Summary Maillard reaction can enhance the functional characteristics of the protein. Herein, we investigated changes in the physicochemical and biological properties of gelatin (GA)‐xylooligosaccharide (XOS) conjugates following Maillard reaction. The results showed that the browning intensity of the conjugates increased with the prolongation of heating time. Additionally, sodium dodecyl sulphate‐polyacrylamide gel electrophoresis revealed some macromolecule polymer formed, and the grafting degree of GA was augmented to 40.05% with prolongation of heating time. GA glycation with XOS decreased α‐Helix content and increased level of β‐Turn level, and exerted a slight impact on contact angle. Moreover, the results of in vitro simulated gastrointestinal digestion analysis showed that the GA‐XOS mixture enhanced resistance to digestion after Maillard reaction for 60 min, affording hydrolysis ratios ranging from 17.86–27.79% and 25.03–35.47% in simulated gastric fluid and intestinal fluid, respectively. The antioxidant ability and heat stability of the conjugates was visibly enhanced following heating treatment. Additionally, increasing the mass of XOS accelerated the antioxidant capacities of GA‐XOS conjugates. This study aimed to evaluate the impact of probiotic Lactiplantibacillus plantarum S61 and prebiotic Xylooligosaccharide (XOS) on the physicochemical (pH, Titratable acidity, and °Brix), functional (color, total phenolic content, antioxidant activity, Vitamin C content), and sensory properties of orange juice during fermentation at 37 °C for 48 h. The fermented juice displayed favorable physicochemical characteristics (pH, Titratable acidity, and °Brix), important growth of the probiotic L. plantarum S61 (p < 0.05), and enhanced color and shelf life of the final product. The results showed that the values of total phenolic content, antioxidant activities, and Vitamin C range between 55.33 and 38.50 µM, 50.68–63.57%, and 17.5-23.75 mg/100 g, respectively, at the end of the fermentation. Furthermore, Probiotic and prebiotic supplementation enhanced significantly (p < 0.05) the total phenolic content, and antioxidant activity, and preserved the degradation of Vitamin C during fermentation. Moreover, there were no significant differences between the formulations regarding color, texture, and overall acceptance (p > 0.05). Consequently, the orange juice supplemented Xylooligosaccharide (prebiotic) and probiotic cultures could be applied to develop a new non-dairy synbiotic beverage product with important and high functional values and acceptance sensory properties. The primary aim of this study was to examine the impact of xylooligosaccharide (XOS) in rice protein concentrate (RPC) based diets on the growth performance, body composition, digestive enzymes, intestinal morphology and blood biochemistry of Labeo rohita fingerlings. Four different XOS levels (0%, 0.5%, 1% and 2%) were used at each RPC (75% and 100%) level. Twenty‐five fish per tank with an average initial weight of 25 ± 0.05 g were randomly assigned (Randomised complete block design) to each of the 8 groups in triplicate aquaria (36 × 16 × 12″) and then fed with respective diets @ 3% body weight for 90 days. The results showed significant improvements in growth performance, such as increased weight gain %, specific growth rate, and protein efficiency ratio and improved feed conversion ratio in 1% XOS supplemented diet at 75% RPC. A significant decrease in serum alkaline phosphatase activity (ALP) and plasma melanodialdehyde (MDA) were observed at 1% XOS level in 75% RPC based diets, respectively. Meanwhile, the lowest total cholesterol and highest lysozyme activity were observed in 1% XOS supplemented diet at 75% RPC levels. Moreover, the serum (alanine aminotransferase and aspartate transaminase) and plasma (superoxide dismutase, triglyceride, high density and low density lipoprotein) activities showed nonsignificant effects among the treatments. Furthermore, the digestive enzymes (protease & lipase) and intestinal morphology were significantly influenced at 1% XOS in the 75% RPC‐based diet. Polynomial regression analysis showed that 1.25% XOS is the optimum requirement for the growth of rohu fingerlings when fed at 75% RPC based diets. Stimbiotics are a new category of feed additives that can increase fibre fermentability by stimulating fibre-degrading microbiota in the gut. The aim of this study was to test, ex vivo, if the microbiota of broilers fed a stimbiotic are better able to ferment different xylose-rich substrates in an ileal and a caecal environment. The ileal and caecal contents from broiler chickens fed a stimbiotic or from a control group were used as an inoculum in the ex vivo fermentation experiment. Different xylose-rich substrates including monomeric xylose (XYL), XOS with DP 2 to 6 (XOS), short DP XOS of 2 to 3 (sDP-XOS), long DP XOS of 4 to 6 (lDP-XOS) and de-starched wheat bran (WB), were added to each ileal and caecal inoculum in fermentation vessels. Total gas, short-chain fatty acids (SCFA) production, bacterial quantification, and carbohydrate utilisation were monitored for 9 h post-inoculation. No significant interactions were observed in any of the parameters measured in either the ileal or caecal contents (p > 0.05). Stimbiotic ileal inocula resulted in higher total gas (p < 0.001) and volatile fatty acid (VFA) (p < 0.001) production, increased numbers of Lactobacillus spp. (p < 0.001), and decreased numbers of Enterococcus spp. (p < 0.01) after 9 h regardless of the xylose-rich substrate added. Stimbiotic caecal inocula resulted in a higher ratio of VFA to branched-chain fatty acids (BCFAs) by up to +9% (p < 0.05). Ileal microbiota were found to preferentially metabolise WB, while caecal microbiota favoured XOS substrates, particularly lDP-XOS. These results indicate that stimbiotics can promote the abundance of lactic acid bacteria involved in the establishment of fibre-degrading bacteria and VFA content in the gut, which could have beneficial effects on broiler performance. The purpose of this study was to explore the ability of different bifidobacterial species to utilize xylooligosaccharide (XOS). It focused on the mechanism for the efficient utilization of XOS by Bifidobacterium pseudolongum JNFEN6 through cell physiological analysis and transcriptomic analysis. The results showed that XOS had different effects on the growth of different Bifidobacterium species, and it had the most significant effect on the growth and acid-producing capacity of B. pseudolongum JNFEN6. When B. pseudolongum JNFEN6 was cultured for 36 h on XOS as the sole carbon source, the activity of β-xylosidase produced by this strain was 0.97 U/mL, and the concentration of propionic acid in the fermentation system was 85.26 μg/mL. The transcriptomic results showed that a total of 297 differentially expressed genes were identified, including 136 up-regulated genes and 161 down-regulated genes. Among them, the ATP-binding cassette (ABC) transport permease, substrate-binding protein and MFS transporter genes promoted the transport of XOS, while the XOS metabolic hydrolase, acetate kinase and lactate dehydrogenase genes promoted the metabolism of XOS. In summary, B. pseudolongum JNFEN6 can efficiently utilize XOS, which can take in XOS, mainly through the ABC transport system and the MFS transport system, and metabolize XOS through the ‘bifidus shunt’ pathway, finally producing short-chain fatty acids or other organic compounds. This study provides a theoretical basis for the application of XOS as a bifidus factor.

