1. Genomic characteristics and clinical significance of CD56+ circulating tumor cells in small cell lung cancer

Abstract: Abstract Circulating tumor cells (CTC) have been studied in various solid tumors but clinical utility of CTC in small cell lung cancer (SCLC) remains unclear. The aim of the CTC-CPC study was to develop an EpCAM-independent CTC isolation method allowing isolation of a broader range of living CTC from SCLC and decipher their genomic and biological characteristics. CTC-CPC is a monocentric prospective non-interventional study including treatment-naïve newly diagnosed SCLC. CD56+ CTC were isolated from whole blood samples, at diagnosis and relapse after first-line treatment and submitted to whole-exome-sequencing (WES). Phenotypic study confirms tumor lineage and tumorigenic properties of isolated cells for the 4 patients analyzed with WES. WES of CD56+ CTC and matched tumor biopsy reveal genomic alteration frequently impaired in SCLC. At diagnosis CD56+ CTC were characterized by a high mutation load, a distinct mutational profile and a unique genomic signature, compared to match tumors biopsies. In addition to classical pathways altered in SCLC, we found new biological processes specifically affected in CD56+ CTC at diagnosis. High numeration of CD56+ CTC (> 7/ml) at diagnosis was associated with ES-SCLC. Comparing CD56+ CTC isolated at diagnosis and relapse, we identify differentially altered oncogenic pathways (e.g. DLL3 or MAPK pathway). We report a versatile method of CD56+ CTC detection in SCLC. Numeration of CD56+ CTC at diagnosis is correlated with disease extension. Isolated CD56+ CTC are tumorigenic and show a distinct mutational profile. We report a minimal gene set as a unique signature of CD56+ CTC and identify new affected biological pathways enriched in EpCAM-independent isolated CTC in SCLC.

Abstract: The expansion of the cerebral cortex is one of the most distinctive changes in the evolution of the human brain. Cortical expansion and related increases in cortical folding may have contributed to emergence of our capacities for high-order cognitive abilities. Molecular analysis of humans, archaic hominins, and non-human primates has allowed identification of chromosomal regions showing evolutionary changes at different points of our phylogenetic history. In this study, we assessed the contributions of genomic annotations spanning 30 million years to human sulcal morphology measured via MRI in more than 18,000 participants from the UK Biobank. We found that variation within brain-expressed human gained enhancers, regulatory genetic elements that emerged since our last common ancestor with Old World monkeys, explained more trait heritability than expected for the left and right calloso-marginal posterior fissures and the right central sulcus. Intriguingly, these are sulci that have been previously linked to the evolution of locomotion in primates and later on bipedalism in our hominin ancestors.

Abstract: Imaging genetic studies of large control cohorts such as UK Biobank enable to assess the range of normal variations in brain structures. Previous studies by our group have shown that the width of several cortical sulci is associated with a variant in the upstream region of KCNK2 gene even if this effect is corrected with age. Here we propose to analyze in a multivariate setup the associations between sets of genetic variants and multiple sulci widths. The genetic variants we consider are sets of SNPs of known phase called haplotypes, taken from the upstream region of KCNK2 gene. To the best of our knowledge, multivariate analysis in imaging genetics has never been used in haplotype studies. Our method was able to recover the expected association signal and uncover new associations between imaging data and genetic variants.

