Background/Objective: While it has been established that physical activity (PA) is key to promote overall health and well-being, insufficient physical activity among children and adolescents is a global problem, including Singapore. It is important to understand the local PA landscape among children and adolescents to decrease surveillance gaps and identify areas for improvement in promoting PA. The present article provides an overview of the development of the 2022 Active Healthy Kids Singapore Report Card and the results, as well as underscore limitations and gaps in the available evidence related to PA among children and adolescents in Singapore. Methods: Following the Global Matrix 4.0, the available data between July 2010 to July 2020 was synthesized for all 10 indicators by the work group and reviewed by a panel of experts. Data sources included published scientific articles, government and non-government reports, national surveys, and unpublished data from on-going research studies. Where possible, grades were informed by nationally representative surveys or large-scale longitudinal studies. Results: The grades assigned were: Overall Physical Activity (C-), Organized Sport and Physical Activity (B-), Active Play (C-), Active Transportation (C), Sedentary Behaviours (C-), Physical Fitness (Incomplete), Family and Peers (C-), School (Incomplete), Community and Environment (A+), Government (B). Conclusion: This is the first comprehensive evaluation of PA among children and adolescents in Singapore. It provides baseline grades valuable for future comparison. It also illustrates gaps in the existing evidence which can inform future surveillance, facilitate international comparisons and enable global efforts in promoting physical activity.

Abstract Background The aim of this study was to determine the expression and function of heterogeneous nuclear ribonucleoprotein R (HNRNPR) in esophageal carcinoma (ESCA), the correlation between its expression and 18F-fluorodeoxyglucose (18F-FDG) positron emission tomography/computerized tomography scan (PET/CT)-related parameters. We also investigated whether 18F-FDG PET/CT can be used to predict the expression of HNRNPR in ESCA. Methods We analyzed patients with ESCA who underwent 18F-FDG PET/CT before surgery, and their tissues were stained with HNRNPR IHC. The associated parameters were derived using the 18F-FDG PET imaging data, and the correlation with the IHC score was evaluated. The Oncomine, TCGA, and GEO datasets were used to investigate HNRNPR expression in the pan- and esophageal cancers, as well as its relationship with N6-methyladenosine (m6A) modification and glycolysis. The R software, LinkedOmics, GeneMANIA, and StringOnline tools were used to perform GO/KEGG, GGI, and PPI analyses on the HNRNPR. Results HNRNPR is highly expressed in the majority of pan-cancers, including ESCA, and is associated with BMI, weight, and history of reflux in patients with ESCA. Coherence is intrinsically related to projective measurement. When the fixed projective measurement involves higher-rank projectors, the coherence resource is referred to as block coherence, which comes from the superposition of orthogonal subspaces. Here, we establish a set of quantitative relations for the interconversion between block coherence and multipartite entanglement under the framework of the block-incoherent operations. It is found that the converted multipartite entanglement is upper bounded by the initial block coherence of single-party system. Moreover, the generated multipartite entanglement can be transferred to its subsystems and restored to block coherence of the initial single-party system by means of local block-incoherent operations and classical communication. In addition, when only the coarse-grained quantum operations are accessible for the ancillary subsystems, we further demonstrate that a lossless resource interconversion is still realizable, and give a concrete example in three four-level systems. Our results provide a versatile approach to utilize different quantum resources in a cyclic fashion.

We investigate quantum phase transitions (QPTs) in various spin chain systems using the multipartite entanglement measure $\tau_\mathrm{SEF}$ based on the monogamy inequality of squared entanglement of formation (EOF). Our results demonstrate that $\tau_\mathrm{SEF}$ is more effective and reliable than bipartite entanglement or bipartite correlation measures such as EOF, von Neumann entropy, and quantum discord in characterizing QPTs. $\tau_\mathrm{SEF}$ not only detects critical points that may go unnoticed by other detectors but also avoids the issue of singularity at non-critical points encountered by other measures. Furthermore, by applying $\tau_\mathrm{SEF}$ , we have obtained the phase diagram for the XY spin chain with three and four interactions and discovered a new quantum phase.