Glycosylated whey protein (GWP) was prepared from whey protein and xylooligosaccharide by wet glycosylation method. Its advantages for use as a fat substitute were analyzed, and the application of this fat substitute in reduced-fat processed cheese was explored. The results showed that the optimal preparation conditions were determined as: xylooligosaccharide concentration of 6%, reaction temperature of 85 ℃ and reaction time of 2 h. The grafting degree of GWP prepared using these conditions was (29.28 ± 1.53)%, and a browning phenomenon occurred during the preparation process. Compared with whey protein, the contents of α-helix, β-fold, β-turn and random coil structures changed in GWP, and the water and oil-holding properties increased by 60.18% and 103.97%, respectively; the emulsifying capacity and emulsion stability increased by 30.81% and 13.57%, respectively. The surface of GWP was smoother. The surface hydrophobicity at pH 7.0 was 2.48 times higher than that of whey protein. After shearing at 10 000 r/min for 10 minutes, the median particle diameter of GWP was 7.57 µm, and the stability of the solution was obviously improved, so that GWP could be used to simulate fat. The hardness of reduced-fat processed cheese with 60% butter replacement by GWP was significantly lower than that of full-fat processed cheese (P 0.05), and the total sensory score was equivalent to that of full-fat processed cheese. These results proved that the glycosylated whey protein has a potential application as a fat substitute in reduced-fat food while guaranteeing that the quality of the product is good.

ABSTRACT: Xylooligosaccharide (XOS) is known as a prebiotic, however, it is unknown whether XOS can directly protect against bacterial infection. This study aimed to investigate the direct inhibitory effects of XOS on Salmonella Typhimurium colonization and the inductive impairments in gut health and growth performance in broilers. We first probed the inhibitory effects of XOS on S. Typhimurium adhesion and its induction of intestinal epithelial cell (IPEC-J2) injuries. Afterward, 168 one-day-old yellow-feathered broilers were randomly divided into 3 groups (7 replicates/group): negative control (NC, received a basal diet), positive control (PC, received a basal diet with S. Typhimurium challenge) and XOS group (PC birds + 1,500 mg/kg XOS). All birds except those in NC were orally challenged with S. Typhimurium from 8 to 10 d of age. Parameters were analyzed on d 11. The results showed that XOS inhibited S. Typhimurium adhesion and the inductive injuries of IPEC-J2 cells by lowering (P < 0.05) certain adhesion-related genes expression of this bacterium. It also alleviated S. Typhimurium-induced increase (P < 0.05) in the expression of certain inflammatory cytokines and tight junction (TJ) proteins of IPEC-J2 cells. Supplementing XOS to S. Typhimurium-challenged broilers attenuated the elevations (P < 0.05) in S. Typhimurium colonization of ileal mucosa and its translocation to the liver and spleen, as well as increased (P < 0.05) certain TJ proteins expression of ileum. Besides, XOS addition normalized S. Typhimurium-induced impairments (P < 0.05) in ileal morphology, final body weight and average daily gain in broilers. Collectively, supplemental XOS directly suppressed intestinal colonization of S. Typhimurium by diminishing its adhesiveness and subsequently mitigated destructions in intestinal barriers, thus contributing to weaken growth retardation in challenged broilers. Our findings provide a new insight into the mechanisms of XOS limiting Salmonella infection in chickens.

Endo-xylanase hydrolyzing xylan in cellulosic residues releasing xylobiose as the major product at neutral pH are desirable in the substitute sweeteners industry. In this study, two endo-xylanases were obtained from Streptomyces rochei and Bacillus velezensis. SrocXyn10 showed the highest identity of 77.22%, with a reported endo-xylanase. The optimum reaction temperature and pH of rSrocXyn10-Ec were pH 7.0 and 60°C, with remarkable stability at 45°C or pHs ranging from 4.5 to 11.0. rBvelXyn11-Ec was most active at pH 6.0 and 50°C, and was stable at 35°C or pH 3.5 to 10.5. Both rSrocXyn10-Ec and rBvelXyn11-Ec showed specific enzyme activities on wheat arabinoxylan (685.83 ± 13.82 and 2809.89 ± 21.26 U/mg, respectively), with no enzyme activity on non-xylan substrates. The Vmax of rSrocXyn10-Ec and rBvelXyn11-Ec were 467.86 U mg−1 and 3067.68 U mg−1, respectively. The determined Km values of rSrocXyn10-Ec and rBvelXyn11-Ec were 3.08 g L−1 and 1.45 g L−1, respectively. The predominant product of the hydrolysis of alkaline extracts from bagasse, corncob, and bamboo by rSrocXyn10-Ec and rBvelXyn11-Ec were xylooligosaccharides. Interestingly, the xylobiose content in hydrolysates by rSrocXyn10-Ec was approximately 80%, which is higher than most reported endo-xylanases. rSrocXyn10-Ec and rBvelXyn11-Ec could be excellent candidates to produce xylooligosaccharides at neutral/near-neutral pHs. rSrocXyn10-Ec also has potential value in the production of xylobiose as a substitute sweetener.

