Abstract In order to better understand the crustal shortening and orogenic uplift in the northeastern margin of the Tibetan Plateau, as well as the geothermal resource effects formed during this process, we used ModEM software to perform 3D MT imaging on broadband magnetotelluric survey points deployed at 710 points in the Gonghe Basin and its surrounding areas. The resistivity model suggests that the Gonghe Basin exhibits a low–high–low overall electrical structure, with high conductivity widely distributed in the middle and lower crust. The resistivity model also reveals a significant discontinuity between high and low resistivity blocks at various depths in the upper and middle crust. These discontinuities are align with the faults observed on the surface related to strong crustal fluctuations, which are connected to high conductors in the middle and upper crust. Using empirical formulas for high-temperature and high-pressure testing of granite, it is estimated that the melting volume of these high conductors ranges from 3 to 43%, demonstrating good "plasticity". These high conductors can act as detachment layers for crustal shortening and deformation during the expansion of the Tibetan Plateau towards the northeast edge and can continuously conduct heat energy upwards, creating a high thermal background in the Gonghe Basin.

The emergence of taxi sharing enhances urban transport efficiency and reduces carbon emissions. Using GPS tracking data from taxis in Chengdu, China, this study first outlines conditions for identifying shareable taxi orders based on their origins and destinations. We then develop a three-phase computational model to optimize matches among all potential shareable orders, calculating the shareable mileage and the proportion of original mileage that could be shared. Our comprehensive temporal and spatial analysis reveal a significant market for taxi sharing in Chengdu, with higher potential on workdays than non-workdays and four distinct demand peaks throughout the day. The morning peak on workdays and the night peak on non-workdays are particularly pronounced. Most shareable orders originate within major city districts. We find a positive correlation between the potential of taxi sharing and average traffic speed, and negative correlations with order volume, regional economic development, and population density. Functional zones related to Enterprises, Motorcycle Services, and Transportation Services exhibit significantly higher sharing potential. Compared to traditional taxi operations, taxi sharing significantly reduces total travel mileage. This quantitative analysis offers insights into the potential demand for taxi sharing among urban residents and may help government authorities optimize taxi resources for the sustainable development of urban transport.

Abstract Hydrogen-isotope storage materials are essential for the controlled nuclear fusion. However, the currently used smelting-ZrCo alloy suffers from rapid degradation of performance due to severe disproportionation. Here, we reveal a defect-derived disproportionation mechanism and report a nano-single-crystal strategy to solve ZrCo’s problems. Single-crystal nano-ZrCo is synthesized by a wet-chemistry method and exhibits excellent comprehensive hydrogen-isotope storage performances, including ultrafast uptake/release kinetics, high anti-disproportionation ability, and stable cycling, far superior to conventional smelting-ZrCo. Especially, a further incorporation of Ti into nano-ZrCo can almost suppress the disproportionation reaction. Moreover, a mathematical relationship between dehydrogenation temperature and ZrCo particle size is established. Additionally, a microwave method capable of nondestructively detecting the hydrogen storage state of ZrCo is developed. The proposed disproportionation mechanism and anti-disproportionation strategy will be instructive for other materials with similar problems.

Abstract Inverse tone mapping technique is widely used to restore the lost textures from a single low dynamic range image. Recently, many stack‐based deep inverse tone mapping networks have achieved impressive results by estimating a set of multi‐exposure images from a single low dynamic range input. However, there are still some limitations. On the one hand, these methods usually set a fixed length for the estimated multi‐exposure stack, which may introduce computational redundancy or cause inaccurate results. On the other hand, they neglect that the difficulties of estimating each exposure value are different and use the identical model to increase or decrease exposure value. To solve these problems, the authors design an exposure decision network to adaptively determine the number of times the exposure of low dynamic range input should be increased or decreased. Meanwhile, the authors decouple the increasing/decreasing process into two sub‐modules, exposure adjustment and optional detail recovery, based on the characteristics of different variations of exposure values. With these improvements, this method can fast and flexibly estimate the multi‐exposure stack from a single low dynamic range image. Experiments on several datasets demonstrate the advantages of the proposed method compared to state‐of‐the‐art inverse tone mapping methods.

This study investigates the propagation dynamics of plasma streamers in a packed-bed dielectric barrier discharge using a 2D particle-in-cell/Monte Carlo collision model. To accurately simulate the high-intensity discharge and streamer propagation mechanism at atmospheric pressure, additional algorithms for particle merging and a new electron mechanism are incorporated into the traditional particle-in-cell/Monte Carlo collision model. To validate the accuracy of this improved model, qualitative comparisons are made with experimental measurements from the existing literature. The results show that the speed of streamer propagation and the distribution of plasma are strongly influenced by the dielectric constant of the packed pellet, which is commonly used as a catalyst. In cases with a moderate dielectric constant, the presence of a strong electric field between the pellet and dielectric layer on the electrode significantly enhances the discharge. This enables the streamer to propagate swiftly along the pellet surface and results in a wider spread of plasma. Conversely, a very high dielectric constant impedes streamer propagation and leads to localized discharge with high intensity. The improved model algorithms derived from this research offer valuable insights for simulating high-density plasma discharge and optimizing plasma processing applications.

Abstract A genetic system, ProTracer, has been recently developed to record cell proliferation in vivo. However, the ProTracer is initiated by an infrequently used recombinase Dre, which limits its broad application for functional studies employing floxed gene alleles. Here we generated Cre-activated functional ProTracer (fProTracer) mice, which enable simultaneous recording of cell proliferation and tissue-specific gene deletion, facilitating broad functional analysis of cell proliferation by any Cre driver.

Summary: CRISPR-Cas system has been repurposed to the promising strategy of CRISPR-based transcriptional interference/activation (CRISPRi/CRISPRa) without eliciting DNA breaks that enables Cas complex a block for transcription initiation or elongation, which greatly expands its application fields and values. However, loss of Cas nuclease ability, especially the endogenous nuclease, may affect genome stability seriously. Here, we found a transcriptional enhancer for genes (CRISPRe) in type I-C system of industrial strain Ketogulonicigenium vulgare by maintaining the natural activity of Cas3 nuclease and introducing the specific motifs that do not trigger immunity. CRISPRe greatly improved the expression of heterologous and endogenous genes and the biosynthesis of products by facilitating transcriptional elongation. Besides, the mechanism for pyrroloquinoline quinone (PQQ) biosynthesis regulated by coupling transcriptional-translational elongation in operon was elucidated. Hence, we enrich the toolbox for CRISPR-Cas system and provide a new framework for gene regulation at transcription.