Abstract: Uncovering the genes that contribute to the variability in brain regions involved in language processing might shed light on the evolution of brain structures essential to the emergence of language in Homo sapiens. The superior temporal asymmetrical pit (STAP), which is not observed in chimpanzees, represents an ideal phenotype to investigate the genetic variations that support human communication. The depth of the left STAP was significantly associated with a DACT1 enhancer region in the UK Biobank British discovery sample (N=16,515). This association was replicated in the IMAGEN cohort (N=1,726) and the UK Biobank non-British validation sample (N=2,161). This genomic region was also associated to a lesser extent with the right STAP depth and the formation of sulcal interruptions, plis de passage, in the bilateral STAP but not with other structural brain MRI phenotypes, highlighting its specific expression in the superior temporal regions. Diffusion MRI emphasized an association with the fractional anisotropy of the left auditory fibers of the corpus callosum and with networks involved in linguistic processing in resting-state functional MRI. Finally, DACT1 is mainly expressed during the first two trimesters of pregnancy just before the superior temporal sulcus becomes visible and the first evidence of speech processing is observed in preterm neonates. Overall, this evidence demonstrates a significant, specific relationship between variants in this genomic region and the establishment of the superior temporal regions that support human communication. Significance statement The search for “language genes” has mainly focused on families with language disorders. Here, using a macroscopic marker of cerebral asymmetry observed only in humans, the STAP, we discovered a genetic variant that modulates many anatomical and functional features of the superior temporal sulcus, particularly in the left hemisphere, in the general population. Whole-brain analyses showed that this modulation is nearly limited to the superior temporal region. Finally, the gene effect is most significant during the first six months of gestation, decreasing just before the appearance of thalamocortical connectivity that enables external auditory inputs. Thus, we uncovered a genomic region that probably contributes to building the networks underlying human language.

Abstract: full-scale mechanical prototype line was deployed to a depth of 2500 m to test the leak tightness of the electronics containers and the pressure-resistant properties of an electromechanical cable under evaluation for use in the ANTARES deep-sea neutrino telescope. During a month-long immersion study, line parameter data were taken using miniature autonomous data loggers and shore-based optical time domain reflectometry. Details of the mechanical prototype line, the electromechanical cable and data acquisition are presented. Data taken during the immersion study revealed deficiencies in the pressure resistance of the electromechanical cable terminations at the entry points to the electronics containers. The improvements to the termination, which have been integrated into subsequent detection lines, are discussed. The line also allowed deep-sea acoustic measurements with a prototype hydrophone system. The technical setup of this system is described, and the first results of the data analysis are presented. 9. Infarctus rénal et splénique après consommation massive de cannabis et cocaïne chez un homme jeune

Abstract: Resume Le cannabis est la drogue la plus consommee dans le monde, particulierement chez les sujets jeunes. La cocaine arrive au troisieme rang. Quelques cas de la litterature rapportent l’association entre cannabis et cocaine et infarctus mesenterique et renal. Nous rapportons le cas d’un homme, âge de 24 ans, ayant presente un infarctus renal et splenique apres consommation massive de cannabis et de cocaine. Les deux evenements vasculaires survinrent sur des arteres saines et le bilan etiologique exhaustif se revela negatif. La responsabilite de l’association entre le cannabis et la cocaine fut retenue. Le cannabis et la cocaine sont associes aux accidents vasculaires par differents mecanismes parmi lesquels un probable phenomene thrombotique. La recherche de cannabis et de cocaine doit etre systematique chez tous patients jeunes admis pour un accident vasculaire quel que soit le territoire.

Abstract: Resume Les troubles du sommeil chez les patients porteurs de cancer ont fait l’objet de nombreuses etudes. L’evaluation de la qualite de sommeil a ete realisee par differents outils : questionnaires, actimetrie, polysomnographie. Les resultats des travaux publies montrent un sommeil fragmente et une vigilance diurne alteree. L’anxiete et la depression, souvent liees a l’annonce diagnostique et les symptomes induits par la maladie cancereuse (douleur, toux, dyspnee, signes fonctionnels urinaires, bouffees de chaleur…) semblent affecter la qualite de sommeil. Les programmes incluant une prise en charge cognitivo-comportementale pourraient etre benefiques aux patients.