The study was to investigate the effect of different levels of dietary xylooligosaccharides on growth performance, immune function, nutrient apparent digestibility and ruminal fermentation parameters of finishing Hu sheep. Sixty male lambs of 2.5 months old and weighing (20.10±1.59) kg were randomly divided into four groups, with three replicates in each group and five lambs in each replicate. The control group, group A, group B and group C were supplemented with 0, 150, 300 and 450 mg/kg xylooligosaccharides in basal diet, respectively. The pre-test period was 7 d, and the formal test period was 90 d. The results showed that there were no significant differences in body weight, average daily gain, average daily feed intake and feed to gain ratio among all groups ( P>0.05). The apparent digestibility of calcium of Hu sheep in group B and group C was significantly higher than that in control group ( P<0.05). Serum IgA content of Hu sheep in group C was extremely higher than that in control group ( P<0.01), and serum IgA content of lake sheep in group B was significantly higher than that in control group ( P<0.05). The serum IgG content of Hu sheep in groups B and group C was extremely higher than that in control group ( P<0.01). The study indicates that supplementation of 300 and 450 mg/kg xylooligosaccharides significantly increased the content of IgA and IgG in serum and the apparent digestibility of calcium, and increased the average daily gain of finishing Hu sheep. According to the production practice, it is suggested that 300 mg/kg of xylooligosaccharides should be added to the diet of finishing Hu sheep. [ABSTRACT FROM AUTHOR]

Xylooligosaccharides (XOS) are one of the emerging oligomers exhibiting prebiotic activity. They are mainly produced by the hydrolysis of cheap and abundant lignocellulosic feedstocks. It has been demonstrated that the oligomers with low degree of polymerization (2–4) are more selective for the microbiota. Therefore, several attempts have been made to develop efficient processes for the production of such types of XOS by employing different strategies. Microbial xylanases possess a crucial role in these processes. They are highly specific biocatalysts that do not produce any undesirable byproducts during oligomer production. This review provides detailed information about the xylanases belonging to different glycoside hydrolase families with respect to their ability to produce XOS. The enzyme integrated methodologies for XOS production are described from a commercial perspective. The studies focusing on XOS production by xylanase enzymes from different microbial sources were summarized in a strain specific manner for the first time. Lytic polysaccharide monooxygenases (LPMOs) are redox-enzymes involved in biomass degradation. All characterized LPMOs possess an active site of two highly conserved histidine residues coordinating a copper ion (the histidine brace), which are essential for LPMO activity. However, some protein sequences that belong to the AA9 LPMO family display a natural N-terminal His to Arg substitution (Arg-AA9). These are found almost entirely in the phylogenetic fungal class Agaricomycetes, associated with wood decay, but no function has been demonstrated for any Arg-AA9. Through bioinformatics, transcriptomic, and proteomic analyses we present data, which suggest that Arg-AA9 proteins could have a hitherto unidentified role in fungal degradation of lignocellulosic biomass in conjunction with other secreted fungal enzymes. We present the first structure of an Arg-AA9, LsAA9B, a naturally occurring protein from Lentinus similis The LsAA9B structure reveals gross changes in the region equivalent to the canonical LPMO copper-binding site, whereas features implicated in carbohydrate binding in AA9 LPMOs have been maintained. We obtained a structure of LsAA9B with xylotetraose bound on the surface of the protein although with a considerably different binding mode compared with other AA9 complex structures. In addition, we have found indications of protein phosphorylation near the N-terminal Arg and the carbohydrate-binding site, for which the potential function is currently unknown. Our results are strong evidence that Arg-AA9s function markedly different from canonical AA9 LPMO, but nonetheless, may play a role in fungal conversion of lignocellulosic biomass.

Abstract Background Two experiments were conducted to establish an optimal NE challenge model and evaluate the efficacy of stimbiotic (STB) supplementation in necrotic enteritis (NE) challenged broilers. In Exp. 1, a total of 120 Arbor Acres (AA) broilers (45.0 ± 0.21 g) were randomly assigned to 6 treatments in a 3 × 2 factorial arrangement. Vaccine treatments included non-challenge (0), × 10 the recommended dose (× 10) or × 20 the recommended dose (× 20) by the manufacturer. Clostridium perfringens (CP) treatments were non-challenge (No) or 3 mL of 2.2 × 107 CFU CP challenge (Yes). Improvements in the abundance and diversity of ileal microbial communities within the EXA, EXF, and XAF treatments were observed (P < 0.05). Positive correlations between microbiota and XOS were revealed, with xylobiose and xylotriose being critical for ten beneficial bacteria (P < 0.05). EXF increased the BWG and FCR of broiler chickens in this phase (P < 0.05), which was attributed to the thriving networks modified by Lactobacillus. The intracecal contents of acetic acid, butyric acid, and propionic acid were greatly enhanced in most ADE groups, such as EXF (P < 0.05). Conclusions: Debranching enzymes appreciably targeted corn AX to release prebiotic XOS in the posterior ileum and facilitated intracaecal fermentation. Xylans, a family of xylose-based polysaccharides, are dietary fibers resistant to digestion. They therefore reach the large intestine intact; there, they are utilized by members of the gut microbiota. They are initially broken down by primary degraders that utilize extracellular xylanases to cleave xylan into smaller oligomers. The resulting xylooligosaccharides (XOS) can either be further metabolized directly by primary degraders or cross-feed secondary consumers, including Bifidobacterium. While several Bifidobacterium species have metabolic systems for XOS, most grow poorly on longer-chain XOS and xylan substrates. In this study, we isolated strains of Bifidobacterium pseudocatenulatum and observed that some, including B. pseudocatenulatum ED02, displayed growth on XOS with a high degree of polymerization (DP) and straight-chain xylan, suggesting a primary degrader phenotype that is rare in Bifidobacterium. In silico analyses revealed that only the genomes of these xylan-fermenting (xylan+) strains contained an extracellular GH10 endo-β-1.4 xylanase, a key enzyme for primary degradation of xylan. The presence of an extracellular xylanase was confirmed by the appearance of xylan hydrolysis products in cell-free supernatants. Extracellular xylanolytic activity was only detected in xylan+ strains, as indicated by the production of XOS fragments with a DP of 2 to 6, identified by thin-layer chromatography (TLC) and high-performance liquid chromatography (HPLC). Additionally, in vitro fecal fermentations revealed that strains with a xylan+ phenotype can persist with xylan supplementation. These results indicate that xylan+ B. pseudocatenulatum strains may have a competitive advantage in the complex environment of the gastrointestinal tract, due to their ability to act as primary