Background/purpose: Recurrent aphthous stomatitis (RAS) is a common oral mucosal disease. Despite a variety of scientific articles have been available till date, merely a few scientometric analyses have been systematically carried out in this field. The objective of this study was to recognize the hotspots and research trends related to RAS via bibliometric approach. Materials and methods: The Elsevier's Scopus database was searched to retrieve qualified literature through an advanced search strategy on 9 Feb 2023. The basic information was collected as following: article type, publication year, journals, impact factor, the count of citations, citation density, keywords, authors, contributing institutions and country. Results: A list of 986 publications were identified from 1933 to 2022, and the number of citations for each paper varied from 0 to 283. A steady increasing trend in the number of documents could be observed each decade with the summit in 2010s. Controlled study (n = 334) and major clinical study (n = 192) were the most common types of study design. Scully C (n = 26) was identified as the most productive author. United States (n = 166) and Turkey (n = 101) top the list of dedicating countries. Conclusion: This report would offer profound insight into the current status of RAS research and serve as a reference source for anyone planning to enhance the quality of future work.

Abstract To evaluate the predictive and prognostic value of fibroblast growth factor 21 (FGF21) levels in retinal artery occlusion (RAO) patients. In this case–control study, serum FGF21 levels were detected by using the ELISA method. Multivariable logistic regression analyses were performed to evaluate the significance of FGF21 in assessing the risk of developing RAO and its impact on vision and concurrent ischemic stroke. Compared with control group, serum FGF21 levels were significantly higher (median [IQR] = 230.90[167.40,332.20] pg/ml) in RAO patients. Multivariate logistic regression analysis showed that elevated serum FGF21 levels were associated with a higher risk of RAO occurrence (P = 0.025, OR [95%CI] = 9.672 [2.573, 36.359]) after adjustment for multiple confounding factors. Higher serum FGF21 levels were negatively associated with visual acuity improvement (P = 0.029, OR [95%CI] = 0.466[0.235, 0.925]) and positively correlated with concurrent ischemic stroke (P = 0.04, OR [95% CI] = 1.944[1.029, 3.672]) in RAO patients. Elevated serum FGF21 levels could promote the development of RAO and indicate worse visual prognosis and increase the risk of concurrent ischemic stroke, which might help clinicians early diagnose and treat RAO patients.

Abstract Clinical research has suggested that chronic HBV infection exerts a certain effect on the occurrence of cardiovascular disease by regulating cholesterol metabolism in liver cells. High serum apolipoprotein B/apolipoprotein A1 (ApoB/ApoA1) ratio plays a certain role in the above regulation, and it serves as a risk factor for cardiovascular disease. However, whether the ApoB/ApoA1 ratio is correlated with chronic HBV infection and its disease progression remains unclear. In accordance with the inclusion and exclusion criteria, all 378 participants administrated at Renmin Hospital of Wuhan University from March 2021 to March 2022, fell into Healthy Control (HC) group (50 participants), Hepatocellular carcinoma (HCC) group (107 patients), liver cirrhosis (LC) group (64 patients), chronic hepatitis B (CHB) group (62 patients), chronic hepatitis C (CHC) group (46 patients) and Hepatitis E Virus (HEV) group (49 patients). Serum ApoA1 and ApoB concentrations were measured at admission, and the ApoB/ApoA1 ratio was determined. The levels of laboratory parameters in the respective group were compared and ApoB/ApoA1 ratios in HCC patients and LC patients with different severity were further analyzed. ROC curves were plotted to analyze the early diagnostic ability of ApoB/ApoA1 ratio for HBV-associated HCC. Logistic regression and restricted cubic spline analysis were used to explore the correlation between ApoB/ApoA1 ratio and LC and HCC risk. Compelling evidence indicates that dyslipidemia is positively associated with oral lichen planus (OLP). The types and magnitude of lipid metabolism disturbance in peripheral blood of OLP patients have been investigated in different studies. Yet, consensus on how these different lipid components varied in levels for the development of OLP lesions has not been reached so far. Herein, a total of 8 eligible studies were recognized, which enrolled 533 cases of OLP and 499 healthy controls. The analysis showed that the average total triglyceride (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C) levels were considerably higher, and high-density lipoprotein cholesterol (HDL-C) levels were significantly lower in OLP patients compared to healthy controls. Collectively, the lipid profile panel maybe serve as the potential predictive indicator for screening OLP.

Glioblastoma multiforme (GBM) is a formidable cancer to treat due to the lack of effective drugs that can also efficiently cross the blood–brain barrier (BBB). Herein, a novel strategy involving the synthesis of p28 peptide‐conjugated, lexiscan (LEX)‐loaded, bardoxolone methyl (BM) self‐assembled nanoparticles, designated as p28‐LBM NPs, is introduced. These NPs are designed to overcome the dual challenges of effectively killing GBM cells and efficiently penetrating the brain. The p28 peptide is chosen for targeted delivery to brain tumors, and LEX is employed to enhance drug penetration across the BBB. The successful penetration of brain tumors by the p28‐LBM NPs after intravenous administration is demonstrated, with BM delivered as part of the NPs significantly inhibiting GBM tumor growth and extending the survival of mice with tumors. In conclusion, the p28‐LBM NPs present a promising approach for GBM treatment, with potential for effective and safe clinical applications in the future.

Abstract Central retinal artery occlusion (CRAO) is an acute retinal ischaemic disease, but early diagnosis is challenging due to a lack of biomarkers. Blood samples were collected from CRAO patients and cataract patients. Gene expression profiles were distinct between arterial/venous CRAO blood (A–V group) and venous CRAO/control blood (V–C group) samples. Differentially expressed genes (DEGs) were subjected to GO and KEGG enrichment analyses. Hub genes were identified by Cytoscape and used to predict gene interactions via GeneMANIA. Immune cell infiltration was analysed by CIBERSORT. More than 1400 DEGs were identified in the A–V group and 112 DEGs in the V–C group compared to controls. The DEGs in both groups were enriched in the ribosome pathway, and those in the V–C group were also enriched in antigen processing/MHC pathways. Network analysis identified ribosomal proteins (RPS2 and RPS5) as the core genes of the A–V group and MHC genes (HLA-F) as the core genes of the V–C group. Coexpression networks showed ribosomal involvement in both groups, with additional immune responses in the V–C group. Immune cell analysis indicated increased numbers of neutrophils and T cells. Ribosomal and MHC-related genes were identified as potential CRAO biomarkers, providing research directions for prevention, diagnosis, treatment and prognosis.

Abstract Inertial Measurement Unit-based methods have great potential in capturing motion in large-scale and complex environments with many people. Sparse Inertial Measurement Unit-based methods have more research value due to their simplicity and flexibility. However, improving the computational efficiency and reducing latency in such methods are challenging. In this paper, we propose Fast Inertial Poser, which is a full body motion estimation deep neural network based on 6 inertial measurement units considering body parameters. We design a network architecture based on recurrent neural networks according to the kinematics tree. This method introduces human body shape information by the causality of observations and eliminates the dependence on future frames. During the estimation of joint positions, the upper body and lower body are estimated using separate network modules independently. Then the joint rotation is obtained through a well-designed single-frame kinematics inverse solver. Experiments show that the method can greatly improve the inference speed and reduce the latency while ensuring the reconstruction accuracy compared with previous methods. Fast Inertial Poser runs at 65 fps with 15 ms latency on an embedded computer, demonstrating the efficiency of the model.

