Bladder cancer (BC) is the most expensive cancer to manage on a per-patient basis, costing about $4 billion in total healthcare expenditure per annum in America alone. Therefore, identifying a natural compound for prevention of BC is of tremendous importance for managing this disease. Previous studies have identified isorhapontigenin (ISO) as having an 85% preventive effect against invasive BC formation induced by N-butyl-N-(4-hydroxybutyl) nitrosamine (BBN). The results showed here that ISO treatment inhibited EGF-induced cell transformation of human urothelial cells through induction of tumor suppressor p27 transcription secondary to activation of an E2F1-dependentpathway.ISOtreatmentrenderedcellsresistanttoEGF-induced anchorage-independent growth concurrent with p27 protein induction in both UROtsa and SV-HUC-1 cells. ISO inhibition of EGF-induced cell transformation could be completely reversed by knockdown of p27, indicating that this protein was essential for the noted ISO inhibitory action. Mechanistic studies revealed that ISO treatment resulted in increased expression of E2F1, which in turn bound to its binding site in p27 promoter and initiated p27 transcription. The E2F1 induction was due to the elevation of its translation caused by ISO-induced miR-205 downregulation. Consistently, miR-205 was found to be overexpressed in human BCs, and ectopic expression of miR-205 mitigated ISO inhibitory effects against EGF-induced outcomes. Collectively, the results here demonstrate that ISO exhibits its preventive effect on EGF-induced human urothelial cell transformation by induction of p27 through a miR-205/E2F1 axis. This is distinct from what has been described for the therapeutic effects of ISO on human BC cells.

To reduce the ripple of the magnetic flux and torque of motors and the reduce the hysteresis in motor speed control, an improved grey model predictive fuzzy direct torque control (DTC) method based on function transformation is proposed. First, a function transformation is used to transform the sampled sequences to nonnegative values. This overcomes the disadvantages caused by fluctuant and random sampling of the motor torque and stator flux linkage. Second, an equal dimensional new information model is used to keep the dimensions unchanged, which reduces the time to predict the motor parameters through the model. Moreover, the voltage space vector plane is divided into six sectors, which simplifies the fuzzy control system rules. Simulation results show that the proposed fuzzy direct torque control based on the improved grey model method reduces the influence of hysteresis on the control system, decreases the motor flux chain and torque ripple, improves the response speed of the torque and rotational speed, reduces overshoot, achieves good effects in terms of anti-interference capability and dynamic response, and improves the real-time performance and accuracy of the fuzzy control system.

We describe a kind of Zr(iv)-based nano-MOF that is well suited for specific adsorption and selective fluorescence sensing of phosphate based on specific interactions between Zr(iv) and phosphate which provides a new approach for treatment of hyperphosphatemia., Hyperphosphatemia is closely associated with the occurrence of multiple organ dysfunctions in patients with end-stage renal disease (ESRD). The application of phosphorus binders as an effective clinical approach for such diseases still suffers from serious side effects. Therefore, development of new phosphorus binders for the treatment of hyperphosphatemia remains a great challenge. Herein, we describe a kind of zirconium (Zr(iv))-based nano-MOF that is well suited for specific adsorption and selective fluorescence sensing of phosphate, and is based on the particular interactions between Zr(iv) and phosphate. The reduced levels of phosphate were quantitatively monitored using the MOF-based fluorescence nanosensor. Notably, the MOFs exhibit a greater reduction in phosphorus levels than commercially available phosphorus binders, and comparable therapeutic effects in the treatment of hyperphosphatemia of a mice model. Hence, the MOF acts as a promising medication for hyperphosphatemia by directly adsorbing phosphorus in the blood, which offers new perspectives in future applications of MOFs.

Urban power supply network plays a vital role in maintaining the city operation. The vulnerability of power supply network in the face of events has been one of significant concerns. This study presents a new methodology and framework for “dose-response” vulnerability assessment of urban power supply network. This framework can explore the vulnerability of power supply network under two types of events: random type and intentional type. It also integrates a new metric that calculates the vulnerability of power supply network in both structural dimension and functional dimension. Taking the power supply network of a city in east China as an example, network vulnerability under different types of events was assessed, and the “dose-response” interrelationships between network performance and event scale under different types of events were thoroughly discussed. The results demonstrated that power supply network was more vulnerable to intentional events in both dimensions. For intentional events, power supply network was more vulnerable to degree-based attack than to betweenness-based attack. After that, the redundancy coefficient α of power supply network was optimized. The conclusion and some suggestions for future research were given in the end.