degraders of xylan through extracellular enzymatic degradation. This study explores the production of prebiotic xylooligosaccharide (XOS) from cassava pulp waste and its effectiveness for the growth of Lactobacillus acidophilus (L. acidophilus). We successfully produced and characterized XOS from cassava pulp xylan using a Bacillus sp. endo-β-1,4-D-xylanase. The XOS was added to modify the MRS medium (MRSm) in various concentrations (0, 1, 3 and 5%) in which the L. acidophilus was inoculated. The growth of L. acidophilus was observed every 12 h for 2 days, and the fermentation products were analyzed for pH, sugar content, and short-chain fatty acids (SCFA) in terms of types and amount. The study showed that L. acidophilus grew well in MRSm. The optimum XOS concentration in MRSm was 5%, indicated by the highest growth of L. acidophilus (8.61 log CFU mL−1). The profile of SCFA products is 14.42 mM acetic acid, 0.25 mM propionic acid, 0.13 mM isobutyric acid, 0.41 mM n-butyric acid, 0.02 mM n-valeric acid, 0.25 mM isovaleric acid, and 25.08 mM lactic acid. Response surface methodology (RSM)-Box–Behnken design was used to optimize recovery of polyphenols, dietary fiber and xylooligosaccharide (XOS) from rice and corn bran in an integrated process. Same parameters were used to train artificial neural network (ANN) model with Levenberg–Marquardt algorithm. ANN showed better prediction accuracy as compared to RSM due to higher coefficient of determination (R2). Optimized conditions for extraction of polyphenols from rice bran were 70% ethanol, 1:10 solvent to sample ratio at 75 °C for 12 h. Moreover, extraction of corn bran polyphenols was optimized at 80% ethanol, 1:10 solvent to sample ratio at 73.6 °C for 10.5 h. After the extraction of polyphenol, left-over bran residue was subjected for dietary fiber extraction. Further, conditions were also optimized for the extraction of XOS from rice bran dietary fiber and found to be pH 6.92, sample to buffer ratio 1:11 with 243.86 units of xylanase enzyme at 46.77 °C for 5.23 h. In case of corn bran dietary fiber XOS, optimization conditions were pH 5.09, sample to buffer ratio 1:12 with 229.12 units of xylanase enzyme at 53.22 °C for 8.02 h. The optimized conditions gave a yield of 8.5% and 9% of polyphenols; followed by 45.1% and 49.2% of dietary fiber and later on 32% and 33.9% of XOS from rice and corn bran, respectively. All extracted bioactive compounds were found to be of good quality. In recent decades, one of the major challenges of biorefinery is the full utilization of lignocellulosic components for coproduction of value-added materials. In this context, we proposed an integrated process of acetic acid (AC) pretreatment, Papir Forsknings Institutet (PFI) refining and enzymatic hydrolysis to coproduce xylooligosaccharides, fermentable sugars and lignin-based porous carbon materials. Results showed AC pretreatment (170 ºC, 60 min, 5 w/v % acetic acid and 5% 2-naphthol addition based on dry mass of biomass) resulted in maximum xylooligosaccharids (XOS) yield of 51.08%. The integrated process of AC pretreatment assisted by 2-naphthol-7-sulfonate addition and PFI post-refining led to cellulose hydrolysis yield of 98.20% and lignin-based porous carbon materials with methylene blue adsorption capacity of up to 749.17 mg/g. Moreover, it was found lignin repolymerization inhibitors (i.e. 2-naphthol, 2-naphthol-7-sulfonate) in acid pretreatment helped increase the specific surface area of substrate and decrease surface lignin coverage, which enhanced cellulose accessibility to cellulase enzymes. Moreover, they enhanced lignin detaching during enzymatic hydrolysis, which reduced cellulase aggregation and promoted the ease of cellulose hydrolysis. The proficient synthesis of xylose and xylooligosaccharides (XOS) from lignocellulosic biomass proved to be a formidable undertaking, particularly in the absence of preliminary hemicellulose isolation. In this study, the potential of benzoic acid (BZA), a non-toxic and edible acidic catalyst, was assessed for the pretreatment of poplar in order to produce xylose and xylooligosaccharides (XOS). At low temperatures (< 160°C) or low acid concentrations (< 2%), the main product obtained from the depolymerization of hemicellulose was XOS, and no significant depolymerization occurred on a large scale. The results showed that the highest extraction yield of hemicellulose was 93.1% with 2% BZA at 160°C for 0.75 h, hemicellulose was mainly converted into xylose and XOS, with xylose accounting for 27.1% and XOS accounting for 46.7%. Meanwhile, the results of the process simulation of industrial energy consumption indicated that the amount of water added was the main factor affecting energy consumption in industrialization, followed by temperature. Background: Two experiments were conducted to establish an optimal NE challenge model and evaluate the efficacy of stimbiotic (STB) supplementation in necrotic enteritis (NE) challenged broilers. In Exp. 1, a total of 120 Arbor Acres (AA) broilers (45.0 ± 0.21 g) were randomly assigned to 6 treatments in a 3 × 2 factorial arrangement. Vaccine treatments included non-challenge (0), × 10 the recommended dose (× 10) or × 20 the recommended dose (× 20) by the manufacturer. Clostridium perfringens (CP) treatments were non-challenge (No) or 3 mL of 2.2 × 107 CFU CP challenge (Yes). In Exp. 2, a total of 72 AA broilers (40.17 ± 0.27 g) were randomly assigned to 6 treatments in a 3 × 2 factorial arrangement. Dietary treatments included non-additive (CON), 100 mg/kg STB (STB) and 100 mg/kg STB on top of a typical commercial blend including an essential oil, probiotics, and enzyme (CB). Challenge treatments included non-NE challenge (No) and NE challenge (Yes) as established in Exp. 1. Results: In Exp. 1, CP and vaccine challenge decreased (P < 0.05) body weight (BW), body weight gain (BWG) and feed intake (FI), and increased (P < 0.05) the number of broilers with diarrhea and intestinal lesions. The oral administration of × 20 recommended dose of vaccines coupled with 3 mL of 2.2 × 107 CFU CP resulted in (P < 0.01) a significantly increased incidence of wet litter and intestinal lesions. Thus, this treatment was chosen as the challenge model for the successful inducement of NE in Exp. 2. In Exp. 2, the NE challenge negatively affected (P < 0.01) growth performance, ileal morphology, immunoglobulin contents in blood, caecal microbiota in the caecum, footpad dermatitis, intestinal lesion scores, tumour necrosis factor (TNF-α) and endotoxin in the serum compared with the non-NE challenged birds. The supplementation of STB and CB in diets enhanced (P < 0.05) growth performance, intestinal microbiota, and blood profiles by stimulating ileal morphology (VH and VH:CD) and propionate production in the cecum, and there were no differences in measured variables between STB and CB supplemented birds. Conclusion: Overall, these results indicate that STB supplementation was able to reduce the inflammatory response and improve the performance of NE challenged birds, and the supplementation of STB alone was as effective as a typical commercial blend containing a number of other additives. The experiment was to study the technology of preparing xylooligosaccharide feed by using Pleurotus eryngii mushroom bran. Using Pleurotus eryngii mushroom bran as raw material and xylooligosaccharide yield as index, single factor experiment was used to study the effect of pretreatment conditions and