Abstract Traffic sign detection is critical for autonomous driving technology. However, accurately detecting traffic signs in complex traffic environments remains challenge despite the widespread use of one‐stage detection algorithms known for their real‐time processing capabilities. In this paper, the authors propose a traffic sign detection method based on YOLO v8. Specifically, this study introduces the Space‐to‐Depth (SPD) module to address missed detections caused by multi‐scale variations of traffic signs in traffic scenes. The SPD module compresses spatial information into depth channels, expanding the receptive field and enhancing the detection capabilities for objects of varying sizes. Furthermore, to address missed detections caused by complex backgrounds such as trees, this paper employs the Select Kernel attention mechanism. This mechanism enables the model to dynamically adjust its focus and more effectively concentrate on key features. Additionally, considering the uneven distribution of training data, the authors adopted the WIoUv3 loss function, which optimizes loss calculation through a weighted approach, thereby improving the model's detection performance across various sizes and frequencies of instances. The proposed methods were validated on the CCTSDB and TT100K datasets. Experimental results demonstrate that the authors’ method achieves substantial improvements of 3.2% and 5.1% on the mAP50 metric compared to YOLOv8s, while maintaining high detection speed, significantly enhancing the overall performance of the detection system. Abstract The limited of texture details information in low‐resolution facial or eye images presents a challenge for gaze estimation. To address this, FSKT‐GE (feature maps similarity knowledge transfer for low‐resolution gaze estimation) is proposed, a gaze estimation framework consisting of both a high resolution (HR) network and low resolution (LR) network with the identical structure. Rather than mere feature imitation, this issue is addressed by assessing the cosine similarity of feature layers, emphasizing the distribution similarity between the HR and LR networks. This enables the LR network to acquire richer knowledge. This framework utilizes a combination loss function, incorporating cosine similarity measurement, soft loss based on probability distribution difference and gaze direction output, along with a hard loss from the LR network output layer. This approach on low‐resolution datasets derived from Gaze360 and RT‐Gene datasets is validated, demonstrating excellent performance in low‐resolution gaze estimation. Evaluations on low‐resolution images obtained through 2×, 4×, and 8× down‐sampling are conducted on two datasets. On the Gaze360 dataset, the lowest mean angular errors of 10.97°, 11.22°, and 13.61° were achieved, while on the RT‐Gene dataset, the lowest mean angular errors of 6.73°, 6.83°, and 7.75° were obtained.

Objective: Inflammation plays an important role in the transformation of pulmonary nodules (PNs) from benign to malignant. Prediction of benignancy and malignancy of PNs is still lacking efficacy methods. Although Mayo or Brock model have been widely applied in clinical practices, their application conditions are limited. This study aims to construct a diagnostic model of PNs by machine learning using inflammation-related biological markers (IRBMs). Methods: Inflammatory related genes (IRGs) were first extracted from GSE135304 chip data. Then, differentially expressed genes (DEGs) and infiltrating immune cells were screened between malignant pulmonary nodules (MN) and benign pulmonary nodule (BN). Correlation analysis was performed on DEGs and infiltrating immune cells. Molecular modules of IRGs were identified through Consistency cluster analysis. Subsequently, IRBMs in IRGs modules were filtered through Weighted gene co-expression network analysis (WGCNA). An optimal diagnostic model was established using machine learning methods. Finally, external dataset GSE108375 was used to verify this result. Results: 4 hub IRGs and 3 immune cells showed significantly difference between MN and BN, C1 and C2 module, namely PRTN3, ELANE, NFKB1 and CTLA4, T cells CD4 naïve, NK cells activated and Monocytes. IRBMs were screened from black module and yellowgreen module through WGCNA analysis. The Support vector machines (SVM) was identified as the optimal model with the Area Under Curve (AUC) was 0.753. A nomogram was established based on 5 hub IRBMs, namely HS.137078, KLC3, C13ORF15, STOM and KCTD13. Finally, external dataset GSE108375 verified this result, with the AUC was 0.718. Conclusion: SVM model established by 5 hub IRBMs was able to effectively identify MN or BN. Accumulating inflammation and immune dysfunction were important to the transformation from BN to MN.

Background: Stroke often damages the basal ganglia, leading to atypical and transient aphasia, indicating that post-stroke basal ganglia aphasia (PSBGA) may be related to different anatomical structural damage and functional remodeling rehabilitation mechanisms. The basal ganglia contain dense white matter tracts (WMTs). Hence, damage to the functional tract may be an essential anatomical structural basis for the development of PSBGA. Methods: We first analyzed the clinical characteristics of PSBGA in 28 patients and 15 healthy controls (HCs) using the Western Aphasia Battery and neuropsychological test batteries. Moreover, we investigated white matter injury during the acute stage using diffusion magnetic resonance imaging scans for differential tractography. Finally, we used multiple regression models in correlation tractography to analyze the relationship between various language functions and quantitative anisotropy (QA) of WMTs. Results: Compared with HCs, patients with PSBGA showed lower scores for fluency, comprehension (auditory word recognition and sequential commands), naming (object naming and word fluency), reading comprehension of sentences, Mini-Mental State Examination, and Montreal Cognitive Assessment, along with increased scores in Hamilton Anxiety Scale-17 and Hamilton Depression Scale-17 within 7 days after stroke onset (P < 0.05). Differential tractography revealed that patients with PSBGA had damaged fibers, including in the body fibers of the corpus callosum, left cingulum bundles, left parietal aslant tracts, bilateral superior longitudinal fasciculus II, bilateral thalamic radiation tracts, left fornix, corpus callosum tapetum, and forceps major, compared with HCs (FDR < 0.02). Correlation tractography highlighted that better comprehension was correlated with a higher QA of the left inferior fronto-occipital fasciculus (IFOF), corpus callosum forceps minor, and left extreme capsule (FDR < 0.0083). Naming was positively associated with the QA of the left IFOF, forceps minor, left arcuate fasciculus, and uncinate fasciculus (UF) (FDR < 0.0083). Word fluency of naming was also positively associated with the QA of the forceps minor, left IFOF, and thalamic radiation tracts (FDR < 0.0083). Furthermore, reading was positively correlated with the QA of the forceps minor, left IFOF, and UF (FDR < 0.0083). Conclusion: PSBGA is primarily characterized by significantly impaired word fluency of naming and preserved repetition abilities, as well as emotional and cognitive dysfunction. Damaged limbic pathways, dorsally located tracts in the left hemisphere, and left basal ganglia pathways are involved in PSBGA pathogenesis. The results of connectometry analysis further refine the current functional localization model of higher-order neural networks associated with language functions.

The optimal load capacity provides sustainable production and high quality in 'Fuji' apple production. Determining optimal crop loads with simple and affordable thinning methods is critical for apple. We set different load capacities on different apple tree rootstocks and evaluated their effects on tree development, fruit yield and quality. The optimal load capacity for vigorous stock, dwarfing rootstock, and dwarfing interstock were 240, 90, and 100 for apples above 80 mm in diameter, respectively. To explore the optimal strategies of diverse fruit thinning agents (carbaryl, 6-BA, NAA, and Metamitron) used in the 'Fuji' apple tree, we assess from thinning agents' types, varying spraying concentrations, and application time in the most widely used dwarfing interstock. The results showed that the best use of fruit thinning agents is 2,000 mg/L carbaryl at 10 d after flowering could significantly reduce fruit set rate and improve the fruit quality in the dwarfing interstock. During the research, only the fruit thinner NAA treatment suppresses fruit development. Thus, we performed the transcriptome analysis on the NAA-treated and control fruits at 60 (FS1), 90 (FS2), 120 (FS3), and 150 (FS4) days after flowering stages to investigate the potential transcriptional regulations of NAA on fruit development and ripening. Transcriptome results showed that genes related to fruit expansion (expansin A15, expansin B3), phytohormone-related genes (HVA22C, PRE1, AHP1, etc.), fruit coloring-related genes (PAL), and many ripening-related transcription factors (EIN3, ERF, ARF, etc.) might be regulated by exogenous NAA. Our results provide an important reference for the sustainable production and optimal use of fruit thinning agents.

