Background Colorectal cancer (CRC) remains a global health concern with persistently high incidence and mortality rates. However, the specific pathogenesis of CRC remains poorly understood. This study aims to investigate the role and pathogenesis of serine and arginine rich splicing factor 10 (SRSF10) in colorectal cancer. Methods Bioinformatics analysis was employed to predict SRSF10 gene expression in CRC patients. Functional experiments involving SRSF10 knockdown and overexpression were conducted using CCK8, transwell, scratch assay, and flow cytometry. Additionally, the PRIdictor website was utilized to predict the SRSF10 interaction site with RFC5. The identification of different transcripts of SRSF10-acting RFC5 pre-mRNA was achieved through agarose gel electrophoresis. Result The knockdown of SRSF10 inhibited the proliferation and migration ability of CRC cells, while promoting apoptosis and altering the DNA replication of CRC cells. Conversely, when SRSF10 was highly expressed, it enhanced the proliferation and migration ability of CRC cells and caused changes in the cell cycle of colorectal cancer cells. This study revealed a change in the replicating factor C subunit 5 (RFC5) gene in colorectal cancer cells following SRSF10 knockdown. Furthermore, it was confirmed that SRSF10 increased RFC5 exon2-AS1(S) transcription variants, thereby promoting the development of colorectal cancer through AS1 exclusion to exon 2 of RFC5. Conclusion In summary, this study demonstrates that SRSF10 promotes the progression of colorectal cancer by generating an aberrantly spliced exclusion isoform of AS1 within RFC5 exon 2. These findings suggest that SRSF10 could serve as a crucial target for the clinical diagnosis and treatment of CRC.

Abstract Rare earth doped up-conversion luminescent nano-materials exhibit abundant emission colors under suitable excitation condition. In this work, NaYF4:Er/Ho@NaYF4 and NaYbF4:Tm@NaYF4 nanoparticles were synthesized by co-precipitation method. The pure red emission can be realized by the designed NaYF4:Er/Ho@NaYF4 nanocrystals and the R/Gs reach 23.3 and 25 under excitations of 980 and 1550 nm lasers, respectively. The R/G declines as the power increasing with the emission color changing from red to yellow, which is due to the quick saturation of the energy levels, radiating red emissions. Meanwhile, the emission intensity of NaYbF4:Tm@NaYF4 nanocrystals increases by 58.3 folds after encasing the inert shell NaYF4 and the CIE color coordinate reaches (0.1646, 0.0602) under 980 nm laser excitation. Furthermore, broad range multicolor from blue to red and yellow up-conversion emissions is achieved by mixing NaYF4:Er/Ho@NaYF4 and NaYbF4:Tm@NaYF4 nanocrystals, which could be applied to colorful displaying, security anti-counterfeiting and information coding.

BackgroundThe relationship between ferroptosis and the progression and treatment of hematological tumors has been extensively studied, although its precise association with chronic myeloid leukemia (CML) remains uncertain.MethodsMulti-transcriptome sequencing data were utilized to analyze the ferroptosis level of CML samples and its correlation with the tumor microenvironment, disease progression, and treatment response. Machine learning algorithms were employed to identify diagnostic ferroptosis-related genes (FRGs). The consensus clustering algorithm was applied to identify ferroptosis-related molecular subtypes. Clinical samples were collected for sequencing to validate the results obtained from bioinformatics analysis. Cell experiments were conducted to investigate the therapeutic efficacy of induced ferroptosis in drug-resistant CML.ResultsFerroptosis scores were significantly lower in samples from patients with CML compared to normal samples, and these scores further decreased with disease progression and non-response to treatment. Most FRGs were downregulated in CML samples. A high ferroptosis score was also associated with greater immunosuppression and increased activity of metabolic pathways. Through support vector machine recursive feature elimination (SVM-RFE), least absolute shrinkage selection operator (LASSO), and random forest (RF) algorithms, we identified five FRGs (ACSL6, SLC11A2, HMOX1, SLC38A1, AKR1C3) that have high diagnostic value. The clinical diagnostic value of these five FRGs and their effectiveness in differentiating CML from other hematological malignancies were validated using additional validation cohorts and our real-world cohort. There are significant differences in immune landscape, chemosensitivity, and immunotherapy responsiveness between the two ferroptosis-related molecular subtypes. By conducting cellular experiments, we confirmed that CML-resistant cells are more sensitive to induction of ferroptosis and can enhance the sensitivity of imatinib treatment.ConclusionOur study unveils the molecular signature of ferroptosis in samples from patients with CML. FRG identified by a variety of machine learning algorithms has reliable clinical diagnostic value. Furthermore, the characterization of different ferroptosis-related molecular subtypes provides valuable insights into individual patient characteristics and can guide clinical treatment strategies. Targeting and inducing ferroptosis holds great promise as a therapeutic approach for drug-resistant CML.

