Frosting is a common phenomenon of the ASHP under the heating mode in winter, and the outdoor air flow rate flowing through the evaporator of the ASHP was always thought to be a major contributor. In order to validate its contribution, effects of outdoor fan airflow rate on the performance of air source heat pumps (ASHPs) were investigated under the winter heating condition. The experiment was conducted in a laboratory at the standard 2 °C air dry bulb temperature (DB)/ 1 °C air wet bulb temperature (WB) frosting condition, which enabled the analysis of the operating performance, frosting performance, and heating performance of the ASHP unit by changing the airflow rate of the outdoor fan. Results showed that as the airflow rate of the outdoor fan reduced from 100% to 36%, the operating performance decline and the elevated frosting-defrosting loss were observed. Meanwhile, both the frosting rate and the operating efficiency during frosting-defrosting cycles showed an increasing trend then followed by decreasing tendency. The maximum frosting rate and operating efficiency were 0.92 g/m2.min and 2.92, respectively, which were observed at 74% airflow rate of the outdoor fan of the ASHP unit. The observation implied the existence of the “minimum frosting suppression airflow rate”. At 36% airflow rate of the outdoor fan of the ASHP unit, however, the performance of the ASHP unit was attenuated greatly, with the frosting-defrosting efficiency loss coefficient of 0.47, the heating capacity and COP reduction by 51.5 and 38.8%, respectively. These findings provided significant references to the optimization of ASHPs performance with variable airflow rate of the outdoor fan under frosting conditions.

Abstract Liquid-solid contact electrification (CE) is essential to diverse applications. Exploiting its full implementation requires an in-depth understanding and fine-grained control of charge carriers (electrons and/or ions) during CE. Here, we decouple the electrons and ions during liquid-solid CE by designing binary superhydrophobic surfaces that eliminate liquid and ion residues on the surfaces and simultaneously enable us to regulate surface properties, namely work function, to control electron transfers. We find the existence of a linear relationship between the work function of superhydrophobic surfaces and the as-generated charges in liquids, implying that liquid-solid CE arises from electron transfer due to the work function difference between two contacting surfaces. We also rule out the possibility of ion transfer during CE occurring on superhydrophobic surfaces by proving the absence of ions on superhydrophobic surfaces after contact with ion-enriched acidic, alkaline, and salt liquids. Our findings stand in contrast to existing liquid-solid CE studies, and the new insights learned offer the potential to explore more applications.

Natural plant fibers are hierarchical structures with multi-level microstructures. With advances in composite material science, these fibers have been widely used in various polymer products. Therefore, it is crucial to quantitatively understand the relationship between their microstructures and mechanical behavior. This paper utilizes the Mori-Tanaka micromechanics model, viscoelasticity theory, and Zakian’s inversion method to study the impact of plant fiber microstructure on the viscoelastic behavior of multiscale structures. At the microscopic scale, the macromolecular polymer (matrix) and cellulose (fiber) are first homogenized. The second homogenization involves the cell wall microstructure, and the third homogenization considers the porosity of the cell wall and lumen to predict the effective modulus of fiber bundles. By applying the principle of elastic-viscoelastic correspondence, the viscoelastic mechanical parameters of plant fibers are calculated. The study examines the effects of cellulose crystallinity and lumen porosity on the structural stiffness and viscoelastic properties of fibers, identifying these factors as key influences on the mechanical behavior of plant fibers. Given their significant economic potential, the feasibility of using tobacco plant fibers as bio-based materials is also explored.

Faecalibacterium prausnitzii (F. prausnitzii) has been recognized for its various intestinal and extraintestinal benefits to human. And reduction of F. prausnitzii has been linked to an increased risk of intestinal fibrosis in patients of Crohn’s disease (CD). In this study, oral administration of either live F. prausnitzii or its extracellular vesicles (FEVs) can markedly mitigate the severity of fibrosis in mice induced by repetitive administration of DSS. In vitro experiment revealed that FEVs were capable of directing the polarization of peripheral blood mononuclear cells (PBMCs) towards an M2b macrophage phenotype, which has been associated with anti-fibrotic activities. This effect of FEV was found to be stable under various conditions that promote the development of pro-fibrotic M1/M2a/M2c macrophages. Proteomics and RNA sequencing were performed to uncover the molecular modulation of macrophages by FEVs. Notably, we found that FEVs reprogramed every metabolism of macrophages by damaging the mitochondria, and inhibited oxidative phosphorylation and glycolysis. Moreover, FEV-treated macrophages showed a decreased expression of PPARγ and an altered lipid processing phenotype characterized by decreased cholesterol efflux, which may promote energy reprogramming. Taken together, these findings identify FEV as a driver of macrophage reprogramming, suggesting that triggering M2b macrophage polarization by oral admiration of FEV may serve as strategy to alleviate hyperfibrotic intestine conditions in CD.