Commensal microbial–host interaction is crucial for host metabolism, growth, development, and immunity. However, research on microbial–host immunity in large animal models has been limited. This study was conducted to investigate the effects of the commensal microbiota on immune function in two model groups: germ-free (GF) and specific-pathogen-free (SPF) piglets. The weight and organ index of the spleen of the GF piglet were larger than those in the SPF piglet (P < 0.05). The histological structure of the red pulp area and mean area of germinal centers were larger in the SPF piglet than in the GF piglet (P < 0.05), whereas the areas of staining of B cells and T cells in the spleen and mesenteric lymph nodes (MLNs) were lower in the GF piglet (P < 0.05). We identified immune-related genes in the spleen and MLNs using RNA sequencing, and used real-time quantitative PCR to analyze the expression of core genes identified in gene set enrichment analysis. The expression levels of genes in the transforming growth factor-β/SMAD3 signaling pathway, Toll-like receptor 2/MyD88/nuclear factor-κB signaling pathway, and pro-inflammatory factor genes IL-6 and TNF-α in the spleen and MLNs were higher in the SPF piglet and in splenic lymphocytes compared with those in the GF and control group, respectively, under treatment with acetic acid, propionic acid, butyric acid, lipopolysaccharide (LPS), or concanavalin A (ConA). The abundances of plasma cells, CD8++ T cells, follicular helper T cells, and resting natural killer cells in the spleen and MLNs were significantly greater in the SPF piglet than in the GF piglet (P < 0.05). In conclusion, the commensal microbiota influenced the immune tissue structure, abundances of immune cells, and expression of immune-related pathways, indicating the importance of the commensal microbiota for spleen and MLNs development and function. In our study, GF piglet was used as the research model, eliminating the interference of microbiota in the experiment, and providing a suitable and efficient large animal research model for exploring the mechanism of “microbial-host” interactions.

The flower coloration of Brassica crops possesses significant application and economic value, making it a research hotspot in the field of genetics and breeding. In recent years, great progress has been made in the research on color variation and creation of Brassica crops. However, the underlying molecular mechanisms and evolutional processes of flower colors are poorly understood. In this paper, we present a comprehensive overview of the mechanism of flower color formation in plants, emphasizing the molecular basis and regulation mechanism of flavonoids and carotenoids. By summarizing the recent advances on the genetic mechanism of flower color formation and regulation in Brassica crops, it is clearly found that carotenoids and anthocyanins are major pigments for flower color diversity of Brassica crops. Meantime, we also explore the relationship between the emergence of white flowers and the genetic evolution of Brassica chromosomes, and analyze the innovation and multiple utilization of Brassica crops with colorful flowers. This review aims to provide theoretical support for genetic improvements in flower color, enhancing the economic value and aesthetic appeal of Brassica crops.

Pigs are widely used as animal models in various studies related to humans. The interaction between the gut microbiota and the host has significant effects on the host’s health and disease status. However, although there have been many studies investigating the pig gut microbiota, the findings have been inconsistent due to variations in rearing conditions. Interactions between the gut microbiota and host have not been fully explored in pigs. Specific pathogen-free (SPF) pigs are ideal non-primate large animals to study the interactions between the gut microbiota and the host. In this study, we performed high-throughput sequencing analysis of the gut microbiota and the gut tissue transcriptome of six SPF pigs to provide a systematic understanding of the composition, function, and spatial distribution of gut microbiota in SPF pigs. We identified significant differences in microbial diversity and functionality among different gastrointestinal tract sites. Metagenomics data analysis revealed significant differences in alpha diversity and beta diversity of microbiota in different gastrointestinal sites of SPF pigs. Additionally, transcriptomic data indicated significant differences in gene expression as well as KEGG and GO functional enrichment between the small intestine and large intestine. Furthermore, by combining microbial metagenomics and host transcriptomics analyses, specific correlations were found between gut microbiota and host genes. These included a negative correlation between the TCN1 gene and Prevotella dentalis, possibly related to bacterial metabolic pathways involving vitamin B12, and a positive correlation between the BDH1 gene and Roseburia hominis, possibly because both are involved in fatty acid metabolism. These findings lay the groundwork for further exploration of the co-evolution between the microbiota and the host, specifically in relation to nutrition, metabolism, and immunity. In conclusion, we have elucidated the diversity of the gut microbiota in SPF pigs and conducted a detailed investigation into the interactions between the gut microbiota and host gene expression. These results contribute to our understanding of the intricate dynamics between the gut microbiota and the host, offering important references for advancements in life science research, bioproduct production, and sustainable development in animal husbandry.

Background and purposeEarly blood–brain barrier (BBB) disruption in patients with acute ischemic stroke (AIS) can be detected on perfusion computed tomography (PCT) images before undergoing reperfusion therapy. In this study, we aimed to determine whether early disruption of the BBB predicts intracranial hemorrhage transformation (HT) in patients with AIS undergoing endovascular therapy and further identify factors influencing BBB disruption.MethodsWe retrospectively analyzed general clinical and imaging data derived from 159 consecutive patients with acute anterior circulation stroke who were admitted to the Department of Neurology of the First Hospital of Jilin University, and who underwent endovascular treatment between January 1, 2021, and March 31, 2023. We evaluated the relationship between BBB destruction and intracranial HT before endovascular reperfusion therapy and examined the risk factors for early BBB destruction.ResultsA total of 159 patients with assessable BBB leakage were included. The median (interquartile range, IQR) age was 63 (54–70) years, 108 (67.9%) patients were male, and the median baseline National Institutes of Health Stroke Scale (NHISS) score was 12 (10–15). Follow-up non-contrast computed tomography (NCCT) detected HT in 63 patients. After logistic regression modeling adjustment, we found that BBB leakage in the true leakage area was slightly more than 2-fold risk of HT (odds ratio [OR], 2.01; 95% confidence interval [CI] 1.02–3.92). Heart rate was also associated with HT (OR, 1.03, 95% CI, 1.00–1.05). High Blood–brain barrier permeability (BBBP) in the true leakage area was positively correlated with infarct core volume (OR, 1.03; 95% CI, 1.01–1.05).ConclusionEarly BBB destruction before endovascular reperfusion therapy was associated with HT, whereas high BBBP correlated positively with infarct core volume.

Subway station fires frequently result in massive casualties, economic losses and even social panic due to the massive passenger flow, semiconfined space and limited conditions for escape and smoke emissions. The combination of different states of fire hazard factors increases the uncertainty and complexity of the evolution path of subway station fires and causes difficulty in assessing fire risk. Traditional methods cannot describe the development process of subway station fires, and thus, cannot assess fire risk under different fire scenarios. To realise scenario-based fire risk assessment, the elements that correspond to each scenario state during fire development in subway stations are identified in this study to explore the intrinsic driving force of fire evolution. Accordingly, a fire scenario evolution model of subway stations is constructed. Then, a Bayesian network is adopted to construct a scenario evolution probability calculation model for calculating the occurrence probability of each scenario state during subway station fire development and identifying critical scenario elements that promote fire evolution. Xi’an subway station system is used as a case to illustrate the application of Bayesian network-based scenario evolution model, providing a practical management tool for fire safety managers. The method adopted in this study enables managers to predict fire risk in each scenario and understand the evolution path of subway station fire, supporting the establishment of fire response strategies based on “scenario–response” planning.