Abstract: Resume Etat des connaissances Le pronostic des cancers bronchopulmonaires a petites cellules (CPC) en rechute est toujours reserve. Il n’y a pas de traitement de reference dans cette situation et une attention particuliere doit etre portee a la qualite de vie et aux consequences economiques. Methodes L’objectif de cette etude (essai GFPC0501), de phase II randomisee, est, chez des patients porteurs d’un CPC en progression apres une premiere ligne de chimiotherapie a base de platine, de comparer une polychimiotherapie orale (CCNU, Cyclophosphamide, etoposide) a un traitement intraveineux classique : cyclophosphamide, doxorubicine, vincristine (CAV) en terme de tolerance, d’efficacite (taux de reponses, survie mediane a un an et globale), de qualite de vie et de consommations en ressource de soins. Avec un schema de Briant and Day en deux temps le nombre total de patients a inclure est de 138 avec une analyse intermediaire a 38 patients inclus. Resultats attendus Cet essai permettra d’ameliorer les connaissances sur plusieurs aspects relatifs a la chimiotherapie de seconde ligne des patients traites pour cancer a petites cellules. Fin decembre 2006, 36 patients ont ete inclus par 16 centres. Les resultats de l’analyse intermediaire seront disponibles en juin 2007.

Abstract: Pemetrexed has recently been approved for use in combination with cisplatin as first-line chemotherapy for malignant pleural mesothelioma (MPM). Radiation therapy is frequently administered to the thoracic orifices and no data are available about the interactions between radiotherapy and pemetrexed. We report the first case of radiation recall dermatitis occurring after pemetrexed chemotherapy in a patient with MPM previously treated with radiation therapy to the thoracoscopy and drainage orifices. The patient received chemotherapy with pemetrexed and cisplatin 19 days after completion of chest wall radiation therapy delivering 21 gray in 3 days. Conventional premedication by folic acid and intramuscular administration of Vitamin B12 and prednisolone was correctly performed. Twelve days later, confluent erythematous and pruritus rash of the irradiated skin was observed. The toxicity grade of this lesion was evaluated at 2 according to the Acute Radiation Morbidity Scoring Criteria proposed by the Radiation Therapy Oncology Group. Pemetrexed challenge was performed without worsening of skin lesions. Three weeks later, skin cicatrisation was observed after a desquamative phase. Persistent hyperpigmentation was seen in the irradiated skin. Pemetrexed could also act as a radiosensitizing agent that should be used with care for several weeks after radiotherapy.

13. Traitement d’un kyste bronchogénique compressif par ponction sous tomodensitométrie

Abstract: Resume Introduction Les kystes bronchogeniques sont des tumeurs mediastinales benignes dont le traitement de reference est la resection chirurgicale complete. Nous rapportons l’observation d’une patiente âgee, presentant un kyste bronchogenique symptomatique non operable, traite par ponction-aspiration sous controle tomodensitometrique. Cas clinique Une femme de 92 ans etait admise pour une dyspnee inspiratoire associee a un stridor. L’interrogatoire rapportait la decouverte d’une masse mediastinale laterotracheale droite, 2 ans auparavant. L’examen tomodensitometrique et l’endoscopie bronchique revelaient une compression tracheale par un volumineux kyste bronchogenique. La chirurgie etait contre-indiquee en raison de l’âge et des antecedents cardiaques de la patiente. Une ponction du kyste sous controle tomodensitometrique a permis d’evacuer 250 ml de liquide et de lever rapidement les signes de compression tracheale. La patiente est restee asymptomatique pendant plusieurs mois. L’operation a ete repetee un an plus tard du fait d’une recidive de la compression. Conclusion Cette observation souligne l’interet potentiel de la ponction sous controle tomodensitometrique en tant qu’alternative a la chirurgie en cas de kyste bronchogenique compressif inoperable.

