A significant amount of research has been done on the segmentation of coronary arteries. However, the resulting automated boundary delineation is still not suitable for clinical utilization. The convolutional neural network was driving advances in the medical image processing. We propose the brief convolutional network (BCN) that automatically produces the labeled mask with the luminal and wall boundaries of the coronary artery. We utilized 50 patients of CCTA - intravascular ultrasound matched image data sets. Training and testing were performed on 40 and 10 patient data sets, respectively. The prediction of luminal and wall mask was performed using stacked BCN on the each image view: axial, coronal, and sagittal of straightened curved planar reformation. We defined the vector that includes probability from BCN result on each image view and proposed amplified probability. We used an Adaptive Boost regressor with an extremely randomized tree regressor to determine the label for unknown probability vector.

In this paper, a model predictive current control (MPCC) strategy with a fixed switching frequency to control the dual boost bridgeless power factor corrected (PFC) converter is proposed. The improved MPCC strategy solves the problem of filter design and the high electromagnetic interference (EMI) which caused by the traditional MPCC. With the consideration of a nonlinear inductor, a formula is fitted to represent the change of the inductance, and the improved strategy has strong robustness. The simulation and experiment shows that the quality of current and power factor of the system is improved by the proposed control strategy.

This case report presents spontaneous resolution of acute rejection in a 66-year-old man who underwent a kidney transplant and developed acute rejection and pneumonia. Our main concern in this case was how to treat the concurrent infection while maintaining the immunosuppressive therapy with a narrow available therapeutic range, in order to save the renal allograft without increasing antirejection therapy.

This study aimed to determine the expression of integrin β1 and vascular endothelial growth factor (VEGF) and microvascular density (MVD) by CD105 staining in hypopharyngeal squamous cell carcinoma to determine their association with clinicopathologic characteristics, and to determine their role and the effects of their interactions in the development and progression of hypopharyngeal squamous cell carcinomas. The expression of integrin β1 and VEGF and MVD in hypopharyngeal squamous cell carcinomas and normal hypopharyngeal tissues were evaluated using immunohistochemistry. The Image-Pro Plus software was used to determine the mean optical density of the immunohistochemical images. Integrin β1 expression was significantly higher in hypopharyngeal squamous cell carcinoma tissues (78.00%) than in normal hypopharyngeal tissues (35.00%; P = 0.001) and significantly differed across pathologic grades and different T stages, and regarding the presence of cervical lymph node metastasis (P0.05). VEGF expression was significantly higher in hypopharyngeal squamous cell carcinoma tissues (74.00%) than in normal hypopharyngeal tissues (30.00%; P = 0.002), VEGF overexpression differed significantly across different pathologic grades and different T stages, and regarding the presence of cervical lymph node metastasis (P0.05). The MVD count was significantly higher in hypopharyngeal squamous cell carcinoma tissues (37.10 ± 5.95) than in normal hypopharyngeal tissues (8.70 ± 3.34; P = 0.000). MVD differed significantly across different pathologic grades and different T stages, and regarding the presence of cervical lymph node metastasis (P0.05). The expression of integrin β1 and VEGF and the MVD count exhibited no significant differences in terms of age, gender, history of smoking, and clinical stages (P0.05). VEGF expression was positively associated with the MVD count of hypopharyngeal squamous cell carcinomas (r = 0.582, P = 0.000); however, integrin β1 was not associated with VEGF or MVD (P0.05). Integrin β1 and VEGF are overexpressed and MVD increased in hypopharyngeal squamous cell carcinomas. VEGF is positively correlated with MVD.

In the normal retinotectal topography established during the embryonic development of the chick visual system, retinal ganglion cell axons from the nasal retina connect to the posterior part and temporal retinal axons connect to the anterior part of the optic tectum. For the investigation of position-specific gene expression along the nasal-temporal axis of the retinal neuroepithelium (RN), differential display PCR was carried out from the nasal or temporal part of the RN at HH11 (E2). We found several genes that are differentially expressed either in the nasal or in the temporal part of the RN and the analysis of the asymmetrically expressed fragments showed at least one cDNA fragment to be exclusively expressed in the nasal RN. This fragment was 550 bp in size.

