Abstract: We report the discovery of GRB 221009A, the highest flux gamma-ray burst (GRB) ever observed by the Fermi Gamma-ray Burst Monitor (Fermi-GBM). This GRB has continuous prompt emission lasting more than 600 s, which smoothly transitions to afterglow emission visible in the Fermi-GBM energy range (8 keV–40 MeV), and total energetics higher than any other burst in the Fermi-GBM sample. By using a variety of new and existing analysis techniques we probe the spectral and temporal evolution of GRB 221009A. We find no emission prior to the Fermi-GBM trigger time ( t _0 ; 2022 October 9 at 13:16:59.99 UTC), indicating that this is the time of prompt emission onset. The triggering pulse exhibits distinct spectral and temporal properties suggestive of the thermal, photospheric emission of shock breakout, with significant emission up to ∼15 MeV. We characterize the onset of external shock at t _0 + 600 s and find evidence of a plateau region in the early-afterglow phase, which transitions to a slope consistent with Swift-XRT afterglow measurements. We place the total energetics of GRB 221009A in context with the rest of the Fermi-GBM sample and find that this GRB has the highest total isotropic-equivalent energy ( E _γ _,iso = 1.0 × 10 ^55 erg) and second highest isotropic-equivalent luminosity ( L _γ _,iso = 9.9 × 10 ^53 erg s ^–1 ) based on its redshift of z = 0.151. These extreme energetics are what allowed us to observe the continuously emitting central engine of Fermi-GBM from the beginning of the prompt emission phase through the onset of early afterglow.

Abstract: We present 294 pulsars found in GeV data from the Large Area Telescope (LAT) on the Fermi Gamma-ray Space Telescope. Another 33 millisecond pulsars (MSPs) discovered in deep radio searches of LAT sources will likely reveal pulsations once phase-connected rotation ephemerides are achieved. A further dozen optical and/or X-ray binary systems colocated with LAT sources also likely harbor gamma-ray MSPs. This catalog thus reports roughly 340 gamma-ray pulsars and candidates, 10% of all known pulsars, compared to ≤11 known before Fermi. Half of the gamma-ray pulsars are young. Of these, the half that are undetected in radio have a broader Galactic latitude distribution than the young radio-loud pulsars. The others are MSPs, with six undetected in radio. Overall, ≥236 are bright enough above 50 MeV to fit the pulse profile, the energy spectrum, or both. For the common two-peaked profiles, the gamma-ray peak closest to the magnetic pole crossing generally has a softer spectrum. The spectral energy distributions tend to narrow as the spindown power $\dot{E}$ decreases to its observed minimum near 10 ^33 erg s ^−1 , approaching the shape for synchrotron radiation from monoenergetic electrons. We calculate gamma-ray luminosities when distances are available. Our all-sky gamma-ray sensitivity map is useful for population syntheses. The electronic catalog version provides gamma-ray pulsar ephemerides, properties, and fit results to guide and be compared with modeling results.

Abstract: The Cherenkov Telescope Array (CTA) is a next-generation ground-based observatory for gamma-ray astronomy at very high energies. The Large-Sized Telescope prototype (LST-1) is located at the CTA-North site, on the Canary Island of La Palma. LSTs are designed to provide optimal performance in the lowest part of the energy range covered by CTA, down to ≃20 GeV. LST-1 started performing astronomical observations in 2019 November, during its commissioning phase, and it has been taking data ever since. We present the first LST-1 observations of the Crab Nebula, the standard candle of very-high-energy gamma-ray astronomy, and use them, together with simulations, to assess the performance of the telescope. LST-1 has reached the expected performance during its commissioning period—only a minor adjustment of the preexisting simulations was needed to match the telescope’s behavior. The energy threshold at trigger level is around 20 GeV, rising to ≃30 GeV after data analysis. Performance parameters depend strongly on energy, and on the strength of the gamma-ray selection cuts in the analysis: angular resolution ranges from 0.°12–0.°40, and energy resolution from 15%–50%. Flux sensitivity is around 1.1% of the Crab Nebula flux above 250 GeV for a 50 hr observation (12% for 30 minutes). The spectral energy distribution (in the 0.03–30 TeV range) and the light curve obtained for the Crab Nebula agree with previous measurements, considering statistical and systematic uncertainties. A clear periodic signal is also detected from the pulsar at the center of the Nebula.