The global hospitality industry is fast-turning sustainable and environmentally friendly. Behaviour-driven energy conservation is an emerging green hotel operation strategy to support this change. The long-stay accommodation services have gained momentum in the hospitality sector since the COVID-19 pandemic. However, the characteristics of long-stay hotel guests are often overlooked in sustainable interventions. Based on an empirical survey in China, this study aims to explore the factors driving energy-saving behaviours of long-stay hotel guests and to compare their effects on guests for different visiting purposes (leisure, business, and extended-stay resident). The analysis indicates that attitude, personal norm and place attachment present a direct contribution to energy-saving behaviour. Besides, the results support that attitude and personal norm connect environmental values and energy-saving behaviour. Both altruistic and biospheric values have positive effects, while egoistic values seem to play a negative role. Biospheric values have stronger impact on attitude and personal norm of business guests. Place attachment has a stronger influence on extended-stay residents while its contribution to energy-saving behaviours of business guests is smaller than other guests. Besides, leisure guests are more sensitive to moral obligations. This research sheds novel lights on the psychological perspectives of the observed heterogeneity of energy-saving behaviours of hotel guests with different visiting purposes. The findings provide hotel operators with a novel theoretical reference for targeted energy-saving interventions to promote energy-saving actions of long-term hotel guests. The study, therefore, can contribute to sustainable tourism policymaking and behaviour-driven hotel energy management.

Abstract Introduction There are only four sizes of implantable collamer lens (ICL) available for selection, which cannot completely fit all patients as a result of the discontinuity of ICL sizes. Sizing an optimal ICL and predicting postoperative vault are still unresolved problems. This study aimed to develop and validate innovative data-level data-balancing machine learning-based models for predicting ICL size and postoperative vault. Methods The patients were randomly assigned to training and internal validation sets in a 4:1 ratio. Feature selection was performed using analysis of variance (ANOVA) and Kruskal–Wallis feature importance methods. Traditional linear regression model and machine learning-based models were used. The accuracy of models was assessed using the area under the curve (AUC) and confusion matrix. Results A total of 564 patients (1127 eyes) were eligible for this study, consisting of 808 eyes in the training set, 202 eyes in the internal validation set, and 117 eyes in the external validation set. Compared with the traditional linear regression method, the machine learning model bagging tree showed the best performance for ICL size selection, with an accuracy of 84.5% (95% confidence interval (CI) 83.2–85.8%), and the AUC ranged from 0.88 to 0.99; the prediction accuracy of 12.1 mm and 13.7 mm ICL sizes was improved by 49% and 59%, respectively. The bagging tree model achieved the best accuracy [90.2%, (95% CI 88.9–91.5%)] for predicting the postoperative vault, and the AUC ranged from 0.90 to 0.94. The prediction accuracies of internal and external validation dataset for ICL sizing were 82.2% (95% CI 81.1–83.3%) and 82.1% (95% CI 81.1–83.1%), respectively. Conclusions The innovative data-level data balancing-based machine learning model can be used to predict ICL size and postoperative vault more accurately, which can assist surgeons in choosing optimal ICL size, thus reducing risks of postoperative complications and secondary surgery.

Abstract Background To investigate the control effect on the axial length elongation using corneal refractive therapy (CRT) with different optical zone diameters (BOZDs). We also sought to compare the difference in higher-order aberrations (HOAs), treatment zone (TZ) size and Zernike defocus coefficient with different BOZDs and seek the optimal parameter for predicting axial elongation. Methods This prospective cohort study included 7- to 14-year-olds fitted with orthokeratology (ortho-K) lenses of 5-mm (5-mm group) or 6-mm BOZD (6-mm group). Axial length (AL), corneal topography, HOAs and Zernike defocus coefficient were obtained at baseline, and 1, 3, 6, 9 and 12 months follow-up visits. Multivariate regression analyses were used to explore the association between AL change and ocular biometric parameters. Receiver operating characteristic (ROC) curve analysis was used to determine the best diagnostic value for AL change in ocular biometric parameters. Results In total, 301 participants completed the one-year follow-up. The mean AL change in the 5-mm group (0.13 ± 0.18 mm) was less than that in the 6-mm group (0.27 ± 0.15 mm) at the 12 months visit. DNA methylation plays an important role in the development and tissue differentiation of eukaryotes. In this study, bisulfite sequencing (BS-seq) technology was used to analyze the DNA methylation profiles of liver tissues taken from Rongchang pigs at three postnatal feeding stages, including newborn, suckling, and adult. The DNA methylation pattern across the genomes or genic region showed little difference between the three stages. We observed 419 differentially methylated regions (DMRs) in promoters, corresponding to 323 genes between newborn and suckling stages, in addition to 288 DMRs, corresponding to 134 genes, between suckling and adult stages and 351 DMRs, corresponding to 293 genes, between newborn and adult stages. These genes with DMRs were mainly enriched in metabolic, immune-related functional processes. Correlation analysis showed that the methylation level of gene promoters was significantly negatively correlated with gene expression. Further, we found that genes related to nutritional metabolism, e.g., carbohydrate metabolism (FAHD1 and GUSB) or fatty acid metabolism (LPIN1 and ACOX2), lost DNA methylation in their promoter, with mRNA expression increased in newborn pigs compared with those in the suckling stage. A few fatty acid metabolism-related genes (SLC27A5, ACOX2) were hypomethylated and highly expressed in the newborn stage, which might satisfy the nutritional requirements of Rongchang pigs with high neonatal birth rates. In the adult stage, HMGCS2—which is related to fatty acid β-oxidation—was hypomethylated and highly expressed, which explains that the characteristics of high energy utilization in adult Rongchang pigs and their immune-related genes (CD68, STAT2) may be related to the establishment of liver immunity. This study provides a comprehensive analysis of genome-wide DNA methylation patterns in pig liver postnatal development and growth. Our findings will serve as a valuable resource in hepatic metabolic studies and the agricultural food industry.

Background: Angiogenesis is essential for various physiological and pathological processes, such as embryonic development and cancer cell proliferation, migration, and invasion. Long noncoding RNAs (lncRNAs) play pivotal roles in normal homeostasis and disease processes by regulating gene expression through various mechanisms, including competing endogenous RNAs (ceRNAs) of target microRNAs (miRNAs). The lncRNA MYU is known to promote prostate cancer proliferation via the miR-184/c-Myc regulatory axis and to be upregulated in vascular endothelial cells under hypoxic conditions, which often occurs in solid tumors. In the present study, we investigated whether MYU might affect cancer growth by regulating angiogenesis in vascular endothelial cells under hypoxia. Methods: The expression of MYU-regulated miR-23a-3p and interleukin-8 (IL-8) in HUVEC cell lines was examined using qRT-PCR. The CCK-8 assay, EdU assay, wound-healing assay, and tube-formation assay were used to assess the effects of MYU on cell proliferation, migration, and tube formation of HUVEC cells in vitro. The dual-luciferase reporter assay was performed to examine the effects of miR-23a-3p on MYU and IL-8 expression. Results: We found that the overexpression of MYU and knockdown of miR-23a-3p in human umbilical vein endothelial cells (HUVECs) under hypoxia promoted cell proliferation, migration, and tube formation. Mechanistically, MYU was shown to bind competitively to miR-23a-3p, thereby preventing miR-23a-3p binding to the 3′ untranslated region of IL-8 mRNA. In turn, increased production of pro-angiogenic IL-8 promoted HUVEC proliferation, migration, and tube formation under hypoxia. Conclusion: This study identified a new role for lncRNA MYU as a ceRNA for miR-23a-3p and uncovered a novel MYU–miR-23a-3p–IL-8 regulatory axis for angiogenesis. MYU and/or miR-23a-3p may thus represent new targets for the treatment of hypoxia-related diseases by promoting angiogenesis.

Abstract. Esophageal cancer (EC) is one of the most common aggressive malignant tumors in the digestive system with a severe epidemiological situation and poor prognosis. The early diagnostic rate of EC is low, and most EC patients are diagnosed at an advanced stage. Multiple multimodality treatments have gradually evolved into the main treatment for advanced EC, including surgery, chemotherapy, radiotherapy, targeted therapy, and immunotherapy. And the emergence of targeted therapy and immunotherapy has greatly improved the survival of EC patients. This review highlights the latest advances in targeted therapy and immunotherapy for EC, discusses the efficacy and safety of relevant drugs, summarizes related important clinical trials, and tries to provide references for therapeutic strategy of EC.