E-mode hybrid ferroelectric charge storage gate (FEG) GaN HEMTs have shown promising performances for future power GaN device applications. The FEG-HEMT demonstrates a combination of ferroelectric polarization and charge trapping process in the ferro-charge-storage gate stack, leading to a positive threshold voltage shift for E-mode operations. In this work, FEG-HEMTs with various Hf-based and Zr-based charge trapping layers are systematically studied. FEG-HEMT which employed nitrogen incorporated HfO2 (HfON) as the charge trapping layer shows an E-mode operation with the highest $V_{\mathrm{ th}}$ (+2.3 V) after initialization. Moreover, the gate leakage of the HfON sample was further reduced due to the nitrogen incorporation, leading to a more complete charging process during initialization. The $V_{\mathrm{ th}}$ instability is also addressed and investigated. The FEG-HEMT with HfON as the charge trapping layer showed a negligible $V_{\mathrm{ th}}$ hysteresis (−43mV) and the highest $V_{\mathrm{ th}}$ stability in both the PBTI (positive bias threshold voltage instability) and NBTI (negative bias threshold voltage instability) test measurements.

Since tectorigenin has been reported to possess anti-inflammation, redox balance restoration, and anti-apoptosis properties, we determine to unravel whether tectorigenin has potential in alleviating spinal cord injury (SCI). Herein, PC12 cells were induced by lipopolysaccharide (LPS) to establish in vitro SCI models. The cell viability and apoptosis were detected through cell counting kit-8 and flow cytometry assays. The caspase-3/8/9 content was measured by colorimetric method. Western blot was conducted to quantify the expressions of cleaved caspse-3/8/9, IGFBP6, TLR4, IκBα, p-IκBα, RELA proto-oncogene, p65, and p-p65. Enzyme-linked immunosorbent assay and real-time quantitative polymerase chain reaction were carried out to quantitate expressions of IGFBP6, interleukin-1β (IL-1β), interleukin-6 (IL-6), and tumor necrosis factor-α (TNF-α). SwissTargetPrediction and GSE21497 database were utilized to predict the potential therapeutic targets of tectorigenin. Comparison of IGFBP6 expression in SCI tissues and normal tissues was analyzed by GEO2R. Our study found that LPS induced the declined cell viability, elevated cell apoptosis, upregulation of caspase-3/8/9, cleaved caspase-3/8/9, IL-1β, IL-6, TNF-α, IGFBP6, and TLR4, and the activation of IκBα and p65 in PC12 cells. Tectorigenin reversed the above effects of LPS. IGFBP6 was predicted to be the potential therapeutic target of tectorigenin and was overexpressed in SCI tissues. Notably, IGFBP6 overexpression offset the effects of tectorigenin on PC12 cells. In conclusion, tectorigenin could alleviate the LPS-induced apoptosis, inflammation, and activation of NF-κB signaling in SCI cell models via inhibiting IGFBP6.

BackgroundThe targeted therapy for lung cancer relies on prognostic genes and requires further research. No research has been conducted to determine the effect of endothelin-converting enzyme 2 (ECE2) in lung cancer.MethodsWe analyzed the expression of ECE2 in lung adenocarcinoma (LUAD) and normal adjacent tissues and its relationship with clinicopathological characteristics from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus database (GEO). Immunohistochemical staining was used to further validate the findings. GO/KEGG enrichment analysis and gene set enrichment analysis (GSEA) of ECE2 co-expression were performed using R software. Data from TIMER, the GEPIA database, and TCGA were analyzed to determine the relationship between ECE2 expression and LUAD immune infiltration. To investigate the relationship between ECE2 expression levels and LUAD m6A modification, TCGA data and GEO data were analyzed.ResultsECE2 is highly expressed in various cancers including LUAD. ECE2 showed high accuracy in distinguishing tumor and normal sample results. The expression level of ECE2 in LUAD was significantly correlated with tumor stage and prognosis. GO/KEGG enrichment analysis showed that ECE2 was closely related to mitochondrial gene expression, ATPase activity and cell cycle. GSEA analysis showed that ECE2-related differential gene enrichment pathways were related to mitotic cell cycle, MYC pathway, PLK1 pathway, DNA methylation pathway, HIF1A pathway and Oxidative stress-induced cellular senescence. Analysis of the TIMER, GEPIA database, and TCGA datasets showed that ECE2 expression levels were significantly negatively correlated with B cells, CD4+ cells, M2 macrophages, neutrophils, and dendritic cells. TCGA and GEO datasets showed that ECE2 was significantly associated with m6A modification-related genes HNRNPC, IGF2BP1, IGF2BP3 and RBM1.ConclusionECE2 is associated with m6A modification and immune infiltration and is a prognostic biomarker in LUAD.