It is important to evaluate any effects that biochar may have on tomato yield under deficit-irrigation to develop water-saving and yield-increasing best management practices for greenhouse tomato production. For this purpose, greenhouse trials were conducted in 2021 and 2022 with five biochar (B) application rates and three irrigation (W) levels. The B treatments were B0: 0 t ha−1; B1: 15 t ha−1; B2: 30 t ha−1; B3: 45 t ha−1; and B4: 60 t ha−1, and the W levels were W1: 50–70% of field capacity (θf); W2: 60–80% of θf; and W3B0: 70–90% of θf full irrigation without biochar, which was designated as the control. Our objective was to quantify the effects of biochar on the characteristics of tomato sources and sinks to determine, first, the optimal irrigation–biochar combination to achieve high yield tomato production, and, second, evaluate the mechanisms of this effect. At W1 and W2 irrigation levels, the addition of 30–60 t ha−1 and 45–60 t ha−1 biochar could compensate for the adverse effects of deficit-irrigation on the tomato source and sink characteristics. Our results show that in both years the highest tomato yield was obtained with the W2 irrigation level and the B3 biochar application rate, with 52% higher tomato yield compared to the lowest value obtained with the W1 irrigation level and B0 application rate. We conclude that biochar application can improve tomato yield by promoting the filling rather than the building of the sink capacity. The tomato yield was mainly affected by the net photosynthetic rate (Pn), followed by the leaf area, and the leaf chlorophyll content indirectly affected tomato yield through Pn. The W2 irrigation level combined with the B3 biochar application rate resulted in the best water–biochar combination under the experimental conditions.

CO2 geological storage combined with saline recovery (CO2-EWR) is considered to be one of the effective storage methods. Taking the lead in carrying out CO2-EWR technology in the eastern Junggar of Xinjiang can achieve CO2 emission reduction and mean while produce saline water, which can alleviate the local water resources shortage problem to a certain extent, and obtain dual benefits of environment and economy. Previous research mainly focused on generalized models, and the support of engineering practices is lacking. Based on the evaluation results of the suitability of CO2 source - sink matching in the eastern Junggar Basin and the geological data of the first CO2-EWR field pilot test site in China, a 3D heterogeneous model of the Xishanyao Formation of the CO2-EWR test site in the eastern Junggar Basin is constructed to study the potential of the CO2-EWR technology. The results show that the the oretical storage capacity of CO2 at the test site is 1.72 × 106 (P50) tons, and the dynamic storage capacity is 2.14 × 106 tons. When the CO2-EWR technology is adopted, the CO2 dynamic storage capacity can reach 11.18 × 106 tons, which is 5.22 times the CO2 geological storage only, and may increase the production of the saline water resources by 10.17 × 106 tons with a mass ratio of 1 to 0.91 of CO2 sweeping out saline water. Meanwhile, the CO2-EWR technology can effectively slow down the accumulation of reservoir pressure caused by the massive injection of CO2, improve the efficiency of CO2 storage, and increase the saline water production potential. This study can provide theoretical basis and technical support for the implementation of large-scale CO2 geological storage combined with deep saline water production project in the eastern Junggar of Xinjiang.

Abstract Background To evaluate trends in the in-hospital mortality rate for pediatric cardiac surgery procedures between 2005 and 2017 in our center, and to discuss the mortality characteristics of children’s CHD after thoracotomy. Methods This retrospective data were collected from medical records of children underwent CHD surgery between 2005 and 2017. Results A total of 19,114 children with CHD underwent surgery and 444 children died, with the in-hospital mortality was 2.3%. Complex mixed defect CHD had the highest fatality rate (8.63%), left obstructive lesion CHD had the second highest fatality rate (4.49%), right to left shunt CHD had the third highest mortality rate (3.51%), left to right shunt CHD had the lowest mortality rate (χ2 = 520.3,P

Abstract Background Phylogenetic profiling is widely used to predict novel members of large protein complexes and biological pathways. Although methods combined with phylogenetic trees have significantly improved prediction accuracy, computational efficiency is still an issue that limits its genome-wise application. Results Here we introduce a new tree-based phylogenetic profiling algorithm named GFICLEE, which infers common single and continuous loss (SCL) events in the evolutionary patterns. We validated our algorithm with human pathways from three databases and compared the computational efficiency with current tree-based with 10 different scales genome dataset. Our algorithm has a better predictive performance with high computational efficiency. Conclusions The GFICLEE is a new method to infers genome-wide gene function. The accuracy and computational efficiency of GFICLEE make it possible to explore gene functions at the genome-wide level on a personal computer.

Actin depolymerization factors (ADFs), as actin-binding proteins, act a crucial role in plant development and growth, as well as in response to abiotic and biotic stresses. Here, we found that CARK3 plays a role in regulating hypocotyl development and links a cross-talk between actin filament and drought stress through interaction with ADF4. By using bimolecular fluorescence complementation (BiFC) and GST pull-down, we confirmed that CARK3 interacts with ADF4 in vivo and in vitro. Next, we generated and characterized double mutant adf4cark3-4 and OE-ADF4:cark3-4. The hypocotyl elongation assay indicated that the cark3-4 mutant seedlings were slightly longer hypocotyls when compared with the wild type plants (WT), while CARK3 overexpressing seedlings had no difference with WT. In addition, overexpression of ADF4 significantly inhibited long hypocotyls of cark3-4 mutants. Surprisingly, we found that overexpression of ADF4 markedly enhance drought resistance in soil when compared with WT. On the other hand, drought tolerance analysis showed that overexpression of CARK3 could rescue adf4 drought susceptibility. Taken together, our results suggest that CARK3 acts as a regulator in hypocotyl elongation and drought tolerance likely via regulating ADF4 phosphorylation.

Traditional magnetothermal nanoparticles showed great limitations for the diagnosis of tumor location in a dynamic physiological environment in vivo. Here, we show an one-step photo-click method based on the composite structure of γ-Fe2O3@Poly(sucrose allyl ether) magnetic microspheres to meet this challenge. The advantages are to prepare the magnetic microspheres not in extreme conditions and using sucrose as the polymeric matrix to prevent Fe2O3 nanoparticles from agglomeration. A series of in vitro and in vivo experiments showed that γ-Fe2O3@PSAE has excellent stability in biological medium and inappreciable cytotoxicity. Moreover obvious tumor magnetic resonance imaging contrasts were observed based on response of γ-Fe2O3@PSAE. Under an alternating magnetic field, the heat generated by the magnetic microspheres γ-Fe2O3@PSAE thermally ablated H22 hepatoma cells from Kunming male mice, thereby inhibiting tumors. The histopathological studies showed that the tumor growth was significantly inhibited with the increase in the dosage of γ-Fe2O3@PSAE.

