In order to realize effective monitoring of Zhixincun landslide, this paper selected 27 sentinel-1A data in 2017, and conducted deformation monitoring of Zhixincun landslide based on small baseline radar interferometry technology (SBAS-InSAR), and analyzed its temporal evolution situation. Using ALOS-2 data from 2016 and 2017, differential radar interferometry (D-InSAR) was used to monitor the characteristics of the landslide variant. SBAS-InSAR monitors the temporal evolution situation of landslide deformation, while D-InSAR mainly monitors the deformation of specific landslide shape and variation. Moreover, the penetration of L-band ALOS-2 data is stronger than that of C-band sentinel-1A data, which can obtain more complete interference information. The monitoring results of both can be cross-verified. Improve the reliability of the results. The SBAS-InSAR monitoring results showed that the slope end of the landslide catchment area in Zhixincun had subsidence during the monitoring period, and the surface subsidence at the landslide end reached 12.47mm from July 5 to July 29, with an average subsidence rate of 2.88mm/a during the monitoring period. Uplift occurred in the threatened residential areas in the valley, with an average cumulative uplift of 19.59mm on December 8 and an average uplift rate of 19.99mm/a during the monitoring period. The D-InSAR results showed that there were five major deformations on the slope of Zhixincun landslide catchment area. The largest deformations with an area of 17 973m2 were located on the west side of the slope, and the most unstable deformations were located on the east side of the slope. The average cumulative shape variable reached 49.9mm during the monitoring period. Both monitoring methods showed that the threat of landslide disaster mainly came from the west slope with poor vegetation cover, and the rainy season was the key period of landslide disaster prevention and control in Zhixincun.

To explore the antimicrobial activity of epidermis mucus, the basic components and amino acid composition were analyzed and the antimicrobial substances in the mucus were extracted in this study. Their antimicrobial activity, preservation effects on salmon were explored. The components with antimicrobial activity were isolated and purified by solid phase extraction, gel filtration chromatography and reverse phase high performance liquid chromatography. And the peptide sequences were identified. Then, the antimicrobial peptides were synthesized accordingly and their antimicrobial activities were verified. The results exhibited that the main composition of epidermis mucus of sturgeon was protein with 47.44%±0.59%. The total amino acid content was 459.68 mg/g, with a high content of basic amino acids and hydrophobic amino acids. The crude antimicrobial peptide exhibited significant antimicrobial activity against Bacillus subtilis. Crude antimicrobial peptide (50 mg/mL) significantly inhibited the growth of microorganisms and protein degradation during salmon preservation. The highest active component after final liquid chromatography purification exhibited an inhibition rate of 92.40%±4.50% against Bacillus subtilis and 75.43%±5.02% against Escherichia coli. The sequences of novel peptides were identified as HSETLHDV and PLTDWQL. The synthetic antimicrobial peptides of sturgeon epidermis mucus had obvious inhibitory effects on Bacillus subtilis, Escherichia coli and Shewanella baltica. Therefore, this study would provide a theoretical basis for exploring the antimicrobial value of sturgeon mucus and lay a foundation for the development and application of natural preservatives.

Forest canopy height data are essential to assessing forest biomass and carbon storage, and they can be efficiently retrieved using the Advanced Terrain Laser Altimetry System (ATLAS) onboard ICESat-2. However, due to the sparse and discrete distribution of ATLAS footprints, the wall-to-wall mapping of forest canopy heights requires a combination of other ancillary data. In order to match the ATLAS data with ancillary data, or estimate higher-resolution canopy heights, the standard ATLAS canopy height products (ATL08), reported at a fixed step size of 100 m (12 m × 100 m), are typically divided into 30 m step sizes. There is little concern about the accuracy of the generated 30 m segment (12 m × 30 m) dataset. Furthermore, previous studies have primarily evaluated the along-track performance of the canopy height product, rather than the ability of ATLAS to provide an accurate representation of canopy heights at the pixel-wise level. In this study, we use airborne LiDAR data as references with which to evaluate the along-track accuracy of 30 m segment canopy height products and their accuracy in representing canopy height at the pixel-wise level. A framework method for spatially matching ATLAS canopy height estimate data with ancillary data accurately is also established in this study. This enables the use of ATLAS data to characterize pixel-wise canopy heights more precisely. The results show that the accuracy of the 30 m segment version dataset in representing the pixel-wise canopy heights (R2 = 0.38, RMSE = 8.37 m) is lower than its along-track accuracy (R2 = 0.44, RMSE = 7.63 m). Using only the nighttime data for the Taranaki region, the proposed method improves the estimation of pixel-wise forest canopy heights, with the R2 increasing from 0.49 to 0.59, the RMSE decreasing from 7.48 m to 5.51 m, and the %RMSE decreasing from 36.7% to 27.6%. This study contributes to understanding the accuracy of the ATLAS in reflecting pixel-wise canopy height and provides a new way of spatially matching ATLAS canopy height data with other remote sensing data.

