Given global agricultural challenges such as population growth, climate change, and limitations on resources and the environment, as well as increasing diversity in breeding goals, relying on traditional breeding methods is inadequate to provide food security requirements and promote sustainable development. Genetic transformation technology has become an effective tool for performing functional genomics research and molecular breeding. In this study, we conducted an in-depth analysis of 1669 literary works to investigate the potential of developmental regulators (DRs) in enhancing the efficiency of plant genetic transformation, with a concentration on their use in maize. Through multi-omics data analysis, we identified 12 homologous DRs from various species that are potentially applicable to maize. We identified a total of 41 possible disease resistances (DRs) for maize genetic transformation. Further experimental verification of ZmWIND1, a novel regulator belonging to the ERF/AP2 transcription factor (TF) family, showed that it significantly improved the efficiency of plant regeneration and transformation efficiency in maize. Specifically, compared to the control group, the callus induction rates for the pG3GB411-ZmWIND1 vector increased to 60.22 % and 47.85 % in Xiang249 and Zheng58, respectively. Transformation efficiency increased to 37.5 % in Xiang249 and 16.56 % in Zheng58, both significantly surpassing the control group. These findings have the potential to broaden the range of transformable maize varieties and lines, as well as introduce new genetic transformation methods in agricultural biotechnology, underscoring the immense potential to enhance genetic transformation efficiency through systematic exploration and application of DRs in maize.

Enhancing the genetic transformation efficiency of major crops remains a significant challenge, primarily due to their suboptimal regeneration efficiency. Developmental regulators, known as key regulatory genes, involved in plant meristem and somatic embryo formation, play a crucial role in improving plant meristem induction and regeneration. This review provides a detailed summary of the molecular mechanisms and regulatory networks of many developmental regulators, in the context of enhancing the genetic transformation efficiency in major crops. We also propose strategies for exploring and utilizing additional developmental regulators. Further investigation into the mechanisms of these regulators will deepen our understanding of the regenerative capacity and genetic transformation processes of plants, offering valuable support for future crop improvement efforts. The discovery of novel developmental regulators is expected to further advance crop transformation and the effective manipulation of various developmental regulators could provide a promising approach in order to enhance genetic transformation efficiency.

Plant male reproduction is a fine-tuned developmental process that is susceptible to stressful environments and influences crop grain yields. Phytohormone signaling functions in control of plant normal growth and development as well as in response to external stresses, but the interaction or crosstalk among phytohormone signaling, stress response, and male reproduction in plants remains poorly understood. Cross-species comparison among 514 stress-response transcriptomic libraries revealed that ms33-6038, a genic male sterile mutant deficient in the ZmMs33/ZmGPAT6 gene, displayed an excessive drought stress-like transcriptional reprogramming in anthers triggered mainly by disturbed jasmonic acid (JA) homeostasis. An increased level of JA appeared in ZmMs33-deficient anthers at both meiotic and post-meiotic stages and activated genes involved in JA biosynthesis and signaling as well as genes functioning in JA-mediated drought response. Excessive accumulation of JA elevated expression level of a gene encoding a WRKY transcription factor that activated the ZmMs33 promoter. These findings reveal a feedback loop of ZmMs33-JA-WRKY-ZmMs33 in controlling male sterility and JA-mediated stress response in maize, shedding light on the crosstalk of stress response and male sterility mediated by phytohormone homeostasis and signaling.