Abstract: [EN] The MOSCAB experiment (Materia OSCura A Bolle) uses the Geyser technique for dark matter search. The results of the first 0.5 kg mass prototype detector using superheated C3F8 liquid were very encouraging, achieving a 5 keV nuclear recoil threshold with high insensitivity to gamma radiation. Additionally, the technique seems to be easily scalable to higher masses for both in terms of complexity and costs, resulting in a very competitive technique for direct dark matter search, especially for the spin dependent case. Here, we report as well in the construction and commissioning of the big detector of 40 kg at the Milano-Bicocca University. The detector, the calibration tests and the evaluation of the background will be presented. Abstract: The ANTARES neutrino telescope is being constructed in the Mediterranean Sea. It consists of a large three-dimensional array of photo-multiplier tubes. The data acquisition system of the detector takes care of the digitisation of the photo-multiplier tube signals, data transport, data filtering, and data storage. The detector is operated using a control program interfaced with all elements. Abstract: ANTARES is a neutrino telescope being deployed in the Mediterranean Sea. It consists of a three dimensional array of photomultiplier tubes that can detect the Cherenkov light induced by charged particles produced in the interactions of neutrinos with the surrounding medium. High angular resolution can be achieved, in particular when a muon is produced, provided that the Cherenkov photons are detected with sufficient timing precision. Considerations of the intrinsic time uncertainties stemming from the transit time spread in the photomultiplier tubes and the mechanism of transmission of light in sea water lead to the conclusion that a relative time accuracy of the order of 0.5 ns is desirable. Accordingly, different time calibration systems have been developed for the ANTARES telescope. In this article, a system based on Optical Beacons, a set of external and well-controlled pulsed light sources located throughout the detector, is described. This calibration system takes into account the optical properties of sea water, which is used as the detection volume of the ANTARES telescope. Abstract: The ANTARES neutrino telescope, to be immersed depth in the Mediterranean Sea, will consist of a 3 dimensional matrix of 900 large area photomultiplier tubes housed in pressure resistant glass spheres. The selection of the optimal photomultiplier was a critical step for the project and required an intensive phase of tests and developments carried out in close collaboration with the main manufacturers worldwide. 19. [Renal and spleen infarction after massive consumption of cannabis and cocaine in a young man]

Abstract: Cannabis is the most widely consumed drug in the world, particularly among young subjects. Cocaine is the third leading illicit drug. Cases of renal infarction associated with combined consumption of cannabis and cocaine have been reported in the literature. We describe the case of a 24-year-old man who presented renal and spleen infarction after massive consumption of cannabis and cocaine. Both vascular events arose on healthy arteries. Etiological tests were negative leading to the conclusion that the events resulted from a toxic cause related to cannabis and cocaine consumption. Different mechanisms, potentially including thrombosis, might explain the association of cannabis and cocaine with vascular events. We suggest that a systematic search for cannabis and cocaine consumption among young victims of vascular disease might be useful.

20. CO2 Storage—Managing the Risk Associated With Well Leakage over Long Timescales

Abstract: One of the major challenges associated with CO2 geological storage is the performance of the confining system over long timescales. In particular, the occurrence of CO2 leakage through existing wells could not only defeat the purpose of storage but also badly affect human health or the environment. Indeed, cement degradation and casing corrosion in injection, production or abandoned wells can create preferential channels over time, allowing migration of CO2 from the reservoir to shallower formations (e.g. aquifers), and/or to the surface. In this paper, a risk-based approach is proposed for well integrity and confinement performance management. The approach, based on Performance and Risk Management methodology (P&R™), serves as a decision support tool. The major steps are (i) identifying the system and the sources of degradation through characterization and system analysis; (ii) quantifying their criticity through modeling, in terms of probability and severity, and (iii) establishing a risk mitigation plan. This methodology is based on experience in material aging and risk assessment of complex systems, like nuclear structures, where probabilistic simulations are performed. It accounts for all stakes involved in well integrity management and enables the full integration of uncertainties as part of risk estimation. The methodology presented here greatly improves common approaches based on "Features, Events, and Processes" as it quantifies risk levels. It provides useful and reliable tools to support decisions for well integrity management strategies or emergency plans. To that purpose, mitigation actions such as characterization/inspection, remediation (workover), design improvement or monitoring are valued based on a cost/benefits ratio. Moreover, updating risk assessment with incoming data allows for an evolving vision of risk levels to optimize interventions in time. This approach is successfully applied, leading to recommendations for safer and more efficient design, maintenance, and monitoring strategies.

