Signal transducer and activator of transcription (STAT) proteins with transcriptional activity, which are involved in the pathogenesis and formation of skin lesions of atopic dermatitis, psoriasis, skin tumors and other dermatosis, and are related to the severity of dermatosis. Different STAT has different pathways in dermatosis. STAT regulate cell signal transduction, affect the expression of downstream factors, and exert biological effects. Further study of STAT expression level and targeted genes to explore the relationship between STAT and dermatosis will help to find potential therapeutic targets for dermatosis.

Two isostructural lanthanide complexes were synthesized by solvent evaporation with 3-dimethylaminobenzoic acid and 5,5′-dimethyl-2,2′-bipyridine as ligands. The general formula of the structure is a [Ln(3-N,N-DMBA)3(5,5′-DM-2,2′-bipy)]2·2(3-N,N-DMHBA), Ln = (Gd(1), Tb(2)), 3-N,N-DMBA = 3-Dimethylamino benzoate, 5,5′-DM-2,2′-bipy = 5,5′-dimethyl-2,2′ bipyridine. Both complexes exhibited dimeric structures based on X-ray diffraction analysis. At the same time, infrared spectroscopy and Raman spectroscopy were used to measure the spectra of the complex. A thermogravimetric infrared spectroscopy experiment was performed to investigate the thermal stability and decomposition mechanism of the complexes. Measurements of the low-temperature heat capacity of the complexes were obtained within the temperature range of 1.9 to 300 K. The thermodynamic function was calculated by heat capacity fitting. In addition, the fluorescence spectra of complex 2 were studied and the fluorescence lifetime values were determined, and the energy transfer mechanism of complex 2 was elucidated. Objective The aim of this study was to examine serum leukocyte cell-derived chemotaxin 2 (LECT2) levels in osteoporosis subjects to confirm its association with osteoporosis. Methods A total of 204 adult subjects were recruited. Bone mineral densities (BMD) were assessed and blood samples were collected for measurements of biomedical parameters and the bone turnover markers. Serum LECT2 levels were measured by enzyme-linked immunosorbent assay. The relationships between serum LECT2 levels and other parameters were analyzed using the Spearman correlation coefficient. Results Serum LECT2 levels were significantly increased in osteoporosis subjects over controls. We found a significantly negative correlation of serum LECT2 with BMD, 25-hydroxy-vitamin D, and creatinine and a significantly positive correlation with C-terminal telopeptide of type 1 collagen and total cholesterol. Conclusion Serum LECT2 levels were significantly upregulated in osteoporosis subjects and correlated with the severity of bone loss. A new binuclear complex [Sm( p-MOBA)3bipy]2 · 2C2H5OH ( p-MOBA = p-methoxybenzoate, bipy = 2,2′-bipyridine) has been prepared via dissolution reaction in an ethanol/water mixed solution and characterized by elemental analysis, IR, UV, XRD and molar conductance, respectively. Its single crystal was characterized by X-ray single crystal diffraction. The crystal belongs to triclinic system, space group P $$ \bar 1 $$. The crystallographic data are: a = 10.970(3) Å, b = 11.507(4) Å, c = 13.931(4) Å, α = 84.449(4)°, β = 76.403(4)°, γ = 87.284(4)°, Z = 1, D r = 1.574 Mg/m3, FW = 1612.02, F(000) = 814, R1 = 0.0275, wR 2 = 0.0646. (1.10-Phenanthroline)-tris(4-amidobenzoate)dysprosium, [Dy( p-ABA)3Phen · H2O] · 1.5H2O (where p-ABA = p-amidobenzoate and Phen = 1.10-phenanthroline) has been synthesized. The complex was characterized by elemental analysis, UV, IR spectroscopy, and molar conductance. The crystal structure was determined by X-ray crystallography. The coordination number of the mononuclear complex is nine. The crystals are triclinic, space group P1 with a = 10.4484(13) Å, b = 12.2015(15) Å, c = 14.0170(17) Å; α = 92.800(2)°, β = 102.7220(10)°, γ = 108.880(2)°. Z = 2, d c = 1.617 mg m−3, F(000) = 786. The title complex [Dy(p-ClBA)3Phen]2·2H2O was synthesized, where p-ClBA is p-chlorobenzoate and Phen is 1,10-phenanthroline. The complex was characterized by various techniques including elemental analysis, IR, XRD, and molar conductance. The thermal decomposition of the complex was studied under the nonisothermal condition by TG-DTG and IR techniques. Complexes of [Sm2(m-ClBA)6(phen)2] · 2H2O and [Sm2(m-BrBA)6(phen)2] · 2H2O (m-ClBA = m-chlorobenzoate, m-BrBA = m-bromobenzoate, and phen = 1,10-phenanthroline) were prepared and characterized by elemental analysis and IR spectra. The thermal decomposition processes of the two complexes were studied by means of TG–DTG and IR techniques. Abstract: The title complex [Sm(o-MBA)3phen]2 (o-MBA, o-methylbenzoate; phen, 1,10-phenanthroline) was prepared and characterized by single crystal X-ray diffraction, elemental analysis, IR spectra and TG-DTG techniques. The results show that the crystal is triclinic, space group P ī with a =13.1364(10)Å, b =14.3739(11)Å, c =17.4356(13)Å; α =83.1100(10)°, β =83.7760(10)°, γ =73.0520(10)°, Z =2, D c =1.568Mg/m3, F (000)=1476. The crystal is consisted of two similar types of binuclear molecule. Each Sm3+ is nine-coordinated by one bidentate chelating carboxylate group, two bidentate bridging and two tridentate chelating-bridging carboxylate groups, as well as one 1,10-phenanthroline molecule. The thermal decomposition mechanism of [Sm(o-MBA)3phen]2 has been determined on the basis of thermal analysis. The dependences of E −α for the first-stage decomposition process were determined by using Ozawa-iterative procedure, which show the E values varied with the extent of conversion. In addition, the lifetime equation at weight-loss of 10% was deduced as ln τ =−25.5547+18511.68/T by isothermal thermogravimetric analysis. [Copyright &y& Elsevier]