enzymatic hydrolysis conditions on xylooligosaccharide yield, and the factors with great influence on xylooligosaccharide yield were optimized by orthogonal experiment. The results showed that the appropriate pretreatment process was pretreatment temperature 50 ℃, H2O2 3%, Na2CO3 4%, liquid-solid ratio 2.5 mL/g, pretreatment time 3 h. The suitable enzymatic hydrolysis process was xylanase 400 U/g, cellulase 400 U/g, mannanase 400 U/g, liquid-solid ratio 4 mL/g, enzymatic hydrolysis temperature 50 ℃, initial pH value 6.0, enzymatic hydrolysis time 4 h. Under these conditions, the yield of xylooligosaccharide in the enzymatic hydrolysis material reached 97.67 mg/g. Xylooligosaccharide (XOS) has been considered to be an effective prebiotic, but its exact mechanisms remain unknown. This research was conducted to evaluate the effects of XOS on pig intestinal bacterial community and mucosal barrier using a lipopolysaccharide (LPS)-caused gut damage model. Twenty-four weaned pigs were assigned to 4 treatments in a 2 × 2 factorial design involving diet (with or without XOS) and immunological challenge (saline or LPS). After 21 d of feeding 0% or 0.02% commercial XOS product, piglets were treated with saline or LPS. After that, blood, small intestinal mucosa and cecal digesta were obtained. Dietary XOS enhanced intestinal mucosal integrity demonstrated by higher villus height, villus height-to-crypt depth ratio, disaccharidase activities and claudin-1 protein expression and lower crypt depth. XOS also caused down-regulation of the gene expression of toll-like receptor 4 and nucleotide-binding oligomerization domain protein signaling, accompanied with decreased pro-inflammatory cytokines and cyclooxygenase 2 contents or mRNA expression and increased heat shock protein 70 mRNA and protein expression. Additionally, increased Bacteroidetes and decreased Firmicutes relative abundance were observed in the piglets fed with XOS. At the genus level, XOS enriched the relative abundance of beneficial bacteria, e.g., Faecalibacterium, Lactobacillus, and Prevotella. Moreover, XOS enhanced short chain fatty acids contents and inhibited histone deacetylases. The correlation analysis of the combined datasets implied some potential connections between the intestinal microbiota and pro-inflammatory cytokines or cecal metabolites. These results suggest that XOS inhibits inflammatory response and beneficially modifies microbes and metabolites of the hindgut to protect the intestine from inflammation-related injury.

Xylooligosaccharide (XOS) is an attractive prebiotic mainly due to its bifidogenic effect. However, commercial XOS with different compositions is often applied in the food industry at different doses without specifications. In this study, we evaluated the bifidogenic activity of XOS at different doses with either mixtures or pure fractions with different degrees of polymerization (DP), using three strains of Bifidobacterium spp., including B. breve ATCC 15700, B. bifidum ATCC 29521, and B. animalis subsp. lactis HN019. Three growth indicators showed strain-specific bifidogenic activity of XOS, and the activity was both dose- and fraction-dependent as only certain fractions stimulated significant growth. Adding 0.25% XOS (w/v) also promoted increase in total bifidobacterial population of rat fecal samples fermented in vitro. Albeit the antibacterial activity of XOS fractions can be demonstrated, significant growth inhibition can only be achieved when 4.0% XOS mixture was added in Staphylococcus aureus ATCC 6538 pure culture. In contrast, in the presence of B. lactis HN019, 1.0% XOS showed significant antibacterial activity against S. aureus ATCC 6538 in milk. In addition, RNA sequencing suggested downregulation of genes involved in S. aureus ATCC 6538 infection, pathogenesis, and quorum sensing, by XOS. In conclusion, the report urges scientific specifications on XOS chemistry for its effective application as a novel food ingredient or functional food and provides novel insights into its bifidogenic and antibacterial activities.

Over the last decade, xylooligosaccharides (XOS) have attracted great attentions because of their unique chemical properties and excellent prebiotic effects. Among the current strategies for XOS production, enzymatic hydrolysis is preferred due to its green and safe process, simplicity in equipment, and high control of the degrees of polymerization. This paper comprehensively summarizes various lignocellulosic biomass and marine biomass employed in enzymatic production of XOS. The importance and advantages of enzyme immobilization in XOS production are also discussed. Many novel immobilization techniques for xylanase are presented. In addition, bioinformatics techniques for the mining and designing of new xylanase are also described. Moreover, XOS has exhibited great potential applications in the food industry as diverse roles, such as a sugar replacer, a fat replacer, and cryoprotectant. This review systematically summarizes the current research progress on the applications of XOS in food sectors, including beverages, bakery products, dairy products, meat products, aquatic products, food packaging film, wall materials, and others. Prebiotics are non-digestible compounds that promote intestinal microbiota growth and/or activity. Xylooligosaccharides (XOS) are new prebiotics derived from the hemicellulose fraction of lignocellulosic materials. Challenges in using those materials as sources for prebiotic compounds lie in the hemicellulose extraction efficiency and the safety of those ingredients. In this sense, this work aims to optimize hemicellulose extraction and XOS production through direct enzymatic hydrolysis of alkali pre-treated wheat straw without undesired byproducts. By increasing the temperature of the enzymatic step from 40 °C to 65 °C we achieved an improvement in the extraction yield from 55 % to 80 %. Products with different degrees of polymerization were also noticed: while XOS ≤ X 6 where the main products at 40 °C, a mixture of long arabinoxylan derived polymers (ADPo) and XOS ≤ X 6 was obtained at 65 °C, irrespective of the extraction yield. Thus, a modulatory effect of temperature on the product profile is suggested here. Among the XOS ≤ X 6 produced, X 2 -X 3 were the main products, and X 4 was the minor one. Xylooligosaccharides (XOS) have been employed as prebiotics containing oligomers of varying sizes or molecular ratios. XOS with a low degree of polymerization (DP) has been demonstrated to have high prebiotic potential. However, there is limited information regarding the specific chain length of XOS required to elicit distinct responses in the gut microbiota. In this study, we aimed to explore whether variations in XOS DP could alter the fate of colonic fermentation. Five XOS fractions (BWXFs) with DP ranges of >40, 20–40, 10–20, 5–10, and 2–4 were prepared by beechwood xylan autohydrolysis and tested on human gut microbiota. Extracellular XOS degradation was observed for molecules with a DP exceeding 5. BWXF treatments altered the microbial community structures, and substrate size-dependent effects on the microbial composition and metabolic outputs were observed. Bacteroidaceae were specifically enriched by xylan. Lachnospiraceae were particularly stimulated by XOS with a DP of 20–40 and 2–4. Bifidobacteriaceae were notably enriched by XOS with a DP of 5–20. High butyrate yields were obtained from cultures containing long-chain BWXFs. Xylan