Abstract: International audience; Geological repositories subject to the injection of large amounts of anthropogenic carbon dioxide will undergo chemical and mechanical instabilities for which there are currently little experimental data. This study reports on experiments where low and high P co 2 (8 MPa) aqueous fluids were injected into natural rock samples. The experiments were performed in flow-through triaxial cells, where the vertical and confining stresses, temperature, and pressure and composition of the fluid were separately controlled and monitored. The axial vertical strains of two limestones and one sandstone were continuously measured during separate experiments for several months, with a strain rate resolution of 10 À11 s À1. Fluids exiting the triaxial cells were continuously collected and their compositions analyzed. The high P co 2 fluids induced an increase in strain rates of the limestones by up to a factor of 5, compared to the low P co 2 fluids. Injection of high P co 2 fluids into the sandstone resulted in deformation rates one order of magnitude smaller than the limestones. The creep accelerating effect of high P co 2 fluids with respect to the limestones was mainly due to the acidification of the injected fluids, resulting in a significant increase in solubility and reaction kinetics of calcite. Compared to the limestones, the much weaker response of the sandstone was due to the much lower solubility and reactivity of quartz in high P co 2 fluids. In general, all samples showed a positive correlation between fluid flow rate and strain rate. X-ray tomography results revealed significant increases in porosity at the inlet portion of each core; the porosity increases were dependent on the original lithological structure and composition. The overall deformation of the samples is interpreted in terms of simultaneous dissolution reactions in pore spaces and intergranular pressure solution creep.

22. [Treatment of a compressive bronchogenic cyst by computed tomography-guided needle aspiration]

Abstract: Bronchogenic cysts are benign tumours of the posterior or middle mediastinum. Treatment usually consists of complete surgical resection. We report the case of an elderly woman presenting with an inoperable bronchogenic cyst causing tracheal compression which was treated successfully using computed-tomography (CT)-guided needle aspiration.A 92 years old woman was admitted for inspiratory dyspnoea associated with stridor. She reported the incidental discovery of a right paratracheal mass, 2 years previously. Thoracic CT scan and bronchoscopy revealed a large mediastinal bronchogenic cyst causing tracheal compression. Surgery was contraindicated because of the patient's age and cardiac disease. CT-guided needle aspiration of 250 ml of viscous fluid was performed, followed by rapid clinical improvement. The patient remained symptom free for several months. The procedure was repeated successfully one year later because of a recurrence of compression.This observation supports the potential use of CT guided transthoracic needle aspiration as an alternative to surgical treatment in cases of inoperable symptomatic bronchogenic cyst.

Abstract: When carbon dioxide (CO2 ) is injected into an aquifer or a depleted geological reservoir, its dissolution into solution results in acidification of the pore waters. As a consequence, the pore waters become more reactive, which leads to enhanced dissolution-precipitation processes and a modification of the mechanical and hydrological properties of the rock. This effect is especially important for limestones given that the solubility and reactivity of carbonates is strongly dependent on pH and the partial pressure of CO2 . The main mechanism that couples dissolution, precipitation and rock matrix deformation is commonly referred to as intergranular pressure solution creep (IPS) or pervasive pressure solution creep (PSC). This process involves dissolution at intergranular grain contacts subject to elevated stress, diffusion of dissolved material in an intergranular fluid, and precipitation in pore spaces subject to lower stress. This leads to an overall and pervasive reduction in porosity due to both grain indentation and precipitation in pore spaces. The percolation of CO2 -rich fluids may influence on-going compaction due to pressure solution and can therefore potentially affect the reservoir and its long-term CO2 storage capacity. We aim at quantifying this effect by using a 2D numerical model to study the coupling between dissolution-precipitation processes, local mass transfer, and deformation of the rock over long time scales. We show that high partial pressures of dissolved CO2 (up to 30 MPa) significantly increase the rates of compaction by a factor of ~50 to ~75, and also result in a concomitant decrease in the viscosity of the rock matrix.