Resumo Fundamento A doença cardiovascular (DCV) é uma série de doenças que afetam o coração ou os vasos sanguíneos. Objetivos Avaliar a relação entre os níveis de pressão arterial (PA) definidos pelo American College of Cardiology/American Heart Association (ACC/AHA) de 2017 diretriz e risco de DCV/doença cardiovascular aterosclerótica (DCVA) para pessoas de meia-idade e idosos na China. Métodos Um total de 6.644 pessoas de meia-idade e idosas do Estudo Longitudinal de Saúde e Aposentadoria da China CHARLS (China Health and Retirement Longitudinal Study) foram finalmente incluídas. De acordo com a diretriz ACC/AHA de 2017, todos os indivíduos foram divididos em quatro grupos: PA normal, PA elevada, hipertensão estágio 1 e hipertensão estágio 2. O desfecho deste estudo foi considerado o risco de DCV e DCVA. Modelos de regressão COX univariados e multivariados foram adotados para examinar a relação da classificação de PA ACC/AHA de 2017 com o risco de DCV. Modelos de regressão logística univariada e multivariada foram utilizados para investigar a associação entre os níveis de PA e o risco de DCVA. Biomass and its derivatives have broad applications in the fields of bio-catalysis, energy storage, environmental remediation. The structure and components of biomass, which are vital parameters affecting corresponding performances of derived products, need to be fully understood for further regulating the biomass and its derivatives. Herein, tobacco is taken as an example of biomass to introduce the typical characterization techniques in unraveling the structural information, chemical components, and properties of biomass and its derivatives. Firstly, the structural information, chemical components and application for biomass are summarized. Then the characterization techniques together with the resultant structural information and chemical components are introduced. Finally, to promote a wide and deep study in this field, the perspectives and challenges concerning structure and composition charaterization in biomass and its derivatives are put forward.

Ground source heat pump (GSHP) systems have been widely used in the field of shallow geothermal heating and cooling because of their high thermal efficiency and environmental friendliness. A borehole heat exchanger (BHE) is the key part of a ground source heat pump system, and its performance and investment cost have a direct and significant impact on the performance and cost of the whole system. The ground temperature gradient, air temperature, seepage flow rate, and injection flow rate affect the heat exchange performance of BHEs, but most of the research on BHEs lacks field test verification. Therefore, this study relied on the results of a field thermal response test (TRT) based on a distributed optical fiber temperature sensor (DOFTS) and site hydrological, geological, and geothermal data to establish a corrected numerical model of buried pipe heat transfer and carry out the heat transfer performance analysis of a buried pipe in the heating season. The results showed that the ground temperature gradient of the test site was about 3.0 °C/100 m, and the temperature of the constant-temperature layer was about 9.17 °C. Increasing the air temperature could improve the heat transfer performance. The temperature of the surrounding rock and soil mass of the single pipe spread uniformly, and the closer it was to the buried pipe, the lower the temperature. When there is groundwater seepage, the seepage carries the cold energy generated by a buried pipe’s heat transfer through heat convection to form a plume zone, which can effectively alleviate the phenomenon of cold accumulation. With an increase in seepage velocity, the heat transfer of the buried pipe increases nonlinearly. The heat transfer performance can be improved by appropriately reducing the temperature and velocity of the injected fluid. Selecting a backfill material with higher thermal conductivity than the ground body can improve the heat transfer performance. These research results can provide support for the optimization of the heat transfer performance of a buried tube heat exchanger.