Solanum pinnatisectum exhibits strong resistance to late blight caused by Phytophthora infestans but only an incomplete genome assembly based on short Illumina reads has been published. In this study, we generated the first chromosome-level draft genome for the wild-type potato species S. pinnatisectum in China using Oxford Nanopore technology sequencing and Hi-C technology. The high-quality assembled genome size is 664 Mb with a scaffold N50 value of 49.17 Mb, of which 65.87% was occupied by repetitive sequences, and predominant long terminal repeats (42.51% of the entire genome). The genome of S. pinnatisectum was predicted to contain 34,245 genes, of which 99.34% were functionally annotated. Moreover, 303 NBS-coding disease resistance (R) genes were predicted in the S. pinnatisectum genome to investigate the potential mechanisms of resistance to late blight disease. The high-quality chromosome-level reference genome of S. pinnatisectum is expected to provide potential valuable resources for intensively and effectively investigating molecular breeding and genetic research in the future.

Abstract Shading in combination with extended photoperiods can cause exaggerated stem elongation (ESE) in soybean, leading to lodging and reduced yields when planted at high-density in high-latitude regions. However, the genetic basis of plant height in adaptation to these regions remains unclear. Here, through a genome-wide association study, we identify a plant height regulating gene on chromosome 13 (PH13) encoding a WD40 protein with three main haplotypes in natural populations. We find that an insertion of a Ty1/Copia-like retrotransposon in the haplotype 3 leads to a truncated PH13H3 with reduced interaction with GmCOP1s, resulting in accumulation of STF1/2, and reduced plant height. In addition, PH13 H3 allele has been strongly selected for genetic improvement at high latitudes. Deletion of both PH13 and its paralogue PHP can prevent shade-induced ESE and allow high-density planting. This study provides insights into the mechanism of shade-resistance and offers potential solutions for breeding high-yielding soybean cultivar for high-latitude regions.

ABSTRACT: As one of the main ingredients in some milk powders, whey powder is sometimes added to pure goat milk products, which can cause health risks, economic fraud, and unfair competition of food industries. This study is the first to explore qualitative and quantitative methods to identify adulteration of bovine whey powder in goat dairy products based on DNA. We extracted DNA from whey powder using a modified DNA extraction method; this exhibited good quality and integrity, with purity of 1.53 to 1.75 and concentration of 122 to 179 ng/μL. Conventional PCR and real-time PCR were compared for qualitative detection of bovine whey powder; real-time PCR demonstrated sensitivity of 0.01 ng/μL, which was higher than the 0.05 ng/μL detected by the conventional PCR method. Furthermore, real-time PCR was conducted for DNA quantitative detection, with good linearity (R2 = 0.9858) obtained for bovine whey powder contents from 0.1% to 30%. Relative error decreased with increase of the mixing proportion of whey powder; the coefficient of variation above 0.1% of the mixing ratio was close to or less than 5%; and the relative standard deviation of repeatability results was less than 5%. Considering the economic costs of testing, conventional PCR could be performed first, and samples with obvious intentional adulteration detected can be further accurately quantified by real-time PCR. Overall, this research provides a realistic and effective method for qualitative and quantitative identification of bovine whey powder in goat dairy products, thus laying a good foundation for verification of goat dairy product label claims and industrial control.