Abstract: The ANTARES collaboration is building a deep sea neutrino telescope in the Mediterranean Sea. This detector will cover a sensitive area of typically 0.1 km-squared and will be equipped with about 1000 optical modules. Each of these optical modules consists of a large area photomultiplier and its associated electronics housed in a pressure resistant glass sphere. The design of the ANTARES optical module, which is a key element of the detector, has been finalized following extensive R & D studies and is reviewed here in detail., 26 pages, 15 figures, to be published in NIM

Abstract: The purpose of this work was to assess results obtained with the MIP (mitomycin 6 mg/m(2) day 1, ifosfamide 1500 mg/m(2) days 1,2, 3, cisplatin 30 mg/m(2) days 1,2, 3) chemotherapy protocol combined with cisplatin-sensitized chest radiotherapy as developed in the French multicentric trial on perioperative chemotherapy.Thirty-five patients with grade III non-small-cell lung cancer (NSCLC) were given two or three starter MIP cycles every 4 weeks then underwent radiation therapy for six weeks for a total dose of 60 Gy with injection of 8 mg/m(2) cisplatin every day for the first two weeks and the last two weeks of treatment. In case of objective response (OR) to MIP before radiotherapy, the MIP protocol was repeated for one to four supplementary cycles according to tolerance.The rate of OR after MIP was 51% and after chemoradiotherapy it was 69%. Toxic effects were limited to one death due to aplasia and 18 cases of grade 3-4 toxicity, mainly due to hematology disorders. Moderate esophagitis was observed in ten cases. Median survival was 13.5 months. Survival rates were 57% and 29% at one and two years.This novel scheme, which can be improved, has demonstrated its efficacy. Tolerance is satisfactory and the cost is low compared with associations using "new" drugs.

Abstract: Dyspnea is the principal clinical feature of pulmonary leukostasis, a syndrome called "poumon hyperleucocytaire" in French. It is a common complication of chronic myeloid leukemia. In the course of pulmonary carcinoma, leukocytosis is frequently noted. One of the etiologies is a paraneoplastic syndrome with production of colony stimulating factor. We report a case of pulmonary hyperleukostasis following antineoplastic chemotherapy for pulmonary carcinoma.

Abstract: The pathogenesis of the sensory retinal detachment of the macula associated with congenital pit of the optic nerve remains controversal. Based on our cases and the cases reported in literature, we propose a management plan for macular retinal detachment associated with optic nerve pit.In this retrospective study ten eyes were treated for progressive visual loss. Treatment modalities were different depending on the time period: laser photocoagulation (to the juxtapapillary region) alone, laser combined with intraocular gas injection (C3 F8), laser combined with vitrectomy and intraocular gas tamponnade (S F6, C3 F8), laser combined with vitrectomy and temporary silicone oil tamponade.The follow-up period ranged between one and five years (mean follow-up thirty three months). We had eight successes and two definitive failures (an old macular detachment treated only by one laser session, and a young girl who had not kept the prone position after gas injection). In the group of six eyes treated by laser alone, only two retinal treatments were directly successfully treated; in three other eyes, the detachment recurred and was treated with success in a second step by a gas injection; the sixth eye is the first definitive failure (old detachment). In the five eyes treated by laser combined gas injection with or without pars plana vitrectomy (including three failures of laser alone), the retina remained attached in four eyes during the follow-up period; the sixth eye was the second definitive failure (the young girl). In the both eyes treated by vitrectomy and silicon oil injection, the retina has flattened.The results suggest that laser photocoagulation alone is not so efficient and that vitrectomy is not necessary with gas injection. Complete resorption of subretinal fluid occurred in eight eyes: two with laser photocoagulation alone, and six with a long term tamponnade combined with a peripapillary laser photocoagulation. In first treatment, this technique (laser with tamponade) is a valuable approach to manage serous macular detachment associated with optic nerve pit.

