycyrrhetinic acid further activating PI3K-Akt and MAPK signaling pathways by regulating key targets Kit, Ppard, Ppara, Fabp4, and Tymp, and thus improving lipid metabolism disorder, oxidative stress, and inflammation levels in T2DM rats. On the topic, more research into the pharmacological ingredient foundation and mechanism of YQW intervention in T2DM rats can be done.

In recent days, the green synthesized nanomagnetic biocomposites have been evolved with tremendous potential as the future catalysts. This has encouraged us to design and synthesis of a novel Cu NPs fabricated potato starch functionalized magnetic nanomaterial (Fe3O4@starch/Cu nanocomposite). The prepared nanocomposite were characterized using advanced analytical techniques like FT-IR, FESEM, TEM, EDX, elemental mapping and ICP-OES. The biogenic synthesized Fe3O4@starch/Cu nanocomposite was explored in the anti-ovarian cancer investigations against ovarian cancer cell lines (PA-1, Caov-3, SW 626, and SK-OV-3). The material exhibited significant cytotoxicities against the ovarian cancer cell lines. However, it was considerably inactive against the normal HUVEC cell line. The antioxidant potential of Fe3O4@starch/Cu nanocomposite was also investigated by DPPH method. The Fe3O4@starch/Cu nanocomposite revealed the significant antioxidant potentials according to the IC50 value.

BackgroundKinesin is a molecular motor for transporting “goods” within cells and plays a key role in many types of tumors. The multi-angle study of kinesin at the pan-cancer level is conducive to understanding its role in tumorigenesis and development and clinical treatment potential.MethodsWe evaluated the expression of KIF genes, performed differential analysis by using the R package limma, and explored the pan-cancer prognosis of KIF genes by univariate Cox regression analysis. To evaluate the pan-cancer role of KIF genes as a whole, we defined the KIFscore with the help of gene set variation analysis (GSVA) and explored the KIFscores across normal tissues, tumor cell lines, and 33 tumor types in TCGA. Next, we used spearman correlation analysis to extensively study the correlation between the KIFscore and tumor prognosis and be-tween the KIFscore and clinical indicators. We also identified the relationship between the KIFscore and genomic variation and immune molecular signatures by multiplatform analysis. Finally, we identified the key genes in clear cell renal cell carcinoma (ccRCC) through machine learning algorithms and verified the candidate genes by CCK8, wound healing assay, Transwell assay, and flow cytometry.ResultsIn most cancers, KIFscores are high and they act as a risk factor for cancer. The KIFscore was significantly associated with copy number variation (CNV), tumor mutation burden (TMB), immune subtypes, DNA repair deficiency, and tumor stemness indexes. Moreover, in almost all cancer species, the KIFscore was positively correlated with T cell CD4+ TH2, the common lymphoid pro-genitor, and the T cell follicular helper. In addition, it was negatively correlated with CXCL16, CCL14, TNFSF13, and TNFRSF14 and positively correlated with ULBP1, MICB, and CD276. Machine learning helped us to identify four hub-genes in ccRCC. The suitable gene, KIF14, is highly expressed in ccRCC and promotes tumor cell proliferation, migration, and invasion.ConclusionOur study shows that the KIF genes play an important pan-cancer role and may become a potential new target for a variety of tumor treatments in the future. Furthermore, KIF14, a key molecule in the KIF genes, can provide a new idea for the ccRCC treatment.

Background: Osteosarcoma initially metastasing to bone only shows distinct biological features compared to osteosarcoma that firstly metastasizes to the lung, which suggests us underlying different genomic pathogenetic mechanism. Methods: We analyzed whole-exome sequencing (WES) data for 38 osteosarcoma with paired samples in different relapse patterns. We also sought to redefine disease subclassifications for osteosarcoma based on genetic alterations and correlate these genetic profiles with clinical treatment courses to elucidate potential evolving cladograms. Results: We investigated WES of 12/38 patients with high-grade osteosarcoma (31.6%) with initial bone metastasis (group A) and 26/38 (68.4%) with initial pulmonary metastasis (group B), of whom 15/38 (39.5%) had paired samples of primary lesions and metastatic lesions. We found that osteosarcoma in group A mainly carries single-nucleotide variations displaying higher tumor mutation burden and neoantigen load and more tertiary lymphoid structures, while those in group B mainly exhibits structural variants. High conservation of reported genetic sequencing over time in their evolving cladograms. Conclusions: Osteosarcoma with mainly single-nucleotide variations other than structural variants might exhibit biological behavior predisposing toward bone metastases as well as better immunogenicity in tumor microenvironment.