The photon point clouds collected by the high-sensitivity single-photon detector on the Ice, Cloud, and Land Elevation Satellite-2 (ICESat-2) are utilized in various applications. However, the discretely distributed noise among the signal photons greatly increases the difficulty of signal extraction, especially the edge noise adjacent to signals. To detect signal photons from vegetation coverage areas at different slopes, this paper proposes a density-based multilevel terrain-adaptive noise removal method (MTANR) that identifies noise in a coarse-to-fine strategy based on the distribution of noise photons and is evaluated with high-precision airborne LiDAR data. First, the histogram-based successive denoising method was used as a coarse denoising process to remove distant noise and part of the sparse noise, thereby increasing the fault tolerance of the subsequent steps. Second, a rotatable ellipse that adaptively corrects the direction and shape based on the slope was utilized to search for the optimal filtering direction (OFD). Based on the direction, sparse noise removal was accomplished robustly using the Otsu’s method in conjunction with the ordering points to identify the clustering structure (OPTICS) and provide a nearly noise-free environment for edge searching. Finally, the edge noise was removed by near-ground edge searching, and the signal photons were better preserved by the surface lines. The proposed MTANR was validated in four typical experimental areas: two in Baishan, China, and two in Taranaki, New Zealand. A comparison was made with three other representative methods, namely differential, regressive, and Gaussian adaptive nearest neighbor (DRAGANN), used in ATL08 products, local distance statistics (LDS), and horizontal ellipse-based OPTICS. The results demonstrated that the values of the F1 score for the signal photon identification achieved by the proposed MTANR were 0.9762, 0.9857, 0.9839, and 0.9534, respectively, which were higher than those of the other methods mentioned above. In addition, the qualitative and quantitative results demonstrated that MTANR outperformed in scenes with steep slopes, abrupt terrain changes, and uneven vegetation coverage.

Red blood cells (RBCs) have unique properties that have been exploited for therapeutic uses. Here the authors engineer RBCs to co-express tumor associated antigens on MHC I, 4-1BBL and IL-12, generating artificial antigen presenting cells that can induce antigen-specific T cell responses and antitumor immune responses in preclinical models.

BackgroundHepatocellular carcinoma (HCC) is a common abdominal cancer with poor survival outcomes. Although there is growing evidence that N7-methylguanosine (m7G) is closely associated with tumor prognosis, development, and immune response, few studies focus on this topic.MethodsThe novel m7G risk signature was constructed through the Lasso regression analysis. Its prognostic value was evaluated through a series of survival analyses and was tested in ICGC-LIRI, GSE14520, and GSE116174 cohorts. CIBERSORT, ssGSEA, and ESTIMATE methods were applied to explore the effects of the m7G risk score on tumor immune microenvironment (TIM). The GSEA method was used to evaluate the impacts of the m7G risk score on glycolysis, ferroptosis, and pyroptosis. The human protein atlas (HPA) database was used to clarify the histological expression levels of five m7G signature genes. The biofunctions of NCBP2 in hepatocellular cancer (HC) cells were confirmed through qPCR, CCK8, and transwell assays.ResultsFive m7G regulatory genes comprised the novel risk signature. The m7G risk score was identified as an independent prognostic factor of HCC and could increase the decision-making benefit of traditional prognostic models. Besides, we established a nomogram containing the clinical stage and m7G risk score to predict the survival rates of HCC patients. The prognostic value of the m7G model was successfully validated in ICGC and GSE116174 cohorts. Moreover, high m7G risk led to a decreased infiltration level of CD8+ T cells, whereas it increased the infiltration levels of Tregs and macrophages. The glycolysis and pyroptosis processes were found to be enriched in the HCC patients with high m7G risk. Finally, overexpression of NCBP2 could promote the proliferation, migration, and invasion of HC cells.ConclusionsThe m7G risk score was closely related to the prognosis, antitumor immune process, glycolysis, and malignant progression of HCC. NCBP2 has pro-oncogenic abilities, showing promise as a novel treatment target.