Abstract: We study the broadband emission of Mrk 501 using multiwavelength observations from 2017 to 2020 performed with a multitude of instruments, involving, among others, MAGIC, Fermi's Large Area Telescope (LAT), NuSTAR, Swift, GASP-WEBT, and the Owens Valley Radio Observatory. Mrk 501 showed an extremely low broadband activity, which may help to unravel its baseline emission. Nonetheless, significant flux variations are detected at all wave bands, with the highest occurring at X-rays and very-high-energy (VHE) γ -rays. A significant correlation (>3 σ ) between X-rays and VHE γ -rays is measured, supporting leptonic scenarios to explain the variable parts of the emission, also during low activity. This is further supported when we extend our data from 2008 to 2020, and identify, for the first time, significant correlations between the Swift X-Ray Telescope and Fermi-LAT. We additionally find correlations between high-energy γ -rays and radio, with the radio lagging by more than 100 days, placing the γ -ray emission zone upstream of the radio-bright regions in the jet. Furthermore, Mrk 501 showed a historically low activity in X-rays and VHE γ -rays from mid-2017 to mid-2019 with a stable VHE flux (>0.2 TeV) of 5% the emission of the Crab Nebula. The broadband spectral energy distribution (SED) of this 2 yr long low state, the potential baseline emission of Mrk 501, can be characterized with one-zone leptonic models, and with (lepto)-hadronic models fulfilling neutrino flux constraints from IceCube. We explore the time evolution of the SED toward the low state, revealing that the stable baseline emission may be ascribed to a standing shock, and the variable emission to an additional expanding or traveling shock.

Abstract: The Fermi Large Area Telescope (LAT) lightcurve repository (LCR) is a publicly available, continually updated library of gamma-ray lightcurves of variable Fermi-LAT sources generated over multiple timescales. The Fermi-LAT LCR aims to provide publication-quality lightcurves binned on timescales of 3, 7, and 30 days for 1525 sources deemed variable in the source catalog of the first 10 yr of Fermi-LAT observations. The repository consists of lightcurves generated through full likelihood analyses that model the sources and the surrounding region, providing fluxes and photon indices for each time bin. The LCR is intended as a resource for the time-domain and multimessenger communities by allowing users to search LAT data quickly to identify correlated variability and flaring emission episodes from gamma-ray sources. We describe the sample selection and analysis employed by the LCR and provide an overview of the associated data access portal.

Abstract: Numerous studies and surveys have examined the utilization of Information and Communication Technologies (ICTs) in Higher Education (HE), but the lack of studies on the utilization of ICTs in Quality Assurance and Accreditation (QAA) of HE led us to perform a Systematic Literature Review (SLR). This review mainly benefits QAA agencies and researchers interested in utilizing ICTs for QAA of HE. Several contributions of ICTs to QAA of HE are identified. 1) Compared to QAA agencies, Higher Education Institutions (HEIs) are more likely to implement ICTs. 2) Most ICTs in HEIs are for Quality Assurance (QA). 3) Most previous studies were conducted in Europe, demonstrating that efforts have been made to align QAA agencies and HEI QAA units with European Standards. Consequently, ICTs are more often utilized by European HEIs. Furthermore, we investigated the QAA status in a developing country (Afghanistan) as a case study.