Abstract Although chimeric antigen receptor (CAR)-engineered T cells have shown great success in the treatment of B cell malignancies, this strategy has limited efficacy in patients with solid tumors. In mouse CAR-T cells, IL-7 and CCL19 expression have been demonstrated to improve T cell infiltration and CAR-T cell survival in mouse tumors. Therefore, in the current study, we engineered human CAR-T cells to secrete human IL-7 and CCL19 (7 × 19) and found that these 7 × 19 CAR-T cells showed enhanced capacities of expansion and migration in vitro. Furthermore, 7 × 19 CAR-T cells showed superior tumor suppression ability compared to conventional CAR-T cells in xenografts of hepatocellular carcinoma (HCC) cell lines, primary HCC tissue samples and pancreatic carcinoma (PC) cell lines. We then initiated a phase 1 clinical trial in advanced HCC/PC/ovarian carcinoma (OC) patients with glypican-3 (GPC3) or mesothelin (MSLN) expression. In a patient with advanced HCC, anti-GPC3-7 × 19 CAR-T treatment resulted in complete tumor disappearance 30 days post intratumor injection. In a patient with advanced PC, anti-MSLN-7 × 19 CAR-T treatment resulted in almost complete tumor disappearance 240 days post-intravenous infusion. Our results demonstrated that the incorporation of 7 × 19 into CAR-T cells significantly enhanced the antitumor activity against human solid tumor. Trial registration: NCT03198546. Abstract V–VI antimony chalcogenide semiconductors have shown exciting potentials for thin film photovoltaic applications. However, their solar cell efficiencies are strongly hampered by anomalously large voltage loss (>0.6 V), whose origin remains controversial so far. Herein, by combining ultrafast pump–probe spectroscopy and density functional theory (DFT) calculation, the coupled electronic and structural dynamics leading to excited state self‐trapping in antimony chalcogenides with atomic level characterizations is reported. The electronic dynamics in Sb2Se3 indicates a ≈20 ps barrierless intrinsic self‐trapping, with electron localization and accompanied lattice distortion given by DFT calculations. Furthermore, impulsive vibrational coherences unveil key SbSe vibrational modes and their real‐time interplay that drive initial excited state relaxation and energy dissipation toward stabilized small polaron through electron–phonon and subsequent phonon–phonon coupling. This study's findings provide conclusive evidence of carrier self‐trapping arising from intrinsic lattice anharmonicity and polaronic effect in antimony chalcogenides and a new understanding on the coupled electronic and structural dynamics for redefining excited state properties in soft semiconductor materials.

To better understand the geothermal system of the Gonghe Basin, we deployed 471 magnetotelluric survey points with an average distance of 2~3 km, covering the eastern and southern areas of the Basin. We used ModEM inversion software to carry out 3D inversion of 431 survey points and established a 3D-electrical model at a depth of 50 km in the area. The resistivity model shows that the low resistivity in the shallow part of the basin is related to the Cenozoic loose sedimentary cover, while the resistivity values of the mountains around the basin and the magmatic rock uplift zone are higher. The electrical model also shows that the high-conductivity layer is widely distributed in the middle and lower crust (15~35 km) of the basin, and direction of the high-conductivity layer is consistent with that of NW–SE fault in the basin. These high-conductivity layers may be the principal reason for the high heat flow values in the Gonghe Basin. Our resistivity model also shows that there is an obvious discontinuity between high- and low-resistivity blocks at different depths in the middle and upper crust. These discontinuities are consistent with the faults observed on the surface, which are related to the strong topographic relief. Our electrical model shows that these faults in the middle and upper crust are connected with the high-conductivity layer as the channel of heat transfer to the shallow part. Finally, the heat energy is enriched in the Triassic granite to form dry hot rock (HDR). The 3D-magnetotelluric imaging results depict the 3D-distribution characteristics of the geothermal system in the eastern and southern parts of the Gonghe Basin.

The study of short-term creep is essential for understanding the concrete creep process and deformation under alternating stress. Researchers are concentrating on the nano- and micron-scale creep of cement pastes. In the latest RILEM creep database, short-term concrete creep data at hourly or minutely levels are still rare and scarce. In order to describe the short-term creep and creep-recovery behavior of concrete specimens more accurately, the short-term creep and creep-recovery experiments were carried out firstly. The load-holding time varied from 60 s to 1800 s. Secondly, the accuracy of current creep models (B4, B4s, MC2010, and ACI209) in predicting the short-term creep of concrete was compared. It was discovered that the B4, B4s, and MC2010 models all overestimate concrete’s short-term creep, and the ACI model does the opposite. Thirdly, the applicability of the fractional-order-derivative viscoelastic model (with a derivative order between 0 and 1) in the calculation of the short-term creep and creep recovery of concrete is investigated. The calculation results show that the fractional-order derivatives are more suitable for analyzing the static viscoelastic deformation of concrete while the classical viscoelastic model requires a large number of parameters. Therefore, a modified fractional-order viscoelastic model is proposed considering the residual deformation characteristics of concrete after unloading, and the values of the model parameters under different conditions are given with the experimental data.

Breakthrough pressure plays an important role in gas reservoir exploitation and caprock sealing evaluation.In order to simulate the effects of temperature and pressure on CH4 breakthrough pressure in low-permeability sandstones, a step-by-step method was used to conduct CH4 breakthrough experiments on an unsaturated low-permeability sandstone from the Ordos Basin under different combinations of temperature and pressure.The results show that with the increase of temperature or pressure, the breakthrough pressure and time of CH4 show a downward trend, and the pressure change has a more significant impact on the breakthrough process of CH4.It was found that the effect of temperature and pressure on the breakthrough pressure of CH4 is due to the change of interfacial tension and contact angle, and then controls the breakthrough pressure.In addition, it was found that the breakthrough pressure decreases with the increase of the two-phase viscosity ratio; the higher the temperature and pressure, the smaller the breakthrough pressure.Therefore, within this P-T range, for the unsaturated low-permeability sandstone reservoir, the higher the temperature and pressure, the more favorable for gas reservoir development; for the unsaturated low-permeability sandstone caprock, the lower the temperature and pressure, the better the sealing property of the cap rock and the safer the caprock.

Preparing polymers from allyl monomers without being hindered by degradative chain transfer (DCT) remains a challenge. Herein, we report the synthesis and characterization of embolic microspheres based on allyl monomers for transcatheter arterial embolization using a photodriven radical-mediated [3 + 2] cyclization reaction (PRMC) without metal catalysis. The mechanism of PRMC was validated by thermal and spectral experiments, indicating that PRMC overcomes the restrictions of DCT and makes it possible to synthesize a variety of well-defined polymers from inert allyl monomers. The embolization microspheres based on poly(sucrose allyl ether) were characterized by morphological determination, degradation and swelling tests. Cell experiments show that the microspheres have good biocompatibility. The complete arterial occlusion of the embolized kidneys was observed, indicating that microspheres have broad application prospects in embolization interventional therapy.

Gas hydrate is a type of ice like crystals, which could widely form and plug the offshore oil and gas pipelines. In order to reduce the cost of hydrate control in oil and gas transportation, search for new anti-agglomerates (AAs) is always needed. In this work, fourteen different types of polymers from drilling fluid additives were selected and their anti-agglomeration ability on gas hydrate was experimentally studied by using a high-pressure rocking cell. The hydrate volume percentage, slider moving range, and moving velocity were measured for evaluation. Two polymers [acrylamide polymer (AAP) and hydrolyzed polyacrylonitrile ammonium salt (HAPS)] were found to be effective to avoid agglomerating at low hydrate concentration (≤6%). AAP and HAPS could prevent plugging, while the hydrate concentration is lower than 15.12% and 16.92%, respectively. It was speculated that the hydrogen bonding capability of the hydrophilic groups, the length of the hydrophobic chains and polymer molecules, and the antagonism of different functional groups might affect the anti-agglomerating performance of polymers.