10. Spinal deformities in Noonan syndrome: a clinical review of sixty cases

Skeletal abnormalities, including spinal deformities, in Noonan syndrome have been described, but no detailed and systematic study of such spinal deformities has been presented in the literature.The cases of sixty patients with Noonan syndrome were reviewed retrospectively, and the general appearance, growth disturbance, and mental status of the patients were documented. Spinal deformities were evaluated radiographically, and the frequency, pattern, and severity of the curves were documented.Spinal deformity was present in eighteen (30%) of the sixty patients. Two patients had congenital spinal deformity. Of the remaining sixteen patients with scoliosis, nine had a single thoracic curve, four had a single thoracolumbar curve, and three had a double major curve. Thoracic lordosis was also present in three of these sixteen patients. No patient had only increased kyphosis or lordosis. The mean age when the spinal deformities were detected was nine years; seven deformities were detected before the age of seven years. Overall, surgery was recommended to eleven of the eighteen patients; it was recommended for the treatment of scoliosis (mean, 68.5 degrees; range, 45 degrees to 125 degrees ) in eight patients and for the treatment of an associated thoracic lordosis (8 degrees, 15 degrees, and 18 degrees ) in three. Seven of the eleven patients underwent spinal arthrodesis. The operation was deferred in one patient because malignant hyperthermia developed during the induction of anesthesia.Scoliosis with an associated thoracic lordosis occurs more frequently in Noonan syndrome than has been reported previously. Since the deformities tend to develop early and are relatively severe, a clinical and, if necessary, radiographic assessment of the spine with careful follow-up should be performed for early detection and treatment of spinal deformity. Although malignant hyperthermia is rare, all patients with Noonan syndrome should be considered to be at risk for the development of this complication before operative treatment.

11. Expression of the CIP1 gene induced under cadmium stress in Candida sp

Cadmium is a toxic heavy metal with no known biological activity. Differential display of mRNA was employed to isolate cDNA corresponding to the transcript that is induced in cadmium-treated Candida sp. In this report we describe the molecular characterization of the CIP1 gene, which was shown to be rapidly induced after cadmium treatment. Northern blot analysis showed that the CIP1 transcript was not present in normal cells, but accumulated at higher levels in cadmium-treated cells. Treatment of other heavy metals such as copper, mercury, lead, zinc, or heat-shock had no effect on the expression of the CIP1 gene. Sequence analysis of CIP1 revealed that it encodes a 32 kDa hydrophobic protein that contains a putative transmembrane domain. The deduced amino acid sequence of CIP1 showed a little homology with isoflavone reductase of plants. From the promoter sequence analysis, we also identified a sequence similar to pas, a cadmium-responsive element of the ParA gene in tobacco. Our results suggest that Candida CIP1 may play a crucial role in the establishment of specific cellular response to stress evolved by the cadmium treatment.

12. Purification and characterization of protease Ci, a cytoplasmic metalloendoprotease in Escherichia coli

Protease Ci, a cytoplasmic metalloprotease in Escherichia coli, has been purified to apparent homogeneity by conventional chromatographic procedures using 125I-labeled oxidized insulin B-chain as a substrate. The purified enzyme behaves as a 54-kDa protein under both denaturing and nondenaturing conditions, suggesting that it consists of a single polypeptide chain. It is inhibited by metal-chelating agents, including o-phenanthroline and NaCN, but not by inhibitors of serine proteases or thiol-blocking agents. Furthermore, protease Ci was found to contain 1.1 mol of zinc per mol of the enzyme upon analysis by HR ICP mass spectroscopy. Thus, protease Ci must be a zinc metalloprotease. Among the polypeptides tested as substrates, oxidized insulin B-chain and glucagon are most rapidly hydrolyzed. Intact insulin is a much poorer substrate than oxidized insulin B-chain, even though the affinity of the enzyme to intact insulin is approximately 100-fold greater than that to the B-chain. Since unlabeled oxidized insulin A-chain is capable of inhibiting the hydrolysis of 125I-labeled insulin B-chain, it also appears to be a substrate. Protease Ci also degrades lysozyme and lactalbumin, although to a much lesser extent than oxidized insulin B-chain. However, it shows little or no activity against proteins larger than 15 kDa (e.g. ovalbumin and denatured bovine serum albumin). Hydrolysis of oxidized insulin B-chain followed by amino acid composition analyses of the cleavage products reveals that as many as 10 of its 29 peptide bonds are hydrolyzed by protease Ci. This ability to hydrolyze relatively small polypeptides suggests that protease Ci may catalyze the later steps in the pathway for intracellular protein breakdown.