ABSTRACTThe present study aimed to characterize the gut microbiota and serum metabolome changes associated with sleep deprivation (SD) as well as to explore the potential benefits of multi-probiotic supplementation in alleviating SD-related mental health disorders. Rats were subjected to 7 days of SD, followed by 14 days of multi-probiotics or saline administration. Open-field tests were conducted at baseline, end of SD (day 7), and after 14 days of saline or multi-probiotic gavage (day 21). Metagenomic sequencing was conducted on fecal samples, and serum metabolites were measured by untargeted liquid chromatography tandem-mass spectrometry. At day 7, anxiety-like behaviors, including significant decreases in total movement distance (P = 0.0002) and staying time in the central zone (P = 0.021), were observed. In addition, increased levels of lipopolysaccharide (LPS; P = 0.028) and decreased levels of uridine (P = 0.018) and tryptophan (P = 0.01) were detected in rats after 7 days of SD. After SD, the richness of the gut bacterial community increased, and the levels of Akkermansia muciniphila, Muribaculum intestinale, and Bacteroides caecimuris decreased. The changes in the host metabolism and gut microbiota composition were strongly associated with the anxiety-like behaviors caused by SD. In addition, multi-probiotic supplementation for 14 days modestly improved the anxiety-like behaviors in SD rats but significantly reduced the serum level of LPS (P = 0.045). In conclusion, SD induces changes in the gut microbiota and serum metabolites, which may contribute to the development of chronic inflammatory responses and affect the gut-brain axis, causing anxiety-like behaviors. Probiotic supplementation significantly reduces serum LPS, which may alleviate the influence of chronic inflammation.IMPORTANCEThe disturbance in the gut microbiome and serum metabolome induced by SD may be involved in anxiety-like behaviors. Probiotic supplementation decreases serum levels of LPS, but this reduction may be insufficient for alleviating SD-induced anxiety-like behaviors.

The profens belong to a class of nonsteroidal anti-inflammatory drugs (NSAIDs), which exert significant anti-inflammatory, analgesic, antipyretic and other pharmacological effects. A considerable number of catalytic asymmetric strategies for the synthesis of enantioenriched profens have been introduced. Herein are outlined recent trends and developments of promising catalytic enantioselective systems for the generation of profens and their derivatives. According to the reaction type, we divided these transformations into three categories: Transition metal-catalyzed asymmetric hydrogenations, transition metal-catalyzed asymmetric cross-couplings and organocatalytic asymmetric transformations. Overviews of generic reaction mechanisms are presented. Ideally, this tutorial review will motivate further interest in the catalytic asymmetric synthesis of highly enantioenriched profens.

Abstract Background Effective risk prediction models are lacking for personalized endoscopic screening of gastric cancer (GC). We aimed to develop, validate, and evaluate a questionnaire-based GC risk assessment tool for risk prediction and stratification in the Chinese population. Methods In this three-stage multicenter study, we first selected eligible variables by Cox regression models and constructed a GC risk score (GCRS) based on regression coefficients in 416,343 subjects (aged 40–75 years) from the China Kadoorie Biobank (CKB, development cohort). In the same age range, we validated the GCRS effectiveness in 13,982 subjects from another independent Changzhou cohort (validation cohort) as well as in 5348 subjects from an endoscopy screening program in Yangzhou. Finally, we categorized participants into low (bottom 20%), intermediate (20–80%), and high risk (top 20%) groups by the GCRS distribution in the development cohort. Results The GCRS using 11 questionnaire-based variables demonstrated a Harrell’s C-index of 0.754 (95% CI, 0.745–0.762) and 0.736 (95% CI, 0.710–0.761) in the two cohorts, respectively. In the validation cohort, the 10-year risk was 0.34%, 1.05%, and 4.32% for individuals with a low (≤ 13.6), intermediate (13.7~30.6), and high (≥ 30.7) GCRS, respectively. In the endoscopic screening program, the detection rate of GC varied from 0.00% in low-GCRS individuals, 0.27% with intermediate GCRS, to 2.59% with high GCRS. A proportion of 81.6% of all GC cases was identified from the high-GCRS group, which represented 28.9% of all the screened participants. Conclusions The GCRS can be an effective risk assessment tool for tailored endoscopic screening of GC in China. Risk Evaluation for Stomach Cancer by Yourself (RESCUE), an online tool was developed to aid the use of GCRS.