C-ros oncogene 1 (ROS1) fusion is a pathogenic driver gene in non-small cell lung cancer (NSCLC). Currently, clinical guidelines from the National Comprehensive Cancer Network (NCCN) have recommended molecular pathologic tests for patients with NSCLC, including the detection of the ROS1 gene. Crizotinib is a small molecule tyrosine kinase inhibitor of anaplastic lymphoma kinase (ALK), ROS1, and mesenchymal-epithelial transition (MET). In recent years, the efficacy of crizotinib in NSCLC patients with ROS1 fusion has been reported. Here, a 77-year-old woman was diagnosed with stage IVA lung adenocarcinoma harboring a novel low-density lipoprotein receptor (LDLR)-ROS1 fusion variant. This novel LDLR-ROS1 fusion was identified by targeted DNA next-generation sequencing (NGS) panel and then verified by RNA fusion panel based on amplicon sequencing. This patient benefited from subsequent crizotinib therapy and achieved progression-free survival of 15 months without significant toxic symptoms. Our case report recommended a promising targeted therapeutic option for patients with metastatic NSCLC with LDLR-ROS1 fusion and highlighted the importance of genetic testing for accurate treatment.

Maternal erythrocyte alloimmunization is one of the most important causes of fetal anemia. The standard treatment for anemic fetuses is intrauterine blood transfusion (IUT). However, IUT may have adverse effects, particularly before 20 weeks of gestation. In this report, two women who had previously had severely affected alloimmunized pregnancy developed high titers of anti-D antibodies before 20 weeks of gestation. Ultrasound Doppler showed severe fetal anemia, and intrauterine transfusion was expected to be unavoidable. To prolong pregnancy to a gestation in which intravascular IUT was possible, we used repeated double filtration plasmapheresis (DFPP) as a rescue therapy. The titers of IgG-D, IgG-A, and IgG-B decreased after DFPP treatment. One woman successfully prolonged pregnancy until 20 weeks of gestation. Subsequently, she underwent four cycles of IUTs and delivered at 30 weeks of gestation by emergency cesarean section due to fetal bradycardia during the fifth intrauterine transfusion. The other woman successfully delayed intrauterine transfusion until 26 weeks of gestation. The favorable results of the two patients indicate that DFPP may be an effective and safe treatment modality for RhD immunity in pregnant women. Moreover, DFPP is potentially helpful for reducing the occurrence of ABO hemolytic disease in neonates due to the clearance of IgG-A and IgG-B antibodies (e.g., O pregnant women harbored A/B/AB neonates). However, more clinical trials are needed to verify the results.

IntroductionDiabetic kidney disease (DKD) is a long-term complication of diabetes and causes renal microvascular disease. It is also one of the main causes of end-stage renal disease (ESRD), which has a complex pathophysiological process. Timely prevention and treatment are of great significance for delaying DKD. This study aimed to use bioinformatics analysis to find key diagnostic markers that could be possible therapeutic targets for DKD.MethodsWe downloaded DKD datasets from the Gene Expression Omnibus (GEO) database. Overexpression enrichment analysis (ORA) was used to explore the underlying biological processes in DKD. Algorithms such as WGCNA, LASSO, RF, and SVM_RFE were used to screen DKD diagnostic markers. The reliability and practicability of the the diagnostic model were evaluated by the calibration curve, ROC curve, and DCA curve. GSEA analysis and correlation analysis were used to explore the biological processes and significance of candidate markers. Finally, we constructed a mouse model of DKD and diabetes mellitus (DM), and we further verified the reliability of the markers through experiments such as PCR, immunohistochemistry, renal pathological staining, and ELISA.ResultsBiological processes, such as immune activation, T-cell activation, and cell adhesion were found to be enriched in DKD. Based on differentially expressed oxidative stress and inflammatory response-related genes (DEOIGs), we divided DKD patients into C1 and C2 subtypes. Four potential diagnostic markers for DKD, including tenascin C, peroxidasin, tissue inhibitor metalloproteinases 1, and tropomyosin (TNC, PXDN, TIMP1, and TPM1, respectively) were identified using multiple bioinformatics analyses. Further enrichment analysis found that four diagnostic markers were closely related to various immune cells and played an important role in the immune microenvironment of DKD. In addition, the results of the mouse experiment were consistent with the bioinformatics analysis, further confirming the reliability of the four markers.ConclusionIn conclusion, we identified four reliable and potential diagnostic markers through a comprehensive and systematic bioinformatics analysis and experimental validation, which could serve as potential therapeutic targets for DKD. We performed a preliminary examination of the biological processes involved in DKD pathogenesis and provide a novel idea for DKD diagnosis and treatment.