Abstract Background Preoperative prediction of International Federation of Gynecology and Obstetrics (FIGO) stage in patients with epithelial ovarian cancer (EOC) is crucial for determining appropriate treatment strategy. This study aimed to explore the value of contrast-enhanced CT (CECT) radiomics in predicting preoperative FIGO staging of EOC, and to validate the stability of the model through an independent external dataset. Methods A total of 201 EOC patients from three centers, divided into a training cohort (n = 106), internal (n = 46) and external (n = 49) validation cohorts. The least absolute shrinkage and selection operator (LASSO) regression algorithm was used for screening radiomics features. Five machine learning algorithms, namely logistic regression, support vector machine, random forest, light gradient boosting machine (LightGBM), and decision tree, were utilized in developing the radiomics model. The optimal performing algorithm was selected to establish the radiomics model, clinical model, and the combined model. The diagnostic performances of the models were evaluated through receiver operating characteristic analysis, and the comparison of the area under curves (AUCs) were conducted using the Delong test or F-test. Results Seven optimal radiomics features were retained by the LASSO algorithm. The five radiomics models demonstrate that the LightGBM model exhibits notable prediction efficiency and robustness, as evidenced by AUCs of 0.83 in the training cohort, 0.80 in the internal validation cohort, and 0.68 in the external validation cohort. The multivariate logistic regression analysis indicated that carcinoma antigen 125 and tumor location were identified as independent predictors for the FIGO staging of EOC. The combined model exhibited best diagnostic efficiency, with AUCs of 0.95 in the training cohort, 0.83 in the internal validation cohort, and 0.79 in the external validation cohort. Abstract Handheld robots offer accessible solutions with a short learning curve to enhance operator capabilities. However, their controllable degree-of-freedoms are limited due to scarce space for actuators. Inspired by muscle movements stimulated by nerves, we report a handheld time-share driven robot. It comprises several motion modules, all powered by a single motor. Shape memory alloy (SMA) wires, acting as “nerves”, connect to motion modules, enabling the selection of the activated module. The robot contains a 202-gram motor base and a 0.8 cm diameter manipulator comprised of sequentially linked bending modules (BM). The manipulator can be tailored in length and integrated with various instruments in situ, facilitating non-invasive access and high-dexterous operation at remote surgical sites. The applicability was demonstrated in clinical scenarios, where a surgeon held the robot to conduct transluminal experiments on a human stomach model and an ex vivo porcine stomach. The time-share driven mechanism offers a pragmatic approach to build a multi-degree-of-freedom robot for broader applications.

ContextDespite the recognition of attention deficit hyperactivity disorder (ADHD) as a multifaceted neurodevelopmental disorder, its core causes are still ambiguous. The objective of this study was to explore if the traits of circulating immune cells contribute causally to susceptibility to ADHD.MethodsBy employing a unified GWAS summary data covering 731 immune traits from the GWAS Catalog (accession numbers from GCST0001391 to GCST0002121), our analysis focused on the flow cytometry of lymphocyte clusters, encompassing 3,757 Sardinians, to identify genetically expected immune cells. Furthermore, we obtained summarized GWAS statistics from the Psychiatric Genomics Consortium to evaluate the genetic forecasting of ADHD. The studies employed ADHD2019 (20,183 cases and 35,191 controls from the 2019 GWAS ADHD dataset) and ADHD2022 (38,691 cases and 275,986 controls from the 2022 GWAS ADHD dataset). Through the examination of genome-wide association signals, we identified shared genetic variances between circulating immune cells and ADHD, employing the comprehensive ADHD2022 dataset. We primarily utilized inverse variance weighted (IVW) and weighted median methods in our Mendelian randomization research and sensitivity assessments to evaluate diversity and pleiotropy.ResultsAfter adjusting for false discovery rate (FDR), three distinct immunophenotypes were identified as associated with the risk of ADHD: CD33 in Im MDSC (OR=1.03, CI: 1.01~1.04, P=3.04×10−5, PFDR=0.015), CD8br NKT %T cell (OR=1.08, 95%CI: 1.04~1.12, P=9.33×10−5, PFDR=0.023), and CD8br NKT %lymphocyte (OR=1.08, 95%CI: 1.03~1.12, P=3.59×10−4, PFDR=0.066). Furthermore, ADHD showed no statistical effects on immunophenotypes. It’s worth noting that 20 phenotypes exist where ADHD’s appearance could diminish 85% of immune cells, including FSC-A in myeloid DC (β= -0.278, 95% CI: 0.616~0.931, P=0.008), CD3 in CD45RA- CD4+ (β= -0.233, 95% CI: 0.654~0.960, P=0.017), CD62L- monocyte AC (β=0.227, 95% CI: 0.038~1.518, P=0.019), CD33 in CD33br HLA DR+ CD14dim (β= -0.331, 95% CI: 0.543~0.950, P=0.020), and CD25 in CD39+ resting Treg (β=0.226, 95% CI: 1.522, P=0.022), and FSC-A in monocytes (β= -0.255, 95% CI: 0.621~0.967, P=0.234), among others.ConclusionStudies indicate that the immune system’s response influences the emergence of ADHD. The findings greatly improve our understanding of the interplay between immune responses and ADHD risk, aiding in the development of treatment strategies from an immunological perspective.

ObjectiveTo describe the return to work of patients with severe acute pancreatitis within 6 months after discharge, and to explore the influence of demographic, clinical, and psychosocial factors on their return to work.Research designProspective 6 months follow-up study.SettingA third class hospital in Guizhou Province. Adult of severe acute pancreatitis(18-60years), with a job before admission, in the intensive care unit ≥ 24 h, were included.Main outcome measuresTo study return to work and influencing factors one, three and six months severe acute pancreatitis patients discharge. several measurements were used, including General Health Questionnaire (Demographic, disease-related, job-related and health behavior data), Readiness for Return-To-Work Scale and the Hospital Anxiety and Depression Scale.ResultsForty-three severe acute pancreatitis patients were included in our study, with mean age 41.53 years. Twenty-nine (67.44%) patients returned to work within 6 months, and fourteen patients did not return to work. The status of Readiness for Return-To-Work Scale: fourteen severe acute pancreatitis patients who did not return to work were mainly in the precontemplation dimension and prepared for action-self-evaluative dimension both 5 cases (35.71%), and the 29 patients who had returned to work were in the Proactive maintenance stage. Altered cholesterol metabolism has been identified as a critical feature of cancers. Cholesterol functions as the main component of cell membrane, cholesterol and is required for sustaining membrane integrity and mediating signaling transduction for cell survival. The intracellular level of cholesterol is dynamically regulated. Excessive cholesterol could be converted to less toxic cholesteryl esters by acyl-coenzyme A:cholesterol acyltransferases (ACATs). While ACAT2 has limited value in cancers, ACAT1 has been found to be widely participated in tumor initiation and progression. Moreover, due to the important role of cholesterol metabolism in immune function, ACAT1 is also essential for regulating anti-tumor immunity. ACAT1 inhibition may be exploited as a potential strategy to enhance the anti-tumor immunity and eliminate tumors. Herein, a comprehensive understanding of the role of ACAT1 in tumor development and anti-tumor immunity may provide new insights for anti-tumor strategies.