The traditional method of obtaining rock quality designation (RQD) cannot fully reflect the anisotropy of the rock mass and thus cannot accurately reflect its quality. In the method of calculating RQD based on three-dimensional network simulation of discontinuities, due to the limited number of samples and low accuracy of discontinuity data obtained by manual contact measurement, a certain deviation in the network is generated based on the data, which has an impact on the calculation result. Taking a typical slope in Dongsheng quarry in Changchun City as an example, in this study, we obtained the discontinuity data of the slope based on digital close-range photogrammetry, which greatly enlarged the sample size of discontinuity data and improved the data quality. Based on the heterogeneity of the rock mass, the optimum threshold of discontinuity spacing was determined when surveying lines were laid parallel to different coordinate axes to calculate the generalized RQD, and the influence of measuring blank areas on the slope caused by vegetation coverage or gravel accumulation was eliminated. The real generalized RQD of the rock mass after eliminating the influence of blank areas was obtained. Experiments showed that, after eliminating the influence of blank areas, the generalized RQD of the slope rock mass more truly represented the complete quality of rock mass and offers a new idea for the quality evaluation of engineering rock mass.

Thrust of shield cutters is the major parameter of tunnel construction and an important index for shield machine design. The thrust bearing resistance of a shield machine has a significant impact on its construction efficiency and safe operation. The use of quasirectangular shield not only can increase the space utilization rate but also avoid the deformation of the back soil when compared with a conventional circular or rectangular shield. In this paper, structural analysis of quasirectangular shield cutterhead is carried out and a corresponding mathematical thrust model is developed. Both the stress and displacement distributions of cutterhead are calculated. It is found that the stress value in most regions of the cutterhead is between 5 MPa and 45 MPa. The maximum stress is 208.44 MPa, which is at the middle part of the rib and is below the yield limit. The maximum deformation is found in the center area of the chest plate, the value of which is also within the design requirement. In addition, a monitoring method suitable for quasirectangular shield is proposed. The appropriateness and reliability of the proposed monitoring method are demonstrated by the comparison between the numerical simulation and monitoring method.

Current models cannot distinguish detailed topological relations between regions with holes. In order to solve this problem, a new detailed representation model for topological relations is proposed in this study. In this model, the key is to describe topological relations caused by multi-holes. Definitions of regions with holes and real objects expressed by elements of regions with holes are comprehensively analyzed, and then a practical definition of regions with holes is presented. Based on the 9-intersection model, a generalized 9-intersection model is proposed, which can completely describe detailed topological relations between regions with holes. For showing the description ability, the model is applied to describe detailed topological relations between regions with holes and detailed topological relations between objects of different complexities with the same major categories of topological relations, and a case study is designed to show the practicality of the model. The results show that the model is valid.

The discrepancies in existing land cover data are relatively high, indicating low local precision and application limitations. Multisource data fusion is an effective way to solve this problem; however, the fusion procedure often requires resampling to unify the spatial resolution, causing a lower spatial resolution. To solve this problem, this study proposes a multisource product fusion mapping method of filtering training samples and product correction at a fine resolution. Based on the Superpixel algorithm, principal component analysis (PCA), and statistical extraction techniques, combined with the Google Earth Engine (GEE) platform, reliable land cover data were acquired. GEE and machine-learning algorithms correct the unreliable information of multiple products into a new land cover fusion result. Compared to the common method of extracting consistent pixels from existing products, our proposed method effectively removes nearly 38.75% of them, with a high probability of classification error. The overall accuracy of fusion in this study reached 85.80%, and the kappa coefficient reached 0.82, with an overall accuracy improvement of 11.75–24.17% and a kappa coefficient improvement of 0.16 to 0.3 compared to other products. For existing single-category products, we corrected the phenomenon of overinterpretation in inconsistent areas; the overall accuracy improvement ranged from 2.99% to 20.71%, while the kappa coefficient improvement ranged from 0.22 to 0.56. Thus, our proposed method can combine information from multiple products and serve as an effective method for large areas and even as a global land cover fusion product.