Abstract Despite extensive astrocyte activation in patients suffering from HIV-associated neurocognitive disorders (HAND), little is known about the contribution of astrocytes to HAND neuropathology. Here, we report that the robust activation of neurotoxic astrocytes (A1 astrocytes) in the CNS promotes neuron damage and cognitive deficits in HIV-1 gp120 transgenic mice. Notably, knockout of α7 nicotinic acetylcholine receptors (α7nAChR) blunted A1 astrocyte responses, ultimately facilitating neuronal and cognitive improvement in the gp120tg mice. Furthermore, we provide evidence that Kynurenic acid (KYNA), a tryptophan metabolite with α7nAChR inhibitory properties, attenuates gp120-induced A1 astrocyte formation through the blockade of α7nAChR/JAK2/STAT3 signaling activation. Meanwhile, compared with gp120tg mice, mice fed with tryptophan showed dramatic improvement in cognitive performance, which was related to the inhibition of A1 astrocyte responses. These initial and determinant findings mark a turning point in our understanding of the role of α7nAChR in gp120-mediated A1 astrocyte activation, opening up new opportunities to control neurotoxic astrocyte generation through KYNA and tryptophan administration.

Background: By observing the changes of lung imaging airway structure in patients with advanced lung squamous cell carcinoma（ALUSC）, the relationship between the different types of COPD pulmonary structural remodeling and the prognosis of patients with ALUSC was analyzed. Methods: We reviewed the medical records of 278 patients with ALUSC. The degree of emphysema and the percentage of bronchial wall thickness（WT%） on chest HRCT were calculated by Synapse3D software, Lung structural remodeling can be divided into there types: airway remodeling dominated, emphysema dominated, and mixed types. Results: Compared with the diagnosis, the Goddard score increased, the proportion of airway remodeling dominated type decreased and the proportion of mixed type increased during the progression of ALUSC. In Kaplan-Meier analysis, whether with or without COPD, the mPFS and mOS of patients with mixed type were the shortest, and the difference was statistically significant. Univariate and multivariate Cox proportional hazard regression analysis showed that mixed type was an independent risk factor for poor PFS and OS in patients with ALUSC. Conclusion: Patients with ALUSC all have varying degrees of lung structural remodeling, and patients with mixed lung structural remodeling have the worst prognosis. In addition, the aggravation of emphysema during tumor progression is more pronounced than the thickening of the airway wall, and the changes of emphysema was more powerful in predicting the progression of ALUSC.Clinicians must pay more attention to the changes of COPD (especially emphysema) in the process of diagnosis and treatment of ALUSC.

''Gannan navel orange'' was used as the test fruit, the effects of the solution of Heracleum hemsleyanum Diels extracts，carboxymethyl cellulose sodium/polyvinyl pyrrolidone and γ-aminobutyric acid with different concentrations on citrus fruit fresh-keeping were studied by response surface methodology with decay rate, weight loss rate and the content of ascorbic acid as evaluation parameters. And then the best formula was determined and its fresh-keeping effects on citrus were explored. The optimal formulation was determined as follow: The concentration of Heracleum hemsleyanum Diels extracts, carboxymethyl cellulose sodium/polyvinyl pyrrolidone and γ-aminobutyric acid were 2.0%, 0.6%/0.4% and 1.0%, respectively. The experimental results of the fresh-keeping effect test can be obtained：Compared with the control group, adding the Heracleum hemsleyanum Diels extracts could significantly reduce the decay rate, delay the decrease of fruit hardness, and keep the activities of superoxide dismutase and β-1,3-glucanase, reduce the content of malondialdehyde. The composite coating could prevent the loss of water in citrus fruits, and keep soluble solids, titratable acid, ascorbic acid and total sugar content, and maintain fruit flavor. The combination of the two components to prepare composite coating preservative could play a role of effect superposition, the comprehensive fresh-keeping effect is better than the two alone.