Abstract: The fourth catalog of active galactic nuclei (AGNs) detected by the Fermi Gamma-ray Space Telescope Large Area Telescope (4LAC) between 2008 August 4 and 2016 August 2 contains $2863$ objects located at high Galactic latitudes (| b| > 10^9). It includes 85% more sources than the previous 3LAC catalog based on 4 yr of data. AGNs represent at least 79% of the high-latitude sources in the fourth Fermi-Large Area Telescope Source Catalog (4FGL), which covers the energy range from 50 MeV to 1 TeV. In addition, 344 gamma-ray AGNs are found at low Galactic latitudes. Most of the 4LAC AGNs are blazars (98%), while the remainder are other types of AGNs. The blazar population consists of 24% Flat Spectrum Radio Quasars (FSRQs), 38% BL Lac-type objects, and 38% blazar candidates of unknown types (BCUs). On average, FSRQs display softer spectra and stronger variability in the gamma-ray band than BL Lacs do, confirming previous findings. All AGNs detected by ground-based atmospheric Cerenkov telescopes are also found in the 4LAC.

Abstract: We present the fourth Fermi Large Area Telescope catalog (4FGL) of gamma-ray sources. Based on the first eight years of science data from the Fermi Gamma-ray Space Telescope mission in the energy range from 50 MeV to 1 TeV, it is the deepest yet in this energy range. Relative to the 3FGL catalog, the 4FGL catalog has twice as much exposure as well as a number of analysis improvements, including an updated model for the Galactic diffuse gamma-ray emission, and two sets of light curves (one-year and two-month intervals). The 4FGL catalog includes 5064 sources above 4 sigma significance, for which we provide localization and spectral properties. Seventy-five sources are modeled explicitly as spatially extended, and overall, 358 sources are considered as identified based on angular extent, periodicity, or correlated variability observed at other wavelengths. For 1336 sources, we have not found plausible counterparts at other wavelengths. More than 3130 of the identified or associated sources are active galaxies of the blazar class, and 239 are pulsars.

Abstract: We report on the observations of gamma-ray burst (GRB) 190114C by the Fermi Gamma-ray Space Telescope and the Neil Gehrels Swift Observatory. The prompt gamma-ray emission was detected by the Fermi GRB Monitor (GBM), the Fermi Large Area Telescope (LAT), and the Swift Burst Alert Telescope (BAT) and the long-lived afterglow emission was subsequently observed by the GBM, LAT, Swift X-ray Telescope (XRT), and Swift UV Optical Telescope. The early-time observations reveal multiple emission components that evolve independently, with a delayed power-law component that exhibits significant spectral attenuation above 40 MeV in the first few seconds of the burst. This power-law component transitions to a harder spectrum that is consistent with the afterglow emission observed by the XRT at later times. This afterglow component is clearly identifiable in the GBM and BAT light curves as a slowly fading emission component on which the rest of the prompt emission is superimposed. As a result, we are able to observe the transition from internal-shock- to external-shock-dominated emission. We find that the temporal and spectral evolution of the broadband afterglow emission can be well modeled as synchrotron emission from a forward shock propagating into a wind-like circumstellar environment. We estimate the initial bulk Lorentz factor using the observed high-energy spectral cutoff. Considering the onset of the afterglow component, we constrain the deceleration radius at which this forward shock begins to radiate in order to estimate the maximum synchrotron energy as a function of time. We find that even in the LAT energy range, there exist high-energy photons that are in tension with the theoretical maximum energy that can be achieved through synchrotron emission from a shock. These violations of the maximum synchrotron energy are further compounded by the detection of very high-energy (VHE) emission above 300 GeV by MAGIC concurrent with our observations. We conclude that the observations of VHE photons from GRB 190114C necessitates either an additional emission mechanism at very high energies that is hidden in the synchrotron component in the LAT energy range, an acceleration mechanism that imparts energy to the particles at a rate that is faster than the electron synchrotron energy-loss rate, or revisions of the fundamental assumptions used in estimating the maximum photon energy attainable through the synchrotron process.