High-pressure gas direct injection (DI) technology benefits engines with high efficiency and clean emissions, and the gas jet process causes crucial effects especially inside an mm-size space. This study presents an investigation on the high-pressure methane jet characteristics from a single-hole injector by analysing jet performance parameters including jet impact force, gas jet impulse, and jet mass flow rate. The results show that the methane jet exhibited a two-zone behaviour along the jet direction in the spatial dimension induced by high-speed jet flow from the nozzle: zone 1 near the nozzle—the jet impact force and jet impulse increased consistently except for a fluctuation due to shock wave effects induced by the sonic jet and no entrainment occurs, and zone II farther away from the nozzle—the jet impact force and jet impulse became stable when the shock wave effects became weak and the jet impulse was conserved with a linear conservation boundary. The Mach disk height was exactly the turning point of two zones. Moreover, the methane jet parameters, such as the methane jet mass flow rate, jet initial jet impact force, jet impulse, and Reynolds number had a monotonous and linearly increasing correlation with injection pressure.

Bag-of-Tasks (BoT) applications consisting of multiple tasks are widely used in various fields.Different from the traditional deadline constraint in scheduling problems,a due date constraint allows the BoT applications to finish more than a predetermined due date,but would result in the tardiness penalty.In this case,in order to reduce the total cost,the efficient harmony search (EHS) algorithm is proposed to optimize the scheduling tasks in hybrid clouds.The algorithm obtains the initial task sequence by random search and fine tuning.By improving the harmony search steps and the way of generating new harmony memory,a large number of high-quality harmony can be obtained in one search process,which greatly improves the efficiency of harmony search and speeds up the convergence speed of the algorithm.Through continuous iteration,the global optimal solution is obtained,that is to say,BoT application scheduling scheme has the lowest total cost.Experimental results show that compared with other algorithms,the proposed algorithm has significant improvement in performance,which can effectively reduce the total cost of scheduling BoT applications.

Two new oxidovanadium(V) complexes, [VOL1(aha)]DMF (1) and [VOL2(mat)] (2), where L1 and L2 are the dianionic form of N'-(4-bromo-2-hydroxybenzylidene)-3-methyl-4-nitrobenzohydrazide and N'-(3,5-dibromo-2-hydroxybenzylidene)pivalohydrazide, respectively, and aha and mat are the monoanionic form of acetohydroxamic acid and maltol, respectively, have been synthesized and structurally characterized by physico-chemical methods and single crystal X-ray determination. X-ray analysis indicates that the V atoms in the complexes are in octahedral coordination. Crystal structures of the complexes are stabilized by hydrogen bonds. The catalytic property for epoxidation of styrene by the complexes was evaluated.

Selective laser melting (SLM) can effectively replace traditional processing methods to prepare parts with arbitrary complex shapes through layer-by-layer accumulation. However, SLM Ti-6Al-4V alloy typically exhibits low ductility and significant mechanical properties anisotropy due to the presence of acicular α′ martensite and columnar prior β grains. Post-heat treatment is frequently used to obtain superior mechanical properties by decomposing acicular α′ martensite into an equilibrium α + β phase. In this study, the microstructure and tensile properties of SLM Ti-6Al-4V alloy before and after various heat treatments were systematically investigated. The microstructure of the as-fabricated Ti-6Al-4V sample was composed of columnar prior β grains and acicular α′ martensite, which led to high strength (~1400 MPa) but low ductility (~5%) as well as significantly tensile anisotropy. The single heat treatment samples with lamellar α + β microstructure exhibited improved elongation to 6.8–13.1% with a sacrifice of strength of 100–200 MPa, while the tensile anisotropy was weakened. A trimodal microstructure was achieved through multi-step high-to-low-temperature (HLT) heat treatment, resulting in an excellent combination of strength (~1090 MPa) and ductility (~17%), while the tensile anisotropy was almost eliminated. The comprehensive mechanical properties of the HLT samples were superior to that of the conventional manufactured Ti-6Al-4V alloy.