Introduction: ATP Binding Cassette G (ABCG) transporters are associated with plant male reproduction, while their regulatory mechanisms underlying anther and pollen development remain largely unknown. Objectives: Identify and characterize a male-sterility gene ZmMs13 encoding an ABCG transporter in modulating anther and pollen development in maize. Methods: Phenotypic, cytological observations, and histochemistry staining were performed to characterize the ms13-6060 mutant. Map-based cloning and CRISPR/Cas9 gene editing were used to identify ZmMs13 gene. RNA-seq data and qPCR analyses, phylogenetic and microsynteny analyses, transient dual-luciferase reporter and EMSA assays, subcellular localization, and ATPase activity and lipidomic analyses were carried out to determine the regulatory mechanisms of ZmMs13 gene. Results: Maize ms13-6060 mutant displays complete male sterility with delayed callose degradation, premature tapetal programmed cell death (PCD), and defective pollen exine and anther cuticle formation. ZmMs13 encodes a plasm membrane (PM)- and endoplasmic reticulum (ER)-localized half-size ABCG transporter (ZmABCG2a). The allele of ZmMs13 in ms13-6060 mutant has one amino acid (I311) deletion due to a 3-bp deletion in its fourth exon. The I311 and other conserved amino acid K99 are essential for the ATPase and lipid binding activities of ZmMS13. ZmMs13 is specifically expressed in anthers with three peaks at stages S5, S8b, and S10, which are successively regulated by transcription factors ZmbHLH122, ZmMYB84, and ZmMYB33-1/-2 at these three stages. The triphasic regulation of ZmMs13 is sequentially required for callose dissolution, tapetal PCD and pollen exine development, and anther cuticle formation, corresponding to transcription alterations of callose-, ROS-, PCD-, sporopollenin-, and anther cuticle-related genes in ms13-6060 anthers. Conclusion: ms13-6060 mutation with one key amino acid (I311) deletion greatly reduces ZmMS13 ATPase and lipid binding activities and displays multiple effects during maize male reproduction. Our findings provide new insights into molecular mechanisms of ABCG transporters controlling anther and pollen development and male fertility in plants.

Plant cell elongation depends on well-defined gene regulations, adequate nutrients, and timely cell wall modifications. Anther size is positively correlated with the number and viability of pollen grains, while little is known about molecular mechanisms underlying anther cell elongation. Here, we found that properly activated cell elongation regulators at transcriptional levels in loss-of-function ZmMs33 mutant (ms33-6038) anthers failed to promote maize anther elongation. ZmMs33 deficiency disrupted metabolic homeostasis mainly by inhibiting both photosynthesis in anther endothecium and lipid accumulation in anther tapetum. Importantly, ms33-6038 anthers displayed ectopic, premature and excessive secondary cell wall thickening in anther middle layer, which constrained cell elongation structurally and blocked nutrient flows across different anther wall layers. The metabolic disorder was only found in ms33-6038 mutant rather than several representative male-sterility lines at transcriptional and post-translational levels. Collectively, the disordered metabolisms and blocked nutrient flows defeated the activated cell elongation regulators, and finally inhibited anther elongation and growth with a unique “idling effect” in ms33-6038 mutant.

Anther development is a programmed biological process crucial to plant male reproduction. Genome-wide analyses on the functions of transcriptional factor (TF) genes and their microRNA (miRNA) regulators contributing to anther development have not been comprehensively performed in maize. Here, using published RNA-Seq and small RNA-Seq (sRNA-Seq) data from maize anthers at ten developmental stages in three genic male-sterility (GMS) mutants (ocl4, mac1, and ms23) and wild type W23, as well as newly sequenced maize anther transcriptomes of ms7-6007 and lob30 GMS mutants and their WT lines, we analyzed and found 1079 stage-differentially expressed (stage-DE) TF genes that can be grouped into six (premeiotic, meiotic, postmeiotic, premeiotic-meiotic, premeiotic-postmeiotic, and meiotic-postmeiotic clusters) expression clusters. Functional enrichment combined with cytological and physiological analyses revealed specific functions of genes in each expression cluster. In addition, 118 stage-DE miRNAs and 99 miRNA-TF gene pairs were identified in maize anthers. Further analyses revealed the regulatory roles of zma-miR319 and zma-miR159 as well as ZmMs7 and ZmLOB30 on ZmGAMYB expression. Moreover, ZmGAMYB and its paralog ZmGAMYB-2 were demonstrated as novel maize GMS genes by CRISPR/Cas9 knockout analysis. These results extend our understanding on the functions of miRNA-TF gene regulatory pairs and GMS TF genes contributing to male fertility in plants.

How to predict wind speed with high accuracy is a fundamental issue for the generation of wind power and energy management of power systems. Moreover, the nonlinear and non-stationary characteristics of wind speed make the task extremely challenging. To resolve this issue, a novel hybrid interval prediction model based on long short-term memory (LSTM) networks and variational mode decomposition (VMD) algorithm is developed in the frame of lower upper bound estimation in this study. Firstly, VMD is applied to decompose sequences of raw wind speed into several different modal components containing different information. Then, a LSTM-based model for each component is built to predict the corresponding wind speed information. In the LSTM structure, lower and upper bounds of the prediction intervals (PIs) are constructed and two customized cost functions based on the probability of convergence and width of the PIs are proposed to optimize the model with gradient descent algorithm. Additionally, particle swarm optimization (PSO) is applied to optimize the hyperparameters in the two cost functions of each LSTM. Finally, experiments with different scales of datasets were conducted to show the feasibility and effectiveness of the proposed method by comparing it with traditional methods. The results demonstrate that the proposed model yielded significantly superior performance with a clear enhancement in the PI quality.