Abstract: Artículo escrito por un elevado número de autores, solo se referencian el que aparece en primer lugar, los autores pertenecientes a la UAM y el nombre del grupo de colaboración, si lo hubiere, The Fermi Large Area Telescope (LAT) lightcurve repository (LCR) is a publicly available, continually updated library of gamma-ray lightcurves of variable Fermi-LAT sources generated over multiple timescales. The Fermi-LAT LCR aims to provide publication-quality lightcurves binned on timescales of 3, 7, and 30 days for 1525 sources deemed variable in the source catalog of the first 10 yr of Fermi-LAT observations. The repository consists of lightcurves generated through full likelihood analyses that model the sources and the surrounding region, providing fluxes and photon indices for each time bin. The LCR is intended as a resource for the time-domain and multimessenger communities by allowing users to search LAT data quickly to identify correlated variability and flaring emission episodes from gamma-ray sources. We describe the sample selection and analysis employed by the LCR and provide an overview of the associated data access portal, M.N. and J.V. acknowledge that the material is based upon work supported by NASA under award number 80GSFC21M0002. Abstract: The HAWC Collaboration released the 2HWC catalogue of TeV sources, in which 19 show no association with any known high-energy (HE; E ≳ 10 GeV) or very-high-energy (VHE; E ≳ 300 GeV) sources. This catalogue motivated follow-up studies by both the Major Atmospheric Gamma-ray Imaging Cherenkov (MAGIC) and Fermi-LAT (Large Area Telescope) observatories with the aim of investigating gamma-ray emission over a broad energy band. In this paper, we report the results from the first joint work between High Altitude Water Cherenkov (HAWC), MAGIC, and Fermi-LAT on three unassociated HAWC sources: 2HWC J2006+341, 2HWC J1907+084*, and 2HWC J1852+013*. Although no significant detection was found in the HE and VHE regimes, this investigation shows that a minimum 1° extension (at 95 per cent confidence level) and harder spectrum in the GeV than the one extrapolated from HAWC results are required in the case of 2HWC J1852+013*, whilst a simply minimum extension of 0.16° (at 95 per cent confidence level) can already explain the scenario proposed by HAWC for the remaining sources. Moreover, the hypothesis that these sources are pulsar wind nebulae is also investigated in detail.

Abstract: We use joint observations by the Neil Gehrels Swift X-ray Telescope (XRT) and the Fermi Large Area Telescope (LAT) of gamma-ray burst (GRB) afterglows to investigate the nature of the long-lived high-energy emission observed by Fermi LAT. Joint broadband spectral modeling of XRT and LAT data reveal that LAT non-detections of bright X-ray afterglows are consistent with a cooling break in the inferred electron synchrotron spectrum below the LAT and/or XRT energy ranges. Such a break is sufficient to suppress the high-energy emission so as to be below the LAT detection threshold. By contrast, LAT-detected bursts are best fit by a synchrotron spectrum with a cooling break that lies either between or above the XRT and LAT energy ranges. We speculate that the primary difference between GRBs with LAT afterglow detections and the non-detected population may be in the type of circumstellar environment in which these bursts occur, with late-time LAT detections preferentially selecting GRBs that occur in low wind-like circumburst density profiles. Furthermore, we find no evidence of high-energy emission in the LAT-detected population significantly in excess of the flux expected from the electron synchrotron spectrum fit to the observed X-ray emission. The lack of excess emission at high energies could be due to a shocked external medium in which the energy density in the magnetic field is stronger than or comparable to that of the relativistic electrons behind the shock, precluding the production of a dominant synchrotron self-Compton (SSC) component in the LAT energy range. Alternatively, the peak of the SSC emission could be beyond the 0.1–100 GeV energy range considered for this analysis.