BackgroundSepsis-associated encephalopathy (SAE) is a common complication in septic patients with a higher ICU and hospital mortality in adults and poorer long-term outcomes. Clinical presentation may range from mild confusion to convulsions and deep coma; however, little is known about SAE in children. We aimed to retrospectively analyze the data for children with sepsis, to illustrate the epidemiology, performance, and adverse outcome, and to evaluate the association between risk factors and SAE in children.MethodsAll children with sepsis who were admitted to the Department of Pediatrics, Guangdong Provincial People's Hospital, Guangzhou, Guangdong, China from January 2010 to December 2020 were retrospectively analyzed.ResultsA total of 210 patients with sepsis were retrospectively assigned to the SAE and non-SAE groups, of which 91 (43.33%) were diagnosed with SAE with a mortality of 6.70% (14/210). Significant differences were observed in the level of white blood platelet, platelets, international normalized ratio, prothrombin time, activated partial thromboplastin time, total protein, Ccr, UREA, blood urea nitrogen, alanine transaminase, aspartate transaminase, creatine kinase, creatine kinase isoenzymes, lactate dehydrogenase, procalcitonin, and lactic acid (p < 0.05). In the risk assessment scales, significant differences were observed in the modified Glasgow Coma score, PCIS, Pediatric Logistic Organ Dysfunction Score 2 (PELOD-2), Pediatric Sequential Organ Failure Assessment Score, and Pediatric Risk of Mortality III (p < 0.05). The incidence of septic shock, acute kidney disease, liver dysfunction, and coagulation disorder were higher in the SAE group (p < 0.05). The mechanical ventilation time ([6.57 d ± 16.86 d] vs. [2.05 d ± 5.79 d]; p < 0.001), CRRT time ([1.74 d ± 6.77 d] vs. [0.11 d ± 0.63 d]; p < 0.001), ICU stay time ([299.90 h ± 449.50 h] vs. [177.67 h ± 245.36 h]); p < 0.001 was longer than that of non-SAE. Both the PCT, Ca2+, septic shock, PELOD-2, and midazolam were identified as independent risk factors, and fentanyl was a protective factor for SAE in pediatric patients (p < 0.05). The main clinical neurological symptoms consisted of agitation, hypnosia, hypnosis alternates agitated, anterior fontanelle full/bulging/high tension, coma, muscle hypertonia, muscle hypotonia, hyperreflexia, focal seizure, and generalized seizure.ConclusionsThe incidence of SAE in children was found high and the prognosis poor. In this retrospective study, the identified patients were more susceptible to SAE, with an inflammatory storm with hypocalcemia or septic shock. The use of midazolam will increase the occurrence of SAE, whereas fentanyl will reduce the incidence of SAE, and PELOD-2 may predict the occurrence of SAE.

Abstract Background Phylogenetic species trees are widely used in inferring evolutionary relationships. Existing software and algorithms mainly focus on phylogenetic inference. However, less attention has been paid to intermediate steps, such as processing extremely large sequences and preparing configure files to connect multiple software. When the species number is large, the intermediate steps become a bottleneck that may seriously affect the efficiency of tree building. Results Here, we present an easy-to-use pipeline named PhySpeTree to facilitate the reconstruction of species trees across bacterial, archaeal, and eukaryotic organisms. Users need only to input the abbreviations of species names; PhySpeTree prepares complex configure files for different software, then automatically downloads genomic data, cleans sequences, and builds trees. PhySpeTree allows users to perform critical steps such as sequence alignment and tree construction by adjusting advanced options. PhySpeTree provides two parallel pipelines based on concatenated highly conserved proteins and small subunit ribosomal RNA sequences, respectively. Accessory modules, such as those for inserting new species, generating visualization configurations, and combining trees, are distributed along with PhySpeTree. Conclusions Together with accessory modules, PhySpeTree significantly simplifies tree reconstruction. PhySpeTree is implemented in Python running on modern operating systems (Linux, macOS, and Windows). The source code is freely available with detailed documentation (https://github.com/yangfangs/physpetools).

As an important equipment for high-speed railway (HSR) to obtain electric power from outside, the state of the pantograph will directly affect the operation safety of HSR. In order to solve the problems that the current pantograph detection method is easily affected by the environment, cannot effectively deal with the interference of external scenes, has a low accuracy rate and can hardly meet the actual operation requirements of HSR, this study proposes a pantograph detection algorithm. The algorithm mainly includes three parts: the first is to use you only look once (YOLO) V4 to detect and locate the pantograph region in real-time; the second is the blur and dirt detection algorithm for the external interference directly affecting the high-speed camera (HSC), which leads to the pantograph not being detected; the last is the complex background detection algorithm for the external complex scene “overlapping” with the pantograph when imaging, which leads to the pantograph not being recognized effectively. The dirt and blur detection algorithm combined with blob detection and improved Brenner method can accurately evaluate the dirt or blur of HSC, and the complex background detection algorithm based on grayscale and vertical projection can greatly reduce the external scene interference during HSR operation. The algorithm proposed in this study was analyzed and studied on a large number of video samples of HSR operation, and the precision on three different test samples reached 99.92%, 99.90% and 99.98%, respectively. Experimental results show that the algorithm proposed in this study has strong environmental adaptability and can effectively overcome the effects of complex background and external interference on pantograph detection, and has high practical application value.

With the development of molecular sequencing approaches, many taxonomic and phylogenetic problems of the genus Allium L. have been solved; however, the phylogenetic relationships of some subgenera or sections, such as section Bromatorrhiza, remain unresolved, which has greatly impeded our full understanding of the species relationships among the major clades of Allium. In this study, the complete chloroplast (cp) genomes of nine species in the Allium sect. Bromatorrhiza were determined using the Illumina paired-end sequencing, the NOVOPlasty de novo assembly strategy, and the PGA annotation method. The results showed that the cp genome exhibited high conservation and revealed a typical circular tetrad structure. Among the sect. Bromatorrhiza species, the gene content, SSRs, codon usage, and RNA editing site were similar. The genome structure and IR regions’ fluctuation were investigated while genes, CDSs, and non-coding regions were extracted for phylogeny reconstruction. Evolutionary rates (Ka/Ks values) were calculated, and positive selection analysis was further performed using the branch-site model. Five hypervariable regions were identified as candidate molecular markers for species authentication. A clear relationship among the sect. Bromatorrhiza species were detected based on concatenated genes and CDSs, respectively, which suggested that sect. Bromatorrhiza is monophyly. In addition, there were three genes with higher Ka/Ks values (rps2, ycf1, and ycf2), and four genes (rpoC2, atpF, atpI, and rpl14) were further revealed to own positive selected sites. These results provide new insights into the plastome component, phylogeny, and evolution of Allium species.