In this work, we present and analyze methods and mechanisms for interconnecting a network slice control and management system of the mobile network, with an IEEE Time-Sensitive Network (TSN) control plane. IEEE TSN is gaining momentum as a key technology that is able to provide network service guarantees for Ethernet-based communications. Although Ultra-Reliable Low-Latency Communications (URLLC) have been thoroughly investigated in 5G, incorporating TSN technologies in the Transport Network is expected to unleash the potential of end-to-end deterministic communications, especially in industrial environments and time-critical applications like factory automation.We elaborate on the concepts of a TSN-aware Xhaul network, present a novel architecture, and describe a set of amendments required in order to enable network slicing. With the devised approach, a slice-aware TSN-enabled transport network can be controlled and managed in an end-to-end orchestrated way. Implementation experience and evaluation results are reported using TSN-enabled prototype devices, OpenAirInterface (OAI), and JOX slice orchestrator.

Model representation of carbon uptake and storage is essential for accurate projection of the response of the arctic-boreal zone to a rapidly changing climate. Land model estimates of LAI and aboveground biomass that can have a marked influence on model projections of carbon uptake and storage vary substantially in the arctic and boreal zone, making it challenging to correctly evaluate model estimates of Gross Primary Productivity (GPP). To understand and correct bias of LAI and aboveground biomass in the Community Land Model (CLM), we assimilated the 8-day Moderate Resolution Imaging Spectroradiometer (MODIS) LAI observation and a machine learning product of annual aboveground biomass into CLM using an Ensemble Adjustment Kalman Filter (EAKF) in an experimental region including Alaska and Western Canada. Assimilating LAI and aboveground biomass reduced these model estimates by 58% and 72%, respectively. The change of aboveground biomass was consistent with independent estimates of canopy top height at both regional and site levels. The International Land Model Benchmarking system assessment showed that data assimilation significantly improved CLM's performance in simulating the carbon and hydrological cycles, as well as in representing the functional relationships between LAI and other variables. To further reduce the remaining bias in GPP after LAI bias correction, we re-parameterized CLM to account for low temperature suppression of photosynthesis. The LAI bias corrected model that included the new parameterization showed the best agreement with model benchmarks. Combining data assimilation with model parameterization provides a useful framework to assess photosynthetic processes in LSMs.

With the remarkable growth of natural gas consumption and the development of renewable energy worldwide in recent years, the penetration capacity of gas-fired generators and uncertain renewable energy has significantly become larger, which poses a great challenge to the reliable and economic operation of the gas–electricity interconnected system. This paper proposes an interval optimization based coordination scheduling model for the gas–electricity coupled system considering the dynamic characteristics of natural gas flow, wind power integration and demand response management. By introducing the arithmetic and order relation of interval numbers, the objective function and corresponding constraints for the interval-based dispatch model can be converted into the deterministic expressions with degrees of pessimism, then the proposed model can be solved in the master-subproblem framework. On this basis, two case studies are implemented on the 6-bus power system integrated with 6-node natural gas system and the modified IEEE 118-bus system with 10-node natural gas system to investigate the impact of gas flow dynamic process, demand response management, wind power uncertainty on the scheduling solution of the coupled systems. Moreover, the scenario-based stochastic optimization and robust optimization methods are carried out for comparison. Simulation results demonstrate the effectiveness of the proposed interval optimization method.

Gene editing (GE) has become one of the mainstream bioengineering technologies over the past two decades, mainly fueled by the rapid development of the CRISPR/Cas system since 2012. To date, plenty of articles related to the progress and applications of GE have been published globally, but the objective, quantitative and comprehensive investigations of them are relatively few. Here, 13,980 research articles and reviews published since 1999 were collected by using GE-related queries in the Web of Science. We used bibliometric analysis to investigate the competitiveness and cooperation of leading countries, influential affiliations, and prolific authors. Text clustering methods were used to assess technical trends and research hotspots dynamically. The global application status and regulatory framework were also summarized. This analysis illustrates the bottleneck of the GE innovation and provides insights into the future trajectory of development and application of the technology in various fields, which will be helpful for the popularization of gene editing technology.