We propose how to achieve synthetic PT $\mathcal{PT}$ symmetry in optomechanics without using any active medium. We find that harnessing the Stokes process in such a system can lead to the emergence of exceptional point (EP), i.e., the coalescing of both the eigenvalues and the eigenvectors of the system. By encircling the EP, both nonreciprocal optical amplification and chiral mode switching can be achieved. As a result, our synthetic PT $\mathcal{PT}$ -symmetric optomechanics works as a topological optomechanical amplifier. This provides a surprisingly simplified route to realize PT $\mathcal{PT}$ -symmetric optomechanics, indicating that a wide range of EP devices can be created and utilized for various applications such as topological optical engineering and nanomechanical processing or sensing.

The control of hot electron (HE) and thermal effects induced by plasmonic nanostructures has recently attracted considerable attention. When illuminated by light with different circular polarization states, the circular dichroism signal of molecules adsorbed by plasmonic chiral nanostructures can control HE and thermal effects. These effects have the potential to enhance reaction rates and to change selectivity patterns in photothermal catalysis. Here, we propose an aluminum L-shaped chiral nanostructure system in which HE and thermal effects can be controlled in different regions of the nanostructure by changing the chirality of the excitation light. A large difference of 12.75% in the HE effect but a virtually identical thermal effect can be achieved in different regions of the nanostructure by selecting the appropriate probed region, while a large thermal effect difference of 65.67% but a virtually identical HE effect can be achieved in one region of the nanostructure by changing the polarization state of the excitation light. In addition, the HE and thermal chiral selectivity effects of double L-shaped nanostructures are investigated as these structures can be more easily controlled during asymmetric chiral growth and crystallization. This work combined with plasmonic chirality is beneficial for quantifying HE and thermal effects in photochemical reactions and provides theoretical support for designing catalysts and optimizing plasmonic platforms. Additionally, the local controllability of HE and thermal effects plays an essential role in high-resolution photochemical reactions, especially in single-molecule photochemical reactions.

Psittacosis accounts for 1–2 % of community-acquired pneumonia. In recent years, reports of psittacosis are increasing. Most reported cases of psittacosis are sporadic. Here, we report a familial cluster of five patients infected with Chlamydophila in a northwest Chinese region and share our diagnosis and treatment experience. The epidemiological characteristics, clinical features, laboratory examinations of family cluster psittacosis were collected and analyzed. We closely followed up all the family members and analyzed their clinical outcome. Five cases of family clustered pneumonia were mainly characterized by fever, cough and fatigue. mNGS rapidly identified the infecting agent as Chlamydophila in case 1 followed by RT-PCR analysis. A newly purchased pet parrot, which had diarrhea, was probably the primary source of infection. The main change of inflammation index in five patients was the decrease of lymphocyte counts. Chest CT showed peripheral or subpleural involvement of patchy high-density shadows with bronchial ventilation signs and blurred edges, mostly unilateral lesions. Five cases were completely cured with moxifloxacin and azithromycin. Our findings suggest that a familial cluster of Chlamydophila infection maybe caused by contact with sick pet parrot or human to human transmission in one close family. For this community-acquired pneumonia, epidemiological characteristics and use of mNGS is very important for improving accuracy in the early diagnosis.

Herein, a kind of three-dimensional hollow Mg-Ca-Al hydrotalcite-like compounds (HTLCs) microsphere was prepared by self-assembly of hydrotalcite-like nanosheets. Mg-Ca-Al HTLCs microsphere (MS) has large specific surface area and large pore size, and the modification of KF·2H2O increases numerous alkaline active sites on the surface of the catalyst. The prepared catalyst has excellent catalytic effect for the production of biodiesel by transesterification. Under the optimal conditions of the catalyst addition amount which accounts for 2% of the weight of oil, the biodiesel yield of the best catalyst is as high as 92% within 30 minutes. This article also provides a paradigm of a rational structural design for regulating the morphology of HTLCs.

Rapeseed (Brassica napus L.) is an important oilseed crop globally. However, its growth and production are significantly influenced by cold stress. To reveal the protective role of exogenous melatonin (MEL) in cold tolerance, rapeseed seedlings were pretreated with different concentrations of MEL before cold stress. The results indicated that the survival rate was increased significantly by the MEL pretreatment under cold stress. Seedlings pretreated with 0.01 g L−1 MEL were all survived and were used to analyze the physiological characteristics and the expression level of various genes related to cold tolerance. Under cold stress, exogenous MEL significantly increased the contents of proline, soluble sugar, and soluble protein; while the malondialdehyde content was decreased by exogenous MEL under cold stress. On the other hand, the activities of antioxidant defense enzymes such as catalase, peroxidase, and superoxide dismutase were also significantly enhanced. The results also showed that MEL treatment significantly upregulated the expression of Cu-SOD, COR6.6 (cold-regulated), COR15, and CBFs (C-repeat binding factor) genes under cold stress. It was suggested exogenous MEL improved the content of osmotic regulatory substances to maintain the balance of cellular osmotic potential under cold stress and improved the scavenging capacity of reactive oxygen species by strengthening the activity of antioxidant enzymes and the cold-related genes expression.