PurposeTo characterize features of central retinal artery occlusion (CRAO) using multicolor (MC) imaging and to assess the differences in CRAO grading between color fundus photography (CFP) and MC image qualitatively and quantitatively.MethodsWe conducted a prospective, cross-sectional study in the Department of Ophthalmology of Renmin Hospital of Wuhan University. In total, 86 acute CRAO patients were included. Spectral-domain optical coherence tomography (SD-OCT), CFP, and MC examinations were taken at baseline. Based on the findings of these three examinations, CRAO was divided into three grades (incomplete, subtotal, and total). Based on OCT grading criteria, we qualitatively compared the ability of grading CRAO by CFP and MC. CRAO patient's visual acuity (VA) was obtained from the initial visit. The retinal thickness was measured by SD-OCT. Superficial capillary plexus (SCP) and deep capillary plexus (DCP) were obtained from optical coherence tomography angiography (OCTA) examinations. Quantitative data were compared across the three acute CRAO subgroups and against three examination findings.ResultsMC image had significantly higher power of acute CRAO detection than CFP (P = 0.03). In the same group of CRAO patients, there was no significant difference in VA when comparing OCT with the MC grading system or with the CFP grading system (all P > 0.05). Significant differences in VA were found between the three CRAO subgroups only under MC grading (P = 0.016). In incomplete CRAO patients, significant differences were found in central fovea thickness (CFT) when comparing OCT with the CFP grading system (P = 0.019). In the same group of CRAO patients, there was no significant difference in retinal thickness when comparing OCT with the MC grading system (All P > 0.05). Significance differences in CFT (P < 0.001), innermost retinal layer (IMRL; P < 0.01), middle retinal layer (MRL; P < 0.001), and outer retinal layer (ORL; P = 0.021) were found between the three CRAO subgroups by MC grading. Vessel density of SCP showed a statistically increased as the severity of three CRAO subgroups (P = 0.03), whereas DCP did not have significant differences (P = 0.745). Comparisons were made between the OCT grading method and the MC and CFP grading methods; there is no significant difference in vessel density of SCP and DCP (All P > 0.05).ConclusionThe images obtained by MC are superior to those obtained by CFP in CRAO grading, retinal thickness, and vessel density measurement. MC imaging may be more capable of CRAO grading than OCT. We recommend MC imaging to determine CRAO severity to guide disease treatment and predict visual prognosis.

Single-phase body-centered cubic (BCC) lightweight Ti–V–Nb–Zrx solid-solution alloys were fabricated by reducing the Zr concentration within the range of 0–5 at% to improve the mechanical properties of equimolar TiVNbZr alloy at elevated temperatures. The fabricated Zrx alloys indeed exhibited greater strength above 800 °C than TiVNbZr, but some special mechanical responses occurred at 800 °C: (ⅰ) superficial cracks with different lengths and amounts along the high-angle grain boundaries (HAGBs) on the compressed sample surface, and (ⅱ) near-brittle fracture of the tensile stress-strain curves. Based on the analysis of the deformed and undeformed surface, local stress concentration and phase transformation were responsible for the cracks and difference in compression behavior, and atmospheric oxygen, deformation localization and phase transformation were mainly responsible for the tensile fracture difference at 800 °C. Phase transformation promote stress dispersion, combined with better oxidation resistance, which make the Zr-containing alloys fractured via the transgranular mode in tensile test. In particular, the Zr3 alloy exhibited work hardening and tensile ductility. Based on the tensile and compression properties of the Zrx alloys, it was found that at RT and 600 °C, atomic size difference (δ) governed alloy strength; from 800 to 1000 °C, δ and melting temperature (Tm) co-operatively governed alloy strength; at 1200 °C, alloy strength was independent of δ, but dependent on Tm. The fabricated Ti–V–Nb–Zrx alloys have medium mechanical properties but a small strength loss at high temperatures, and are suitable for various RT deformation processing methods.

BackgroundRecent studies have revealed a significant decrease in serum fetuin-A levels in atherosclerotic aneurysms, indicating that fetuin-A may play a protective role in the progression of arterial calcification. However, the specific mechanism behind this phenomenon remains unclear. We aimed to examine the association between fetuin-A levels in thoracic aortic aneurysms (TAAs) and risk of TAAs and to evaluate whether this association was causal.MethodsA total of 26 SNPs were selected as instrumental variables for fetuin-A in 9,055 participants of European ancestry from the CHARGE consortium, and their effects on thoracic aortic aneurysm and decreased descending thoracic aortic diameter were separately estimated in 353,049 and 39,688 individuals from FinnGen consortium. We used two-sample Mendelian randomization (MR) analysis to examine the causal association. At the same time, we employed various methods, including random-effects inverse variance weighting, weighted median, MR Egger regression, and MR PRESSO, to ensure the robustness of causal effects. We assessed heterogeneity using Cochran’s Q value and examined horizontal pleiotropy through MR Egger regression and retention analysis.ResultsFetuin-A level was associated with a significantly decreasing risk of thoracic aortic aneurysm (odds ratio (OR) 0.64, 95% CI 0.47 - 0.87, P = 0.0044). Genetically predicted fetuin-A was also correlated with the decreased descending thoracic aortic diameter (β = -0.086, standard error (SE) 0.036, P = 0.017).ConclusionsSerum fetuin-A level was negatively associated with risk of TTAs and correlated with the decreased descending thoracic aortic diameter. Mendelian randomization provides support for the potential causal relationship between fetuin-A and thoracic aortic aneurysm.

Amylopectin and amylose components are natural polymers within rice starch granules, intertwined in specific conditions to form gel polymerized with pore crosslink network, has potential printing properties. In this study, a rice starch gel preparation scheme is proposed for stable properties, and starch granule phase transition mechanism is analyzed based on RVA test during preparation, it can be divided into four-stage, swelling, reacting, homogenizing and self-assembling stages. Gel surface tension and contact angle tested with starch concentration effect, a correlation is developed, reflecting a competition result to gel droplet macro-morphology between the intermolecular cohesion and crosslink network. SEM is used to reveal typical crosslink structures of different starch molecular component proportions, providing objective support for starch gel rheologic property change. Results indicate gel interior crosslink network formed under concentration 12 %, the gel with amylose 4.475 % presents better printing accuracy. Gel shear modulus positively correlated with amylose proportion. Japonica gel under 20 % is of higher viscosity and rapid reassembly ability after interior crosslink network is broken. Max dynamic viscosity is positively correlated with starch concentration. The study aims to provide theoretical and practical support for in-depth analysis of rice starch material application in direct-write 3D printing.

Abstract To estimate the association between central retinal artery occlusion (CRAO) and major adverse cardiovascular and cerebrovascular events (MACCE), including their clinical characteristics, blood markers, and the contribution of CRAO to MACCE, as well as to assess any sex differences. This retrospective cohort study included continuous new-onset CRAO patients and 1:4 controls during the same period. Correlations of CRAO with the incidence of MACCE during follow-up and the sex-related differences were studied. One hundred and twenty-four CRAO patients and four hundred and ninety-six controls were enrolled. Neutrophil-to-lymphocyte ratio (NLR, P = 0.014) and high-sensitivity C-reactive protein (hs-CRP, P = 0.038) were tended to be higher in CRAO patients. After the follow-up period, 78 patients experienced MACCE. Multivariate Cox regression analysis showed that CRAO was a predictor of the occurrence of MACCE (HR 2.321, 95% CI 1.439–3.744, P = 0.001). Background: Human papillomavirus (HPV) vaccination is a key initiative to promote the WHO global strategy to accelerate the elimination of cervical cancer, and this study aimed to investigate the current status of HPV infection and genotypic characteristics of the population under the impact of age-expansion of nine-valent HPV vaccination policy in China. Methods: The clinical data of 60,685 subjects who were admitted in the Renmin Hospital of Wuhan University and underwent HPV genotyping from January 2017 to October 2022 were retrospectively analyzed. Results: The total number of positive HPV genotyping in the included population was 10,303, with a positivity rate of 17.0 %. Within the rapidly growing urban tourism industry, the development of urban landscapes plays a crucial role in shaping a city’s image and competitiveness; however, standardized and mismatched landscapes often have a negative impact, highlighting the importance of assessing urban landscape perceptions. Although existing studies have discussed this through subjective questionnaires and physiological methods, the underlying neural mechanisms have not been thoroughly explored. The research focuses on Jingdezhen, a renowned historical and cultural city in China, as its case study. Utilized the event-related potential (ERP) method to explore individuals’ perceptual consistency and neural activity toward different types of urban landscapes. We adopted a 2 (landscape type: historical, modern) X 2 (perceptual match: consistent, inconsistent) within-subject design while recording behavioral data and electrophysiological responses. The results showed that, under any condition, there were no significant differences in people’s behavioral data. Neurophysiological results indicate that consistent perceptions of modern landscapes elicited greater P200 responses, suggesting increased attention driven by visual aesthetics and emotional activation. Under conditions of perceptual inconsistency, historical landscapes elicited higher N400 amplitudes than modern landscapes, revealing cognitive conflict and effort. This study demonstrates that P200 and N400 components are effective indicators for assessing urban perception, proving the viability of the event-related potential method in urban landscape research. Additionally, the research reveals the neural mechanisms of urban environmental perception from the early stages of attention and emotional distribution to the later stages of cognitive decision-making, which involve cognitive processes from “bottom-up” to “top-down”. This study not only provides a reference for efficient design planning for those involved in urban science but also inspires the coordination between the developmental needs of historical and modern urban landscapes. Moreover, it offers a new perspective for an interdisciplinary approach to urban perception assessment.