The notorious lithium dendrite growth, causing the safety concern, hinders the practical application of high-capacity Li metal anodes for rechargeable batteries. Here, a robust and highly ionic conductive solid electrolyte interphase (SEI) layer to protect Li metal anode is in-situ constructed by introducing trace additive of tetrapotassium heptaiodobismuthate (K4BiI7) into electrolyte. The K4BiI7-added electrolyte enables Li metal anode to display a stable cycling for over 600 cycles at 1.0 mA cm−2/1.0 mAh cm−2 and over 400 cycles at 5.0 mA cm−2/5.0 mAh cm−2. In situ optical microscopy observations also conform the suppression of Li dendrites at high current density. Moreover, the in-situ SEI layer modified Li anode exhibits an average Coulombic efficiency of 99.57% and less Li dendrite growth. The Li-S full sells with the modified electrolyte also show improved electrochemical performance. This research provides a cost-efficient method to achieve a highly ionic conductive and stable SEI layer toward advanced Li metal anodes.

Abstract Background To investigate the predictive capability of microRNAs (miRNAs) prior treatment for HBsAg clearance in chronic hepatitis B (CHB) treated with pegylated interferon α-2a (PEG-IFNα-2a). Methods The treatment effect was determined by HBsAg clearance and subjects were classified into HBsAg clearance group and non HBsAg clearance group. Differential miRNAs expression in peripheral blood mononuclear cells (PBMC) was screened using microarrays in an identification cohort (n = 20) and validated by quantitative reverse-transcription polymerase chain reaction (qRT-PCR) in a confirmation cohort (n = 47). Receiver operating characteristic curve (ROC), logistic regression and gene ontology (GO)/Pathway analyses were used to evaluate the predictive capability of selected miRNAs for HBsAg clearance and determine their mechanistic roles. Results Twenty-seven subjects (40.3%) acquired HBsAg clearance, ten in the identification cohort and seventeen in the confirmation cohort. Four miRNAs out of twelve (miR-3960, miR-126-3p, miR-335-5p, miR-23a-3p) were verified to be differential expressed by qRT-PCR in the confirmation cohort. Their expression patterns were consistent with the microarray results. Their levels were lower in the response group compared with the nonresponse group (p 0.05). Conclusions We identified differential expressed miRNAs between response and nonresponse groups of PEG-IFNα-2a treatment and demonstrated that miR-3960 was the optimal predictor for HBsAg clearance compared with other miRNAs, but it requires to be further comfired in larger cohort studies. Light detection and ranging (LiDAR) can quickly and accurately obtain 3D point clouds on the surface of rock masses, and on the basis of this, discontinuity information can be extracted automatically. This paper proposes a new method to automatically extract discontinuity information from 3D point clouds on the surface of rock masses. This method first applies the improved K-means algorithm based on the clustering algorithm by fast search and find of density peaks (DPCA) and the silhouette coefficient in the cluster validity index to identify the discontinuity sets of rock masses, and then uses the hierarchical density-based spatial clustering of applications with noise (HDBSCAN) algorithm to segment the discontinuity sets and to extract each discontinuity from a discontinuity set. Finally, the random sampling consistency (RANSAC) method is used to fit the discontinuities and to calculate their parameters. The 3D point clouds of the typical rock slope in the Rockbench repository is used to extract the discontinuity orientations using the new method, and these are compared with the results obtained from the classical approach and the previous automatic methods. The results show that, compared to the results obtained by Riquelme et al. in 2014, the average deviation of the dip direction and dip angle is reduced by 26% and 8%, respectively; compared to the results obtained by Chen et al. in 2016, the average deviation of the dip direction and dip angle is reduced by 39% and 40%, respectively. The method is also applied to an artificial quarry slope, and the average deviation of the dip direction and dip angle is 5.3° and 4.8°, respectively, as compared to the manual method. Furthermore, the related parameters are analyzed. The study shows that the new method is reliable, has a higher precision when identifying rock mass discontinuities, and can be applied to practical engineering.