This study was conducted to evaluate the effect of flavor on reproductive performance and fecal microbiota of sows during late gestation and lactation. A total of 20 healthy Yorkshire sows were fed a corn-soybean basal diet unsupplemented or supplemented with 0.1% flavor compound from d 90 of gestation to 25 d post-farrowing, and then the piglets were weaned. The reproductive performance and the fecal microbiota of sows were analyzed. Compared with the controls, flavor supplementation in maternal diets increased (P 0.05). BackgroundAlong with uric acid, which is the primary driving factor of gout, downstream inflammatory mediators have been shown to be involved in the pathogenesis of gouty arthritis flares. Extracorporeal haemadsorption is an emerging technology for the treatment of dysregulated inflammatory states by effectively removing cytokines from the bloodstream. Whether haemadsorption was effective in refractory gout flares has not been reported in the literature.Case summaryWe report the case of a 52-year-old male who presented with refractory gouty arthropathy for 30 years. His uric acid levels were poorly controlled due to poor diet and treatment compliance. Tophi were found to have precipitated in multiple joints and subcutaneous tissue. In the last 2 years, his incidents of gouty flares had become more frequent, and resistant to the medications, including colchicine, allopurinol, febuxostat, glucocorticoids, and NSAID analgesics. He had experienced a triad of chills, high fever and arthritis for the past 2 weeks. Therefore, he took 2 mg colchicine twice daily for 2 weeks with no improvement in his pain. Proinflammatory cytokines, such as interleukin-6 (IL-6) and tumour necrosis factor-α (TNF-α), were found to be remarkably elevated. Given that conventional treatment was unsuccessful, we tried to employ plasma adsorption (PA) to remove inflammatory cytokines. After 4 sessions, symptoms, such as fever, joint swelling and pain, were greatly improved. Meanwhile, the levels of proinflammatory factors such as IL-6 and TNF-α were found to be decreased, while the anti-inflammatory factor IL-10 remained the same during the course. He was followed up for 8 months and arthritis have flared up twice in response to a high-purine diet.ConclusionOur study suggests that plasma adsorption (PA) may be a promising and feasible treatment for refractory gout when conventional treatments are unsatisfactory or contraindicated. However, more clinical trials are needed to verify the efficacy and safety of the treatment.Core tipChronic gouty arthritis flares are refractory to conventional treatment, such as uric acid-lowering drugs and NSAID analgesics. Due to the involvement of inflammatory cytokines, plasma adsorption was employed to alleviate flares by removing inflammatory mediators. Herein, we report a 52-year-old male who presented with refractory gouty arthropathy for 30 years, manifested with a triad of chills, high fever and arthritis. He underwent several sessions of plasma adsorption, and his symptoms soon improved, along with a drop in inflammatory mediators. We conclude that plasma adsorption may be a promising and feasible treatment for refractory gout when conventional treatments are unsatisfactory or contraindicated.

Abstract Aimed at the problem that the quality of the formation of aero-rotor blades with complex shaped structures is difficult to predict due to the coupling of each physical field in the process of electrochemical machining, a mathematical model of electrochemical machining characterized by the conductivity of the multi-physics field is established firstly by studying the coupling action mechanism of the flow field and temperature field on electric field conductivity in the machining process. Then, utilizing the Runge–Kutta method, the relationship between the conductivity and the machine path is analysed; Finally, based on this relationship, the multi-physics field electrochemical machining erosion model is compared with the traditional single electric field electrochemical machining erosion model. The results show that the error of the multi-physical field coupling prediction model proposed in this study is in the range of 1.27–2.35%, while the accuracy of the single electric field prediction model is in the range of 4.35–5.88%. The theoretical value of the multi-physics field coupling simulation is closer to the measured value of the test and can accurately simulate the actual electrochemical machining process, which can provide a theoretical basis for the design of cathode tools and parameter optimization in the actual processing process and is of great significance to improve the quality and efficiency of the electrochemical machining of aero-rotor blades.

