The ca. 2.97 to 2.80 Ga Witwatersrand Supergroup, South Africa, represents the oldest intracontinental sedimentary basin of the Kaapvaal craton. Two volcanic units occur in this supergroup: the widespread Crown Formation lavas in the marine shale-dominated West Rand Group and the more geographically restricted Bird Member lavas, intercalated with fluvial to fluvio-deltaic sandstone and conglomerate of the Central Rand Group. These units remain poorly studied as they are rarely exposed and generally deeply weathered when cropping out. We report whole-rock major and trace elements, Hf and Nd-isotope whole-rock analyses of the lavas from core samples drilled in the south of the Witwatersrand basin and underground samples from the Evander Goldfield in the northeast. In the studied areas, both the Crown Formation and Bird Member are composed of two units of lava separated by sandstone. Whereas all the Crown Formation samples show a similar geochemical composition, the upper and lower volcanic units of the Bird Member present clear differences. However, the primitive mantle-normalized incompatible trace element concentrations of all Crown Formation and Bird Member samples show variously enriched patterns and marked negative Nb and Ta anomalies relative to Th and La. Despite the convergent geodynamic setting of the Witwatersrand Supergroup suggested by the literature, the Crown Formation and Bird Member are probably not related to subduction-related magmatism but more to decompression melting. Overall, the combined trace element and Sm-Nd isotopic data indicate melts from slightly to moderately depleted sources that were variably contaminated with crustal material. Greater contamination, followed by differentiation in different magma chambers, can explain the difference between the two signatures of the Bird Member. Finally, despite previous proposals for stratigraphically correlating the Witwatersrand Supergroup to the Mozaan Group of the Pongola Supergroup, their volcanic units are overall geochemically distinct.

Numerous Mesoproterozoic alkaline intrusions belonging to the Pilanesberg Alkaline Province are present within the Transvaal sub-basin of the Kaapvaal Craton. The Pilanesberg Complex is the best-known example; it represents one of the world’s largest alkaline complexes, and is associated with a northwest-southeast trending dyke swarm that extends from Botswana to the southwest of Johannesburg. This paper documents the results of a petrological and geochemical study of a thin mafic sill (here referred to as an alkaline igneous body, AIB), which intrudes the ca. 2 200 Ma Silverton Formation close to the southernmost part of the Pilanesberg dyke swarm. The AIB has only been observed in cores from a borehole drilled close to Carletonville. It is hypocrystalline, containing randomly oriented elongated skeletal kaersutite crystals and 6 to 8 mm varioles mainly composed of radially oriented acicular plagioclase. These two textures are related to undercooling, probably linked to the limited thickness (70 cm) of the AIB coupled with a probable shallow emplacement depth. Ar-Ar dating of the kaersutite gives an age of ca. 1 400 Ma, similar to the age of Pilanesberg Complex. However, the AIB is an alkaline basaltic andesite and is thus notably less differentiated than the Pilanesberg Complex and some of its associated dykes, such as the Maanhaarrand dyke, for which we provide whole-rock geochemical data. Literature data indicate that the Pilanesberg dyke swarm also contains mafic hypabyssal rocks suggesting a link between the dyke swarm and the AIB. The AIB is characterized by strongly negative εNd and εHf, that cannot be related to crustal contamination, as shown by positive Ti and P anomalies, and the absence of negative Nb-Ta anomalies in mantle-normalised trace element diagrams. The AIB magma is interpreted to have been derived from a long-lived enriched, probably lithospheric mantle reservoir. The AIB thus provides important information on the magma source of the Pilanesberg Alkaline Province.

The aim of this study was to evaluate the production of chicken anaemia virus VP3 protein in different Escherichia coli strains and to address the diagnostic application of purified E. coli-expressed VP3 protein for the detection of chicken anaemia virus (CAV) infection and the development of an ELISA kit. Three E. coli strains, BL21, BL21 codonplus RP and BL21 pLysS, each harbouring a VP3 protein expressing plasmid, were investigated after induction to produce recombinant VP3 protein. After isopropyl-β-D-thiogalactoside (IPTG) induction, VP3 protein was successfully expressed in all three E. coli strains. The BL21 pLysS strain gave the best performance in terms of protein productivity and growth profile. In addition, the optimal culture temperature and IPTG concentration were found to be 0.25 mM and 20 °C, respectively. Using Ni-NTA-purified VP3 protein as an ELISA coating antigen, the purified VP3 was shown to be highly antigenic and able to discriminate sera from chickens infected with CAV from those that were uninfected during an evaluation of CAV infection serodiagnosis. A VP3-based ELISA demonstrated 100% (6/6 x 100%) specificity and sensitivities of 91.3% (21/23 x 100%) and 82.6% (19/23 x 100%) using cut-off values of the mean plus 2 SD and the mean plus 3 SD, respectively.