A digital twin is a simulator of a physical system, which is built upon a series of models and computer programs with real-time data (from sensors or devices). Digital twins are used in various industries, such as manufacturing, healthcare, and transportation, to understand complex physical systems and make informed decisions. However, predictions and optimizations with digital twins can be time-consuming due to the high computational requirements and complexity of the underlying computer programs. This poses significant challenges in making well-informed and timely decisions using digital twins. This paper proposes a novel methodology, called the “digital triplet”, to facilitate real-time prediction and decision-making. A digital triplet is an efficient representation of a digital twin, constructed using statistical models and effective experimental designs. It offers two noteworthy advantages. Firstly, by leveraging modern statistical models, a digital triplet can effectively capture and represent the complexities of a digital twin, resulting in accurate predictions and reliable decision-making. Secondly, a digital triplet adopts a sequential design and modeling approach, allowing real-time updates in conjunction with its corresponding digital twin. We conduct comprehensive simulation studies to explore the application of various statistical models and designs in constructing a digital triplet. It is shown that Gaussian process regression coupled with sequential MaxPro designs exhibits superior performance compared to other modeling and design techniques in accurately constructing the digital triplet.

Personal information and other types of private data are valuable for both data owners and institutions interested in providing targeted and customized services that require analyzing such data. In this context, privacy is sometimes seen as a commodity: institutions (data buyers) pay individuals (or data sellers) in exchange for private data. In this study, we examine the problem of designing such data contracts, through which a buyer aims to minimize his payment to the sellers for a desired level of data quality, while the latter aim to obtain adequate compensation for giving up a certain amount of privacy. Specifically, we use the concept of differential privacy and examine a model of linear and nonlinear queries on private data. We show that conventional algorithms that introduce differential privacy via zero-mean noise fall short for the purpose of such transactions as they do not provide a sufficient degree of freedom for the contract designer to negotiate between the competing interests of the buyer and the sellers. Instead, we propose a biased randomized algorithm to generate differentially private output and show that this algorithm allows us to customize the privacy-accuracy tradeoff for each individual. We use a contract design approach to find the optimal contracts when using this biased algorithm to provide privacy and show that under this combination the buyer can achieve the same level of accuracy with a lower payment as compared to using the conventional, unbiased algorithms, while at the same time incurring lower privacy loss for the sellers.

Fractals have emerged as a promising platform for realizing broadband absorption metasurfaces. In this paper, we propose an ultra-broadband absorber with high absorption operational in the long-wave infrared regime by means of a second-order Peano fractal curve (PFC) metasurface with a four-layer insulator–metal–insulator–metal (Ge/Cr/Ge/Cr) waveguide structure. The unique space-filling shape of the PFC enables the generation of multiple gap plasmon resonance modes, which expands the bandwidth of the absorber. In addition, the hybridization of these modes with cavity mode resonances and localized surface plasmon resonances enhances the average absorption of the device in the spectrum of operation, with an average absorption of 94.3% obtained for TM-polarized light with wavelengths between 12.3 µm and 32 µm in our simulations. A second-order quasi-PFC is designed for polarization-insensitive broadband absorption. Our findings establish general and systematic strategies for guiding the design of metamaterial absorbers with excellent broadband absorption.

All‐optical control of magnetization vectors is considered as a powerful solution that empowers the development of multifunctional integrated optomagnetic devices. The interaction between vectorial light and magnetism has recently experienced a surge of interest due to its prominent ability to steer the magnetic polarization orientations and spatial textures within the subwavelength domain, which, however, usually requires complex wavefront coding optimizers or arduous material fabrication procedures. Herein, an optimization‐free all‐vectorial‐optical strategy for first realizing spatially twist‐controllable and successively polarization‐tunable magnetization at the subdiffraction scale through the inverse Faraday effect is conceived and demonstrated. This facile method relies on the coherent coupling of double crossed azimuthally polarized doughnut‐Gaussian vortex beams in a single optically configured geometry. It is found that both the magnetic twisting manifolds and polarization states, rotating from perpendicular (longitudinal), via hybrid (3D) to in‐plane (transverse) orientations, can be on‐demand mediated by judiciously tuning the crossing angles. It is further unraveled that the orbital angular momenta associated with topological charges of the vortex beams can be viewed as a vital knob to energetically maneuver magnetic twisting and guide magnetization switching. It is believed that the proposed route and presented findings are not only of great theoretical implication in twistronics and optomagnetic topology but also of considerable technological significance in multidimensional high‐density and low‐energy consumption optomagnetic storage and engineering of magnetic topologic materials.