Root system architecture (RSA) and tiller are important agronomic traits. However, the mechanisms of the IGT family genes regulate RSA and tiller development in different rice varieties remain unclear. In this study, we demonstrated that 38 rice varieties obtained from Yuanyang Hani’s terraced fields with different RSA and could be classified into six groups based on the ratio of root length and width. We found a positive correlation between RSA (including root width, length, and area) and tiller number in most of rice varieties. Furthermore, the IGT family genes Deeper Rooting 1 (DRO1), LAZY1, TAC1, and qSOR1 showed different expression patterns when rice grown under irrigation and drought conditions. Moreover, the qSOR1 gene had higher levels in the roots and tillers, and accompanied with higher levels of PIN1b gene in roots when rice grown under drought environmental condition. DRO1 gene had two single nucleotide polymorphisms (SNPs) in the exon 3 sequences and showed different expression patterns in the roots and tillers of the 38 rice varieties. Overexpression of DRO1 with a deletion of exon 5 caused shorter root length, less lateral roots and lower levels of LAZY1, TAC1, and qSOR1. Further protein interaction network, microRNA targeting and co-expression analysis showed that DRO1 plays a critical role in the root and tiller development associated with auxin transport. These data suggest that the RSA and tiller development are regulated by the IGT family genes in an intricate network way, which is tightly related to rice genetic background in rice adapting to different environmental conditions.

A new bromido-coordinated mononuclear copper(II) complex [Cu(HL1)Br2] (1), and a new mononuclear copper(II) complex [CuL2(HL2)]ClO4·0.5H2O (2), with the hydrazone ligands 4-tert-butyl-N’-(1-(pyridin-2-yl)ethylidene)benzohydrazide (HL1) and 4-bromo-N’-(pyridin-2-ylmethylene)benzohydrazide (HL2), have been synthesized and structurally characterized by physico-chemical methods and single crystal X-ray determination. X-ray analysis indicates that the Cu atom in complex 1 is in distorted square pyramidal coordination, and that in complex 2 is in octahedral coordination. The catalytic property for epoxidation of styrene by the complexes was evaluated.

Speech feature learning is the key to speech mental health recognition. Deep feature learning can automatically extract the speech features but suffers from the small sample problem. The traditional feature extract method is effective, but cannot find the inter-feature structure to generate the new high-quality features. This paper proposes an embedded hybrid feature deep sparse stacked autoencoder ensemble method to solve this problem. Firstly, the speech features are extracted based on prior knowledge and called original features. Secondly, the original features are embedded into the deep network (Sparse Stacked Autoencoder) to filter the output of the hidden layer, to enhance the complementarity between the deep features and the original features. Thirdly, the L1 regularized feature selection mechanism is designed to reduce the hybrid feature set formed by the combination of deep features and original features. Finally, a manifold projection classifier ensemble is designed to enhance the stability of classification. Besides, this paper for the first time proposes a speech collection scheme for mental health recognition. We construct a large-scale Chinese mental health speech database for verification of the proposed algorithm of mental health. In the experimental section, the proposed algorithm is verified and compared with the representative related algorithms. The experimental results show that the proposed algorithm has better classification accuracy than the other representative algorithms. The proposed method combines the advantages of deep feature learning and traditional feature extraction methods more efficiently to solve the small sample problem.

Abstract Background High concentrate (HC) diet-induced oxidative stress causes gut epithelial damages associated with apoptosis. Selenium (Se) being an integral component of glutathione peroxidase (GSH-Px) plays an important role in antioxidant defense system. Therefore, increasing dietary Se level would alleviate HC diet-induced injuries in gut mucosa. The present study investigated eighteen cross-bred goats, randomly divided into three groups (n = 6/group) fed either low concentrate (LC, roughage: concentrate ratio 65:35), high concentrate (HC, 35:65) or HC plus Se (HC-SY) diets for 10 weeks. Se was supplemented at the dose rate of 0.5 mg Se kg− 1 diet in the form of selenium yeast. The background Se level in HC and LC diets were 0.15 and 0.035 mg.kg− 1 diet, respectively. The Se at the dose of 0.115 mg.kg− 1 diet was added in LC diet to make its concentration equivalent to HC diet and with the supplementation of 0.5 mg Se kg− 1, the goats in group HC-SY received total Se by 0.65 mg.kg− 1 diet.