Abstract: A new binuclear samarium (III) complex [Sm(p-BrBA)3bipy·H2O]2·H2O (p-BrBA= p-bromobenzoic acid; bipy=2,2′-bipyridine) has been synthesized and characterized by elemental analysis, UV, IR, molar conductance and TG–DTG techniques. The structure of the complex was established by single crystal X-ray diffraction. It crystallizes in triclinic, space group with a =8.2476(7)Å, b =13.3483(10)Å, c =15.9035(13)Å, α =73.9160(10)°, β =78.9630(10)°, γ = 74.4770(10)°, Z =1, D c =1.947gcm−3, F(000)=910. The carboxylic groups are bonded to the samarium ion in two modes: bidentate bridging, monodentate. Each center Sm3+ ion is eight-coordinated by one 2,2′-bipyridine molecular, four bidentate bridging and a monodentate carboxylic group, as well as one water molecular. The coordination polyhedron around each Sm3+ ion can be described as bi-capped triangular prism geometry. Abstract: The complex of [Sm(BA)3bipy]2 (BA=benzoic acid; bipy=2,2′-bipyridine) was obtained and characterized by elemental analysis, molar conductance, IR spectra, UV spectra, single crystal X-ray diffraction and TG–DTG techniques. The crystal is monoclinic with space group P2(1)/n. The unit cell parameters are: a =14.157(2)Å, b =15.416(3)Å, c =26.079(4)Å, β =103.445(2)°. Two Sm3+ ions are connected by four carboxylic groups through bridging bidentate mode. Each center Sm3+ is eight-coordinated with a trigondodecahedron. The thermal decomposition behaviors of the title complex in a static air atmosphere were discussed by TG–DTG and IR techniques. The non-isothermal kinetics was investigated by using double equal-double step method and nonlinear integral isoconversional method. The mechanism function of the first decomposition step was determined. Meanwhile, the thermodynamic parameters (ΔH ≠, ΔG ≠ and ΔS ≠) and kinetic parameters (activation energy E and pre-exponential factor A) were also calculated. The lifetime equation at mass-loss of 10% was deduced as ln τ =−34.56+19299.15/T by isothermal thermogravimetric analysis. The study was aimed to investigate the mechanism of mannan-binding lectin (MBL) on bacterial lipopolysaccharide (LPS)-induced human peripheral blood monocyte-derived dendritic cell (DC) maturation. The monocytes were prepared from the peripheral blood of healthy adult volunteers. The immature dendritic cells (imDC) were induced by 5-day-culture in medium supplemented with rhGM-CSF and rhIL-4. FACS was used to investigate the interaction of MBL with imDC and the impact of MBL on LPS binding to imDC. ELISA and Western blot was used to analyze the interaction of MBL with soluble TLR4 ectodomain protein (sTLR4); Western blot was used to detect LPS-induced NF-κB translocation in imDC. The results showed that MBL could directly bind to imDC in the presence of calcium. sTLR4 protein or LPS could competitively inhibit the binding of MBL to imDC. ELISA and Western blot showed that MBL could evidently bind to sTLR4 protein in a concentration-dependent manner. FACS showed that MBL could competitively inhibit the binding of LPS to imDC by binding to imDC directly. Western blot showed that MBL decreased LPS-induced NF-κB translocation in imDC. It is concluded that MBL may competitively inhibit the binding of LPS to imDC by binding to TLR4 expressed on imDC, resulted in inhibition of LPS-induced DC maturation, suggesting that MBL can regulate DC maturation through ligand-binding. This study provides the good foundation to clarify the mechanism of MBL inhibiting the LPS-induced DC maturation.