BackgroundMorus alba L. (Sangzhi) alkaloid (SZ-A) is a new antidiabetic drug approved by the China National Medical Products Administration in 2020. Diabetic nephropathy (DN) is a common diabetic complication and an important cause of morbidity and mortality in patients with diabetes. The effects of SZ-A on DN remain unknown.PurposeThis study evaluated the effects of SZ-A on DN in Zucker diabetic fatty (ZDF) rats and explored the underlying mechanisms based on nitrosative stress, inflammation, and fibrosis.MethodsDiabetic ZDF rats were orally administered 100 and 200 mg/kg of SZ-A once daily for 9 weeks. The glucose metabolism and kidney function were assayed. The pathological injury and fibrosis of the kidneys were separately evaluated using hematoxylin and eosin staining and Masson’s staining. The oxidative and nitrosative stress and inflammation were assayed by determining the levels of related indices in the blood and kidneys and quantifying the related gene and protein expression. The expression of transforming growth factor β1 (TGFβ1) gene and protein were assayed by quantitative real-time PCR and immunohistochemistry, respectively. The renal transcriptomics was analyzed using RNA sequencing.ResultsRepeated treatment with SZ-A significantly improved glucose metabolism, dose-dependently decreased the levels of blood urea nitrogen, urinary albumin, and β2-microglobulin, and evidently relieved the renal injury in diabetic ZDF rats. As for the mechanisms, SZ-A remarkably ameliorated systemic nitrosative stress through lowering the levels of blood inducible nitric oxide synthase and nitric oxide, and significantly relieved systemic and renal inflammation by reducing the levels of blood interleukin-1β and monocyte chemoattractant protein-1 (MCP-1) and decreasing the levels of renal C-reactive protein content and expression of tumor necrosis factor-α in the kidneys. SZ-A also improved renal fibrosis by lowering the expression of TGFβ1 in the kidneys. Additionally, SZ-A significantly lowered the expression of stimulator of chondrogenesis 1 in the kidneys.ConclusionRepeated treatments with SZ-A significantly ameliorates DN by regulating systemic nitrosative stress, renal inflammation, and renal fibrosis partially through inhibition of the cytokine-NO and TGF-β1 signaling in ZDF rats, providing evidence for the additional application of SZ-A in clinical use for the treatment of DN.

Abstract Background Colorectal cancer (CRC) with pulmonary metastasis usually indicates a poor prognosis, whereas patients may benefit from adoptive cell therapy. Tumor-specific cytotoxic T lymphocytes (CTLs) have been reported as a promising treatment for CRC. However, the antitumor effect of CTLs remains limited partially due to insufficient production of effector cells via the activation by antigen-presenting dendritic cells (DCs). Method This study showed that a combination of CD40 mAb and Picibanil (OK-432) could significantly enhance the activation of CTLs by DCs, both in vitro and in vivo. Flow cytometry, colon cancer mouse model, and pathological staining were employed to demonstrate the specific functions. Results This approach promoted the maturation of DCs, augmented the production of stimulatory cytokines, and suppressed the secretion of inhibitory cytokines. Additionally, it facilitated the killing efficiency of CTLs via stimulating their proliferation while restraining the number of Tregs, concomitantly with the positive regulation of corresponding cytokines. Furthermore, the combined unit could hurdle the expansion of tumor cells on metastatic lungs in the colon cancer mouse model. Conclusion Collectively, the combination of CD40-mAb and OK-432 facilitated the maturation of DCs and enhanced the cytotoxicity of T cells, promising therapeutic approach against CRC. Objective: To study the anti-fatigue effect of irradiated Inonotus obliquus polysaccharide on mice. Method: 60Co-γ rays were irradiated to obtain 5, 10, 20, 30 kGy irradiation doses and unirradiated Inonotus obliquus bacteria powder to produce polysaccharides. Divided into a control group, an irradiated high-medium-low-dose group, and a non-irradiated high-dose group. Male Kunming mice were given four weeks continuous gavage to determine the rod climbing, exhausted swimming time, changes in liver glycogen(LG), muscle glycogen(MG), blood lactate(BLA), lactate dehydrogenase(LDH), serum urea nitrogen(SUN), superoxide dismutase(SOD) and malondialdehyde(MDA) in skeletal muscle. Result: The optimal radiation dose measured in the experiment was 20 kGy. Each dose group had no significant effect on the body weight of the mice(P>0.05). The high and medium dose groups and the non-irradiated groups significantly prolonged the rod climbing time and exhausted swimming time of the mice(P0.05). Conclusion: This test proves that the anti-fatigue effect of the irradiated Inonotus obliquus polysaccharide is further improved compared with the non-irradiated group, and that the irradiated Inonotus obliquus polysaccharide can relieve fatigue symptoms to a certain extent.