The importance of the P1 reactive site for the specificity of ecotin on target proteases was examined by site-directed mutagenesis. The replacement of Met at the P1 site with Ile, Arg, Glu, or Tyr showed little or no effect on the ability of ecotin to inhibit trypsin. Similar results were obtained for chymotrypsin, except that its replacement with Glu caused about 40% reduction of the inhibitory activity of ecotin. On the other hand, the replacement of the Met residue with Arg, Tyr, or Glu dramatically reduced its ability to inhibit elastase, while that with Ile showed little or no effect. Nevertheless, elastase could be completely inhibited upon incubation with excess amounts of the mutant ecotin containing Arg, Glu, or Tyr. Moreover, all the mutant forms of ecotin could be cleaved at the mutated P1 site upon incubation with trypsin at pH 3.75. In addition, the replacement of a Cys residue in the disulfide bridge with Ser showed little or no effect on the ability of ecotin to inhibit trypsin, chymotrypsin, or elastase. However, the mutant ecotin containing Ser was more sensitive to inactivation by heating at 100 degrees C than the wild-type inhibitor. Furthermore, the wild-type ecotin whose disulfide bond had been reduced and alkylated was also more easily inactivated by heat treatment than the untreated control. These results strongly suggest that the P1 site of ecotin is not crucial for its specificity on target proteases and that the disulfide bridge in ecotin appears to play an important role in maintenance of its structural stability.

15. Purification of Acidic Fibroblast Growth Factor from Bovine Heart and Its Localization in the Cardiac Myocytes

We found an endogenous growth factor, referred to here as heart-derived growth factor (HDGF), that stimulates the proliferation of vascular endothelial cells. HDGF was purified from bovine myocardium using a procedure that involves denaturation of undesired proteins with methanol and chloroform. Soluble HDGF was purified essentially to homogeneity in a single step by heparin affinity chromatography. The purified HDGF was identified to be acidic fibroblast growth factor based on the following properties: molecular weight of 18,000, isoelectric point of 5.2, amino acid composition and sequence, its dissociation from a heparin affinity column at 0.9 M NaCl, potentiation of activity in the presence of heparin, and antigenicity. Our yield of HDGF was 500 µg/kg of tissue. Antiserum raised to HDGF localized HDGF in the cardiac myocytes in culture. These data indicate that a large amount of acidic fibroblast growth factor is present in the heart, and the cardiac myocytes are likely to be a major source of it.

16. Enhancement of Calcium Sensitivity of Lipocortin I in Phospholipid Binding Induced by Limited Proteolysis and Phosphorylation at the Amino Terminus as Analyzed by Phospholipid Affinity Column Chromatography

A phospholipid column was prepared by coating siliconized porous glass beads with phospholipids. The analysis of the Ca2+ requirement of lipocortin I and its derivatives in the binding to phospholipids was carried out with this column. The Ca2+ concentration required for 50% binding to the phospholipid column at room temperature was about 30 microM for lipocortin I, while that was reduced to 15 microM when lipocortin I was phosphorylated by the epidermal growth factor receptor/kinase, and a further reduction in the Ca2+ requirement was observed with proteolytic cleavage at the N-terminal region. Cathepsin D and calpain I (low calcium-requiring form of calcium-activated neutral protease) rapidly cleaved human placental lipocortin I at Trp-12 and Lys-26, respectively. These N-terminal-truncated proteins required only 5 microM Ca2+ for 50% binding to the phospholipid column. This enhancement of Ca2+ sensitivity by limited proteolysis was also observed for porcine lung lipocortin I. Essentially the same results were obtained when the Ca2+ sensitivities of the modified lipocortins I were analyzed using dispersed phospholipid vesicles instead of the phospholipid affinity column. Equilibrium dialysis indicated that the release of the N-terminal region markedly increased the affinity of lipocortin I for Ca2+ in the presence of phosphatidylserine, without any appreciable change of the number of Ca2+-binding sites. Limited proteolysis by endogenous proteases such as calpain may be an important regulatory mechanism for the Ca2+ sensitivity of lipocortin I in phospholipid binding.

To determine the phosphate binding sites in hen egg white riboflavin binding protein (RBP), a highly phosphorylated peptide, which consisted of 23 amino acid residues including eight phosphoserines, was isolated from the tryptic digest of reduced and carboxymethylated RBP. The conditions of the beta-elimination-addition reaction to convert phosphoserine residues in the peptide to cysteic acids, S-methylcysteines, alanines, and beta-methylaminoalanines (DL-alpha-amino-beta-methylamino propionic acid) were examined. These converted peptides were purified by HPLC and subjected to Edman degradation. The results of Edman degradation indicated that the S-methylcysteine derivative of the peptide gave the most satisfactory result for determining the phosphate binding sites in the peptide. The phosphorylation sites of the peptide determined by the method mentioned above are as follows: His182-Leu-Leu-Ser185-Glu-Ser(P)-Ser(P)-Glu-Glu190-Ser (P)-Ser(P)-Ser(P)-Met-Ser195(P)-Ser(P)-Ser(P)-Glu-Glu-. These studies indicated that the conversion of phosphoserines in phosphoproteins to S-methylcysteines followed by Edman analysis was a useful method for the elucidation of the phosphorylation sites in phosphopeptides.