Abstract: Very-high Energy (VHE) gamma-ray astroparticle physics is a relatively young field, and observations over the past decade have surprisingly revealed almost two hundred VHE emitters which appear to act as cosmic particle accelerators. These sources are an important component of the Universe, influencing the evolution of stars and galaxies. At the same time, they also act as a probe of physics in the most extreme environments known - such as in supernova explosions, and around or after the merging of black holes and neutron stars. However, the existing experiments have provided exciting glimpses, but often falling short of supplying the full answer. A deeper understanding of the TeV sky requires a significant improvement in sensitivity at TeV energies, a wider energy coverage from tens of GeV to hundreds of TeV and a much better angular and energy resolution with respect to the currently running facilities. The next generation gamma-ray observatory, the Cherenkov Telescope Array Observatory (CTAO), is the answer to this need. In this talk I will present this upcoming observatory from its design to the construction, and its potential science exploitation. CTAO will allow the entire astronomical community to explore a new discovery space that will likely lead to paradigm-changing breakthroughs. In particular, CTA has an unprecedented sensitivity to short (sub-minute) timescale phenomena, placing it as a key instrument in the future of multi-messenger and multi-wavelength time domain astronomy. Abstract: GRB 131108A is a bright long Gamma-Ray Burst (GRB) detected by the Large Area Telescope and the Gamma-ray Burst Monitor on board the Fermi Gamma-ray Space Telescope. Dedicated temporal and spectral analyses reveal three $\gamma$-ray flares dominating above 100 MeV, which are not directly related to the prompt emission in the GBM band (10 keV - 10 MeV). The high-energy light curve of GRB 131108A (100 MeV - 10 GeV) shows an unusual evolution: a steep decay, followed by three flares with an underlying emission, and then a long-lasting decay phase. The detailed analysis of the $\gamma$-ray flares finds that the three flares are 6 - 20 times brighter than the underlying emission and are similar to each other. The fluence of each flare, (1.6 $\sim$ 2.0) $\times$ 10$^{-6}$ erg cm$^{-2}$, is comparable to that of emission during the steep decay phase, 1.7 $\times$ 10$^{-6}$ erg cm$^{-2}$. The total fluence from three $\gamma$-ray flares is 5.3 $\times$ 10$^{-6}$ erg cm$^{-2}$. The three $\gamma$-ray flares show properties similar to the usual X-ray flares that are sharp flux increases, occurring in $\sim$ 50\% of afterglows, in some cases well after the prompt emission. Also, the temporal and spectral indices during the early steep decay phase and the decaying phase of each flare show the consistency with a relation of the curvature effect ($\alpha$ = 2 + $\beta$), which is the first observational evidence of the high-latitude emission in the GeV energy band., Comment: ApJL, 10 pages, 3 figures, 2 tables

Abstract: InterPlanetary Network (IPN) data are presented for the gamma-ray bursts in the second Fermi Gamma-Ray Burst Monitor (GBM) catalog. Of the 462 bursts in that catalog between 2010 July 12 and 2012 July 11, 428, or 93%, were observed by at least 1 other instrument in the 9-spacecraft IPN. Of the 428, the localizations of 165 could be improved by triangulation. For these bursts, triangulation gives one or more annuli whose half-widths vary between about 2.′3° and 16°, depending on the peak flux, fluence, time history, arrival direction, and the distance between the spacecraft. We compare the IPN localizations with the GBM 1σ, 2σ, and 3σ error contours and find good agreement between them. The IPN 3σ error boxes have areas between about 8 square arcminutes and 380 square degrees, and are an average of 2500 times smaller than the corresponding GBM 3σ localizations. We identify four bursts in the IPN/GBM sample whose origins were given as “uncertain,” but may in fact be cosmic. This leads to an estimate of over 99% completeness for the GBM catalog.

37. Methods for immunoblot detection and electron microscopic localization of septin subunits in mammalian nervous systems

Abstract: The minimal functional units of the mammalian septin system are diverse heterooligomers of SEPT1-14 subunits, which are most abundantly and differentially expressed in postmitotic neurons and glia. The subunit compositions of such heterooligomers are thought to differentiate their affinity for other proteins and lipids, and subcellular localization. Thus, high-precision quantification and mapping of each subunit is necessary to understand their subcellular functions and physiological roles. However, systematic information on the localization of individual septin subunits in the mammalian nervous system is limited. Here, we present our experimental workflows for the study of septin expression and localization in the rodent brain by immunoblot and serial section immunoelectron microscopy. Our protocols, based on standard methods, have been rigorously optimized and simplified for universality and reproducibility to aid non-experts in the field.