Taxonomic composition and emergence phenologies of Chironomidae inhabiting an intermittent stream in eastern Kansas were determined over a three year period using collections of surface-floating pupal exuviae. Duration of surface flows varied substantially over the period of study, ranging from 266 days in 1993-1994 to 215 days in 1994-1995 and 137 days in 1995-1996. The stream bed dried completely during summer and/or fall in all three years. Fifty-five species emerged from the stream during the study, with Orthocladiinae having the greatest species richness (26 taxa), followed by Chironomini (14), Tanytarsini (10) and Tanypodinae (5). No Diamesinae, Prodiamesinae, Podonominae nor Pseudochironomini were present. Annual emergence varied considerably with 29 species in 1993-1994, 49 species in 1994-1995 and only 6 species in 1995-1996. During the first two years the phenological patterns resembled those expected for small, cooler-water streams, with winter and early spring emergence dominated by Orthocladiinae. The peak in emergence of Orthocladiinae was followed by a small but distinct increase in Tanytarsini in early spring, then Chironomini became more common as water temperatures increased and surface discharge declined. The timing of emergence was strongly influenced by the date at which surface discharge began, and resulted in about a 2 month shift between 1993-1994 and 1994-1995. Averaged over the three years of the study, emergence was dominated by seven species: Corynoneura sp. 1, Corynoneura sp. 2, Orthocladius lapponicus, Parametriocnemus cf. lundbecki, Thienemanniella sp., Larsia decolorata, and Zavrelimyia sinuosa. The remaining species consisted of a mixture of species common in perennial streams plus (1) winter/early spring emerging species that likely persist in the stream in desiccation-resistant stages or in the hyporheic during periods of intermittency, (2) semiaquatic or semiterrestrial species, and (3) species of genera in which larvae are known to produce cocoons and diapause, or occur in moist sediments. We estimate that as many as 60 % of the species of Chironomidae are facultative or specialist species that characterize this intermittent stream.

Intravenous lidocaine has been widely used for suppressing the autonomic activation from tracheal intubation during induction of general anesthesia. Conventionally, researches of its effectiveness through assessment of heart rate and blood pressure changes obtained by common clinical methods result in the conclusions deduced of much controversy. Heart rate variability is a noninvasive measurement of autonomic regulation and is suitable for the study of this subject.36 ASA class I-II patients undergoing general anesthesia were divided into 3 groups. Besides induction agents, intravenous lidocaine was given 5 min before tracheal intubation in group A, 3 min before intubation in group B and nothing in group C. HRV spectral powers were measured at awake state, anesthetized state before tracheal intubation and anesthetized state after tracheal intubation by time frequency spectral analysis method and comparison was made between the three groups.The HRV spectral power in high frequency (HF) and mid-frequency (MF) power bands and their ratios (MF/HF) were not significantly different among the 3 groups during the 3 observation periods.There was no evidence to indicate the effectiveness of intravenous lidocaine on the autonomic regulation during tracheal intubation under the influence of induction agents used in general anesthesia.

A complementary DNA library was constructed from mRNA of rat liver induced by an initiating dose of a chemical carcinogen, diethylnitrosamine. Using a differential hybridization technique, a complementary DNA clone which is induced more than 10-fold by an acute single dose of diethylnitrosamine was identified. The DNA sequence of this clone was matched with rat microsomal epoxide hydrolase. This gene may be of great interest, since it was found to be highly expressed in neoplastic nodules and primary hepatocellular carcinomas induced by different carcinogenic regimes. The inducible high level expression of this gene becomes constitutive during the process of hepatocarcinogenesis. The gene was also found to be inducibly expressed during partial hepatectomy in a similar manner as a multidrug-resistant gene (mdr-I). No change in the transcriptional initiation site was observed in the gene expression between induced and uninduced rat livers. The 5' upstream region of the gene was characterized and some potential controlling elements for gene regulation, such as Sp-1, AP-2, and Hepatitis B virus enhancer, were found. Based on our own and published results, we hypothesize that the altered expression of this xenobiotic enzyme in nodules and cancer cells could be a result of constitutive internal stimuli which might be associated with cell growth.