The natural gas hydrate resources in the South China Sea alone are about 85 trillion cubic meters. In the drilling process of marine gas hydrate, the natural gas hydrate will decompose and produce gas, as the rising of temperature and dropping of the pressure in the annulus. This process will have a significant impact on drilling safety. Therefore, it is necessary to study the wellbore temperature distribution during the drilling of marine hydrate layer. In this paper, the wellbore temperature distribution of safe drilling in hydrated formation is taken as the research goal, and the research status of relevant domestic and international wellbore temperatures was investigated. According to the characteristics of the marine environment and reservoir-forming characteristics of hydrate reservoirs in the South China Sea, the wellbore temperature distribution model of offshore drilling wellbore under the condition of hydrate decomposition was established. The temperature distribution curve of drilling straight wellbore in hydrate layer of South China Sea was obtained. When drilling the hydrate reservoir, the distribution regularity of the wellbore temperature is similar to that of the conventional offshore drilling wellbore. However, the temperature of the wellbore annulus near the hydrate decomposition site is lower than the ambient temperature, mainly due to the hydrate decomposition endothermic. In this paper, the sensitivity analysis of several main parameters of the wellbore temperature distribution of drilling straight wellbore in hydrate layer of South China Sea was carried out. Through the conduction of experiment and numerical simulation, we have get some new findings: (1) The hydrate saturation has little effect on the wellbore temperature; (2) As the drilling fluid displacement increases, the annulus temperature of the wellbore above the mudline increases, and the temperature of the wellbore below the mudline decreases continuously; (3) As the density of the drilling fluid increases, the temperature at the wellhead decreases, and the temperature at the bottom of the well increases slightly; (4) The greater the rate of penetration of the well, the temperature at the upper part of the wellbore decreases, and the temperature at the bottom of the wellbore increases; Among them, the penetration rate has the most obvious effect on the annular temperature. The results are expected to be helpful to guide the drilling process of marine gas hydrate and offer some references.

Abstract Introduction Diosgenin is a natural steroidal compound with reported antidiabetic and many other protective properties. This study aimed to explore the protective effect of diosgenin on high‐glucose (HG)‐induced retinal pigment epithelial cells. Methods HG‐induced ARPE‐19 cells were considered as a cell model of diabetic retinopathy (DR). The viability and apoptosis of ARPE‐19 cells induced by HG treated with either diosgenin or Compound C (CC; dorsomorphin) were detected by Cell Counting Kit‐8 assay and flow cytometric analysis. The expression of apoptosis‐related proteins, inflammation‐related proteins, and AMPK/Nrf2/HO‐1 pathway‐related proteins was detected by western blotting. The levels of inflammatory cytokines and detection of oxidative stress indexes were performed using the appropriate assay kits. The messenger RNA expression of inflammatory cytokines was detected by real‐time quantitative polymerase chain reaction. Results There was no obvious effect of diosgenin on the viability of ARPE‐19 cells and the viability of ARPE‐19 cells was significantly reduced after HG induction. However, diosgenin increased the viability, inhibited the apoptosis, and reduced the inflammatory response and oxidative stress of ARPE‐19 cells induced by HG. In addition, diosgenin could activate the AMPK/Nrf2/HO‐1 pathway. CC, an AMPK inhibitor, could reverse the above changes caused by diosgenin treatment in ARPE‐19 cells induced by HG. Conclusions Diosgenin could protect ARPE‐19 cells from inflammatory damage and oxidative stress induced by HG, by activating the AMPK/Nrf2/HO‐1 pathway.

