Flower color is one of the most important ornamental traits of tulips (Tulipa gesneriana). Five typical tulip cultivars were selected to identify the flavonoid components and analyze their key gene expression in their tepals. Firstly, after preliminary determination of the pigment type, the flavonoids were identified by UPLC-Q-TOF-MS. A total of 17 anthoxanthins were detected in the five cultivars. The total anthoxanthin content in the white tulip and the red tulip showed a similar decreasing trend, while an increasing trend was observed in the black tulip. Similarly, a total of 13 anthocyanins were detected in five tulip cultivars. The black tulip contained the largest number of anthocyanins, mainly delphinidin derivatives (Dp) and cyanidin derivatives (Cy). The total anthocyanin content (TAC) in the orange, red, and black cultivars was higher than that in the white and yellow cultivars and presented an overall increase trend along with the flower development. TgCHS, TgFLS, TgF3H, TgF3′H, TgF3′5′H, and TgDFR, as key structural genes, were involved in the flavonoid synthesis pathway, and the expression patterns of these genes are basically consistent with the components and accumulation patterns of flavonoids mentioned above. Taken together, the flower color in tulips was closely related to the composition and content of anthocyanins and anthoxanthins, which were indeed regulated by certain key structural genes in the flavonoid pathway.

A cloud computing multi-objective task scheduling optimization based on fuzzy self-defense algorithm is proposed. Select the shortest time, the degree of resource load balance and the cost of multi-objective task completion as the goal of cloud computing multi-objective task scheduling, establish a mathematical model to measure the effect of multi-objective task scheduling, and construct the objective function of cloud computing multi-objective task scheduling. The experimental results show that the cloud computing multi-objective task scheduling optimization method based on fuzzy self-defense algorithm can improve the performance of multi-objective maximum completion time, deadline violation rate and virtual machine resource utilization.

Abstract Virtual screening (VS) based on molecular docking has emerged as one of the mainstream technologies of drug discovery due to its low cost and high efficiency. However, the scoring functions (SFs) implemented in most docking programs are not always accurate enough and how to improve their prediction accuracy is still a big challenge. Here, we propose an integrated platform called ASFP, a web server for the development of customized SFs for structure-based VS. There are three main modules in ASFP: (1) the descriptor generation module that can generate up to 3437 descriptors for the modelling of protein–ligand interactions; (2) the AI-based SF construction module that can establish target-specific SFs based on the pre-generated descriptors through three machine learning (ML) techniques; (3) the online prediction module that provides some well-constructed target-specific SFs for VS and an additional generic SF for binding affinity prediction. Our methodology has been validated on several benchmark datasets. The target-specific SFs can achieve an average ROC AUC of 0.973 towards 32 targets and the generic SF can achieve the Pearson correlation coefficient of 0.81 on the PDBbind version 2016 core set. To sum up, the ASFP server is a powerful tool for structure-based VS.

Kisspeptins are neuropeptides encoded by the kiss1 gene, and little is known about them outside the vertebrate lineage. Two kisspeptin-type neuropeptides (KPs) have been discovered in Apostichopus japonicus (AjK1 and AjK2), an edible sea cucumber, and have been linked to reproductive and metabolic regulation. In this study, we evaluated how KPs affected locomotor behavior in one control group and two treatment groups (AjK1 and AjK2). We discovered that AjK1 had a significant dose effect, primarily by shortening the stride length and duration of movement to reduce the sea cucumber movement distance, whereas AjK2 had little inhibitory effect at the same dose. The levels of phosphatidylethanolamine (PE), phosphatidylcholine (PC), uridine, glycine, and L-serine in the longitudinal muscle of A. japonicus treated with AjK1 differed significantly from those of the control, which may explain the observed changes in locomotor behavior. Treatment with AjK2 induced changes in aspartate levels. Our results imply that AjK1 is more likely than AjK2 to have a role in the regulation of A. japonicus locomotion.