Friction stir processing (FSP) with high rotating speed were successfully employed to fabricate a novel AZ31 magnesium matrix composites (MMCs) reinforced by biological particles of nano-hydroxyapatite (nHA) and nano-magnesium oxide (nMgO). The influences of tool rotating speed on the microstructure evolution, mechanical properties and corrosion resistance of MMCs were investigated. In results, high rotating speed not only significantly refined the grains of the stirred zone (SZ) in MMCs due to sufficient dynamic recrystallization (DRX), but also reduced the size of β-Al12Mg17 precipitates and caused them homogeneously distributed. With the increase of rotating speed, the bio-reinforced particles exhibited a larger range with uniform distribution in SZ of MMCs, and the microhardness, tensile strength and corrosion resistance of MMCs were also improved. At high rotating speed of 6000 rpm, MMCs had a maximum tensile strength of 235.5 MPa with cleavage fracture of ductile-brittle mixed mode. Meanwhile, the self-corrosion potential of MMCs (−1.408 V) was 10.4% higher than the matrix (−1.571 V), while the self-corrosion current density (9.328 × 10−5 A/cm2) was 96.4% much lower than the matrix (2.607 × 10−3 A/cm2), indicating that the corrosion resistance of MMCs was significantly enhanced than matrix. And the corrosion morphologies of MMCs presented mud cracking due to relatively uniform corrosion.

Increasing evidence indicates that inflammatory cytokines are involved in the pathogenesis of recurrent aphthous stomatitis (RAS). A wide range of over ten cytokines in peripheral blood of RAS patients have been investigated in different studies. Yet, which of the ones are the most prominent indicators contributed for the process of RAS are uncertain. Herein, a total of 16 eligible case–control studies including 1051 cases of RAS and 616 health controls were summarized. The bubble chart analysis showed that the most prominent cytokines for RAS were interleukin (IL)-6 (646 cases, 308 controls), TNF-α (498 cases, 298 controls), and IL-2 (371 cases, 264 controls). On the other hand, 9 studies on cytokines as therapeutic indicators of RAS were identified. The effect of levamisole and thalidomide on cytokines mainly were IL-6, TNF-α, and IL-8. Collectively, an optimum panel of IL-6, TNF-α, and IL-2 maybe serve as the potential significant indicators for RAS investigations.

ABSTRACTIt is known to all that Wnt signaling pathway plays an important role in the early development of tooth. Our previous research found that Wnt signaling pathway played crucial roles in dental development, and mutations in antagonist of Wnt signaling pathway may lead to the formation of supernumerary teeth. However, the expression pattern of Wnt signaling molecules in early development of tooth, especially genes with stage specificity, remains unclear. Hence, we applied RNA-seq analysis to determine the expression levels of wnt signal molecules at five different stages of rat first molar tooth germ. In addition, after literature review we summarized the function of Wnt signaling molecules during tooth development and the relationship between Wnt signaling molecules variation and tooth agenesis. Our research may have implications for exploring the role of Wnt signaling molecules in different stages of tooth development.

Abstract：: Fog seal technology has the advantages of convenient construction and rapid opening traffic, and it has a broad application prospect in the maintenance of airport asphalt pavement. However, the current evaluation indices of fog seal materials are not relevant enough to reflect their performance strengths and weaknesses comprehensively and systematically. Moreover, there is a lack of research on the non-emulsification class of fog seal materials. This research aims to compare and contrast various types of fog seal materials for airport asphalt pavement and provide suggestions for optimizing the fog seal materials and construction technology to achieve better results. This research tested the material, construction, and road performance of three fog seal materials: A, B, and C. Materials A and B are emulsified asphalt as matrix additive fog sealer, while material C is a special material fog sealer. The content of evaporation residue, surface curing performance, and rejuvenation performance test were optimized. The adhesion performance test was redesigned. Furthermore, the research performed a horizontal comparison of the material properties, construction performance, and road performance of the three kinds of fog seal materials. The key test methods and evaluation indices of fog seal were recommended by using statistical methods and discrimination analysis. The results showed that the emulsified asphalt as matrix additive fog sealer had a high content of evaporation residue and poor adhesion performance under low-temperature conditions, with a spalling ratio of over 75%. However, they had better adhesion performance with the aggregate under high-temperature conditions. The special material fog sealer had a low content of evaporation residue and excellent adhesion performance under low-temperature conditions, with a spalling ratio of nearly 0%. It also had the shortest surface curing time among the three materials. The special material fog seal has better comprehensive performance and better application prospects. According to the results of discrimination analysis, evaporation residue content, residue on sieve, and water dilution stability were recommended as evaluation indices of material performance. Adhesion of the material surface at the construction site and changes in material quality at 20 °C in the laboratory were recommended as evaluation indices of the surface curing performance. Adhesion (impact method) − 10 °C and adhesion (boiling method) amount of sticky sand were recommended as evaluation indices of road performance. This research aims to provide guidance for the optimization of fog seal material and construction technology of airport asphalt pavement.

N4-acetylcytidine (ac4C) is a modification of cytidine at the nitrogen-4 position, playing a significant role in the translation process of mRNA. However, the precise mechanism and details of how ac4C modifies translated mRNA remain unclear. Since identifying ac4C sites using conventional experimental methods is both labor-intensive and time-consuming, there is an urgent need for a method that can promptly recognize ac4C sites. In this paper, we propose a comprehensive ensemble learning model, the Stacking-based heterogeneous integrated ac4C model, engineered explicitly to identify ac4C sites. This innovative model integrates three distinct feature extraction methodologies: Kmer, electron-ion interaction pseudo-potential values (PseEIIP), and pseudo-K-tuple nucleotide composition (PseKNC). The model also incorporates the robust Cluster Centroids algorithm to enhance its performance in dealing with imbalanced data and alleviate underfitting issues. Our independent testing experiments indicate that our proposed model improves the Mcc by 15.61% and the ROC by 5.97% compared to existing models. To test our model’s adaptability, we also utilized a balanced dataset assembled by the authors of iRNA-ac4C. Our model showed an increase in Sn of 4.1%, an increase in Acc of nearly 1%, and ROC improvement of 0.35% on this balanced dataset. This study aimed to explored the association between depressive symptoms and infertility among U.S. women, and the effect of serum calcium on this association. We used data from the National Health and Nutrition Examination Survey (2013–2018), relating to women aged 20–45 years. Depressive symptoms were determined using the nine-item Patient Health Questionnaire (PHQ-9 scores ≥10), and interview data were used to identify self-reported infertility. Of 2708 women (mean age: 32.7 ± 7.5 years), 274 were depressed and 12.0 % self-reported being “ever-infertile.” Depressive symptoms were associated with infertility in multivariable logistic regression (OR, 1.62; 95 % CI, 1.11–2.38). Depressive symptoms were associated with infertility among participants who were obese (OR, 1.68; 95 % CI, 1.03–2.74), had not received psychological counseling (OR, 1.60; 95 % CI, 1.03–2.50), were antidepressant users (OR 3.22; 95 % CI, 1.15–9.00), and had high serum calcium levels (OR, 2.05; 95 % CI, 1.25–3.35). A significant interaction between serum calcium and depression was observed for infertility (P = .038, interaction likelihood ratio test). In sensitivity analyses, the association between depressive symptoms and infertility remained after excluding women aged ≥35 years (OR, 1.87; 95 % CI, 1.08–3.23), lowering the cut-off for PHQ-9 scores (≥5) (OR, 1.48; 95 % CI, 1.12–1.96), excluding women with some gynecological diseases (OR, 1.63; 95 % CI, 1.07–2.49), and using inverse probability of treatment weighting (OR, 1.64; 95 % CI, 1.17–2.31). Conclusion: Our findings indicate that depression is associated with infertility among U.S. women and serum calcium may have an effect on the association. Interventions such as serum calcium reduction, weight management and psychosocial counseling for infertility treatment in individuals with depression may be integrated into routine clinical practice. Additionally, more caution could be exercised when using antidepressants.