Abstract Background Osteoarthritis (OA) is responsible for the impotent disability in old people. Circular RNA (circRNA) has been reported to be related to the development of diseases. The lack of research on the role of circRNA spastic paraplegia 11 (circ-SPG11) results in conducting this study. Methods The expression of circ-SPG11, microRNA-337-3p (miR-337-3p), and aggrecanases like a disintegrin and metalloproteinase with thrombospondin motifs 5 (ADAMTS5) mRNA was detected by quantitative real-time polymerase chain reaction (qRT-PCR). Western blot was used to measure the protein expression of extracellular matrix (ECM) degradation-related markers and ADAMTS5. Ribonuclease R (RNase R) was applied to test the stability of circ-SPG11 in CHON-001 cells. The viability, apoptosis, TNF-α and IL-6 production were determined by cell counting kit-8 (CCK-8) assay, flow cytometry assay, and enzyme-linked immunosorbent assay (ELISA), respectively. Meanwhile, the interaction between miR-337-3p and circ-SPG11 or ADAMTS5 was respectively predicted by Circinteractome or Starbase2.0, which was further verified by dual-luciferase reporter system and RNA binding protein immunoprecipitation (RIP) assay. Results Circ-SPG11 and ADAMTS5 were upregulated and miR-337-3p was downregulated in OA tissues and OA model cells. Circ-SPG11 knockdown allayed interleukin 1β (IL-1β)-induced restraint in viability and promotion in apoptosis, TNF-α, and IL-6 generation and ECM degradation in CHON-001 cells. Anti-miR-337-3p or ADAMTS5 overexpression correspondingly reversed si-circ-SPG11 or miR-337-3p overexpression-mediated facilitation in viability, and inhibition in apoptosis, TNF-α and IL-6 generation and ECM degradation in OA model cells. Moreover, anti-miR-337-3p ameliorated si-circ-SPG11-mediated inhibition in ADAMTS5 mRNA and protein expression in OA model cells. Conclusion Circ-SPG11 facilitated OA development via regulating miR-337-3p/ADAMTS5 axis. This finding might contribute to the improvement of OA therapy.

As the penetration rate of variable renewable energy such as wind power increases in the power system, the composition and balance of the system also change gradually. The intermittency of renewable energy poses great stability challenges to the traditional centralized generation and load-oriented transmission and distribution methods. Therefore, the Active Distribution Network Operator (ADNO) with distributed installation at the local level has a good application prospect in the new scenario. However, ADNO needs to improve its operational efficiency based on the types of local generation and storage devices and the nature of the market environment. To address this issue, this paper proposes a forecasting method that considers the coupling fluctuations of spot electricity prices and renewable energy, and a bi-level optimization operation method based on the Stackelberg game for optimizing the operation of small-scale ADNO under high wind power penetration rate. Simulation results show that the proposed methods achieve greater positive impact on the operational efficiency of ADNO than conventional methods. In addition, the proposed methods ensure the long-term profitability of ADNO, even with fluctuations in external factors.

Abstract Background The fecal microbiota in pancreatic ductal adenocarcinoma (PDAC) and in autoimmune pancreatitis (AIP) patients remains largely unknown. We aimed to characterize the fecal microbiota in patients with PDAC and AIP, and explore the possibility of fecal microbial biomarkers for distinguishing PDAC and AIP. Methods 32 patients with PDAC, 32 patients with AIP and 32 age- and sex-matched healthy controls (HC) were recruited and the fecal microbiotas were analyzed through high-throughput metagenomic sequencing. Alterations of fecal short-chain fatty acids were measured using gas chromatographic method. Results Principal coordinate analysis (PCoA) revealed that microbial compositions differed significantly between PDAC and HC samples; whereas, AIP and HC individuals tended to cluster together. Significant reduction of phylum Firmicutes (especially butyrate-producing bacteria, including Eubacterium rectale, Faecalibacterium prausnitzii and Roseburia intestinalis) and significant increase of phylum Proteobacteria (especially Gammaproteobacteria) were observed only among PDAC samples. At species level, when compared with HC samples, we revealed 24 and 12 differently enriched bacteria in PDAC and AIP, respectively. Functional analysis showed a depletion of short-chain fatty acids synthesis associated KO modules (e.g. Wood-Ljungdahl pathway) and an increase of KO modules associated with bacterial virulence (e.g. type II general secretion pathway). Consistent with the downregulation of butyrate-producing bacteria, gas chromatographic analysis showed fecal butyrate content was significantly decreased in PDAC group. Eubacterium rectale, Eubacterium ventrisum and Odoribacter splanchnicus were among the most important biomarkers in distinguishing PDAC from HC and from AIP individuals. Receiver Operating Characteristic analysis showed areas under the curve of 90.74% (95% confidence interval [CI] 86.47–100%), 88.89% (95% CI 73.49–100%), and 76.54% (95% CI 52.5–100%) for PDAC/HC, PDAC/AIP and AIP/HC, respectively. Conclusions In conclusion, alterations in fecal microbiota and butyrate of patients with PDAC suggest an underlying role of gut microbiota for the pathogenesis of PDAC. Fecal microbial and butyrate as potential biomarkers may facilitate to distinguish patients with PDAC from patients with AIP and HCs which worth further validation.