We investigated the anti-tumor effects of Verbena officinalis extract on H22 tumor-bearing mice and its effect on immune function. Mice model of H22 solid tumor was established, the mice were divided into five groups and administered the extract, later, tumors were removed and inhibition rates were calculated; spleens were removed and spleen indices were calculated, and the sheep red blood cell-delayed-type hypersensitivity (SRBC-DTH) and the serum hemolysin level were determined. The Verbena officinalis extract had anti-tumor effect, with the inhibition rate reaching 38.78%, it also increased the spleen index to a certain extent, in addition, the changes in DTA and HA were not obvious compared with the model group. The Verbena officinalis extract had in vivo anti-tumor effect, while causing no damage on the immune function.

Objective: To analyze the clinical value of dual-source CT (DSCT) in the diagnosis of coronary artery disease., Methods: Fifty-five patients with suspected coronary heart disease underwent both DSCT coronary angiography (DSCTCA) and selective coronary angiography (CAG) examination, and the diagnostic sensitivity, specificity and accuracy of the DSCTCA was evaluated., Results: The sensitivity, specificity, positive and negative predictive value, and accuracy of DSCT in the diagnosis of coronary heart disease were 97.7%, 72.6%, 93.5%, 88.9% and 92.7% by the number of patients, respectively; by calculating the coronary arteries, the sensitivity, specificity, positive and negative predictive value, accuracy were 94.9%, 95.8%, 92.5%, 97.1%, 95.5%, respectively. According to the lesion segment, these values were 88.2%, 96.9%, 90.5%, 96.1%, 94.7%, respectively. DSCTCA showed no significant difference from CAG for a diagnostic purpose, nor did their vessel sensitivity, specificity, positive and negative predictive value, and accuracy in different coronaries differ significantly., Conclusion: DSCT has a diagnostic accuracy of coronary heart disease close to that CAG and can on some occasion serve as an alternative to CAG in the screening of coronary artery disease.

A lymphocyte cDNA library of normal human was constructed in order to obtain specific gene and prepare lymphocyte gene chips to detect the relative genes between psychiatric diseases and immunity. The lymphocyte was abstracted from fresh normal human blood and cultured in vitro. Total RNA of lymphocyte was extracted from the cultured cells and then mRNA was extracted further. Moreover,single-strand cDNA and double-strand cDNA were synthesized in turn. The double-strand cDNAs were ligated to SalI and NotI adaptor,which were later ligated to arms of gammaZipLox. Ligated-cDNAs were packed in vitro, and then infected E. coli Y1090. Titering the phage and amplifying the library. The lymphocyte cDNA library consisted of 2.6 x 10(6) recombinants with the length of 1-5 kb and the cloning efficiency was 5 x 10(12) recombinants/g cDNA. The amplified library was 3 x 10(7) recombinants/microl in concentration and the number of bacteriophage plagues was the most suitable in density after it was diluted to 10(-6) in concentration. The constructed cDNA library of normal human lymphocyte would be helpful to further detecting target genes and preparing gene chips etc.