Abstract Our previous studies unexpectedly indicated that the level of serum hepatitis B core antibody (anti-HBc) was positively correlated with the serum alanine aminotransferase (ALT) level. The aim of this study was to determine whether anti-HBc could serve as a potential biomarker for the detection of liver inflammation in chronic hepatitis B (CHB) patients, especially in patients with normal ALT levels. Serum anti-HBc levels were quantified in 655 treatment-naïve CHB patients, including 45 patients who underwent two liver biopsies (baseline phase and the 78th weeks of antiviral-treatment). Serum anti-HBc levels increased significantly along with the increasing histology activity index (HAI) score. After antiviral-treatment, patients with HAI score reduction had significant decline in serum anti-HBc level. Multivariate analysis showed that anti-HBc was independently associated with moderate-to-severe hepatic inflammation in patients with normal ALT level. Furthermore, serum anti-HBc showed a high diagnostic accuracy for predicting moderate-to-severe inflammation in both hepatitis B e antigen (HBeAg)-positive and HBeAg-negative CHB patients with normal ALT levels (area under the curve, AUC = 0.87 and 0.75; respectively). Thus, anti-HBc may be a strong indicator for assessing the hepatic inflammatory degree and used for antiviral treatment decisions in CHB patients with normal ALT levels.

This paper uses a locking-free nonconforming Crouzeix–Raviart finite element to solve the planar linear elastic eigenvalue problem with homogeneous pure displacement boundary condition. Making full use of the Poincaré inequality, we obtain the guaranteed lower bounds for eigenvalues. Besides, we also use the nonconforming Crouzeix–Raviart finite element to the planar linear elastic eigenvalue problem with the pure traction boundary condition, and obtain the guaranteed lower eigenvalue bounds. Finally, numerical experiments with MATLAB program are reported.

Bordetella holmesii, a species closely related to B. pertussis, has been reported sporadically as a cause of whooping cough–like symptoms. To investigate whether B. pertussis–induced immunity is protective against infection with B. holmesii, we conducted an analysis using 11 human respiratory B. holmesii isolates collected during 2005–2009 from a highly B. pertussis–vaccinated population in Massachusetts. Neither whole-cell (wP) nor acellular (aP) B. pertussis vaccination conferred protection against these B. holmesii isolates in mice. Although T-cell responses induced by wP or aP cross-reacted with B. holmesii, vaccine-induced antibodies failed to efficiently bind B. holmesii. B. holmesii–specific antibodies provided in addition to wP were sufficient to rapidly reduce B. holmesii numbers in mouse lungs. Our findings suggest the established presence of B. holmesii in Massachusetts and that failure to induce cross-reactive antibodies may explain poor vaccine-induced cross-protection.

The activation of host cells by interferon gamma (IFNγ) is essential for inhibiting the intracellular replication of most microbial pathogens. Although significant advances have been made in identifying IFNγ-dependent host factors that suppress intracellular bacteria, little is known about how IFNγ enables cells to recognize, or restrict, the growth of pathogens that replicate in the host cytoplasm. The replication of the cytosolic bacterial pathogen Shigella flexneri is significantly inhibited in IFNγ-stimulated cells, however the specific mechanisms that mediate this inhibition have remained elusive. We found that S. flexneri efficiently invades IFNγ-activated mouse embryonic fibroblasts (MEFs) and escapes from the vacuole, suggesting that IFNγ acts by blocking S. flexneri replication in the cytosol. This restriction on cytosolic growth was dependent on interferon regulatory factor 1 (IRF1), an IFNγ-inducible transcription factor capable of inducing IFNγ-mediated cell-autonomous immunity. To identify host factors that restrict S. flexneri growth, we used whole genome microarrays to identify mammalian genes whose expression in S. flexneri-infected cells is controlled by IFNγ and IRF1. Among the genes we identified was the pattern recognition receptor (PRR) retanoic acid-inducible gene I (RIG-I), a cytoplasmic sensor of foreign RNA that had not been previously known to play a role in S. flexneri infection. We found that RIG-I and its downstream signaling adaptor mitochondrial antiviral signaling protein (MAVS)--but not cytosolic Nod-like receptors (NLRs)--are critically important for IFNγ-mediated S. flexneri growth restriction. The recently described RNA polymerase III pathway, which transcribes foreign cytosolic DNA into the RIG-I ligand 5'-triphosphate RNA, appeared to be involved in this restriction. The finding that RIG-I responds to S. flexneri infection during the IFNγ response extends the range of PRRs that are capable of recognizing this bacterium. Additionally, these findings expand our understanding of how IFNγ recognizes, and ultimately restricts, bacterial pathogens within host cells.

