14. Baicalin Inhibits VSMC Phenotypic Transition and Ameliorates Atherosclerosis via GSK3β-TCF21 Signaling Pathway.

Abstract

Background: Atherosclerosis (AS) is the pathological basis of acute myocardial infarction, acute stroke, and other acute critical illnesses. Vascular smooth muscle cell (VSMC) phenotypic transformation plays a remarkable role in the occurrence and development of AS. Previous studies have found that GSK3β-TCF21 is a key signaling pathway regulating VSMC phenotypic transformation. Baicalin is an important herbal monomer for cardiovascular protection, but whether it can regulate the GSK3β-TCF21 signaling pathway and VSMC phenotypic transformation remains unclear. Purpose: The study aimed to explore the role of Baicalin in AS and VSMC phenotypic transformation and whether the GSK3β-TCF21 signaling pathway participates in this process. Materials and Methods: In this study, through the AS mouse model and ox-LDL-induced VSMC phenotypic transformation model in vitro, the effect of Baicalin on the GSK3β-TCF21 signaling pathway and VSMC phenotypic transformation was investigated. Results: Pathological staining showed that the AS plaque area of the Baicalin group decreased significantly (p < 0.001). The expression of the marker gene (TAGLN, ACTA2, CNN1, and MYH11) of VSMC phenotypic transformation was significantly increased (p < 0.001) after Baicalin treatment. Baicalin could activate GSK3β and increase the expression of TCF21 significantly (p < 0.001) to inhibit VSMC phenotypic transition. Inhibiting the GSK3β-TCF21 signaling pathway was able to reduce the effects of Baicalin on reducing VSMC phenotypic transition and protecting AS. Conclusion: Our study indicates that Baicalin is protective in inhibiting VSMC phenotypic transformation and ameliorating AS by regulating GSK3β-TCF21. [ABSTRACT FROM AUTHOR]

Abstract

Background This study aimed to estimate population-level and state-level lead-attributable mortality burdens stratified by socioeconomic status (SES) class in the USA. Methods Based on the National Health and Nutrition Examination Survey (NHANES), we constructed individual-level SES scores from income, employment, education and insurance data. We assessed the association between the blood lead levels (BLL) and all-cause mortality by Cox regression in the NHANES cohort (n  = 31 311, 4467 deaths). With estimated hazard ratios (HR) and prevalences of medium (2–5 μg/dL) and high (≥ 5 μg/dL) BLL, we computed SES-stratified population-attributable fractions (PAFs) of all-cause mortality from lead exposure across 1999–2019. We additionally conducted a systematic review to estimate the lead-attributable mortality burden at state-level. Results The HR for every 2-fold increase in the BLL decreased from 1.23 (1.10–1.38) for the lowest SES class to 1.05 (0.90–1.23) for the highest SES class. Across all SES quintiles, medium BLL exhibited a greater mortality burden. Individuals with lower SES had higher lead-attributable burdens, and such disparities haver persisted over the past two decades. In 2017–19, annually 67 000 (32 000–112 000) deaths in the USA were attributable to lead exposure, with 18 000 (2000–41 000) of these deaths occurring in the lowest SES class. Substantial disparities in the state-level mortality burden attributable to lead exposure were also highlighted. Conclusions These findings suggested that disparities in lead-attributable mortality burden persisted within US adults, due to heterogeneities in the effect sizes of lead exposure as well as in the BLL among different SES classes. Abstract

In order to further develop efficient ultraviolet light-driven photocatalysts for environmental application, α-zirconium phosphate (α-ZrP) and carbon nitride (C3N4) were synthesized, respectively. Then, C3N4–ZrP compositing nanomaterials were prepared by compositing α-ZrP nanocrystals and C3N4 with different mass ratios. C3N4–ZrP compositing nanomaterials were characterized by X-ray diffraction, scanning electron microscopy, Fourier transform infrared spectroscopy, and X-ray photoelectron spectroscopy. The results illustrated that α-ZrP and C3N4 were successfully composited, and the polarization of the compositing nanomaterials was reduced compared with raw materials. The photocatalytic performances of C3N4–ZrP compositing nanomaterials with different mass ratios were studied by photodegradation of RhB under ultraviolet irradiation. All of the degradation rates of the C3N4–ZrP compositing nanomaterials system were achieved more than 90% after 18 min. When the mass ratio of C3N4–ZrP compositing nanomaterials is 2:1, the degradation efficiency achieved 99.95%, which is more efficient than other tested mass ratios. The result indicated the possibility of utilizing C3N4–ZrP compositing nanomaterials for environmental pollutants degradation.

Abstract

Amphiphilic graphene oxide (A-GO) with grafted octylamine was prepared via a one-step method of N,N′-dicyclohexylcarbodiimide coupling and epoxide ring opening at a mild temperature of 40 °C. The phase of oil–water emulsion stabilized by the complexes of A-GO and the cetyltrimethyl ammonium bromide (CTAB) surfactant could invert three times by adding CTAB or A-GO. This process was called triple phase inversion, which was a function of the concentration of A-GO or CTAB surfactants. The conductivity and zeta potential measurements confirmed that CTAB could influence the carboxyl ionization of A-GO. In addition, the turbidity of the A-GO and CTAB mixed dispersion system revealed that the appearance and disappearance of precipitation occurred when CTAB or A-GO concentration was increased. Meanwhile, the emulsion prepared by mixing the dispersion with an equal volume of xylene at a fixed CTAB concentration also showed triple phase inversion as the A-GO concentration varied. Results indicated that the stability of the emulsion and the size of the emulsion droplets had a certain correlation during the phase inversion process, which varied with the concentration of CTAB or A-GO.