This study presents an in-depth investigation into the intersection of Building Information Modeling (BIM) and green building practices within China’s rapidly evolving construction industry. As China intensifies its efforts to mitigate environmental impacts and promote sustainable growth, the integration of BIM into green building practices emerges as a crucial area of study. A qualitative research method was adopted in this research. In the first step, utilizing semi-structured interviews with a diverse array of industry professionals, this research provides nuanced insights into the current state and prospects of BIM in the green building landscape. Secondly, thematic analysis is used to formalize the views and points from interviewees. Finally, a novel conceptual framework is proposed, addressing these challenges through technological innovation, supportive policies, cultural and educational reform, economic incentives, and collaborative dynamics. This study contributes a systematic approach to amalgamate BIM with sustainable construction, offering insights for industry professionals and policymakers to promote environmentally conscious building practices in China.

The normal growth and development of skeletal muscle is essential for the health of the body. The regulation of skeletal muscle by intestinal microorganisms and their metabolites has been continuously demonstrated. Acetate is the predominant short-chain fatty acids synthesized by gut microbiota through the fermentation of dietary fiber; however, the underlying molecular mechanisms governing the interaction between acetate and skeletal muscle during the rapid growth stage remains to be further elucidated. Herein, specific pathogen-free (SPF) mice, germ-free (GF) mice, and germ-free mice supplemented with sodium acetate (GS) were used to evaluate the effects of acetate on the skeletal muscle growth and development of young mice with gut microbiota deficiency. We found that the concentration of serum acetate, body mass gain, succinate dehydrogenase activity, and expression of the myogenesis maker gene of skeletal muscle in the GS group were higher than those in the GF group, following sodium acetate supplementation. Furthermore, the transcriptome analysis revealed that acetate activated the biological processes that regulate skeletal muscle growth and development in the GF group, which are otherwise inhibited due to a gut microbiota deficiency. The in vitro experiment showed that acetate up-regulated Gm16062 to promote skeletal muscle cell differentiation. Overall, our findings proved that acetate promotes skeletal muscle growth and development in young mice via increasing Gm16062 expression.

Abstract Using an adult female miniature pig model with diet-induced weight gain/weight loss, we investigated the regulatory mechanisms of three-dimensional (3D) genome architecture in adipose tissues (ATs) associated with obesity. We generated 249 high-resolution in situ Hi-C chromatin contact maps of subcutaneous AT and three visceral ATs, analyzing transcriptomic and chromatin architectural changes under different nutritional treatments. We find that chromatin architecture remodeling underpins transcriptomic divergence in ATs, potentially linked to metabolic risks in obesity development. Analysis of chromatin architecture among subcutaneous ATs of different mammals suggests the presence of transcriptional regulatory divergence that could explain phenotypic, physiological, and functional differences in ATs. Regulatory element conservation analysis in pigs and humans reveals similarities in the regulatory circuitry of genes responsible for the obesity phenotype and identified non-conserved elements in species-specific gene sets that underpin AT specialization. This work provides a data-rich tool for discovering obesity-related regulatory elements in humans and pigs.

Abstract Background Ocular trauma is a leading cause of vision loss. Penetrating ocular injury is a major type of open globe injury(OGI), while its epidemiology and clinical characteristics are still uncertain. The aim of this study is to reveal the prevalence and prognostic factors of penetrating ocular injury in the Shandong province. Methods A retrospective study of penetrating ocular injury was performed at the Second Hospital of Shandong University, from January 2010 to December 2019. Demographic information, injury causes, ocular trauma types, and initial and final visual acuity(VA) were analyzed. To obtain more precise characteristics of penetrating injury, the eye global was divided into three zones and analyzed. Results Among 210 OGI, there are 83 penetrating injuries which account for 39.5% of all. In addition, the final VA of 59 penetrating injuries recovered to 0.1 or better, which possesses the highest frequency among OGI. In order to research the relationship between the wound location and the final VA, we took 74 cases of penetrating injuries without retina or optic nerve damage for analysis. Results show that 62 were male and 12 were female. The average age was 36.01 ± 14.15. The most frequent occupation is the worker followed by the peasant. Abstract Background Clinical tissue adhesives remain some critical drawbacks for managing emergency injuries, such as inadequate adhesive strength and insufficient anti-infection ability. Herein, a novel, self-healing, and antibacterial carboxymethyl chitosan/polyaldehyde dextran (CMCS/PD) hydrogel is designed as the first-aid tissue adhesive for effective trauma emergency management. Methods We examined the gel-forming time, porosity, self-healing, antibacterial properties, cytotoxicity, adhesive strength, and hemocompatibility. Liver hemorrhage, tail severance, and skin wound infection models of rats are constructed in vivo, respectively. Results Results demonstrate that the CMCS/PD hydrogel has the rapid gel-forming (~ 5 s), good self-healing, and effective antibacterial abilities, and could adhere to tissue firmly (adhesive strength of ~ 10 kPa and burst pressure of 327.5 mmHg) with excellent hemocompatibility and cytocompatibility. This suggests the great prospect of CMCS/PD hydrogel in acting as a first-aid tissue adhesive for trauma emergency management. The CMCS/PD hydrogel is observed to not only achieve rapid hemostasis for curing liver hemorrhage and tail severance in comparison to commercial hemostatic gel (Surgiflo ®) but also exhibit superior anti-infection for treating acute skin trauma compared with clinical disinfectant gel (Prontosan ®). Conclusions Overall, the CMCS/PD hydrogel offers a promising candidate for first-aid tissue adhesives to manage the trauma emergency. Because of the rapid gel-forming time, it could also be applied as a liquid first-aid bandage for mini-invasive surgical treatment. Graphical Abstract

Abstract Heat stress, which is caused by global warming, threatens crops yield and quality across the world. As a kind of post-translation modification, SUMOylation involves in plants heat stress response with a rapid and wide pattern. Here, we identified small ubiquitin modifiers (SUMO), which affect drought tolerance in apple, also participated in thermotolerance. Six isoforms of SUMOs located on six chromosomes in apple genome, and all the SUMOs were up-regulated in response to heat stress condition. The MdSUMO2 RNAi transgenic apple plants exhibited higher survival rate, lower ion leakage, higher catalase (CAT) activity, and Malondialdehyde (MDA) content under heat stress. MdDREB2A, the substrate of MdSUMO2 in apple, was accumulated in MdSUMO2 RNAi transgenic plants than the wild type GL-3 at the protein level in response to heat stress treatment. Further, the inhibited SUMOylation level of MdDREB2A in MdSUMO2 RNAi plants might repress its ubiquitination, too. The accumulated MdDREB2A in MdSUMO2 RNAi plants further induced heat-responsive genes expression to strengthen plants thermotolerance, including MdHSFA3, MdHSP26.5, MdHSP18.2, MdHSP70, MdCYP18-1 and MdTLP1. In summary, these findings illustrate that interfering small ubiquitin modifiers (SUMO) in apple improves plants thermotolerance, partly by facilitating the stability and activity of MdDREB2A.