Abstract Background This study aimed to construct a biological age assessment formula for the Chinese population and to explore the effectiveness of double filtration plasmapheresis for anti-ageing and longevity. Methods 915 subjects were recruited, including 584 (63.8%) males and 331 females (36.2%). Male age was 50.94±10.60 (mean±SD), and female age was 51.20±11.84 (mean±SD). 34 blood markers were detected in the laboratory. The ageing biomarkers were determined by statistical correlation analysis and redundancy analysis, and the biological age assessment formula was established by multiple linear regression analysis. Paired sample T test was used to analyse the elimination effect of double filtration plasmapheresis on aging biomarkers. Results Based on the comprehensive blood test and analysis, the ageing biomarkers were screened, and the male and female biological age assessment formulas were established. Then, the elimination of ageing biomarkers by double filtration plasmapheresis was examined. Double filtration plasmapheresis can eliminate ageing biomarkers, with an average of 4.47 years decrease in age for males and 8.36 years for females. Conclusion So, biological age provides a scientific tool for assessing ageing, and double filtration plasmapheresis is safe and might be effective for anti-ageing and longevity. However, the effect of plasmapheresis is expected to be transient, so further studies are needed to plan the number and range of the plasmapheresis procedures necessary to consistently lower the parameters under study.

Abstract Herein, a tumor-targeted multifunctional theranostic agent was synthetized using a facile method, combining four clinically approved materials: artesunate (Arte), human serum albumin (HSA), folic acid (FA), and indocyanine green (ICG). The obtained nanocomposites (FA-IHA NPs) showed an excellent photo- and physiological stability. The ICG in the FA-IHA NPs was used not only for near infrared (NIR) fluorescence imaging, but also for photothermal and photodynamic (PTT-PDT) therapy under a single NIR irradiation. In addition, the NIR irradiation (808 nm, 1 W/cm2) could trigger Arte release that showed enhanced chemotherapeutic effect. Through fluorescence imaging, the cell uptake and tumor accumulation of FA-IHA NPs were observed in vitro and in vivo, analyzed by confocal microscopy and NIR fluorescence imaging in tumor xenograft mice. Based on the diagnostic results, FA-IHA NPs at 24 h post injection and combined with NIR irradiation (808 nm, 1 W/cm2) could efficiently suppress tumor growth through a photo-chemo combination therapy, with no tumor recurrence in vitro and in vivo. The obtained results suggested that FA-IHA NPs are promising photo-chemo theranostic agents for future clinical translation.

There are many kinds of vibration forms of drillstring in petroleum drilling. Whirl is one of the main vibration modes when drilling tools are drilling, and it is also the main reason of fatigue failure of the drillstring. In this paper, the causes of drillstring whirl are analyzed. The results show that the speed of rotation (RPM) is a major factor that affects the severity of the whirl. The greater the speed of rotation, the more intense the whirl. The whirl of the drillstring will produce bending stress. The severity of the whirl of the drillstring is the main factor affecting the bending stress. The increase of RPM and weight on bit (WOB) will lead to the increase of bending stress. The RPM has a great influence on the bending stress. Given the complex stress condition of drillstring, the calculation model of safety factor is established. The bending stress and safety factor are calculated under the different WOB and RPM. The relationship between dynamic bending stress and the safety factor of drilling tools is analyzed. Bending stress has great influence on the safety factor of drillstring. The variation trend of safety factor is opposite to that of bending stress caused by whirl. It provides a theoretical reference for the optimization of drilling tools in drilling engineering construction.

The water film depth is a key variable that affects traffic safety under rainfall conditions. According to the Federal Highway Administration, approximately 5700 people are killed and more than 544 700 people are injured in crashes on wet pavements annually. While several studies have attempted to address water film depth issues by establishing prediction models, a few focused on the relationship among road geometric features, capacity of drainage facilities and water film depth. To ascertain the influence of the geometric features of road and facility drainage capacities on the water film depth, the road geometry features were first classified into four types, and the facility drainage capacities were considered from three aspects in this study. Furthermore, the concept of short-time rainfall grade was proposed according to the results of the field test. Finally, the theoretical prediction model for the water film depth was conceived, based on the geometric features of road and facility drainage capacities with different rainfall intensities. Compared with the traditional regression prediction models, the theoretical prediction model clearly shows the effects of the geometric features of road and facility drainage capacities. When the road drainage facilities have no drainage capacity, the water film depth increases rapidly with the rainfall intensity. This model can be used to predict the water film depth of road surfaces on rainy days, evaluate the effect of rainfall on the driving environment, and provide guidance for determining safety control measures on rainy days.

Most of the existing research on shared autonomous vehicles (SAVs) and road congestion pricing have studied the short-term impact on traffic flow. These types of studies focused on the influences on mobility and ignored the long-term impacts on regional job accessibility. Given this, from the perspective of land use and transportation integration, this study explored the long-term effects of SAVs and cordon-based congestion pricing on regional land use, transportation, and job accessibility. The contributions of this study have been summarized by the following three purposes. First, to the best of the authors’ knowledge, this study was the first attempt to identify the long-term impact of the combination of these two technologies on regional job accessibility. Second, compared to the previous research methodology, this study adopted the land use and transportation integrated model (TRANUS model) and scenario planning to ensure the comprehensiveness and validity of the results. Third, this study analyzed the spatial heterogeneity of the impact of the combination of the two technologies on regional job accessibility in different areas with different built-environment attributes. To realize this purpose, this study quantitatively classified traffic analysis zones (TAZs) using data mining technology, i.e., factor analysis and clustering analysis. Results showed that the introduction of SAVs will contribute to job and population development in the charging zone and reduce the negative effect of road congestion pricing. From the perspective of reducing the average travel time between TAZs, the best strategy will be to implement SAVs alone, followed by integrated use of SAVs and road congestion pricing, while the worst strategy will be to implement the cordon-based congestion pricing policy alone. By comparison, from the perspective of improving regional job accessibility, the effect of introducing SAVs was better than that of road congestion pricing, while the combination of these two technologies was not superior to implementing SAVs alone.

It is of great significance to explore the strategy of regulated deficit irrigation (RDI) under mulched drip irrigation to stabilize tomato yield and improve quality and efficiency. This experimental study was conducted on a drip-irrigated greenhouse in two consecutive years (2020 and 2021). Three deficit levels were set for the flowering and fruit development stage (Stage I), and three were set for the fruit-ripening stage (Stage II). As a combination evaluation method, the fuzzy Borda model was used to optimize the RDI strategy of greenhouse tomato. The results showed that the net photosynthetic rate, stomatal conductance, transpiration rate, and total shoot biomass of tomato decreased with an increase in the water deficit, while the intercellular CO2 concentration had an opposite trend. The mild and moderate water deficit at Stage I reduced tomato yield by 16–24% and 30–40% compared to full irrigation. The water deficit at Stage II was able to improve various quality parameters and the water-use efficiency of tomato; the irrigation water-use efficiency (32.8–33.9 kg/m3) and leaf water-use efficiency (3.2–3.6 μmol/mmol) were the highest when the soil water content was 70–90% θf (field capacity) at Stage I and 40–60% θf at Stage II (T3). Based on the fuzzy Borda combination evaluation model, T3 was determined as the treatment with stable yield, high quality, and efficient irrigation under the experimental conditions. The irrigation regime was as follows: irrigating 20–25 mm in the transplanting stage, no irrigation in the seedling stage, irrigating 193.2–220.8 mm at Stage I, and then irrigating 27.6 mm at Stage II.