Abstract: Le faisceau de protons polarises du synchrocyclotron d'Orsay est decrit. L'energie est de (141 ± 2 MeV), la polarisation de (61,6 ± 2,5) %. La polarisation dans la diffusion elastique p - 12C et p - d a ete mesuree dans l'intervalle 10° — 25° lab. pour le carbone et 23° — 160° CM pour le deuterium. Les resultats sont compares a ceux deja existants et aux calculs theoriques pour le deuterium.

Abstract: In this paper, we describe the construction of a proton beam which can also be vertically polarized in both orientations. To reverse the beam polarization, the incident scattering angle was reversed. This polarized beam has a sufficiently high intensity (2·106 protons per second) and polarization (near 40%) to be used in experiments. Measurements of the analysing power of carbon at 600 MeV are also given for four scattering angles.

Abstract: In order to increase the pharmacological activities of the molecules described in the first part of the paper, an acetic or a 2-propionic group was introduced in to the fundamental structure, either on the aromatic ring, on the pyrrole nitrogen or on carbon 3 of the heterocycle. Moreover, a 4-chlorobenzoyl group was fixed on carbon 3 of the pyrrole via condensation of 4-chloroaniline with 4-chlorobenzoyl triketones. The structure of the adducts was demonstrated both by N.M.R. spectra and by using another unambiguous synthetic route. Finally, derivatives with the salicylic substructure on the nitrogen of the pyrrole are described. Their analgesic and anti-inflammatory properties are reported as well as some effects on the central nervous system.

Abstract: The derivatives in question are prepared by reaction of substituted anilines with 1,4-ketones in an acidic medium. The diketones are themselves obtained either using Friedel and Crafts reaction with levulinic acid chloride, or by Stetter's method reacting Mannich bases with aryl aldehydes in the presence of sodium cyanide. The results of test of acute toxicity and of analgesic and anti-inflammatory activity are reported.

Abstract: In 2005, the ANTARES Collaboration deployed and operated at a depth of 2500 m a so-called Mini Instrumentation Line equipped with Optical Modules (MILOM) at the ANTARES site. The various data acquired during the continuous operation from April to December 2005 of the MILOM confirm the satisfactory performance of the Optical Modules, their front-end electronics and readout system, as well as the calibration devices of the detector. The in-situ measurement of the Optical Module time response yields a resolution better than 0.5 ns. The performance of the acoustic positioning system, which enables the spatial reconstruction of the ANTARES detector with a precision of about 10 cm, is verified. These results demonstrate that with the full ANTARES neutrino telescope the design angular resolution of better than $0.3^\circ$ can be realistically achieved., Comment: 18 pages, 12 figures, submitted to Astroparticle Physics