The EarlyâMiddle Ordovician peri-Gondwana and Baltica acritarch provinces are easily recognizable, illustrating a clear provincialism of global phytoplankton. However, acritarch assemblages have been reported to become increasingly similar towards the Late Ordovician, revealing a general cosmopolitanism, although, based on more recent studies, their assemblages from Laurentia and Baltica are reported to be rather different from those from peri-Gondwana. In this decade, new acritarch assemblages reported from South America, the Baltic region, Iran and Siberia, as well as the new material from South China and Tarim, provide additional data that help to understand the palaeobiogeographical distribution and evolution of phytoplankton during the Late Ordovician in more detail. The binary dataset was compiled based on published information from about 120 localities in South China, the Middle East, North Africa, South America, India, Bohemia, Avalonia, Laurentia, Baltica, North China, Tarim and Siberia. This dataset was used for Cluster Analysis (CA), Nonmetric Multidimensional Scaling (NMDS), and Network Analysis (NA) to understand acritarch palaeogeography in the Late Ordovician. Re-evaluation of the global palaeobiogeographical distribution of acritarchs revealed their clear provincialism also during the Late Ordovician, particularly in the Katian. The Late Ordovician acritarch provinces were related to global climatic zones. The highly diverse acritarch assemblage normally existed around 30° in the Southern Hemisphere, which is similar to the latitudinal diversity gradient model proposed by Zacaï et al. (2021) for the Late Ordovician. The total diversity of acritarchs was highest and their palaeobiogeographical differentiation strongest in the Katian. The higher acritarch diversity recorded in this time interval may have resulted from their clearly developed provincialism. Acritarch diversity decreased rapidly at the end of the Ordovician, evidently due to the Late Ordovician Mass Extinction (LOME). The relative proportion of endemic taxa in the assemblages in different regions was very high during the main part of the Late Ordovician. However, as a consequence of the LOME, the relative proportion of cosmopolitan taxa increased in the Hirnantian and became higher than in the Sandbian and Katian. Acritarch assemblages from different plates (regions) reacted to the LOME differently and exhibited various diversity trends.

BackgroundSolute carrier family 17 member 9 (SLC17A9) encodes a member of a family of transmembrane proteins that are involved in the transport of small molecules. SLC17A9 is involved in the occurrence and development of various cancers, but its biological role in liver hepatocellular carcinoma (LIHC) is unclear.MethodsThe expression level of SLC17A9 was assessed using The Cancer Genome Atlas (TCGA) database and immunohistochemistry of tumor tissues and adjacent normal liver tissues. The receiver operating characteristic (ROC) and R software package performed diagnosis and prognosis. Gene Ontology/Kyoto Encyclopedia of Genes and Genomes functional enrichment and co-expression of SLC17A9, gene–gene interaction (GGI), and protein–protein interaction (PPI) networks were performed using R, GeneMANIA, and STRING. Western blot, real-time quantitative PCR (RT-qPCR), immunofluorescence, colony formation, wound scratch assay, ATP production assays, and high connotation were applied to determine the effect of SLC17A9 knockdown on HEPG2 (hepatocellular liver carcinoma) cells. TIMER, GEPIA, and TCGA analyzed the relationship between SLC17A9 expression and immune cells, m6A modification, and ferroptosis.ResultsSLC17A9 expression in LIHC tissues was higher than in normal liver tissues (p < 0.001), and SLC17A9 was related to sex, DSS (disease-specific survival), and PFI (progression-free interval) (p = 0.015, 0.006, and 0.023). SLC17A9 expression has diagnostic (AUC: 0.812; CI: 0.770–0.854) and prognostic potential (p = 0.015) in LIHC. Gene Ontology/Kyoto Encyclopedia of Genes and Genomes (GO/KEGG) functional enrichment analysis showed that SLC17A9 was closely related to neuronal cell body, presynapse, axonogenesis, PI3K/Akt signaling pathway. GGI showed that SLC17A9 was closely related to MYO5A. PPI showed that SLC17A9 was closely related to SLC18A3. SLC17A9 silencing inhibited HepG2 cells proliferation, migration, colony formation, and reduced their ATP level. SLC17A9 expression level was related to immune cells: B cells (r = 0.094, P = 8.06E-02), CD4+ T cells (r = 0.184, P = 5.95E-04), and macrophages (r = 0.137, P = 1.15E-02); m6A modification: HNRNPC (r = 0.220, p < 0.001), METTL3 (r = 0.180, p < 0.001), and WTAP (r = 0.130, p = 0.009); and ferroptosis: HSPA5 (r = 0.240, p < 0.001), SLC7A11 (r = 0.180, p < 0.001), and FANCD2 (r = 0.280, p < 0.001).ConclusionOur data show that SLC17A9 may influence LIHC progression. SLC17A9 expression correlates with tumor immune infiltration, m6A modification, and ferroptosis in LIHC and may have diagnostic and prognostic value in LIHC.