Abstract Background The hypolipidemic effect of phytosterols has been wildely recognized, but its application is limited due to its insolubility in water and low solubility in oil. In this study, β-sitosterol ester with linoleic acids and β-sitosterol self-microemulsions were prepared and their hypolipidemic effects on hyperlipidemia mice were studied. Methods Firstly, the mice were randomly divided into normal group and model group,they were fed with basic diet and high-fat diet for 70 days respectively. After high-fat model mice was successfully established, the model group was further divided into eight groups: HFD (high-fat diet feeding), SELA-TSO(8 ml/kg, SELA:700 mg/kg), TSO (8 ml/kg), SSSM (8 ml/kg,SS:700 mg/kg), NLSM (8 ml/kg), SSHT-TSO (8 ml/kg, SS: 700 mg/kg) and SS-TSO (8 ml/kg, SS: 700 mg/kg) groups, and treated with β-sitosterol ester with linoleic acid, β-sitosterol self-microemulsion, commercial β-sitosterol health tablets and β-sitosterol powder for 35 days, respectively, and blank control groups were established. At the end of the treatment period, the blood lipid level, tissues, cholesterol and lipids in feces of mice in each group were investigated. Statistical and analytical data with SPSS 17.0 Software,statistical significance was set at p* SELA-TSO > SSHT-TSO > SS-TSO, which shows that β-sitosterolself-microemulsion have the highest treatment effect among the experimental groups. Conclusions In this study, a new formulation of β-sitosterol was developed, and its hypolipidemic effect was investigated. The results showed that β-sitosterol self-microemulsion has a good blood lipid lowering effect.

A novel two-branched twistacene (PyDN) has been designed and synthesized for application on ultrafast optical limiting. This twistacene exhibits excellent two photon absorption and two photon absorption-induced excited singlet state absorption, which was systematically investigated with a femtosecond Z-scan experiment, transient absorption spectrum, and two-photon excited fluorescence experiments. The admirable two photon absorption is attributed to the high degree of π electron delocalization in twistacene which is caused by introduction of two strong donors. The excited singlet state absorption cooperates with two-photon absorption to provide an excellent ultrafast optical limiting behavior with high linear transmittance, where the thresholds are 2.3–5.3 mJ/cm2 in the spectral region of 532–800 nm of femtosecond laser and 133 mJ/cm2 for picosecond pulse at 532 nm. These thresholds are lower than that of most of the optical limiters reported previously, which indicates PyDN is a promising candidate for ultrafast optical limiting.

Objective To clarify the efficacy of functional magnetic stimulation (FMS) in improving hemiplegic upper extremity function in patients with sub-acute stroke. Methods In this randomized controlled trial, 40 sub-acute stroke patients with hemiplegia were recruited from inpatient wards in the Department of Rehabilitation and randomly assigned to two groups. In the FMS group, magnetic stimulation was applied to extensor muscle groups of the affected upper extremity. In the low-frequency repetitive transcranial magnetic stimulation (LF-rTMS) group, stimulation was applied to the contralesional primary motor cortex. All patients received occupational therapy. Hand and upper extremity motor function was evaluated using the Fugl–Meyer Assessment for upper extremity (FMA-UE), and the Barthel Index (BI) evaluated daily living abilities. Results The FMA-UE and BI scores were significantly increased in both groups following stimulation. Furthermore, a significant between-group difference was observed in both FMA-UE and BI scores after 2 weeks of therapy. In the FMS group, 6 of 19 patients regained wrist and finger extension abilities, but only 2 patients regained equivalent motor skills in the LF-rTMS group Conclusions FMS improves paretic upper extremity function and leads to better recovery of motor activity than LF-rTMS. FMS may be a novel modality to improve motor function.