Abstract: We present early phase observations in optical and near-infrared wavelengths for the extremely luminous Type Ia supernova (SN Ia) 2009dc. The decline rate of the light curve is Δm 15(B) = 0.65 ± 0.03, which is one of the slowest among SNe Ia. The peak V-band absolute magnitude is estimated to be MV = -19.90 ± 0.15 mag if no host extinction is assumed. It reaches MV = -20.19 ± 0.19 mag if we assume the host extinction of AV = 0.29 mag. SN 2009dc belongs to the most luminous class of SNe Ia, like SNe 2003fg and 2006gz. Our JHKs -band photometry shows that this SN is also one of the most luminous SNe Ia in near-infrared wavelengths. We estimate the ejected 56Ni mass of 1.2 ± 0.3 M sun for the no host extinction case (and of 1.6 ± 0.4 M sun for the host extinction of AV = 0.29 mag). The C II λ6580 absorption line remains visible until a week after the maximum brightness, in contrast to its early disappearance in SN 2006gz. The line velocity of Si II λ6355 is about 8000 km s-1 around the maximum, being considerably slower than that of SN 2006gz. The velocity of the C II line is similar to or slightly less than that of the Si II line around the maximum. The presence of the carbon line suggests that the thick unburned C+O layer remains after the explosion. Spectropolarimetric observations by Tanaka et al. indicate that the explosion is nearly spherical. These observational facts suggest that SN 2009dc is a super-Chandrasekhar mass SN Ia.

Abstract: Suzaku launched in July 2005 is equipped with the wideband instruments covering 0.3–700 keV. In addition to these main instruments, active shield counters of the hard X-ray detector are designed to be a wideband all-sky monitor (WAM). The WAM is a powerful gamma-ray burst monitor with a wide energy range of 50–5000 keV and with a large effective area of 400 cm 2 even around 1 MeV. The paper describes the strategy for the prompt GRB observation with the WAM, and the follow-up observation of X-ray afterglows with the narrow field instruments. So far, the WAM has detected 53 GRBs and half of them were detected simultaneously with other satellites. In addition to that, from at least 8 GRBs, including a bright and hard GRB 051008, it is succeeded to observe significant gamma-ray emission up to 1 MeV. The X-ray afterglow observations were carried out on January 5 and on September 4, 2006. Their results are reported in separate paper.

42. Developing networks between workers and community for health promotion of workers in small businesses

Abstract: The purpose of this study is to assess preventive factors to workers' health care in small businesses, and to develop the network between workers and the community health resources. The life expectancy of 65 years of the Fuji City citizen, and their SMR of cancer, stroke and cardiovascular disease were worst in the Shizuoka prefecture. The ratio of workers in small business with workers under 50 employees was 97.5% in this city. To assess the preventive factor for the worker's health promotion, we obtained information both of the health care system and the health consciousness of the employee of small business, through a questionnaire by mail. 22.7% in business did not execute the periodic health check. The awareness of illness was in 18.4% of them. 48.4% of them became aware of the anxiety of mental health, and 82.2% of them were conscious of the uncertainty of the future health. 41.5% of them did not receive the detailed medical examination and treatment. We hope employers in small businesses would set up the opportunity of periodic health checks of employees, for getting rid of the anxiety of mental health and the uncertainty for the future health, and develop the network between the workers and the community.

Abstract: The Gamma Ray Large Area Space Telescope (GLAST) Large Area Telescope (LAT) is a pair-production high-energy (>20 MeV) gamma-ray telescope being built by an international partnership of astrophysicists and particle physicists for a satellite launch in 2006, designed to study a wide variety of high-energy astrophysical phenomena. As part of the development effort, the collaboration has built a Balloon Flight Engineering Model (BFEM) for flight on a high-altitude scientific balloon. The BFEM is approximately the size of one of the 16 GLAST-LAT towers and contains all the components of the full instrument: plastic scintillator anticoincidence system (ACD), high-Z foil/Si strip pair-conversion tracker (TKR), CsI hodoscopic calorimeter (CAL), triggering and data acquisition electronics (DAQ), commanding system, power distribution, telemetry, real-time data display, and ground data processing system. The principal goal of the balloon flight was to demonstrate the performance of this instrument configuration under conditions similar to those expected in orbit. Abstract: International audience; To uniformly determine the properties of supernova remnants (SNRs) at high energies, we have developed the first systematic survey at energies from 1 to 100 GeV using data from the Fermi Large Area Telescope (LAT). Based on the spatial overlap of sources detected at GeV energies with SNRs known from radio surveys, we classify 30 sources as likely GeV SNRs. We also report 14 marginal associations and 245 flux upper limits. A mock catalog in which the positions of known remnants are scrambled in Galactic longitude allows us to determine an upper limit of 22% on the number of GeV candidates falsely identified as SNRs. We have also developed a method to estimate spectral and spatial systematic errors arising from the diffuse interstellar emission model, a key component of all Galactic Fermi LAT analyses. By studying remnants uniformly in aggregate, we measure the GeV properties common to these objects and provide a crucial context for the detailed modeling of individual SNRs. Combining our GeV results with multiwavelength (MW) data, including radio, X-ray, and TeV, we demonstrate the need for improvements to previously sufficient, simple models describing the GeV and radio emission from these objects. We model the GeV and MW emission from SNRs in aggregate to constrain their maximal contribution to observed Galactic cosmic rays.

