Functional roles of SIGLEC15 in hepatocellular carcinoma (HCC) were not clear, which was recently found to be an immune inhibitor with similar structure of inhibitory B7 family members. SIGLEC15 expression in HCC was explored in public databases and further examined by PCR analysis. SIGLEC15 and PD-L1 expression patterns were examined in HCC samples through immunohistochemistry. SIGLEC15 expression was knocked-down or over-expressed in HCC cell lines, and CCK8 tests were used to examine cell proliferative ability in vitro. Influences of SIGLEC15 expression on tumor growth were examined in immune deficient and immunocompetent mice respectively. Co-culture system of HCC cell lines and Jurkat cells, flow cytometry analysis of tumor infiltrated immune cells and further sequencing analyses were performed to investigate how SIGLEC15 could affect T cells in vitro and in vivo. We found SIGLEC15 was increased in HCC tumor tissues and was negatively correlated with PD-L1 in HCC samples. In vitro and in vivo models demonstrated inhibition of SIGLEC15 did not directly influence tumor proliferation. However, SIGLEC15 could promoted HCC immune evasion in immune competent mouse models. Knock-out of Siglec15 could inhibit tumor growth and reinvigorate CD8+ T cell cytotoxicity. Anti-SIGLEC15 treatment could effectively inhibit tumor growth in mouse models with or without mononuclear phagocyte deletion. Bulk and single-cell RNA sequencing data of treated mouse tumors demonstrated SIGLEC15 could interfere CD8+ T cell viability and induce cell apoptosis. In all, SIGLEC15 was negatively correlated with PD-L1 in HCC and mainly promote HCC immune evasion through inhibition of CD8+ T cell viability and cytotoxicity. [ABSTRACT FROM AUTHOR]

Propulsion performance produced by laser ablation of polymer made of acrylonitrile butadiene styrene is experimentally investigated using the first, second, and third harmonics of a Nd: YAG laser. A ballistic pendulum is employed to assess the impulse and coupling coefficient for laser propulsion application. Fast photography, target ablation, and optical emission spectroscopy are proposed to analyze the energy coupling characteristic. The impulse and coupling coefficient under different pressures are demonstrated to depend on the target ablation and plasma properties which are relevant to laser wavelength. As the laser wavelength decreases, the crater depth and ablation mass are enhanced. Meanwhile, the plasma plume separates at atmospheric pressure and its length extends continuously in the low-pressure range. As a result, plasma including more ejected particles with higher velocity contributes to obtaining excellent impulse and coupling coefficient. In addition, the decreased electron density and temperature indicate higher collision frequency and photoionization dominate rather than inverse bremsstrahlung absorption at shorter laser wavelengths. Scientific developing concept of higher education is overall, coordinated and sustained based on facts. The developing planning of a university which is the perspective of a university must be unified, directed and completed scientifically. Take Yanshan University for example, during its 11th Five-Year-Plan period, we hold the opinion that university orientation is a norm of formulating the university developing planning. Meanwhile, scientific developing plan is the prerequisite to implementing scientific developing concept of higher education.

Background: Although CA19-9 is an essential blood biomarker of pancreatic cancer (PC), its sensitivity and specificity are limited for early detection. Methods: We analyzed the serum proprotein convertase subtilisin/kexin type 9 (sPCSK9) in PC patients, benign disease groups (BDG), and healthy controls (HC) by ELISA. Results: Consistently, sPCSK9 was considerably lower in PC patients than in HC (Z = −2.546, P < 0.05), and sPCSK9 in PC patients was statistically significantly higher than in BDG (Z = −5.457, P < 0.001). sPCSK9 was linked to the invasion of lymph nodes (χ2 = 6.846, P < 0.01). According to ROC curves, combining sPCSK9 with CA19-9 could potentially enhance the diagnostic capability of CA19-9 in early-stage PC patients. Furthermore, the low sPCSK9 group (n = 41) exhibited statistically significantly prolonged overall survival compared to the high sPCSK9 group (n = 15), with median survival times of 27 months (95% CI [17.59–36.41]) and 11 months (95% CI [7.21–14.79]), respectively (P = 0.022). Conclusion: The diagnostic performance of CA19-9 for early-stage PC patients could be improved by combining sPCSK9 with CA19-9. Single-stage compression loading experiments were carried out on concrete specimens of various strengths to explore the characteristic parameters of the acoustic emission signal and its damage evolution law in the concrete damage process. These specimens were monitored in real time with acoustic emission and DIC instruments during the loading process, and internal pores and slices were scanned with CT scanning instruments after compression. The acoustic emission phenomenon was expressed using the energy activity coefficient, and the law relating to the phenomenon was summarized. The results show that when the peak and mean values in the first adjacent time domain grow rapidly, the specimen produces a large crack and enters the stage of rapid crack development, which can be taken as an indication of the impending damage to the specimen. The energy activity coefficient reflects the damage development intensity as follows: the smaller the energy activity coefficient, the more the cracks developed; the faster the speed, the larger the deformation. With an increase in the load level, the energy activity coefficient gradually tends to stabilize, and the specimen enters the stage of rapid crack development. However, when the energy activity coefficient suddenly increases again, the specimen is destabilized and destroyed. Therefore, the energy activity coefficient responds to the degree of congenital defects in the specimen. As the load increases, the energy activity coefficient is more stable, and the defects are smaller; in contrast, the energy activity coefficient drastically oscillating indicates that the material is very defective.
