Abstract

Yüksek Lisans Tezi., Vicia cinsi Fabaceae familyasının Papilionoideae alt familyasının bir üyesidir ve ülkemizde Vicia, Cracca, Ervum ve Faba olmak üzere dört seksiyon ile temsil edilmektedir. Vicia cinsinin Türkiye'de yayılış gösteren 66 türü, 27 alttürü ve 29 varyetesi bulunmaktadır. Bu çalışmada Vicia cinsinin Türkiye'de doğal olarak yayılış gösteren yedi taksonunun kromozom sayısı ve morfolojisi Görüntü Analiz Sistemi ile belirlenmiştir. Çalışılan taksonların somatik kromozom sayısı 2n=12 (Vicia cassubica, V. noeana. var. noeana, V. sativa subsp. sativa, V. caesarea) ve 2n=14 (Vicia articulata, V. peregrina, V. villosa subsp. villosa) arasında değişmektedir. Taksonlara ait kromozom morfolojileri ise farklı tespit edilmiş olup karyotip formülleri; Vicia articulata'da 7m, V. caesarea 4m+2sm, V. cassubica 6sm, V. noeana var. noeana 6sm, V. peregrina 5sm+2st, V. sativa subsp. sativa 5sm+1st, V. villosa subsp. villosa 7sm şeklindedir., The genus Vicia is located in Fabaceae and it is a member of subfamily Papilionoideae. It is represented with four sections that name are Vicia, Cracca, Ervum ve Faba in our country. The genus Vicia distrubuted in Turkey with 66 species, 27 subspecies and 29 varieties. In this thesis study naturally grown in Turkey Vicia genus's seven taxa and morphology are determined by Image Analysis System. Studied taxa's somatic chromosome number change between 2n=12 (Vicia cassubica, V. noeana var. noeana, V. sativa subsp. sativa, V. caesarea) and 2n=14 (Vicia articulata, V. peregrina, V. villosa subsp. villosa). Chromosome morphology of belongin to taxa's result are different and that's koryotype formulas are Vicia articulata'da 7m, V. caesarea 4m+2sm, V. cassubica 6sm, V. noeana var. noeana 6sm, V. peregrina 5sm+2st, V. sativa subsp. sativa 5sm+1st, V. villosa subsp. villosa 7sm.

3. THE EFFECTS OF SINAPIC ACID ON TRP CHANNELS EXPRESSION IN LNCAP PROSTATE CANCER CELL LINE.

Abstract

OBJECTIVES: The aim of study was to evaluate effects of SA on TRP channel expression in LNCaP human prostate cancer cells. TRP family of ion channel expression changes were observed in different cancers including breast, brain, prostate, ovary and colon. Prostate cancer is one of the leading threats to men's health in the world. There are still limited therapeutic options for metastatic prostate cancer. Sinapic acid (SA) is a naturally occurring phenolic compound that can be found in free form. SA is also found in various fruits, vegetables, cereals, oilseeds and some spices. MATERIALS AND METHODS: The effect of SA on cell viability was determined using XTT assay. After SA treatment, total RNAs were isolated and cDNAs were synthesized. Expressions of ion channels genes including TRPM2, TRPM7, TRPM8 and TRPV2 were evaluated by qPCR. RESULTS: The IC50 dose of SA in LNCaP cell line was found to be 1000 µM for 72 hour. SA treatment increased the expression of TRPV2. However, expressions of TRPM2 and TRPM8 were significant decreased the in LNCaP cells. An insignificant increase was observed for TRPM7 mRNA expression. CONCLUSIONS: Changes in the expression of TRP ion family may have an effect on cell viability, cell invasion, migration and apoptosis. The decreased expression of TRPM2 and TRPM8 may induce apoptotic cell death by reducing Ca+2 input. Elevated TRPM7 and TRPV2 expressions were found to be statistically insignificant. 5. Detailed chromosome measurements and karyotype asymmetry of some Vicia(Fabaceae) taxa from Turkey

Abstract

ABSTRACTThis study examined the detailed chromosome measurements and karyotype asymmetries of seven taxa in the genus Vicia. The taxa are V. articulata, V. cassubica, V. villosasubsp. villosain sect. Cracca, V. noeanavar. noeana, V. sativasubsp. sativa, V. peregrinain sect. Viciaand V. caesareain sect. Ervum. V. cassubica, V. noeanavar. noeana, V. sativasubsp. sativa, V. caesareahave 2n = 12 chromosomes. V. articulata, V. villosasubsp. villosa, V. peregrinahave 2n = 14 chromosomes in somatic cells. Total chromosome lengths range between 2.93–4.99 µm in V. articulata, 2.09–4.73 µm in V. cassubica, 1.86–3.36 µm in V. villosasubsp. villosa, 4.23–6.05 µm in V. noeanavar. noeana, 2.07–3.72 µm in V. sativasubsp. sativa, 4.32–7.21 µm in V. peregrinaand 2.39–5.78 µm in V. caesarea. The detailed chromosome measurements, relative lengths, centromeric indexes and karyotype asymmetries are also given. V. articulatais the most symmetrical karyotype, while V. villosasubsp. villosais the most asymmetrical karyotype in intrachromosomal asymmetry including parameters of MCA, AsK, TF, Syi, A1, and A. However, the asymmetrical karyotypes are different in interchromosomal asymmetries. While V. noeanavar. noeanais the most symmetrical karyotype in CVCL, Rec, and A2. V. caesareais the most asymmetrical karyotype in only CVCLand A2. Unlike all parameters, V. cassubicais the most asymmetrical karyotype in Rec value. The scatter diagrams are given between MCA–CVCLand Syi–Rec.