Abstract Background Increasing evidence shows that tRNA‐derived small RNAs (tsRNAs) are not only by‐products of transfer RNAs, but they participate in numerous cellular metabolic processes. However, the role of tsRNAs in skeletal muscle regeneration remains unknown. Methods Small RNA sequencing revealed the relationship between tsRNAs and skeletal muscle injury. The dynamic expression level of 5'tiRNA‐Gly after muscle injury was confirmed by real‐time quantitative PCR (q‐PCR). Objective This study aims to investigate the associations of axial length to corneal radius of curvature (AL/CRC) ratio with refractive error and to determine the effect of AL/CRC ratio on hyperopia reserve and myopia assessment among Chinese preschoolers.Methods This was a retrospective cross-sectional study that evaluated subjects aged 4–6 years. AL and CRC were obtained using a non-contact ocular biometer. Correlation analysis was performed to explore the associations of AL/CRC ratio with spherical equivalent refractive error (SER). The accuracy of AL/CRC ratio for hyperopia reserve and myopia assessment was analysed using cycloplegic refraction by drawing receiver operating characteristic (ROC) curves.Results The analysis included 1024 participants (537 boys, 52.4%). Abstract Climate change and the resulting natural vegetation succession can alter vegetation productivity. However, the mechanisms underlying future productivity changes under the two influences remain unclear. Here, we used the comprehensive sequence classification system to simulate changes in global potential natural vegetation under different climate scenarios (SSP1‐2.6, SSP2‐4.5, SSP3‐7.0, and SSP5‐8.5), and combined the Carnegie–Ames–Stanford Approach model with random forest to assess the response of net primary productivity (NPP) to climate change and vegetation succession from 2020 to 2100. Except for SSP126, terrestrial NPP in 2100 decreased by 0.86, 2.39, and 2.54 Pg C·a−1 versus 2020 under SSP2‐4.5, SSP5‐8.5, and SSP3‐7.0, respectively. Forest was the primary contributor to terrestrial NPP changes. The total forest area was projected to increase under all scenarios, with SSP2‐4.5 showing the largest increase (358.57 × 104 km2). However, expanding forest regions exhibited a relatively low mean NPP, while stable regions demonstrated a declining pattern. Consequently, forest NPP increased under SSP1‐2.6 but decreased by 4.03, 3.43, and 0.82 Pg C·a−1 in 2100 versus 2020 under SSP5‐8.5, SSP3‐7.0, and SSP2‐4.5, respectively. In comparison, grassland and desert exerted minor influence on terrestrial NPP changes, their total NPP decreased only under the SSP1‐2.6 scenario. The grassland area decreased, but the mean NPP increased, whereas the desert area expanded, resulting in consistent changes in both total and mean NPP. Our results analyzed the effects of climate change and vegetation distribution under its influence on the change of NPP, which can deepen our understanding of their relationship.

ABSTRACT: Proteins are considered major effectors of sperm function. However, the proteins expressed in pigeon sperm have not been explored. Here, we collected semen from meat and racing pigeons using the electroejaculation method and identified proteins in pigeon sperm using the proteomics approach. A total of 1,641 proteins were identified in the sperm of domesticated pigeons. Of which, 1,541 proteins were reliably quantified, and gene ontology (GO) and associated bioinformatics analyses indicated that annotated proteins were linked to the oxidation-reduction process, integral component membrane, and protein binding, etc. Among quantified proteins, 1,515 and 1,507 proteins were respectively presented in White King pigeons and racing pigeons, and 1,481 proteins were shared between these 2 types of pigeons, including axonemal dynein, solute carrier, cilia- and flagella-associated protein, outer dense fiber protein, etc. Proteins in our constructed protein-protein interaction (PPI) network are involved in oxidative phosphorylation, sperm axoneme assembly, cilium-dependent cell motility, axonemal dynein complex assembly, flagellated sperm motility, etc. In conclusion, this study characterized the sperm proteome of pigeons and provided a foundation for the subsequent research screening markers for fertility evaluation of pigeons.

The Arctic sea ice plays a significant role in climate-related processes and has a considerable effect on humans, however accurately predicting the Arctic sea ice concentration is still challenging. Recently, with the rise and development of artificial intelligence, big data technology, machine learning has been widely used in the field of sea ice prediction. In this study, we utilized a sea ice concentration dataset obtained from satellite remote sensing and applied the k-nearest-neighbors (Ice-kNN) machine learning model to forecast the summer Arctic sea ice concentration and extent on 122 days prediction. Based on the physical characteristics of summer sea ice, different algorithms are employed to optimize the prediction model. A drift-ice correction algorithm is designed to address the unrealistic drift ice around the sea ice edge, and a distance function combined with the spatial pattern is proposed to enhance similarity detection. Deseasonalized and detrended sea ice datasets and an expanded training library are also utilized to improve model performance. Furthermore, sensitivity analysis reveals a positive impact of net surface heat flux on sea ice prediction. The modified Ice-kNN model outperforms climatological and anomaly persistence predictions, demonstrating its applicability to predicting summer Arctic sea ice. The September sea ice extent hindcasts of the modified Ice-kNN model are compared to a variety of models submitted to the Sea Ice Prediction Network, underscoring its potential to improve predictive skill for Arctic sea ice.

Abstract In this study, exosomes from cooked meat were extracted by ultra-high-speed centrifugation. Approximately 80% of exosome vesicles were within 20–200 nm. In addition, the surface biomarkers of isolated exosomes were evaluated using flow cytometry. Further studies showed the exosomal microRNA profiles were different among cooked porcine muscle, fat and liver. Cooked pork-derived exosomes were chronically administered to ICR mice by drinking for 80 days. The mice plasma levels of miR-1, miR-133a-3p, miR-206 and miR-99a were increased to varying degrees after drinking exosome enriched water. Furthermore, GTT and ITT results confirmed an abnormal glucose metabolism and insulin resistance in mice. Moreover, the lipid droplets were significantly increased in the mice liver. A transcriptome analysis performed with mice liver samples identified 446 differentially expressed genes (DEGs). Functional enrichment analysis found that DEGs were enriched in metabolic pathways. Overall, the results suggest that microRNAs derived form cooked pork may function as a critical regulator of metabolic disorder in mice.

Abstract Background To investigate ocular biological characteristics for myopic children with axial length (AL) reduction during orthokeratology (Ortho-K) treatment and provide clinical clues for better myopia control effects. Methods Changes in ocular parameters and treatment zone (TZ) in 75 subjects who completed one-year Ortho-K treatment were retrospectively reviewed. The subjects were divided into two groups according to one-year AL change: the AL reduction group (n = 37) and the AL elongation group (n = 38). Univariate and multivariate regression analyses were performed to determine the association between TZ, ocular parameters, and AL change. Results There was no significant difference in baseline between the two groups (all P > 0.05). Nitrogen is one of the most critical macroelements required for apple biomass accumulation and quality formation. Thus, identification and characterization of candidate genes involved in nitrogen deficiency stress is of great importance to the sustainable development of the apple industry. SUMOylation is a kind of post-translational modification, which is widely involved in plant life processes, including responses to nitrogen (N) deficiency. Here, we identified apple small ubiquitin modifiers (SUMO) that participated in responding to nitrogen deficiency. Firstly, all isoforms of apple SUMOs were induced by N deficiency stress. Under N deficiency stress conditions, the MdSUMO2 RNAi transgenic apple plants showed better growth potential than the wild type GL-3, including more new leaves, increased height, thicker stems, and heavier fresh weight. In addition, the MdSUMO2 RNAi transgenic apple plants exhibited darker green leaves and greater root volume than GL-3 in response to N deficient conditions. Finally, the activated glutamine synthetase (GS) activity, nitrate reductase (NR) activity, as well as the induced expression of genes involved in nitrogen assimilation in the root might contribute to the tolerance of MdSUMO2 RNAi transgenic apple plants in response to nitrogen deficiency. Taken together, our finding provides insight to further study the mechanisms underlying the essential roles of apple MdSUMOs in responding to N deficient conditions.

Optimization challenges are becoming more complex as the world advances. Since deterministic and heuristic approaches are no longer sufficient to deal with such complex problems, metaheuristics have recently emerged as a viable option to address optimization difficulties. Since Sand Cat Swarm Optimization (SCSO) is a famous meta-heuristic algorithm, SCSO has a weak ability to balance search between exploration and exploitation and slow convergence, so it may not be effective in finding the global optima, particularly for complex problems. Hence, this paper proposes an intensified SCSO with multiple strategies (IMSCSO). The performance of the IMSCSO algorithm was evaluated on 23 standard test functions and test suites of CEC 2017, CEC 2019, and CEC 2020. Experimental results show that the IMSCSO algorithm performs significantly better than or is on par with other state-of-the-art optimizers. The statistical results obtained from the Wilcoxon signed-rank test and the Friedman test also indicate that the IMSCSO algorithm has a high ability to significantly outperform and rank first among all methods. Moreover, seven typical engineering issues were employed to estimate the efficacy of IMSCSO in optimizing constrained problems. The experimental findings show that the suggested IMSCSO method can efficiently handle real-world application issues.