18. Insulin-stimulating peptide from a tryptic digest of bovine serum albumin: purification and characterization

A procedure was established for isolation of a low molecular weight polypeptide with insulin-stimulating activity in apparent homogeneity from a tryptic digest of bovine serum albumin on a semipreparative scale. Purification of this insulin-stimulating peptide (ISP) was monitored by an adipose-explant assay in which stimulation of fatty acid synthesis from glucose by insulin was measured. The polypeptide was purified by a combination of DEAE-cellulose column chromatography, gel filtration on Bio-Gel P-10, hydrophobic chromatography on a semipreparative C18 reversed-phase HPLC column, and ion exchange chromatography on an SP-5PW HPLC column. The primary structure of ISP was deduced. ISP is a two-chain polypeptide consisting of 71 amino acid residues, and corresponds essentially to residues 115-143 and 144-184 (185) of bovine serum albumin connected to each other by a disulfide bridge. But comparison of the sequence of ISP with that of the relevant regions of bovine serum albumin determined by Brown indicated the presence of one tyrosine insertion between residues 155 and 156 of albumin. Therefore, the molecular weight of ISP was calculated to be 8,496.

A bacteriolytic enzyme obtained from the culture fluid of Staphylococcus aureus FDA 209P was purified to homogeneity utilizing dye-ligand affinity column chromatography, hydrophobic interaction high pressure liquid chromatography (HPLC) and hydroxyapatite HPLC. Subsequent characterizations indicated that the purified enzyme acted as endo-beta-N-acetylglucosaminidase. The molecular weight determined by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) was 51,000 and the isoelectric point was higher than 10. The optimum pH for the enzyme activity on whole cells of Micrococcus luteus as a substrate was 8.0. Some heavy metal cations (Cu2+ and Zn2+) inhibited the enzyme activity at a concentration of 0.1 mM and others (Ba2+, Mg2+ and Co2+) showed a stimulating effect at a concentration of 1 mM.

The sources of noise present in coherent optical systems are reviewed and classified according to their origin. It is shown that the ultimate limit of image quality is the noise present in the film. The measurable film parameters (MTF, granularity, resolution limit, and threshold modulation) influencing the performance of coherent optical processing systems are combined to set forth a performance criterion that defines the image quality attainable in a given experiment. Sources of noise originating within the system, rather than from the film, can be lessened or eliminated by various techniques that effectively reduce the degree of spatial and/or temporal coherence of the system. These techniques create an ensemble of random noise patterns superimposed upon a stationary image. This superposition averages the noise to a uniform background and reduces the contrast of the image. This system noise reduction leaves the film noise as the limiting parameter in the system. Both holographic and coherent imaging experiments are compared in terms of noise reduction techniques. Five experimental procedures to be considered when designing a coherent optical system with minimum noise are recommended, and examples of their application are demonstrated.

The development and application of second generation high temperature superconducting (2G-HTS) tapes have attracted much attention in China recently. Progress in upscaling high performance 2G-HTS tape production at Shanghai Superconductor Technology (SST) is reported in this paper. With ion beam assisted deposition, biaxially textured buffer layers with a configuration of CeO
2 /LaMnO3 /MgO/Y2 O3 /Al2 O3 /C-276 have successfully been fabricated. In-plane and out-of-plane texture degrees of CeO2 films achieve 2°–4° and 2°, respectively. A multi-plume multi-turn pulsed laser deposition (PLD) system combined with the so-called ‘radiation assisted conductive heater’ has been proposed and further developed for REBCO layer deposition. Our effort was focused on minimizing the temperature variations in the deposition region by modifying the heating shield that assists the conductive heater of the drum-like cylinder. A tape travelling speed of 100–180 m h−1 can be achieved with a steady temperature profile when passing through the deposition zone, which is very beneficial for the growth of the REBCO layer. Taking advantage of the liquid phase growth mode, several compositions of superconducting films with a thickness in the range of 1–2.5 μm have been grown with high growth rates of over 40 nm s−1 . Furthermore, the microstructures and superconducting performance were investigated in detail. Based on these studies, superconducting tapes with piece lengths of up to 500 meters have been developed. High Ic values at 77 K, self-field (over 520 A cm−1 width) or at low temperature, high magnetic field conditions (over 560 A/4 mm width at 4.2 K, 10 T, perpendicular field) have been achieved.