is one of the major structural components of the plant cell wall. Xylan present in the human diet reaches the large intestine undigested and becomes a substrate to species of the gut microbiota. Here, we characterised the capacity of Limosilactobacillus reuteri and Blautia producta strains to utilise xylan derivatives. We showed that L. reuteri ATCC 53608 and B. producta ATCC 27340 produced β-D-xylosidases, enabling growth on xylooligosaccharide (XOS). The recombinant enzymes were highly active on artificial (p-nitrophenyl β-D-xylopyranoside) and natural (xylobiose, xylotriose, and xylotetraose) substrates, and showed transxylosylation activity and tolerance to xylose inhibition. The enzymes belong to glycoside hydrolase family 120 with Asp as nucleophile and Glu as proton donor, as shown by homology modelling and confirmed by site-directed mutagenesis. In silico analysis revealed that these enzymes were part of a gene cluster in L. reuteri but not in Blautia strains, and quantitative proteomics identified other enzymes and transporters involved in B. producta XOS utilisation. Based on these findings, we proposed a model for an XOS metabolism pathway in L. reuteri and B. producta strains. Production of defatted rice bran arabinoxylan (DRB‐AX) from defatted rice bran (DRB) is maximized using alkali extraction. Conversion of DRB‐AX into xylooligosaccharide (XOS) by endoxylanase from Neocallimastix partriciarum, and refining enzymatic XOS liquor using activated carbon are also investigated. Chemical composition analysis of DRB indicates the richness of nutrients at 60.76% carbohydrate, 10.42% protein, and 32.69% total dietary fiber. Alkali extraction performed at 2.5 M NaOH solution for 12 h is the optimal condition to produce DRB‐AX, with a crude yield of 52.82 g 100 g−1 containing 26.96% arabinoxylan, 26.07% arabinose, and 21.42% xylose with arabinose/xylose ratio 1.22. Endoxylanase from N. partriciarum partially hydrolyzes DRB‐AX at 50 °C, pH 6.0 at enzyme concentration of 80 U g−1 substrate for 24 h. This enzymatic XOS production design yields 26.67 ± 0.69 mg g−1 DRB. Xylobiose and xylotriose including an unknown saccharide are short chain oligomer products. Refining XOS from DRB with 15% (w/v) activated carbon recover 55.7% (w/w) of initial XOS. Background: Autohydrolysis is an extensively investigated pretreatment method due to its environmental friendliness. During autohydrolysis, most xylan from hemicellulose can be converted into xylooligosaccharides (XOS), and cellulose in the autohydrolyzed residues can be transformed into glucose after enzymatic hydrolysis. Both of these are value-added biochemicals in the biorefining process. In this work, paper mulberry (PM), which contains abundant protein, was utilized as a raw material to coproduce XOS and single-cell protein (SCP) through autohydrolysis and fermentation technologies. Results: The results showed that 8.3 g of XOS and 1.8 g of amino acids could be recovered in the autohydrolysate (based on 100 g raw material) after autohydrolysis (170 °C, 1 h). Moreover, 5.7 g of low-DP XOS along with 1.8 g of amino acids could be further obtained from the autohydrolysate after hydrolysis with endo-β-1-4-xylanase. In addition, 20.1 g of fermentable monosaccharides was recovered after hydrolyzing the autohydrolyzed PM with cellulase, which can be used to produce 4.8 g of SCP after fermentation with Candida utilis. Conclusion: As a valuable application of PM, a novel process is proposed to coproduce amino acid-rich XOS and SCP through autohydrolysis. Background: This study evaluated the effects of supplemental xylanase and xylooligosaccharides (XOS) in a corn-soybean meal (SBM)-based diet on growth performance and intestinal health of broilers. A total of 288 day-old chicks (Cobb 500) were allocated to 36 floor pens (8 birds/pen) equally in 9 dietary treatments in a 3 × 3 factorial arrangement. The treatments were combinations of 3 levels of xylanase (0, 0.005% and 0.01% Econase XT) and 3 levels of prebiotics (0, 0.005% and 0.01% XOS) added to basal mash diets formulated in three phases (starter, d 0–14; grower, d 15–28; finisher, d 29–42). The feed intake and body weights were recorded weekly. On d 42, ileal sections were collected for histomorphometric and gene expression analysis, and cecal content was collected for determining short-chain fatty acids (SCFA) and microbiota. Results: Xylanase linearly (P < 0.01) increased the average daily gain (ADG) in both the finisher and total period and the final body weight gain (FBWG, 2940 & 2932 vs. 2760 g) of broilers. XOS did not significantly increase either ADG or FBWG (P > 0.05). Supplemental xylanase and XOS did not affect average daily feed intake and feed conversion ratio (P > 0.05). Xylanase and XOS did not change villus height (VH) or crypt depth (CD) ratio (P > 0.05). However, xylanase exhibited a trend (P = 0.097) on VH:CD ratio. The inclusion of 0.01% XOS without xylanase increased the level of IL-10 (a marker of anti-inflammatory cytokine) and IL-4 (a T-cell differentiation cytokine) genes compared with control (P < 0.05). The acetate production was increased by xylanase (P < 0.01) and XOS (P < 0.05) without an additive effect. Xylanase increased total SCFA (P < 0.01) while XOS had a tendency to increase (P = 0.052). Alpha and beta diversity of microbiota among treatments were not different (P > 0.05). However, the mean proportion of family Ruminococcaceae was increased by the supplemental 0.01% xylanase (P < 0.01). Conclusion: It can be concluded that XOS can enhance cecal fermentation, while xylanase can increase the body weight gain along with the fermentation metabolites in the ceca of broilers fed the corn-SBM-based diet but the effects may not always translate into an improved mucosal absorptive capacity and a better feed efficiency. In Brazil, the production and consumption of palm heart, especially from the Bactris gasipaes Kunth, generates a large number of lignocellulosic by-products. This study reports the obtainment of xylooligosaccharides (XOS) from xylans extracted from these residues. Xylans from peach palm waste (inner sheath and peel) were extracted using a mild alkali treatment with recovery yields of 82% and 80%, respectively. XOS were obtained through enzymatic hydrolysis employing a commercial xylanase with yields from xylan inner sheath and xylan peel of 50.1% and 48.8%, respectively. The antioxidant potential of XOS was measured employing five of the most commonly used procedures. In overall terms, the XOS from the xylans of peach palm wastes showed higher antioxidant capacity than the XOS obtained from the commercial xylans. The chemical structures of the XOS were determined by mass spectrometry (ESI–MS). The ESI–MS spectra suggest that XOS with grouped xylose or arabinose units ranging from 2 to 5 (differing by 132 Da) and as sodium adduct ions [M + Na]+ in the range of 100–1000 m/z. These results indicate that peach palm wastes can be explored to XOS production, which could be applied as natural antioxidants in functional food and pharmaceutical preparations. We used agricultural residue, corn cob, with biorefinery and bioeconomy concepts. At short-time cultivation in corn cob (12 h), Bacillus firmus K-1 produced cellulase-free xylanolytic enzyme, with xylooligosaccharides (XOSs), X5 and X6, as the main products, which can be used in a variety of applications. The xylanolytic enzyme produced from B. firmus K-1 effectively degraded xylan in corn