The occurrence of cracks in brown rice kernels has a substantial impact on grain quality. The timely and accurate detection of rice grains with cracks is crucial for enhancing the overall quality and flavor of processed rice. In this study, we developed an optical observation platform and optimized the original ResNet-18 neural network structure to improve the detection and classification of grain cracks. We established image datasets for japonica and indica rice varieties, and employed image augmentation and model migration techniques during training. In addition, we compared the performance of the optimized model with DenseNet-121 and GoogLeNet. The results demonstrate a notable enhancement in crack detection accuracy for japonica, reaching 96%, which is a 3.67% improvement over the original model. Furthermore, we achieved a substantial reduction in average training time, reduced by 58.66%. For indica rice, after model optimization and migration, the accuracy reached 96.67%. It’s important to note that the optimized model has limitations and is not suitable for mixed datasets with limited training data. This technology offers the capability to accurately identify and detect cracks in brown rice kernels under visible light conditions, presenting a promising solution for enhancing grain quality during processing.

Abstract Background It has been hypothesized that higher growth differentiation factor 15 (GDF15) level and lower testosterone/ estradiol (T/E) ratio are associated with major depressive disorder (MDD), yet the underlying effect of serum GDF15 on hinting the T/E ratio imbalance is not fully understood. We observed the correlation between serum T/E ratio and circulating GDF15 in male depressed cohort. Methods The sample consisted of participants (aged 18 ~ 65 years) from the Renmin Hospital of Wuhan University with MDD (n = 412) defined according to a Structured Clinical Interview for DSM-V (SCID), and male healthy controls (n = 137). Serum levels of testosterone, estradiol, and depression risk biomarkers (thyroid hormone, lipids, hs-CRP, Tenascin-C [TNC], GDF15, KLF4, Gas6, and sgp130) were measured. The associations among log-transformed T/E ratio and these biomarkers were analyzed using univariate correlation analysis, category analyses, and linear regression adjusting for standard risk factors. Results Of the sample, 36.89% had lower T/E ratio ( 20:1). After multivariable adjustment, T/E ratio was negatively associated with GDF15 (-0.095 [95% CI -0.170 ~ -0.023] standard deviation [SD] change per SD increase in lg[T/E], P = 0.015) and inversely related to TNC (-0.085 [95% CI -0.167 ~ 0.003] standard deviation [SD] change per SD increase in lg[T/E], P = 0.048). Serum T/E ratio was negatively associated with GDF15 level in both FT3, TSH and HDL strata, whereas this association was not observed in TNC. In T/E ratio strata analyses, there is a significant and negative correlation among T/E ratio and GDF15 in depressive patients with sex hormone imbalance, yet this relationship was not investigated in patients with sex hormone balance. Conclusion In our community-based observation, circulating GDF-15 level was greatly and inversely associated with serum T/E ratio, indicating that higher GDF-15 alerts sex hormone imbalance in patients with MDD.

Snake robots have great potential for exploring and operating in challenging unstructured environments, such as rubble, caves, and narrow pipelines. However, due to the complexity and unpredictability of unstructured environments, designing a controller that can achieve adaptive motion is crucial. This paper proposes a self-adaptive torque-based rolling controller for snake robots, enabling compliant motion in unstructured environments. First, a controller is designed to modify the torque of each motor by focusing on the different motion states of the rolling gait. Second, an experimental platform is established for snake robots to verify the effectiveness of the controller. Finally, a series of rolling experiments are conducted using the torque-based rolling controller. In conclusion, the self-adaptive torque-based rolling controller enhances snake robot adaptability and mobility.

The high temperature compression is performed by isothermal compression tests using Gleeble 3500 thermo-mechanical simulator at a constant strain rate for the hot-pressed Nb-22.5Cr-2.5Mo alloy. The deformation temperature ranges from 800 to 1200 °C and the strain rate ranges from 0.001 to 0.1 s−1. The high temperature flow behavior and deformation mechanism of the alloy are investigated based on the compression test data and TEM observation of the deformed microstructure. The results show that the ductile–brittle transition temperature of the alloy is about 800 °C, and the flow stress is sensitive to the deformation temperature and strain rate, in which the plastic deformation ability increases obviously with the increase of deformation temperature and the decrease of strain rate. Besides, the hyperbolic sine Arrhenius model with stress exponent of 3.131 and apparent activation energy of 402 kJ/mol is established for the peak flow stress. The value of apparent activation energy is higher than that of Nb metal (246 kJ/mol), which implies the existence of Laves phase NbCr2 makes the deformation of the alloy more difficult. Moreover, it is found that the deformation mechanisms in the Nbss matrix are mainly DRV controlled by the glide and climb of the dislocations and DRX formed by bulging nucleation mechanism, while the deformation mechanisms in the Laves phase NbCr2 particles are mainly twinning and synchroshear of the Shockley partial dislocation.

ContextCortisol, a hormone regulated by the hypothalamic-pituitary-adrenal (HPA) axis, has been linked to attention deficit hyperactivity disorder (ADHD). The nature of the relationship between cortisol and ADHD, and whether it is causal or explained by reverse causality, remains a matter of debate.ObjectiveThis study aims to evaluate the bidirectional causal relationship between morning plasma cortisol levels and ADHD.MethodsThis study used a bidirectional 2-sample Mendelian randomization (MR) design to analyze the association between morning plasma cortisol levels and ADHD using genetic information from the authoritative Psychiatric Genomics Collaboration (PGC) database (n = 55,347) and the ADHD Working Group of the CORtisol NETwork (CORNET) Consortium (n = 12,597). MR analyses were employed: inverse variance weighting (IVW), MR-Egger regression, and weighted medians. OR values and 95% CI were used to evaluate whether there was a causal association between morning plasma cortisol levels on ADHD and ADHD on morning plasma cortisol levels. The Egger-intercept method was employed to test for level pleiotropy. Sensitivity analysis was performed using the “leave-one-out” method, MR pleiotropy residual sum, and MR pleiotropy residual sum and outlier (MR-PRESSO).ResultsFindings from bidirectional MR demonstrated that lower morning plasma cortisol levels were associated with ADHD (ADHD-cortisol OR = 0.857; 95% CI, 0.755–0.974; P = 0.018), suggesting there is a reverse causal relationship between cortisol and ADHD. However, morning plasma cortisol levels were not found to have a causal effect on the risk of ADHD (OR = 1.006; 95% CI, 0.909–1.113; P = 0.907), despite the lack of genetic evidence. The MR-Egger method revealed intercepts close to zero, indicating that the selected instrumental variables had no horizontal multiplicity. The “leave-one-out” sensitivity analysis revealed stable results, with no instrumental variables significantly affecting the results. Heterogeneity tests were insignificant, and MR-PRESSO did not detect any significant outliers. The selected single-nucleotide polymorphisms (SNPs) F were all >10, indicating no weak instrumental variables. Thus, the overall MR analysis results were reliable.ConclusionThe study findings suggest a reverse causal relationship between morning plasma cortisol levels and ADHD, with low cortisol levels associated with ADHD. No genetic evidence was found to support a causal relationship between morning plasma cortisol levels and the risk of ADHD. These results suggest that ADHD may lead to a significant reduction in morning plasma cortisol secretion.