The linear and nonlinear optical properties of two BODIPY derivatives, 1,7-Diphenyl-3,5-bis(9,9-dimethyl-9H-fluoren-2-yl)-boron-diuoride-azadipyrromethene (ZL-61) and 1,7-Diphenyl-3,5-bis(4-(1,2,2-triphenylvinyl)phenyl)-boron-diuoride-azadipyrromethene (ZL-22), were comprehensively investigated based on experimental and theoretical studies. It was found that both compounds show a strong two-photon absorption response in the near-infrared regime, and the two-photon-absorption cross-section values of ZL-61 and ZL-22 were determined to be 8321 GM and 1864 GM at 800 nm, respectively. The improvement of the two-photon absorption cross section in ZL-61 was attributed to the enhancement of the donor group, which was confirmed by transient absorption measurements and DFT calculation. Our results indicate that these BODIPY derivatives are a promising candidate for optical limiting and two-photon imaging applications.

Developing countries account for about 86.5% of the world’s population and are experiencing rapid urbanization. Globally, the increase in the urban population is generally accompanied by the expansion of the latter and construction lands, as well as the reduction in the rural population and rural construction lands. However, with the rapid development of urbanization in China, the rural population has decreased, while the proportion of rural construction lands has increased, resulting in a significant waste of land resources. In order to quantitatively characterize the degree of deviation between the permanent rural population and rural construction lands based on the 2009–2016 demographic data and land survey data in China, we comprehensively used the decoupling model and the coordination degree model to analyze the temporal change characteristics, spatial distribution law, and the degree of deviation of rural construction land areas and the number of rural permanent residents. Firstly, according to the decoupling model, the type of decoupling between the area of rural construction lands and the number of rural permanent residents at the national scale was strongly negative. Secondly, according to the coordination degree model, the coordination type between rural construction land areas and the rural resident population was uncoordinated; at the provincial scale, the coordination system involved one city and one district (Beijing and the Tibet Autonomous Region) and the basic coordination of two cities (Tianjin and Shanghai). Xinjiang and Qinghai belonged to the reconcilable type, and the other 25 provinces belonged to the uncoordinated type. Finally, according to the comprehensive measurement model, the number of rural permanent residents and rural construction lands showed two types of decoupling: highly strong negative decoupling incoordination and moderately and weakly strong negative decoupling incoordination.

Accurate and reliable non-contact temperature sensors are imperative for industrial production and scientific research. Here, Er3+/Tm3+/Yb3+ co-doped NaYF4 phosphors were studied as an optical thermometry material. The typical hydrothermal method was used to synthesize hexagonal Er3+/Tm3+/Yb3+ co-doped NaYF4 phosphors and the morphology was approximately rod-like. The up-conversion emissions of the samples were located at 475, 520, 550, 650, 692 and 800 nm. Thermo-responsive emissions from the samples were monitored to evaluate the relative sensing sensitivity. The thermal coupled energy level- and non-thermal coupled energy level-based luminescence intensity ratio thermometry of the sample demonstrated that these two methods can be used to test temperature. Two green emissions (520 and 550 nm), radiated from 2H11/2/4S3/2 levels, were monitored, and the maximum relative sensing sensitivities reached to 0.013 K−1 at 297 K. The emissions located in the first biological window (650, 692 and 800 nm) were monitored and the maximum relative sensing sensitivities reached to 0.027 (R692/650) and 0.028 K−1 (R692/800) at 297 K, respectively. These results indicate that Er3+/Tm3+/Yb3+ co-doped NaYF4 phosphors have potential applications for temperature determination in the visible and the first biological window ranges.