cob, which was examined by chemical composition, scanning electron microscope (SEM), and Fourier transform infrared spectroscopy (FTIR). After cultivation, the xylan contained in the corn cob residue was decreased (as biological pretreatment), causing morphological and structural changes, including creating porosity and increasing the surface area and the exposure of cellulose of pretreated corn cob. These results lead to an improvement of cellulose access by cellulases. Commercially available cellulases, Accellerase® 1500 and Cellic® CTec2, yielded significantly higher glucose concentrations from pretreated corn cob compared to untreated corn cob. After saccharification, the lignin-rich corn cob residue can be used as a raw material for other purposes. Moreover, the B. firmus cells, with a low risk to human health, can be used in some applications. This study presents an efficient method for producing high-value-added products from agricultural residue (corn cob) through biological processes which are environmentally friendly and economically viable. An alkali-mediated hydrothermal approach was optimized for partial extraction of high-purity hemicellulose (xylan) from a timber waste product (unbleached eucalyptus sawdust). The extraction process was: a) extractives removal using sequential solvent extraction; b) alkali-mediated hydrothermal extraction of hemicellulose; and c) purification of the isolated hemicellulose fraction. Biomass was extracted in an autoclave at a fixed temperature of 121 °C while varying the extraction time (1 h to 2.5 h) and alkali concentration (10%, 15%, and 20% w/v). Using this simple approach, high purity polymeric hemicellulose with molecular mass (21 to 30 kDa) was recovered in yields of 35 to 37%. The isolated hemicellulose can be chemically transformed into high-value commercial products such as prebiotics (xylooligosaccharides), surfactants, hydrogels, and food packaging materials. The purified residue can be utilized in existing kraft pulping processes. In this study, crude and partially purified xylanase from Aureobasidium pullulans CCT 1261 were characterized and applied in beechwood xylan hydrolysis to obtain xylooligosaccharides (XOS). Fractional precipitation (0–30 %/30–60 %) with ammonium sulfate allowed partial purification of xylanases in 6.8-fold and recovery of 69.4 %. Both extracts exhibited optimum activity at 50 °C and pH 4.5, and greater stability at 40 °C within the studied range. The thermostability parameters (k d , t 1/2 , ΔH*, ΔG*, ΔS*) of the extracts indicated the enzyme denaturation by the increment in temperature from 40 °C to 60 °C; its process was endothermic and led to protein unfolding in a more random structure. Crude xylanase showed a lower value of the Michaelis-Menten constant (25 mg/mL) than the partially purified enzyme (50 mg/mL). In hydrolysis reactions, total XOS contents (7.7 mg/mL and 7.9 mg/mL) and XOS yields (25.7 % and 26.5 %) were equal statistically using the crude or partially purified extracts, differing only in the amount of xylose released. The hydrolysates were mainly composed of potential prebiotics, i.e., xylobiose (50.9 % and 53.8 %) and xylotriose (30.9 % and 32.8 %). Cooking with active oxygen and solid alkali (CAOSA) is a mild and environmentally friendly method for separation of lignin components. The degradation mechanism of hemicellulose associated with this method has been investigated. It was found that the hemicellulose-derived polysaccharides were directly oxidized rather than hydrolyzed to monosaccharides for degradation. To avoid the undesirable degradation of hemicellulose components by CAOSA, the idea of separating hemicellulose first in the form of xylan to produce xylooligosaccharide was presented by this study. Although hydrothermal autocatalysis has been commonly used in hemicellulose separation, the effects of acid-enhanced hydrothermal method and alkali-suppressed hydrothermal method on hydrothermal autocatalysis were systematically compared for the first time, and it was found that the acid-enhanced method was beneficial for highly selective extraction of xylan. Then, the hemicellulose-removed bamboo was treated with CAOSA process to separate lignin from cellulose. It was found that solid alkali dosage of CAOSA could be evidently decreased for hemicellulose-removed bamboo, which was helpful to reduce the cost of CAOSA. Coupling hemicellulose pre-extraction and CAOSA lignin fractionation strategy, a comprehensive biorefinery case for bamboo biomass could be anticipated. [ABSTRACT FROM AUTHOR]

Xylooligosaccharide (XOS) is known as a prebiotic, however, it is unknown whether XOS can directly protect against bacterial infection. This study aimed to investigate the direct inhibitory effects of XOS on Salmonella Typhimurium colonization and the inductive impairments in gut health and growth performance in broilers. We first probed the inhibitory effects of XOS on S. Typhimurium adhesion and its induction of intestinal epithelial cell (IPEC-J2) injuries. Afterward, 168 one-day-old yellow-feathered broilers were randomly divided into 3 groups (7 replicates/group): negative control (NC , received a basal diet), positive control (PC , received a basal diet with S. Typhimurium challenge) and XOS group (PC birds + 1,500 mg/kg XOS). All birds except those in NC were orally challenged with S. Typhimurium from 8 to 10 d of age. Parameters were analyzed on d 11. The results showed that XOS inhibited S. Typhimurium adhesion and the inductive injuries of IPEC-J2 cells by lowering (P < 0.05) certain adhesion-related genes expression of this bacterium. It also alleviated S. Typhimurium-induced increase (P < 0.05) in the expression of certain inflammatory cytokines and tight junction (TJ) proteins of IPEC-J2 cells. Supplementing XOS to S. Typhimurium-challenged broilers attenuated the elevations (P < 0.05) in S. Typhimurium colonization of ileal mucosa and its translocation to the liver and spleen, as well as increased (P < 0.05) certain TJ proteins expression of ileum. Besides, XOS addition normalized S. Typhimurium-induced impairments (P < 0.05) in ileal morphology, final body weight and average daily gain in broilers. Collectively, supplemental XOS directly suppressed intestinal colonization of S. Typhimurium by diminishing its adhesiveness and subsequently mitigated destructions in intestinal barriers, thus contributing to weaken growth retardation in challenged broilers. Xylooligosaccharide (XOS), an oligosaccharide with great prebiotic potential, has been shown to modulate the gut microbiota. Our aim was to determine the effects of dietary XOS supplementation on body composition, lipid metabolism and intestinal microbiota using a mouse model. Our data showed that XOS dietary supplementation at 2.1 and 7.0% significantly changed the gut microbial composition. A decrease in visceral fat depots, concentration of inflammatory cytokine MCP-1, and abundance of the two most important obesity associated gut microbial phyla Firmicutes and Bacteroidetes were observed with 7% XOS supplementation. No change in blood or liver lipids was observed with both XOS doses. The production of SCFAs in the cecum was increased and relative abundance of Actinobacteria was decreased with supplementation of both doses of XOS. To conclude, XOS supplementation reduced adiposity through decreasing gene expression of markers of adipogenesis and fat synthesis and induced changes in intestinal microbial composition.