Freesia hybrida is rich in flower colors with beautiful flower shapes and pleasant aroma. Flavonoids are vital to the color formation of its flowers. In this study, five Freesia cultivars with different flower colors were used to study on the level of accumulation of their flavonoids and expression of flavonoid-related genes and further explore new novel transcription factor (TF). Ultra-high-performance liquid chromatography and VION ion mobility quadrupole time-of-flight mass spectrometer (UPLC-Q-TOF-MS) were used to determine the flavonoids. Combined with transcriptome sequencing technology, the molecular mechanism of the flavonoid metabolism difference in Freesia was revealed. A total of 10 anthoxanthin components and 12 anthocyanin components were detected using UPLC-Q-TOF-MS. All six common anthocyanin aglycones in high plants, including cyanidin, delphinidin, petunidin, peonidin, malvidin, and pelargonidin, were detected in Freesia at first time in this study. In orange, yellow, and white cultivars, anthoxanthins gradually decreased with the opening of the petals, while in red and purple cultivars, anthoxanthins first increased and then decreased. No anthocyanin was detected in yellow and white cultivars, while anthocyanins increased with the opening of the petals and reached their maximum at the flowering stage (S3) in other three cultivars. The correlation analysis revealed that the color of Freesia petals was closely related to the composition and content of anthoxanthins and anthocyanins. Petals of five cultivars at S3 were then selected for transcriptome sequencing by using the Illumina Hiseq 4000 platform, and a total of 100,539 unigenes were obtained. There were totally 5,162 differentially expressed genes (DEGs) when the four colored cultivars were compared with the white cultivar at S3. Comparing all DEGs with gene ontology (GO), KEGG, and Pfam databases, it was found that the genes involved in the flavonoid biosynthesis pathway were significantly different. In addition, AP2, WRKY, and bHLH TF families ranked the top three among all differently expressed TFs in all DEGs. Quantitative real-time PCR (qRT-PCR) technology was used to analyze the expression patterns of the structural genes of flavonoid biosynthesis pathway in Freesia. The results showed that metabolic process was affected significantly by structural genes in this pathway, such as CHS1, CHI2, DFR1, ANS1, 3GT1, and FLS1. Cluster analysis was performed by using all annotated WRKY and AP2 TFs and the above structural genes based on their relatively expression. Four novel candidate TFs of WRKY and AP2 family were screened. Their spatiotemporal expression patterns revealed that these four novel TFs may participate in the regulation of the flavonoid biosynthesis, thus controlling its color formation in Freesia petals.

Objectives: To investigate the association between muscle dysmorphia (MD), social physique anxiety, and body-checking behavior in male college students with weight exercise, and to reveal the association between them.Methods: A total of 492 male college students with weight exercise from 18 Fitness Clubs or bodybuilding centers in Chengdu, China, participated in this study.Results: First, the social physique anxiety scores, body checking frequency, and weight exercise behavior (i.e., frequency, time, and intensity) in male college students with MD were significantly higher than those without MD; it indicated that the higher the exercise frequency they had, the longer the exercise time they cost, and the higher exercise intensity carried out, and the higher the social physique anxiety scores tended to be, the higher the frequency of body checking on “global muscles,” “chest and shoulder muscles,” “comparison with others” and “posture measurement” they did. Second, the mediating effect of the social physique anxiety on MD and body checking was established in the “MD → global muscle checking,” “MD → chest and shoulder muscle checking,” “MD → comparison with others,” and “muscle dysmorphia → posture measurement.”Conclusion: Male college students with MD not only have a higher social physique anxiety, but also a higher frequency of body-checking behavior than the ordinary individuals. Social physique anxiety is one of the important mediating factors to those with MD which affects the body-checking behavior.

Tulipa gesneriana has rich cultivars with abundant flower colors; among them, black tulips have become precious cultivars for their rareness in nature. It is of great significance to understand its color formation mechanism for breeding new cultivars with a blackish flower color in the future. In this present study, petals at five developmental stages of Tulipa gesneriana ‘Queen of Night’ (shorted for ‘QN’), a typical black tulip cultivar, were collected to identity the flavonoid composition and compare the accumulation along with the flower development process, aiming to explore the metabolic mechanism of its flower coloration. By using UPLC-Q-TOF-MS, three anthocyanins and nineteen anthoxanthins (including eighteen flavonols and one flavone) were detected in the petals of ‘QN’. The anthocyanins were identified as delphinidin 3-o-rutinoside, cyanidin 3-o-rutinoside and pelargonidin 3-o-rutinoside, respectively. The main flavonols were identified as quercetin, kaempferol, isorhamnetin, naringin and their glycosides. The only one flavone substance was identified as an apigenin derivative. By comparing the content, anthocyanins were the most abundant substance in the petals of ‘QN’, and showed obvious regularity in the development process. With the flower opening, the anthocyanin content accumulated continuously, and reached the highest level at 3575.9 μg·g−1·FW at S5, accounting for 80% of the total flavonoids. Among them, delphinidin 3-o-rutinoside and cyanidin 3-o-rutinoside made the main contribution to the coloration of ‘QN’. The content of anthoxanthins increased first and then decreased, reaching the highest to 1114.8 μg·g−1·FW at S3. Quercetin and its glycosides were the most important flavonol substances in the petals of ‘QN’, accounting for more than 60% of the total anthoxanthins in the five stages. The content of a flavone identified was extremely low, suggesting a limited role in the flower color of ‘QN’. Taken together, the flower color presentation of ‘QN’ was closely related to the composition and accumulation of anthocyanins and flavonols; anthocyanins were the main substances that determine the petal coloration of ‘QN’, and the high content of flavonols played a role of co-pigmentation with these anthocyanins.