Fragaria nilgerrensis Schlecht. (wild strawberry) is widely distributed in Southwest China, characterized by stress tolerance and the fruits of a notable peach aroma. So far there is only limited knowledge of variability and genetic structure in this species. Using AFLP markers, we investigated the genetic variability of 37 plants of F. nilgerrensis sampled in six main mountain areas of Sichuan Province and analyzed their genetic structure. Genetic similarity according to Nei and Li was used for cluster analysis based on UPGMA method and Agglomerative Hierarchical Clustering. Stratification of plants into more distinctive genetic groups was determined using Bayesian structure analysis. Six primer combinations produced a total of 1302 fragments of which 818 (62.8%) were polymorphic. Bayesian analysis showed the 37 plants of F. nilgerrensis grouped into five distinctive genetic groups. Most of the plants from the same mountain area clustered into the same genetic group, indicating each area as an area with the unique genetic profile. The genetic parameters analyzed here indicate a huge genetic variability of F. nilgerrensis in Sichuan Province. Our results provide reference data for surveying and protecting the germplasm resources of F. nilgerrensis that could be used in strawberry breeding programs.

Abstract The heat shock response is crucial for organisms against heat-damaged proteins and maintaining homeostasis at a high temperature. Heterologous expression of eukaryotic molecular chaperones protects Escherichia coli from heat stress. Here we report that expression of the plant E3 ligase BnTR1 significantly increases the thermotolerance of E. coli. Different from eukaryotic chaperones, BnTR1 expression induces the accumulation of heat shock factor σ32 and heat shock proteins. The active site of BnTR1 in E. coli is the zinc fingers of the RING domain, which interacts with DnaK resulting in stabilizing σ32. Our findings indicate the expression of BnTR1 confers thermoprotective effects on E. coli cells, and it may provide useful clues to engineer thermophilic bacterial strains.

Background: The incidence of chronic diseases, such as cardiovascular disease, diabetes, overweight, obesity, cancer and other diseases has been increasing. It is a huge challenge to public health industry about how to provide risk intervention and preventive medical services and explore advanced technology platform for effective prevention and control of chronic diseases. Methods: We collaborated domestic and international experts on preventive medicine, and analyzed pathogenesis and risk factors for the major chronic diseases. Results: We established Target Elimination--denatured and unstable proteins, environmental toxins, metabolic wastes, immunosuppressive factors and chronic inflammatory factor (TE-PEMIC) system that offer us the standard and methods to eliminate and intervene pathogenic factors of the chronic diseases. Conclusion: It provides new researches and exploring new ideas to prevent and intervene chronic diseases by applying the TE-PEMIC chronic diseases prevention medical technology system.

CO2 geological storage in deep saline aquifers is an effective way to reduce CO2 emissions. The injection of CO2 inevitably causes a significant pressure increase in reservoirs. When there exist faults which cut through a deep reservoir and shallow aquifer system, there is a risk of the shallow aquifer being impacted by the changes in reservoir hydrodynamic fields. In this paper, a radial model and a 3D model are established by TOUGH2-ECO2N for the reservoir system in the CO2 geological storage demonstration site in the Junggar Basin to analyze the impact of the CO2 injection on the deep reservoir pressure field and the possible influence on the surrounding shallow groundwater sources. According to the results, the influence of CO2 injection on the reservoir pressure field in different periods and different numbers of well is analyzed. The result shows that the number of injection wells has a significant impact on the reservoir pressure field changes. The greater the number of injection wells is, the greater the pressure field changes. However, after the cessation of CO2 injection, the number of injection wells has little impact on the reservoir pressure recovery time. Under the geological conditions of the site and the constant injection pressure, although the CO2 injection has a significant influence on the pressure field in the deep reservoir, the impact on the shallow groundwater source area is minimal and can be neglected and the existing shallow groundwater sources are safe in the given project scenarios.

CO2 geological storage (CGS) proved to be an effective way to mitigate greenhouse gas emissions, and CO2-enhanced water recovery (CO2-EWR) technology may improve the efficiency of CO2 injection and saline water production with potential economic value as a means of storing CO2 and supplying cooling water to power plants. Moreover, the continuous injection of CO2 may cause a sharp increase for pressure in the reservoir system, so it is important to determine reasonable reservoir pressure control strategies to ensure the safety of the CGS project. Based upon the typical formation parameters of the China Geological Survey CO2-EWR test site in the eastern Junggar Basin, a series of three-dimensional (3D) injection-extraction models with fully coupled wellbores and reservoirs were established to evaluate the effect of the number of production wells and the well spacing on the enhanced efficiency of CO2 storage and saline production. The optimal key parameters that control reservoir pressure evolution over time are determined. The numerical results show that a smaller spacing between injection and production wells and a larger number of production wells can enhance not only the CO2 injection capacity but also the saline water production capacity. The effect of the number of production wells on the injection capacity and production capacity is more significant than that of well spacing, and the simulation scenario with 2 production wells, one injection well, and a well spacing of 2 km is more reasonable in the demonstration project of Junggar Basin. CO2-EWR technology can effectively control the evolution of the reservoir pressure and offset the sharp increase in reservoir pressure caused by CO2 injection and the sharp decrease of reservoir pressure caused by saline production. The main controlling factors of pressure evolution at a certain spatial point in a reservoir change with time. The monitoring pressure drops at the beginning and is controlled by the extraction of water. Subsequently, the injection of CO2 plays a dominant role in the increase of reservoir pressure. Overall, the results of analysis provide a guide and reference for the CO2-EWR site selection, as well as the practical placement of wells.

Wire-arc additive manufacturing (WAAM) has been considered as one of the potential additive-manufacturing technologies to fabricate large components. However, its industrial application is still limited by the existence of stress and distortion. During the process of WAAM, the scanning pattern has an important influence on the temperature field, distortion and final quality of the part. Four kinds of deposition patterns, including sequence, symmetry, in–out and out–in, were designed to deposit H13 steel in this study. An in situ measurement system was set up to record the temperature history and the progress of accumulated distortion of the parts during deposition. An S value was proposed to evaluate the distortion of the substrate. It was shown that the distortion of the part deposited by sequence was significantly larger than those of other parts. The distortion deposited by the out–in pattern decreased by 68.6% compared with sequence. The inherent strain method and strain parameter were introduced to expose the mechanism of distortion reduction caused by pattern variation.