Abstract Aberrant DNA methylation is often related to the diagnosis, prognosis, and therapeutic response of acute myeloid leukemia (AML); however, relevant studies on the relationship between bone marrow myeloblast percentage and the DNA methylation level in AML have not been reported. We evaluated the effects of AML blast percentage on DNA methylation level using the MethylC-capture sequencing (MCC-Seq) approach based on next-generation sequencing (NGS) and found that the methylation level of both genome-wide and promoter regions significantly increased when the percentage of AML blasts reached ≥ 40%, indicating that an accurate DNA methylation level in cancer cells can be obtained when the bone marrow samples of AML patients have more than 40% myeloblasts.

The present study aimed to explore the effects of α-cyperone, an extract of Cyperus rotundus, on PC12 cells, as well as the underlying mechanism. Following the cells were induced by oxygen-glucose deprivation/reoxygena-tion (OGD/R), the viability, morphology, inflammation, oxidative stress and apoptotic levels in the cells were evaluated. To explore the mechanism of α-cyperone, cells were treated with 3-TYP, a sirtuin-3 (SIRT3) inhibitor, and then the effects of 3-TYP on the function of α-cyperone were assessed. α-Cyperone was found to reduce OGD/R-induced damage to neuronal viability and alleviate inflammation, oxidative stress, and apoptosis. In addition, α-cyperone could elevate SIRT3 and decline acetyl-forkhead box O1 (FOXO1) levels, and 3-TYP broke the effects of α-cyperone on the aforementioned aspects in the PC12 cells. In conclusion, α-cyperone activated SIRT3 and FOXO1 deacetylation, and alleviated OGD/R-induced cell inflammation, oxidative stress, and apoptosis.

Abstract Background Rho GTPase activating protein 9 (ARHGAP9) is expressed in various types of cancers and can inactivate Rho GTPases that mainly regulate cytoskeletal dynamics. However, the exact role of ARHGAP9 in acute myeloid leukemia (AML) has yet to be clarified. Methods We compared the transcriptional expression, prognosis, differentially expressed genes, functional enrichment, and hub genes in AML patients on the basis of the data published in the following databases: UALCAN, GEPIA, Gene Expression Omnibus, the Human Protein Atlas, Cancer Cell Line Encyclopedia, LinkedOmics, Metascape, and String. Data from the Cancer Genome Atlas database was used to evaluate the correlations between ARHGAP9 expression and various clinicopathological parameters, as well as the significantly different genes associated with ARHGAP9 expression. Results We found that ARHGAP9 expression was higher in the tissues and cell lines extracted from patients with AML than corresponding control tissues and other cancer types. ARHGAP9 overexpression was associated with decreased overall survival (OS) in AML. Compared with the ARHGAP9low group, the ARHGAP9high group, which received only chemotherapy, showed significantly worse OS and event-free survival (EFS); however, no significant difference was observed after treatment with autologous or allogeneic hematopoietic stem cell transplantation (auto/allo-HSCT). The ARHGAP9high patients undergoing auto/allo-HSCT also had a significantly better prognosis with respect to OS and EFS than those receiving only chemotherapy. Most overlapping genes of the significantly different genes and co-expression genes exhibited enriched immune functions, suggesting the immune regulation potential of ARHGAP9 in AML. A total of 32 hub genes were identified from the differentially expressed genes, within which the KIF20A had a significant prognostic value for AML. Conclusions ARHGAP9 overexpression was associated with poor OS in AML patients and can be used as a prognostic biomarker. AML patients with ARHGAP9 overexpression can benefit from auto/allo-HSCT rather than chemotherapy.