Neuropeptides are endogenous active substances that are present in nervous tissues and participate in behavioral and physiological processes of the animal system. Locomotor behavior is basic to predation, escape, reproduction in animals, and neuropeptides play an important role in locomotion. In this study, the function of pedal peptide-type neuropeptide (PDP) in the process of locomotor behavior of the sea cucumber Apostichopus japonicus was evaluated. The locomotor behavior of A. japonicus was recorded by infrared camera before and after PDP administration, and muscle physiology was studied by ultra performance liquid chromatography and quadrupole time-off-light mass spectrometry (UPLC-Q-TOF-MS) to clarify the potential physiological mechanisms. The results showed that PDP enhanced the cumulative duration of moving significantly at the 7th h after injection, and reduced the mean and maximum velocity by 16.90 and 14.22% in A. japonicus. The data of muscle metabolomics suggested that some significantly changed metabolites were related to locomotor behavior of sea cucumbers. The decreases of phosphatidylethanolamine (PE) and phosphatidylcholine (PC) might result in the increases of lysophosphatidylcholines (lysoPC) and lysophosphatidylethanolamine (lysoPE), and suggested the change of fluidity and permeability in the muscle cell membrane, which would affect the physiology and function of muscle cells, and finally alter the locomotor behavior. In addition, the increased level of arachidonic acid (ARA) might activate K+ ion channels and then affect the signaling of muscle cells, or promote the sensitivity of muscle cells to Ca2+ and then result in the contractility of longitudinal muscles in sea cucumbers. ARA was also involved in the linoleic acid metabolism which was the only pathway that disturbed significantly after PDP administration. In conclusion, PDP participated in the regulation of locomotor behavior in the sea cucumber, and the decreased PE and PC, increased lysoPC, lysoPE and ARA might be the potential physiological mechanisms that responsible for behavioral effects of PDP in A. japonicus.

The Qinghai-Tibet Plateau sensu lato (QTP s.l.) harbors an exceptionally high biodiversity, especially at its southeastern margin: this area encompasses the Hengduan Mountains and the eastern Himalayas, which have been listed as biodiversity hotspots. To the contrary, the plateau interior (namely the Qinghai-Tibet Plateau sensu stricto, QTP s.s.) is relatively species-poor because of its particularly harsh climate. With contrasting geological histories and environmental conditions of the Hengduan Mountains, the Himalayas, and the QTP s.s., it would be expected that floristic compositions and diversity patterns of these three regions would differ between each other. To compare the floristic diversity of these three regions, we assembled data on seed plant's distribution in the three regions based on county-level mapping from published monographs and online databases, and we then analyzed their floristic features and species diversity patterns (horizontal and elevational). We found that the Hengduan Mountains hosted the most seed plant species (8,439), as expected. The highest percentage of shrub (22.88%) and tree species (9.80%) were in the Himalayas, whereas herbaceous species (81.50%) were relatively more prominent in the QTP s.s. The Hengduan Mountains also had the most species-rich genera (10) with more than 50% of their total species diversity in China. Also, temperate genera dominated across these three regions, with a highest percentage (77.61%) within the QTP s.s. Across the QTP s.l., species diversity gradually decreased from the southeastern part to the northwest, and most of seed plants were distributed in the southern and eastern margin of the Hengduan Mountains and East Himalayas. Along elevational gradients, species richness all demonstrated a hump-shape curve, but the most species-rich elevation zone differed for each type of life-form across the three regions. Our study sets a base for exploring the origin and evolution of mountain taxa, as well as provides a snapshot of the current plant distribution, which will certainly be modified by climate change.

A small, novel wheat drying oven was designed, and tested in order to validate the possibility of recovering diesel engine waste heat as thermal energy. Prior to experiments, the theoretical probability was conducted, and it was confirmed that the thermal energy required for drying wheat could be satisfied by reusing the diesel engine waste heat. Three different experiment parameters, including air temperature, baffle angle, and drying time, were controlled to optimize the results. Based on the experiment data, the optimum temperature for drying wheat was 55°C, and the optimum baffle angle was 23.25°. The drying efficiency decreased as the drying temperature and drying time increased. The moisture content of wheat decreased from 26.0% to 20.0% with 55°C hot air when the proposed wheat drying oven was used. The test results demonstrated that the proposed oven could be made to work.