48. Role of the X Chromosome in Alzheimer Disease Genetics.

Abstract: Importance: The X chromosome has remained enigmatic in Alzheimer disease (AD), yet it makes up 5% of the genome and carries a high proportion of genes expressed in the brain, making it particularly appealing as a potential source of unexplored genetic variation in AD., Objectives: To perform the first large-scale X chromosome-wide association study (XWAS) of AD., Design, Setting, and Participants: This was a meta-analysis of genetic association studies in case-control, family-based, population-based, and longitudinal AD-related cohorts from the US Alzheimer's Disease Genetics Consortium, the Alzheimer's Disease Sequencing Project, the UK Biobank, the Finnish health registry, and the US Million Veterans Program. Risk of AD was evaluated through case-control logistic regression analyses. Data were analyzed between January 2023 and March 2024. Genetic data available from high-density single-nucleotide variant microarrays and whole-genome sequencing and summary statistics for multitissue expression and protein quantitative trait loci available from published studies were included, enabling follow-up genetic colocalization analyses. A total of 1 629 863 eligible participants were selected from referred and volunteer samples, 477 596 of whom were excluded for analysis exclusion criteria. The number of participants who declined to participate in original studies was not available., Main Outcome and Measures: Risk of AD, reported as odds ratios (ORs) with 95% CIs. Associations were considered at X chromosome-wide (P < 1 × 10-5) and genome-wide (P < 5 × 10-8) significance. Primary analyses are nonstratified, while secondary analyses evaluate sex-stratified effects., Results: Analyses included 1 152 284 participants of non-Hispanic White, European ancestry (664 403 [57.7%] female and 487 881 [42.3%] male), including 138 558 individuals with AD. Six independent genetic loci passed X chromosome-wide significance, with 4 showing support for links between the genetic signal for AD and expression of nearby genes in brain and nonbrain tissues. One of these 4 loci passed conservative genome-wide significance, with its lead variant centered on an intron of SLC9A7 (OR, 1.03; 95% CI, 1.02-1.04) and colocalization analyses prioritizing both the SLC9A7 and nearby CHST7 genes. Of these 6 loci, 4 displayed evidence for escape from X chromosome inactivation with regard to AD risk., Conclusion and Relevance: This large-scale XWAS of AD identified the novel SLC9A7 locus. SLC9A7 regulates pH homeostasis in Golgi secretory compartments and is anticipated to have downstream effects on amyloid β accumulation. Overall, this study advances our knowledge of AD genetics and may provide novel biological drug targets. The results further provide initial insights into elucidating the role of the X chromosome in sex-based differences in AD.

49. Safety and efficacy of immunotherapy using a double-dose regimen in advanced non-small cell lung cancer (NSCLC): results of IDEE study.

Abstract: Background: Pembrolizumab 400 mg every six weeks (Q6W) and nivolumab 480 mg every four weeks (Q4W) are used since 2020 and the coronavirus disease 2019 (COVID-19) pandemic. This recommendation relied on pharmacokinetic and pharmacodynamic models. The objective of the IDEE (Immunothérapie Double dose Etendue: Experience bretonne) study is to determine the safety and efficacy of this treatment regimen in real life conditions., Methods: We conducted an observational, retrospective, multicentric study including 117 patients with advanced non-small cell lung cancer (NSCLC) who received pembrolizumab Q6W or nivolumab Q4W between March 2020 and March 2021., Results: The median age was 67 years, 68% were men with predominantly lung adenocarcinoma. The median time to double-dose regimen failure (TDDF) was 9.2 months. The survival rate at 12 months was 79%. TDDF was not influenced by sex, line of treatment, pathologic subtypes or anti-programmed cell death protein 1 (PD-1) antibody. There was no correlation between TDDF and duration of prior exposition to immunotherapy before switching. Sixty-eight patients experienced double-dose treatment failure, 28% because of toxicity including five definitive discontinuations. Five grade ≥3 immune-adverse events were reported included two cases of pneumonitis, all responding to corticosteroid therapy., Conclusions: Our multicentric cohort supports the feasibility of pembrolizumab Q6W and nivolumab Q4W for patients with advanced NSCLC. 50. Plasma proteomics in the UK Biobank reveals youthful brains and immune systems promote healthspan and longevity.

Abstract: Organ-derived plasma protein signatures derived from aptamer protein arrays track organ-specific aging, disease, and mortality in humans, but the robustness and clinical utility of these models and their biological underpinnings remain unknown. Here, we estimate biological age of 11 organs from 44,526 individuals in the UK Biobank using an antibody-based proteomics platform to model disease and mortality risk. Organ age estimates are associated with future onset of heart failure (heart age HR=1.83), chronic obstructive pulmonary disease (lung age HR=1.39), type II diabetes (kidney age HR=1.58), and Alzheimer's disease (brain age HR=1.81) and sensitive to lifestyle factors such as smoking and exercise, hormone replacement therapy, or supplements. Remarkably, the accrual of aged organs progressively increases mortality risk while a youthful brain and immune system are uniquely associated with disease-free longevity.