As is well-known, endo-1,4-β-xylanase and β-xylosidase are the rate-limiting enzymes in the degradation of xylan (the major hemicellulosic component), main functions of which are cleavaging xylan to release xylooligosaccharides (XOS) and xylose that these two compounds have important application value in fuel, food, and other industries. This study focuses on enzymatic hydrolysis of poplar sawdust xylan for production of XOS and xylose by a GH11 endo-1,4-β-xylanase MxynB-8 and a GH39 β-xylosidase Xln-DT. MxynB-8 showed excellent ability to hydrolyze hemicellulose of broadleaf plants, such as poplar. Under optimized conditions (50°C, pH 6.0, dosage of 500 U/g, substrate concentration of 2 mg/mL), the final XOS yield was 85.5%, and the content of XOS2−3 reached 93.9% after 18 h. The enzymatic efficiency by MxynB-8 based on the poplar sawdust xylan in the raw material was 30.5%. Xln-DT showed excellent xylose/glucose/arabinose tolerance, which is applied as a candidate to apply in degradation of hemicellulose. In addition, the process and enzymatic mode of poplar sawdust xylan with MxynB-8 and Xln-DT were investigated. The results showed that the enzymatic hydrolysis yield of poplar sawdust xylan was improved by adding Xln-DT, and a xylose-rich hydrolysate could be obtained at high purity, with the xylose yield of 89.9%. The enzymatic hydrolysis yield was higher (32.2%) by using MxynB-8 and Xln-DT together. This study provides a deep understanding of double-enzyme synergetic enzymolysis of wood polysaccharides to valuable products.

Xylan is one of the major structural components of the plant cell wall. Xylan present in the human diet reaches the large intestine undigested and becomes a substrate to species of the gut microbiota. Here, we characterised the capacity of Limosilactobacillus reuteri and Blautia producta strains to utilise xylan derivatives. We showed that L. reuteri ATCC 53608 and B. producta ATCC 27340 produced β-D-xylosidases, enabling growth on xylooligosaccharide (XOS). The recombinant enzymes were highly active on artificial (p-nitrophenyl β-D-xylopyranoside) and natural (xylobiose, xylotriose, and xylotetraose) substrates, and showed transxylosylation activity and tolerance to xylose inhibition. The enzymes belong to glycoside hydrolase family 120 with Asp as nucleophile and Glu as proton donor, as shown by homology modelling and confirmed by site-directed mutagenesis. In silico analysis revealed that these enzymes were part of a gene cluster in L. reuteri but not in Blautia strains, and quantitative proteomics identified other enzymes and transporters involved in B. producta XOS utilisation. Based on these findings, we proposed a model for an XOS metabolism pathway in L. reuteri and B. producta strains. Together with phylogenetic analyses, the data also revealed the extended xylanolytic potential of the gut microbiota.

There is an increasing emphasis on the transformation of lignocellulosic biomass into versatile products. The feasibility of preparing xylooligosaccharides (XOS) by hydrolysis of sorghum stalk (SS) using organic and inorganic acids was studied. The influences of different acids (gluconic acid, acetic acid, sulfuric acid, and oxalic acid), process time and temperature on the hydrolysis of SS were explored. The findings indicated XOS yield can be maintained at a high level under different conditions with organic acid pretreatments. Optimum yield of XOS (39.4%) was obtained using sulfuric acid (pH 2.2) at 170 °C and 75 min of process time. It is suggested when reaction temperature and time were increased, both X5 and X6 are cracked into XOS with lower molecular mass such as X2, X3, and X4. Moreover, the results based on mass balance showed that up to 110 g (XOS) plus 117 g (glucose) can be recovered from 1000 g of SS. Results will give insights into establishing an efficient acid pretreatment of sorghum stalk to coproduction of XOS and glucose.

Date palm (phonix dactylifera L.) is an important tropical fruit growing in central and southern regions of Iran. Date seed is composed of cellulose, hemicellulose, and lignin, that make it an excellent candidate for xylooligosaccharide (XOS) production. In this study, two different protocols are used for the extraction of hemicellulose from date seeds. In the first protocol, hemicellulose (xylan1) was extracted by 2.25 M alkaline solution at room temperature for 24 hr. In the second protocol, date seed was treated with LCHTA (low concentration, 0.1 M, high temperature, 80°C, alkaline solution) for 3 hr, and thereafter, hemicellulose (xylan2) was extracted by 2.25 M alkaline solution at room temperature for 24 hr. The carbohydrate units of xylan1 and xylan2 were qualified and quantified by HPAEC‐ PAD. Side groups of xylan1 and xylan2 were detected by FTIR. In the next step, xylan1 and xylan2 were exposed to two commercial endoxylanases namely veron 191 and pentopan mono BG. Temperature, pH, time, and enzyme dosage of hydrolyzation were optimized to maximize XOS and minimize xylose. The results showed that the enzymes successfully hydrolyzed xylan2 and produced XOS, but cannot hydrolyze xylan1. Pentopan mono BG and veron 191 produced the highest amount of XOS after 4 (1.17 mmol/g) and 6 hr (1.13 mmol/g) of incubation, respectively. Conversion factors of xylan2 to XOS for pentopan mono BG and veron were 0.41 and 0.36, respectively.