45. Pioglitazone attenuates tactile allodynia and microglial activation in mice with peripheral nerve injury

Abstract: To test the possibility of a peroxisome proliferator activated receptor (PPAR) γ agonist to treat neuropathic pain, we examined the effects of pioglitazone, a PPARγ agonist, on tactile allodynia and expression of activated microglia in the dorsal horn of spinal cord using neuropathic pain model. The unilateral sciatic nerve was partially ligated (PSL) in male ICR mice. Pioglitazone (1-25 mg/kg p.o.) was administrated to mice once daily for five days immediately after PSL. We stimulated the footpad of the hind paw of mice using a von Frey filament to estimate tactile allodynia on day 5 of PSL. The activated microglia in the lumbar spinal cord was observed by immunohistochemistry with anti-Iba1 antibody, a marker for activated microglia. The number of Iba1-immunoreactive cells was counted in the dorsal horn spinal cord. On day 5, significant allodynia was developed in PSL mice. Pioglitazone significantly attenuated the tactile allodynia in a dose of 1-25 mg/kg. However, these doses of pioglitazone did not affect nociceptive responses in sham mice. Moreover, on day 6, the number of activated microglia was significantly increased in the ipsilateral dorsal horn of mice. The increase in the number of activated microglia induced by PSL was significantly suppressed by pioglitazone (1-25 mg/kg p.o.). Pioglitazone did not affect the number of activated microglia in sham mice. These results suggest that PPARγ activation inhibits the development of tactile allodynia and the expression of activated microglia in the dorsal horn of spinal cord in mice with PSLinduced peripheral nerve injury.

46. CC-chemokine ligand 4/macrophage inflammatory protein-1β participates in the induction of neuropathic pain after peripheral nerve injury

Abstract: Neuropathic pain is caused by neural damage or dysfunction and neuropathic pain-related symptoms are resistant to conventional analgesics. Neuroinflammation due to the cytokine-chemokine network may play a pivotal role in neuropathic pain. We demonstrate that macrophage inflammatory protein-1β (MIP-1β) participates in neuropathic pain.Mice received partial sciatic nerve ligation (PSL), and tactile allodynia and thermal hyperalgesia were assessed by von Frey test and Hargreaves test, respectively. Agents were administered into the region surrounding the sciatic nerve (SCN).Using reverse transcription polymerase chain reaction, the mRNA expressions of MIP-1β and its receptor (CC-chemokine receptor 5; CCR5) in the injured SCN were up-regulated after PSL. MIP-1β immunoreactivity was localized in macrophages and Schwann cells and increased in the injured SCN on day 1. PSL-induced tactile allodynia on days 4 to 7 was prevented by the administration of MIP-1β neutralizing antibody (anti-MIP-1β; on days 0, 3 and 6). PSL-induced up-regulations of inflammatory cytokine-chemokine mRNAs in the injured SCN were suppressed with anti-MIP-1β treatment on day 7. Administration of CCR5 antagonist, D-ala-peptide T-amide (on days 0, 3 and 6) prevented tactile allodynia and thermal hyperalgesia on days 4 to 14. Single administration of recombinant mouse MIP-1β (rmMIP-1β) elicited tactile allodynia. Moreover, rmMIP-1β increased the mRNA expression of inflammatory mediators in the SCN on day 1 after administration.These results suggest that MIP-1β is a novel key mediator, and the peripheral MIP-1β-CCR5 axis contributes to neuropathic pain. Therefore, investigation of this cascade might be a validated approach for the elucidation of neuropathic pain mechanisms.