In plants, oxidative stress and metabolic reprogramming frequently induce male sterility, however our knowledge of the underlying molecular mechanism is far from complete. Here, a maize genic male-sterility (GMS) mutant (ms33-6038) with a loss-of-function of the ZmMs33 gene encoding glycerol-3-phosphate acyltransferase 6 (GPAT6) displayed severe deficiencies in the development of a four-layer anther wall and microspores and excessive reactive oxygen species (ROS) content in anthers. In ms33-6038 anthers, transcriptome analysis identified thousands of differentially expressed genes that were functionally enriched in stress response and primary metabolism pathways. Further investigation revealed that 64 genes involved in ROS production, scavenging, and signaling were specifically changed in expression levels in ms33-6038 anthers compared to the other five investigated GMS lines. The severe oxidative stress triggered premature tapetal autophagy and metabolic reprogramming mediated mainly by the activated SnRK1-bZIP pathway, as well as the TOR and PP2AC pathways, proven by transcriptome analysis. Furthermore, 20 reported maize GMS genes were altered in expression levels in ms33-6038 anthers. The excessive oxidative stress and the metabolic reprogramming resulted in severe phenotypic deficiencies in ms33-6038 anthers. These findings enrich our understanding of the molecular mechanisms by which ROS and metabolic homeostasis impair anther and pollen development in plants.

The phase 3 INO-VATE trial demonstrated higher rates of remission, measurable residual disease negativity, and improved overall survival for patients with relapsed/refractory (R/R) acute lymphoblastic leukemia (ALL) who received inotuzumab ozogamicin (InO) vs standard-of-care chemotherapy (SC). Here, we examined associations between genomic alterations and the efficacy of InO. Of 326 randomized patients, 91 (InO, n = 43; SC, n = 48) had samples evaluable for genomic analysis. The spectrum of gene fusions and other genomic alterations observed was comparable with prior studies of adult ALL. Responses to InO were observed in all leukemic subtypes, genomic alterations, and risk groups. Significantly higher rates of complete remission (CR)/CR with incomplete count recovery were observed with InO vs SC in patients with BCR::ABL1-like ALL (85.7% [6/7] vs 0% [0/5]; P = .0076), with TP53 alterations (100% [5/5] vs 12.5% [1/8]; P = .0047), and in the high-risk BCR::ABL1- (BCR::ABL1-like, low-hypodiploid, KMT2A-rearranged) group (83.3% [10/12] vs 10.5% [2/19]; P < .0001). This retrospective, exploratory analysis of the INO-VATE trial demonstrated potential for benefit with InO for patients with R/R ALL across leukemic subtypes, including BCR::ABL1-like ALL, and for those bearing diverse genomic alterations. Further confirmation of the efficacy of InO in patients with R/R ALL exhibiting the BCR::ABL1-like subtype or harboring TP53 alterations is warranted. Male sterility represents an important trait for hybrid breeding and seed production in crops. Although the genes required for male fertility have been widely studied and characterized in many plant species, most of them are single genic male-sterility (GMS) genes. To investigate the role of multiple homologous genes in anther and pollen developments of maize, we established the CRISPR/Cas9-based gene editing method to simultaneously mutate the homologs in several putative GMS gene families. By using the integrated strategies of multi-gene editing vectors, maize genetic transformation, mutation-site analysis of T0 and F1 plants, and genotyping and phenotyping of F2 progenies, we further confirmed gene functions of every member in ZmTGA9-1/-2/-3 family, and identified the functions of ZmDFR1, ZmDFR2, ZmACOS5-1, and ZmACOS5-2 in controlling maize male fertility. Single and double homozygous gene mutants of ZmTGA9-1/-2/-3 did not affect anther and pollen development, while triple homozygous gene mutant resulted in complete male sterility. Two single-gene mutants of ZmDFR1/2 displayed partial male sterility, but the double-gene mutant showed complete male sterility. Additionally, only the ZmACOS5-2 single gene was required for anther and pollen development, while ZmACOS5-1 had no effect on male fertility. Our results show that the CRISPR/Cas9 gene editing system is a highly efficient and convenient tool for identifying multiple homologous GMS genes. These findings enrich GMS genes and mutant resources for breeding of maize GMS lines and promote deep understanding of the gene family underlying pollen development and male fertility in maize.