Objective Pigs, an ideal biomedical model for human diseases, suffer from about 50% early embryonic and fetal death, a major cause of fertility loss worldwide. However, identifying the causal variant remains a huge challenge. This study aimed to detect single nucleotide polymorphisms (SNPs) and candidate genes for the number of mummified (NM) piglets using the imputed whole-genome sequence (WGS) and validate the potential candidate genes. Methods The imputed WGS was introduced from genotyping-by-sequencing (GBS) using a multi-breed reference population. We performed genome-wide association studies (GWAS) for NM piglets at birth from a Landrace pig populatiGWAS peak located on SSC11: 0.10 to 7.11 Mbp (Top SNP, SSC11:1,889,658 bp; p = 9.98E-13) was identified in cyclin dependent kinase on. A total of 300 Landrace pigs were genotyped by GBS. The whole-genome variants were imputed, and 4,252,858 SNPs were obtained. Various molecular experiments were conducted to determine how the genes affected NM in pigs. Results A strong GWAS peak located on SSC11: 0.10 to 7.11 Mbp (Top SNP, SSC11:1,889,658 bp; p = 9.98E-13) was identified in cyclin dependent kinase 8 (CDK8) gene, which plays a crucial role in embryonic retardation and lethality. Based on the molecular experiments, we found that Y-box binding protein 1 (YBX1) was a crucial transcription factor for CDK8, which mediated the effect of CDK8 in the proliferation of porcine ovarian granulosa cells via transforming growth factor beta/small mother against decapentaplegic signaling pathway, and, as a consequence, affected embryo quality, indicating that this pathway may be contributing to mummified fetal in pigs. Conclusion A powerful imputation-based association study was performed to identify genes associated with NM in pigs. CDK8 was suggested as a functional gene for the proliferation of porcine ovarian granulosa cells, but further studies are required to determine causative mutations and the effect of loci on NM in pigs.

In recent years, railway safety accidents have repeatedly occurred. Any omission in the process of management or operation can easily have very serious consequences. This study aimed to examine the causes and transmission mechanisms of safety risks in railway engineering departments. First, the multi-objective particle swarm optimization algorithm was employed to determine the key risk factors, allowing for indicator screening that was in line with the requirements of practical applications. Then, Bayesian networks were used, and their structure was optimized to analyze the propagation diagnosis and probability of key risk indicators, obtaining the causal logic chain that produces accidents and, from that, the four aspects (human, machine, environment, management) of the corresponding prevention of risk recommendations. Finally, in this article, it is shown that combining the indicators and Bayesian networks can improve the accuracy of risk prediction and provide more accurate results than using existing research and, hence, it can fill the gap in research on railway safety risks in risk transmission mechanisms.

Objective This work was to determine coat inheritance and evaluate production performance for crossbred pigs from Berkshire×Chenghua (BC) compared with Chinese indigenous Chenghua (CH) pigs. Methods The coat color phenotypes were recorded for more than 16,000 pigs, and the genotypes of melanocortin 1 receptor (MCIR) gene were identified by sequencing. The reproductive performance of 927 crossbred BC F4 gilts and 320 purebred CH gilts was recorded. Sixty pigs of each breed were randomly selected at approximately 60 days of age to determine growth performance during fattening period, which lasted for 150 days for BC pigs and 240 days for CH pigs. At the end of the fattening period, 30 pigs of each breed were slaughtered to determine carcass composition and meat quality. Results The coat color of BC pigs exhibits a “dominant black” hereditary pattern, and all piglets derived from boars or sows genotyped ED1ED1 homozygous for MC1R gene showed a uniform black coat phenotype. The BC F4 gilts displayed a good reproductive performance, showing a higher litter and tear size and were heavier at farrowing litter and at weaning litter than the CH gilts, but they reached puberty later than the CH gilts. BC F4 pigs exhibited improved growth and carcass characteristics with a higher average daily live weight gain, lower feed-to-gain ratio, and higher carcass lean meat rate than CH pigs. Like CH pigs, BC F4 pigs produced superior meat-quality characteristics, showing ideal pH and meat-color values, high intramuscular fat content and water-holding capacity, and acceptable muscle-fiber parameters. C18:1, C16:0, C18:0, and C18:2 were the main fatty acids in M. longissimus lumborum in the two breeds, and a remarkably high polyunsaturated/saturated fatty acid ratio of ~0.39 was observed in the BC F4 pigs. Conclusion The BC F4 pigs exhibit a uniform black coat pattern and acceptable total production performance.

To study the effect of fortified Zygosaccharomyces rouxii (JL-02) on the quality of soy sauce，salt tolerance characteristic of strain JL-02, pH and neutral proteinase activity of koji, the physicochemical properties and volatile flavor substances of two soy sauces in different brewing processes were analyzed. The results showed that salt tolerance of the strain JL-02 was up to 16%, the pH of koji was 6.85±0.08, and the neutral proteinase activity reached to (2109.2±45.2) U/g. Compared with the control group, the soy sauce treated with fortified Zygosaccharomyces rouxii exhibited the increased total acid content ((1.14±0.01) g/100 mL), decreased reducing sugar content ((1.06±0.01) g/100 mL) and pH value (5.09±0.03). But there was no significant difference in the content of amino nitrogen, and all samples reached the standard of special grade soy sauce. In addition, there were 15, 52 and 57 volatile flavor compounds in the koji, control group, and experimental group, respectively. And the furan flavor substances (2-pentylfuran, 4-methoxy-2,5-dimethyl-3(2H)-furanone and 2,3-dihydrobenzofuran) were increased in the experimental group. The principal component analysis of the volatile flavor compounds in the three groups of samples showed a good result. This research provides a theoretical basis and technical reference for the soy sauce enterprises to carry out the intensified yeast strain brewing soy sauce technology in the future.

Abstract Background Lupus nephritis (LN) is a common and serious complication of systemic lupus erythematosus (SLE). However, the aetiology and pathogenesis of LN remain unknown. 1,25-dihydroxyvitamin D3 [1,25-(OH)2-VitD3] is the active form of vitamin D, and it has been shown to perform important functions in inflammatory and immune-related diseases. In this study, we investigated the time-dependent effects of 1,25-dihydroxyvitamin D3 and explored the underlying mechanism in MRL/lpr mice, a well-studied animal model of LN. Methods Beginning at 8 weeks of age, 24-h urine samples were collected weekly to measure the levels of protein in the urine. We treated female MRL/lpr mice with 1,25-dihydroxyvitamin D3 (4 μg/kg) or 1% DMSO by intraperitoneal injection twice weekly for 3 weeks beginning at the age of 11 weeks. The mice were separately sacrificed, and serum and kidney samples were collected at the ages of 14, 16, 18, and 20 weeks to measure creatinine (Cr) levels, blood urea nitrogen (BUN) levels, histological damage, immunological marker (A-ds DNA, C1q, C3, IgG, IgM) levels, and inflammatory factor (TNF-α, IL-17, MCP-1) levels. Furthermore, the nuclear factor kappa B (NF-κB) and the mitogen-activated protein kinase (MAPK) signalling pathways were also assessed to elucidate the underlying mechanism. Different anatomical locations of the body skin show differences in their gene expression patterns depending on different origins, and the inherent heterogeneous information can be maintained in adults. However, highly resolvable cellular specialization is less well characterized in different anatomical regions of the skin. Pig is regarded as an excellent model animal for human skin research in view of its similar physiology to human. In this study, single-cell RNA sequencing was performed on pig skin tissues from six different anatomical regions of Chenghua (CH) pigs, with a superior skin thickness trait, and the back site of large white (LW) pigs. We obtained 233,715 cells, representing seven cell types, among which we primarily characterized the heterogeneity of the top three cell types, including smooth muscle cells (SMCs), endothelial cells (ECs), and fibroblasts (FBs). Then, we further identified several subtypes of SMCs, ECs, and FBs, and discovered the expression patterns of site-specific genes involved in some important pathways such as the immune response and extracellular matrix (ECM) synthesis in different anatomical regions. By comparing differentially expressed genes of skin FBs among different anatomical regions, we considered TNN, COL11A1, and INHBA as candidate genes for facilitating ECM accumulation. These findings of heterogeneity in the main skin cell types from different anatomical sites will contribute to a better understanding of inherent skin information and place the potential focus on skin generation, transmission, and transplantation, paving the foundation for human skin priming.