Although the prevalence of Bordetella parapertussis varies dramatically among studies in different populations with different vaccination regimens, there is broad agreement that whooping cough vaccines, composed only of B. pertussis antigens, provide little if any protection against B. parapertussis. In C57BL/6 mice, a B. pertussis whole-cell vaccine (wP) provided modest protection against B. parapertussis, which was dependent on IFN-gamma. The wP was much more protective against an isogenic B. parapertussis strain lacking O-antigen than its wild-type counterpart. O-antigen inhibited binding of wP-induced antibodies to B. parapertussis, as well as antibody-mediated opsonophagocytosis in vitro and clearance in vivo. aP-induced antibodies also bound better in vitro to the O-antigen mutant than to wild-type B. parapertussis, but aP failed to confer protection against wild-type or O antigen-deficient B. parapertussis in mice. Interestingly, B. parapertussis-specific antibodies provided in addition to either wP or aP were sufficient to very rapidly reduce B. parapertussis numbers in mouse lungs. This study identifies a mechanism by which one pathogen escapes immunity induced by vaccination against a closely related pathogen and may explain why B. parapertussis prevalence varies substantially between populations with different vaccination strategies.

Although B. bronchiseptica efficiently infects a wide range of mammalian hosts and efficiently spreads among them, it is rarely observed in humans. In contrast to the many other hosts of B. bronchiseptica, humans are host to the apparently specialized pathogen B. pertussis, the great majority having immunity due to vaccination, infection or both. Here we explore whether immunity to B. pertussis protects against B. bronchiseptica infection. In a murine model, either infection or vaccination with B. pertussis induced antibodies that recognized antigens of B. bronchiseptica and protected the lower respiratory tract of mice against three phylogenetically disparate strains of B. bronchiseptica that efficiently infect naïve animals. Furthermore, vaccination with purified B. pertussis-derived pertactin, filamentous hemagglutinin or the human acellular vaccine, Adacel, conferred similar protection against B. bronchiseptica challenge. These data indicate that individual immunity to B. pertussis affects B. bronchiseptica infection, and suggest that the high levels of herd immunity against B. pertussis in humans could explain the lack of observed B. bronchiseptica transmission. This could also explain the apparent association of B. bronchiseptica infections with an immunocompromised state.

Eco-friendly ceramic capacitors gradually become an important section of pulsed power devices. However, the synchronous realization of ultra-high energy storage density (Wrec > 6 J/cm³) and efficiency (η > 90%) is difficult. Thus, a novel multiscale amelioration strategy in Na0.5Bi0.5TiO3-based ceramics is proposed to achieve ultra-high energy storage density and efficiency. The multiscale amelioration strategy for (Na0.5Bi0.47La0.03)0.94Ba0.06TiO3 (NBLBT) ceramic focuses on grain size, bandgap width, and dielectric relaxor behavior, which can be regulated by introducing Sr(Al0.5Nb0.25Ta0.25)O3 (SANT). On the one hand, the refined grain size and increased bandgap width are conducive to improving the breakdown strength. On the other hand, the optimized dielectric relaxation behavior is beneficial to suppress the remanent polarization. Accordingly, an ultrahigh Wrec = 6.89 J/cm³ and η = 90.1% are simultaneously achieved in 0.84NBLBT- 0.16SANT ceramic. In addition, the sample synchronously possesses excellent thermal and frequency stability (a variation within 5% in Wrec and η), transient discharge rate of t0.9 ~ 78.8 ns and a high-power density of PD ~ 114.5 MW/cm³.