The mortality and morbidity rates of ovarian cancer (OC) are high, but the underlying mechanisms of OC have not been characterized. In this study, we determined the role of Rho GTPase Activating Protein 30 (ARHGAP30) in OC progression. We measured ARHGAP30 abundance in OC tissue samples and cells using immunohistochemistry (IHC) and RT-qPCR. EdU, transwell, and annexin V/PI apoptosis assays were used to evaluate proliferation, invasiveness, and apoptosis of OC cells, respectively. The results showed that ARHGAP30 was overexpressed in OC tissue samples and cells. Inhibition of ARHGAP30 suppressed growth and metastasis of OC cells, and enhanced apoptosis. Knockdown of ARHGAP30 in OC cells significantly inhibited the PI3K/AKT/mTOR pathway. Treatment with the PI3K/AKT/mTOR pathway inhibitor buparlisib simulated the effects of ARHGAP30 knockdown on growth, invasiveness, and apoptosis of OC cells. Following buparlisib treatment, the expression levels of p-PI3K, p-AKT, and p-mTOR were significantly decreased. Furthermore, buparlisib inhibited the effects of ARHGAP30 upregulation on OC cell growth and invasiveness. In conclusion, ARHGAP30 regulated the PI3K/AKT/mTOR pathway to promote progression of OC.

Reflective cracking is a common distress of old pavement overlaid with an asphalt layer. The asphalt rubber stress-absorbing membrane interlayer can effectively mitigate and prevent reflective cracking. However, the existing test methods and evaluation indices for the crack resistance of the asphalt rubber stress-absorbing membrane interlayer are insufficient and unsystematic. They do not account for the significant effect of gradation parameters on the crack resistance in a comprehensive way. Therefore, this research aims to explore the impact of gradation parameters on the performance of the asphalt rubber stress-absorbing membrane interlayer. Based on the Chinese and U.S. standards, three kinds of 10 types of gradation were selected, forming a total of seven groups. The asphalt rubber stress-absorbing membrane interlayer was subjected to −10 °C and 15 °C beam bending test, low-temperature semi-circular bend test, crack expansion semi-circular bend test, and overlay test to evaluate its cracking resistance. The correlation and influence law between the key sieve hole method, graded fractal method, and Bayley method parameters of different grades and beam bending test, low-temperature semi-circular bend test, crack expansion semi-circular bend test, and overlay test indexes were quantitatively analyzed by the coefficient of variation and Pearson correlation analysis method. The results showed that the performance of the mixtures with different gradation ranges varied significantly in different tests, as indicated by the maximum difference of 56.07% in stress absorption. This implied that gradation is a critical factor that affects the stress absorption performance of mixes. The different sensitivities of different tests to the parameters of the key sieve method, the graded fractal method, and the Bailey method indicated that the stress absorption performance was affected by a combination of factors. Therefore, in order to evaluate and optimize the stress absorption performance, it was necessary to comprehensively consider the interactions among the parameters of the key sieve method, the graded fractal method, and the Bailey method. The stress absorption performance included crack resistance and crack expansion resistance, which were inversely related and needed to be balanced and optimized during design. The −10 °C beam bending test and crack expansion semi-circular bend tests were more suitable test methods for evaluating stress absorption performance, and maximum flexural–tensile strain, strain energy density, fracture energy, and flexibility index were recommended as evaluation indicators. This research provides a reference for the optimization of the grading design of asphalt rubber stress-absorbing membrane interlayers, and provides test methods and indicators for the evaluation of crack resistance.

As urbanization quickens, the role of public art in urban landscape design gains prominence. Nevertheless, how stylistic characteristics of landscape public art affect aesthetic preferences remains insufficiently discussed, particularly with objective assessment methods. The use of event-related potential (ERP) can offer neurophysiological evidence to support research and practice in landscape art design. We employed a 2 (artistic features) × 2 (professional proficiency) repeated-measures design, involving abstract and figurative experimental stimuli; both experts and non-experts participated, with their aesthetic reactions and relevant electroencephalographic data recorded. Behavioral findings show a preference for figurative public artworks regardless of professional background. From neurophysiological outcomes, stimuli elicit an elevated N100 during early perceptual processing, signifying increased attentional resources. During aesthetic processing, figurative stimuli more effectively evoke positive emotions, particularly among professionals, yielding a heightened P200 response. Conversely, abstract stimuli may evoke a higher N200 amplitude, reflecting augmented negative biases. Nevertheless, non-experts exhibit no marked differences in their stimulus responses during aesthetic processing. Research indicates that low-level physical attributes of public artworks are initially noted, while the visual processing of artistic traits lies at a higher perceptual level, necessitating specialized expertise involvement. Furthermore, the complexity of visual perceptual processing plays a significant role in the assessment of landscape art preferences. This study not only offers crucial reference indices for designing urban landscapes that satisfy diverse public aesthetic needs but also lays the foundation for neural techniques to assess landscape design preferences and expands the field of landscape design research.

Abstract Background In research on new drug discovery, the traditional wet experiment has a long period. Predicting drug–target interaction (DTI) in silico can greatly narrow the scope of search of candidate medications. Excellent algorithm model may be more effective in revealing the potential connection between drug and target in the bioinformatics network composed of drugs, proteins and other related data. Results In this work, we have developed a heterogeneous graph neural network model, named as HGDTI, which includes a learning phase of network node embedding and a training phase of DTI classification. This method first obtains the molecular fingerprint information of drugs and the pseudo amino acid composition information of proteins, then extracts the initial features of nodes through Bi-LSTM, and uses the attention mechanism to aggregate heterogeneous neighbors. In several comparative experiments, the overall performance of HGDTI significantly outperforms other state-of-the-art DTI prediction models, and the negative sampling technology is employed to further optimize the prediction power of model. In addition, we have proved the robustness of HGDTI through heterogeneous network content reduction tests, and proved the rationality of HGDTI through other comparative experiments. These results indicate that HGDTI can utilize heterogeneous information to capture the embedding of drugs and targets, and provide assistance for drug development. Conclusions The HGDTI based on heterogeneous graph neural network model, can utilize heterogeneous information to capture the embedding of drugs and targets, and provide assistance for drug development. In the context of the global COVID-19 pandemic, the phenomenon that the elderly have higher morbidity and mortality is of great concern. Existing evidence suggests that senescence and viral infection interact with each other. Viral infection can lead to the aggravation of senescence through multiple pathways, while virus-induced senescence combined with existing senescence in the elderly aggravates the severity of viral infections and promotes excessive age-related inflammation and multiple organ damage or dysfunction, ultimately resulting in higher mortality. The underlying mechanisms may involve mitochondrial dysfunction, abnormal activation of the cGAS-STING pathway and NLRP3 inflammasome, the role of pre-activated macrophages and over-recruited immune cells, and accumulation of immune cells with trained immunity. Thus, senescence-targeted drugs were shown to have positive effects on the treatment of viral infectious diseases in the elderly, which has received great attention and extensive research. Therefore, this review focused on the relationship between senescence and viral infection, as well as the significance of senotherapeutics for the treatment of viral infectious diseases.