Antarctic sea ice prediction has garnered increasing attention in recent years, particularly in the context of the recent record lows of February 2022 and 2023. As Antarctica becomes a climate change hotspot, as polar tourism booms, and as scientific expeditions continue to explore this remote continent, the capacity to anticipate sea ice conditions weeks to months in advance is in increasing demand. Spurred by recent studies that uncovered physical mechanisms of Antarctic sea ice predictability and by the intriguing large variations of the observed sea ice extent in recent years, the Sea Ice Prediction Network South (SIPN South) project was initiated in 2017, building upon the Arctic Sea Ice Prediction Network. The SIPN South project annually coordinates spring-to-summer predictions of Antarctic sea ice conditions, to allow robust evaluation and intercomparison, and to guide future development in polar prediction systems. In this paper, we present and discuss the initial SIPN South results collected over six summer seasons (December-February 2017-2018 to 2022-2023). We use data from 22 unique contributors spanning five continents that have together delivered more than 3000 individual forecasts of sea ice area and concentration. The SIPN South median forecast of the circumpolar sea ice area captures the sign of the recent negative anomalies, and the verifying observations are systematically included in the 10-90% range of the forecast distribution. These statements also hold at the regional level except in the Ross Sea where the systematic biases and the ensemble spread are the largest. A notable finding is that the group forecast, constructed by aggregating the data provided by each contributor, outperforms most of the individual forecasts, both at the circumpolar and regional levels. This indicates the value of combining predictions to average out model-specific errors. Finally, we find that dynamical model predictions (i.e., based on process-based general circulation models) generally perform worse than statistical model predictions (i.e., data-driven empirical models including machine learning) in representing the regional variability of sea ice concentration in summer. SIPN South is a collaborative community project that is hosted on a shared public repository. The forecast and verification data used in SIPN South are publicly available in near-real time for further use by the polar research community, and eventually, policymakers.

Abstract The predictive skill of summer sea ice concentration (SIC) in the Arctic presents a steep decline when initialized before June, which is the so‐called spring predictability barrier for Arctic sea ice. This study explores the potential influence of surface heat flux, cloud and water vapor anomalies on monthly to seasonal predictions of Arctic SIC anomalies. The results show an enhancement in skill predicting Arctic September SIC in the models that use surface fluxes, clouds, or water vapor in combination with SIC and surface sea temperature as predictors when initialized in boreal spring. This result shows the potential to reduce the spring barrier for Arctic SIC predictions by including the surface heat budget. The enhanced predictive skill can be very likely linked to the improved representation of the thermodynamics associated with water vapor and cloudiness anomalies in spring.

In order to reduce the effect of nonlinear friction and time-varying factors on the servo system of a computer numerical control (CNC) machine tool and improve its motion control accuracy, this paper uses an adaptive sliding mode control (ASMC) method based on model reference adaptive control (MRAC). The method adopts ASMC in the control outer loop and obtains the optimal control parameters by making the sliding mode control (SMC) law continuous and adaptively estimating the control parameters. At the same time, MRAC is used in the control inner loop to enhance the “invariance” of the controlled object so that the switching gain of SMC can satisfy the disturbance matching condition even under lesser conditions. Simulation and experimental results show that compared with the traditional SMC, the ASMC based on MRAC proposed in this paper effectively reduces the influence of nonlinear friction on the system performance, and the reduction in following error reaches 71.2%, which significantly improves the motion control accuracy of the control system. The spectral analysis of the following errors shows that the maximum magnitude reduction rate of the high-frequency chattering is 89.02%, which significantly reduces the effect of the high-frequency chattering and effectively improves the stability performance of the control system.

Abstract. Gastric cancer (GC) is one of the most common malignant tumors worldwide. Its incidence ranks the 5th among all malignant tumors globally, and it is the 3rd leading cause of death among patients with cancer. Surgical treatment is the first choice in clinical practice. However, targeted therapy, immunotherapy, and other treatment methods have also become research hotspots at home and abroad with the development of individualized precision therapy in recent years, besides traditional radiotherapy and chemotherapy. At present, targeted therapy and immunotherapy are methods used for treating GC, and they have important clinical application value and prospects. This study aimed to review the research progress of targeted therapy and immunotherapy for GC, focusing on its mechanism of action and related important clinical trials, hoping to provide references for the clinical treatment of GC.

Abstract Liver development is a complex process that is regulated by a series of signaling pathways. Three-dimensional (3D) chromatin architecture plays an important role in transcriptional regulation; nonetheless, its dynamics and role in the rapid transition of core liver functions during development and obesity-induced metabolic stress remain largely unexplored. To investigate the dynamic chromatin architecture during liver development and under metabolic stress, we generated high-resolution maps of chromatin architecture for porcine livers across six major developmental stages (from embryonic day 38 to the adult stage) and under a high-fat diet-induced obesity. The characteristically loose chromatin architecture supports a highly plastic genome organization during early liver development, which fundamentally contributes to the rapid functional transitions in the liver after birth. We reveal the multi-scale reorganization of chromatin architecture and its influence on transcriptional regulation of critical signaling processes during liver development, and show its close association with transition in hepatic functions (i.e., from hematopoiesis in the fetus to metabolism and immunity after birth). The limited changes in chromatin structure help explain the observed metabolic adaptation to excessive energy intake in pigs. These results provide a global overview of chromatin architecture dynamics associated with the transition of physiological liver functions between prenatal development and postnatal maturation, and a foundational resource that allows for future in-depth functional characterization.

Abstract Skin thickness is closely related to the appearance of human skin, such as sagging and wrinkling, which primarily depends on the level of collagen I synthesized by dermal fibroblasts (DFs). Small extracellular vesicles (SEVs), especially those derived from human DFs (HDFs), are crucial orchestrators in shaping physiological and pathological development of skin. However, the limited supply of human skin prevents the production of a large amount of HDFs-SEVs, and pig skin is used as a model of human skin. In this study, SEVs derived from DFs of Chenghua pigs (CH-SEVs), considered to have superior skin thickness, and Large White pigs (LW-SEVs) were collected to compare their effects on DFs and skin tissue. Our results showed that, compared with LW-SEVs, CH-SEVs more effectively promoted fibroblast proliferation, migration, collagen synthesis and contraction; in addition, in mouse model injected with both SEVs, compared with LW-SEVs, CH-SEVs increased the skin thickness and collagen I content more effectively. Some differentially expressed miRNAs and proteins were found between CH-SEVs and LW-SEVs by small RNA-seq and LC–MS/MS analysis. Interestingly, we identified that CH-SEVs were enriched in miRNA-218 and ITGBL1 protein, which played important roles in promoting fibroblast activity via activation of the downstream TGFβ1-SMAD2/3 pathway in vitro. Furthermore, overexpression of miRNA-218 and ITGBL1 protein increased the thickness and collagen I content of mouse skin in vivo. These results indicate that CH-SEVs can effectively stimulate fibroblast activity and promote skin development and thus have the potential to protect against and repair skin damage. A comprehensive dynamic model is introduced try to further reduce dimensional errors of micromilled micro straight thin walls based on an in-process cutting parameter compensation device. First, a mathematical model is established to predict the dynamic elastic deformation of the thin walls in the micromilling process. Next, the radial deformation and the runout of the cutter are modeled. Then, the comprehensive in-process parametric compensation model and strategy are established. Finally, a real-time in-process cutting force measurement and cutting parameter compensation device is applied to the micromilling system and thin wall fabrication experiments have been conducted. It shows that the comprehensive dimensional error model and iterative compensation method are effective, and the dimension and shape precision of the micro straight thin wall has been obviously improved after the compensation of the radial cutting depth. The average relative errors have been reduced from 6.86% to 1.10%∼1.70%. The in-process compensation method introduced in this study is efficient and convenient as long as compensation strategies are integrated in the controlling unit for a specific micromilling process.