The miniaturization and integration of photonic devices are new requirements in the fast-growing optics field. In this paper, we focus on a feature-rich sub-wavelength nanograting-coated single-layer metal film. The numerical results show that the reflection behaviors of this proposed structure can realize bidirectional dual-channel ultra-narrowband polarized filtering and bidirectional wavelength-modulated sensing in a wide refractive index (RI) range from 1.0 to 1.4 for incident angle of 10° with transverse-magnetic (TM) polarized illumination at wavelengths between 550 nm to 1500 nm. Moreover, the bidirectional properties of filtering and sensing are not obviously decreased when increasing incident angle from 10° to 30°, and decreasing incident angle from 10° to 0°. The calculated RI sensitivity can be up to 592 nm/RIU with a high figure of merit (FOM) of 179.4 RIU−1. More to the point, this nanograting has a simple structure and is less sensitive to the height and shape of grating ridge, which provides great convenience for the fabrication of devices. The other thing that is going on is that this structure can also realize synchronously tunable color filtering, including green to red, with high color purity in the visible band by choosing the period. The underlying physical mechanism is analyzed in detail, and is primarily attributed to surface plasmon polariton (SPP) resonance and dipole resonance at double plasmon resonance wavelengths. This work has tremendous potential in developing multipurpose and high-performance integrated optical devices such as spectral filters, colored displays and plasmon biomedical sensors.

Apple mosaic disease has a great influence on apple production. In this study, an investigation into the incidence of apple mosaic disease in southwest China was performed, and the pathogen associated with the disease was detected. The results show that 2869 apple trees with mosaic disease were found in the Sichuan, Yunnan, and Guizhou Provinces, with an average incidence of 9.6%. Although apple mosaic virus (ApMV) is widespread in apples worldwide, the diseased samples were negative when tested for ApMV. However, a novel ilarvirus (apple necrotic mosaic virus, ApNMV) was identified in mosaic apple leaves which tested negative for ApMV. RT-PCR analysis indicated that ApNMV was detected in 322 out of 357 samples with mosaic symptoms. Phylogenetic analysis of coat protein (CP) sequences of ApNMV isolates suggested that, compared with ApMV, ApNMV was closer to prunus necrotic ringspot virus (PNRSV). The CP sequences of the isolates showed the diversity of ApNMV, which may enable the virus to adapt to the changeable environments. In addition, the pathology of mosaic disease was observed by microscope, and the result showed that the arrangement of the tissue and the shape of the cell, including the organelle, were seriously destroyed or drastically changed.

The multistage and discontinuous nature of the injection process used in the geological storage of CO2 causes reservoirs to experience repeated loading and unloading. The reservoir permeability changes caused by this phenomenon directly impact the CO2 injection process and the process of CO2 migration in the reservoirs. Through laboratory experiments, variations in the permeability of sandstone in the Liujiagou formation of the Ordos CO2 capture and storage (CCS) demonstration project were analyzed using cyclic variations in injection pressure and confining pressure and multistage loading and unloading. The variation in the micropore structure and its influence on the permeability were analyzed based on micropore structure tests. In addition, the effects of multiple stress changes on the permeability of the same type of rock with different clay minerals content were also analyzed. More attention should be devoted to the influence of pressure variations on permeability in evaluations of storage potential and studies of CO2 migration in reservoirs in CCS engineering.

In China, south of the Yangtze River, there are a large number of carbon sources, while the Sichuan Basin is the largest sedimentary basin; it makes sense to select the targets for CO2 geological storage (CGUS) early demonstration. For CO2 enhanced oil and gas, coal bed methane recovery (CO2-EOR, EGR, and ECBM), or storage in these depleted fields, the existing oil, gas fields, or coal seams could be the target areas in the mesoscale. This paper proposed a methodology of GIS superimposed multisource information assessment of geological suitability for CO2 enhanced water recovery (CO2-EWR) or only storage in deep saline aquifers. The potential per unit area of deep saline aquifers CO2 storage in Central Sichuan is generally greater than 50 × 104 t/km2 at P50 probability level, with Xujiahe group being the main reservoir. CO2 storage potential of depleted gas fields is 53.73 × 108 t, while it is 33.85 × 108 t by using CO2-EGR technology. This paper recommended that early implementation of CGUS could be carried out in the deep saline aquifers and depleted gas fields in the Sichuan Basin, especially that of the latter because of excellent traps, rich geological data, and well-run infrastructures.

Abscisic acid (ABA) signaling plays important roles in plant growth, development and adaptation to various stresses. RCAR1/PYL9 has been known as a cytoplasm and nuclear ABA receptor in Arabidopsis. To obtain further insight into the regulatory mechanism of RCAR1/PYL9, a yeast two-hybrid approach was performed to screen for RCAR1/PYL9-interacting proteins and an R2R3-type MYB transcription factor, AtMYB44, was identified. The interaction between RCAR1/PYL9 and AtMYB44 was further confirmed by glutathione S-transferase (GST) pull-down and bimolecular fluorescence complementation (BiFC) assays. Gene expression analysis showed that AtMYB44 negatively regulated the expression of ABA-responsive gene RAB18, in contrast to the opposite role reported for RCAR1/PYL9. Competitive GST pull-down assay and analysis of phosphatase activity demonstrated that AtMYB44 and ABI1 competed for binding to RCAR1/PYL9 and thereby reduced the inhibitory effect of RCAR1/PYL9 on ABI1 phosphatase activity in the presence of ABA in vitro. Furthermore, transient activation assay in protoplasts revealed AtMYB44 probably also decreased RCAR1/PYL9-mediated inhibition of ABI1 activity in vivo. Taken together, our work provides a reasonable molecular mechanism of AtMYB44 in ABA signaling.

Drought is a major limiting factor for plant growth and crop productivity. Many Calcineurin B-like interacting protein kinases (CIPKs) play crucial roles in plant adaptation to environmental stresses. It is particularly essential to find the phosphorylation targets of CIPKs and to study the underlying molecular mechanisms. In this study, we demonstrate that CIPK11 acts as a novel component to modulate drought stress in plants. The overexpression of CIPK11 (CIPK11OE) in Arabidopsis resulted in the decreased tolerance of plant to drought stress. When compared to wild type plants, CIPK11OE plants exhibited higher leaf water loss and higher content of reactive oxygen species (ROS) after drought treatment. Additionally, a yeast two hybrid screening assay by using CIPK11 as a bait captures Di19-3, a Cys2/His2-type zinc-finger transcription factor that is involved in drought stress, as a new interactor of CIPK11. Biochemical analysis revealed that CIPK11 interacted with Di19-3 in vivo and it was capable of phosphorylating Di19-3 in vitro. Genetic studies revealed that the function of CIPK11 in regulating drought stress was dependent on Di19-3. The transcripts of stress responsive genes, such as RAB18, RD29A, RD29B, and DREB2A were down-regulated in the CIPK11OE plants. Whereas overexpression of CIPK11 in di19-3 mutant background, expression levels of those marker genes were not significantly altered. Taken together, our results demonstrate that CIPK11 partly mediates the drought stress response by regulating the transcription factor Di19-3.