Several studies have shown that the dysregulation of the oral microbiota plays a crucial role in human health conditions, such as dental caries, periodontal disease, oral cancer, other oral infectious diseases, cardiovascular diseases, diabetes, bacteremia, and low birth weight. The use of traditional detection methods in conjunction with rapidly advancing molecular techniques in the diagnosis of harmful oral microorganisms has expanded our understanding of the diversity, location, and function of the microbiota associated with health and disease. This review aimed to highlight the latest knowledge in this field, including microbial colonization; the most modern detection methods; and interactions in disease progression. The next decade may achieve the rapid diagnosis and precise treatment of harmful oral microorganisms.

IntroductionIn live streaming shopping, exploring the influencing mechanism of consumers’ participation is an important prerequi site for understanding consumer behavior in social commerce activities. The purpose of this study is to explore the relationship betw een technological and social factors (visibility, media richness, guidance shopping and real-time interactivity) in live streaming shop ping and consumers’ purchase intention. The mediating roles of attraction and cognitive assimilation were also examined.MethodsThis study collected 425 pieces of data through questionnaire survey. The structural equation model is established based on S-O-R frame. The hypothesis is tested by structural equation model.ResultsOur study found that that real-time interactive and media richness positively affect attraction; visibility, guidance shopping and media richness positive affect cognitive assimilation; cognitive assimilation and attraction positive affect consumers’ purchase intention; cognitive assimilation and attraction play a partial mediating role in the influence of technology and social factors on cons umers’ purchase intention of live streaming shopping.DiscussionFrom the perspective of socio-technical, this study explores the influence mechanism of different influencing factors on consumers’ purchase intention in live streaming shopping. This study expands the application of IT affordance theory in the context of live streaming shopping, and reveals the mediating role of attraction and cognitive assimilation between social, technological factors and consumers’ purchase intention.

Aiming at the difficulty of feeding filler wire in the dual laser-beam synchronous welding (DLSW) of Ti–6Al–4V titanium alloys T-joints, the self-fusion DLSW process based on prefabricated welding materials on the stringer was developed. Prefabricated bosses on both sides of the stringer were used as welding materials. In this paper, the influence of boss size and laser power on the weld formation was studied. The influence of microstructure transformation on mechanical properties was demonstrated. It is found that T-joints with desirable weld formation can be obtained when the boss size of 0.8 × 0.8 mm2 was applied. The temperature distribution of the T-joint during laser welding process was obtained by finite element simulation method. It indicates that the cooling rate of fusion zone is 140 °C/s, indicating that massive transformation occurs in the fusion zone. The microstructure of fusion zone characterized by acicular martensite plates α′ and a small amount of intergranular α phase meets with the cooling rate of FZ. Due to multiple strengthening effects, the average microhardness of fusion zone is 351.6 HV, which is higher than the average microhardness of base material. The pull-out strength of T-joint is not related to the weld penetration, and its average pull-out strength is 1035.5 MPa. The fracture occurred in the weld toe of the pull-out specimen, and fracture surface presents quasi-cleavage fracture characteristics.

Sorting algorithms usually generate strictly descending sequences. However, almost descending sequences may be sufficient to fulfill our needs in practice, and the number of movements needed for generating such sequences can be greatly reduced. We call a sequence $S$ almost descending if $S[j]-S[i]< C$ for any $i< j$ and a positive number $C$ , which depends on the practical circumstances. Unless the difference between $S[j]$ and $S[i]$ exceeds the bound $C$ , we deem them indistinguishable. For example, when people are arranged in a line from tall to short, two people with heights of 1.751 meters and 1.752 meters can be considered visually indistinguishable (we can set the bound to $C=0.005$ meters in this case). Therefore, we do not need to adjust their positions, which leads to fewer movements in sorting. In this paper, three algorithms are provided to generate almost descending sequences, and their correctness is proven. The experimental results of the proposed algorithms demonstrate that our technique can reduce the number of movements in sorting. In addition, almost descending sequences are more stable than strictly descending sequences in dynamic sorting, which is useful in online sorting and sorting in noisy environments.

Protein S-nitrosylation is one of the most important post-translational modifications, a well-grounded understanding of S-nitrosylation is very significant since it plays a key role in a variety of biological processes. For an uncharacterized protein sequence, it is a very meaningful problem for both basic research and drug development when we can firstly identify whether it is a S-nitrosylation protein or not, and then predict the specific S-nitrosylation site(s). This work has proposed two models for identifying S-nitrosylation protein and its PTM sites. Firstly, three kinds of features are extracted from protein sequence: KNN scoring of functional domain annotation, PseAAC and bag-of-words based on the physical and chemical properties of amino acids. Secondly, the synthetic minority oversampling technique is used to balance the data sets, and some state-of-the-art classifiers and feature fusion strategies are performed on the balanced data sets. In the five-fold cross-validation for predicting S-nitrosylation proteins, the results of Accuracy (ACC), Matthew's correlation coefficient (MCC) and area under ROC curve (AUC) are 81.84%, 0.5178, 0.8635, respectively. Finally, a model for predicting S-nitrosylation sites has been constructed on the basis of tripeptide composition (TPC) and the composition of k-spaced amino acid pairs (CKSAAP). To eliminate redundant information and improve work efficiency, elastic nets are employed for feature selection. The five-fold cross-validation tests have indicated the promising success rates of the proposed model. Depression, one of the most common causes of disability, has a high prevalence rate in patients with metabolic syndrome. Type 2 diabetes patients are at an increased risk for depression. However, the molecular mechanism coupling diabetes to depressive disorder remains largely unknown. Here we found that the neuroinflammation, associated with high-fat diet (HFD)-induced diabetes and obesity, activated the transcription factor CCAAT/enhancer binding protein β (C/EBPβ) in hippocampal neurons. This factor repressed brain-derived neurotrophic factor (BDNF) expression and caused depression-like behaviors in male mice. Besides, the loss of C/EBPβ expression in C/EBPβ heterozygous knockout male mice attenuated HFD-induced depression-like behaviors, whereas Thy1-C/EBPβ transgenic male mice (overexpressing C/EBPβ) showed depressive behaviors after a short-term HFD. Furthermore, HFD impaired synaptic plasticity and decreased surface expression of glutamate receptors in the hippocampus of wild-type (WT) mice, but not in C/EBPβ heterozygous knockout mice. Remarkably, the anti-inflammatory drug aspirin strongly alleviated HFD-elicited depression-like behaviors in neuronal C/EBPβ transgenic mice. Finally, the genetic delivery of BDNF or the pharmacological activation of the BDNF/TrkB signaling pathway by 7,8-dihydroxyflavone reversed anhedonia in a series of behavioral tests on HFD-fed C/EBPβ transgenic mice. Therefore, our findings aim to demonstrate that the inflammation-activated neuronal C/EBPβ promotes HFD-induced depression by diminishing BDNF expression.