Abstract Automatic image cropping can change the composition to improve the aesthetic quality of the images. Most of the existing automatic image cropping methods based on specific features need to generate a large number of candidate cropping windows. It is very time‐consuming and can only produce a limited aspect ratio results. In the face of these situations, a reinforcement learning cropping method based on comprehensive feature and aesthetics assessment is proposed. It does not need to produce a large number of candidate windows. Its gradually cropping mode is more in line with the process of image cropping by human. What is more, the proposed method takes the image aesthetic assessment into consideration. Experimental results show that the proposed method improves the cropping efficiency and achieves excellent cropping effect on the open Flickr Cropping Dataset and CUHK Image Cropping Dataset. The proposed method can overcome the shortages of existing methods.

BackgroundStroke is one of the most severe diseases worldwide, resulting in physical and mental problems. Dl-3-n-butylphthalide, a compound derived from celery seed, has been approved for treating ischemic stroke in China. No study has evaluated how Dl-3-n-butylphthalide affects the ferroptosis SLC7A11/GSH/GPX4 signal pathway and blood–brain barrier (BBB) PDGFRβ/PI3K/Akt signal pathways in the rat middle cerebral artery occlusion/reperfusion (MCAO/R) model of ischemic stroke.MethodsSprague–Dawley rats were used to develop the MCAO/R model. Our study used three incremental doses (10, 20, and 30) of Dl-3-n-butylphthalide injected intraperitoneally 24 h after MCAO/R surgery. The neuroprotective effect and success of the model were evaluated using the neurofunction score, brain water content determination, and triphenyl-tetrazolium chloride-determined infarction area changes. Pathological changes in the brain tissue and the degree of apoptosis were examined by hematoxylin and eosin, Nissl, and terminal deoxynucleotidyl transferase dUTP nick end labeling staining. In addition, pathway proteins and RNA expression levels were studied to verify the effects of Dl-3-n-butyphthalide on both pathways. At the same time, commercial kits were used to detect glutathione, reactive oxygen species, and malondialdehyde, to detect oxidative stress in brain tissues.ResultsThe middle dose of Dl-3-n-butylphthalide not only improved MCAO-induced brain dysfunction and alleviated pathological damage, brain inflammatory response, oxidative stress, and apoptosis but also protected against ferroptosis and reduced BBB damage. These changes resulted in improved neurological function in the cerebral cortex.ConclusionWe speculate that Dl-3-n-butylphthalide has a neuroprotective effect on focal cerebral ischemia/reperfusion, which may be mediated through ferroptosis-dependent SLC7A11/GSH/GPX4 signal pathway and PDGFRβ/PI3/Akt signal pathway.

As the world's demand for rare earths soars, the supply of rare earths is in danger of being depleted. The recycling rate of rare earth is only 1 % of all rare earth. Therefore, the recovery of rare earth is crucial and challenging. A phosphoric acid modified kaolin (P-K) was prepared by a simple mechanochemical method, and the selective recovery of rare earth element lanthanum (La) was studied. The P-K was characterized and analyzed by SEM, EDS, and FT-IR. Additionally, the different modification conditions of P-K preparation such as milling speed, time, and mass ratio of H3PO4/kaolin on La adsorption were investigated, and the effects of operating conditions such as solution initial pH, adsorption time, and adsorbent concentration on La adsorption were studied. The adsorption behavior of La by P-K was consistent with the Langmuir isothermal adsorption model with a saturated adsorption capacity of 18.55 mg/g, and the adsorption kinetics accorded with the pseudo-second-order kinetic model. Additionally, La in the mixture of La and magnesium (Mg) can be selectively adsorbed through P-K, while the adsorption rate of Mg is almost 0. P-K load with La could be eluted by 0.03 mol/L HCl, and the elution rate of La can reach 99.9 %. As a result, this study demonstrates a method for the recovery of La from rare earth wastewater.

Image dehazing is one of the problems that need to be solved urgently in the field of computer vision. In recent years, more and more algorithms have been applied to image dehazing and achieved good results. However, the image after dehazing still has color distortion, contrast and saturation disorder, and other challenges; in order to solve these problems, in this paper, an effective image dehazing method is proposed, which is based on improved color channel transfer and multiexposure image fusion to achieve image dehazing. First, the image is preprocessed using a color channel transfer method based on k-means. Second, gamma correction is introduced on the basis of guided filtering to obtain a series of multiexposure images, and the obtained multiexposure images are fused into a dehazed image through a Laplacian pyramid fusion scheme based on local similarity of adaptive weights. Finally, contrast and saturation corrections are performed on the dehazed image. Experimental verification is carried out on synthetic dehazed images and natural dehazed images, and it is verified that the method proposed is superior to existing dehazed algorithms from both subjective and objective aspects.

In this study, we aimed to identified CpG sites at which DNA methylation levels are associated with meat quality traits in 140 Yorkshire pigs, including pH at 45 min (pH45min), pH at 24 h (pH24h), drip loss (DL), meat redness value (a*), yellowness (b*) and lightness (L*). Genome-wide methylation levels were measured in muscular tissue using reduced representation bisulfite sequencing (RRBS). Associations between DNA methylation levels and meat quality traits were examined using linear mixed-effect models that were adjusted for gender, year, month and body weight. A Bonferroni-corrected p-value lower than 7.79×10−8 was considered statistically significant threshold. Eight CpG sites were associated with DL, including CpG sites annotated to RBM4 gene (cpg301054, cpg301055, cpg301058, cpg301059, cpg301066, cpg301072 and cpg301073) and NCAM1 gene (cpg1802985). Two CpG sites were associated with b*, including RNFT1 and MED13 (cpg2272837) and TRIM37 gene (cpg2270611). Five CpG sites were associated with L*, including GSDMA and LRRC3C gene (cpg2252750) and ENSSSCG00000043539 and IRX1 gene (cpg2820178, cpg2820179, cpg2820181 and cpg2820182). No significant associations were observed with pH45min, pH24h or a*. We reported associations of meat quality traits with DNA methylation and identified some candidate genes associated with these traits, such as NCAM1, MED13 and TRIM37 gene. These results provide new insight into the epigenetic molecular mechanisms of meat quality traits in pigs.

To investigate the static and dynamic trends of scientific research efficiency of different types of universities in China during the period of 2016 - 2020. Methods: Based on the scientific research data of six types of universities nationwide from 2016 to 2020, which are classified by the Compendium of Science and Technology Information of Higher Education Institutions published by the Department of Science and Technology of the Ministry of Education. We selected the full-time personnel (person-years) of research and development personnel and Internal expenditure for the year of science and technology funds (thousand yuan) as input evaluation indexes, and the number of Academic papers published abroad (articles), the number of international projects acceptance (item), the number of patent authorizations (item), and the number of Actual income of technology patents in the current year (thousands of yuan) as output evaluation indexes, and constructed the evaluation index system of scientific research efficiency of six types of universities nationwide. SPSS version 23.0 software was used for descriptive data statistics, and DEA-BCC model and DEA-Malmquist index model of DEAP2.1 software were used for static and dynamic evaluation of its scientific research efficiency from 2016 to 2020, respectively. Results ① Overall analysis: the level of scientific research efficiency of all types of universities is high, but the total factor productivity of scientific research shows a trend of rising and then declining during the 13th Five-Year Plan period, and the overall scientific research efficiency of universities has limited room for improvement. ② Comparative analysis: universities of comprehensive, science and technology, medicine and other universities have the highest level of scientific research efficiency, followed by universities of teacher training and lower universities of agriculture and forestry. ③ From 2017 - 2020, full-time personnel, internal expenditures of agriculture and forestry universities are input redundancy, patent authorization number and the actual income of agriculture and forestry universities are insufficient output. In 2020, full-time personnel, internal expenditures of normal universities are input redundancy, patent authorization number of normal universities is output insufficient. Conclusion: During the period of 2016 - 2020, all kinds of universities nationwide have achieved high research efficiency with high input and high output, which provides a strong reference for the national research management to allocate university research funds more scientifically and reasonably. This result to optimize the allocation of resources of university's scientific research in China and improve the economic benefit of university's scientific research has important theoretical and practical significance.