We present, in this work, a new method to measure the chromatic dispersion of liquid based on the surface plasmon resonance sensor. The liquid sample is serving as the liquid prism at the same time. Both angular and spectral interrogations are used in the experiments. The chromatic dispersions of six kinds of liquids with the wavelengths from 450nm to 1050nm are obtained, respectively. The experimental data are also compared with the data of Abbe refractometer, and they are in good agreement. The method provides a practical application for measuring the chromatic dispersion of liquid in a wide spectral range. Keywords: Surface plasmon resonance, Chromatic dispersion

In this work, we present a feasible pathway for initially constructing light-induced isotropic super-resolved magnetization along with steerable orientations and trivial side-lobe levels. Toward this end, the counter-propagating azimuthally polarized vortex Hermite–Gauss beams are tailored with angular rotators at the exit pupil planes and then focused by using high numerical aperture objective lenses in a 4π optical microscopic configuration. By wilfully regulating the rotatable azimuth angle and judiciously optimizing the scaling parameter, the spherical super-resolved (λ3/24), orientation-tunable (any direction), and sidelobe-negligible (

meso-Tetraphenylporphyrin (TPP) and its two substituted derivatives (meso-tetrakis(4-cyanophenyl)porphyrin [TPP(CN)4] and meso-tetrakis(4-methoxyphenyl)porphyrin [TPP(OMe)4]) were synthesized. Their nonlinear absorption and refraction properties were studied using the Z-scan technique in the picosecond (ps) and nanosecond (ns) regimes. The open aperture Z-scan results reveal that TPP and TPP(CN)4 display an identical reverse saturable absorption (RSA) character in the ps and ns regimes. While TPP(OMe)4 exhibits a transition from saturable absorption (SA) to RSA in the ps regime and a typical RSA character in the ns regime. The closed aperture Z-scan results show that TPP(CN)4 and TPP(OMe)4 have regular enhancement of the magnitude of nonlinear refraction as compared to their parent TPP in both the ps and ns regimes. In addition, the second-order molecular hyperpolarizabilities (γ) of these three porphyrins are calculated, and the γ values of TPP(CN)4 and TPP(OMe)4 are remarkable larger than that of TPP. The introduction of the electron-withdrawing group CN and the electron-donating group OMe into TPP has enhanced its nonlinear refraction and γ value, and tuned its nonlinear absorption (TPP(OMe)4), which could be useful for porphyrin-related applications based on the desired NLO properties.

“Smart” materials are an emerging category of multifunctional materials with physical or chemical properties that can be controllably altered in response to an external stimulus. By combining the standard properties of the advanced material with the unique ability to recognize and adapt in response to a change in their environment, these materials are finding applications in areas such as sensing and drug delivery. While the majority of these materials are responsive to physical or chemical changes, a particularly exciting area of research seeks to develop smart materials that are sensitive to specific molecular or biomolecular stimuli. These systems require the integration of a molecular recognition probe specific to the target molecule of interest. The ease of synthesis and labeling, low cost, and stability of DNA aptamers make them uniquely suited to effectively serve as molecular recognition probes in novel smart material systems. This review will highlight current work in the area of aptamer-based smart materials and prospects for their future applications.

It has been reported that backward surface waves of Tamm state can be supported by a multilayered metal–dielectric metamaterial with different partner materials, such as left-handed metamaterial or dissimilar metallic–dielectric metamaterial. In this paper, the transfer-matrix method is employed to reveal that transverse-magnetic (TM)-polarized backward surface waves can be realized by a multilayered metal–dielectric metamaterial in contact with a conventional homogenous dielectric medium. Owing to the strong optical nonlocality, the existence of such backward surface waves is proved to be dependent on the order of the metallic/dielectric layers. The relevant anomalous dispersion relations can also be dramatically engineered by varying the unit-cell thickness and the filling factor. Additionally, the distribution of the energy flow is presented to further unfold the physical mechanism of the backward surface waves. Finally, a numerical simulation of backward surface wave excited by a TM-polarized Gaussian beam based on a prism-coupled configuration is displayed.