ObjectivesSystemic sclerosis (SSc) is an autoimmune disease caused by various pathogenic factors, including hypoxia. Hypoxia stimulates the production of the extracellular matrix to promote fibrosis. However, the integrated function and the underlying mechanism of hypoxia in SSc are unclear.MethodsIn the present study, we used Agilent SurePrint G3 Human Gene Expression v3 for the transcriptional sequencing of fibroblasts with and without hypoxia to detect differentially expressed genes (DEGs) in hypoxia. We analyzed the results with the transcriptome data of SSc lesions (GSE95065) to select the co-DEGs. Then, Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses were performed on the basis of the co-DEGs using the R package ClusterProfiler, which showed that hypoxia and cross talk of hypoxia with other pathogenic factors are involved in the pathogenesis of SSc. Furthermore, we constructed a protein–protein interaction (PPI) network of co-DEGs and screened two significant functional expression modules.ResultsWe identified nine hub genes (ALDH1A1, EGF, NOX4, LYN, DNTT, PTGS2, TKT, ACAA2, and ALDH3A1). These genes affect the pentose phosphate pathway, oxidative stress, and lipolysis.ConclusionOur study provides insights into the mechanisms underlying the effects of hypoxia on SSc pathogenesis, which will help to better understand SSc pathogenesis and develop new therapeutic strategies for SSc.

Redundant manipulators are flexible enough to adapt to complex environments, but their controller is also required to be specific for their extra degrees of freedom. Inspired by the morphology of snakes, we propose a path planning algorithm named Swinging Search and Crawling Control, which allows the snake-like redundant manipulators to explore in complex pipeline environments without collision. The proposed algorithm consists of the Swinging Search and the Crawling Control. In Swinging Search, a collision-free manipulator configuration that of the end-effector in the target point is found by applying reinforcement learning to self-motion, instead of designing joint motion. The self-motion narrows the search space to the null space, and the reinforcement learning makes the algorithm use the information of the environment, instead of blindly searching. Then in Crawling Control, the manipulator is controlled to crawl to the target point like a snake along the collision-free configuration. It only needs to search for a collision-free configuration for the manipulator, instead of searching collision-free configurations throughout the process of path planning. Simulation experiments show that the algorithm can complete path planning tasks of hyper-redundant manipulators in complex environments. The 16 degrees of freedom and 24 degrees of freedom manipulators can achieve 83.3% and 96.7% success rates in the pipe, respectively. In the concentric pipe, the 24 degrees of freedom manipulator has a success rate of 96.1%.

Abstract Background Population-based screening was essential for glaucoma management. Although various studies have investigated the cost-effectiveness of glaucoma screening, policymakers facing with uncontrollably growing total health expenses were deeply concerned about the potential financial consequences of glaucoma screening. This present study was aimed to explore the impact of glaucoma screening with artificial intelligence (AI) automated diagnosis from a budgetary standpoint in Changjiang county, China. Methods A Markov model based on health care system’s perspective was adapted from previously published studies to predict disease progression and healthcare costs. A cohort of 19,395 individuals aged 65 and above were simulated over a 15-year timeframe. Fur illustrative purpose, we only considered primary angle-closure glaucoma (PACG) in this study. Prevalence, disease progression risks between stages, compliance rates were obtained from publish studies. We did a meta-analysis to estimate diagnostic performance of AI automated diagnosis system from fundus image. Screening costs were provided by the Changjiang screening programme, whereas treatment costs were derived from electronic medical records from two county hospitals. Main outcomes included the number of PACG patients and health care costs. Cost-offset analysis was employed to compare projected health outcomes and medical care costs under the screening with what they would have been without screening. One-way sensitivity analysis was conducted to quantify uncertainties around model results. Results Among people aged 65 and above in Changjiang county, it was predicted that there were 1940 PACG patients under the AI-assisted screening scenario, compared with 2104 patients without screening in 15 years’ time. Specifically, the screening would reduce patients with primary angle closure suspect by 7.7%, primary angle closure by 8.8%, PACG by 16.7%, and visual blindness by 33.3%. Due to early diagnosis and treatment under the screening, healthcare costs surged dramatically to $107,761.4 dollar in the first year and then were constantly declining over time, while without screening costs grew from $14,759.8 in the second year until peaking at $17,900.9 in the 9th year. However, cost-offset analysis revealed that additional healthcare costs resulted from the screening could not be offset by decreased disease progression. The 5-, 10-, and 15-year accumulated incremental costs of screening versus no screening were estimated to be $396,362.8, $424,907.9, and $434,903.2, respectively. As a result, the incremental cost per PACG of any stages prevented was $1464.3. Conclusions This study represented the first attempt to address decision-maker’s budgetary concerns when adopting glaucoma screening by developing a Markov prediction model to project health outcomes and costs. Population screening combined with AI automated diagnosis for PACG in China were able to reduce disease progression risks. However, the excess costs of screening could never be offset by reduction in disease progression. Further studies examining the cost-effectiveness or cost-utility of AI-assisted glaucoma screening were needed.

Drug–target interactions (DTIs) are regarded as an essential part of genomic drug discovery, and computational prediction of DTIs can accelerate to find the lead drug for the target, which can make up for the lack of time-consuming and expensive wet-lab techniques. Currently, many computational methods predict DTIs based on sequential composition or physicochemical properties of drug and target, but further efforts are needed to improve them. In this article, we proposed a new sequence-based method for accurately identifying DTIs. For target protein, we explore using pre-trained Bidirectional Encoder Representations from Transformers (BERT) to extract sequence features, which can provide unique and valuable pattern information. For drug molecules, Discrete Wavelet Transform (DWT) is employed to generate information from drug molecular fingerprints. Then we concatenate the feature vectors of the DTIs, and input them into a feature extraction module consisting of a batch-norm layer, rectified linear activation layer and linear layer, called BRL block and a Convolutional Neural Networks module to extract DTIs features further. Subsequently, a BRL block is used as the prediction engine. After optimizing the model based on contrastive loss and cross-entropy loss, it gave prediction accuracies of the target families of G Protein-coupled receptors, ion channels, enzymes, and nuclear receptors up to 90.1, 94.7, 94.9, and 89%, which indicated that the proposed method can outperform the existing predictors. The proposed method may also be a potential option for other DITs.

Pupylation is an important posttranslational modification in proteins and plays a key role in the cell function of microorganisms; an accurate prediction of pupylation proteins and specified sites is of great significance for the study of basic biological processes and development of related drugs since it would greatly save experimental costs and improve work efficiency. In this work, we first constructed a model for identifying pupylation proteins. To improve the pupylation protein prediction model, the KNN scoring matrix model based on functional domain GO annotation and the Word Embedding model were used to extract the features and Random Under-sampling (RUS) and Synthetic Minority Over-sampling Technique (SMOTE) were applied to balance the dataset. Finally, the balanced data sets were input into Extreme Gradient Boosting (XGBoost). The performance of 10-fold cross-validation shows that accuracy (ACC), Matthew’s correlation coefficient (MCC), and area under the ROC curve (AUC) are 95.23%, 0.8100, and 0.9864, respectively. For the pupylation site prediction model, six feature extraction codes (i.e., TPC, AAI, One-hot, PseAAC, CKSAAP, and Word Embedding) served to extract protein sequence features, and the chi-square test was employed for feature selection. Rigorous 10-fold cross-validations indicated that the accuracies are very high and outperformed its existing counterparts. Glucose and cholesterol engage in almost all human physiological activities. As the primary energy substance, glucose can be assimilated and converted into diverse essential substances, including cholesterol. Cholesterol is mainly derived from de novo biosynthesis and the intestinal absorption of diets. It is evidenced that glucose/insulin promotes cholesterol biosynthesis and uptake, which have been targeted by several drugs for lipid-lowering, e.g., bempedoic acid, statins, ezetimibe, and proprotein convertase subtilisin/kexin type 9 (PCSK9) inhibitors. Inversely, these lipid-lowering drugs may also interfere with glucose metabolism. This review would briefly summarize the mechanisms of glucose/insulin-stimulated cholesterol biosynthesis and uptake, and discuss the effect and mechanisms of lipid-lowering drugs and genetic mutations on glucose homeostasis, aiming to help better understand the intricate relationship between glucose and cholesterol metabolism.