Abscisic acid (ABA) plays a fundamental role in plant growth and development, as well as in the responses to abiotic stresses. Previous studies have revealed that many components in ABA and drought stress signaling pathways are ubiquitinated by E3 ligases. In this study, AtPPRT1, a putative C3HC4 zinc-finger ubiquitin E3 ligase, was explored for its role in abiotic stress response in Arabidopsis thaliana. The expression of AtPPRT1 was induced by ABA. In addition, the β-glucuronidase (GUS) gene driven by the AtPPRT1 promoter was more active in the root hair zone and root tips of primary and major lateral roots of young seedlings in the presence of ABA. The assays for seed germination, stomatal aperture, root length, and water deficit demonstrated that the AtPPRT1-overexpressing Arabidopsis was insensitive to ABA and sensitive to drought stress compared with wild-type (WT) plants. The analysis by quantitative real-time PCR (qRT-PCR) revealed that the expression of three stress-inducible genes (AtRAB18, AtERD10, and AtKIN1) were upregulated in the atpprt1 mutant and downregulated in AtPPRT1-overexpressing plants, while two ABA hydrolysis genes (AtCYP707A1 and AtCYP707A3) were downregulated in the atpprt1 mutant and upregulated in AtPPRT1-overexpressing plants in the presence of ABA. AtPPRT1 was localized in the mitochondria. Our findings indicate that AtPPRT1 plays a negative role in ABA and drought stress responses.

The plant hormone abscisic acid (ABA) play essential roles in numerous physiological processes such as seed dormancy, seed germination, seeding growth and responses to biotic and abiotic stresses. Such biological processes are tightly controlled by a complicated regulatory network including ABA homoeostasis, signal transduction as well as cross-talking among other signaling pathways. It is known that ABA homoeostasis modulated by its production, inactivation, and transport pathways is considered to be of great importance for plant development and stress responses. Most of the enzymes and transporters involved in ABA homoeostasis have been largely characterized and they all work synergistically to maintain ABA level in plants. Increasing evidence have suggested that transcriptional regulation of the genes involved in either ABA production or ABA inactivation plays vital roles in ABA homoeostasis. In addition to transcription factors, such progress is also regulated by microRNAs and newly characterized root to shoot mobile peptide-receptor like kinase (RLKs) mediated long-distance signal transduction. Thus, ABA contents are always kept in a dynamic balance. In this review, we survey recent research on ABA production, inactivation and transport pathways, and summarize some latest findings about the mechanisms that regulate ABA homoeostasis.

Drought stress hinders plant growth and development, and abscisic acid (ABA) stimulates plants to respond to drought. Here, to increase plant tolerance to drought, we designed three synthetic promoters (Ap, Dp, ANDp) to determine transcription activity and drought stress resistance in plants resulting from combinations of (1) synthetic promoters and (2) the functional genes CARK1 (cytosolic ABA receptor kinase 1) and RCAR11 (regulatory components of ABA receptor 11). Transient expression of eGFP and the dual-luciferase assay demonstrated that the basal transcriptional activities of Ap and ANDp were present at low levels under normal conditions, while the synthetic promoters were apparently induced upon either treatment of exogenous ABA or co-transformation with effector DREB2A (dehydration-responsive element binding protein 2A). Analysis of the transgenic plants (Ap:CARK1, Dp:CARK1, ANDp:CARK1, and Dp:RCAR11-Ap:CARK1) showed that the synthetic promoters Ap, Dp, and ANDp increased the expression of exogenous genes in transgenic plants upon treatment of ABA or d-mannitol. ANDp:CARK1 and Dp:RCAR11-Ap:CARK1 transgenic plants were sensitive to ABA and d-mannitol during cotyledon greening and root growth. A drought tolerance assay revealed that ANDp:CARK1 and Dp:RCAR11-Ap:CARK1 exhibited a higher survival rate than others upon drought stress. These results indicate that the combinations ANDp:CARK1 and Dp:RCAR11-Ap:CARK1 can be used to generate drought stress resistance in plants.

The phytohormone abscisic acid (ABA) regulates plant growth, the developmental process, and abiotic stresses. ABA signaling is induced in response to mediate plant acclimation to environmental challenges, including high salinity and drought. The ABA-binding receptors (RCAR/PYR1/PYL), composing of 14 members, are the core components of the ABA-signaling pathway. Here, we observed that the three subfamilies within the RCARs showed different expression patterns at the basal and exogenous ABA levels. Subsequently, we generated transgenic plants overexpressing subfamily III, RCAR11–RCAR14, respectively. The transgenic plants showed increased ABA sensitivity in seed germination and post-germination seedling establishment and root length. Further studies revealed that the overexpressing subfamily III transgenic plants enhanced drought resistance, increased water-use efficiency, and accelerated stress-responsive gene expression compared with the wild-type plants. These findings confirm that the subfamily III plays a key role in ABA-mediated developmental processes and, more importantly, is involved in drought tolerance in the ABA-dependent pathway.

The voltage-dependent anion channel (VDAC) is the major transport protein in the outer membrane of mitochondria and plays crucial roles in energy metabolism, apoptosis, and metabolites transport. In plants, the expression of VDACs can be affected by different stresses, including drought, salinity and pathogen defense. In this study, we investigated the expression pattern of AtVDAC2 in A. thaliana and found ABA suppressed the accumulation of AtVDAC2 transcripts. Further, phenotype analysis of this VDAC deregulated-expression transgenic Arabidopsis plants indicated that AtVDAC2 anti-sense line showed an ABA-insensitivity phenotype during the early seedling development under ABA treatment. The results suggested that AtVDAC2 might be involved in ABA signaling in A. thaliana.

DNA methylation is a critical epigenetic mechanism involved in key cellular processes. Its deregulation has been linked to many human cancers including esophageal squamous cell carcinoma (ESCC). This study was designed to explore the whole methylation status of ESCC and to identify potential plasma biomarkers for early diagnosis. We used Infinium Methylation 450k array to analyze ESCC tissues (n = 4), paired normal surrounding tissues (n = 4) and normal mucosa from healthy individuals (n = 4), and combined these with gene expression data from the GEO database. One hundred and sixty eight genes had differentially methylated CpG sites in their promoter region and a gene expression pattern inverse to the direction of change in DNA methylation. These genes were involved in several cancer-related pathways. Three genes were validated in additional 42 ESCC tissues and paired normal surrounding tissues. The methylation frequency of EPB41L3, GPX3, and COL14A1 were higher in tumor tissues than in normal surrounding tissues (P < 0.017). The higher methylation frequency of EPB41l3 was correlated with large tumor size (P = 0.044) and advanced pT tumor stage (P = 0.001). The higher methylation frequency of GPX3 and COL14A1 were correlated with advanced pN tumor stage (P = 0.001 and P < 0.001). The methylation of EPB41L3, GPX3, and COL14A1 genes were only found in ESCC patients' plasma, but not in normal individuals upon testing 42 ESCC patients and 50 healthy individuals. Diagnostic sensitivity was increased when methylation of any of the 3 genes were counted (64.3% sensitivity and 100% specificity). These differentially methylated genes in plasma may be used as biomarkers for early diagnosis of ESCC.