Organic carbonyls are emerging as promising electrodes for potassium-ion batteries (PIBs) owing to their remarkable structural adaptability. Nevertheless, most small-molecule carbonyls still face significant challenges related to solubility issues and sluggish kinetics. Herein, a simple but efficient "two in one" strategy is employed, i.e., perylenetetracarboxylic diimide is firstly linked by low-weight ethylene to form a dimer (labelled as PTCDI2) to enhance the intramolecular interactions, followed by tuning through amorphization via electrochemical activation aiming to improve the charge transfer. The resulting PTCDI2 anode delivers high reversible capacity (310 mA h g−1 at 0.5 A g−1), outstanding rate capability (160 mA h g−1 at 10 A g−1) and remarkable long-term cycle stability (over 2000 cycles with a retention of 75%). Ex situ characterization methods demonstrate that improved electrochemical performance of PIBs with PTCDI2 anodes benefits from the following aspects: (1) the ethylene bridge strongly enhances the intramolecular interactions between two PTCDI monomers, thereby effectively overcoming the dissolution issue and (2) the transformation of PTCDI2 from crystalline to amorphous during cycling provides multiple pathways for ions, resulting in accelerated ion dynamics. The concept of incorporating ethylene-bridged structures and inducing amorphization in carbonyl anodes offers an effective avenue to tailor high-performance organic electrode materials for PIBs. Background: YTHDF1 is highly expressed in multiple tumors and affects tumor progression. However, there are only a few comprehensive studies on the analysis of YTHDF1 in esophageal cancer.Methods: We analyzed YTHDF1 expression in pan-cancer by comparing both the GEPIA and TCGA cohorts, and further verified the differences in YTHDF1 expression between the ESCA and normal groups by the GEO ESCA cohort and in vitro experiments. The correlation of YTHDF1 expression and the clinical characteristics of ESCA patients was analyzed using the TCGA ESCA clinical data. The GO and KEGG enrichment analyses of the YTHDF1 coexpressed genes were completed by bioinformatics analysis, and the GGI and PPI were constructed for the YTHDF1, respectively. The relationship between YTHDF1 expression and the infiltration of ESCA immune cells was analyzed by using the TIMER database and the TCGA ESCA cohort. The relationships between YTHDF1 expression levels and glycolysis and ferroptosis-related genes were analyzed using the TCGA and GEPIA ESCA cohorts. Finally, the ceRNA network that may be involved in YTHDF1 in ESCA was predicted and constructed through a variety of databases.Results: YTHDF1 was overexpressed in various cancers, and in vitro experiments confirmed that YTHDF1 expression was higher in ESCA samples than in normal samples. The expression of YTHDF1 has some accuracy in predicting the tumor outcome. Expression of YTHDF1 was significantly associated with multiple clinical features in ESCA patients. GO and KEGG enrichment analyses indicated that YTHDF1 coexpressed genes involved multiple biological functions. There is a potential association between YTHDF1 expression and multiple immune cell infiltration, glycolysis, and ferroptosis-related genes in ESCA. YTHDF1 may be involved in multiple ceRNA regulatory networks in ESCA, including PAXIP1-AS1/hsa-miR-376c-3p/YTHDF1 axis, THUMPD3-AS1/hsa-miR-655-3p/YTHDF1 axis, and SNHG20/hsa-miR-655-3p/YTHDF1 axis, respectively.Conclusion: YTHDF1 can serve as a biomarker of ESCA, related to the immune cell infiltration of ESCA, regulation of glycolysis and ferroptosis, and the ceRNA regulatory network.