Abstract: All-sky observations from hard X-ray to soft gamma-ray band is very important to study non-thermal processes for many bright astronomical objects such as Gamma-ray Bursts (GRBs), soft gamma-ray repeaters (SGRs), and solar flares. The Wide-band All-sky Monitor (WAM) onboard Suzaku enables unique observations utilizing its wide energy coverage between 50 keV to 5000 keV with very large effective area of 400 cm2 even at 1 MeV. After six year observations, the WAM has detected more than 850 GRBs. Thanks to its largest effective area above 300 keV among all all-sky instruments, many data with good photon statistics is available even for both timeresolved spectroscopy and short duration GRBs and some implications for emission mechanism and physical condition of emission region of GRBs have been suggested. The WAM also observed many solar flares and SGRs. We summarized the observational results from 105 solar flares and their characteristics and addressed discussion on particle acceleration and heating process in solar flares. In addition to above transient objects, many bright gamma-ray sources such as Crab, Cyg X-1, GRB 1915-105, and NGC 4541 are monitored by the WAM using earth occultation technique.

Abstract: We report on the discovery of \geq 100 MeV {\gamma} rays from the binary system PSR B1259-63/LS 2883 using the Large Area Telescope (LAT) on board Fermi. The system comprises a radio pulsar in orbit around a Be star. We report on LAT observations from near apastron to ~ 60 days after the time of periastron, tp, on 2010 December 15. No {\gamma}-ray emission was detected from this source when it was far from periastron. Faint {\gamma}-ray emission appeared as the pulsar approached periastron. At ~ tp + 30d, the \geq 100 MeV {\gamma}-ray flux increased over a period of a few days to a peak flux 20-30 times that seen during the pre-periastron period, but with a softer spectrum. For the following month, it was seen to be variable on daily time scales, but remained at ~ 1 - 4 \times 10^-6 cm^-2 s^-1 before starting to fade at ~ tp + 57d. The total {\gamma}-ray luminosity observed during this period is comparable to the spin-down power of the pulsar. Simultaneous radio and X-ray observations of the source showed no corresponding dramatic changes in radio and X-ray flux between the pre-periastron and post-periastron flares. Abstract: We report on the first year of Fermi gamma-ray observations of pulsed high-energy emission from the old PSR J2043+2740. The study of the gamma-ray efficiency of such old pulsars gives us an insight into the evolution of pulsars' ability to emit in gammma rays as they age. The gamma-ray lightcurve of this pulsar above 0.1 GeV is clearly defined by two sharp peaks, 0.353+/-0.035 periods apart. We have combined the gamma-ray profile characteristics of PSR J2043+2740 with the geometrical properties of the pulsar's radio emission, derived from radio polarization data, and constrained the pulsar-beam geometry in the framework of a Two Pole Caustic and an Outer Gap model. The ranges of magnetic inclination and viewing angle were determined to be {alpha,zeta}~{52-57,61-68} for the Two Pole Caustic model, and {alpha,zeta}~{62-73,74-81} and {alpha,zeta}~{72-83,60-75} for the Outer Gap model. Based on this geometry, we assess possible birth locations for this pulsar and derive a likely proper motion, sufficiently high to be measurable with VLBI. At a characteristic age of 1.2 Myr, PSR J2043+2740 is the third oldest of all discovered, non-recycled, gamma-ray pulsars: it is twice as old as the next oldest, PSR J0357+32, and younger only than the recently discovered PSR J1836+5925 and PSR J2055+25, both of which are at least 5 and 10 times less energetic, respectively., 17 pages, 6 figures, Accepted for publication in ApJ
